In order to quantitatively study the influence of the shape of the stern on the resistance of the projectile, the computer-aided fluid dynamics method was used to numerically analyze the shape of the stern model with different mach numbers under the condition of supersonic speed, by analyzing and comparing the pressure coefficient distribution of the stern flow field, the stern surface and the bottom of the projectile ship. The results showed that the tail resistance of the projectile increases linearly with the increase of the tail cone angle; the bottom resistance of the projectile is affected by the area of the bottom of the projectile (shrinkage ratio) and the tail cone angle, and increases linearly with the increase of the shrinkage ratio; The overall drag coefficient of the projectile is related to the bottom resistance and the tail resistance. When the tail length is fixed, there is a tail cone angle that minimizes the drag coefficient, and its size is related to the flight speed of the projectile. When the flight speed of the arrow increases to 3Ma, the optimal solution of the tail cone angle reaches a stable value and its value is about 9.8°, which has a certain reference value for the design of the aerodynamic shape of the high Mach number arrow. &nbsp

Introduction: Kidney cancer is one of the most common and lethal urological malignancies. Discovering a biomarker that can predict prognosis and potential drug treatment sensitivity is necessary for managing patients with kidney cancer. SUMOylation is a type of posttranslational modification that could impact many tumor-related pathways through the mediation of SUMOylation substrates. In addition, enzymes that participate in the process of SUMOylation can also influence tumorigenesis and development.Methods: We analyzed the clinical and molecular data which were obtanied from three databases, The Cancer Genome Atlas (TCGA), the National Cancer Institute’s Clinical Proteomic Tumor Analysis Consortium (CPTAC), and ArrayExpress.Results: Through analysis of differentially expressed RNA based on the total TCGA-KIRC cohort, it was found that 29 SUMOylation genes were abnormally expressed, of which 17 genes were upregulated and 12 genes were downregulated in kidney cancer tissues. A SUMOylation risk model was built based on the discovery TCGA cohort and then validated successfully in the validation TCGA cohort, total TCGA cohort, CPTAC cohort, and E-TMAB-1980 cohort. Furthermore, the SUMOylation risk score was analyzed as an independent risk factor in all five cohorts, and a nomogram was constructed. Tumor tissues in different SUMOylation risk groups showed different immune statuses and varying sensitivity to the targeted drug treatment.Discussion: In conclusion, we examined the RNA expression status of SUMOylation genes in kidney cancer tissues and developed and validated a prognostic model for predicting kidney cancer outcomes using three databases and five cohorts. Furthermore, the SUMOylation model can serve as a biomarker for selecting appropriate therapeutic drugs for kidney cancer patients based on their RNA expression.

ObjectivesThis retrospective analysis aimed to evaluate the early and midterm outcomes of thoracic endovascular aortic repair (TEVAR) with fenestrated surgeon-modified stent-graft (f-SMSG) for type B aortic dissections (TBAD) involving the aortic arch.MethodsFrom March 2016 to April 2021, 47 consecutive patients were treated using TEVAR with f-SMSG. All patients were diagnosed with TBAD involving the aortic arch.ResultsIn total, 47 patients with TBAD involving the aortic arch were treated with f-SMSGs. There were 21 zone 1 and 26 zone 2 TEVAR, and 65 arteries were revascularized successfully with fenestrations. Technical success was achieved in 46 patients (97.88%). The 30-day estimated survival (± SE) and reintervention was 93.6 ± 1.0% (95% Confidence Interval [CI], 92.6–94.6%) and 91.5 ± 1.2% (95% CI, 90.3–92.7%), respectively. During a median follow-up of 51 months (range, 16–71 months), 1 patient died of rupture of aortic dissection (AD) and 3 patients died of non-aortic-related reasons. Reintervention was performed for four patients, including two patients of type IA entry flow and two patients of type IB entry flow. No occlusion of the supra-aortic trunk was observed. The estimated survival and reintervention (± SE) at 4 years was 88.7 ± 1.4% (95% CI, 87.3–90.1%) and 84.8 ± 1.5% (95% CI, 83.3–86.3%), respectively.ConclusionThoracic endovascular aortic repair with f-SMSG is an alternative treatment option for TBAD involving the aortic arch in high-volume centers.

