Diabetes is a chronic metabolic disorder that is accompanied by the inability of the body to regulate blood sugar level. The prevalent lifestyle and diet habits have increased the number of diabetic individuals to a large number. The novel treatment options have been explored to manage diabetes and overcome the risks associated with the existing therapy. Some of the recent researchers have prepared advanced microspheres either for the delivery of anti-diabetic drugs and insulin or for diagnostic purposes. These microspheres allow the development of controlled or sustained delivery products with rapid onset of action and improved bioavailability. The fabrication of surface-modified microspheres has shown significant promise in achieving the therapeutic goal of diabetes. Glucose-sensitive microspheres can control the release of insulin for a longer duration and improve the hypoglycemic effect of the product. Apart from these, specially designed microspheres were utilized for early detection of diabetes and study disease progression. This review is aimed at providing a perspective over some of the important recent researches with regards to treatment and diagnosis of diabetes. Microspheres have offered a lot of potential for integrating innovative strategies in the treatment and diagnosis of diabetes. The scalability and real-life interventions of these approaches might still be a challenge that need to be addressed.

This study aims to evaluate the current condition of ecotourism in North India in light of the effects of climate change, balance the benefits and drawbacks of implementing more eco-friendly regulations, and highlight innovative new strategies. To find areas of unresolved study and possible future research topics, three papers were evaluated. In this research, the management of ecotourism was examined from the standpoint of a specific place in relation to the use of information and communication technology (ICT). It underlines the opportunity for destination management to utilise ICT to promote sustainable tourism development and offers a summary of a number of ICT-based tools and applications that might be used by destination managers. Method: The study involves the collection of primary data through a survey of 125 tourists in North India. Conclusion: There is awareness among the majority of respondents that sustainable tourism practices can mitigate the impact of climate change in the region. There is a clear need for further education and awarenessraising initiatives to decrease the effects of climate change via sustainable tourism, although respondents demonstrated knowledge with current trends and best practises in green tourism. The city of Rajsamand has experienced a vast change in land use and land cover in the last few decades. The main objective of this study is to analyze the change in land use/land cover (LULC) of Rajsamand urban area. Urbanization refers to industrialization and rapid growth in urban population. Land management is one of the most important elements in the development of the city. According to the present scenario, valuable agricultural land is being converted into non-agricultural land on the pretext of urban expansion of Rajsamand. The findings of this study provide information on changes in land use patterns that Rajsamand city officials can use to develop sustainable development plans.

Immune dysregulation plays a key role in determining COVID-19 disease severity. We aimed to analyze the T cell activation profile in COVID - 19 cases and its predictive role in disease severity and outcome.This was a prospective observational pilot study from a tertiary care COVID-19 hospital. Peripheral blood samples obtained between the fifth and seventh day of COVID-19 illness, were subjected to lymphocyte subset analysis using multicolor flowcytometry using a single tube, 8 antibodies (CD45, CD19, CD3, CD4, CD8, CD38, HLADR, and CD56) analysis. Correlation between lymphocyte subset analysis and clinical profile was determined.26 patients including 11 with mild disease and 15 with severe disease were enrolled. The median age was 58 years (range: 33-81), with a male: female ratio of 1.36:1. Significant lymphopenia was observed in the severe group compared to the mild group (p 0.02). The absolute numbers of CD3+, CD4+, CD8 + T cells, B cells, and NK cells were significantly reduced in the severe group as compared to the mild group (p 0.05). In patients with severe disease, the proportion of CD8 + and CD4 + T cells were significantly higher than those in patients with mild disease (p = 0.0372). Using ROC analysis, a CD4:8 T cell ratio of ≥ 2.63 and an activated (CD38 + HLA-DR+) CD8 T cell proportion of 15.85% of the total CD8 T cell population, significantly determined the severe disease category.Severe COVID-19 is associated with severe lymphopenia, altered CD4/CD8 ratio and markedly increased CD8 T cell activation profile.The online version contains supplementary material available at 10.1007/s12288-022-01558-6.

