In recent years, breast cancer has become one of the biggest threats to women's health, accounting for the majority of cancer deaths among women. Because early treatment of breast cancer has a great effect on the recovery of breast cancer, the diagnosis of breast cancer is particularly important. Machine learning, as the most popular method, is also used for model construction in this field. This study is based on data from breast tumors, which contain 10 morphological features of breast tumor nucleus. In this study, homogenization, standardization and feature selection were used to process the data and KNN algorithm was used to construct the classification prediction model, with principal component analysis (PCA) used to optimize the model. Finally, the original 30 variables were reduced to 3 variables and the model parameters were adjusted in order to achieve the best model with the accuracy of 98.54%. The final model achieves the highest operating efficiency and accuracy. In this study, through the visualization of PCA and the comparison of different models, the classification effect of the final model can be the best. This model can be applied to the clinical diagnosis of breast cancer patients, which is helpful to the early treatment efficiency and greatly reduce the mortality of breast cancer patients

Oil content (OC) is one of the important evaluation indicators in oilfield wastewater (OW) treatment. The purpose of this study is to realize online real-time detection of OC in OW by combining ultraviolet spectrophotometry with the convolutional neural network (CNN). In this paper, 80 groups of OW transmission data were measured for model establishment. Three CNN models with different structures are established to generalize the super parametric optimization process of the model. Furthermore, as a common method used in spectroscopy, the synergy interval partial least squares (siPLS) model is built in order to compare its accuracy with the CNN model. The results indicated the CNN model has a better performance than siPLS, in which the CNN model numbered Model 3 has the lowest root mean square error (MSE) of prediction (RMSEP) of 1.606 mg/L. As a consequence, the CNN model can be used in the monitoring of OW. This article will guide a rapid analysis of the OC of OW.

Diaphragm compressors have become the primary source of on-site hydrogen compression for hydrogen fuelling stations around the world. The most common hydrogen-compressor-failure mechanism has been identified as diaphragm fracture due to low tensile strength and poor surface properties. In this study, a new type of GH4169 diaphragm with high yield strength and low surface roughness was fabricated. Microstructures, tensile behaviours and surface properties of the specimens were characterized by using scanning electron microscopy, an electronic universal testing machine and atomic force microscopy, respectively. The evolution of the γ′′ and γ′ phases with temperature has been examined. Both γ′′ and γ′ phases were precipitant-strengthened phases for GH4169 and the volume and size of γ′′ and γ′ phases increase with ageing time. In addition, the γ′′ phase is the main strengthening phase and γ′ is the supportive strengthening phase. Tensile strength was increased due to the strengthening phases at the cost of ductility. The roughness of the diaphragm could reach an arithmetic average roughness (Ra) of 0.1 μ m by applying a cooling-assisted polishing process. This work improves the reliability and reduces the auxiliary steps and down-time for diaphragm compressors.

A novel strategy for the synthesis of conductive shear thickening fluids (STFs) which manifest both shear thickening behavior and strain-sensing properties.

With the wide application of intelligent manufacturing and the development of diversified functions of industrial manipulator, the requirements for the control accuracy and stability of the manipulator servo system are also increasing. The control of industrial manipulator is a time-varying system with nonlinear and strong coupling, which is often affected by uncertain factors, including parameter changing, environmental interference, joint friction and so on. Aiming at the problem of the poor control accuracy of the manipulator. Under the complex disturbance environment, control accuracy of the manipulator will be greatly affected, so this paper proposes an adaptive type-2 fuzzy sliding mode control (AT2FSMC) method applied to the servo control of the industrial manipulator, which realizes the adaptive adjustment of the boundary layer thickness to suppress the trajectory error caused by the external disturbance and weakens the chattering problem of the sliding mode control. The simulation results on a two-axis manipulator indicate that, with the presence of external disturbances, the proposed control method can help the manipulator maintain control signal stability and improve tracking accuracy. It also suppressed chattering produced by sliding mode control (SMC) and strengthening the robustness of the system. Compared with other conventional trajectory tracking control methods, the effectiveness of the proposed method can be reflected. Finally, the proposed method is tested in an actual manipulator to complete a practical trajectory to prove its feasibility.