As a potential revolutionary topic in future information processing, mechanical computing has gained tremendous attention for replacing or supplementing conventional electronics vulnerable to power outages, security attacks, and harsh environments. Despite its potential for constructing intelligent matter towards nonclassical computing systems beyond the von Neumann architecture, most works on mechanical computing demonstrated that the ad hoc design of simple logic gates cannot fully realize a universal mechanical processing framework involving interconnected arithmetic logic components and memory. However, such a logic-with-memory computing architecture is critical for complex and persistent state-dependent computations such as sequential logic. Here we propose a mechanical transistor (M-Transistor), abstracting omnipresent temperatures as the input-output mechanical bits, which consists of a metamaterial thermal channel as the gate terminal driving a nonlinear bistable soft actuator to selectively connect the output terminal to two other variable thermal sources. This M-Transistor is an elementary unit to modularly form various combinational and sequential circuits, such as complex logic gates, registers (volatile memory), and long-term memories (non-volatile memory) with much fewer units than the electronic counterparts. Moreover, they can establish a universal processing core comprising an arithmetic circuit and a register in a compact, reprogrammable network involving periodic read, write, memory, and logic operations of the mechanical bits. Text-based delivery addresses, as the data foundation for logistics systems, contain abundant and crucial location information. How to effectively encode the delivery address is a core task to boost the performance of downstream tasks in the logistics system. Pre-trained Models (PTMs) designed for Natural Language Process (NLP) have emerged as the dominant tools for encoding semantic information in text. Though promising, those NLP-based PTMs fall short of encoding geographic knowledge in the delivery address, which considerably trims down the performance of delivery-related tasks in logistic systems such as Cainiao. To tackle the above problem, we propose a domain-specific pre-trained model, named G2PTL, a Geography-Graph Pre-trained model for delivery address in Logistics field. G2PTL combines the semantic learning capabilities of text pre-training with the geographical-relationship encoding abilities of graph modeling. Specifically, we first utilize real-world logistics delivery data to construct a large-scale heterogeneous graph of delivery addresses, which contains abundant geographic knowledge and delivery information. Then, G2PTL is pre-trained with subgraphs sampled from the heterogeneous graph. Comprehensive experiments are conducted to demonstrate the effectiveness of G2PTL through four downstream tasks in logistics systems on real-world datasets. G2PTL has been deployed in production in Cainiao's logistics system, which significantly improves the performance of delivery-related tasks.

Background: Establishing R, S-enantiomer (S-KT and R-KT) chiral resolution and determination methods for KT are of great significance. Objective: This study aimed to establish a high-performance liquid chromatography (HPLC) method for the resolution and determination of ketorolac tromethamine (KT) enantiomers. Methods: A CHIRALPAK AGP column (0.4 × 10 cm, 5 μm) was used with 10 mmol/L ammonium acetate (pH 5.5) and isopropanol (97:3) as the mobile phase. The detection wavelength was 324 nm, the flow rate was 1.0 mL/min, the column temperature was 25°C, and the injection volume was 5 μL. Results: The resolution between S-KT and R-KT was 2.8. S-KT and R-KT demonstrated a good linear relationship in the range of 3-60 μg/mL (r > 0.999). The average recoveries of S-KT and RKT were 99.2% and 99.8%, with relative standard deviations of 2.0% and 2.4%, respectively. Conclusion: The established method can be used for the resolution and determination of S-KT and R-KT.

