Currently, feed enzymes are primarily obtained through fermentation of fungi, bacteria, and other microorganisms. Although the manufacturing technology for feed enzymes has evolved rapidly, the activities of these enzymes decline during the granulating process and the cost of application has increased over time. An alternative approach is the use of genetically modified plants containing complex feed enzymes for direct utilization in animal feedstuff. We co-expressed three commonly used feed enzymes (phytase, β-glucanase, and xylanase) in barley seeds using the Agrobacterium-mediated transformation method and generated a new barley germplasm. The results showed that these enzymes were stable and had no effect on the development of the seeds. Supplementation of the basal diet of laying hens with only 8% of enzyme-containing seeds decreased the quantities of indigestible carbohydrates, improved the availability of phosphorus, and reduced the impact of animal production on the environment to an extent similar to directly adding exogenous enzymes to the feed. Feeding enzyme-containing seeds to layers significantly increased the strength of the eggshell and the weight of the eggs by 10.0%-11.3% and 5.6%-7.7%, respectively. The intestinal microbiota obtained from layers fed with enzyme-containing seeds was altered compared to controls and was dominated by Alispes and Rikenella. Therefore, the transgenic barley seeds produced in this study can be used as an ideal feedstuff for use in animal feed.

Nucleobase oxidation and alkylation can destroy Watson-Crick base-pairing to challenge the genomic integrity. Human 8-oxoguanine glycosylase 1 (hOGG1) and alkyladenine glycosylase (hAAG) are evolved to counter these two cytotoxic lesions through base-excision repair, and their deregulations are implicated with multifactorial diseases and cancers. Herein, we demonstrate activatable self-dissociation of Watson-Crick structures with fluorescent nucleotides for sensing multiple human glycosylases at single-cell level. The presence of hOGG1 and hAAG catalyzes 8-oxoG and deoxyinosine removal in functional probe 1 to release two trigger probes (1 and 2). Then, trigger probes hybridize with functional probe 2 to activate the autocatalytic degradation of functional probes 2 (Cycle I) and 3 (Cycle II), replicating abundant trigger probes (1-4) and releasing two fluorophores (2-aminopurine (2-AP) and pyrrolo-dC (P-dC)). New trigger probes (1, 2) and (3, 4), in turn, hybridize with free functional probes 2 and 3, repeating Cycles I and II turnovers. Through multicycle self-dissociation of Watson-Crick structures, 2-AP and P-dC are exponentially accumulated for the simultaneous quantification of hOGG1 and hAAG. Dance, as a unique form of expression, is usually accompanied by music and presented to the audience visually, improving people’s cultural and spiritual lives while also strengthening their creative energy. And dance choreography is usually created by a few skilled choreographers, either individually or together, with a high level of expertise and complexity. With the introduction of motion capture technology and artificial intelligence, computers can now do autonomous choreography based on music, and science and technology are changing the way artists produce art today. Computer music choreography must solve two fundamental issues: how to create realistic and creative dance moves without relying on motion capture and manual creation and how to improve music and dance synchronization utilizing appropriate music and movement elements and matching algorithms. This article employs a hybrid density network to generate dances that fit the target music in three steps, action generation, action screening, and feature matching, to address the aforementioned two concerns.

Halomonas bluephagenesis, a robust and contamination-resistant microorganism has been developed as a chassis for "Next Generation Industrial Biotechnology". The non-model H. bluephagenesis requires efficient tools to fine-tune its metabolic fluxes for enhanced production phenotypes. Here we report a highly efficient gene expression regulation system (PrrF1-2-HfqPa) in H. bluephagenesis, small regulatory RNA (sRNA) PrrF1 scaffold from Pseudomonas aeruginosa and a target-binding sequence that downregulate gene expression, and its cognate P. aeruginosa Hfq (HfqPa), recruited by the scaffold to facilitate the hybridization of sRNA and the target mRNA. The PrrF1-2-HfqPa system targeting prpC in H. bluephagenesis helps increase 3-hydroxyvalerate fraction in poly(3-hydroxybutyrate-co-3-hydroxyvalerate) to 21 mol% compared to 3.1 mol% of the control. This sRNA system repressed phaP1 and minD simultaneously, resulting in large polyhydroxybutyrate granules. Further, an sRNA library targeting 30 genes was employed for large-scale target identification to increase mevalonate production. This work expands the study on using an sRNA system not based on Escherichia coli MicC/SgrS-Hfq to repress gene expression, providing a framework to exploit new powerful genome engineering tools based on other sRNAs.