Musculoskeletal manifestations as the sole presentation in acute leukemia is rare in adults. Acute promyelocytic leukemia (APML) is a subtype of acute myeloid leukemia (AML) with reported incidence of 10 to 15% of total AML cases. APML presenting as polyarticular arthritis has never been reported in the literature. We present an interesting case of 20-year-old male patient who manifested with polyarticular arthritis mainly of small joints as the initial presentation, followed by pancytopenia and eventually was diagnosed as a case of APML on bone marrow morphology and molecular analysis for PML-RARα transcript. He was successfully treated with all-trans-retinoic acid (ATRA) and arsenic trioxide (ATO). Arthritis also resolved with complete remission of APML. Arthritis in a case with pancytopenia should promptly be evaluated prior to treatment with steroids and anti-metabolites. Arthritis can be a presenting manifestation of APML and responds to prompt management of leukemia as in other cases of leukemic arthritis.

Dry eye syndrome is a disease commonly seen in all age groups with a greater ration in elderly population. Large scale studies have determined that increasing age, female sex, and Asian race are all associated with higher DED prevalence. Their occurs tear deficiency and ocular surface disorders, which produces symptoms like discomfort, visual disturbance and tear film instability. Clinically and etymologically, similar entity to Dry eye syndrome is Shushkakshipaka. In modern ophthalmology, conventional approach for Dry eye syndrome is lubricating eye drops which are not satisfactory. Ayurveda has a holistic approach to deal with this problem. This clinical study was done to evaluate the effect of Jivaniya Ghritapana in Shushkakshipaka (dry eye syndrome). An open clinical trial for 10 patients was planned. They were presenting with clinical features of Shushkakshipaka (Dry Eye Syndrome). Snehapana was done in all patients with Jivaniya Ghrita. Results: On assessing the selected criteria, significant results were found in subjective parameters like Rukshta (feeling of dryness), Gharshana (gritty sensation), Daruna vartmakshi (hardness of eyelids), Karachaunmilana (difficulty in opening and closing of eyes), Daha (burning sensation) & intolerance to light. Significant result was found in all objective parameters. Conclusion: Thus we can conclude that, for successful treatment Shushkakshipaka (Dry Eye Syndrome), Snehapana can be advised in such patients.

Background: Benign prostatic hyperplasia (BPH) is a benign enlargement of prostate gland commonly found in elderly men (50% found above 40 year of age) and responsible for major affecting factor for quality of life. The overall incidence rate of BPH is 15/1000 men per year. The present treatment modalities have their own complication and limitations. Hence, here is a need to approach a treatment aspect which can minimize the complication along with major relief effect. According to Ayurveda, considering Vata dosha (ApanaVata) as a main culprit for BPH/Mootraghata, line of treatment was planned to evaluate and compare the clinical efficacy of Kanchanar Guggulu and Varuna Shigru Guggulu, along with Matra basti of Bala Taila svata-kaphahar, mutrakrichahara, Lekhan, and shothahara along with oral medication. Materials and Methods: The patients having signs and symptoms of Mootraghata/BPH were registered and allocated into two groups by computer generated randomization method. Patients were treated with Kanchanar Guggulu orally (1 g thrice a day) in Group A (n = 40) and (1 g thrice a day) Group B (n = 41) for 21 days. Matra basti with 60 ml Bala Taila once a day was given in both groups for 21 days. Results: The result was drawn using appropriate statistical testes in objective as well subjective parameters and also to compare efficacy of both group treatment modalities. The trial drug depicted highly significant results in international prostate symptoms score (IPSS) symptoms, average urine flow rate and prostate size, and post-void residual urine (PVRU) in Group A, whereas Group B possessed highly significant effect on all the mentioned IPSS symptoms with improvement in the QOL, but significant changes were observed in PVRU. Conclusion: Final study concluded that Kanchanar Guggulu and Varuna Shigru Guggulu orally along with Bala Taila Matra bastiis safe and effective in the management of Mootraghata (BPH).

