Sweet potato yellow curling (YC), the most severe disease of sweet potato detected in Argentina, causes symptoms and damage to sweet potato crops in all cultivated regions. Since 2010/11, the presence of four viruses has been detected in symptomatic cv. Arapey INIA: two potyviruses non-persistently transmitted by Myzus persicae (sweet potato feathery mottle virus, SPFMV and sweet potato virus G, SPVG); a closterovirus, sweet potato chlorotic stunt virus (SPCSV) and a geminivirus, sweet potato leaf curl virus (SPLCV), both transmitted by Bemisia tabaci in a semi-persistent and persistent manner, respectively. All the plants were collected from fields in Colonia Caroya, Córdoba province, Argentina. The objectives of the present work are to isolate and identify the virus or viruses involved in YC disease of sweet potato, and to elucidate the viral combination that reproduces YC symptoms. The most severe YC symptoms for this genotype in the field were only reproduced by a combination of the four viruses. The symptoms include chlorosis, stunting, mosaic, blistering, leaf curling, chlorotic spots, chlorotic patterns, leaf area reduction and distortion, and upward curling of leaf edges. The presence of each virus was detected by serological (DAS, NCM and TAS-ELISA) and molecular (PCR) tests. It is concluded that the interaction of SPFMV, SPVG, SPCSV and SPLCV is needed for the development of YC symptoms. El encrespamiento amarillo (EA), la enfermedad más severa detectada en Argentina, causa síntomas y daños en cultivos de batata en toda la región productora. Desde 2010/11 se ha detectado la presencia de cuatro virus en plantas sintomáticas del cv. Arapey INIA recolectadas en lotes de Colonia Caroya, provincia de Córdoba. Los virus son sweet potato feathery mottle virus (SPFMV) y sweet potato virus G (SPVG), dos potyvirus transmitidos de forma no persistente por Myzus persicae; un closterovirus: sweet potato chlorotic stunt virus (SPCSV) y un geminivirus: sweet potato leaf curl virus (SPLCV), ambos transmitidos por Bemisia tacabi de manera semipersistente y persistente, respectivamente. Los objetivos de este trabajo fueron aislar e identificar el o los virus involucrados en la enfermedad EA de la batata y determinar la combinación de virus que reproduce la sintomatología de EA Solo la combinación de los cuatro virus permitió reproducir la sintomatología más severa del encrespamiento amarillo observada a campo en dicho genotipo. Los síntomas incluyen clorosis, achaparramiento, mosaico, ampollado, enrulado de la hoja, manchas cloróticas, diseños cloróticos, reducción y distorsión del área foliar, bordes de la hoja curvados hacia arriba. La presencia de cada uno de los virus se detectó mediante pruebas serológicas (DAS, NCM y TAS-ELISA) y moleculares (PCR). Potato stands first as a non-cereal food crop for global consumption purposes. During the year 2019–20, dry rot disease symptoms were observed on potato tubers of the cultivar ‘Kufri Pukhraj’ kept in a cold store in the Moga district of Punjab State of India. Initial symptoms were observed as wrinkled and shriveled tubers with necrotic lesions on the tuber skin. The cottony white mycelial growth was also evident on some cracked tubers. During the incubation of these tubers at 25 °C for 7 days, the symptoms were more prominent with intensive whitish mycelial growth throughout the tuber. The pure culture of isolated fungus showed sparsely white colonies, which turned violet after 2 weeks. The oval or club-shaped microconidia were abundantly present on monophialides and multiphialides. The slender or straight macroconidia having 3–5 septa were also observed. The long and short chains of microconidia were observed in the fresh colonies. The molecular identification based on translation elongation factor 1 α (TEF1 α) and internal transcribed spacer region primers revealed the fungus identity as Fusarium proliferatum. The pathogenicity test was also performed to prove the Koch postulates on the initially identified host cultivar. To the best of our knowledge, this is the first report of dry rot disease caused by Fusarium proliferatum in India.

In 2017, Xylella fastidiosa, a quarantine plant-pathogenic bacterium in Europe, was detected in almond trees associated with leaf scorch symptoms in Alicante, a Mediterranean area in southeastern mainland Spain. The bacterium was detected by serological and molecular techniques, isolated in axenic culture from diseased almond trees, and identified as X. fastidiosa subsp. multiplex sequence type (ST) 6. Inoculation experiments on almond plants in greenhouse trials with a characterized strain of X. fastidiosa subsp. multiplex ST6 isolated in the outbreak area have proved that it was able to multiply and systemically colonize inoculated plants. Disease symptoms characteristic of leaf scorch like those observed in the field were observed in the inoculated almond trees after 1 year. Furthermore, the pathogen was reisolated and identified by molecular tests. With the fulfillment of Koch’s postulates, we have demonstrated that X. fastidiosa is the causal agent of the almond leaf scorch disease in the Alicante outbreak.

Nannizziopsis guarroi is an ascomycete fungus associated with a necrotizing dermatitis in captive green iguanas (Iguana iguana) and bearded dragons (Pogona vitticeps) across both Europe and North America. Clinical signs of the disease include swelling and lesion formation. Lesions develop from white raised bumps on the skin and progress into crusty, yellow, discolored scales, eventually becoming necrotic. The clinical signs are the basis of a colloquial name yellow fungal disease (YFD). However, until now, N. guarroi has not been confirmed as the primary agent of the disease in bearded dragons. In this experiment, we fulfill Koch's postulates criteria of disease, demonstrating N. guarroi as the primary agent of YFD in bearded dragons.

In the 90's, pear blister canker viroid (PBCVd), apple dimple fruit viroid (ADFVd), peach latent mosaic viroid (PLMVd) and chrysanthemum chlorotic mottle viroid (CChMVd) were identified and characterized in the Ricardo Flores' laboratory. In these studies, the autonomous replication of these infectious RNAs and their involvement in the elicitation of diseases in their natural hosts were also shown. Their discovery was achieved by classical approaches based on the physical purification of the viroid RNAs from polyacrylamide gels followed by the sequencing of their genomic RNAs and by bioassays to assess their autonomous replication and the fulfillment of Koch's postulates. The molecular characterization of these four viroids, including the study of their sequence variability, contributed to the establishment of the concept of quasispecies for viroids and to the development of reliable molecular diagnostic methods that have facilitated the control of the diseases they caused. Most importantly, some of these viroids became valuable experimental model systems that are still used nowadays to study structural-functional relationships in RNAs and to dissect evolutionary and pathogenic pathways underlying plant-viroid interaction. The differences between early viroid discovery strategies, relying on biological and pathogenic issues, and the current high-throughput sequencing-based approaches, that frequently allow the discovery of new viroids and viroid-like RNAs in symptomless hosts, is also discussed, clarifying why the traditional molecular and biological studies mentioned above are still required to conclusively define the nature of any novel viroid-like RNA.

