Obesity is rising globally and is associated with neurodevelopmental and psychiatric disorders among children, adolescents and young adults. Whether obesity is the cause or the consequence of these disorders remains unclear. To examine the behavioural effects of obesity systematically, locomotion, anxiety and social behaviour were assessed in male and female C57Bl/6J mice using the open field, elevated plus maze and social preference task. First, the effects of age and sex were examined in control mice, before investigating post-weaning consumption of a high fat-high sugar diet commonly consumed in human populations with high rates of obesity. In the open field and elevated plus maze, locomotor activity and anxiety-related behaviours reduced with aging in both sexes, but with different sex-specific profiles. The high fat-high sugar diet reduced food and calorie intake and increased body mass and fat deposition in both sexes. In the open field, both male and female mice on the obesogenic diet showed reduced locomotion; whereas, in the elevated plus maze, only females fed with the obesogenic diet displayed reduced anxiety-related behaviours. Both male and female mice on the obesogenic diet had a significantly higher social preference index than the control group. In conclusion, the findings demonstrate that the behavioural effects of age and diet-induced obesity all depend on the sex of the mouse. This emphasises the importance of considering the age of the animal and including both sexes when assessing behavioural phenotypes arising from dietary manipulations.

This thesis concerns the relationship between Kant and Durkheim as expressed in the philosophy of Charles Renouvier. His 'Neo-Criticisme' presents itself as both a retention and critique of the principles of Kant's 'Criticisme'. And it contains the theoretical premises of Durkheim's socioloay. The thesis traces the transformation of thought effected by Penouvier to Kant's Critical Philosophy. In particular it is claimed that the Copernicanism which marks the interpretation of human reality for Renouvier and Durkheim, has its oriqin in the philosophical revolution in epistemology and morality effected by Kant. Part One examines the historical and analytic preliminaries for the development of the ideas in Part II, III and IV. The development of Durkheim's sociology is seen as occurring during a period of thought marked by a return to Kant. Part One traces the development of Durkheim's idea of the sociology of knowledge as a form of post-Kantian Copernican science. The goal of this is to discover the principles involved in human knowledge: these are the categories which express the a priori in knowledge. For Durkheim the origin is social, whereas for Kant these principles are intellectual. Renouvier's critique of Kant Abstract 1 ~ ��� is seen as providing a transition between the two. Part III looks at the conflict of reasons in the Kantian system entailed by determinism and how Renouvier develops a non-deterministic conception of causality that is available to a human science, and through this reconciles the reasons split by Kant. This analysis reveals that for Renouvier and Durkheim in contradistinction to Kant, the necessity which governs all of human experience is a moral necessity. Part IV looks at the laws of the moral order: the principles of a Copernican morality are Duty, Unity and Autonomy

Background: The B-MaP-C study aimed to determine alterations to breast cancer (BC) management during the peak transmission period of the UK COVID-19 pandemic and the potential impact of these treatment decisions. Methods: This was a national cohort study of patients with early BC undergoing multidisciplinary team (MDT)-guided treatment recommendations during the pandemic, designated ‘standard’ or ‘COVID-altered’, in the preoperative, operative and post-operative setting. Findings: Of 3776 patients (from 64 UK units) in the study, 2246 (59%) had ‘COVID-altered’ management. ‘Bridging’ endocrine therapy was used (n = 951) where theatre capacity was reduced. There was increasing access to COVID-19 low-risk theatres during the study period (59%). In line with national guidance, immediate breast reconstruction was avoided (n = 299). Where adjuvant chemotherapy was omitted (n = 81), the median benefit was only 3% (IQR 2–9%) using ‘NHS Predict’. There was the rapid adoption of new evidence-based hypofractionated radiotherapy (n = 781, from 46 units). Only 14 patients (1%) tested positive for SARS-CoV-2 during their treatment journey. Conclusions: The majority of ‘COVID-altered’ management decisions were largely in line with pre-COVID evidence-based guidelines, implying that breast cancer survival outcomes are unlikely to be negatively impacted by the pandemic. However, in this study, the potential impact of delays to BC presentation or diagnosis remains unknown.