Methionine is an essential sulfur-containing amino acid that finds widespread applications in agriculture, medicine, and the food industry. However, the complex and multibranched biosynthetic pathway of methionine has posed significant challenges to its efficient fermentation production. In this study, we employed a modularized synthetic biology strategy to improve the weakest branched pathway of methionine biosynthesis. Three exogenous modules were constructed and assembled to provide methyl donors, which are the primary limiting factors in methionine biosynthesis. The first module utilized added methanol, which was converted into 5,10-methylene-tetrahydrofolate for methionine production but was hindered by the toxicity of methanol. To circumvent this issue, a non-toxic formate module was constructed, resulting in a visible improvement in the methionine titer. Finally, an exogenous betaine module was constructed, which could directly deliver methyl to methionine. The final strain produced 2.87 g/L of methionine in a flask, representing a 20% increase over the starting strain. This study presents a novel strategy for improving and balancing other metabolites that are synthesized through complex multibranched pathways.

Purpose: Chimeric antigen receptor–modified T cells (CAR-T) have demonstrated encouraging results in early-phase clinical trials. Successful adaptation of CAR-T technology for CEA-expressing adenocarcinoma liver metastases, a major cause of death in patients with gastrointestinal cancers, has yet to be achieved. We sought to test intrahepatic delivery of anti-CEA CAR-T through percutaneous hepatic artery infusions (HAIs).Experimental Design: We conducted a phase I trial to test HAI of CAR-T in patients with CEA+ liver metastases. Six patients completed the protocol, and 3 received anti-CEA CAR-T HAIs alone in dose-escalation fashion (108, 109, and 1010 cells). We treated an additional 3 patients with the maximum planned CAR-T HAI dose (1010 cells × 3) along with systemic IL2 support.Results: Four patients had more than 10 liver metastases, and patients received a mean of 2.5 lines of conventional systemic therapy before enrollment. No patient suffered a grade 3 or 4 adverse event related to the CAR-T HAIs. One patient remains alive with stable disease at 23 months following CAR-T HAI, and 5 patients died of progressive disease. Among the patients in the cohort that received systemic IL2 support, CEA levels decreased 37% (range, 19%–48%) from baseline. Biopsies demonstrated an increase in liver metastasis necrosis or fibrosis in 4 of 6 patients. Elevated serum IFNγ levels correlated with IL2 administration and CEA decreases.Conclusions: We have demonstrated the safety of anti-CEA CAR-T HAIs with encouraging signals of clinical activity in a heavily pretreated population with large tumor burdens. Autoimmune diseases such as ankylosing spondylitis (AS) can be driven by emerging neoantigens that disrupt immune tolerance. Here, we developed a workflow to profile posttranslational modifications involved in neoantigen formation. Using mass spectrometry, we identified a panel of cysteine residues differentially modified by carboxyethylation that required 3-hydroxypropionic acid to generate neoantigens in patients with AS. The lysosomal degradation of integrin αIIb [ITGA2B (CD41)] carboxyethylated at Cys96 (ITGA2B-ceC96) generated carboxyethylated peptides that were presented by HLA-DRB1*04 to stimulate CD4 + T cell responses and induce autoantibody production. Immunization of HLA-DR4 transgenic mice with the ITGA2B-ceC96 peptide promoted colitis and vertebral bone erosion. Thus, metabolite-induced cysteine carboxyethylation can give rise to pathogenic neoantigens that lead to autoreactive CD4 + T cell responses and autoantibody production in autoimmune diseases.