A devastating complex pathological condition involving a blend of symptoms such as profuse gum exudation from the stem and slow vine decline; sudden and complete death of vines; and foliar spots progressing into blight in bottle gourd fields was observed for the first time during the year Kharif 2013–14 in Indian Punjab. Over the last five years, this problem has become widespread to emerge as one of the major limitations for bottle gourd cultivation. Through systematic studies, the pathogenesis of three simultaneously occurring syndromes in bottle gourd viz. stem gummosis and slow vine decline; quick wilting and leaf blight were deciphered and their primary etiological causes were established. Pathogenicity tests were conducted to prove Koch postulates and identification of organisms was done through morphological as well as molecular studies. The causal organism of stem gummosis and slow vine decline was established as Fusarium verticillioides (Sacc.) Nirenberg. The pathogen associated with quick wilting was identified as Fusarium oxysporum Schltdl. and the pathogen associated with leaf blight was identified as Lasiodiplodia theobromae (Pat.) Griffon & Maubl. This is the first study to decipher three different diseases caused by three distinct pathogens leading to wilt or blight type symptoms.

Roots of plants characteristic of the Brazilian caatinga showing necrosis symptoms were observed in Iguatu, CE, Brazil. To identify the species, morphological characterization was performed, through the morphometry of females, and molecular analysis of the ITS and 28S rDNA regions. The nematodes Pratylenchus zeae and P. brachyurus were identified as causal agents, confirming pathogenicity by Koch postulates. This is the first report of P. zeae and P. brachyurus in caaatinga plants in the state of Ceará, Brazil.

Xylella fastidiosa (Xf) is an emerging insect-vectored, xylem-limited bacterium that can cause disease on several economically important fruit and tree crops including almond, blueberry, citrus, grapevine, peach, and pecan. On blueberry, Xf causes bacterial leaf scorch (BLS), which is prevalent in the southeastern United States. This disease, previously reported to be caused by Xf subsp. multiplex (Xfm), can result in rapid plant decline and death of southern highbush (SHB) blueberry cultivars. In 2017, a survey of blueberry plantings in southern Georgia (U.S.A.) confirmed the presence of Xf-infected plants in eight of nine sites examined, and seven isolates were cultured from infected plants. Genetic characterization of these isolates through single-locus and multilocus sequence analysis revealed that three isolates from two sites belonged to Xf subsp. fastidiosa (Xff), with significant similarity to isolates from grapevine. After these three isolates were artificially inoculated onto greenhouse-grown SHB blueberries (cv. ‘Rebel’), symptoms typical of BLS developed, and Xff infection was confirmed through genetic characterization and reisolation of the bacterium to fulfill Koch’s postulates. Because all previously reported Xf isolates from blueberry have been characterized as Xfm, this is the first time that isolation of Xff has been reported from naturally infected blueberry plantings. The potential impact of Xff isolates on disease management in blueberry requires further exploration. Furthermore, given that isolates from both Xfm and Xff were obtained within a single naturally infected blueberry planting, blueberry in southern Georgia may provide opportunities for intersubspecific recombination between Xff and Xfm isolates.

Awareness of the pathogenicity and aggressiveness nature of each pathogenic microbe is necessary not only to understand the expression of symptoms, but also contribute to the sheath blight disease control programme. The intensive survey of sheath blight infected farmers fields during the kharif 2015-16 at forty one locations of fifteen districts of Chhattisgarh plane were carried out and the disease samples were collected at maximum tillering stage of rice. Sheath blight and banded blight symptoms were examined for detail study. Naturally infected plant samples of sheath blight have been obtained from the fields in the laboratory for isolation and purification of sheath blight fungus. The pathogen was isolated on PDA and the subcultures were purified using a single hyphal tip cutting method and kept on PDA at 28 ± 2 °C. Samples infected with the sheath blight disease of paddy were collected from different districts of Chhattisgarh for systematic study of sheath blight symptoms in rice. The total number of fifty-eight isolates were confirmed by the proof of the Koch postulates and classified in the category as weakly aggressive, moderately aggressive, aggressive and highly aggressive. The seven isolates were recorded as highly aggressive and the maximum PDI 98.15 was shown by the isolate no. RS57.

Effective control disease measures require a great deal of knowledge about the pathogen, which is also applied to dieback disease of the nutmeg tree in Aceh Selatan District, Province of Aceh, Indonesia. The objectives of this study were to identify the causal agent of dieback disease on a nutmeg plantation and to assess the effect of drought stress on the development of dieback disease. Pathogen identification was carried out by observing the symptoms of infected plants, morphological characters of the isolated pathogens, Koch’s postulates, and molecular analysis and pure pathogen cultures. The samples were taken from an infected part of the nutmeg plant (twigs, stems, and roots) in 6 subdistricts at the center of the nutmeg plantation in Aceh Selatan. Koch's postulates were performed by using the mycelia of pathogens on 1-year-old nutmeg seedlings. Drought stress was simulated everyday, and then once every 2 weeks, to study the effect of drought stress on dieback. The results showed that there were 4 genera of fungal pathogens (Botryodiplodia theobromae, Fusarium solani, Graphium euwallaceae, and Rigidoporus microporus) out of 6 fungi associated with dieback disease. However, B. theobromae Pat. turned out to be the dominant fungi associated with dieback on the nutmeg tree in Aceh Selatan. Drought stress was proven to be a predisposing factor for explaining this problem epidemic in Aceh Selatan.

