AtNPF4.5/AIT2, which was predicted to be a low-affinity transporter capable for nitrate uptake, was screened by ABA receptor complex in Arabidopsis ten years ago. However, the molecular and biochemical characterizations of AtNPF4.5 in plants remained largely unclear. In this study, the function of a plasma-membrane-localized and root-specifically-expressed gene MeNPF4.5 (Manihot-esculenta NITRATE TRANSPORTER 1 PTR FAMILY4.5), an ortholog of the Arabidopsis thaliana NPF4.5, was investigated in cassava roots as a nitrate efflux transporter on low nitrate medium and an influx transporter following exposure to high concentration of external nitrates. Moreover, RNA interference (RNAi) of MeNPF4.5 reduced the nitrate efflux capacity but the overexpressing cassava seedlings increased the ability of efflux from the elongation to the mature zone of root under low nitrate treatments. Besides, MeNPF4.5-RNAi expression reduced the nitrate influx capacity but enhanced nitrate absorption in parts of overexpressing plants from the meristem, elongation to mature zone of roots under high nitrate conditions. Furthermore, MeNPF4.5-RNAi seedlings survived owing to roots that could grow normally, but the MeNPF4.5-over-expressors showed adverse growth under 7% PEG6000 stress, suggesting that MeNPF4.5 negatively regulated the osmotic stress and was involved in nitrate flux through cassava seedlings.

Objectives Immune checkpoint inhibitors are associated with adverse cardiovascular events. However, there are no data characterizing cardiovascular events among Asians on immune checkpoint inhibitors. We aim to determine the incidence and risk of cardiac events associated with immune checkpoint inhibitors in an Asian population. Methods We performed a retrospective, propensity score-matched cohort study at two tertiary referral centers in Taiwan. Immune checkpoint inhibitor users were matched with non-immune checkpoint inhibitor users based on predetermined clinical variables. The primary outcome was major adverse cardiovascular events, defined as a composite of myocardial infarction, ischemic stroke, acute peripheral occlusive disease, pulmonary embolism, deep venous thrombosis, heart failure, pericardial disease, myocarditis, cardiac arrhythmias and conduction block. Results Between January 2010 and November 2021, 868 immune checkpoint inhibitor users were matched 1:1 with non-immune checkpoint inhibitor users. Among immune checkpoint inhibitor users, 67 (7.7%) patients developed major adverse cardiovascular events. During a median follow-up period of 188 days, the incidence rate of major adverse cardiovascular events for immune checkpoint inhibitor and non-immune checkpoint inhibitor users was 94.8 and 46.2 per 1000 patient-years, respectively, resulting in an incidence rate ratio of 2.1 [95% confidence interval: 1.5–2.9]. In multivariate Cox proportional hazard models, immune checkpoint inhibitor users had a 60% increased risk for major adverse cardiovascular events [hazard ratio, 1.6 (95% confidence interval: 1.1–2.3)]. Immune checkpoint inhibitors use was independently associated with increased risk of ischemic stroke [hazard ratio, 3.0 (95% confidence interval: 1.0–9.0)] and pulmonary embolism [hazard ratio, 5.5 (95% confidence interval: 1.4–21.3)]. In multivariate logistic regression analysis, age > 65, metastatic disease, hypertension and baseline platelet-to-lymphocyte ratio Conclusions Among Asians, immune checkpoint inhibitors were associated with an increased risk of major adverse cardiovascular events, particularly ischemic stroke and pulmonary embolism.

Glutaredoxins (GRXs) are essential for reactive oxygen species (ROS) homeostasis in responses of plants to environment changes. We previously identified several drought-responsive CC-type GRXs in cassava, an important tropical crop. However, how CC-type GRX regulates ROS homeostasis of cassava under drought stress remained largely unknown. Purpose The purpose of this paper is to design a unified operational design domain (ODD) monitoring framework for mitigating Safety of the Intended Functionality (SOTIF) risks triggered by vehicles exceeding ODD boundaries in complex traffic scenarios. Design/methodology/approach A unified model of ODD monitoring is constructed, which consists of three modules: weather condition monitoring for unusual weather conditions, such as rain, snow and fog; vehicle behavior monitoring for abnormal vehicle behavior, such as traffic rule violations; and road condition monitoring for abnormal road conditions, such as road defects, unexpected obstacles and slippery roads. Additionally, the applications of the proposed unified ODD monitoring framework are demonstrated. The practicability and effectiveness of the proposed unified ODD monitoring framework for mitigating SOTIF risk are verified in the applications. Findings First, the application of weather condition monitoring demonstrates that the autonomous vehicle can make a safe decision based on the performance degradation of Lidar on rainy days using the proposed monitoring framework. Second, the application of vehicle behavior monitoring demonstrates that the autonomous vehicle can properly adhere to traffic rules using the proposed monitoring framework. Third, the application of road condition monitoring demonstrates that the proposed unified ODD monitoring framework enables the ego vehicle to successfully monitor and avoid road defects. Originality/value The value of this paper is that the proposed unified ODD monitoring framework establishes a new foundation for monitoring and mitigating SOTIF risks in complex traffic environments.

