Liver transplantation (LT) is the best treatment for patients with hepatocellular carcinoma (HCC). However, the surgical technique needs to be improved. The present study aimed to evaluate the "no-touch" technique in LT.From January 2018 to December 2019, we performed a prospective randomized controlled trial on HCC patients who underwent LT. The patients were randomized into two groups: a no-touch technique LT group (NT group, n = 38) and a conventional LT technique group (CT group, n = 46). Operative outcomes and survival in the two groups were analyzed.The perioperative parameters were comparable between the two groups (P0.05). There was no significant difference between the two groups in disease-free survival (DFS) (P = 0.732) or overall survival (OS) (P = 0.891). Of 36 patients who were beyond the Hangzhou criteria for LT, the DFS of the patients in the NT group was significantly longer than that in the CT group (median 402 vs. 126 days, P = 0.025). In 31 patients who had portal vein tumor thrombosis (PVTT), DFS and OS in the NT group were significantly better than those in the CT group (median DFS 420 vs. 167 days, P = 0.022; 2-year OS rate 93.8% vs. 66.7%, P = 0.043). In 14 patients who had diffuse-type HCCs, DFS and OS were significantly better in the NT group than in the CT group (median DFS 141 vs. 56 days, P = 0.008; 2-year OS rate 75.0% vs. 33.3%, P = 0.034). Multivariate analysis showed that for patients with PVTT and diffuse-type HCCs, the no-touch technique was an independent favorable factor for OS (PVTT: HR = 0.018, 95% CI: 0.001-0.408, P = 0.012; diffuse-type HCCs: HR = 0.034, 95% CI: 0.002-0.634, P = 0.024).The no-touch technique improved the survival of patients with advanced HCC compared with the conventional technique. The no-touch technique may provide a new and effective LT technique for advanced HCCs.

In a coil magnetometer, the size and uniformity of the bias magnetic field generated by the Helmholtz coil directly determine the accuracy of the solution of the geomagnetic direction. The design of traditional spherical coils relies heavily on the manual experience or mathematical derivation, making it difficult to obtain optimal parameters or requiring larger spherical coils. To address the problem, first, a coaxial symmetrical spherical coil model that improves space utilization was established. Second, an optimal design method for the spherical magnetic field generation coil based on a differential evolution algorithm was proposed. Third, the optimal bias magnetic field was obtained without increasing the volume of the coil. The verification results showed that the magnetic non-uniformity and magnetic gradient of the bias field generated by the optimized coil were reduced by 63.2% and 82.8%, respectively.

Real neural networks are characterized by large-scale and complex topology. However, the current dynamical analysis is limited to low-dimensional models with simplified topology. Therefore, there is still a huge gap between neural network theory and its application. This article proposes a class of large-scale neural networks with a ring-hub structure, where a hub node is connected to n peripheral nodes and these peripheral nodes are linked by a ring. In particular, there exists a hybrid coupling mode in the network topology. The mathematical model of such systems is described by fractional-order delayed differential equations. The aim of this article is to investigate the local stability and Hopf bifurcation of this high-dimensional neural network. First, the Coates flow graph is employed to obtain the characteristic equation of the linearized high-dimensional neural network model, which is a transcendental equation including multiple exponential items. Then, the sufficient conditions ensuring the stability of equilibrium and the existence of Hopf bifurcation are achieved by taking time delay as a bifurcation parameter. Finally, some numerical examples are given to support the theoretical results. It is revealed that the increasing time delay can effectively induce the occurrence of periodic oscillation. Moreover, the fractional order, the self-feedback coefficient, and the number of neurons also have effects on the onset of Hopf bifurcation.