An experiment was conducted during 2017–18 to study the effect of different N, P and K doses on vegetative growth and leaf nutrient content of apple nursery plants of cultivar Starking Delicious and to find out the optimum dose of nutrients for production of healthy planting material for Merton 793 rootstock. The experiment was laid out in a randomized block design comprising 12 combinations of N, P2O5 and K2O {3 levels of N (12, 15 and 18 g/m2 nursery bed), 2 levels of P (3 and 6 g/m2 nursery bed) and 2 levels of K (4.5 and 9 g/m2 nursery bed)}, Jeevamrut and control. The results revealed that maximum linear growth of plant, number of leaves, leaf area, length of main root, total root length, fresh and dry weight of roots, leaf P, Mg and Ca contents were recorded in the plants fertilized with NPK @18:6:4.5 g/m2. However, radial growth of plant, internodal length, fresh and dry weight of shoots, total biomass, leaf N and K were highest in the plants treated with NPK @18:6:9 g/m2 nursery bed. NPK @18:6:4.5 g/m2 nursery bed and NPK @18:6:9 g/m2 nursery bed were found statistically at par with each other with respect to plant growth and leaf nutrients. Thus, nursery bed with 18.0 g N, 6.0 g P and 4.5 g K per m2 was found most suitable for production of quality nursery plants of apple plantlets on Merton 793 clonal rootstock.

The outcome of acute myeloid leukemia (AML) in low-middle-income countries (LMIC) is dismal due to delayed clinical presentation and infection-related complications. We aimed to analyze the outcome of patients with AML and the factors associated with its prognosis.A retrospective observational study was conducted at a tertiary care university hospital in North India from January 2015 to December 2019.A total of 137 AML patients (median age 32 year (3-66 years) received intensive chemotherapy during study period. The median delay from diagnosis to treatment was 45 days (6-177 days). Among the 352 febrile neutropenia (FN) episodes analyzed, 175 (49.7%) were culture positive; Gram-negative multi-drug resistant organism (MDRO) sepsis during induction being 57.4% with 34.5% infections due to carbapenem-resistant Enterobacteriaceae (CRE) leading to a mortality rate of 14.6%. The median EFS and OS were 12.0 ± 1.57 (95% CI 8.91-15.08) and 15.0 ± 2.44 (95% CI 10.21-19.78) months respectively. Multivariable analysis revealed significant difference in median OS between favorable vs high risk AML groups (20.0 (95% CI: 12.50-27.49) vs 9.0 (95% CI: 2.99-15.01) months; p = 0.002); time from diagnosis to treatment ( 30 days vs ≥ 30 days; not reached vs 9.0 (95% CI: 6.81-11.18) months; p = 0.001), performance status (1 vs 2 vs 3; not reached vs 12.0 (95% CI: 10.32-13.67) vs 4.0 (95% CI:2.77-5.22); p = 0.001), and attainment of complete remission vs induction failure (not reached vs 6.0 (95% CI: 3.78-8.21); p = 0.002).Patient-related factors like delayed treatment initiation and high incidence of MDRO-associated sepsis are critical determinants of AML outcome in LMIC.

Ralstonia mannitolilytica is a Gram-negative, nonfermentative, soil bacterium that is reported to cause opportunistic infections in immunocompromised patients in nosocomial settings. After extensive review of literature, it was found that this is second outbreak reported from India. This study is a retrospective analysis of the clinical features, outcome, and source identification of R. mannitolilytica infection outbreak in a hemato-oncology unit of a tertiary care center of North India between February 2020 and March 2020. We report an outbreak of R. mannitolilytica bacteremia (with or without septic shock) in five patients admitted in hemato-oncology unit at a tertiary care institute in North India for 1 month period. Four patients were cured after administration of appropriate antibiotics as per sensitivity reports, while one patient died of septicemia due to delayed diagnosis. Environmental cultures revealed multidose saline bottles used for administration of drugs as the source of outbreak. Following implementation of use of single dose diluents and flushing solutions in patients with central venous catheter, no new case was reported. Clinicians and microbiologists should keep high index of suspicion to identify these organisms as timely diagnosis is the only key to improve outcomes.