The normal function of the mammalian reproductive axis is strongly influenced by physiological, metabolic and environmental factors. Kisspeptin neuropeptides, encoded by the Kiss1 gene, are potent regulators of the mammalian reproductive axis by stimulating gonadodropin releasing hormone secretion from the hypothalamus. To understand how the reproductive axis is modulated by higher order neuronal inputs we have mapped the afferent circuits into arcuate (ARC) Kiss1 neurons. We used a transgenic mouse that expresses the CRE recombinase in Kiss1 neurons for conditional viral tracing with genetically modified viruses. CRE-mediated activation of these viruses in Kiss1 neurons allows the virus to move transynaptically to label neurons with primary or secondary afferent inputs into the Kiss1 neurons. Several regions of the brain showed synaptic connectivity to arcuate Kiss1 neurons including proopiomelanocortin neurons in the ARC itself, kisspeptin neurons in the anteroventral periventricular nucleus, vasopressin neurons in the supraoptic and suprachiasmatic nuclei, thyrotropin releasing neurons in the paraventricular nucleus and unidentified neurons in other regions including the subfornical organ, amygdala, interpeduncular nucleus, ventral premammilary nucleus, basal nucleus of stria terminalis and the visual, somatosensory and piriform regions of the cortex. These data provide an insight into how the activity of Kiss1 neurons may be regulated by metabolic signals and provide a detailed neuroanatomical map for future functional studies.

With the increasing popularity of herbomineral preparations in healthcare, a new proprietary herbomineral formulation was formulated with ashwagandha root extract and three minerals viz. zinc, magnesium, and selenium. The aim of the study was to evaluate the immunomodulatory potential of Biofield Energy Healing (The Trivedi Effect®) on the herbomineral formulation using murine splenocyte cells. The test formulation was divided into two parts. One was the control without the Biofield Energy Treatment. The other part was labelled the Biofield Energy Treated sample, which received the Biofield Energy Healing Treatment remotely by twenty renowned Biofield Energy Healers. Through MTT assay, all the test formulation concentrations from 0.00001053 to 10.53 µg/mL were found to be safe with cell viability ranging from 102.61% to 194.57% using splenocyte cells. The Biofield Treated test formulation showed a significant (p≤0.01) inhibition of TNF-α expression by 15.87%, 20.64%, 18.65%, and 20.34% at 0.00001053, 0.0001053, 0.01053, and 0.1053, µg/mL, respectively as compared to the vehicle control (VC) group. The level of TNF-α was reduced by 8.73%, 19.54%, and 14.19% at 0.001053, 0.01053, and 0.1053 µg/mL, respectively in the Biofield Treated test formulation compared to the untreated test formulation. The expression of IL-1β reduced by 22.08%, 23.69%, 23.00%, 16.33%, 25.76%, 16.10%, and 23.69% at 0.00001053, 0.0001053, 0.001053, 0.01053, 0.1053, 1.053 and 10.53 µg/mL, respectively compared to the VC. Additionally, the expression of MIP-1α significantly (p≤0.001) reduced by 13.35%, 22.96%, 25.11%, 22.71%, and 21.83% at 0.00001053, 0.0001053, 0.01053, 1.053, and 10.53 µg/mL, respectively in the Biofield Treated test formulation compared to the VC. The Biofield Treated test formulation significantly down-regulated the MIP-1α expression by 10.75%, 9.53%, 9.57%, and 10.87% at 0.00001053, 0.01053, 0.1053 and 1.053 µg/mL, respectively compared to the untreated test formulation. The results showed the IFN-γ expression was also significantly (p≤0.001) reduced by 39.16%, 40.34%, 27.57%, 26.06%, 42.53%, and 48.91% at 0.0001053, 0.001053, 0.01053, 0.1053, 1.053, and 10.53 µg/mL, respectively in the Biofield Treated test formulation compared to the VC. The Biofield Treated test formulation showed better suppression of IFN-γ expression by 15.46%, 13.78%, 17.14%, and 13.11% at concentrations 0.001053, 0.01053, 0.1053, and 10.53 µg/mL, respectively compared to the untreated test formulation. Overall, the results demonstrated that The Trivedi Effect®- Biofield Energy Healing (TEBEH) has the capacity to potentiate the immunomodulatory and anti-inflammatory activity of the test formulation. Biofield Energy may also be useful in organ transplants, anti-aging, and stress management by improving overall health and quality of life.