Kidney cancer is one of the most common urological cancers worldwide, and kidney renal clear cell cancer (KIRC) is the major histologic subtype. Our previous study found that von-Hippel Lindau (VHL) gene mutation, the dominant reason for sporadic KIRC and hereditary kidney cancer-VHL syndrome, could affect VHL disease-related cancers development by inducing telomere shortening. However, the prognosis role of telomere-related genes in kidney cancer has not been well discussed. In this study, we obtained the telomere-related genes (TRGs) from TelNet. We obtained the clinical information and TRGs expression status of kidney cancer patients in The Cancer Genome Atlas (TCGA) database, The International Cancer Genome Consortium (ICGC) database, and the Clinical Proteomic Tumor Analysis Consortium (CPTAC) database. Totally 353 TRGs were differential between tumor and normal tissues in the TCGA-KIRC dataset. The total TCGA cohort was divided into discovery and validation TCGA cohorts and then using univariate cox regression, lasso regression, and multivariate cox regression method to conduct data analysis sequentially, ten TRGs (ISG15, RFC2, TRIM15, NEK6, PRKCQ, ATP1A1, ELOVL3, TUBB2B, PLCL1, NR1H3) risk model had been constructed finally. Adult brainstem gliomas are rare central nervous system tumors that represent a heterogeneous group of tumors. Somatic IDH mutations are uncommon in adult brainstem gliomas and there are few relevant clinical studies. Here, we reported five patients with IDH1 mutations associated with brainstem gliomas, including four cases of IDH1 R132H mutations and one case of R132G mutation. All patients were treated with focal intensity-modulated radiation therapy (IMRT) with concurrent temozolomide (TMZ). One patient died, one relapsed, and three survived to date. All these cases carried a pathogenic variant of TP53, among whom 1 harbored ATRX mutation and 1 had H3K27M mutation. Moreover, we also found some genes related to a worse prognosis, such as CDK4/6 amplification. These findings demonstrate that the specific characteristics of IDH-mutant brainstem gliomas should be considered in diagnostic workflows to make therapeutic regimens and improve the prognosis.

Background: SMILE is one of the most leading-edge corneal refractive surgery.In our study, we aim to investigate the stability of anterior segments in patients with moderate and high myopia one year after SMILE. Methods: 30 eyes of 15 patients who underwent SMILE surgery from July 2020 to January 2021 were collected, including 7 males and 8 females, aged from 20 to 37 years (average 27.20 ± 5.40 years old). Pentacam AXL anterior segment analyzer was performed before operation, 1 month and 1 year after operation to measure the relevant data of anterior segments, including the elevation of posterior corneal surface, Q value of anterior and posterior corneal surface (8mm), curvature of anterior and posterior corneal surface, central anterior chamber depth (ACD), anterior chamber volume and peripheral anterior chamber angle (ACA). Results: all the operations were successful without complications. There were significant differences in preoperative and postoperative equivalent spherical lens, average curvature(km) of anterior and posterior corneal surface , posterior elevation at the thinnest corneal point and Q value of anterior and posterior corneal surface (F = 755.055, P < 0.05; F = 700.001, P < 0.05; F = 3.494, P = 0.037, P < 0.05; F = 8.374, P = 0.001; F = 393.012, P < 0.05; F = 28.179, P < 0.05). Conclusions: the corneal surface morphology before and after SMILE will change in the early stage, but it will gradually stabilize in the medium and long term, and the anterior chamber parameters change, which needs more attention.

Staphylococcus haemolyticus (S. haemolyticus) is the second most commonly isolated coagulase-negative staphylococcus (CoNS) in patients with hospital-acquired infections. It can produce phenol-soluble modulin (PSM) toxins and form biofilms. Compared with the wealth of information on Staphylococcus aureus and Staphylococcus epidermidis, very little is known about S. haemolyticus. There is an urgent need to find an effective preparation to combat the harm caused by S. haemolyticus infection. Chinese herbs have been utilized to cure inflammation and infectious diseases and have a long history of anticancer function in China. Here, we modified fusaric acid characterized from the metabolites of Gibberella intermedia, an endophyte previously isolated from Polygonum capitatum. This study shows that fusaric acid analogs (qy17 and qy20) have strong antibacterial activity against S. haemolyticus. In addition, crystal violet analyses and scanning electron microscopy observations demonstrated that qy17 inhibited biofilm formation and disrupted mature biofilms of S. haemolyticus in a dose-dependent manner. Additionally, it reduced the number of live bacteria inside the biofilm. Furthermore, the antibiofilm function of qy17 was achieved by downregulating transcription factors (sigB), transpeptidase genes (srtA), and bacterial surface proteins (ebp, fbp) and upregulating biofilm-related genes and the density-sensing system (agrB). To further elucidate the bacteriostatic mechanism, transcriptomic analysis was carried out. The following antibacterial mechanisms were uncovered: (i) the inhibition of heat shock (clpB, groES, groL, grpE, dnaK, dnaJ)-, oxidative stress (aphC)- and biotin response (bioB)-related gene expression, which resulted in S. haemolyticus being unable to compensate for various stress conditions, thereby affecting bacterial growth; and (ii) a reduction in the expression of PSM-beta (PSMβ1, PSMβ2, PSMβ3) toxin- and Clp protease (clpP, clpX)-related genes. These findings could have major implications for the treatment of diseases caused by S. haemolyticus infections. Our research reveals for the first time that fusaric acid derivatives inhibit the expression of biofilm formation-related effector and virulence genes of S. haemolyticus. These findings provide new potential drug candidates for hospital-acquired infections caused by S. haemolyticus.