The tumor-suppressive activity of FOXP3 has been observed in tumor initiation, but the underlying mechanism still remains largely unknown. Here, we identified a FOXP3–microRNA-146 (miR-146)–NF-κB axis in vitro and in vivo in prostate cancer cells. We observed that FOXP3 dramatically induced the expression of miR-146a/b, which contributed to transcriptional inhibition of IRAK1 and TRAF6, in prostate cancer cell lines. Tissue-specific deletion of Foxp3 in mouse prostate caused a significant reduction of miR-146a and upregulation of NF-κB activation. In addition, prostatic intraepithelial neoplasia lesions were observed in miR-146a–mutant mice as well as in Foxp3-mutant mice. Notably, the NF-κB inhibitor bortezomib inhibited cell proliferation and induced apoptosis in prostate epithelial cells, attenuating prostatic intraepithelial neoplasia formation in Foxp3-mutant mice. Our data suggest that the FOXP3–miR-146–NF-κB axis has a functional role during tumor initiation in prostate cancer. Background: Mechanical neck pain (MNP) is the most common type of pain confined in the posterior aspect of neck, also known as nonspecific or simple neck pain affecting two thirds of the general population at some time in their lives. Prevalence: Neck pain is most common muscular skeletal condition affecting up to 70% of individual and 5% to 10% of adults having a disabling neck pain problem. Neck pain is more common in women than men. The prevalence of neck pain in the general population has been reported to be 15% for men and 23% for women. Objectives of the Study: To find out the effect of Post Isometric Relaxation (PIR) along with Deep Neck Flexor (DNF) exercises in reducing pain and disability and improving ROM in patient with sub-acute mechanical neck pain. To find out the effect of Deep Neck Flexor exercise alone in reducing pain and disability and improving ROM in patient with sub-acute mechanical neck pain. To compare whether Post Isometric Relaxation (PIR) along with Deep Neck Flexor exercises or Deep Neck Flexor exercises alone are effective in reducing pain and disability and improving ROM in patient with sub-acute mechanical neck pain Methods: Study included 30 patients with lateral epicondylitis between ages 30-50 years. The subjects were randomly divided into 2 groups: Group-A and Group-B. Group A (n = 15) was treated using Cyriax physiotherapy, group B (n = 15) was treated with movement with mobilization. Patients were treated for 3 times a week for 4 weeks. Outcome measures are NPRS (Numeric Pain Rating Scale) and DASH (The Disabilities of the Arm, Shoulder and Hand). Results: The results of the study indicate that there were highly statistically significant improvement in functional disability on NDI, cervical lateral flexion(Right & Left), rotation (Right & Left) ROM and reduction in pain on VAS in Group-A as compared to Group-B. There was no significant difference observed when the groups were compared for cervical flexion and extension ROM. Conclusion: From the result it is concluded that Post isometric relaxation with DNF exercise is effective in mechanical neck pain. There is limited evidence on various clinical aspects of SARS-CoV-2 infection in patients with haematological cancers. The risk factors, prognosis, and outcome of patients with haematological cancers with coexistent COVID-19 need to be explored in different subsets of population. A single-institutional prospective observational study was conducted at a tertiary level medical institute in North India. The clinical details of the recruited patients having haematological malignancies and diagnosed with COVID-19 between 15 March 2020 and 31 May 2021 were prospectively collected through the electronic patient database system. The outcomes with respect to 28-day and 56-day mortality and the associated risk factors for prognostication were analysed. Of the 5750 hospital admissions (inpatient and day-care) during the study period, two hundred and forty-two patients (4.2%) were diagnosed with COVID-19. Acute leukaemia was the most common haematological malignancy, seen in 117 (48.3%) patients. Eighty-nine (36.8%) patients had moderate-to-severe COVID-19 while 153 (63.2%) patients presented with mild infection. The 28-day and 56-day mortality rates in our cohort were 13.3% and 19.8% respectively. Amongst the risk factors associated with poor outcome, the severity of COVID-19 (HR = 1.8, 95% CI 1.16-10.35; p = 0.04), presence of secondary infection (HR = 2.1, 95% CI 2.45-21.3; p = 0.023), and need for invasive mechanical ventilation (HR = 2.3, 95% CI 1.8-18.43; p = 0.01) were prognostically significant on multivariate log rank analysis. The risk of SARS-CoV-2 infection does not increase with haematological malignancies; however, the outcome remains poor in patients with severe COVID-19, requirement of invasive mechanical ventilation, and pre-existing bacterial/fungal infection at presentation.

