The activation of melanocortin 1 receptor (MC1R) on melanocytes stimulates the production of eumelanin. A tridecapeptide α melanocyte-stimulating hormone (αMSH) is known to induce skin pigmentation.We characterised the properties of a novel oral MC1R agonist dersimelagon (MT-7117) with respect to its specific binding to MC1R, downstream signalling and eumelanin production in experimental models.The competitive binding and production of intracellular cyclic adenosine 3', 5'-monophosphate in cells expressing recombinant melanocortin receptors were examined. A mouse melanoma cell line B16F1 was used for the evaluation of in vitro melanin production. The in vitro activity of MT-7117 was determined with αMSH and [NleMT-7117 showed the highest affinity for human MC1R compared to the other melanocortin receptors evaluated and agonistic activity for human, cynomolgus monkey and mouse MC1R, with ECMT-7117 is a novel oral MC1R agonist that induces melanogenesis in vitro and in vivo, suggesting its potential application for the prevention of phototoxic reactions in patients with photodermatoses, such as erythropoietic protoporphyria and X-linked protoporphyria.

Background: Excessive alcohol intake has been shown to be associated with cardiovascular disease via metabolic pathways. However, the relationship between alcohol intake and obesity has not been fully elucidated. We aimed to examine the association of alcohol consumption with fat deposition and anthropometric measures. Methods: From 2006–2008, we conducted a cross-sectional study in a population-based sample of Japanese men aged 40 through 79 years. Areas of abdominal visceral adipose tissue (VAT) and subcutaneous adipose tissue (SAT) were calculated using computed tomography imaging. Based on a questionnaire, we classified participants into five groups according to weekly alcohol consumption, excluding former drinkers: non-drinkers (0 g/week), 0.1–160.9, 161–321.9, 322–482.9, and ≥483 g/week. Multivariable linear regression was used to estimate adjusted means of obesity indices for each group. Results: We analyzed 998 men (mean age and body mass index [BMI], 63.8 years and 23.6 kg/m2, respectively). 8. Empathy with Human's and Robot's Embarrassments in Virtual Environments

We feel embarrassed not only when we are embarrassed but also when we are watching others embarrassed. Humans show empathy for pain not only human others but also robots. However, it has not been investigated whether humans show empathy for robot's embarrassment. Thus, we aimed to test whether humans can empathize with robot's embarrassment in virtual environments. Four situations both of non-embarrassing and embarrassing stimuli were presented on an HMD, and participants were asked to rate their own feeling of embarrassment and the actor's feeling of embarrassment. We found that the own feeling of embarrassment was higher in human than robot actors, and higher in embarrassing than non-embarrassing conditions. The actor's feeling of embarrassment was rated higher in embarrassing than non-embarrassing conditions, and the effect was much larger in human than robot actors. Lipomyces starkeyi is an oil-producing yeast that can produce triacylglycerol (TAG) from glycerol as a carbon source. The TAG was mainly produced after nitrogen depletion alongside reduced cell proliferation. To obtain clues for enhancing the TAG production, cell metabolism during the TAG-producing phase was characterized by metabolomics with 13C labeling. The turnover analysis showed that the time constants of intermediates from glycerol to pyruvate (Pyr) were large, whereas those of tricarboxylic acid (TCA) cycle intermediates were much smaller than that of Pyr. Surprisingly, the time constants of intermediates in gluconeogenesis and the pentose phosphate (PP) pathway were large, suggesting that a large amount of the uptaken glycerol was metabolized via the PP pathway. To synthesize fatty acids that make up TAG from acetyl-CoA (AcCoA), 14 molecules of nicotinamide adenine dinucleotide phosphate (NADPH) per C16 fatty acid molecule are required. Because the oxidative PP pathway generates NADPH, this pathway would contribute to supply NADPH for fatty acid synthesis. To confirm that the oxidative PP pathway can supply the NADPH required for TAG production, flux analysis was conducted based on the measured specific rates and mass balances. Flux analysis revealed that the NADPH necessary for TAG production was supplied by metabolizing 48.2% of the uptaken glycerol through gluconeogenesis and the PP pathway. This result was consistent with the result of the 13C-labeling experiment. Furthermore, comparison of the actual flux distribution with the ideal flux distribution for TAG production suggested that it is necessary to flow more dihydroxyacetonephosphate (DHAP) through gluconeogenesis to improve TAG yield.