Purpose of Review Chronic diseases remain a daunting challenge for clinicians and researchers alike. While difficult to completely understand, most chronic diseases, including late-onset dementias, are thought to arise as an interplay between host genetic factors and environmental insults. One of the most diverse and ubiquitous environmental insults centers on infectious agents. Associations of infectious agents with late-onset dementia have taken on heightened importance, including our investigations of infection by the intracellular respiratory bacterium, Chlamydia pneumoniae (Cpn), in late-onset dementia of the Alzheimer’s type. Recent Findings Over the last two decades, the relationship of this infection to pathogenesis in late-onset dementia has become much clearer. This clarity has resulted from applying contemporary molecular genetic, biochemical, immunochemical, and cell culture techniques to analysis of human brains, animal models, and relevant in vitro cell culture systems. Data from these studies, taken in aggregate form, now can be applied to evaluation of proof of concept for causation of this infection with late-onset disease. In this evaluation, modifications to the original Koch postulates can be useful for elucidating causation. Summary All such relevant studies are outlined and summarized in this review, and they demonstrate the utility of applying modified Koch postulates to the etiology of late-onset dementia of the Alzheimer’s type. Regardless, it is clear that even with strong observational evidence, in combination with application of modifications of Koch’s postulates, we will not be able to conclusively state that Cpn infection is causative for disease pathogenesis in late-onset dementia. Moreover, this conclusion obtains as well for the putative causation of this condition by other pathogens, including herpes simplex virus type 1, Borrelia burgdorferi, and Porphyromonas gingivalis.

Curvularia includes plant pathogenic species with a worldwide distribution and a wide host range, particularly cereals and grass (Poaceae). Chinese fir (Cunninghamia lanceolata) is an important afforestation tree species in southern China, with a high economic value. Leaf blight disease on Chinese fir was discovered in Hunan and Zhejiang, China, and two fungal species were found to be associated with the symptoms. Using morphological and phylogenetic approaches, they were identified as Curvularia spicifera and Curvularia muehlenbeckiae. They were determined to be the pathogens by fulfilling Koch's postulates. Both showed a pathogenicity on Citrus reticulata and Citrus tangerina, which have a similar geographic distribution to Chinese fir. The similar geographic distribution of these hosts may increase the risk of disease. Based on a literature review, leaf blight caused by C. spicifera and C. muehlenbeckiae is recorded here on a gymnosperm for the first time.

Anthracnose is one of the major problems for cultivating many crops, including vegetables, fruits, and trees. It is a continual threat for fruits grower worldwide. Colletotrichum fructicola was isolated from Shine Muscat berries showing typical anthracnose symptom in Korea. It was identified as C. fructicola based on morphology, pathological signs and concatenated sequences of internal transcribed spacer region of rDNA, glyceraldehyde-3-phosphate dehydrogenase, β-tubulin-2, chitin synthase-1, calmodulin, and the Apn2-Mat1-2 intergenic spacer and partial mating type (Mat1-2) gene. To the best of our knowledge, this is the first report first report of anthracnose of Shine Muscat caused by C. fructicola in Korea.

La soya es una de las leguminosas más importantes en el mundo. En años recientes en el norte de Sinaloa se ha observado incidencia de mancha foliar en la etapa de llenado de grano e inicio de la madurez. El objetivo del presente trabajo fue identificar el agente causal de esta enfermedad. Se colectaron muestras de foliolos sintomáticos en ocho lotes comerciales de soya, de los cuales se obtuvieron ocho aislados fúngicos, y se identificaron morfológica y molecularmente. Se realizaron pruebas de patogenicidad en plantas de soya var. Nainari y se confirmaron los postulados de Koch, mediante el reaislamiento de los aislados originales. Los análisis morfológicos determinaron al género Curvularia asociado a la mancha foliar de la soya. El análisis filogenético del gen gadph permitió la identificación de Curvularia ahvazensis, C. muehlenbeckiae, C. spicifera y Curvularia sp. Estas especies resultaron patogénicas en soya var. Nainari mostrando variación en virulencia, pues el área foliar afectada varió de 3 a 31%. Los resultados abren nuevas líneas de investigación relacionadas con el impacto de la enfermedad en el rendimiento y calidad de la cosecha de la soya en Sinaloa.

One of the causal of the low rubber production is the presence of plant diseases. Important diseases in rubber plants generally cause symptoms of leaf fall, due to Colletotrichum spp., Corynespora sp., and Oidium sp. Currently, there is an incidence of new rubber leaf fall disease with symptoms that are different from the diseases previously found in rubber plantations. This disease is widespread in Sumatra, Kalimantan, and Southeast Asia. There are many suspicions about the pathogens that cause the disease, but there is no precise diagnosis. Therefore, an accurate diagnosis is needed as a basis for determining an effective and efficient disease control strategy. This research was carried out to determine the causal agents of leaf fall disease with symptoms of round leaf spots that can cause leaf fall. Diagnosis was carried out by isolating the pathogen from several rubber clones, inoculating the pathogen to healthy rubber plants, identifying morphologically and molecularly, and re-isolating from inoculated plants. The results of Koch's postulates and morphological and molecular identification determined that the causal agents of leaf fall disease at rubber plants with round spots was the fungus Pestalotiopsis microspora.

Advances in the science have promoted all aspects of human's life; these, in turn, have changed many principles and scientific postulates. Koch's postulates, since the beginning of their implementation, have been one of the important subjects involving complications and misinterpretations regarding the causal relationship of microbe-hosts. These postulates have been shown not to be correct in some cases including the inability of some microbes to grow in the culture medium, viruses, or anaerobic bacteria. Today, due to some new scientific facts like the social behaviors of bacteria, such as quorum sensing, there are serious problems regarding the definition of whole microbial effects; these include microbiomes and viromes, as well as their interaction with the existing eukaryotics, the complicated relations between bacteria, L-forms, and cell wall-deficient bacteria, and the important role of microbes in the development of non-infectious diseases. So, the application of Koch's postulates to explain the causal relationships between host-microbes could be difficult. Therefore, nowadays, even the molecular Koch's postulates are not accountable. Also, according to the new scientific discoveries, various criteria such as changes in the immune system, pathology, and clinical findings, along with the results of daily laboratory tests, should be used to apply Koch's postulates in the etiologic studies. Otherwise, the possible etiologic relationships between the host-microbes cannot be verified due to numerous complications; certainly, the relationship between the doctor and the lab is ultimately weakened. Therefore, public health, prevention, and much of the antimicrobial treatments will also remain in a state of ambiguity.