Epstein-Barr virus (EBV)-associated T-/natural killer (T/NK)-cell lymphoproliferative diseases clinically take on various forms, ranging from an indolent course to an aggressive condition.Clinically, failure to establish precise diagnosis and provide proper treatment makes it difficult to help patients. We sought to better understand the underlying pathogenesis and to identify genetic prognostic factors to achieve better treatment efficacy.In this study, 119 cases of EBV-associated lymphoproliferative diseases, including EBV-associated hemophagocytic lymphohistiocytosis (n = 46) and chronic active EBV disease of T/NK cell type (n = 73), were retrospectively examined.Adults aged20 years at onset accounted for 71.4% of our cohort. About 54.6% patients with unfavorable overall survival developed hemophagocytic lymphohistiocytosis and had higher plasma EBV load. Allogenic hematopoietic stem-cell transplantation was the sole independent favorable factor. We systematically screened germline and somatic aberrations by whole-exome and targeted sequencing. Among 372 antiviral immunity genes, germline variants of 8 genes were significantly enriched. From a panel of 24 driver genes, somatic mutations were frequently identified in dominant EBV-infected T/NK cells. Patients carrying any germline/somatic aberrations in epigenetic modifiers and RIG-I-like receptor (RLR) pathway had worse overall survival than those without 2 type aberrations. Importantly, patients with IFIH1 and/or DDX3X aberrations in the RLR pathway had higher plasma and NK-cell EBV load. Knockdown of DDX3X in NKYS cells downregulated RLR signaling activities and elevated the expression of EBV-encoded oncogenes such as LMP1 and EBNA1.Genetic defects were prevalent in adult EBV-associated hemophagocytic lymphohistiocytosis patients and patients with chronic active EBV disease of T/NK cell type; these defects were associated with unfavorable prognosis. These findings can help clinicians work out more precise staging of the condition and provide new insights into these EBV-associated diseases.

This study discusses the application of fuzzy adaptive PID and improved genetic algorithm (IGA) in motion control and path optimization of autonomous underwater vehicle (AUV). The fuzzy adaptive PID method is selected because it is considered to be a strongly nonlinear and coupled system. First, this study creates the basic coordinate system of the AUV, and then analyzes the spatial force from the AUV to obtain the control model of the heading angle, climb angle, and depth. Next, the knowledge of fuzzy adaptive PID and IGA technology on AVU are investigated, then fuzzy adaptive PID controllers and path optimization are established, and experimental simulations are carried out to compare and analyze the simulation results. The research results show that controllers and IGA can be used for the motion control and path optimization of AUV. The advantages of fuzzy adaptive PID control are less overload, enhanced system stability, and more suitable for motion control and path optimization of AUV.

A computational study was conducted to investigate the mechanism of a new type of C–N bond metathesis reaction, in which a reversible reductive elimination/oxidative addition mode is involved.

This paper studies the parameter estimation of fractional order equivalent circuit model of lithium-ion batteries. Since intelligent optimization algorithms can achieve parameters with high accuracy by transforming the parameter estimation into optimization problem, coyote optimization algorithm is taken in this paper by modifying two key steps so as to improve the accuracy and convergence speed. Firstly, tent chaotic map is introduced to avoid falling into local optimum and enhance population diversity. Secondly, dual strategy learning is employed to improve the searching ability, accuracy and convergence speed. Non-parametric statistical significance is tested by 6 benchmark functions with the comparison of other 5 optimization algorithms. Furthermore, the proposed algorithm is applied to identify the fractional order model of the Samsung ICR18650 (2600 mAh) and compared with conventional coyote optimization algorithm and particle swarm algorithm, which declared the excellence in identification accuracy.

Industry-scale microfluidizer system (ISMS) is a novel equipment for producing filtered-free whole beverages. This study investigated the color, nutrition and flavor changes of whole mango juice (WMJ) treated by ISMS under various pressure (0-120 MPa). Results showed that ISMS well maintained the total soluble solid content, pH value, and yellow color of WMJ. The carotenoids were not destroyed with treatment pressure below 90 MPa, and the carotenoids bioaccessibility of all the WMJ samples was no change. More importantly, ISMS promoted the release of ascorbic acid and total polyphenols by disrupting cellular integrity, thus improving antioxidant activities of WMJ. Volatile compounds analysis found that WMJ samples had more terpenes and aldehydes when treated at 90 MPa, and possessed more esters at 120 MPa. These results demonstrated that ISMS might offer new opportunities to produce WMJ with excellent qualities at an industrial level.

