Aming to select a suitable system for the synergistic solvent extraction of Ga(III), solvent extraction (SX) experiments were carried out by using acidic organo-phosphorous extractants, namely P204, P507 and Cyanex 272, and their mixtures with neutral extractants (TBP, N503) or amine extractants (Alamine 336). Among the studied mixtures, a remarkable synergistic effect for Ga(III) extraction was resulted by the combination of Cyanex 272 and Alamine 336 and the maximal synergistic enhancement coefficient reached 3.11. Compared to the single Cyanex 272, higher equilibrium pH values were observed when mixture of Cyanex 272 and Alamine 336 was employed. The extraction of hydrogen ions by Alamine 336 during the reaction was considered to be responsible for the synergistic extraction, and thus the the extraction of Ga(III) by Cyanex 272 was promoted. Finally, the synergistic extraction stoichiometry of mixture of Cyanex 272 and Alamine 336 for Ga(III) was projected by slope analysis. Ga(OH)A2 was resulted to be the composition of extracted complexes.

Photodynamic therapy (PDT) with an oxygen-dependent character is a noninvasive therapeutic method for cancer treatment.

A powder thixoforging route combined with slurry based mixing process was proposed to fabricate graphene nanoplatelets (GNPs) reinforced magnesium matrix composites (MgMCs). The originally spherical and ball-milled ZK60 powders were used as matrices, respectively. The mixing of 0.05 wt.% GNPs with the spherical powder led to GNPs clusters and degraded the mechanical properties of the composite. In contrast, with the addition of an optimal content (0.1 wt.%) of GNPs, the composite fabricated from ball-milled powder achieved a joint enhancement in tensile yield strength (52%) and fracture toughness (19%), demonstrating a pronounced strengthening efficiency of 650% and a good balance between strength and toughness. The ball-milled powder endowed the composite with a homogenous distribution of GNPs and a denser microstructure with reduced Mg-Zn eutectics, and the thixoforging process offered a well-bonded Mg/GNP interface, making full use of the strengthening and toughening potential of GNPs. Theoretical predication based on a modified shear-lag model suggested that load transfer dominated the strengthening mechanisms. In-situ tensile tests verified that crack deflection, secondary cracks and GNPs bridging mainly accounted for the toughening mechanisms. A numerical model with consideration of GNPs orientations was also established to understand the toughening effect from GNPs bridging.

The autoclave curing process of the large-scale composite stiffened panel greatly affects the manufacturing quality of the component, especially the curing quality of stiffeners and the bonding quality between stiffeners and the panel. In this paper, a multi-objective optimization method is developed to optimize the curing cycle for the composite T-stiffened fuselage panel, which aims to minimize the temperature difference within the component to achieve better forming quality. The autoclave curing model considering thermal properties of composites is established and verified by the experiment first. The response surface models are then built up with the Finite Element Method (FEM) and the Box-Behnken Design (BBD). The fgoalattain algorithm is applied to solve the multi-objective optimization problem with nonlinear constraints. Results show that the maximum temperature difference of the two groups of monitoring points decreases by 22.91% and 21.42%, respectively. Besides, the total curing time is also reduced by 11 mins.

The copper ion content in the body maintains homeostasis, and when dysregulated, it can produce cytotoxicity and induce cell death through a variety of pathways. Cuproptosis refers to copper ions combining directly with acylated molecules, leading to the accumulation of oligomerization of lipoylated protein and subsequent downregulation of iron-sulfur cluster proteins; this induces proteotoxic stress and cell death. This study on the relationship between cuproptosis-related lncRNAs (CRLns) and the prognosis of primary hepatic carcinoma (PHC) has important clinical guiding significance for the diagnosis and treatment of PHC. Prognosis-related CRLRs were identified via rank-sum tests, correlational analyses, and univariate Cox regression, and a CRLR risk-scoring model (CRLRSM) was constructed using LASSO Cox regression. Patients were divided into high-risk and low-risk groups based on the median CRLRSM scores. Variance analysis for cuproptosis-related genes, gene set enrichment analysis, and correlational analysis for risk and immunity were performed using boxplots. Quantitative polymerase chain reactions were used to verify the CRLR levels in PHC cell lines. The study results showed that patients in the CRLRSM high-risk group had worse survival rates than those in the low-risk group. The PHC stage and risk score were independent prognostic factors for hepatocellular carcinoma. There were 7 CRLRs (MIR210HG, AC099850.3, AL031985.3, AC012073.1, MKLN1-AS, KDM4A-AS1, and PLBD1-AS1) associated with PHC prognosis, primarily through cellular metabolism, growth, proliferation, apoptosis, and immunity. In conclusion, the overexpression of 7 CRLRs in patients with PHC indicates a poor prognosis.