Para identificar el agente causal de la costra negra del nopal (Opuntia ficus-indica) se aislaron e identificaron morfológicamente los hongos asociados a la enfermedad. El estudio se realizó a partir de cladodios con síntomas característicos de la enfermedad colectados en la localidad de Santa Cecilia Clavijero, en el municipio de San Juan Ixcaquixtla, Puebla. Los hongos asociados a la enfermedad se identificaron como: Cladosporium cladosporioides, Aplosporella hesperidica y Didymella glomerata. Para demostrar su patogenicidad, los hongos se inocularon individualmente y en sus combinaciones en cladodios de seis meses de edad. Los síntomas iniciaron 50 días después de la inoculación. El método de inoculación que permitió la reproducción de los síntomas fue la inyección de una suspensión a una concentración de 6×103 conidios mL?1 de la mezcla de los tres hongos; cladodios inoculados con uno y dos hongos, no resultó en el desarrollo de la enfermedad. C. cladosporioides, A. hesperidica y D. glomerata son los agentes causales de la costra negra del nopal siendo este el primer reporte de estos patógenos en el cultivo del nopal.

In India, begomovirus infection causing tomato leaf curl disease (ToLCD) is a major constraint for tomato productivity. Here, we have identified two distinct monopartite begomovirus and betasatellite complexes causing ToLCD in the western part of India. A new monopartite begomovirus (Tomato leaf curl Mumbai virus, ToLCMumV) and betasatellite (Tomato leaf curl Mumbai betasatellite, ToLCMumB) were isolated from the Mumbai sample. A distinct Tomato leaf curl Gandhinagar virus (ToLCGanV) and Tomato leaf curl Gandhinagar betasatellite (ToLCGanB) were identified from the Gandhinagar sample. Both of the cloned begomoviruses were recombinants. The demonstration of systemic infection caused by begomovirus (ToLCGanV or ToLCMumV) alone in N. benthamiana and tomato (a virus resistant variety) emphasizes that they were monopartite begomoviruses. Co-inoculation of cognate begomovirus and betasatellite reduces the incubation period and increases symptom severity. Thus, Koch’s postulates were satisfied for these virus complexes. Further, an enhanced accumulation of ToLCGanV was detected in the presence of cognate ToLCGanB, however ToLCMumB did not influence the level of ToLCMumV in the agro-inoculated tomato plants. Our results indicate that the cloned viruses form potential virus resistance breaking disease complexes in India. This necessitates to investigate the spread of these disease complexes to major tomato growing regions in the country.

Purpose Aphis gossypii and A. punicae are the most damaging pests. The emergence of large populations has created concern among farmers because this pest complex is considered critical as it has a significant effect on major crops around the globe. The lack of new technologies for the control of A. gossypii and A. punicae is also worrying due to the indiscriminate use of chemical insecticides. Besides, this leads to the rapid development of resistance, which strangles their control in the field. Hence, there is a dire need to find the effective biocontrol agent for the management of Aphis gossypii and A. punicae. Methods The present investigation emphasizes the isolation and characterization of Aphidicidal Bacillus thuringiensis from the Andaman and Nicobar Islands, Karnataka, Assam, Arunachal Pradesh, and Sikkim soil samples. Phase contrast and scanning electron microscopy analysis used to characterize and identify the crystal morphology. Molecular profiling of Bt cry genes was determined by PCR using aphidicidal cry gene-specific primers, and molecular cloning and sequencing were carried out. Protein profiling by SDS–PAGE analysis was further studied. Finally, a qualitative bioassay of insecticidal activity was carried out against A. gossypii and A. punicae. Results A total of 65 Bacillus-like colonies were screened; 15 putative Aphidicidal B. thuringiensis isolates were identified based on morphological as well as through microbiological studies. Spherical and amorphous crystal inclusion was predominantly present in 34.28% of the Bt isolates. Crystal protein profiling of Bt isolates by SDS–PAGE analysis showed the presence 130, 73, 34, 25, and 13 kDa bands, among which 50−66 kDa bands were present abundantly. The detection of the cry gene of these isolates was done by PCR analysis, which indicated that cry1, cry2A, cry3A, and cry11A were on plasmid DNA. All cry genes were 80–100% homologous when aligned on alignment tool NCBI-BLASTn and BLASTp. All isolates of Bt were tested for their insecticidal activity against aphids. Three of the 15 isolates are Aphidicidal toxin specific by PCR analysis which were observed to be toxic to Aphis gossypii and A. punicae at a concentration of 35 μg/mL. The observed physical changes were induced by B. thuringiensis infection; these strains had been re-isolated from the dead aphids, and the presented results fulfilled Koch’s postulates. Conclusion The present study brought promising Bt isolates, primarily capable of creating an efficient biocontrol agent for Aphis gossypii and A. punicae and various sucking pests soon.

Mycosphaerella linicola is the causal agent of pasmo, a disease affecting flax (Linum usitatissimum) worldwide. We performed a three-year field survey in France to characterize the dynamics of sexual reproduction in this fungal pathogen on flax straw during the interepidemic period. Cohorts of M. linicola fruiting bodies were sampled from linseed straw during the autumn and winter and carefully observed, focusing on pseudothecia, asci and ascospores. A sequence of experimental steps corresponding to Koch s postulates confirmed, for the first time in France and continental Europe, the widespread presence of the sexual stage of M. linicola in plant host tissues, and its potential role in the disease cycle. The developmental dynamics of pseudothecia on straw, expressed as the change over time in the percentage of mature pseudothecia, was similar in all three years. Pseudothecia appeared in late summer, with peak maturity reached in October. A temporal shift, thought to be due to the climatic conditions in early autumn (rainfall), was highlighted in one of the three years. These observations suggest that sexual reproduction plays a significant role in the epidemiology of pasmo in France. A resurgence of M. linicola infections in spring flax is thought to have occurred in recent years, due to the increase in the area under flax. The presence of the sexual stage of this pathogen probably increased the quantitative impact of residues of winter linseed (used for oil) and flax straw (left on the soil for retting and used for fibers) as an interepidemic "brown bridge". This case study highlights how certain parts of a disease cycle, in this case the sexual phase, can become crucial due to changes in production conditions. Finally, we highlight the importance of further studies of the specific detrimental effects of the sexual stage of this pathogen on flax fiber quality.