Background: SMARCA4-deficient undifferentiated tumor (SMARCA4-UT) is a rare tumor that is independent of lung cancer and is classified as undifferentiated carcinoma of epithelial tumors according to the 2021 WHO classification of thoracic tumors. Loss of BRG1 expression due to SMARCA2 and SMARCA4 gene defects is a possible pathogenic mechanism. The pathological features are mainly nest-like rhabdoid cells with necrosis. Currently, there is a lack of well-recognized and convincing regimens for the treatment of SMARCA4-UT. Because the lesions are mainly located in the mediastinum and lung, patients can present with thoracic-related symptoms. We report the case of a middle-aged male patient with SMARCA4-UT who was admitted to the hospital with polyarthralgia and fever and lacked typical thoracic-related symptoms. This may be the first case report of SMARCA4-UT with fever and polyarthralgia as the initial symptoms, and its diagnostic idea and case characteristics are worthy of consideration and reference. Case Description: A 57-year-old male fisherman with a long history of smoking and emphysema was initially admitted to the Department of Rheumatology and Immunology because of fever and polyarthralgia as the initial symptoms. The imaging examination suggested a mediastinal mass. With the gradual advancement of the examination, the riddle of fever and polyarthralgia was gradually revealed. After symptom control with steroids, he underwent thoracoscopic surgery for mediastinal lymph node biopsy, and SMARCA4-UT was finally diagnosed. He was evaluated and transferred to the oncology department for one cycle of postoperative adjuvant chemotherapy, and he was discharged with good symptom control after discharge.We lost follow-ups of patients after they were discharged. Conclusions: Although fever and multiple arthralgias are not uncommon in neoplastic lesions, as atypical initial symptoms of rare tumors, clinicians should be aware of this when diagnosing tumors and not limited to SMARCA4-UT. In addition, the pathological and imaging features of SMARCA4-UT should be fully understood.

Organosulfur compounds in fossil fuels have been a major concern in the process of achieving zero-sulfur fuel production. Biodesulfurization (BDS) is an environmentally friendly strategy for the removal of refractory organosulfur compounds from fossil fuels. Even though researchers are committed to engineering the desulfurization-specific pathway for improving BDS efficiency, the industrial application of BDS is still difficult. Recently, the sulfur metabolism of Rhodococcus has begun to attract attention due to its influences on the BDS process. In this review, we introduce the sulfur metabolism in Rhodococcus, including sulfur absorption, reduction, and assimilation; and summarize desulfurization in Rhodococcus, including the desulfurization mechanism, the regulation mechanism of the 4S pathway, and the strategies of optimizing the 4S pathway to improve BDS efficiency. In particular, the influence of sulfur metabolism on BDS efficiency is discussed. In addition, we consider the latest genetic engineering strategies in Rhodococcus. An improved understanding of the relationship between sulfur metabolism and desulfurization will enable the industrial application of BDS.

Small-cell lung cancers (SCLC) and large-cell neuroendocrine carcinomas (LCNEC) are two types of high-grade pulmonary neuroendocrine carcinomas (NECs). Diagnostic neuroendocrine markers commonly include synaptophysin, chromogranin A, CD56, and insulinoma-associated protein 1 (INSM1). In this study, the utility of secretagogin (SCGN) was examined in the context of pulmonary NEC diagnosis. The study included 71 pulmonary NEC cases (18 SCLCs, 13 combined-SCLCs, 23 LCNECs, and 17 combined-LCNECs). Immunohistochemical stains of SCGN, synaptophysin, chromogranin A, CD56, and INSM1 were performed on whole tumor sections. The stains were evaluated based on combined staining intensity and the proportion of positive tumor cells. At least mild staining intensity in at least 1% of the cells was considered positive. Bioinformatic studies showed specific SCGN expression in neuroendocrine cells and NECs. SCGN showed diffuse nuclear and cytoplasmic staining in NECs with intra-tumoral heterogeneity. The non-neuroendocrine components were negative. The sensitivity of SCGN was no better than the other established neuroendocrine markers based on all NECs combined or LCNECs/c-LCNECs only. However, the sensitivity of SCGN (71%) was higher than chromogranin A (68%) for SCLCs/c-SCLCs only. The average proportion of SCGN positive tumor cells was 8% higher than chromogranin A (22% versus 14%, P = 0.0332) in all NECs and 18% higher for SCLC and c-SCLC cases only (32% versus 13%, P = 0.0054). The above data showed that SCGN could be used as a supplemental neuroendocrine marker to diagnose SCLC.