Background A non-invasive tool for tumor regression grade (TRG) evaluation is urgently needed for gastric cancer (GC) treated with neoadjuvant chemotherapy (NAC). Purpose To develop and validate a radiomics signature (RS) to evaluate TRG for locally advanced GC after NAC and assess its prognostic value. Material and Methods A total of 103 patients with GC treated with NAC were retrospectively recruited from April 2018 to December 2019 and were randomly allocated into a training cohort (n = 69) and a validation cohort (n = 34). Delineation was performed on both mixed and iodine-uptake images based on dual-energy computed tomography (DECT). A total of 4094 radiomics features were extracted from the pre-NAC, post-NAC, and delta feature sets. Spearman correlation and the least absolute shrinkage and selection operator were used for dimensionality reduction. Multivariable logistic regression was used for TRG evaluation and generated the optimal RS. Kaplan–Meier survival analysis with the log-rank test was implemented in an independent cohort of 40 patients to validate the prognostic value of the optimal RS. Results Three, five, and six radiomics features were finally selected for the pre-NAC, post-NAC, and delta feature sets. The delta model demonstrated the best performance in assessing TRG in both the training and the validation cohorts (AUCs=0.91 and 0.76, respectively; P>0.1). The optimal RS from the delta model showed a significant capability to predict survival in the independent cohort ( PConclusion Delta radiomics based on DECT images serves as a potential biomarker for TRG evaluation and shows prognostic value for patients with GC treated with NAC.

Coronary motion artifacts affect the diagnostic accuracy of coronary CT angiography (CCTA), especially in the mid right coronary artery (mRCA). The purpose is to correct CCTA motion artifacts of the mRCA using a GAN (generative adversarial network).We included 313 patients with CCTA scans, who had paired motion-affected and motion-free reference images at different R-R interval phases in the same cardiac cycle and included another 53 CCTA cases with invasive coronary angiography (ICA) comparison. Pix2pix, an image-to-image conversion GAN, was trained by the motion-affected and motion-free reference pairs to generate motion-free images from the motion-affected images. Peak signal-to-noise ratio (PSNR), structural similarity (SSIM), Dice similarity coefficient (DSC), and Hausdorff distance (HD) were calculated to evaluate the image quality of GAN-generated images.At the image level, the median of PSNR, SSIM, DSC, and HD of GAN-generated images were 26.1 (interquartile: 24.4-27.5), 0.860 (0.830-0.882), 0.783 (0.714-0.825), and 4.47 (3.00-4.47), respectively, significantly better than the motion-affected images (p0.001). At the patient level, the image quality results were similar. GAN-generated images improved the motion artifact alleviation score (4 vs. 1, p0.001) and overall image quality score (4 vs. 1, p0.001) than those of the motion-affected images. In patients with ICA comparison, GAN-generated images achieved accuracy of 81%, 85%, and 70% in identifying no,50%, and ≥ 50% stenosis, respectively, higher than 66%, 72%, and 68% for the motion-affected images.Generative adversarial network-generated CCTA images greatly improved the image quality and diagnostic accuracy compared to motion-affected images.• A generative adversarial network greatly reduced motion artifacts in coronary CT angiography and improved image quality. • GAN-generated images improved diagnosis accuracy of identifying no,50%, and ≥ 50% stenosis.