In March 2019, the California Animal Health and Food Safety Laboratory (CAHFS), Turlock branch, received two submissions of broiler chickens from commercial flocks reporting increased mortality. Submissions consisted of either white or brown broilers. Submitted chickens appeared depressed with ruffled feathers. At necropsy, moderate to severely enlarged and pale kidneys were observed, with gross lesions indicative of dehydration. Microscopically, renal tubules were degenerated and distended with necrotic debris and tubular casts. The kidney parenchyma contained mononuclear inflammatory cell infiltrates and interstitial edema. Infectious bronchitis virus (IBV) was isolated and identified by reverse transcription quantitative PCR from kidney tissue pools and tracheal swab pools from both cases. Partial sequencing of the S1 hypervariable region was most similar to a local California variant, CA1737. The outbreak lasted roughly 1 wk in both flocks, with 2% total mortality in the brown broilers and 20% total mortality in the white broilers. Final proof of the IBV strains causing nephropathy will require fulfillment of Koch postulates. IBV associated with nephropathy has been sporadically reported in California chicken flocks and represents a significant pathogen due to its potential for inducing high flock mortality. The incidence of IBV associated with a nephropathy diagnosis in chicken necropsy submissions to the CAHFS system-wide from 1998 to 2019 is also reviewed.

Despite the detection of Neorickettsia species DNA sequences in various trematode species and their hosts, only three Neorickettsia species have been cell culture isolated and whole-genome sequenced and are known to infect mammals and/or cause disease. The molecular mechanisms that enable the obligatory intracellular bacterium Neorickettsia to colonize trematodes and to horizontally transmit from trematodes to mammals, as well as the virulence factors associated with specific mammalian hosts, are unknown. Potomac horse fever (PHF) is a severe and acute systemic infectious disease of horses, with clinical signs that include diarrhea. Neorickettsia risticii is the only known bacterial species that causes PHF. Ingestion of insects harboring N. risticii-infected trematodes by horses leads to PHF. Our discovery of a new Neorickettsia species that causes PHF and whole-genome sequence analysis of this bacterium will improve laboratory diagnosis and vaccine development for PHF and will contribute to our understanding of Neorickettsia ecology, pathogenesis, and biology., Potomac horse fever (PHF), a severe and frequently fatal febrile diarrheal disease, has been known to be caused only by Neorickettsia risticii, an endosymbiont of digenean trematodes. Here, we report the cell culture isolation of a new Neorickettsia species found in two locations in eastern Ontario, Canada, in 2016 and 2017 (in addition to 10 variable strains of N. risticii) from N. risticii PCR-negative horses with clinical signs of PHF. Gene sequences of 16S rRNA and the major surface antigen P51 of this new Neorickettsia species were distinct from those of all previously characterized N. risticii strains and Neorickettsia species, except for those from an uncharacterized Neorickettsia species culture isolate from a horse with PHF in northern Ohio in 1991. The new Neorickettsia species nonetheless had the characteristic intramolecular repeats within strain-specific antigen 3 (Ssa3), which were found in all sequenced Ssa3s of N. risticii strains. Experimental inoculation of two naive ponies with the new Neorickettsia species produced severe and subclinical PHF, respectively, and the bacteria were reisolated from both of them, fulfilling Koch’s postulates. Serological assay titers against the new Neorickettsia species were higher than those against N. risticii. Whole-genome sequence analysis of the new Neorickettsia species revealed unique features of this bacterium compared with N. risticii. We propose to classify this new bacterium as Neorickettsia finleia sp. nov. This finding will improve the laboratory diagnosis of and vaccine for PHF, environmental risk assessment of PHF, and understanding of PHF pathogenesis and Neorickettsia biology in general.

In Nov 2020, mass mortality of cultured Pacific white shrimp (Penaeus vannamei) occurred due to black spot disease (BSD) in some shrimp farms in China. To investigate the disease cause, a potential pathogenic fungus was isolated from melanized lesions of P. vannamei suffering from the BSD syndrome. The fungal isolate was then identified as Fusarium solani based on TEF1α and ITS-gene sequences. The isolate of fungus was used in infection studies. This study used Koch postulates confirmed that the studied agent was causing BSD symptoms in the white leg shrimp, which were significantly symptoms relieved after molting. Results showed that an identical fungus was re-isolated from black lesions after the artificial infection period. Virulence test revealed that F. solani could cause 88.66% mortality within 30 dpi at a dose of 1.02 × 106 CFU/mL, and the LC50 of F. solani at 30 dpi is 3.37 × 104 CFU/mL. The accumulative mortalities also displayed concentration dependence. The histopathological characterization displayed that responses of P. vannamei against F. solani are haemocytic infiltration, encapsulation, melanization, etc. in the black spot lesions. Furthermore, severe infection of F. solani in P. vannamei causes molting failure, and further contributes to the mortality of the diseased individuals. The findings of our study would be useful to provide a theoretical basis for early prevention and control of BSD.

Koch's postulates are regularly included in the lecture portion of microbiology courses, but rarely are they demonstrated in a microbiology teaching lab. This is understandable given the logistical challenges of undergraduates working with pathogenic bacteria, ethical concerns using animals, and limited time constraints of a weekly lab period. Here we present a cost-effective, time-friendly lab activity that demonstrates the principles of microbial isolation and infection assays that are part of fulfilling Koch's postulates. The disease is “peep pox” caused by a gelatinase-positive bacterial species hydrolyzing marshmallow peeps that proxy as infected animals.

Medical practice is increasingly coming under the guidance of statistical-mathematical models that are, undoubtedly, valuable tools but are also only a partial representation of reality. Indeed, given that statistics may be more or less adequate, a model is still a subjective interpretation of the researcher and is also influenced by the historical context in which it operates. From this opinion, I will provide a short historical excursus that retraces the advent of probabilistic medicine as a long process that has a beginning that should be sought in the discovery of the complexity of disease. By supporting the belonging of this evolution to the scientific domain it is also acknowledged that the underlying model can be imperfect or fallible and, therefore, confutable as any product of science. Indeed, it seems non-trivial here to recover these concepts, especially today where clinical decisions are entrusted to practical guidelines, which are a hybrid product resulting from the aggregation of multiple perspectives, including the probabilistic approach, to disease. Finally, before the advent of precision medicine, by limiting the use of guidelines to the original consultative context, an aged approach is supported, namely, a relationship with the individual patient.