Silicon (Si) is considered a promising anode material for rechargeable lithium-ion batteries (LIBs) due to its high theoretical capacity, low working potential, and safety features. However, the practical use of Si-based anodes is hampered by their huge volume expansion during the process of lithiation/delithiation, and they have relatively low intrinsic electronic conductivity, therefore seriously restricting their application in energy storage. Here, we propose a facile approach to directly transform siliceous biomass (bamboo leaves) into a porous carbon skeleton-wrapped Si nanodot architecture through a partial oxidization strategy and magnesium thermal reaction to obtain a high Si nanodot component composite (denoted as Si/C-O). With the synergistic effect of the porous carbon skeleton structure and uniformly dispersed Si nanodots, the Si/C-O composite anode with a stable structure that can avoid pulverization and accommodate volume expansion during cycling is fabricated. As expected, the biomass-converted Si/C-O anode not only presents a high Si component (59.7 wt %) by TGA but also exhibits an excellent capacity of 1013 mAh g

Bmp5 mutation can lead to microtia in mice models; however, its underlying mechanism is unclear. We analyzed circular RNA (circRNA) expression changes and associated gene regulation during embryonic development of the mouse's external ear after a point mutation occurred naturally in the BMP5 gene. The outer ear tissues of BMP5 short-eared mouse model embryos at embryonic day (E) 15.5 and E17.5 were subjected to RNA sequencing. Changes in the circRNA expression profile were detected using find_circ and the CiRi2 software. Differentially expressed circRNAs were annotated by Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses. The circRNA expression profile differed between wild-type and mutant mouse embryos. At E15.5, differentially expressed RNAs were involved in the Hippo signaling pathway, whereas those at E17.5 were associated with stem cell pluripotency. Therefore, circRNA is involved in regulating embryonic external ear development, thus providing a basis for studying the biological aspect of its regulation.

Disc herniation have shown confirmed biomechanical influence on close neighboring vertebrae but the impact on distant neighboring vertebrae remains unknown. After matching age and gender at a single site, we retrospectively evaluated 516 patients with lumbar disc herniation(LDH group)and an equal number of healthy people༈Control group༉to determine the effect of disc herniation on vertebral axial rotation (VAR) in lumbar spine, and explore potential risk factors for VAR in patients with (LDH). Each lumbar spine vertebra's degree of axial rotation was evaluated using the Nash-Moe index. The prevalence of VAR of lumbar spine is greater in LDH group than in Control group (65.7% vs 46.7%, P༜0.001). L2 vertebra in LDH group had the highest frequency of VAR (49.5%), followed by L1(45.1%), and from L3 to L5༈33.6%,8.9%,3.1%༉. The Control group also displayed a similar pattern (L2,39.8%; L1,34.6%; L3,23.2%; L4,3.1%; L5,0.8%). Disc herniation was associated with a higher incidence of VAR in both close and distant neighboring vertebrae. Multivariate analysis showed that age is an independent risk factor for VAR, (OR = 1.245, 95% CI [1.111,1.394], P༜0.001), while symptoms duration and back pain not.

Ultrafine Si embedded SiO2 nanowires have been prepared by thermal evaporation using Pt catalyst. The ultrafine Si embedded SiO2 nanowires with a diameter of about 10 nm were grown in-situ on the Si substrate, conforming to a vapor-liquid-solid growth mechanism. On account of the existence of the quantum confinement effect, the absorption edge of the ultrafine Si embedded SiO2 nanowires is slightly larger. The photoluminescence result reveals a blue shift in the ultrafine nanowires, which may be due to the macroscopic behavior of the sample becomes more relevant and the typical surface plasmon absorption band appears. The Pt-related light emission characteristics will enable the development of nanowires in the field of optoelectronics.