The (co)variance components and corresponding phenotypic and genetic parameters for growth traits and wool traits of economic importance were estimated in the Alpine Merino sheep population maintained at Gansu Provincial Sheep Breeding Technology Extension Station in northwestern China. Data from a maximum of 49,474 animals sired by 526 rams and born from 22,531 ewes over 20 years from 2000 to 2019 were used in this study. Birth type, age of dam, birth year, sex and/or management group, and age at measurement were initially fitted as fixed effects in an animal model with various random effects. Genetic groups were defined for all animals by the sire breed and breed genotype interacted with dam-strain flocks and were fitted as one of the random effects. Analyses were conducted using a residual maximum likelihood procedure (ASReml). Seven different animal models were fitted for all traits, and the most appropriate model with relevant random effects was selected through log-likelihood ratio testing. After identifying the appropriate model through single-trait analysis, bivariate analyses were used to obtain the phenotypic and genetic correlations among the traits. The estimates of additive direct heritability for birth weight (BWT), weaning weight (WWT), preweaning growth rate (prwADG), postweaning growth rate (powADG), yearling body weight (YWT), average fibre diameter (AFD), greasy fleece weight (GFW), clean fleece weight (CFW), yield (YLD), yearling wool staple length (YSL), coefficient of variation of average fibre diameter (FDcv) and wool visual fineness counts (VFC) were 0.30, 0.18, 0.18, 0.20, 0.29, 0.20, 0.19, 0.20, 0.35, 0.19, 0.16 and 0.13, respectively, with standard errors ranging from 0.02 to 0.05. The corresponding ratios of genetic group variance to additive genetic variance were significant and, respectively, 0.35, 0.80, 0.62, 0.26, 0.13, 1.06, 0.38, 0.64, 0.09, 0.12, 0.06 and 0.58. These results suggest for these traits that there is potential to exploit both the additive genetic variation and between genetic group variation although for most traits the between group variation was smaller than the variation within groups. Favourable genetic correlations were found among the growth traits, and between growth traits and fleece production traits, and among wool traits GFW, CFW, YSL and YLD. This study provides the required estimates of genetic parameters of both growth and wool traits of the new breed for the design of more effective breeding programmes.

One dimensional (1D) organic-inorganic halide hybrid perovskite has the advantages of excellent organic cation modifiability and diversity of inorganic framework structures, which cannot be ignored in the development of multi-functional phase transition materials in photoelectric and photovoltaic devices. Here, we have successfully modified and synthesized an organic-inorganic hybrid perovskite photoelectric multifunctional phase transition material: [C7H13ONCH2F] PbBr3 (1). The synergistic effect of the order double disorder transition of organic cations and the change of the degree of distortion of the inorganic framework leads to its high temperature reversible phase transition point of Tc=374 K/346 K and its ultra-low loss high-quality dielectric switch response. Through in-depth research and calculation, compound 1 also has excellent semiconductor characteristics with a band gap of 3.06 eV and the photoluminescence characteristics of STE broadband emission. Undoubtedly, this modification strategy provides a new choice for the research field of organic-inorganic hybrid perovskite reversible phase transition photoelectric multifunctional materials with rich coupling properties.

Objective Diabetes increases the risk for cancers. However, whether it is associated with hematologic malignancies is not clear. The present study investigated the association between diabetes and acute lymphocytic leukemia (ALL), acute myeloid leukemia (ML), non-Hopkin lymphoma (NHL), and multiple myeloma (MM). Methods Newly diagnosed adult cancer patients were recruited consecutively from our clinical database. Peoples from a local enterprise were recruited to create a small-scale population-based dataset. We compared the diabetes prevalence between the cancer patients and the local people; an increase in diabetes prevalence in the cancer patients suggests an association between diabetes and the cancer(s). Results We found that the prevalence of diabetes was 19.7%, 21.3%, 12.5%, and 12.0% in ALL, AML, NHL, and MM, respectively, which was higher than that (9.1%) in the local people. Despite that there were more male than female cancer patients, there were more female than male diabetic patients. The increase in diabetes prevalence occurred in ALL and NHL patients aged 18 to 39 years old as well as in AML patients over 40. In MM patients, the increase in diabetes prevalence (18.6%) occurred only in females. Approximately 70% of the diabetic patients were undiagnosed before the diagnosis of the blood cancer. Approximately half of the pre-existing diabetic patients had anti-diabetic treatment, with over 70% of them still had poor glycemic control. Conclusions Our results suggest that diabetes is associated with ALL, AML, NHL, and MM, at least in adult patients.

Water stable Zr-metal–organic framework nanoparticles (PCN-224 NPs) have been solvothermally synthesized. PCN-224 NPs show spherical shape with smooth surface and particle size of approximately 200 nm. PCN-224 NPs can be stable in acid and aqueous solutions, as confirmed by powder X-ray diffraction. Gatifloxacin (GTF) adsorption measurements showed that PCN-224 NPs exhibit a high adsorption capacity of 876 mgg-1. Meanwhile, the adsorption factors, adsorption characteristics, and mechanisms of GTF were investigated in batch adsorption experiments.