N-Methyl-d-aspartate receptors (NMDARs) are Ca(2+)-permeable glutamate receptors that play a critical role in synaptic plasticity and promoting cell survival. However, overactive NMDARs can trigger cell death signalling pathways and have been implicated in substantia nigra pars compacta (SNc) pathology in Parkinson's disease. Calcium ion influx through NMDARs recruits Ca(2+)-dependent proteins that can regulate NMDAR activity. The surface density of NMDARs can also be regulated dynamically in response to receptor activity via Ca(2+)-independent mechanisms. We have investigated the activity-dependent regulation of NMDARs in SNc dopaminergic neurones. Repeated whole-cell agonist applications resulted in a decline in the amplitude of NMDAR currents (current run-down) that was use dependent and not readily reversible. Run-down was reduced by increasing intracellular Ca(2+) buffering or by reducing Ca(2+) influx but did not appear to be mediated by the same regulatory proteins that cause Ca(2+)-dependent run-down in hippocampal neurones. The NMDAR current run-down may be mediated in part by a Ca(2+)-independent mechanism, because intracellular dialysis with a dynamin-inhibitory peptide reduced run-down, suggesting a role for clathrin-mediated endocytosis in the regulation of the surface density of receptors. Synaptic NMDARs were also subject to current run-down during repeated low-frequency synaptic stimulation in a Ca(2+)-dependent but dynamin-independent manner. Thus, we report, for the first time, regulation of NMDARs in SNc dopaminergic neurones by changes in intracellular Ca(2+) at both synaptic and extrasynaptic sites and provide evidence for activity-dependent changes in receptor trafficking. These mechanisms may contribute to intracellular Ca(2+) homeostasis in dopaminergic neurones by limiting Ca(2+) influx through the NMDAR.

Across the U.S., there is a paucity of mental health and substance abuse services for Deaf individuals. Without the availability of communication accommodations and specialized clinical expertise, Deaf individuals seeking behavioral health services contend with access limitations, misdiagnoses, and superficial treatments. Moreover, while the assessment and treatment of co-occurring disorders and the infusion of trauma-informed care are key components of effective treatment in the mainstream literature, the investigation and application of these initiatives to the Deaf population has not yet occurred. The proposed poster will outline the current state of specialized behavioral health services for Deaf individuals in Central Massachusetts, identify gaps in the continuum of behavioral health care for this population, and offer recommendations for future behavioral health programming. Currently, Central Massachusetts is home to two specialized programs for Deaf individuals seeking behavioral health services, the Center for Living and Working’s Deaf Independent Living Services and the DMH Inpatient Deaf Services at Worcester State Recovery Center and Hospital. Unfortunately, even with these programs, it is challenging to find appropriate outpatient placement and services that meet the range of psychiatric, intellectual, linguistic, and cultural needs of Deaf individuals. Connections between inpatient units and specialized outpatient services for the Deaf need to be developed or enhanced to increase continuity of care and reduce rehospitalization of Deaf individuals. At the current time, specialized outpatient psychotherapy and psychiatry services for Deaf individuals are not available in this region. Lack of funding for AA/NA interpreters and the recent closure of a Deaf-accessible substance abuse treatment agency highlight the need for the development of programs that provide accessible substance abuse treatment. Culturally-affirmative, linguistically-accessible, trauma-informed outpatient behavioral health services that are tailored to the Deaf population are vital to promoting the wellness and recovery of the Deaf community.