The following are the responses to the “letter to the editor” (“Helicobacter is preserved in yeast vacuoles! Does Koch’s postulates confirm it?”) authored by Nader Alipour and Nasrin Gaeini that rejected the methods, results, discussions and conclusions summarized in the review article authored by Siavoshi F and Saniee P. In the article, 7 papers, published between 1998 and 2013, were reviewed. The 7 papers had been reviewed and judged very carefully by the assigned expertise of the journals involved, including the reviewers of the World Journal of Gastroenterology (WJG), before publication. In the review article, 121 references were used to verify the methods, results and discussions of these 7 papers. The review article was edited by the trustworthy British editor of the (WJG), and the final version was rechecked and finally accepted by the reviewers of (WJG). None of the reviewers made comments like those in this “letter to the editor”, especially the humorous comments, which seem unprofessional and nonscientific. Above all, the authors’ comments show a lack of understanding of basic and advanced microbiology, e.g. bacterial endosymbiosis in eukaryotic cells. Accordingly, their comments all through the letter contain misconceptions. The comments are mostly based on personal conclusions, without any scientific support. It would have been beneficial if the letter had been reviewed by the reviewers of the article by Siavoshi and Saniee.

Camellia japonica is a native tree species with high economic value that is widely cultivated in southern China. In recent years, canker disease has been observed in camellia plantations in Zhejiang Province, China, with the disease incidence rate in some plantations exceeding 20%. Canker disease severely affects the trunks and branches of C. japonica in China, but the causal agent has not yet been identified. In this study, the pathogen was isolated from infected C. japonica tissues through a conventional tissue isolation approach. Species identification was conducted using morphological methods combined with multilocus phylogenetic analysis. Pathogenicity was tested based on Koch’s postulates. The results showed that the pathogen could be isolated from the diseased bark of C. japonica ‘Hongluzhen’. The pathogen was identified as Nectria pseudotrichia based on morphological, cultural, and molecular traits. The inoculation of the pathogen into C. japonica ‘Hongluzhen’ caused necrotic lesions on healthy seedlings, and the fungus N. pseudotrichia could be re-isolated from such lesions. Therefore, N. pseudotrichia is the causal agent of canker disease affecting C. japonica in China.

1. Infectious diseases have a large impact on poultry health and economics. Elucidating the pathogenesis of a certain disease is crucial to implement control strategies. 2. Multiplication of a pathogen and its characterisation in vitro are basic requirements to perform experimental studies. However, passaging of the pathogen in vitro can influence the pathogenicity, a process targeted for live vaccine development, but limits the reproduction of clinical signs. 3. Numerous factors can influence the outcome of experimental infections with some importance on the pathogen, application route and host as exemplarily outlined for Histomonas meleagridis, Gallibacterium anatis and fowl aviadenoviruses (FAdVs). 4. In future, more comprehensive and detailed settings are needed to obtain as much information as possible from animal experiments. Processing of samples with modern diagnostic tools provides the option to closely monitor the host–pathogen interaction.

It is a common laboratory practice to propagate viruses in cell culture. While convenient, these methodologies often result in unintentional genetic alterations, which have lead to adaptation and even attenuation in animal models of disease. An example is the attenuation of hantaviruses (family: Bunyaviridae, genus: Hantavirus) when cultured in vitro. In this case, viruses propagated in the natural reservoir species cause disease in nonhuman primates that closely mimics the human disease, but passaging in cell culture attenuates these viruses to the extent that do not cause any measurable disease in nonhuman primates. As efforts to develop animal models progress, it will be important to take into account the influences that culture in vitro may have on the virulence of viruses. In this review we discuss this phenomenon in the context of past and recent examples in the published literature.

Human microbiome research makes causal connections between entire microbial communities and a wide array of traits that range from physiological diseases to psychological states. To evaluate these causal claims, we first examine a well-known single-microbe causal explanation: of Helicobacter pylori causing ulcers. This apparently straightforward causal explanation is not so simple, however. It does not achieve a key explanatory standard in microbiology, of Koch’s postulates, which rely on manipulations of single-microorganism cultures to infer causal relationships to disease. When Koch’s postulates are framed by an interventionist causal framework, it is clearer what the H. pylori explanation achieves and where its explanatory strengths lie. After assessing this ‘simple’, single-microbe case, we apply the interventionist framework to two key areas of microbiome research, in which obesity and mental health states are purportedly explained by microbiomes. Despite the experimental data available, interventionist criteria for explanation show that many of the causal claims generated by microbiome research are weak or misleading. We focus on the stability, specificity and proportionality of proposed microbiome causal explanations, and evaluate how effectively these dimensions of causal explanation are achieved in some promising avenues of research. We suggest some conceptual and explanatory strategies to improve how causal claims about microbiomes are made.

Foliage symptoms on soybean resembling those of sudden death syndrome were detected in Argentina during 1991 and 1992 in the Pampas Region and during 1992 and 1993 in the Northwest Region. Isolations were made in 1999, 2000, and 2001 from soybean plants (Glycine max (L.) Merr.) showing these symptoms. Five isolates of fungi obtained from taproot tissue and blue sporulation on taproot exteriors were selected for further evaluation. These isolates were plated on potato dextrose agar supplemented with streptomycin (PDAS). Based on the spore morphology, colony growth rate, morphology and pigmentation on PDAS, and lack of microconidia (1) five isolates were identified as Fusarium solani f. sp. glycines. Soybean cvs. Ripley, Spencer, Pioneer 9492RR, and A6445 RG were inoculated in greenhouse tests with each of the isolates using toothpick and soil infestation methods for a total of six experiments. Isolate 171 provided by J. Rupe (University of Arkansas, Fayetteville) was tested as a positive control. Foliar symptoms typical of sudden death syndrome and similar to those in the field were observed 14 and 25 days, respectively, after inoculations using the toothpick and soil infestation methods. Lesions produced on leaves averaged 3.6 for all five isolates and 4 for the reference strain using a disease severity scale where: 1 = no symptoms; 2 = slight symptom development with mottling and mosaic on leaves (1 to 20% foliage affected); 3 = moderate symptom development with interveinal chlorosis and necrosis on foliage (21 to 50% foliage affected); 4 = heavy symptom development with interveinal chlorosis and necrosis (51 to 80% foliage affected); and 5 = severe interveinal chlorosis and necrosis (81 to 100% foliage affected). Noninoculated controls were symptomless. Differences in virulence were observed among the isolates. Based on disease symptoms in the greenhouse and cultural morphology on PDAS, the isolates were classified as Fusarium solani f. sp. glycines. Isolates recovered from symptomatic plants resembled Fusarium solani f. sp. glycines on PDAS and peptone/p-chloro-nitrobenzene agar amended with streptomycin, confirming Koch's postulates. Fusarium solani f. sp. glycines was recovered from 60% of inoculated plants. Reference: (1) K. W. Roy et al. Plant Dis. 81:1100,1997.