Lympho-epithelial Kazal-type-related inhibitor (LEKTI) is a large multidomain serine protease inhibitor that is expressed in epidermal keratinocytes. Nonsense mutations of the SPINK5 gene, which codes for LEKTI, cause Netherton syndrome, which is characterized by hair abnormality, ichthyosis, and atopy. A single nucleotide polymorphism (SNP) of SPINK5, p.K420E, is reported to be associated with the pathogenesis of atopic dermatitis (AD). We studied all 34 exons of the SPINK5 gene in Japanese 57 AD patients and 50 normal healthy controls. We detected nine nonsynonymous variants, including p.K420E; these variants had already been registered in the SNP database. Among them, p.R654H (n=1) was found as a heterozygous mutation in the AD patients, but not in the control. No new mutation was detected. We next compared the data of the AD patients with data from the Human Genetic Variation Database provided by Kyoto University; a significant difference was found in the frequency of the p.S368N genotype distribution. PolyPhen-2 and SIFT, two algorithms for predicting the functional effects of amino acid substitutions, showed significant scores for p.R654H. Therefore, R654H might be a risk factor for epidermal barrier dysfunction in some Japanese AD patients.

Item does not contain fulltext BACKGROUND: Many patients with atrial fibrillation have concomitant coronary artery disease with or without acute coronary syndromes and are in need of additional antithrombotic therapy. There are few data on the long-term clinical outcome of atrial fibrillation patients with a history of acute coronary syndrome. This is a 2-year study of atrial fibrillation patients with or without a history of acute coronary syndromes. METHODS: Adults with newly diagnosed atrial fibrillation and >/=1 investigator-defined stroke risk factor were enrolled in GARFIELD-AF between March 2010 and September 2015. The association between prior acute coronary syndromes and long-term outcomes was determined using a Cox proportional hazards model, adjusting for baseline risk factors, oral anticoagulation (OAC) +/- antiplatelet (AP) therapy, and usual care. RESULTS: Of 39,679 patients, 10.5% had a history of acute coronary syndromes. At 2-year follow-up, patients with prior acute coronary syndromes had higher adjusted risks of stroke/systemic embolism (hazard ratio [HR] 1.39; 95% confidence interval [CI], 1.08-1.78), major bleeding (HR 1.30; 95% CI, 0.95 -1.79), all-cause mortality (HR 1.34; 95% CI, 1.21 -1.49), cardiovascular mortality (HR 1.85; 95% CI, 1.51-2.26), and new acute coronary syndromes (HR 3.42; 95% CI, 2.62-4.45). Comparing antithrombotic therapy in the acute coronary syndromes vs no acute coronary syndromes groups, most patients received OAC +/- AP: 60.8% vs 66.1%, but AP therapy was more likely in the acute coronary syndromes group (68.1% vs 32.9%), either alone (34.9% vs 20.8%) or with OAC (33.2% vs 12.1%). Overall, 17.8% in the acute coronary syndromes group received dual AP therapy with (5.3%) or without OAC (12.5%). Among patients with moderate/high risk for stroke/systemic embolism, fewer in the acute coronary syndromes group received OAC with or without AP therapy (Congestive heart failure, Hypertension, Age 75 years, Diabetes mellitus, prior Stroke, TIA, or thromboembolism, Vascular disease, Age 65-74 years, Sex category [CHA2DS2-VASc] 2: 52.1% vs 64.6%; CHA2DS2-VASc >/=3: 62.0% vs 70.7%), and the majority with a Hypertension (uncontrolled systolic blood pressure >160 mm Hg), Abnormal renal or liver function, previous Stroke, Bleeding history or predisposition, Labile international normalized ratios, Elderly, and concomitant Drugs or alcohol excess (HAS-BLED) score >/=3 were on AP therapy (83.8% vs 65.5%). CONCLUSIONS: In GARFIELD-AF, previous acute coronary syndromes are associated with worse 2-year outcomes and a greater likelihood of under-treatment with OAC, while two-thirds of patients receive AP therapy. Major bleeding was more common with previous acute coronary syndromes, even after adjusting for all risk factors.