Shor's algorithm has seriously challenged information security based on public key cryptosystems. However, to break the widely used RSA-2048 scheme, one needs millions of physical qubits, which is far beyond current technical capabilities. Here, we report a universal quantum algorithm for integer factorization by combining the classical lattice reduction with a quantum approximate optimization algorithm (QAOA). The number of qubits required is O(logN/loglog N), which is sublinear in the bit length of the integer $N$, making it the most qubit-saving factorization algorithm to date. We demonstrate the algorithm experimentally by factoring integers up to 48 bits with 10 superconducting qubits, the largest integer factored on a quantum device. We estimate that a quantum circuit with 372 physical qubits and a depth of thousands is necessary to challenge RSA-2048 using our algorithm. In the management of lung nodules, we are desirable to predict nodule evolution in terms of its diameter variation on Computed Tomography (CT) scans and then provide a follow-up recommendation according to the predicted result of the growing trend of the nodule. In order to improve the performance of growth trend prediction for lung nodules, it is vital to compare the changes of the same nodule in consecutive CT scans. Motivated by this, we screened out 4,666 subjects with more than two consecutive CT scans from the National Lung Screening Trial (NLST) dataset to organize a temporal dataset called NLSTt. In specific, we first detect and pair regions of interest (ROIs) covering the same nodule based on registered CT scans. After that, we predict the texture category and diameter size of the nodules through models. Last, we annotate the evolution class of each nodule according to its changes in diameter. Based on the built NLSTt dataset, we propose a siamese encoder to simultaneously exploit the discriminative features of 3D ROIs detected from consecutive CT scans. Then we novelly design a spatial-temporal mixer (STM) to leverage the interval changes of the same nodule in sequential 3D ROIs and capture spatial dependencies of nodule regions and the current 3D ROI. According to the clinical diagnosis routine, we employ hierarchical loss to pay more attention to growing nodules. The extensive experiments on our organized dataset demonstrate the advantage of our proposed method. Cardiac hypertrophy is a common pathological change accompanied by various cardiovascular diseases; however, its underlying mechanisms remain elusive. Mounting evidence indicates that long non-coding RNAs (lncRNAs) are novel transcripts involved in regulating multiple biological processes. However, little is known about their role in regulating cardiac hypertrophy. This study revealed a novel lncRNA4930473A02Rik (abbreviated as lncRNAA02Rik), which showed considerably increased expression in hypertrophic mouse hearts in vivo and angiotensin-II (Ang-II)-induced hypertrophic cardiomyocytes in vitro. Notably, lncRNAA02Rik knockdown partly ameliorated Ang-II induced hypertrophic cardiomyocytes in vitro and hypertrophic mouse heart function in vivo, whereas lncRNAA02Rik overexpression promoted cardiac hypertrophy in vitro. Furthermore, lncRNAA02Rik acted as a competing endogenous RNA by sponging miR-135a, while forced expression of lncRNAA02Rik could repress its activity and expression. Furthermore, forcing miR-135a overexpression exerted a significant protective effect against cardiac hypertrophy by inhibiting the activity of its downstream target TCF7, a critical member of Wnt signaling, and the protective effect could be reversed by AMO-135a. Luciferase assay showed direct interactions among lncRNAA02Rik, miR-135a, and TCF7. Altogether, our study demonstrated that lncRNAA02Rik upregulation could promote cardiac hypertrophy development via modulating miR-135a expression levels and TCF7 activity. Therefore, lncRNAA02Rik inhibition might be considered as a novel potential therapeutic strategy for cardiac hypertrophy.