MicroRNA (miRNA)-related single-nucleotide polymorphisms (miR-SNPs) are a novel class of genetic variations involved in multiple cellular functions, and they have emerged as promising biomarkers for cancer diagnostics and prognostics. Herein, we demonstrate for the first time the structure-switchable hairpin-powered exponential replications for sensing attomolar miR-SNPs in human cancer tissues with zero background. In the presence of target miR-196a2T, hairpin probes (i.e., HP1 and HP2) are splinted together to construct the dumbbell-shaped probe (DSP) with SplintR ligase catalysis. Once the DSP is formed, the self-primed polymerization extension and linear FIP/BIP-primed strand displacement DNA synthesis (SDS) are automatically repeated to activate self-circulated exponential amplification, producing large amounts of double-stranded DNAs (dsDNAs) which can be real-time monitored using SYBR Green I. This nanodevice can detect miR-196a2T with an ultralow detection limit of 2.46 aM; distinguish rare miR-196a2 SNP with a selectivity factor of 0.001%; and even profile miR-196a2T in human tissues for nonsmall cell lung cancer (NSCLC) diagnosis, risk assessment, and cancer type prediction. Notably, this nanodevice can be rapidly and homogeneously used in one tube in a real-time and label-free manner, providing a powerful point-of-care platform for noninvasive diagnostics and prognostics of miR-SNPs-related human cancers.

BackgroundGestational trophoblastic neoplasia (GTN) is a group of clinically rare tumors that develop in the uterus from placental tissue. Currently, its satisfactory curability derives from the timely and accurately classification and refined management for patients. This study aimed to discover biomarkers that could predict the outcomes of GTN patients after first-line chemotherapy.MethodsA total of 65 GTN patients were included in the study. Patients were divided into the good or poor outcome group and the clinical characteristics of the patients in the two groups were compared. Furthermore, the serum peptide profiles of all patients were uncovered by using weak cation exchange magnetic beads and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Feature peaks were identified by three machine learning algorithms and then models were constructed and compared using five machine learning methods. Additionally, liquid chromatography mass spectrometry was used to identify the feature peptides.ResultsMultivariate logistic regression analysis showed that the International Federation of Gynecology and Obstetrics (FIGO) risk score was associated with poor outcomes. Eight feature peaks (m/z =1287, 2042, 2862, 2932, 2950, 3240, 3277 and 6626) were selected for model construction and validation by the three algorithms. Based on the panel combining FIGO risk score and peptide serum signatures, the neural network (nnet) model showed promising performance in both the training (AUC=0.9635) and validation (AUC=0.8788) cohorts. Peaks at m/z 2042, 2862, 2932, 3240 were identified as the partial sequences of transthyretin, fibrinogen alpha chain (FGA), beta-globin and FGA, respectively.ConclusionWe combined FIGO risk score and serum peptide signatures using the nnet method to construct the model which can accurately predict outcome of GTN patients after first-line chemotherapy. With this model, patients can be further classified and managed, and those with poor predicted outcomes can be given more attention for developing treatment failure.

To investigate the relationship between polymorphism of cytochrome P450, family 2, subfamily A, polypeptide 6(CYP2A6) and periodontitis, the expression of inflammatory cytokines in 123 Han smokers.From October 2018 to October 2019, a total of 123 smokers with periodontitis were selected as the experimental group, and 125 non-smokers as the control group. The general data of the patients were collected, including age, gender, body mass index (BMI), chewing and brushing habits, as well as molar condition; plaque index (PLI), gingival bleeding index (BI), periodontal probing depth (PD) and attachment loss (AL) were detected. CYP2A6 was amplified by PCR. The level of interleukin (IL)-17, IL-1, IL-6, IL-23 and tumor necrosis factor-α (TNF-α) in GCF was detected by enzyme-linked immunosorbent assay(ELISA). SPSS 25.0 software package was used for statistical analysis of the data.There was significant difference in gender, PLI, IL-17, IL-1, IL-6, IL-23, TNF-α level in GCF between the two groups(P＜0.05). All samples were amplified by PCR. Among them, 23 were not amplified, which were identified as CYP2A6 deletion type (CYP2A6del), including 5 in the experimental group and 18 in the control group; 225 were amplified and identified as CYP2A6 wild type(CYP2A6wt), including 118 in the experimental group and 107 in the control group. There was significant difference in CYP2A6 genotype between the two groups(P＜0.05). In the experimental group, the level of IL-1 and PLI of different CYP2A6 genotypes was significantly different(P＜0.05); and in the control group, the level of IL-17 and PLI of different CYP2A6 genotypes was also significantly different(P＜0.05).There are differences in CYP2A6 genotype between smokers and non-smokers in Han population with periodontitis, but the relationship between CYP2A6 genotype and inflammatory cytokines is not clear.