International audience; Ticagrelor may be a more effective antiplatelet therapy than aspirin for the prevention of recurrent stroke and cardiovascular events in patients with acute cerebral ischemia.; We conducted an international double-blind, controlled trial in 674 centers in 33 countries, in which 13,199 patients with a nonsevere ischemic stroke or high-risk transient ischemic attack who had not received intravenous or intraarterial thrombolysis and were not considered to have had a cardioembolic stroke were randomly assigned within 24 hours after symptom onset, in a 1:1 ratio, to receive either ticagrelor (180 mg loading dose on day 1 followed by 90 mg twice daily for days 2 through 90) or aspirin (300 mg on day 1 followed by 100 mg daily for days 2 through 90). The primary end point was the time to the occurrence of stroke, myocardial infarction, or death within 90 days.; During the 90 days of treatment, a primary end-point event occurred in 442 of the 6589 patients (6.7%) treated with ticagrelor, versus 497 of the 6610 patients (7.5%) treated with aspirin (hazard ratio, 0.89; 95% confidence interval [CI], 0.78 to 1.01; P=0.07). Ischemic stroke occurred in 385 patients (5.8%) treated with ticagrelor and in 441 patients (6.7%) treated with aspirin (hazard ratio, 0.87; 95% CI, 0.76 to 1.00). Major bleeding occurred in 0.5% of patients treated with ticagrelor and in 0.6% of patients treated with aspirin, intracranial hemorrhage in 0.2% and 0.3%, respectively, and fatal bleeding in 0.1% and 0.1%.; In our trial involving patients with acute ischemic stroke or transient ischemic attack, ticagrelor was not found to be superior to aspirin in reducing the rate of stroke, myocardial infarction, or death at 90 days. This study was undertaken to examine whether the production of active oxygen species by bleomycin (BLM) could be enhanced by various cytokines in human neutrophils.Peripheral blood neutrophils were obtained from normal volunteers, and the production of superoxide radical (O-2) and the generation of luminol dependent chemiluminescence (LDCL) were measured by the cytochrome C reduction method and an ATP photometer, respectively. IL-1α, IL-1β, IL-2, IL-6, IL-8, G-CSF, IFN-γ, and TNF-α were used as cytokincs, and neutrophils were treated with these cytokines for 10 minutes at 37°C. Phorbol myristate acetate (PMA) and N-formyl-methionyl-leucyl-phenylalanine (FMLP) were used as stimulants of O-2 and LDCL production, respectively.The results of these experiments revealed slight production of O-2 from neutrophils by BLM, and the production of O-2 by PMA was enhanced by BLM concentrations over 0.3 μg/ml. With a concentration of 2, 500 U/ml of IFN-γ, and at concentrations over l ng/ml of TNF-α, small amounts of O-2 wcre produced without the stimulants or BLM. Thc production of O-2 by BLM was enhanced by IL-1α, IL-1β, IL-8, G-CSF, IFN-γ and TNF-α, that produced by PMA was enhanced by IFN-γ or TNF-α, and O-2 produccd by BLM and PMA in combination was enhanced by IL-8, G-CSF, IFN-γ and TNF-α.These results suggest that the production of active oxygen species by BLM in vivo may be enhanced by these cytokines, except for IL-2 and IL-6. Through this mechanism, cytokines might enhance the antitumor activity of BLM.

A diffusive film characterized with a directional property was developed. This new type of diffusive film is achieved with a new technology derived from holography. Directionality of diffusive film is characterized because it diffuses the light from the determinate angle strongly more than other angles. This discriminative property is consequence of the diffusive film structure that consists of different index transparent materials. Additionally, The film is a volume type diffuser, and the surfaces are almost flat so it is possible to laminated it to various part of other elements without remarkable affecting the diffusing properties of the film. Moreover, durability of this film is actually enough to consumer applications. With these remarkable properties, this diffusive film can widely applied to display systems etc.

Holographic reflectors have proved to be a viable solution to help to increase the luminance of current reflection mode LCD displays at a low cost. Here we present some alternative holographic reflectors being considered at Toppan Printing Co. Inc. for liquid crystal display applications. We describe a new holographic optical element to improve the image's quality of a reflective liquid crystal displays (LCDs). This new holographic reflector consists basically of 2 layers: a volume type transmission hologram layer and a metallic reflection layer. Compared with conventional reflectors for reflective LCDs, a high optical efficiency can be obtained because the hologram is able to concentrate the reflected light to the observer's eyes. Also, it avoids the problems of glare in the LCDs by deviating the reflected incident light (used for display) away from the direction of the direct reflection light. The transmission hologram's low wavelength selectivity permits us to obtain a near white color reflector for reflective LCDs which for multiple applications is the preferable color for the background.

Bleomycin (BLM) has been frequently used for the treatment of oral cancer. BLM is thought to produce active oxygen species such as superoxide anion, which have been shown to bring on tissue damage. It is therefore suggested that active oxygen species may have an antitumor effect. This study was undertaken to clarify the effect of BLM on active oxygen species generation in human peripheral blood neutrophils.For this purpose, peripheral blood neutrophils were obtained from normal volunteers and then luminol dependent chemiluminescence (LDCL) was measured to estimate the generation of active oxygen species. Phorbol myristate acetate (PMA), N-formyl-methionyl-leucyl-phenylalanine (FMLP) or serum opsonized zymosan (OPZ) was used as stimulants of LDCL generation.As a result, at concentrations over 5 μg/ml BLM, LDCL without those stimulants was slightly increased. LDCL stimulated by PMA or FMLP is obviously increased by the addition of O.1 μg/ml BLM or 0.01 μg/ml, respectively. While LDCL induced by OPZ was not increased by BLM. 5-fluorouracil (5-Fu) an antitumor agent, had also no effect on LDCL generation.These results suggest that BLM may promote or enhance the generation of active oxygen species in human neutrophils. Through this function, BLM may exhibit an antitumor activity.