Transcription factors (TFs) play key roles in biological processes and are usually used as cell markers. The emerging importance of TFs and related markers in identifying specific cell types in human diseases increases the need for a comprehensive collection of human TFs and related markers sets. Here, we developed the TF-Marker database (TF-Marker, http://bio.liclab.net/TF-Marker/), aiming to provide cell/tissue-specific TFs and related markers for human. By manually curating thousands of published literature, 5905 entries including information about TFs and related markers were classified into five types according to their functions: (i) TF: TFs which regulate expression of the markers; (ii) T Marker: markers which are regulated by the TF; (iii) I Marker: markers which influence the activity of TFs; (iv) TFMarker: TFs which play roles as markers and (v) TF Pmarker: TFs which play roles as potential markers. The 5905 entries of TF-Marker include 1316 TFs, 1092 T Markers, 473 I Markers, 1600 TFMarkers and 1424 TF Pmarkers, involving 383 cell types and 95 tissue types in human. TF-Marker further provides a user-friendly interface to browse, query and visualize the detailed information about TFs and related markers. We believe TF-Marker will become a valuable resource to understand the regulation patterns of different tissues and cells.

Pathway analysis is considered as an important strategy to reveal the underlying mechanisms of diseases. Pathways that are involved in crosstalk can regulate each other and co-regulate downstream biological processes. Furthermore, some genes in the pathways can function with other genes via the relationship of the competing endogenous RNA (ceRNA) mechanism, which has also been demonstrated to play key roles in cellular biology. However, the comprehensive analysis of ceRNA-mediated pathway crosstalk is lacking. Here, we constructed the landscape of the ceRNA-mediated pathway–pathway crosstalk of eight major cardiovascular diseases (CVDs) based on sequencing data from ∼2,800 samples. Some common features shared by numerous CVDs were uncovered. A fraction of the pathway–pathway crosstalk was conserved in multiple CVDs and a core pathway–pathway crosstalk network was identified, suggesting the similarity of pathway–pathway crosstalk among CVDs. Experimental evidence also demonstrated that the pathway crosstalk was functioned in CVDs. We split all hub pathways of each pathway–pathway crosstalk network into three categories, namely, common hubs, differential hubs, and specific hubs, which could highlight the common or specific biological mechanisms. Importantly, after a comparison analysis of the hub pathways of networks, ∼480 hub pathway-induced common modules were identified to exert functions in CVDs broadly. Moreover, we performed a random walk algorithm on the hub pathway-induced sub-network and identified 23 potentially novel CVD-related pathways. In summary, our study revealed the potential molecular regulatory mechanisms of ceRNA crosstalk in pathway–pathway crosstalk levels and provided a novel routine to investigate the pathway–pathway crosstalk in cardiology. Structural variations (SVs), such as inversion and duplication, contribute to important agronomic traits in crops1. Pan-genome studies revealed that SVs were a crucial and ubiquitous force driving genetic diversification2–4. Although genome editing can effectively create SVs in plants and animals5–8, the potential of designed SVs in breeding has been overlooked. Here, we show that new genes and traits can be created in rice by designed large-scale genomic inversion or duplication using CRISPR/Cas9. A 911 kb inversion on chromosome 1 resulted in a designed promoter swap between CP12 and PPO1, and a 338 kb duplication between HPPD and Ubiquitin2 on chromosome 2 created a novel gene cassette at the joint, promoterUbiquitin2::HPPD. Since the original CP12 and Ubiquitin2 genes were highly expressed in leaves, the expression of PPO1 and HPPD in edited plants with homozygous SV alleles was increased by tens of folds and conferred sufficient herbicide resistance in field trials without adverse effects on other important agronomic traits. CRISPR/Cas-based genome editing for gene knock-ups has been generally considered very difficult without inserting donor DNA as regulatory elements. Our study challenges this notion by providing a donor-DNA-free strategy, thus greatly expanding the utility of CRISPR/Cas in plant and animal improvements. This study reports a donor DNA-free approach for gene knock-up based on CRISPR. It induced designed promoter swap or replacement for target genes by triggering genomic inversion or duplication so as to elevate gene expression and generate new traits.