Particulate matter (PM) pollution has become a major problem worldwide, with significant adverse health impacts, but the climatology of annual variations in and the effect of climate change on PM levels are still not comprehensively evaluated. Here we show that the vertical motions of the East Asian winter and summer monsoons regulate the annual variation in PM over Taiwan. The PM pollution season starts and ends earlier during El Niño episodes than during La Niña episodes. Furthermore, the onset, active, break, revival, and retreat phases of the PM pollution lifecycle are innovatively defined. Our results demonstrate that the annual seasonal cycle dominates wintertime PM pollution climate development during the active phase with a minor modulation by El Niño and La Niña episodes, whereas October and March, the seasonal transition periods, are significantly modulated by these two episodes. The findings suggest a new dimension of PM pollution research—the lifecycle evolution effect.

Background Among six extant tiger subspecies, the South China tiger (Panthera tigris amoyensis) once was widely distributed but is now the rarest one and extinct in the wild. All living South China tigers are descendants of only two male and four female wild-caught tigers and they survive solely in zoos after 60 years of effective conservation efforts. Inbreeding depression and hybridization with other tiger subspecies were believed to have occurred within the small, captive South China tiger population. It is therefore urgently needed to examine the genomic landscape of existing genetic variation among the South China tigers. Results In this study, we assembled a high-quality chromosome-level genome using long-read sequences and re-sequenced 29 high-depth genomes of the South China tigers. By combining and comparing our data with the other 40 genomes of six tiger subspecies, we identified two significantly differentiated genomic lineages among the South China tigers, which harbored some rare genetic variants introgressed from other tiger subspecies and thus maintained a moderate genetic diversity. We noticed that the South China tiger had higher FROH values for longer runs of homozygosity (ROH > 1 Mb), an indication of recent inbreeding/founder events. We also observed that the South China tiger had the least frequent homozygous genotypes of both high- and moderate-impact deleterious mutations, and lower mutation loads than both Amur and Sumatran tigers. Altogether, our analyses indicated an effective genetic purging of deleterious mutations in homozygous states from the South China tiger, following its population contraction with a controlled increase in inbreeding based on its pedigree records. Conclusions The identification of two unique founder/genomic lineages coupled with active genetic purging of deleterious mutations in homozygous states and the genomic resources generated in our study pave the way for a genomics-informed conservation, following the real-time monitoring and rational exchange of reproductive South China tigers among zoos.

Background Knee osteoarthritis (KOA) has become an issue of public health. Several Systematic Reviews and Meta-analyses (SRs/Mas) indicate that Traditional Chinese Exercise (TCE) may be an effective treatment for reducing pain, stiffness, and physical function in people with knee osteoarthritis (KOA). Objectives To re-evaluate the literature quality and the evidence for the systematic evaluation of TCE for osteoarthritis of the knee and to offer evidence to support the clinical practice of TCE for osteoarthritis of the knee. Methods Eight electronic databases will be used to retrieve literature from their inception to January 3, 2023, including China National Knowledge Infrastructure (CNKI), Wanfang database (WF), Chinese Scientific Journals Database (VIP), China Biomedical database (CBM), PubMed, EMBASE, Web of Science and Cochrane Library, without restrictions on the study date or language, a systematic evaluation/Meta-analysis of commonly used TCE for knee osteoarthritis was compiled, the methodological and reporting quality of the included literature was evaluated using the AMSTAR-2 scale and PRISMA 2020, and the quality of the evidence was assessed using the GRADE classification. Results A total of 18 papers were included, and the AMSTAR-2 review revealed that the literature's methodological quality could have been better. The study of RPISMA 2020 indicated that the overall reporting standards were deficient. The quality of Chinese and English literature differed, with English literature being superior in methodological quality and reporting quality. There were a total of 93 pieces of evidence obtained, 46 of which were of very low quality (49.46%, 46/93), 34 of low quality (36.56%, 34/93), 13 of medium level (13.98%, 13/93), and none of the high quality. TCE was supported by 76 pieces of evidence (81.72%, 76/93). Conclusion TCE is, therefore, advantageous and secure for KOA. However, clinicians should proceed from these findings with caution in their practice due to the relatively low methodological and evidentiary quality of the included SRs/MAs.

