Impacts of climate change, such as sea-level rise and changes in annual precipitation, are becoming increasingly visible around the world and within Germany, thus increasing pressures to adapt. Forces of stability and change within established policy fields greatly determine the extent to which governance systems can adapt to worsening existing risks and new challenges. Employing a lock-in perspective and a comparative analysis of the governance of coastal risks in Schleswig-Holstein and water scarcity in Thuringia, we show how infrastructures, institutions, actors, and cognitive framing shape policy landscapes and together constitute dynamics of policy stability and change in the face of long-term climate impacts. This paper offers a comprehensive, systemic perspective of how adaptation challenges fit into established policy paradigms and programs as it highlights how non-material and material components are intertwined and can act as constraints to policy-making. Die Auswirkungen des Klimawandels, wie etwa der Anstieg des Meeresspiegels oder zunehmende Dürre und Wassermangel, werden vermehrt auch in Deutschland spürbar und erzeugen einen wachsenden politischen Handlungsbedarf. Widerstreitende Kräfte von Wandel und Stabilität innerhalb etablierter Politikfelder bestimmen dabei, inwieweit sich bestehende Governancesysteme an diese neuen Herausforderungen und Klimarisiken anzupassen vermögen. Mithilfe eines Lock-in-Ansatzes und einer vergleichenden Analyse zweier Fallstudien zur Governance von Küstenrisiken in Schleswig-Holstein und von Niedrigwasser in Thüringen verdeutlicht dieser Beitrag, wie das Zusammenspiel von Infrastrukturen, Institutionen, Akteuren und kognitiven Frames die Stabilität bzw. den Wandel etablierter Politikfelder vor dem Hintergrund langfristiger Klimarisiken beeinflusst. Damit zeigt dieser Beitrag eine übergreifende, systemische Perspektive auf, um zu erfassen, inwieweit bestehende Politikfelder mit ihren etablierten Programmen und Paradigmen fähig und geeignet sind, neuen politischen Problemlagen zu begegnen, und wie materielle und immaterielle Faktoren den politischen Handlungsspielraum begrenzen.

Mule deer were first introduced to Santa Catalina Island, California, in the early 1930s and persist today. Other feral, non-native ungulates have been eradicated (goats, pigs) or significantly reduced in numbers (bison) over the past two decades. Effective management of the deer population is necessary to protect the island’s biodiversity but is dependent upon reliable estimates of population density and demography. We used annual summer spotlight counts, conducted in eight of the past ten years, to estimate deer densities in the island interior. In 2021, we also surveyed transects in the area around Avalon, the largest town on the island. Distance sampling (Program DISTANCE) was used to model density based on line transect data. Island-wide densities varied from 6.3 to 16.9 deer per km2, with an average of 10.2 per km2, and were positively correlated with July-June rainfall during the preceding year. Most (77-96%) of the identifiable deer were adults and most adults were does (58-75%). Deer were spotted most frequently in island chaparral and coastal sage scrub vegetation, the most common vegetation cover types along transects, whereas the use of grasslands and man-made and non-native habitats varied among years. The estimated deer density in Avalon (65.7 per km2) was six to nine times higher than estimates for the interior transects in 2021. The high density of deer near Avalon suggests that gardens, landscaping, golf courses, and intentional feeding subsidize deer numbers in town that have the potential to spill over into the more natural areas of the island.