The present study proposes a framework for the continuous Bayesian calibration of whole building energy simulation (BES) models utilizing data from building information models (BIM) and building energy management systems (BEMS). The ability to import data from BIM and BEMS provides the potential to significantly reduce the time and effort needed for the continuous calibration of BES models. First, five gbXML geometric test cases were used to check the BIM to BES model translation. Translation of the test cases indicates good geometric agreement between the native BIM and the gbXML-based BES model. An actual building calibration case study (with BIM and three years of monthly electrical energy consumption data) was then used to evaluate the proposed continuous calibration method. The results suggest that compared to a non-continuous approach, the continuous Bayesian calibration method showed reduced prediction uncertainty and improved prediction accuracy on a test dataset. The paper also presents information and comparison of the coefficient of variance of the root mean square error (CVRMSE) and the normalized mean biased error (NMBE), recommending looking at their distributions when working with probabilistic BES predictions.

Many applications of wireless power transmission (WPT) require high misalignment tolerance. However, the transmission power of WPT is highly susceptible to the misalignment. To solve this problem, the magnetic resonant WPT control method with high misalignment tolerance is proposed here. First, based on the general mutual model, the complex ratio transformer model is established. This model gives an intuitive physical picture in power transmission process and frequency splitting phenomenon. Second, approximately constant voltage gain at frequency splitting point in a wide range of misalignment is analysed. Considering this frequency splitting characteristics of Magnetic resonant WPT, the maximum peak point tracking control methods for wide misalignment, maintaining constant output power is proposed. The safety and quality of aquatic foods are a public concern due to their content of pollutants, such as arsenic. A formula is derived for quantifying the benefit-risk ratio (HQ) of the essential polyunsaturated fatty acids vs. arsenic in Chinses mitten crabs. Among these arsenic species, the proportion of inorganic arsenic, which is extremely harmful to the human body, is5%, and its level does not exceed the national standard limit. Meanwhile, comparing with the HQ from the original method, the HQs from groups 0 min, 5 min, 15 min are significantly higher(p 0.05). This suggests the original assessment method could underestimate the risk of eating crabs. Eating steamed crabs is easier to digest essential fatty acids (EFAs) than eating raw crabs, and it also protects consumers against arsenic exposure. To achieve a good balance of dietary benefits and risks, the steaming duration of the crabs should exceed 30 min.

Solving the wave equation is one of the most (if not the most) fundamental problems we face as we try to illuminate the Earth using recorded seismic data. The Helmholtz equation provides wavefield solutions that are dimensionally reduced, per frequency, compared to the time domain, which is useful for many applications, like full waveform inversion (FWI). However, our ability to attain such wavefield solutions depends often on the size of the model and the complexity of the wave equation. Thus, we use here a recently introduced framework based on neural networks to predict functional solutions through setting the underlying physical equation as a loss function to optimize the neural network parameters. For an input given by a location in the model space, the network learns to predict the wavefield value at that location, and its partial derivatives using a concept referred to as automatic differentiation, to fit, in our case, a form of the Helmholtz equation. We specifically seek the solution of the scattered wavefield considering a simple homogeneous background model that allows for analytical solutions of the background wavefield. Providing the neural network (NN) a reasonable number of random points from the model space will ultimately train a fully connected deep NN to predict the scattered wavefield function. The size of the network depends mainly on the complexity of the desired wavefield, with such complexity increasing with increasing frequency and increasing model complexity. However, smaller networks can provide smoother wavefields that might be useful for inversion applications. Preliminary tests on a two-box-shaped scatterer model with a source in the middle, as well as, the Marmousi model with a source on the surface demonstrate the potential of the NN for this application. Additional tests on a 3D model demonstrate the potential versatility of the approach.