Non-syndromic sensorineural hearing loss (NSHL) is a group of genetically heterogeneous conditions with broad phenotypic heterogeneity. There is, at present, no curative treatment for genetic hearing loss (HL). Early molecular diagnosis of progressive disorders and elucidation of the causes and pathomechanisms are essential for developing therapeutic strategies. Here, we identified a novel rare frameshift variant of LMX1A (c.915dup), which resulted in the C-terminal-altered and -truncated LMX1A (p.Val306Cysfs*32). This C-terminal frameshift mutation co-segregated with autosomal dominant (AD) NSHL in a four-generation Chinese family, suggesting that the LMX1A non-missense mutation is also contributed to ADNSHL. In this family, the affected individuals exhibited the variable auditory phenotypes ranging from profound congenital deafness at birth or to mild/moderate HL in adulthood. We also found that the embryonic cells carrying with the heterozygous variant significantly expressed several upregulated HL-associated genes at transcriptional level. In vitro splicing assay suggested that the LMX1A mRNA with c.915dup did not cause nonsense-mediated decay and was translated into a truncated LMX1A. In addition, electrophoresis mobility shift assay and luciferase assays have shown that the highly conserved C-terminal domain (amino acid 306–382) of the LMX1A was required for regulating the protein–DNA interaction and transactivation in vitro. Furthermore, apoptosis assays suggested that the C-terminal domain of the LMX1A was important for mediating apoptosis in the cochlear hair cells. Our work provided the multiline of the evidence to support that non-missense mutation of LMX1A leads to ADNSHL and the C-terminal domain of LMX1A is important for mediating transcriptional activity and associated with promoting apoptosis in the cells.

Reservoir computing (RC) is a computational architecture capable of efficiently processing temporal information, which allows low-cost hardware implementation. However, the previously reported memristor-based RC mostly utilized binarized data sets to reduce the difficulty of signal processing of the memristor, which inevitably induces data distortion to a certain extent, leading to poor network computing performance. Here, we report on a RC system in a fully memristive architecture based on solution-processed perovskite memristors. The perovskite memristor exhibits 10000 conductance states with a modulation range of more than 4 orders of magnitude. The obtained tens of thousands of finely spaced conductance states with a near-ideal analog property provide a sufficiently large dynamic range and enough intermediate states, which were further applied as a reservoir to map the feature information on different sequential inputs in an analog way. The computing capability of the image classification task of a Fashion-MNIST data set with a high recognition accuracy of up to 90.1% shows that the excellent analog and short-term properties of our perovskite memristor allow the hardware implementation of neuromorphic computing with a reduced training cost.

The activation of the stimulating factor of the interferon gene (STING) pathway can enhance the immune response within the tumor. Cyclic diguanylate monophosphate (c-di-GMP) is a negatively charged, hydrophilic STING agonist, however, its effectiveness is limited due to the poor membrane permeability and low bioavailability. Herein, we introduced KL-7 peptide derived from Aβ amyloid fibrils that can self-assemble to form nanotubes to load and deliver c-di-GMP, which significantly enhanced c-di-GMP's effectiveness and then exhibited a robust "Supplementary material (synthesis and characterization of KL-7 peptide; the encapsulation rate and cumulative release rate of c-di-GMP-PNT; cytotoxicity of PNT, c-di-GMP, and c-di-GMP-PNT; anti-tumor effect of c-di-GMP-PNT (equivalent to 1 and 5 µg c-di-GMP per mouse); representative immunofluorescence images; and biosafety analysis) is available in the online version of this article at 10.1007/s12274-022-5102-z.

RNA labeling in situ has enormous potential to visualize transcripts and quantify their levels in single cells, but it remains challenging to produce high levels of signal while also enabling multiplexed detection of multiple RNA species simultaneously. Here, we describe clampFISH 2.0, a method that uses an inverted padlock design to efficiently detect and exponentially amplify signals from many RNA species at once, while also reducing time and cost compared to the prior clampFISH method. We leverage the increased throughput afforded by multiplexed signal amplification and sequential detection by demonstrating the ability to detect 10 different RNA species in over 1 million cells. We also show that clampFISH 2.0 works in tissue sections. We expect the advantages offered by clampFISH 2.0 will enable many applications in spatial transcriptomics.

Numerous factors contribute to the low adherence to lung cancer screening (LCS) programs. A theory-informed approach to identifying the obstacles and facilitators to LCS uptake is required. This study aimed to identify, assess, and synthesize the available literature at the individual and healthcare provider (HCP) levels based on a social-ecological model and identify gaps to improve practice and policy decision-making. Systematic searches were conducted in nine electronic databases from inception to December 31, 2020. We also searched Google Scholar and manually examined the reference lists of systematic reviews to include relevant articles. Primary studies were scored for quality assessment. Among 3938 potentially relevant articles, 36 studies, including 25 quantitative and 11 qualitative studies, were identified for inclusion in the review. Fifteen common factors were extracted from 34 studies, including nine barriers and six facilitators. The barriers included individual factors (n = 5), health system factors (n = 3), and social/environmental factors (n = 1). The facilitators included only individual factors (n = 6). However, two factors, age and screening harm, remain mixed. This systematic review identified and combined barriers and facilitators to LCS uptake at the individual and HCP levels. The interaction mechanisms among these factors should be further explored, which will allow the construction of tailored LCS recommendations or interventions for the Chinese context.