Herein, Bacillus subtilis PBE-8′s biocontrol efficacy was evaluated through physiological and metabolic approaches against Fusarium oxysporum f.sp. lycopersici (FOL). The study elaborates on PBE-8′s cell-free filtrate (CFF) antifungal activity through mycelial growth inhibition, metabolite profiling, and substrates utilization patterns. Additionally, under different CFF concentrations, reduction in spore count (94%–55%), biomass (50%), and cytoplasmic bulbous protrusions in mycelia were also observed. Furthermore, the effect of bacterial CFF on FOL metabolism was confirmed through GC-MS. CFF suppresses the concentration of aliphatic amino acids like L-valine, L-leucine, L-Isoleucine, glycine, and fatty acids such as linoleic acid and α- linolenic acid during the co-culturing conditions, which are essential for pathogenicity and resistance against host's systemic acquired resistance. The phenotype microarray assay revealed that CFF-treated FOL shows phenotype loss in 507 (56.58%) out of 896 substrates. Among 507, twenty-seven substrates showed significant phenotype loss, among which four substrates such as L-glutamic acid, L-glutamine, ammonia, and L-arginine are common in different crucial metabolic pathways of FOL, like alanine, aspartate, and glutamate metabolism, arginine and proline, carbon metabolism, arginine biosynthesis, nitrogen metabolism, amino-acyl tRNA synthesis, and biosynthesis of amino acids. The results suggest that PBE-8 CFF has certain antifungal metabolites that hinder the fungal metabolic pathways.

Three new zinc phosphites, [HIm]2[Zn3(HPO3)4] (1), [Zn2(HPO3)2Im2] (2), and [Zn(HPO3)Im] (3) (Im = imidazole), have been synthesized from the hydro/solvothermal reaction of zinc acetate, dimethyl phosphite, and imidazole by varying the temperature and solvent of the reaction medium. The structure of 1 is built from vertex-sharing of four HPO3-capped Zn3P3 units and adopts an open framework with 12-ring channels stabilized by HIm cations via N-H···O hydrogen bonds. For 2, the inorganic skeleton is comprised of alternating ZnO4 and HPO3 tetrahedra, while the coordinatively associated ZnN2O2 fragments occupy the 12-ring hexagonal channels. Compound 3 adopts a ladder-type one-dimensional structure and exhibits N-H···O hydrogen-bonding interactions to afford a supramolecular assembly. A plausible rationale on the genesis of 1-3 has been put forth by reacting the preformed inorganic zinc phosphites Zn{OP(O)(OMe)H}2 or [Zn2(HPO3)2(H2O)4]·H2O with imidazole as the structure-directing ligand. Alternating-current impedance measurements reveal that 1 and 3 exhibit proton conductivities on the order of 10-3-10-4 S cm-1 between 25 and 100 °C under 35 and 77% relative humidity in repeated impedance cycles (Ea = 0.22-0.35 eV). On the contrary, the conduction property is completely impaired in 2 under similar conditions.