Bioleaching is a promising route for electrolytic manganese (Mn) residue (EMR) reutilization due to being eco-friendly and cost-effective. However, microbes with high bioleaching efficiency are scarce. This work aimed to isolate, screen, and characterize a novel fungal strain with high Mn-bioleaching efficiency from EMR, and study the kinetics and mechanism. Confirming the tumor origin is often a diagnostic challenge in pathology and carries significant therapeutic impacts. Cytokeratin 7, estrogen receptor, and GATA binding protein 3 (GATA3) are well-established diagnostic markers frequently used to support a tumor's breast origin. However, their specificities still have room to improve. Many nonbreast tumors express cytokeratin 7 and estrogen receptor, and urothelial tumors frequently express GATA3. There is a practical need for a new breast lineage marker that is sensitive and specific. Wnt family member proteins play critical roles in embryo development, tissue homeostasis and tumor development through β-catenin dependent and independent pathways. The current study evaluated Wnt9b and GATA3 expression in 163 primary breast cancers, 63 metastatic breast cancers, and 525 nonbreast epithelial tumors. The positive rates of Wnt9b and GATA3 in primary breast cancer were both 98.7%. The positive rates in metastatic breast cancer were 87.3% for Wnt9b and 96.8% for GATA3. For nonbreast tumors, including 64 cases of urothelial carcinoma, Wnt9b was negative in all except salivary gland carcinomas. The study demonstrated that Wnt9b is a breast cancer marker with similar sensitivity as GATA3 but with greater specificity than GATA3 and may ultimately become a useful diagnostic tool in routine surgical pathology practice.

Aqueous zinc (Zn)-ion batteries are considered very promising in grid-scale energy storage systems. However, the dendrite, corrosion, and H2 evolution issues of Zn anode have restricted their further applications. Herein, to solve these issues, a hydrophilic layer, consisting of a covalent organic polymer (COP) and carboxylmethyl cellulose (CMC), is designed to in situ construct a multifunctional quasi-gel (COP-CMC/QG) interface between Zn metal and the electrolyte. The COP-CMC/QG interface can significantly improve the rechargeability of the Zn anode through enhancing Zn2+ transport kinetics, guiding uniform nucleation, and suppressing Zn corrosion and H2 evolution. As a result, the COP-CMC-Zn anode exhibits a reduced overpotential (12 mV at 0.25 mA cm-2), prolonged cycle life (over 4000 h at 0.25 mA cm-2 and 2000 h at 5 mA cm-2 in symmetrical cells), and elevated full-cell (Zn/MnO2) performance. This work provides an efficient approach to achieve long-life Zn metal anodes and paves the way toward high-performance Zn-based and other metal-ion batteries.

The primary objective of this study was to assess the patient safety culture in a general hospital in Shanghai, China, through a modified Manchester Patient Safety Framework (MaPSaF).This study has a qualitative interview design. Data were collected through group interviews and analyses performed through content analysis.The MaPSaF was translated into Chinese and used to assess the patient safety culture in a large general hospital in Shanghai, China. Group interviews using the MaPSaF were conducted with 15 nurses in the obstetric ward. Participants rated their safety practice individually on each of the nine MaPSaF safety culture dimensions. The dimensions and scores were then collectively discussed and a practice-wide consensus score for each dimension was agreed. Discussions were recorded, transcribed and analysed to assess patient safety in the obstetric ward.It took about 2 hr to complete the discussion focusing on patients' safety employing the MaPSaF. Most participants recognized the process as acceptable and useful. The MaPSaF directed team discussion about patient safety issues and facilitated communication, prompting some practice changes. All participants responded positively to the discussion and perceived MaPSaF as a good safety culture assessment tool, with clear, comprehensive and understandable entries. The process demonstrated that the department of obstetrics in the hospital already had a positive patient safety culture, but certain areas were highlighted as still needing improvement. Based on participants' positive experience and perception of the MaPSaF, it can be concluded that there is potential benefit in its adaptation and use in obstetrics wards of Chinese hospitals. The MaPSaF has the potential to strengthen existing safety cultures and improve general safety through collaborative measures.