Herein, we aimed to determine the effect of vitamin D (Vit D) and underlying mechanisms on asthma-induced lung injury via regulation of HIF-1α/Notch1 (hypoxia-inducible factor 1 alpha/neurogenic locus notch homolog protein 1) signaling during autophagy. We established an asthma mouse model using respiratory syncytial virus (RSV) nasal drop combined with ovalbumin (OVA) atomization. Mice were treated with different Vit D concentrations. Pathological changes and cell apoptosis were examined using hematoxylin-eosin (HE) staining and TUNEL (terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate (dUTP) nick end-labeling) assay, respectively. Additionally, periodic acid-Schiff (PAS) and Masson's trichrome staining solutions were used to examine changes in lung tissue. Immunofluorescence determined LC 3B (microtubule-associated protein 1 light chain 3B) expression in lung tissues, whereas western blotting and immunohistochemistry were used to evaluate other proteins, including HIF-1α and Notch1. Compared with the normal group, the asthma model group exhibited pathological lung tissue deterioration, elevated fibrosis, increased apoptosis cell numbers, and upregulated autophagy. Vitamin D supplementation ameliorated pathological changes and fibrosis in the lung tissue. Furthermore, Vit D treatment significantly suppressed apoptotic cell numbers and autophagy while enhancing the HIF-1α/Notch1 pathway. Given the HIF-1α/Notch1 agonistic activity, Vit D treatment inhibited apoptosis cell numbers, which were increased following asthma-induced upregulation of autophagy. Vitamin D improved asthma-induced lung tissue injury by suppressing autophagy via regulation of HIF-1α/Notch1 signaling in vivo.

Peripheral nerve injury (PNI) exists widely and seriously affects patients' daily lives. However, the effect of nerve repair is still limited, and only 50% of patients can recover useful functions. To overcome these obstacles, collagen-coated poly(lactic-co-glycolic acid) (PLGA) conduits loaded with CBD-IGF-1 were designed and tested in vitro and in vivo. The physical characterization of the conduit was tested by scanning electron microscopy, and the static water contact angle, release rate, and nerve regeneration ability of the conduit were verified in a rat sciatic nerve injury model. The results showed that the PLGA/col/CBD-IGF-1 conduit had a rough surface and good hydrophilicity. CBD-IGF-1 could be released slowly from the PLGA/col/CBD-IGF-1 conduit. In the in vivo experiment, gait analysis and electrophysiological evaluation showed that the sciatic functional index and electrophysiological parameters were best in the group treated with the PLGA/col/CBD-IGF-1 conduit. The pathological examination results for the sciatic nerve and gastrocnemius muscle in the group treated with the PLGA/col/CBD-IGF-1 conduit were better than those in the other three groups. In short, this study demonstrated the beneficial effects of CBD-IGF-1 in nerve regeneration. The PLGA/col/CBD-IGF-1 conduit has therapeutic potential for use in the treatment of PNI.

Affected by earthquakes and heavy rainfall, multiple landslide dams often cluster closely together along river reaches or gullies. Compared with a single landslide dam, the burst flood produced by the cascading failure of multiple landslide dams can be enhanced, seriously threatening life and property downstream. Here, we conduct a series of experiments in a 42 m flume to investigate the failure mechanisms of single and paired dams with fine-grained, well graded and coarse-grained debris, analyze the effects of dam geometry and initial water level of a downstream dam on the cascading breach, and quantitatively evaluate the amplification effect of cascading breach discharge. Single dams fail by overtopping along with seepage instability for a fine-grained dam, headcutting for a well graded dam and overtopping for a coarse-grained dam, respectively. The type of failure which occurs for a single dam is influenced by the shear strength of the dam material and seepage. However, the downstream dams in cascading tests fail by overtopping irrespective of dam material due to the large outburst floods from the upstream dams. A general flat slope angle is maintained during breaching for the fine-grained and coarse-grained dams, while a step-pool structure is developed for the well graded dams because the finer grains are easier to wash away than coarse grains. The peak breach discharge for a downstream dam is 1.4–1.9 times the value for an upstream dam in the experimental runs, indicating the amplification effect of breach discharge. The amplification effect has a negative linear correlation with the time interval between the peak breach discharges of the two dams because the overlap of breach processes of upstream and downstream dams is gradually reduced as the time interval increases.