This study was conducted with the aim to assess the clinico-pathological profile, treatment outcomes and the challenges faced in Low Middle Income Countries (LMIC) during management of pediatric Burkitt lymphoma cases on intensive chemotherapy protocol. This was a single center retrospective analysis of pediatric Burkitt lymphoma cases (age18 years) managed uniformly with Lymphomes Malins B (LMB) 96 chemotherapy protocol between January 2015 and September 2019. 40 cases were analyzed with a median age 11.5 years (range 4-18 years) and male: female ratio =4.7:1. Patients belonging to different LMB risk groups were: A-3 (7.5%), B-31 (77.5%), and C-06 (15%). 25 (62.5%) patients had abdominal disease at presentation. Nanoscience has become a diverse and unique field of scientific and technical activity. Over the last few years, the research interest in nanomaterials/nanoparticles and their various applications in various electronic devices, effective optoelectronic devices, bio-sensors, photodetectors, solar cells, nanodevices and plasmonic structures have been increasing tremendously. The reasons are-the unique properties of nanostructures and the outstanding performance of nanoscale devices. At the nanoscale, a material’s property can change dramatically, with unique design possibilities and properties; they attract the attention of researchers worldwide. Nano-composites are those materials with a Nano-scale structure which improves the microscopic property of the products. Composite materials have combinations of two or several nanocomponents, which acquire new and unique characteristic properties that the individual constituents, by themselves, cannot obtain. There are number of various methods to synthesis various Nano-composites. Based on the literature survey, this review article explains a brief introduction of nanocomposites and their types, preparation methods and different diverse properties. It also describes the benefits and limitations of it and new challenges/future scope of the nanocomposites for the better future applications in different sectors.

Microglial cells are the main reservoir for HIV-1 within the brain and potential exists for negative immune checkpoint blockade therapies to purge this viral reservoir. Here, we investigated cytolytic responses of CD8(+) T lymphocytes against microglia loaded with peptide epitopes. Initially, flow cytometric analysis demonstrated efficient killing of HIV-1 p24 AI9 or YI9 peptide-loaded splenocytes in MHC-matched recipients. Cytolytic killing of microglia was first demonstrated using ovalbumin (OVA) as a model antigen for in vitro cytotoxic T lymphocyte (CTL) assays. Peptide-loaded primary microglia obtained from programmed death ligand (PD-L) 1 knockout (KO) animals showed significantly more killing than cells from wild-type (WT) animals when co-cultured with activated CD8(+) T-cells isolated from rAd5-OVA primed animals. Moreover, when peptide loaded-microglial cells from WT animals were treated with neutralizing α-PD-L1 Ab, significantly more killing was observed compared to either untreated or IgG isotype-treated cells. Most importantly, significantly increased in vivo killing of HIV-1 p24 YI9 peptide-loaded microglia from PD-L1 KO animals, as well as AI9 peptide-loaded BALB/c microglial cells treated with α-PD-L1, was observed within brains of rAd5-p24 primed-CNS boosted C57BL/6 or BALB/c mice, respectively. Finally, ex vivo responses of brain CD8(+) T-cells in response to AI9 stimulation showed significantly increased IFN-γ and IL-2 production when treated with α-PD-1 Abs. Greater proliferation of CD8+ T-cells from the brain was also observed following blockade. Taken together, these studies demonstrate that PD-L1 induction on microglia restrains CTL responses and indicate that immune checkpoint blockade targeting this pathway may be beneficial in clearing viral brain reservoirs.

Darvyadi Raskriya is a combination of herbal drugs mentioned in Ayurvedic classic Sharangdhar Samhita. The formulation is believed to cure ocular complaints like Netra Daha, Ashru, raag and Ruja if prepared following appropriate methods. Material and Methods: For making the application of drug more convenient present study has been undertaken with the aim to modify Darvyadi Raskriya into ointment form and to develop the physicochemical profile of the final product. This form was designed in two steps viz. preparation of Ghana Satva from decoction of herbal drugs and mixing powdered Satva with Goghrit and paraffin wax used as a base for attaining the final product. The prepared drug was evaluated for organoleptic study, physicochemical study, pH value and also the product was subjected for microbial contamination test. It was tested in analytical laboratory and results were documented. Result and Discussion: The result shows the organoleptic character and sterility of the product. Conclusion: Darvyadi Raskriya ointment was prepared by following the method described in Sharangdhar Samhita. This paper presents the analytical study of the formulation.