Diabetic nephropathy (DN) is one of the most important complications of diabetes mellitus (DM) and has become the second cause of end-stage renal disease (ESRD). This study intends to investigate the molecular mechanism of increased mitochondrial fission in podocytes under the effect of high glucose (HG), and to preliminarily study the role of mitochondrial fission factor (MFF)-mediated mitochondrial fission in podocyte injury of DN. In vitro studies, we found that HG induced increased mitochondrial fission and podocyte damage. At the same time MFF mRNA and protein levels was increased, suggesting that MFF was transcriptional upregulated under HG conditions. Consistent with this, in vivo studies found that mitochondrial fission was also significantly increased in podocytes of diabetic nephropathy mice, and MFF expression was up-regulated. Therefore, our study proves that mitochondrial fission increases in podocytes under DM both in vitro and in vivo, and the up-regulation of MFF expression may be one of the reasons for the increase of mitochondrial fission. After inhibiting the expression of MFF, the survival rate of podocytes was significantly decreased under HG conditions, suggesting that MFF may play a protective role in podocyte injury in DN.

A large number of experiments have proved that the ring structure is a common phenomenon in neural networks. Nevertheless, a few works have been devoted to studying the neurodynamics of networks with only one ring. Little is known about the dynamics of neural networks with multiple rings. Consequently, the study of neural networks with multiring structure is of more practical significance. In this article, a class of high-dimensional neural networks with three rings and multiple delays is proposed. Such network has an asymmetric structure, which entails that each ring has a different number of neurons. Simultaneously, three rings share a common node. Selecting the time delay as the bifurcation parameter, the stability switches are ascertained and the sufficient condition of Hopf bifurcation is derived. It is further revealed that both the number of neurons in the ring and the total number of neurons have obvious influences on the stability and bifurcation of the neural network. Ultimately, some numerical simulations are given to illustrate our qualitative results and to underpin the discussion.

In the process of large-scale group decision making (LSGDM), probabilistic linguistic term set (PLTS) is an useful tool to represent the preferences of expert. There is a common case that experts tend to provide incomplete preferences due to various reasons. However, previous methods which cope with the missing values never took experts′ level of cognition over alternatives and attributes into account. In reality, because of limited knowledge reservation and the complexity of decision problem, experts have diverse familiarity with each scheme and attribute. For handling the defect, we propose a novel method to fill missing preference values, based on the combination of knowledge-match degree and trust degree of experts providing reference information. We obtain the knowledge-match degree through the accuracy and reliability of preference as well as the trust degree through social network analysis technology (SNA), and use the probabilistic linguistic weighted average operator (PLWA) to integrate the referential values into preferences of the missing expert. Moreover, to solve the consensus problem at minimal cost, a consensus model based minimum adjust is developed in which the consensus degree of identified elements are all lowest at three aspects including decision matrix, internal experts and intra-group. On the basis of the trust relationship, revising the preference with low consensus guarantees regulated experts′ real aspiration. In addition, a new approach to measure the weight of sub-group is proposed in the light of trust in-degree which considers the reliability of experts in the same subgroup.The feasibility and validity of the LSGDM method are tested by using a numerical example and comparing with other methods.

All of the patients underwent LLR while in the LLDP or the ROP for the treatment of hepatic lesions located in segment VI and/or VII.Results: The preoperative clinical and laboratory parameters were comparable between the two groups (P > 0.05). All patients completed the surgery successfully. There were two patients required conversion to open resection in the ROP comparing with zero in the LLDP. The mean operative time was 256.9 ± 132.7 minutes in LLDP and 255.7 ± 92.1 minutes in ROP, while the median perioperative blood loss was 100 mL (range: 50– 300 mL) in LLDP and 200 mL (range: 50– 425 mL), respectively. The postoperative pathological examination showed that margin-negative resection was achieved all of the cases. The important postoperative parameters all returned to normal within five days after the LLR. The mean postoperative hospital stay (15.6 vs 19.3 days; p < 0.05) and the extraction of the drainage tube time (7.8 vs 10.4 days; p < 0.05) were shorter for patients in LLDP.Conclusion: The LLDP represents a safe and feasible position for performing LLR in selected patients with lesions in segment VI and/or VII. LLR in the LLDP is helpful in terms of the exposure of the surgical field and the recovery of the patient.Keywords: laparoscopic, liver resection, left lateral decubitus position