Ulcerative colitis (UC) is a complicated disease caused by the interaction between genetic and environmental factors that affects mucosal homeostasis and triggers an inappropriate immune response. Single-cell RNA sequencing (scRNA-seq) can be used to rapidly obtain the precise gene expression patterns of thousands of cells in the intestine, analyze the characteristics of cells with the same phenotype, and provide new insights into the growth and development of intestinal organs, the clonal evolution of cells, and immune cell changes. These findings can provide new ideas for the diagnosis and treatment of intestinal diseases.To identify clinical phenotypes and biomarkers that can predict the response of UC patients to specific therapeutic drugs and thus aid the diagnosis and treatment of UC.Using the Gene Expression Omnibus (GEO) database, we analyzed peripheral blood cell subtypes of patients with UC by scRNA-seq combined with bulk RNA sequencing (RNA-seq) to reveal the core genes of UC. We then combined weighted gene correlation network analysis (WGCNA) and least absolute shrinkage and selection operator (LASSO) analysis to reveal diagnostic markers of UC.After processing the scRNA-seq data, we obtained data from approximately 24340 cells and identified 17 cell types. Through intercellular communication analysis, we selected monocyte marker genes as the candidate gene set for the prediction model. Construction of a WGCNA coexpression network identified RhoB, cathepsin D (CTSD) and zyxin (ZYX) as core genes. Immune infiltration analysis showed that these three core genes were strongly correlated with immune cells. Functional enrichment analysis showed that the differentially expressed genes were closely related to immune and inflammatory responses, which are associated with many challenges in the diagnosis and treatment of UC.Through scRNA-seq analysis, LASSO diagnostic model building and WGCNA, we identified RhoB, CTSD and ZYX as core genes of UC that are closely related to monocyte infiltration that may serve as diagnostic markers and molecular targets for UC therapeutic intervention.

Background Production of vitamin C has been traditionally based on the Reichstein process and the two-step process. However, the two processes share a common disadvantage: vitamin C cannot be directly synthesized from D-glucose. Therefore, significant effort has been made to develop a one-step vitamin C fermentation process. While, 2-KLG, not vitamin C, is synthesized from nearly all current one-step fermentation processes. Vitamin C is naturally synthesized from glucose in Arabidopsis thaliana via a ten-step reaction pathway that is encoded by ten genes. The main objective of this study was to directly produce vitamin C from D-glucose in Escherichia coli by expression of the genes from the A. thaliana vitamin C biosynthetic pathway. Results Therefore, the ten genes of whole vitamin C synthesis pathway of A. thaliana were chemically synthesized, and an engineered strain harboring these genes was constructed in this study. The direct production of vitamin C from D-glucose based on one-step fermentation was achieved using this engineered strain and at least 1.53 mg/L vitamin C was produced in shaking flasks. Conclusions The study demonstrates the feasibility of one-step fermentation for the production of vitamin C from D-glucose. Importantly, the one-step process has significant advantages compared with the currently used fermentation process: it can save multiple physical and chemical steps needed to convert D-glucose to D-sorbitol; it also does not involve the associated down-streaming steps required to convert 2-KLG into vitamin C.

To quantify the impacts of native vegetation on the spatial and temporal variations in hydraulic properties of bank gully concentrated flows, a series of in situ flume experiments in the bank gully were performed at the Yuanmou Gully Erosion and Collapse Experimental Station in the dry-hot valley region of the Jinsha River, Southwest China. This experiment involved upstream catchment areas with one- and two-year native grass (Heteropogon contortus) and bare land drained to bare gully headcuts, i.e., Gullies 1, 2 and 3. In Gully 4, Heteropogon contortus and Agave sisalana were planted in the upstream catchment area and gully bed, respectively. Among these experiments, the sediment concentration in runoff in Gully 3 was the highest and that in Gully 2 was the lowest, clearly indicating that the sediment concentration in runoff obviously decreased and the deposition of sediment obviously increased as the vegetation cover increased. The concentrated flows were turbulent in response to the flow discharge. The concentrated flows in the gully zones with native grass and bare land were sub- and supercritical, respectively. The flow rate and shear stress in Gully 3 upstream catchment area were highest among the four upstream catchment areas, while the flow rate and shear stress in the gully bed of Gully 4 were lowest among the four gully beds, indicating that native grass notably decreased the bank gully flow rate and shear stress. The Darcy-Weisbach friction factor (resistance f) and flow energy consumption in the gully bed of Gully 4 were notably higher than those in the other three gully beds, clearly indicating that native grass increased the bank gully surface resistance and flow energy consumption. The Reynolds number (Re), flow rate, shear stress, resistance f, and flow energy consumption in the gully beds and upstream areas increased over time, while the sediment concentration in runoff and Froude number (Fr) decreased. Overall, increasing vegetation cover in upstream catchment areas and downstream gully beds of the bank gully is essential for gully erosion mitigation.