Endophytes can induce the defence responses and modulates physiological attributes in host plants during pathogen attacks. In the present study, 127 bacterial endophytes (BEs) were isolated from different parts of healthy soybean plant. Among them, two BEs (M-2 and M-4) resulted a significant antagonistic property against Macrophomina phaseolina, causes charcoal rot disease in soybean. The antagonistic potential was evaluated through dual culture plate assay, where M-4 expressed higher antifungal activity than M-2 against M. phaseolina. In the current work, globally based on Koopmans’ approximation, local electron transport characteristics of dihydroxybenzenes have been examined using the density functional theory for understanding their antioxidant activity. Our experimental and theoretical studies show that hydroquinone has better antioxidant activities when compared to resorcinol and catechol. To identify the antioxidant sites for each dihydroxybenzene molecule, an average analytical Fukui analysis was used. The typical Fukui analytical results demonstrate that dihydroxybenzene oxygen atoms serve as antioxidant sites. The experimental and theoretical results are in good agreement with each other; therefore, our results are reliable.

Nutrients are the finite natural resources that are essential for productivity and development of rice and its deficiency causes compromised yield along with reduced immunity against several biotic and abiotic stresses. In this study, the potential of Trichoderma reesei has been investigated as a biofertilizer (BF) to ameliorate nutrient stress in different rice cultivars at physiological, biochemical and molecular levels. The results indicated that cultivar Heena is much more compatible with BF as compared to cultivar Kiran at 50% nutrient limiting condition. Enhancement in physiological attributes and photosynthetic pigments were observed in BF treated Heena seedlings. The localization of biofertilizer in treated roots was further validated by scanning electron micrographs. This result correlated well with the higher levels of Indole acetic acid and Gibberellic acid in biofertilizer treated rice. Similarly, the uptake of micro-nutrients such as Fe, Co, Cu and Mo was found to be 1.4-1.9 fold higher respectively in BF treated Heena seedlings under 50% nutrient deficient condition. Furthermore, different stress ameliorating enzymes Guaiacol peroxidase, Super oxide dismutase, Total Phenolic Content, Phenol Peroxidase, Phenylalanine ammonia lyase and Ascorbate peroxidase in Heena seedlings were also increased by 1.8, 1.4, 1.2, 2.4, 1.2, and 8.3-fold respectively, at 50% nutrient deficient condition. The up-regulation of different micro and macro-nutrients allocation and accumulation; metal tolerance related; auxin synthesis genes in BF treated Heena as compared to 50% nutrient deficient condition was further supported by our findings that the application of biofertilizer efficiently ameliorated the deficiency of nutrients in rice.

Catunaregam spinosa leaves have been ethnopharmacologically accounted for acquiring various pharmacological properties. The present study was undertaken to evaluate anti-inflammatory, analgesic and antipyretic potential of leaves of C. spinosa.The ethanolic extract was selected for this purpose based on phytochemical screening. Inflammation was inhibited at the dose of 200 mg/kg with percent inhibition of inflammation 32.06, 37.28 and 43.16 %, respectively, at 1, 3 and 5 h, while in egg albumin model % inhibition was found to be 47.81%. There was no significant analgesic activity seen in acetic acid induced writhing response method while significant effects were observed in the doses of 25 and 100 mg/kg on hot plate test. No antipyretic activity was shown by ethanolic extracts(25, 100 and 200 mg/kg) against Brewer’s yeast induced pyrexia in rats. Upon reactivation of quiescent neurotropic viruses antigen (Ag)-specific brain resident-memory CD8+ T-cells (bTRM) may respond tode novo-produced viral Ag through the rapid release of IFN-γ, which drives subsequent interferon-stimulated gene expression in surrounding microglia. Through this mechanism, a small number of adaptive bTRMmay amplify responses to viral reactivation leading to an organ-wide innate protective state. Over time, this brain-wide innate immune activation likely has cumulative neurotoxic and neurocognitive consequences. We have previously shown that HIV-1 p24 Ag-specific bTRMpersist within the murine brain using a heterologous prime-CNS boost strategy. In response to Ag restimulation, these bTRMdisplay rapid and robust recall responses, which subsequently activate glial cells. In this study, we hypothesized that repeated challenges to viral antigen (Ag) (modeling repeated episodes of viral reactivation) culminate in prolonged reactive gliosis and exacerbated neurotoxicity. To address this question, mice were first immunized with adenovirus vectors expressing the HIV p24 capsid protein, followed by a CNS-boost using Pr55Gag/Env virus-like particles (HIV-VLPs). Following the establishment of the bTRMpopulation [>30 days (d)], prime-CNS boost animals were then subjected toin vivochallenge, as well as re-challenge (at 14 d post-challenge), using the immunodominant HIV-1 AI9 CD8+ T-cell epitope peptide. In these studies, Ag re-challenge resulted in prolonged expression of microglial activation markers and an increased proliferative response, longer than the challenge group. This continued expression of MHCII and PD-L1 (activation markers), as well as Ki67 (proliferative marker), was observed at 7, 14, and 30 days post-AI9 re-challenge. Additionally,in vivore-challenge resulted in continued production of inducible nitric oxide synthase (iNOS) with elevated levels observed at 7, 14 and 30 days post re-challenge. Interestingly, iNOS expression was significantly lower among challenged animals when compared to re-challenged groups. Furthermore,in vivospecific Ag re-challenge produced lower levels of arginase (Arg)-1 when compared with the challenged group. Taken together, these results indicate that repeated Ag-specific stimulation of adaptive immune responses leads to cumulative dysregulated microglial cell activation.