Objectives To evaluate robustness of dual-energy CT (DECT) radiomics features of virtual unenhanced (VUE) image and virtual monoenergetic image (VMI) among different imaging platforms. Methods A phantom with sixteen clinical-relevant densities was scanned on ten DECT platforms with comparable scan parameters. Ninety-four radiomic features were extracted via Pyradiomics from VUE images and VMIs at energy level of 70 keV (VMI70keV). Test–retest repeatability was assessed by Bland–Altman analysis. Inter-platform reproducibility of VUE images and VMI70keV was evaluated by coefficient of variation (CV) and quartile coefficient of dispersion (QCD) among platforms, and by intraclass correlation coefficient (ICC) and concordance correlation coefficient (CCC) between platform pairs. The correlation between variability of CT number radiomics reproducibility was estimated. Results 92.02% and 92.87% of features were repeatable between scan–rescans for VUE images and VMI70keV, respectively. Among platforms, 11.30% and 28.39% features of VUE images, and 15.16% and 28.99% features of VMI70keV were with CV 0.90 and CCC > 0.90 between platform pairs were 10.00% and 9.86% in VUE images and 11.23% and 11.23% in VMI70keV. The CT number inter-platform reproducibility using CV and QCD showed negative correlations with percentage of the first-order radiomics features with CV 70keV (r2 0.3870–0.6178, all p Conclusions The majority of DECT radiomics features were non-reproducible. The differences in CT number were considered as an indicator of inter-platform DECT radiomics variation. Critical relevance statement: The majority of radiomics features extracted from the VUE images and the VMI70keV were non-reproducible among platforms, while synchronizing energy levels of VMI to reduce the CT number value variability may be a potential way to mitigate radiomics instability. CreTA, CRISPR-regulated toxin-antitoxin (TA), safeguards CRISPR-Cas immune systems by inducing cell dormancy/death upon their inactivation. Here, we characterize a bacterial CreTA associating with the I-F CRISPR-Cas in Acinetobacter. CreT is a distinct bactericidal small RNA likely targeting several essential RNA molecules that are required to initiate protein synthesis. CreA guides the CRISPR effector to transcriptionally repress CreT. We further demonstrate a proof-of-concept antimicrobial strategy named ATTACK, which AssociaTes TA and CRISPR-Cas to Kill multidrug resistant (MDR) pathogens. In this design, CRISPR-Cas is programed to target antibiotic resistance gene(s) to selectively kill MDR pathogens or cure their resistance, and when CRISPR-Cas is inactivated or suppressed by unwanted genetic or non-genetic events/factors, CreTA triggers cell death as the last resort. Our data highlight the diversity of RNA toxins coevolving with CRISPR-Cas, and illuminate a combined strategy of CRISPR and TA antimicrobials to ‘ATTACK’ MDR pathogens.

CRISPR RNAs (crRNAs) and Cas proteins together provide prokaryotes with adaptive immunity against genetic invaders. How Cas expression is fine-tuned to avoid energy burden while satisfying the dynamic need of crRNAs remains poorly understood. Here we experimentally demonstrated widespread degenerated mini-CRISPRs encode CreR (Cas-regulating)RNAs to mediate autorepression of type I-B, I-E and V-A Cas proteins, based on their partial complementarity tocaspromoters. This autorepression decreases energy burden and autoimmune risks, thus mitigating the fitness cost on host cell, and remarkably, senses and responds to alterations in the volume of canonical crRNAs, which compete with CreR for Cas proteins. Moreover, CreR-guided Cas autorepression can be subverted by diverse anti-CRISPR (Acr) proteins that destruct Cas proteins, which in turn replenishes the weapon depot. Our data unveil a general degenerated crRNA-guided autorepression paradigm for diverse Cas effectors, which highlights the intricate (self-)regulation of CRISPR-Cas and its transcriptional counterstrategy against Acr attack.