The Badain Jaran Desert is the third largest desert in China, covering an area of 50000 km2. It lies in Northwest China, where the arid and rainless natural environment has a great impact on the climate, environment, and human living conditions. Based on the results of 1∶250000 regional hydrogeological surveys and previous researches, this study systematically investigates the circulation characteristics and resource properties of the groundwater as well as the evolution of the climate and ecological environment since the Quaternary in the Badain Jaran Desert by means of geophysical exploration, hydrogeological drilling, hydrogeochemistry, and isotopic tracing. The results are as follows. (1) The groundwater in the Badain Jaran Desert is mainly recharged through the infiltration of local precipitation and has poor renewability. The groundwater recharge in the desert was calculated to be 1.8684×108 m3/a using the water balance method. (2) The Badain Jaran Desert has experienced four humid stages since the Quaternary, namely MIS 13-15, MIS 5, MIS 3, and the Early–Middle Holocene, but the climate in the desert has shown a trend towards aridity overall. The average annual temperature in the Badain Jaran Desert has significantly increased in the past 50 years. In detail, it has increased by about 2.5°C, with a higher rate in the south than in the north. Meanwhile, the precipitation amount has shown high spatial variability and the climate has shown a warming-drying trend in the past 50 years. (3) The lakes in the hinterland of the Badain Jaran Desert continuously shrank during 1973–2015. However, the vegetation communities maintained a highly natural distribution during 2000–2016, with the vegetation cover has increased overall. Accordingly, the Badain Jaran Desert did not show any notable expansion in that period. This study deepens the understanding of groundwater circulation and the climate and ecological evolution in the Badain Jaran Desert. It will provide a scientific basis for the rational exploitation of the groundwater resources and the ecological protection and restoration in the Badain Jaran Desert. © 2021 China Geology Editorial Office.

The photophysical and charge transport properties of five non-planar cyclopenta-fused polycyclic aromatic hydrocarbons including dibenzo[a,m]rubicene (molecule 1) and its derivatives are studied through density functional theory (DFT). The absorption and emission spectra (λab and λem) are calculated at the B3LYP/6-31+G(d,p), PBE0/6-31+G(d,p), BMK/6-31+G(d,p), M062X/6-31+G(d,p) and CAM-B3LYP/6-31+G(d,p) levels in dichloromethane solvent. The results show that the BMK and M062X functionals can reproduce experimental λab and λem more accurately than other functionals, respectively. The charge mobilities can be effectively tuned through introducing silyl groups of different sizes (molecules 2, 3 and 4) and the 1,3-dimethoxy benzene group (molecule 5) to dibenzo[a,m]rubicene. The relationship between the structures of five dibenzo[a,m]rubicene derivatives and a set of relevant electronic properties including the molecular geometries, frontier molecular orbitals, ionization potentials, electron affinities, reorganization energies and transfer integrals have been established. The results show that these molecules possess prominent charge carrier mobilities, although they all possess non-planar geometries. The charge transport properties could be effectively modulated via introducing different substituents to molecule 1. The high hole mobilities indicate that all five molecules facilitate hole transport, which is in accordance with the experimental results. Intriguingly, the electron mobility is enhanced maximally by five times through introducing silyl groups of different sizes from 0.06 cm2 V−1 s−1 (molecule 1) to 0.30 cm2 V−1 s−1 (molecule 4). Particularly, molecules 4 and 5 possess balanced electron and hole mobilities, which could be used as bipolar semiconductors and show promising use in optoelectronic devices.