The reactive oxygen species (ROS) signal regulates stress-induced leaf abscission in cassava. The relationship between the function of the cassava transcription factor bHLH gene and low temperature-induced leaf abscission is still unclear. Here, we report that MebHLH18, a transcription factor, involved in regulating low temperature-induced leaf abscission in cassava. The expression of the MebHLH18 gene was significantly related to low temperature-induced leaf abscission and POD level. Under low temperatures, the levels of ROS scavengers in different cassava genotypes were significantly different in the low temperature-induced leaf abscission process. Cassava gene transformation showed that MebHLH18 overexpression significantly decreased the low temperature-induced leaf abscission rate. Simultaneously, interference expression increased the rate of leaf abscission under the same conditions. ROS analysis showed a connection between the decrease in the low temperature-induced leaf abscission rate caused by MebHLH18 expression and the increase in antioxidant activity. A Genome-wide association studies analysis showed a relationship between the natural variation of the promoter region of MebHLH18 and low temperature-induced leaf abscission. Furthermore, studies showed that the change in MebHLH18 expression was caused by a single nucleotide polymorphism variation in the promoter region upstream of the gene. The high expression of MebHLH18 led to a significant increase in POD activity. The increased POD activity decreased the accumulation of ROS at low temperatures and the rate of leaf abscission. It indicates that the natural variation in the promoter region of MebHLH18 increases antioxidant levels under low temperatures and slows down low temperature-induced leaf abscission.

Bazhong City is located on stratum dominated by red-bed conditions. This type of weak geological condition with sand and mudstone interbedding is very easily affected by climatic conditions to produce rainfall-type landslides. On the basis of landslide data statistics collected in Bazhong City from 2011 to 2019, this paper uses ERA5-Land rainfall data to statistically analyze the correlation between rainfall and landslide events in Bazhong City. The landslide events in Bazhong City are greatly affected by rainfall events lasting for 10 days. Considering the influence of rainfall seepage and other processes, an effective cumulative rainfall-duration threshold curve for Bazhong City is obtained via median nonlinear fitting. Then, on the basis of the prediction interval, the rainfall threshold formula under different landslide occurrence probabilities is obtained and the critical threshold curve with a non-exceeding probability of 1% in Bazhong City is calculated and verified. Subsequently, a continuous probability distribution fitting function of landslide occurrence is established and a continuous probability distribution surface with a good fitting effect in Bazhong City is obtained. This allows a definite probability of whether future rainfall events will induce landslides to be obtained, providing an important basis for engineering disaster prevention and mitigation.

This study used the 400 g-ton geotechnical centrifuge model test system at the Nanjing Hydraulic Research Institute (NHRI) to investigate the breach evolution characteristics and hydrograph process of overtopping-induced breaching of landslide dams. It was achieved by taking advantage of the “time-space amplification” effect created by high-speed rotation using a centrifuge overweight force field. The effects of dam height, slope ratio, and soil gradation on the overtopping failure process of landslide dams were investigated by centrifugal model tests for the first time. In addition, a detailed physically-based dam breach model was developed to simulate the overtopping failure of landslide dams. Results show that the breach process of a landslide dam can be divided into four stages based on the measured breach morphology evolution process and the abrupt variations of breach flow discharge: initial scour on the downstream slope, retrogressive erosion to the dammed lake, erosion along the breach channel, and breach stabilization. Moreover, the peak breach flow is most sensitive to the dam height, followed by the average particle size; the time to peak is mainly affected by the slope ratio, and the relative residual dam height is primarily susceptible to the average particle size. In practice, the calculated results are consistent with the measured results. This study provides a scientific reference for the cognition of the overtopping-induced breach mechanism of landslide dams.