The temperature distribution of the mold is directly linked to the manufacturing quality of the composite component. In this paper, a bionic mold based on the analysis of a manta ray in swimming is developed to improve the thermal performance of the mold during the autoclave curing process. The bionic mold has fewer frames and better air permeability, and the mold weight is decreased by 42.95% while the stiffness is proved to satisfy the requirement. The model of the autoclave curing process which is verified by experiments is established to carry out the thermal analysis of the mold. Results show that the air velocity in the mold is increased with the bionic design, which can reduce not only the maximum temperature difference of the mold plate (−30.35%) but also the percentage of low-temperature areas. Besides, the heating rate and the heat preservation time of the mold are also improved.

To investigate the prognostic value of dual-energy CT (DECT) based radiomics to predict disease-free survival (DFS) and overall survival (OS) for patients with advanced gastric cancer (AGC) after neoadjuvant chemotherapy (NAC).From January 2014 to December 2018, a total of 156 AGC patients were enrolled and randomly allocated into a training cohort and a testing cohort at a ratio of 2:1. Volume of interest of primary tumor was delineated on eight image series. Four feature sets derived from pre-NAC and delta radiomics were generated for each survival arm. Random survival forest was used for generating the optimal radiomics signature (RS). Statistical metrics for model evaluation included Harrell's concordance index (C-index) and the average cumulative/dynamic AUC throughout follow-up. A clinical model and a combined Rad-clinical model were built for comparison.The pre-IU (derived from iodine uptake images before NAC) RS performed best for DFS and OS in the testing cohort (C-indices, 0.784 and 0.698; the average cumulative/dynamic AUCs, 0.80 and 0.77). When compared with the clinical model, the radiomics model had significantly higher C-index to predict DFS in the testing cohort (0.784 vs. 0.635, p 0.001), but no statistical difference was found for OS (0.698 vs. 0.680, p = 0.473). The combined Rad-clinical models showed improved performance in the testing cohort, with C-indices of 0.810 and 0.710 for DFS and OS, respectively.DECT-derived radiomics serves as a promising non-invasive biomarker to predict survival for AGC patients after NAC, providing an opportunity for transforming proper treatment.

Peach shoot blight (PSB), caused by Phomopsis amygdali, is a serious threat to the healthy development of the peach industry and leads to 30 to 50% damage to peach production in southern China. In this study, loop-mediated isothermal amplification (LAMP) technology was used to detect the P. amygdali target of a gene of GME6801 that was unique in the whole genome of the pathogen compared with that of Diaporthe (Phomopsis) longicolla TWH P74, Fusarium graminearum PH-1, Colletotrichum gloeosporioides SMCG1 and Magnaporthe oryzae 70-15. Blast comparison of this gene sequence in NCBI database showed that no homologous sequences were found. Therefore, the gene sequence of GME6801 was used to design two pairs of LAMP primers and one pair of PCR primers. The results showed that both primer sets were specific to the 15 strains of P. amygdali, and the other 15 fungal strains presented negative reactions, similar to the control. In addition, 50 pg of genomic DNA of P. amygdali in a 25-μl reaction system could be detected by LAMP assay, which was 100 times more sensitive than PCR. Furthermore, the GME6801 LAMP assay was used to detect artificially inoculated twigs of the pathogen, disease twigs within significantly symptomatic PSB in the fields, and healthy twigs in the same orchard, with detection rates of 100, 75, and 20.8%, respectively. However, detection rates of conventional PCR were separately 100, 62.5, and 16.7%. The results indicated that GME6801-based LAMP could be used for P. amygdali detection as its specificity, sensitivity, and simplicity. This study provides a rapid experimental basis for the identification and prediction of P. amygdali that causes PSB and is beneficial for precise prevention and control of the disease.