Plants require macro and micronutrients for their physiological function and development process. Deficiency of nutrients such as iron (Fe), phosphorus (P), nitrogen (N), and potassium (K) is a major constraint in agricultural production. Plants have adapted to nutrient deficiency by using several strategies including morphological and physiological changes in plant root architecture through extensive root branching. In such nutrient deficient situations plants require higher nutrient acquisition. Under such nutrient deficiency condition, the presence of endophytic microbes (Bradyrhizobium sp, Pseudomonas aeruginosa, Bacillus cereus, Klebsiella pneumoniae, Bacillus subtilis, and Penicillium guanacastense) associated with plants that occur asymptomatically inside the plant organs and tissues, provide benefits to plant health by nutrient acquisition. Application of endophytes gives an impactful solution for management of nutrient deficiency and moves toward sustainability by preventing pollution caused due to excessive use of chemical fertilizers. This chapter illustrates the effect of nutrient deficiency and the role of endophytic microbes for its mitigation in legume crops.

Appraisal of changes in plant functional behavior due to altitudinal gradients have proved its utility for munificent congregation of communities and their rejoinder to universal and restricted ecological drivers. The accessibility of an ample range of nanostructures and its ability to strappingly defy microorganisms make ZnO a superlative material for antimicrobial impending and biotechnology. The current research endeavor demonstrates the upshot of altitudinal variations in phytochemicals, & antimicrobial and antioxidant activities of Ag nanoparticles (AgNPs) and Ag doped ZnO (Ag-ZnO) nanostructures synthesized in the presence of the floral extract of C. officinalis. The aforementioned plant was collected from three diverse altitudes in Himachal Pradesh, (India). Silver (Ag), Zinc oxide (ZnO), and Ag-ZnO NPs were synthesized in the presence of the floral extract of C. officinalis collected from the Shimla (2104masl), Mandi (1235masl) and Kangra (733masl) hills in the Himalayas. Structural, morphological, and vital ocular properties of the same were inspected. Among Ag NPs, the ones from the Kangra floral extract revealed the maximum inhibition zone (20.83±1.20) against S. aureus, whereas ZnO demonstrated the maximum activity (23.65±1.59) against K. pneumonia . Results also indicated that lower ranges favor the increased potential of antimicrobial activity in floral extracts and synthesized AgNPs from among all the altitudinal ranges. Synergistic effects of Ag doped ZnO NPs and streptomycin showed amplified efficacies as signified by the enlarged zones of inhibition in comparison to their individual effects. The ensuing exertion provided a plausible scheme to develop nanostructures with enviable properties, possessing promising prospect in antimicrobial activity