Sadly, despite the discovery of the tuberculosis bacillus over a century ago by Robert Koch, tuberculosis remains a major killer and modern day plague. Progress in the eradication of tuberculosis has been very slow and will require determined efforts on multiple fronts to make substantial inroads to lower the currently stagnant incidence of around 2% globally.

As we learn more and more about the classes of organisms that infect humans, we are discovering that many organisms, including pathogenic organisms, may have a complex relationship with humans in which infection seldom results in the production disease. In some cases, infection may be just one biological event that occurs during a multievent process that develops sequentially, over time, and involves genetic and environmental factors that may vary among individuals. Consequently, the role of infectious organisms in the development of human disease may not meet all of the criteria normally required to determine when an organism can be called the cause of a disease. This chapter reviews the expanding role of infections in the development of human disease. We discuss prion diseases of humans, a fascinating example of an infectious disease-causing agent that is not a living organism. We also discuss the diseases of unknown etiology for which infectious organisms may play a role. In addition, this chapter reviews some of the misconceptions and recurring errors associated with the classification of infectious diseases that have led to misdiagnoses and have impeded our understanding of the role of organisms in the development of human diseases.

We argue that Koch's postulates are best understood within an interventionist account of causation, in the sense described in Woodward (2003). We show how this treatment helps to resolve interpretive puzzles associated with Koch's work and how it clarifies the different roles the postulates play in providing useful, yet not universal criteria for disease causation. Our paper is an effort at rational reconstruction; we attempt to show how Koch's postulates and reasoning make sense and are normatively justified within an interventionist framework and more difficult to understand within alternative frameworks for thinking about causation.

Colletotrichum truncatum was for the first time found to cause severe anthracnose of Chinese flowering cabbage (Brassica parachinensis L. H. Bailey) (Cruciferae), in Guangdong province, China. Identification of this Colletotrichum species with curved conidia was based on morphological and cultural characteristics and multi-gene phylogenetic analysis, which showed that this fungus belongs to C. truncatum. Artificially inoculated seedlings of Chinese flowering cabbage showed typical symptoms of anthracnose similar to those observed in the field. To understand the cross-pathogenicity of C. truncatum isolates infecting Chinese flowering cabbage and soybean, artificial inoculations on both hosts were carried out and the results indicated that the isolates of Chinese flowering cabbage could infect soybean, and vice versa. C. truncatum was consistently re-isolated from the artificially infected tissues of both hosts. Furthermore, a comparison of the effects of environmental factors on C. truncatum isolates CBP001 and XST01 from Chinese flowering cabbage and soybean, respectively, demonstrated that these two isolates have similar biological characteristics. Therefore, it is concluded that C. truncatum is the causal agent of anthracnose of Chinese flowering cabbage.

The objective of this study was to investigate a role of a recently discovered marsupial nidovirus in the development of a neurological disease, termed wobbly possum disease (WPD), in the Australian brushtail possum (Trichosurus vulpecula). Four possums received 1 mL of a standard inoculum that had been prepared from tissues of WPD-affected possums, 4 possums received 1.8 mL (1 × 10(6) TCID50) of a cell lysate from inoculated cultures, and 4 possums received 1 mL (× 10(7) TCID50) of a purified WPD isolate. All but one possum that received infectious inocula developed neurological disease and histopathological lesions characteristic for WPD. High levels of viral RNA were detected in livers from all possums that received infectious inocula, but not from control possums. Altogether, our data provide strong experimental evidence for the causative involvement of WPD virus in development of a neurological disease in infected animals.

This study aimed to demonstrate the association of the ash dieback pathogen Hymenoscyphus fraxineus with leaf symptoms on Fraxinus excelsior and to test its pathogenicity towards leaves of three European ash species, F. excelsior, F. angustifolia and F. ornus, in wound inoculation experiments. On F. excelsior, H. fraxineus was isolated from 94% of leaf rachises with necrotic lesions and from 74% of necrotic leaflet midribs. Following wound inoculation of leaf rachises, in two separate experiments performed in 2010 and 2011, the ash dieback pathogen caused symptoms (necrotic rachis lesions, leaf wilting and premature leaf shedding) on all three ash species, while control leaves remained symptomless. Hymenoscyphus fraxineus was consistently reisolated from fungus‐inoculated rachises. All 10 isolates tested were pathogenic to the three ash species and varied in virulence. Koch's postulates for H. fraxineus as causal agent of leaf symptoms on F. excelsior were fulfilled in this study. Complemented with the isolation of the fungus from naturally infected, symptomatic leaf rachises of F. angustifolia and F. ornus in previous investigations, H. fraxineus was confirmed to be a leaf pathogen of these ash species as well. The leaf inoculation experiments showed that F. excelsior was highly susceptible to H. fraxineus, F. angustifolia was equally or slightly less susceptible, whereas F. ornus was the least affected species; however, F. ornus should also be regarded as a host tree for the ash dieback pathogen. This susceptibility ranking corresponds well with field observations and previous stem inoculation experiments.