Gestational diabetes mellitus (GDM) is a health risk for pregnant women and infants. Emerging evidence suggests that the deregulation of circular RNAs (circRNAs) is associated with the progression of this disorder. The objective of this study was to investigate the role of circ_FOXP1 in GDM. Cell models of GDM were established by treating human trophoblast cells with high glucose (HG). The expression of circ_FOXP1, miR-508-3p and SMAD family member 2 (SMAD2) mRNA was detected by quantitative real-time PCR (qPCR). Cell proliferation was assessed by EdU assay and MTT assay, and cell cycle and cell apoptosis were determined by flow cytometry assay. The protein levels of proliferation- and apoptosis-related markers and SMAD2 were measured by western blot. The relationship between miR-508-3p and circ_FOXP1 or SMAD2 was validated by dual-luciferase reporter assay or pull-down assay. The expression of circ_FOXP1 was downregulated in HG-treated HTR-8/SVneo cells. Circ_FOXP1 overexpression promoted HG-inhibited HTR-8/SVneo cell proliferation and suppressed HG-induced HTR-8/SVneo cell cycle arrest and apoptosis. Circ_FOXP1 positively regulated the expression of SMAD2 by targeting miR-508-3p. MiR-508-3p was overexpressed in HG-treated HTR-8/SVneo cells, and its overexpression reversed the effects of circ_FOXP1 overexpression. MiR-508-3p inhibition also alleviated HG-induced HTR-8/SVneo cell injuries, while the knockdown of SMAD2 abolished these effects. Collectively, circ_FOXP1 promotes the growth and survival of HG-treated human trophoblast cells through the miR-508-3p/SMAD2 pathway, hinting that circ_FOXP1 was involved in GDM progression.

Peach shoot blight (PSB), which kills shoots, newly sprouted leaf buds, and peach fruits, has gradually increased over the last 10 years and resulted in 30 to 50% of total production loss of the peach industry in China. Phomopsis amygdali has been identified as the common causal agent of this disease. In this study, two new species, Phomopsis liquidambaris (strain JW18-2) and Diaporthe eres (strain JH18-2), were also pathogens causing PSB, as determined through molecular phylogenetic analysis based on the sequences of the internal transcribed spacer (ITS) region, translation elongation factor 1-α (EF1-α) and beta-tubulin (TUB), and colony and conidial morphological characteristics. Biological phenotypic analysis showed that the colony growth rate of strain JW18-2 was faster than that of strains JH18-2 and ZN32 (one of the P. amygdali strains that we previously found and identified). All three strains produced α-conidia; however, JW18-2 could not produce β-conidia on alfalfa decoction and Czapek media, and the β-conidia produced by strain JH18-2 were shorter in length and thicker in width than those produced by strain ZN32. Pathogenicity tests showed that JW18-2 presented the strongest pathogenicity for peach fruits and twigs and was followed by strains JH18-2 and ZN32. The results shed light on the etiology of PSB and provide a warning that P. liquidambaris or D. eres might develop into dominant species after a few years while also potentially benefitting the development of effective disease control management strategies.

Aggregation-induced emission luminogens (AIEgens) exhibit efficient cytotoxic reactive oxygen species (ROS) generation capability and unique light-up features in the aggregated state, which have been well explored in image-guided photodynamic therapy (PDT). However, the limited penetration depth of light in tissue severely hinders AIEgens as a candidate for primary or adjunctive therapy for clinical applications. Coincidentally, microwaves (MWs) show a distinct advantage for deeper penetration depth in tissues than light. Herein, for the first time, we report AIEgen-mediated microwave dynamic therapy (MWDT) for cancer treatment. We found that two AIEgens (TPEPy-I and TPEPy-PF6) served as a new type of microwave (MW) sensitizers to produce ROS, including singlet oxygen (1O2), resulting in efficient destructions of cancer cells. The results of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and live/dead assays reveal that the two AIEgens when activated by MW irradiation can effectively kill cancer cells with average IC-50 values of 2.73 and 3.22 μM, respectively. Overall, the ability of the two AIEgens to be activated by MW not only overcomes the limitations of conventional PDT, but also helps to improve existing MW ablation therapy by reducing the MW dose required to achieve the same therapeutic outcome, thus reducing the occurrence of side-effects of MW radiation., Graphical abstract Image 1, Highlights • For the first time, microwave-induced photodynamic therapy using AIEgens is reported. • The AIEgens showed significant cytotoxicity when excited by microwave toward HeLa cells. • AIEgens are promising candidates for image-guided microwave dynamic therapy. • This novel work opens a new door for AIEgens applications.

