Dentale Follikelzellen (DFCs, engl. Dental follicle cells) sind die endogenen Stamm-/Vorläuferzellen des Zahnhalteapparats und können in alveoläre Osteoblasten, Zementoblasten und Fibroblasten des parodontalen Ligaments differenzieren. DFCs besitzen großes Potential für regenerative Therapien von (dentalem) Knochengewebe, wobei zunächst eine genaue Kenntnis der molekularen Mechanismen während der osteogenen Differenzierung, welche sich in vitro durch BMP2 (engl. Bone morphogenetic protein 2) oder Dexamethason induzieren lässt, erforderlich ist. Nachdem Vorarbeiten zeigten, dass klassische Isoformen der Proteinkinase C (PKC) während der Osteogenese von DFCs herunterreguliert werden und diese hemmen, sollten in der vorliegenden Arbeit die zugrundeliegenden Mechanismen evaluiert werden. Hierfür wurde die Regulierung potentieller Zielproteine und Signalwege nach osteogener Induktion und Hemmung klassischer PKCs untersucht sowie deren Einfluss auf osteogene Marker evaluiert. Dabei wurde festgestellt, dass klassische PKCs die Kinase Akt regulieren und stromabwärts durch Phosphorylierung von GSK3β (Glykogensynthase-Kinase 3β) die Aktivität des Proteins β-Catenin beeinflussen, welches eine wichtige Rolle für die Induktion der Osteogenese spielt. Während Akt zwar die Expression von aktivem β-Catenin unterstützte, führte eine Überaktivierung der Kinase jedoch zur Inhibition der Differenzierung und des Signalwegs stromabwärts von BMP2. Darüber hinaus konnte eine Regulierung des NF-κB (engl. Nuclear factor kappa B)-Signalwegs durch klassische PKCs und Akt nachgewiesen werden, wobei NF-κB die Proliferation und osteogene Differenzierung von DFCs unterdrückte. Dahingegen konnte keine direkte Beteiligung von klassischen PKCs oder Akt an der BMP2-induzierten Aktivierung der Proteinkinase A (PKA), welche die Osteogenese unterstützt, gefunden werden. Stattdessen zeigten die Versuche, dass die PKA-Aktivierung von der intrazellulären Expression des Proteins PTHrP (engl. Parathyroid hormone-related protein) abhängt, und dass PKA den Signalweg stromabwärts von BMP2 durch eine negative Rückkopplung hemmt. Des Weiteren konnte eine Induktion des mitochondrialen Energiemetabolismus und des oxidativen Stresses während der osteogenen Differenzierung und nach Inhibition klassischer PKCs festgestellt werden, während eine Hemmung des mitochondrialen Stoffwechsels die Mineralisierung unterdrückte. Eine Evaluierung von Proteinen, welche oxidativen Stress neutralisieren können, zeigte eine Stimulierung der Catalase-Expression und eine Herunterregulierung der Expression von GPX1 (Glutathionperoxidase 1) während der Differenzierung und nach Inhibition klassischer PKCs. Überdies führte eine Hemmung der Catalase zu einer verringerten Mineralisierung. Stromaufwärts von PKC wurde PTHrP für die Aktivität der Kinase benötigt, während das Protein WNT5A (engl. Wnt family member 5A) diese unterdrückte. Die Ergebnisse legten dar, dass klassische Isoformen von PKC die osteogene Differenzierung von DFCs über verschiedene Mechanismen hemmen, wobei insbesondere biologische Prozesse reguliert werden, die in späteren Differenzierungsphasen wichtig sind., Dental follicle cells (DFCs) are the genuine stem/precursor cells of the periodontium and capable of differentiating into alveolar osteoblasts, cementoblasts and periodontal ligament fibroblasts. DFCs possess great potential for regenerative therapies of (dental) bone tissue, but their usage first requires a profound knowledge of the molecular mechanisms during the osteogenic differentiation, which can be induced in vitro by BMP2 (Bone morphogenetic protein 2) or dexamethasone. After previous experiments showed that classical isoforms of PKC (Protein kinase C) are downregulated during osteogenesis of DFCs and inhibit the differentiation, this thesis aimed to evaluate the underlying mechanisms. For this, regulation of potential target proteins and signaling pathways was analyzed after osteogenic induction and inhibition of classical PKCs, and their impact on osteogenic markers was investigated. The results showed that classical PKCs regulate the kinase Akt, which in turn phosphorylates GSK3β (Glycogen synthase kinase 3β) and consequently affects the stability of β-catenin, which