2. Effects of Ambient Air Quality Improvement on Mortality From Acute Air Pollution Exposure in Beijing

Exposure to air pollutants increase the mortality of population. Developing countries have taken measures to control air pollution. To explore the effects of air quality improvement on mortality, air quality and acute exposure-response coefficients of excess death in Beijing since the 1990’s were analyzed. It was divided into five stages according to the concentration level of pollutants. Coefficients for period 1990 – 2013 were obtained by retrieving literatures published before December 31, 2019. The coefficients for period 2015 – 2017 were obtained by analyzing the daily data of air pollutant concentration, meteorological and human mortality conducting Poisson Generalized Additive Model (GAM). Meta-analysis of random effect model was used to estimate the integrated coefficient of multiple studies at each stages. Comparative analysis was used to analyze the variation of air quality and coefficients in different stages. The results showed that the concentrations of sulfur dioxide (SO2), nitrogen dioxide (NO2), particulate matter with aerodynamic diameter ≤10μm (PM10) and ≤2.5μm (PM2.5) decreased by up to 50%, 21%, 22% and 15% in different stages. The coefficient of SO2 on death from respiratory diseases decreased by up to 63.79%. The coefficients of NO2 on mortality from non-accidental causes, cardiovascular disease, and respiratory disease decreased by up to 0.95%, 1.34% and 0.54%. The coefficients of PM10, PM2.5 on mortality from cardiovascular diseases and respiratory disease were decreased by up to 0.19%, 0.31%, 0.65% and 0.36%. Continued improvements in air quality have reduced the acute impact on the health of the local population.

We report here the clinical, genetic, and molecular characteristics of a large Chinese family exhibiting non-syndromic, late-onset autosomal dominant sensorineural hearing loss. Clinical evaluation revealed variable phenotypes of hearing loss in terms of severity and age-at-onset of disease in these subjects. Genome-wide linkage analysis mapped the disease gene to the DFNA5 locus with a maximum two-point log odds score of 5.39 at [theta] = 0 for marker D7S2457. DNA sequencing of DFNA5 revealed a novel heterozygous IVS8+4 A>G substitution in the splice donor site of intron 8. Reverse transcriptase-polymerase chain reaction (RT-PCR) showed skipping of exon 8 in the mutant transcript. This mutation faithfully cosegregated with hearing loss in the family. In addition, the mutation was absent in 100 unrelated control DNA samples of Chinese origin. The IVS8+4 A>G mutation is predicted to create a shift in the reading frame and introduce a stop codon at position 372, thereby resulting in a prematurely truncated DFNA5 protein. Up to date, a total of four mutations in DFNA5 have been reported to lead to hearing impairment, all of them result in skipping of exon 8 at the mRNA level. Our findings provide further support for the hypothesis that DFNA5-associated hearing loss is caused by a very specific gain-of-function mutation.

One hundred pigs (BW=50.7 ± 1.89 kg) were used to investigate the effects of fermented garlic powder (FGP) in growing-finishing pigs. Pigs were allotted to one of five treatments, included: (i) NC (basal diet); (ii) PC (NC+44 mg/kg of Tylosin); (iii) FGP1 (NC + 1 g/kg FGP); (iv) FGP2 (NC + 2 g/kg FGP); and (v) FGP4 (NC+4 g/kg FGP). Supplemental FGP increased average daily gain (ADG) and gain/feed ratio compared with those fed NC diet throughout the experiment (P0.05, linearly). Dietary PC treatment increased the ADG and average daily feed intake during the first 6 weeks (P0.05). The inclusion of FGP2 and FGP4 treatments had higher apparent total tract digestibility of dry matter and nitrogen (P0.05) than those in the NC treatment. Dietary NC treatment exhibited lower (P0.05) lymphocyte count (6 and 12 weeks) and immunoglobulin G (12 weeks) than those in the other treatments. Marbling scores were higher (P0.05) in FGP treatment groups than the NC treatments. Pigs fed on FGP2 and FG4 diets evidenced lower 2-thiobarbituric acid reactive substances value (P0.05) than those in the NC and PC treatments. Collectively, our findings demonstrated that the administration of FGP at a level of 2 or 4 g/kg improved growth performance, nutrient digestibility and meat quality in growing-finishing pigs.

