Actin cytoskeleton dynamic rearrangement is required for tumor cell metastasis and is a key characteristic of Helicobacter pylori (H. pylori)-infected host cells. Actin cytoskeleton modulation is coordinated by multiple actin-binding proteins (ABP). Through Kyoto encyclopedia of gene and genomes database, GEPIA website, and real-time PCR data, we found that H. pylori infection significantly induced L-plastin, a key ABP, in gastric cancer cells. We further explored the regulation and function of L-plastin in H. pylori–associated gastric cancer and found that, mechanistically, H. pylori infection induced gastric cancer cells to express L-plastin via cagA-activated ERK signaling pathway to mediate SP1 binding to L-plastin promoter. Moreover, this increased L-plastin promoted gastric cancer cell proliferation and migration in vitro and facilitated the growth and metastasis of gastric cancer in vivo. Finally, we detected the expression pattern of L-plastin in gastric cancer tissues, and found that L-plastin was increased in gastric cancer tissues and that this increase of L-plastin positively correlated with cagA+ H. pylori infection status. Overall, our results elucidate a novel mechanism of L-plastin expression induced by H. pylori, and a new function of L-plastin–facilitated growth and metastasis of gastric cancer, and thereby implicating L-plastin as a potential therapeutic target against gastric cancer.Implications:Our results elucidate a novel mechanism of L-plastin expression induced by H. pylori in gastric cancer, and a new function of L-plastin–facilitated gastric cancer growth and metastasis, implicating L-plastin as a potential therapeutic target against gastric cancer.

A protein of Eimeria tenella (encoded by the locus ETH_00028350) homologous to Toxoplasma gondii dense granule protein 9, designated as EtHGRA9 hereafter, was reported to be expressed in all life cycle stages of E. tenella. However, no data are currently available regarding its functional properties. In the present study, a recombinant vector harboring a 741 bp gene segment encoding the mature form of EtHGRA9 was constructed and transfected into leghorn male hepatoma (LMH) cells. Then, transcriptomic analysis of the transfected LMH cells was carried out by using a high-throughput RNA-seq technology. The LMH cells overexpressing EtHGRA9 was validated by means of Western blotting as well as indirect immunofluorescence staining. The results demonstrated that the expression of 547 genes (275 upregulated genes and 272 downregulated genes) was altered by EtHGRA9. The quantitative real-time polymerase chain reaction (qRT-PCR) validation of the ten genes with differential expression between the two groups was consistent with the transcriptome analysis. According to pathway enrichment analysis for the obtained differentially expressed genes, seven pathways were significantly affected by EtHGRA9, such as cytokine-cytokine receptor interaction, MAPK signaling pathway, and protein processing in endoplasmic reticulum. Our data reveal several possible roles of EtHGRA9 in immune or inflammatory responses, which paves the way for a better understanding of the molecular interplay between E. tenella and its host.

A sensing system based on a Fabry-Perot cavity formed by two fiber Bragg gratings for simultaneous measurements of temperature and liquid concentration is reported. Detailed analysis and discussion of the spectral transmission characteristics of the device are provided based on Coupled-Mode Theory. Through numerical simulation of spectral transmission, reflection spectra are shown to be influenced by the grating and Fabry-Perot cavity lengths, and sensing properties of temperature and liquid concentration are studied. A fast Fourier transform based demodulation method that simultaneously demodulates temperature and liquid concentration signals of the device was studied. The use of the fast Fourier transform provides absolute measurements with high sensitivity. The temperature sensitivity was 0.0104 nm/°C, the resolution was 1°C, the error of repeatability was 4%, the hysteresis was less than 0.010 nm, and the sensitivity of the measurement on liquid reflection was 0.01%.

Ginkgolic acids may induce malignant cell death via the B-cell lymphoma 2 (Bcl-2)-associated X protein (Bax)/Bcl-2 apoptosis pathway. Concurrently, apoptosis, autophagy and mitochondrial dysfunction may also be involved in bringing about this endpoint. The anticancer effect of Ginkgolic acids (GAs) was investigated using the HepG2 cell line. The median lethal dose of the GAs of the HepG2 was measured via an MTT assay, the dose-response curves were evaluated and changes in cell morphology were monitored by microscopy. Autophagy in HepG2 cells was down regulated using 3-methyladenine (3-MA) or Beclin-1-specific small interfering RNA (siRNA) and the expression of apoptosis associated proteins caspase-3, Bax/Bcl-2, and the autophagy-associated protein 5 and microtubule-associated protein 1A/1B-light chain 3 in the GA-treated HepG2 cells were all measured by western blot analysis. The level of apoptosis in the GA-treated cells was also assessed using terminal deoxynucleotidyl-transferase-mediated dUTP nick-end labeling (TUNEL) assay, and the mitochondrial membrane potential (Δψm) was detected by immunofluorescence. The results of the MTT and TUNEL assays indicated that the proliferation of HepG2 cells treated with GAs was significantly reduced compared with the control group, and the rate of the inhibition was dose-dependent. Western blot analysis indicated that treatment with the Gas induced apoptosis and autophagy in the HepG2 cells. The Δψm of the GA-treated HepG2 cells was decreased compared with the control, as monitored by immunofluorescence. However, upon the administration of 3-MA or Beclin-1-specific siRNAs (inhibitors of the autophagy), the expression levels of the apoptosis- and autophagy-associated proteins were decreased. In conclusion, the results of the present study indicated that GAs are potent anticancer agents that function through a combination of the apoptosis, autophagy and mitochondrial pathways.

The experiment introduce the performance of orange oil cleanser containing natural orange oil, sodium dodecyl sulfate, fatty alcohol polyoxyethylene ether and alcohol that selected in the best ratio by uniform experimental design method, using the stiring-emulsification method to make a cleanser solution of compounded system at normal temperature, and measured the surface tension. We found that with the content of sodium dodecyl sulfate and alcohol increasing, the surface tension reduced significantly, the cleaning effect improved too. The orange oil cleanser with best formula of sodium dodecyl sulfate -14.2 (mass ratio), fatty alcohol polyoxyethylene ether -2.7(mass ratio), Ethanol-5.1(mass ratio), 22 in total, orange oil-10 ml, contributing the surface tension of 17.8 can be completely biodegraded.

Four chiral aromatic amino alcohol drugs were separated by TLC on a general-purpose silica gel plate with ammonium-D-10-camphorsulfonate (CSA) as chiral ion-interaction agent. The chiral aromatic amino alcohols are all of pharmacological importance as β-adrenergic blockers. The influence of eluent composition, temperature and concentration of CSA on the chiral separation is discussed. It is found that the temperature is also one of the important parameters to be varied for optimum separation in ion-pair chiral resolution. Among four drugs, three enantiomeric drugs were resolved at lower temperature (5°C). In this study, analytical reagent grade methanol and dichlormethane can be directly used as mobile phase without using molecular sieve before use.

Xe ions at energies from 50 to 500 keV have been implanted into the polymer ladderlike polyphenylsilsesquioxane ([(C6H5)8Si8O12]n). The corresponding depth profiles have been measured by 2.1‐MeV 4He++ Rutherford backscattering. The experimental data were compared with our calculation based on Biersack’s angular diffusion model and Monte Carlo simulation. The results show that a good agreement is found between the experimental data and calculated values for the mean projected range, but significant deviations from theoretical values are observed for range straggling.