plays an important role for the induction of osteogenesis. While Akt supported the expression of active β-catenin, overactivation of the kinase, however, inhibited the differentiation and the signaling pathway downstream of BMP2. Furthermore, classical PKCs and Akt regulated the NF-κB (Nuclear factor kappa B) signaling pathway, and NF-κB was shown to suppress proliferation and osteogenic differentiation of DFCs. By contrast, classical PKCs and Akt were not directly involved in the BMP2-induced activation of protein kinase A (PKA), which is known to support osteogenesis. Instead, the experiments showed that activation of PKA depends on the intracellular expression of the protein PTHrP (Parathyroid hormone-related protein) and that PKA inhibits the signaling pathway downstream of BMP2 via negative feedback. Moreover, mitochondrial energy metabolism and oxidative stress were induced during the osteogenic differentiation as well as after inhibition of classical PKCs, and inhibition of the mitochondrial metabolism suppressed mineralization. Evaluation of proteins which are capable of neutralizing oxidative stress revealed that expression of catalase was upregulated and expression of GPX1 (Glutathione peroxidase 1) was inhibited during differentiation and after inhibition of classical PKCs. Besides, inhibition of catalase attenuated mineralization. Upstream of PKC, PTHrP was required for kinase activity, while WNT5A (Wnt family member 5A) suppressed it. The results showed that classical isoforms of PKC inhibit the osteogenic differentiation of DFCs via different mechanisms, which especially involve biological processes important for later differentiation periods.

The sodium dependent reduced folate carrier (Rfc1; Slc19a1) provides the major route for cellular uptake of reduced folates and antifolate drugs such as methotrexate (MTX) into various tissues. Despite its essential role in folate homeostasis and cancer treatment, little is known about Rfc1 regulation. A barbiturate recognition box, which as yet has only been found in the promoter region of xenobiotic metabolizing enzymes, particularly those of the CYP450 enzyme family, was predicted in the 5′ untranslated region of rat rfc1 cDNA. We have therefore investigated the regulation of Rfc1 by phenobarbital (PB)-type CYP450 inducers on the functional, transcriptional and translational level in a suitable in vitro model for rat liver. A decrease of >75% in substrate uptake was observed following treatment (48 h) with 1–10 times therapeutic plasma concentrations of PB-type CYP450 inducers like PB, carbamazepine, chlorpromazine, clotrimazole and with 0.1–1 ng/ml of the constitutive androstane receptor agonist TCPOBOP. This was not associated with reduced mRNA and protein levels. Further mechanistic investigations revealed that short-term treatment (2 h) of cells with protein phosphatase 1/2A inhibitor okadaic acid (80.5 ng/ml) and proteinkinase C inducer phorbol 12-myristate 13-acetate (PMA; 0.62 μg/ml) almost abolished Rfc1 mediated MTX uptake. Finally, the reduction in Rfc1 activity caused by PB, TCPOBOP and PMA was reversed by simultaneous incubation with the specific PKC inhibitor bisindolylmaleimide (BIM; 21 ng/ml). These results demonstrate that clinically relevant concentrations of PB-type CYP450 inducers cause a significant PKC-dependent reduction in Rfc1 uptake activity at the posttranscriptional level. © 2007 Elsevier B.V. Cultivation with 4-8 μmol/l hypericin (specific proteinkinase C inhibitor) activated by light induced high inhibition of the rate of HL-60 cell growth. When hypericin treated cells were not exposed to light growth inhibition was insignificant. Cultivation with light activated hypericin in concentration 16 μmol/l slightly inhibited growth rate of AtT20/D16v-F2 cells. AtT20/D16v-F2 cells did not proliferate in presence of light activated 32 μmol/l hypericin. Evidence of apoptosis was found in HL-60 cells treated with 1-8 μmol/l light activated hypericin, with maximum of apoptotic cells detected after 8 μmol/l light activated hypericin. Light activated hypericin induces both apoptosis and necrosis in dose and time dependent manners in HL-60 cells, but causes only necrosis in AtT20/D16v-F2 cells.