Glycoscience-based research that is performed expressly to address medical necessity and improve patient outcomes is called “translational glycobiology”. In the 19th century, Robert Koch proposed a set of postulates to rigorously establish causality in microbial pathogenesis, and these postulates can be reshaped to guide knowledge into how naturally-expressed glycoconjugates direct molecular processes critical to human well-being. Studies in the 1990s indicated that E-selectin, an endothelial lectin that binds sialofucosylated carbohydrate determinants, is constitutively expressed on marrow microvessels, and investigations in my laboratory indicated that human hematopoietic stem cells (HSCs) uniquely express high levels of a specialized glycoform of CD44 called “hematopoietic cell E-/L-selectin ligand” (HCELL) that functions as a highly potent E-selectin ligand. To assess the role of HCELL in directing HSC migration to marrow, a method called “glycosyltransferase-programmed stereosubstitution” (GPS) was developed to custom-modify CD44 glycans to enforce HCELL expression on viable cell surfaces. Human mesenchymal stem cells (MSCs) are devoid of E-selectin ligands, but GPS-based glycoengineering of CD44 on MSCs licenses homing of these cells to marrow in vivo, providing direct evidence that HCELL serves as a “bone marrow homing receptor”. This review will discuss the molecular basis of cell migration in historical context, will describe the discovery of HCELL and its function as the bone marrow homing receptor, and will inform on how glycoengineering of CD44 serves as a model for adapting Koch's postulates to elucidate the key roles that glycoconjugates play in human biology and for realizing the immense impact of translational glycobiology in clinical medicine.

Since the 1950s, medical communities have been facing with emerging and reemerging infectious diseases, and emerging pathogens are now considered to be a major microbiologic public health threat. In this review, we focus on bacterial emerging diseases and explore factors involved in their emergence as well as future challenges. We identified 26 major emerging and reemerging infectious diseases of bacterial origin; most of them originated either from an animal and are considered to be zoonoses or from water sources. Major contributing factors in the emergence of these bacterial infections are: (1) development of new diagnostic tools, such as improvements in culture methods, development of molecular techniques and implementation of mass spectrometry in microbiology; (2) increase in human exposure to bacterial pathogens as a result of sociodemographic and environmental changes; and (3) emergence of more virulent bacterial strains and opportunistic infections, especially affecting immunocompromised populations. A precise definition of their implications in human disease is challenging and requires the comprehensive integration of microbiological, clinical and epidemiologic aspects as well as the use of experimental models. It is now urgent to allocate financial resources to gather international data to provide a better understanding of the clinical relevance of these waterborne and zoonotic emerging diseases.

Cassava is consumed in the Democratic Republic of Congo (DRC) as a staple food for the majority of the Congolese population. This crop is used in several forms: as fufu, chikwangue and pondu; cassava leaves are the most consumed vegetable in the country. In 2002, cassava root symptoms similar to cassava brown streak disease (CBSD) were reported for the first time in western DRC. PCR assays, using primers specific to Cassava brown streak virus (CBSV), failed to detect or identify any viral pathogens in diseased cassava samples from western DRC. Therefore, next generation sequencing (NGS) techniques were used as they are able to sequence full organism genomes and are widely used for the identification of pathogens responsible for new diseases. The main objective of this study was to identify the pathogens causing root necrosis in western DRC. Whatman®FTA™ cards were used to collect 12 cassava leaf samples from plants with symptoms indicative of very severe root necrosis, as well as two asymptomatic samples. These 12 samples were sent to Australia at the University of Western Australia in Perth for next generation sequencing (NGS) using the Illumina HiSeq platform. Additional bioinformatics tools included Geneious, CLC workbench, ParaKraken and Kaijou software for short DNA sequences. No viruses (including CBSV) were found in any of the DRC samples. These preliminary results confirm all the previous negative results obtained using PCR and CBSV primers. However, NGS analyses did reveal the presence of a number of bacterial and fungal taxa. These will require further investigation and tests such as the Koch Postulates, to establish their specific pathogenic role in cassava. This is the first scientific evidence that no currently known virus is responsible for the disease which had been referred to previously as ‘CBSD-like disease’. Consequently, the disease found in DRC cassava samples has been designated ‘Cassava Root Necrosis Disease’ or CRND.

The human pathogen Acinetobacter baumannii has emerged as a frequent cause of hospital-acquired infections, but infection of animals has rarely been observed. Here we analyzed an outbreak of epidemic pneumonia killing hundreds of sheep on a farm in Pakistan and identified A. baumannii as the infecting agent. A pure culture of strain AbPK1 isolated from lungs of sick animals was inoculated into healthy sheep, which subsequently developed similar disease symptoms. Bacteria re-isolated from the infected animals were shown to be identical to the inoculum, fulfilling Koch’s postulates. Comparison of the AbPK1 genome against 2283 A. baumannii genomes from the NCBI database revealed that AbPK1 carries genes for unusual surface structures, including a unique composition of iron acquisition genes, genes for O-antigen synthesis and sialic acid-specific acetylases of cell-surface carbohydrates that could enable immune evasion. Several of these unusual and otherwise rarely present genes were also identified in genomes of phylogenetically unrelated A. baumannii isolates from combat-wounded US military from Afghanistan indicating a common gene pool in this geographical region. Based on core genome MLST this virulent isolate represents a newly emerging lineage of Global Clone 2, suggesting a human source for this disease outbreak. The observed epidemic, direct transmission from sheep to sheep, which is highly unusual for A. baumannii, has important consequences for human and animal health. First, direct animal-to-animal transmission facilitates fast spread of pathogen and disease in the flock. Second, it may establish a stable ecological niche and subsequent spread in a new host. El consumo del zapote mante (Pouteria campechiana) en México se ha incrementado, siendo el municipio de El Mante, Tamaulipas, uno de los principales productores. En un huerto no comercial se observaron en frutos manchas hundidas de color marrón que coalescen. El objetivo de este estudio fue identificar el agente causal de la mancha del fruto de zapote mante. Con los hongos aislados se realizaron los postulados de Koch, se amplificó y secuencio la región ITS del ADNr mediante PCR utilizando los iniciadores universales ITS1-ITS4. El análisis morfológico y molecular indicó que Cladosporium cladosporioides es el agente causal de la mancha del fruto. Hasta donde se sabe, este es el primer reporte de este patógeno afectando a zapote mante en México.

This research aimed to identify the type of fungus that causes the appearance of spots on the leaves of pokem. Descriptive method were used in this research with laboratory observation technique which consisted by preparation, sampling of pokem leaf, isolation, purification, innoculation and the observation of pathogen character. The result showed that the pathogen causing pokem (Setaria italica L.) leaf spot was a fungal pathogen Cercospora jenseana, f. Sp. Setariae which can be seen from the symptoms of the plant through the koch postulate test and the identification of color, shape and character of the colony, mycelium, hyphae, and spores.
