La glutammato deidrogenasi NAD(H)-dipendente (GDH; EC 1.4.1.2) è un enzima abbondante presente in differenti forme isoenzimatiche. In Arabidopsis thaliana sono presenti tre geni (GDH1, GDH1 e GDH3) che codificano per tre differenti subunità (b, a e g) in grado di associarsi casualmente per formare omo- ed etero- esameri della GDH. In questo lavoro di dottorato, sono state analizzate le variazioni del pattern isoenzimatico durante lo sviluppo della pianta Arabidopsis thaliana. In particolare la nostra attenzione si è concentrata sulla regolazione delle isoforme della GDH codificate da GDH3. Per fare questo ci siamo avvalsi della disponibilità di singoli e doppio mutanti della GDH, compromessi nell’espressione dei geni GDH1 e/o GDH2. Il ruolo fisiologico dell’isoforma GDH3 è stato analizzato in piante sottoposte a vari trattamenti nutrizionali e ormonali. I nostri risultati suggeriscono per la GDH3 un possibile ruolo durante la senescenza delle piante e /o durante i processi di rimobilizzazione dei nutrienti. Abbiamo analizzato anche le caratteristiche biochimiche degli isoenzimi della GDH, valutando la loro termostabilità: l’espressione in lievito delle isoforme GDH1 e GDH3 WT o mutate ha indicato che il carbossi- terminale delle subunità della GDH è coinvolto nella stabilizzazione della struttura oligomerica dell’enzima. In Arabidopsis thaliana three genes (GDH1, GDH2 and GDH3) encodes three different GDH subunits (b, a and g) that randomly associate to form a complex array of homo- and hetero- GDH hexhamers. In this report we have analysed the modification of the GDH isozyme pattern during Arabidopsis plant development. In particular our attention was focused on the regulation of the GDH3 encoded GDH isoforms. For this we take advantage of the availability of single and double GDH mutants impaired in the expression of GDH1 and/or GDH2 genes. The physiological role of the GDH3 isoform was investigated in plants subjected to various nutritional and hormonal treatments. Our results suggested a possible role for GDH3 during plant senescence and/or nutrient remobilization processes . We have also analysed the biochemical characteristic of the GDH isoenzymes by evaluating their thermal stability: expression in yeast of WT and mutated GDH1 and GDH3 isoforms indicated that the carboxyl-end of the GDH subunits is involved in the stabilization of the oligomeric structure of the enzyme.