HINTERGRUND: Im Zustand der Herzinsuffizienz sind die Plasmaspiegel des atrialen natriuretischen Peptids (ANP) erhöht. ANP wird durch Herzmuskelzellen bei atrialer (und dadurch kardiomyozytärer) Dehnung freigesetzt. Eine Reihe von Plasmafaktoren, welche bei der Herzinsuffizienz aktiviert sind, bewirken ebenfalls eine ANP-Freisetzung durch die Interaktion mit G(alpha-q)-gekoppelten Rezeptoren und der daraus resultierenden Aktivierung der Proteinkinase C (PKC). Ziel der vorliegenden Studie war es, im Vergleich von normalen und insuffizienten Rattenherzen die dehnungsinduzierte ANP-Sekretion sowie die Rolle der PKC bei der basalen ANP-Freisetzung zu analysieren. METHODIK: Durch Anlegen eines infrarenalen aortokavalen Shunts (4 Wochen) wurde eine volumeninduzierte Herzinsuffizienz in Wistar Ratten erzeugt. Die ANP-Freisetzung wurde am isoliert, retrograd perfundierten Herzmodell analysiert. Durch Umstellen auf anterograde Perfusion (Perfusionsdruck: 10mmHg) wurde eine atriale De! hnung induziert. Die Aktivierung der PKC fand durch Zugabe von PMA (Phorbol-12-Myristat-13-Azetat) zum Perfusat im Vergleich zu Vehikel statt. Alle Versuchsreihen wurden mit Herzen von shunt- sowie scheinoperierten Tieren durchgeführt. ERGEBNISSE UND DISKUSSION: Nach Shuntoperation war die ANP-Basalfreisetzung deutlich gesteigert und könnte die erhöhten ANP-Plasmaspiegel bei Patienten mit Herzinsuffizienz erklären. Dagegen wurde durch atriale Dehnung (als adäquaten Freisetzungsreiz am gesunden Herzen) keine weitere Steigerung der ANP-Freisetzung in der Shuntgruppe bewirkt. Die ANP-Freisetzung am insuffizienten Herzen ist im Vergleich zur normalen Herzfunktion somit verändert. Unter PKC-Stimulation sank die Freisetzung von ANP in der Shuntgruppe erheblich ab, wogegen es in der Kontrollgruppe zu keiner signifikanten Änderung der ANP-Sekretion kam. Damit wurde der Verdacht erhärtet, dass die Proteinkinase C in die abweichende Regulation der ANP-Freisetzung bei Herzinsuffizie! nz involviert ist., BJECTIVE: Plasma levels of atrial natriuretic peptide (ANP) are markedly increased in congestive heart failure. ANP has been shown to be released by cardiomyocytes during atrial (and thereby cardiomyocytical) stretch. Several factors that are activated in heart failure as well enhance ANP release through the interaction with G(alpha-q)-coupled receptors and the consequent activation of proteinkinase C (PKC). The goal of this study was to analyse stretch induced ANP secretion and to investigate the involvement of PKC in the regulation of ANP release in heart failure compared to normal hearts. METHODS: Volume overload induced heart failure was produced by an infrarenal aortocaval shunt (4 weeks) in Wistar rats. ANP release was analysed in an isolated retrogradly perfused heart preparation. Atrial stretch was induced by switching to anterograd perfusion (perfusion pressure: 10mmHg). For PKC activation PMA (phorbol 12 myristate 13 acetate) was added to the perfusate and compar! ed to vehicle treatment. All experiments were performed with shunt and sham operated rats. RESULTS AND CONCLUSIONS: After shunt operation ANP baseline release was considerably augmented and could thus explain elevated ANP plasma levels in heart failure patients. In contrast, atrial stretch as an adequate secretion stimulus in control hearts did not further enhance ANP release in hearts of shunt operated animals. The ANP secretion in heart failure therefore seems to differ from the ANP secretion in the healthy state. Stimulation of PKC markedly decreased ANP release in the shunt group, whereas it did not have any significant effect on ANP release in the control group. In conclusion, the role of the proteinkinase C in ANP release differs between normal and failing hearts and seems to be involved in the deviating regulation of ANP release in states of heart failure.