The hypothalamus is the main integrating center for metabolic control, regulating the response to nutrients and other metabolic signals to maintain energy homeostasis. This response includes not only neuronal circuits that control appetite, but also glial cells that are involved in nutrient processing and inflammatory responses to high fat intake. However, it is now clear that the metabolic response is different depending on the type of fatty acid consumed, but the mechanisms remain largely unknown. Our aim was to analyze the hypothalamic response to the consumption of a diet enriched with sunflower oil (SOD), high in oleic acid content, or palm seed oil (POD), with a high lauric acid content. Adult male C57BL/6N mice were given ad libitum access to a standard rodent chow diet enriched with SOD or POD, or to a standard rodent chow (SD) for 8 weeks. Ahora sabemos que sus causas son variadas y que cada individuo muestra diferente propensión a desarrollar obesidad y/o patologías asociadas a esta. Sin duda, el desarrollo de estrategias terapéuticas y medicamentos contra esta epidemia abarca un amplio rango de posibles dianas. Sin embargo, todavía queda mucho que aprender sobre el control del metabolismo y sus diferencias individuales. El objetivo de esta tesis es analizar los efectos de la edad, el sexo y el trasfondo genético en la respuesta a una dieta alta en grasas en ratones. La pubertad es un período del desarrollo con múltiples cambios que ocurren tanto en el sistema nervioso central como a nivel periférico. Sin embargo, las respuestas específicas a una mala nutrición durante la pubertad no han sido estudiadas en profundidad. Por tanto, hemos analizado la respuesta metabólica de ratones de ambos sexos en edad peri-puberal, sometiéndoles a una dieta alta en grasa durante un corto período de tiempo. Por otra parte, el envejecimiento aumenta la propensión al sobrepeso y ciertas enfermedades, incluyendo trastornos neurodegenerativos como el de Alzheimer. Por ello, hemos utilizado un modelo genético de enfermedad de Alzheimer en ratones, sometiendo a machos y a hembras a una dieta alta o baja en grasas durante un período largo de tiempo para analizar su respuesta metabólica. Además, dado que los astrocitos están implicados tanto en el control metabólico como en la neuroprotección, hemos analizado la respuesta de astrocitos hipotalámicos de machos y hembras al ácido palmítico, un ácido graso saturado que se encuentra comúnmente en nuestra dieta, y al amiloide-β, un compuesto importante en el desarrollo de la enfermedad de Alzheimer. Los resultados que aquí se exponen indican que los ratones en edad puberal/peripuberal son menos propensos a la ganancia de peso tras un período corto de dieta alta en grasas, en comparación con lo que se ha observado en otros estadios del desarrollo. Los ratones de ambos sexos aumentaron su ingesta calórica sin aumentar de peso. Sin embargo, los perfiles metabólicos se vieron alterados de diferente manera en función del sexo, con un mayor efecto en machos que en hembras. Cuando ratones de mediana edad fueron expuestos a una dieta alta en grasas encontramos que las hembras ganaron más peso y grasa que los machos, lo que contrasta con lo que se suele observar en adultos jóvenes., Obesity and its secondary complications continue to increase worldwide and constitute one of the most important health care problems in many societies. It is now clear that the underlying causes of obesity can vary and that not all individuals have similar propensities to become obese and/or to develop obesity-associated pathologies. Indeed, the development of treatment strategies and drugs to curtail this epidemic span a wide range of targets. However, much is yet to be learned regarding metabolic control and individual differences in this process. The aim of this thesis was to analyze the effects of age, sex and genetic background on the response to a high fat diet in mice. Puberty is a period of development with multiple changes occurring both in the central nervous system and systemically. However, little is known regarding the specific response during puberty to poor nutrition. Hence, we analyzed the metabolic response of both male and female peripubertal mice to a short-term high fat diet protocol. On the other extreme, aging increases the overall propensity to weight gain and some diseases, including neurodegenerative diseases such as Alzheimer’s. Hence, we employed a genetic model of Alzheimer’s disease in mice, submitting male and female mice to a long-term high fat diet or low fat diet and analyzing their metabolic response. Moreover, as astrocytes are involved in both metabolic control and neuroprotection, we analyzed the response of hypothalamic astrocytes from the hypothalamus of males and females to palmitic acid, a saturated fatty acid commonly found in our diet, and to amyloid-β, which is an important component in the development of Alzheimer’s disease. The results reported here indicate that during the peripubertal/pubertal period mice are less prone to excess weight gain to short-term high fat intake compared to that reported at other developmental ages. Mice of both sexes increased their energy intake, but body weight was not affected. However, metabolic profiles were modified in a sex dependant manner with males being more affected than females. La CCK tiene un efecto estimulante sobre la liberación de insulina que, junto con la disminución del tamaño de las raciones ingeridas, permitiría la correcta distribución de los nutrientes. De este modo, se evitaría la eventual sobrecarga de los mismos, que podría desencadenar la saturación de los mecanismos celulares involucrados en el metabolismo de la glucosa y los TG. Por tanto, es necesario seguir investigando el papel de la CCK en la regulación hormonal de la digestión y el modo en el que se relaciona con el desarrollo de obesidad, para obtener un conocimiento más completo acerca de los mecanismos fisiopatológicos responsables de esta enfermedad y poder desarrollar tratamientos efectivos. Además, este estudio incide en el concepto de que la obesidad, independientemente de los factores genéticos, no es únicamente una cuestión de ingesta calórica y de composición de la comida sino también de la distribución circadiana de la ingesta diaria. Por tanto, este estudio puede proporcionar información relevante para el diseño de nuevas terapias para el tratamiento y prevención de la obesidad.