The development of the Internet of Things (IoT) technology has achieved remarkable results in recent years. A large number of sensors and machinery are connected, and combined with the Internet, to achieve intelligent management and operation. However, the traditional centralized IoT data management solution inevitably encounters challenges of data trust, security, sustainability, and user privacy is difficult to guarantee. This paper takes the intelligent transportation sensor network as an example and proposes a blockchain-based Internet of Vehicles (IoV) data secure sharing scheme (called IoVChain), which implements automatic registration, rapid authentication, and reliable sharing method of IoV data through smart contract. The smart contract performs homomorphic encryption and zero-knowledge proof processing on the sensitive part of the data, and this part exists in the form of ciphertext on the blockchain. We adopt PBFT consensus mechanism to ensure the consistency of the entire network ledger, and all the IoV data processing and usage procedure is stored in the Merkle-tree-based block, which cannot be tampered with. Compared with the centralized IoV data management solution, the IoVChain scheme avoids the risk of single point of failure, and keeps the IoV data credible, available, and tamper-resistant under the premise of privacy and security, so that the data can be traced when needed. We have implemented the IoVChain scheme based on the consortium blockchain for trusted and secure sharing of IoV data. Finally, analysis and evaluation show that the proposed IoVChain scheme is feasible, safe, and extensible for the secure sharing of IoV data.

LC resonant wireless passive pressure sensor has been receiving increased attention nowadays. In this paper, an innovative LC-C resonant wireless passive pressure sensor based on dual capacitors was designed and manufactured to replace the traditional LC device. In addition to the inductor and capacitor structure required by the traditional LC device, this paper innovatively designs the fixed capacitor as the inter-layer interconnection structure. The key structural dimensions of the sensor were determined through the numerical analysis of the planar inductor theory and the electromechanical coupling simulation of the dual capacitors model in the previous stage, and the processing of the sensor was carried out through MEMS technology. The final test results showed that within the pressure range of 5kPa to 100kPa, the sensor’s average sensitivity was greater than 69.0 kHz/kPa, while the maximum linear fitting error was smaller than 4.18%. The temperature drift in atmospheric environment was 2.7kHz/°C, and the sensitivity temperature drift was 0.0062kHz/(kPa·°C).

The cleavage of DMF into a dimethyl carbamoyl radical under mild electrochemical conditions was revealed for the first time. Meanwhile, an electrochemical decarboxylation of α-ketoacid occurred to produce an acyl radical. The radical cross-coupling reaction of these two electron-deficient acyl radicals was carried out with high selectivity. This discovery provides a new electrochemical way for the cracking of DMF, and a milder and safer method for its application in organic synthesis.

Gardenia jasminoides Ellis, a typical representative for “homology of medicine and food”, can be used to produce gardenia yellow and edible oil. Here, aqueous enzymatic extraction (AEE) combined with puffing pre-treatment was explored to prepare oil from gardenia seeds. Free oil yield (FOY) by AEE was evaluated in term of selection of enzyme, enzyme combination, as well as puffing ways and conditions. Results suggested that seeds taken as raw materials were better than whole fruit, sequential hydrolysis was superior to single enzymatic hydrolysis. Both wet-heating puffing at 90℃ and dry-heating puffing at 1.0MPa facilitated the release of free oil by AEE, while FOY produced the highest oil yields of 21.8% and 23.2% within 3h, and 58.3%, 68.4% higher than that of un-puffed group, active crocin and geniposide were also completely released. The FOY obtained in this study was far higher than pressing method (10.44%), and close to solvent extraction (25.45%). Also, microstructure analysis indicated that gardenia seeds expanded by dry-heating puffing (1.0MPa) had larger and rougher surface and porous structure than other groups. Moreover, there was no significant difference between oils prepared by various methods in term of nutritional and safety characteristics. AEE coupled with puffing pre-treatment developed is an eco-friendly extraction technology with high efficiency that can be employed to oil preparation enriched with active crocin and geniposide from gardenia seeds.