The binding between ADA-07 and TOPK is illustrated by Maestro in the Schrödinger Suite. The binding model shows that the hydrogen atom (white color) of glycine binds with oxygen (red color) and nitrogen (blue color) of ADA-07 to form hydrogen bonds. The oxygen atom of glycine binds with hydrogen of ADA-07 to form a hydrogen bond.

Lung cancer is the leading cause of cancer-related death worldwide. However, promising agents for lung cancer prevention are still very limited. Identification of preventive targets and novel effective preventive agents is urgently needed for clinical applications. In this study, we found that fluvastatin targeted 3-Hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase (HMGCR), which a rate-limiting enzyme in the mevalonate pathway, and inhibited non–small cell lung cancer (NSCLC) tumorigenesis. Initially, we demonstrated that HMGCR is overexpressed in human lung adenocarcinoma tissues compared with normal tissues. Knockdown of HMGCR in NSCLC cells attenuated growth and induced apoptosis in vitro and in vivo. Furthermore, we found that fluvastatin, an inhibitor of HMGCR, suppressed NSCLC cell growth and induced apoptosis. Intriguingly, fluvastastin functions by inhibiting the HMGCR-driven Braf/MEK/ERK1/2 and Akt signaling pathways. Notably, fluvastatin attenuated tumor growth in 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK)-induced lung tumorigenesis and in a patient-derived xenograft lung tumor model. Overall, our findings suggest that fluvastatin might be promising chemopreventive or potential therapeutic drug against NSCLC tumorigenesis, providing hope for rapid clinical translation.

ADA-07 synthesis, MS analysis and cytotoxicity assays.

Cumulative exposure to solar ultraviolet (SUV) irradiation is regarded as the major etiologic factor in the development of skin cancer. The activation of the MAPK cascades occurs rapidly and is vital in the regulation of SUV-induced cellular responses. The T-LAK cell–originated protein kinase (TOPK), an upstream activator of MAPKs, is heavily involved in inflammation, DNA damage, and tumor development. However, the chemopreventive and therapeutic effects of specific TOPK inhibitors in SUV-induced skin cancer have not yet been elucidated. In the current study, ADA-07, a novel TOPK inhibitor, was synthesized and characterized. Pull-down assay results, ATP competition, and in vitro kinase assay data revealed that ADA-07 interacted with TOPK at the ATP-binding pocket and inhibited its kinase activity. Western blot analysis showed that ADA-07 suppressed SUV-induced phosphorylation of ERK1/2, p38, and JNKs and subsequently inhibited AP-1 activity. Importantly, topical treatment with ADA-07 dramatically attenuated tumor incidence, multiplicity, and volume in SKH-1 hairless mice exposed to chronic SUV. Our findings suggest that ADA-07 is a promising chemopreventive or potential therapeutic agent against SUV-induced skin carcinogenesis that acts by specifically targeting TOPK. Malignant melanoma is an aggressive tumor of the skin and still lacks effective preventive and therapeutic treatments. In melanoma, both the BRAF/MEK/ERK and PI3-K/AKT signaling pathways are constitutively activated through multiple mechanisms, which result in cell-cycle progression and prevention of apoptosis. Therefore, the development of novel strategies for targeting BRAF and PI3K are of utmost importance. In this study, we found that Ashitaba (Angelica keiskei) chalcones, 4-hydroxyderricin (4HD) and xanthoangelol (XAG), suppressed melanoma development by directly targeting both BRAFV600E and PI3K, which blocked the activation of downstream signaling. This led to the induction of G1 phase cell-cycle arrest and apoptosis in melanoma cells. Importantly, 4HD or XAG dramatically attenuated tumor incidence and volume in the BRAF-activated Pten-deficient melanoma mouse model. Our findings suggest that 4HD and XAG are promising chemopreventive or potential therapeutic agents against melanomagenesis that act by targeting both BRAF and PI3K, providing hope for rapid clinical translation.
