The detailed analysis and structural characterization of proteoforms by top-down proteomics (TDP) has gained a lot of interest in biomedical research. Data-dependent acquisition (DDA) of intact proteins is non-trivial due to the diversity and complexity of proteoforms. Dedicated acquisition methods thus have the potential to greatly improve TDP. Here, we present FLASHIda, an intelligent online data acquisition algorithm for TDP that ensures the real-time selection of high-quality precursors of diverse proteoforms. FLASHIda combines fast charge deconvolution algorithms and machine learning-based quality assessment for optimal precursor selection. In an analysis of E. coli lysate, FLASHIda increases the number of unique proteoform level identifications from 800 to 1500 or generates a near-identical number of identifications in one third of the instrument time when compared to standard DDA mode. Furthermore, FLASHIda enables sensitive mapping of post-translational modifications and detection of chemical adducts. As a software extension module to the instrument, FLASHIda can be readily adopted for TDP studies of complex samples to enhance proteoform identification rates.

The main purpose of this research was to reveal the effect of selection attributes and innovative restaurant image on perceived value, satisfaction, and behavior intention. This study laid the foundation of the clear concept of fine-dining restaurant in Korea and derived the variables of innovativeness and wine quality as key constructs. A total of 657 samples were collected from experienced customers who visited fine-dining restaurants in Seoul within 3 months. SEM analysis was conducted to test hypotheses. The result indicates that Price Fairness and Food Quality are key variables which needed to be considered more to induce customer satisfaction and revisit intention by providing strategical implications to make fine-dining restaurant as a sustainable business model.

This study aims to present the necessity of categorizing Korean for Hobby Purposes (KHP) as a part of Korean for Specific Purposes (KSP) and to suggest the method to design a KHP course. The number of overseas Korean learners has increased with the influence of the Korean wave, and many of them want to learn Korean as a hobby. To reflect this social change and learners’ needs, it is necessary to add KHP to a category of KSP. This study grouped three types of KHP learners: 1) to enjoy the Korean wave, 2) to prepare a trip to Korea, 3) to learn Korean as leisure. These learners are beginners, but the KHP curriculum is different from Korean for General Purposes (KGP). KHP course focuses on speaking skills, spoken Korean and the Korean wave contents, whereas KGP course focuses on integrated skills, grammar and tests. The principal objective of this study was to assess the presence of Salmonella sp. in imported amphibians and reptiles. A total of 49 samples were collected from commercial pet shops and life science museums in Korea and examined via the traditional detection method (TDM) and the PCR method. Salmonella typhimurium and Salmonella enteritidis were screened via multiplex PCR (mPCR). The results of the TDM showed a positive isolation in 18.4% of the samples, whereas the PCR method had a 28.6% positive detection rate. These results indicate the possibility that humans can contact Salmonella infections from amphibians and reptiles. Our findings are also important from a public health standpoint. Additionally, the presence of Salmonella sp. in these samples indicates the necessity of appropriate quarantine practices to be established.
