We determine from first principles the quark mass anomalous dimension in Nf=3 QCD between the electroweak and hadronic scales. This allows for a fully non-perturbative connection of the perturbative and non-perturbative regimes of the Standard Model in the hadronic sector. The computation is carried out to high accuracy, employing massless O (a) -improved Wilson quarks and finite-size scaling techniques. We also provide the matching factors required in the renormalisation of light quark masses from lattice computations with O (a) -improved Wilson fermions and a tree-level Symanzik improved gauge action. The total uncertainty due to renormalisation and running in the determination of light quark masses in the SM is thus reduced to about 1% ., P.F., C.P. and D.P. acknowledge support through the Spanish MINECO project FPA2015-68541-P and the Centro de Excelencia Severo Ochoa Programme SEV-2012-0249 and SEV-2016-0597. Introduction: Haemophilia management and patients’ quality of life significantly improved. However, data on current patients’, caregivers’ and clinicians’ satisfaction and limitations of treatments and haemophilia management are limited. Aim: Assessing the management satisfaction and unmet needs from the perspective of Italian patients with haemophilia (PWH) without inhibitors (or caregivers if children) and of specialist physicians. Methods: Surveys (for patients≥18 years, caregivers of children and haemophilia specialists) were developed by a multidisciplinary working group and conducted from November 2019 to June 2020. Results: Among 275 participants, 120 (43.6%) were PWH without inhibitors, 79 (28.7%) caregivers and 37 (13.4%) clinicians. Patients and caregivers perceived a higher control of the disease compared to clinicians. However, more than 40% of patients and caregivers reported to feel significantly conditioned by the risk of bleeding during their daily life. PWH reported a 6-month mean/median (range) of bleeds 2.3/.0 (0-24) and caregivers 1.3/.0 (0-16) in children. The treatment burden (frequency of administration) was not satisfactory for more than half adults and caregivers of children treated with prophylaxis. A good access to treatment, haemophilia centres and medical service was reported, with issues associated to the multidisciplinary approach and treatment at emergency department. Conclusions: This large national study provides an updated overview of haemophilia care in Italy from different points of views, highlighting positive aspects and unmet needs. This information can guide future interventions to improve haemophilia management and the assessment of impact of new treatment options.

Stimulation of A(2A) adenosine receptors (AR) promotes anti-inflammatory responses in animal models of allergic rhinitis, asthma, chronic obstructive pulmonary disease, and rheumatic diseases. Herein we describe the results of a research program aimed at identifying potent and selective agonists of the A(2A)AR as potential anti-inflammatory agents. The recent crystallographic analysis of A(2A)AR agonists and antagonists in complex with the receptor provided key information on the structural determinants leading to receptor activation or blocking. In light of this, we designed a new series of 2-((4-aryl(alkyl)piperazin-1-yl)alkylamino)-5'-N-ethylcarboxamidoadenosines with high A(2A)AR affinity, activation potency and selectivity obtained by merging distinctive structural elements of known agonists and antagonists of the investigated target. Docking-based SAR optimization allowed us to identify compound 42 as one of the most potent and selective A(2A) agonist discovered so far (K-i hA(2A)AR = 4.8 nM, EC50 hA(2A)AR = 4.9 nM, K-i hA(1)AR > 10.000 nM, K-i hA(3)AR = 1487 nM, EC50 hA(2B)AR > 10 000 nM).

A series of 4-allyl/benzyl-7,8-dihydro-8-methyl/ethyl-2-[(substituted) isoxazol/pyrazol-3/5-yl]-1H-imidazo[2,1-i]purin-5(4H)-ones has been synthesized and evaluated in radioligand binding assays to determine their affinities at the human A 1, A 2A, and A 3 adenosine receptors. Efficacy at the hA 2B AR and antagonism of selected ligands at the hA 3 AR were also assessed through cAMP experiments. All of the synthesized molecules exhibited high affinity at the hA 3 AR (K i values ranging from 1.46 to 44.8 nM), as well as remarkable selectivity versus A 1, A 2A, and A 2B AR subtypes. Compound (R)-4-allyl-8-ethyl-7,8-dihydro-2-(3-methoxy-1-methyl-1H- pyrazol-5-yl)-1H-imidazo[2,1-i]purin-5(4H)-one (R-33) was found to be the most potent and selective ligand of the series (K i hA 3 = 1.46 nM, K i hA 2A/K i hA 3 > 3425; IC 50 hA 2B/K i hA 3 > 3425; K i hA 1/K i hA 3 = 1729). Molecular modeling studies were helpful in rationalizing the available structure-activity relationships along with the selectivity profiles of the new series of ligands. © 2011 American Chemical Society.

Two new series of inhibitors of tubulin polymerization based on the 2-amino-3-(3,4,5-trimethoxybenzoyl)benzo[b]thiophene molecular skeleton and its 3-amino positional isomer were synthesized and evaluated for antiproliferative activity, inhibition of tubulin polymerization, and cell cycle effects. Although many more 3-amino derivatives have been synthesized so far, the most promising compound in this series was 2-amino-6-methyl-3-(3,4,5-trimethoxybenzoyl)benzo[b]thiophene, which inhibits cancer cell growth at subnanomolar concentrations and interacts strongly with tubulin by binding to the colchicine site.
