Recent studies demonstrate that paternal nutrition prior to conception may determine offspring development and health through epigenetic modification. This study aims to investigate the effects of paternal supplementation of n-3 polyunsaturated fatty acids (n-3 PUFAs) on the brain development and function, and associated gene imprinting in the offspring. Three to four-week-old male C57BL/6J mice (founder) were fed with an n-3 PUFA-deficient diet (n-3 D), and two n-3 PUFA supplementation diets – a normal n-3 PUFA content diet (n-3 N) and a high n-3 PUFA content diet (n-3 H) for 12 weeks. Then they were mated to 10-week-old virgin female C57BL/6J mice to generate the offspring. The results showed that paternal n-3 PUFA supplementation in preconception reduced the anxiety- and depressive-like behavior, and improved sociability, learning and memory in the offspring, along with increased synaptic number, upregulated expressions of neuron specific enolase, myelin basic protein, glial fibrillary acidic protein, brain-derived neurotrophic factor in the hippocampus and cerebral cortex, and altered expressions of genes associated with mitochondria biogenesis, fusion, fission and autophagy. Furthermore, with paternal n-3 PUFA supplementation, the expression of imprinted gene Snrpn was downregulated both in testes of the founder mice and their offspring, but upregulated in the cerebral cortex and hippocampus, with altered DNA methylation in its differentially methylated region. The data suggest that higher paternal intake of n-3 PUFAs in preconception may help to maintain optimal brain development and function in the offspring, and further raise the possibility of paternal nutritional intervention for mental health issues in subsequent generations.

Polymer-based nanocarriers require extensive knowledge of their chemistries to learn functionalization strategies and understand the nature of interactions that they establish with biological entities. In this research, the poly (β-amino ester) (PβAE-447) was synthesized and characterized, aimed to identify the influence of some key parameters in the formulation process. Initially; PβAE-447 was characterized for aqueous solubility, swelling capacity, proton buffering ability, and cytotoxicity study before nanoparticles formulation. Interestingly, the polymer-supported higher cell viability than the Polyethylenimine (PEI) at 100 μg/ml. PβAE-447 complexed with GFP encoded plasmid DNA (pGFP) generated nanocarriers of 184 nm hydrodynamic radius (+7.42 mV Zeta potential) for cell transfection. Transfection assays performed with PEGylated and lyophilized PβAE-447/pDNA complexes on HEK-293, BEAS-2B, and A549 cell lines showed better transfection than PEI. The outcomes toward A549 cells (above 66%) showed the highest transfection efficiency compared to the other cell lines. Altogether, these results suggested that characterizing physicochemical properties pave the way to design a new generation of PβAE-447 for gene delivery.

Ultra-small biomass gasification and power generation systems are a promising technology for disaster areas in developed countries and non-electrified rural areas of developing countries. The gasification characteristics of carbonized wood pellets and carbonized wood briquettes without or with 1 wt% sea salt were tested in a pilot-scale updraft fixed-bed gasifier. The results showed that the carbon balance during gasification was easily achieved, and most of the chlorine in the carbonized briquettes with sea salt remained in the residual char and fly ash after gasification. Inorganic chlorine in the solid fuel was transformed into organic chlorine combined with carbon matrix after heat treatment in the gasifier. Syngas with a low heating value higher than 4 MJ·m−3 could be continuously obtained when the system reached a stable condition, which enabled power generation using a gas engine. A combination of several secondary tar removal processes was adopted for the syngas purification. The tar content in the syngas after the gas cleaning for 425 °C-carbonized pellet, 425 °C-carbonized briquette, 475 °C-carbonized briquette, and 475 °C-carbonized briquette with sea salt was 0.98, 0.26, 1.4, and 0.2 g·Nm−3, respectively. During the stable stage, 425 °C-carbonized pellet showed the highest cold gas efficiency (CGE) of 45.6% and carbon conversion efficiency (CCE) of 57.8%, respectively, with an equivalence ratio (ER) of 0.24. A lower carbonization temperature showed a higher CGE and CCE. The residual char and fly ash after gasification could be considered for reuse as feedstock to further improve the overall process efficiency. The maximum output power was 23 kW at an air flow rate of 40 Nm3/h during the gasification of carbonized pellets, and the corresponding overall power generation efficiency of the engine using the syngas was about 27.8%. For 475 °C carbonized briquette, the maximum output power was 18 kW at an air flow rate of 40 Nm3/h, and the corresponding overall power generation efficiency of the engine using the syngas was about 25.5%.

Small-scale, distributed and low cost biomass power generation technologies are highly required in the modern society. The biomass pretreatment process, that is, a combination of carbonization and pelletization processes of cedar wood were explored in this study. For the carbonization process, a novel carbonizer composed of one carbonization chamber and one combustion chamber was tested, and the mass and energy balances during cedar wood carbonization at 400–500 °C were analyzed, respectively. During the initial heating stage, about 22 kg of wood with dry basis should be supplied into the combustion chamber through several batches. When the temperature in the carbonization chamber reached to the range of 200–220 °C, the combustion chamber could be self-heated by burning the pyrolysis gas (tar vapor, CH4, H2, CO) sent from the carbonization chamber. The specific wood consumption for producing 1 kg of wood char was within 3.6–4.4 kg, which presented an increase trend with increasing the carbonization temperature. Moreover, this carbonization system can deal with different kinds of biomass with various water contents and shapes. For the pelletization process, effects of adding amount of the binder (poval solution), water and the carbonization temperature on the pellet quality were studied so as to explore the optimal condition for making high quality pellets from the crushed wood char. The results indicated that there was an optimal ratio among char, added water, and the binder for making high quality pellets. Considering the energy transfer efficiency and pellet quality, the mass ratio of 0.93:0.32:0.10 for 450 °C-char in the dry basis: added water plus water in char: poval solution was the optimal condition for making high quality pellets.

This work aims to demonstrate the technical feasibility of a portable ultra-small power generation system based on the gasification of carbonized wood pellet/briquette. A combined pretreatment process including carbonization and densification were adopted to deal with various biomass materials. Pelletization/Briquetting of carbonized fuel realized the stable operation of an updraft gasifier. After purification with several secondary clean devices, tar content in syngas was reduced to 0.49 g·m-3 and 1.4 g·m-3 for carbonized pellet and carbonized briquette, respectively. The cold gas efficiency during carbonized wood briquette gasification was about 64.4%, and the corresponding overall efficiency of the engine was 19.0%. Moreover, the steady output of 25 kW power was continuously realized. Therefore, the comprehensive system covering biomass carbonization, densification, gasification, syngas purification, and engine system is feasible for portable, ultra-small power generation