A search for a new scalar resonance decaying to a pair of Z bosons is performed in the mass range from 130 GeV to 3 TeV, and for various width scenarios. The analysis is based on proton-proton collisions recorded by the CMS experiment at the LHC in 2016, corresponding to an integrated luminosity of 35.9 fb−1 at a center-of-mass energy of 13 TeV. The Z boson pair decays are reconstructed using the 4ℓ, 2ℓ2q, and 2ℓ2ν final states, where ℓ = e or μ. Both gluon fusion and electroweak production of the scalar resonance are considered, with a free parameter describing their relative cross sections. A dedicated categorization of events, based on the kinematic properties of associated jets, and matrix element techniques are employed for an optimal signal and background separation. A description of the interference between signal and background amplitudes for a resonance of an arbitrary width is included. No significant excess of events with respect to the standard model expectation is observed and limits are set on the product of the cross section for a new scalar boson and the branching fraction for its decay to ZZ for a large range of masses and widths., Journal of High Energy Physics, 2018 (6), ISSN:1126-6708, ISSN:1029-8479

Ba(Zr0.2Ti0.8)O3 (BZT) and 2 mol% Mn additional doped BZT (Mn-BZT) thin films were deposited by pulsed laser deposition technique under the same growth conditions on LaAlO3 substrates with the bottom electrodes of LaNiO3. The microstructure of the films was characterized by X-ray diffraction (XRD) in the mode of θ–2θ scan and Φ-scan. The results indicated that BZT film was (001)-oriented_with an in-plane relationship of BZT[100]//LNO[100]//LAO[100]. The Mn-BZT film exhibited higher dielectric constant of 225 at zero electric field, larger dielectric tunability of 59.4%, and lower dielectric loss of 1.8% under an applied electric field of 720 kV/cm. The figure of merit for BZT thin film increased from 19.8 to 33 by Mn doping. The enhanced dielectric behavior by Mn doping could be mainly attributed to the decrease of oxygen vacancies and the reorientation of the dipolar defect complex of ${\rm Mn}^{\prime\prime}_{\rm Ti} - {\rm V}_{\ddot{\rm O}}$.

The circadian system cyclically regulates many physiological and behavioral processes within the day. Desynchronization between physiological and behavioral rhythms increases the risk of developing some, including metabolic, disorders. Here we investigate how the oscillatory nature of metabolic signals, resembling feeding-fasting cycles, sustains the cell-autonomous clock in peripheral tissues. By controlling the timing, period and frequency of glucose and insulin signals via microfluidics, we find a strong effect on Per2::Luc fibroblasts entrainment. We show that the circadian Per2 expression is better sustained via a 24 h period and 12 h:12 h frequency-encoded metabolic stimulation applied for 3 daily cycles, aligned to the cell-autonomous clock, entraining the expression of hundreds of genes mostly belonging to circadian rhythms and cell cycle pathways. On the contrary misaligned feeding-fasting cycles synchronize and amplify the expression of extracellular matrix-associated genes, aligned during the light phase. Human induced pluripotent stem cells (hiPSCs) have a number of potential applications in stem cell biology and regenerative medicine, including precision medicine. However, their potential clinical application is hampered by the low efficiency, high costs, and heavy workload of the reprogramming process. Here we describe a protocol to reprogram human somatic cells to hiPSCs with high efficiency in 15 d using microfluidics. We successfully downscaled an 8-d protocol based on daily transfections of mRNA encoding for reprogramming factors and immune evasion proteins. Using this protocol, we obtain hiPSC colonies (up to 160 ± 20 mean ± s.d (n = 48)) in a single 27-mm2 microfluidic chamber) 15 d after seeding ~1,500 cells per independent chamber and under xeno-free defined conditions. Only ~20 µL of medium is required per day. The hiPSC colonies extracted from the microfluidic chamber do not require further stabilization because of the short lifetime of mRNA. The high success rate of reprogramming in microfluidics, under completely defined conditions, enables hundreds of cells to be simultaneously reprogrammed, with an ~100-fold reduction in costs of raw materials compared to those for standard multiwell culture conditions. This system also enables the generation of hiPSCs suitable for clinical translation or further research into the reprogramming process. This protocol describes how to use a microfluidics system to produce human induced pluripotent stem cells at high efficiency and low cost using a small starting cell number.
