To evaluate the national prevalence depression of Chinese cancer patients and clarify its potential associated factors. Twelve databases were searched from database inception through October 31, 2022 for relevant studies published in English (PubMed MEDLINE, Ovid MEDLINE, Embase, Scopus, PsycINFO, and Cochrane and Web of Science) and Chinese (China National Knowledge Infrastructure, Wanfang, Weipu, Chinese Biomedical Literature Service System, Duxiu) that estimated point or period prevalence of depression, depressive disorder, or suicidal ideation as assessed by self-report scale or structured interview. 201 studies were eligible for inclusion in our meta-analyses. We found the national pooled prevalence rates of depression and suicidal ideation were 44.63 % (95 % CI 42.24 %-47.01 %) and 24.95 % (95 % CI 10.96 %-38.95 %), respectively. The pooled prevalence of depression was associated with study location, cancer type, comorbid chronic disease, and female sex (all P 0.05). The distribution of the depression prevalence among cancer patients in mainland China was characterized by significant geographical variation and clustering. The highest pooled prevalence of depression was among gynecologic oncology patients. We recommend that more attention and resources be given to mental health problems such as depression and suicidal ideation among Chinese cancer patients, and that prevention programs be developed, especially in areas with a high prevalence of depression.

Platelet hyperactivation could lead to various cardiovascular and cerebrovascular diseases, while epidemiological analyses have found that long-term tea drinking could prevent and restrain cardiovascular diseases. Existing studies have shown that catechins, especially epigallocatechin gallate (EGCG), are the main functional factors of tea in alleviating thrombosis, which could inhibit arterial thrombosis and platelet aggregation induced by a variety of agonists. However, their structure-activity relationship and the underlying mechanisms are still unclear. Based on the above background, this study took six typical catechins as research objects, constructed platelet activation models with different inducers, and explored the inhibitory effects and potential mechanisms of catechins with different structures on platelet aggregation through flow cytometry, immunoblotting, cell spreading, and other experiments. It was found that ester catechins could inhibit platelet aggregation induced by adenosine diphosphate (ADP), while epigallocatechin (EGC) with three hydroxyls on the B ring in non-ester catechins was also able to effectively inhibit platelet aggregation. Our data suggested that gallic acyl on the C ring and three hydroxyls on the B ring were the main functional groups affecting the antithrombotic effect of catechins, and the effect of gallic acyl on platelets was significantly stronger than that of the hydroxyl.

STUDY QUESTION Would the construction of a competing endogenous RNA (ceRNA) network help identify new drug targets for the development of potential therapies for polycystic ovary syndrome (PCOS)? SUMMARY ANSWER Both Food and Drug Administartion (FDA)-approved and candidate drugs could be identified by combining bioinformatics approaches with clinical sample analysis based on our established ceRNA network. WHAT IS KNOWN ALREADY Thus far, no effective drugs are available for treating PCOS. ceRNAs play crucial roles in multiple diseases, and some of them are in current use as prognostic biomarkers as well as for chemo-response and drug prediction. STUDY DESIGN, SIZE, DURATION For the bioinformatics part, five microarrays of human granulosa cells were considered eligible after applying strict screening criteria and were used to construct the ceRNA network for target identification. For population-based validation, samples from 24 women with and without PCOS were collected from January 2021 to July 2021. PARTICIPANTS/MATERIALS, SETTING, METHODS The public data included 27 unaffected women and 25 women with PCOS, according to the Rotterdam criteria proposed in 2003. The limma and RobustRankAggreg R packages were used to identify differentially expressed messenger RNAs and noncoding RNAs. Gene Ontology, Reactome and Kyoto Encyclopedia of Genes and Gemomes (KEGG) enrichment analyses were performed. A ceRNA network was constructed by integrating the differentially expressed genes and target genes. The population-based validation included human luteinized granulosa cell samples from 12 unaffected women and 12 women with PCOS. Quantitative real-time polymerase chain reaction was conducted to detect the levels of mRNAs and microRNAs (miRNAs). Connectivity map and computational model algorithms were implemented to predict therapeutic drugs from the ceRNA network. Additionally, we compared the predicted drugs with known clinical medications in DrugBank. MAIN RESULTS AND THE ROLE OF CHANCE A set of 10 mRNAs, 11 miRNAs and 53 long non-coding RNAs (lncRNAs) were differentially expressed. Functional enrichment analysis revealed the highest relevance to immune system-related biological processes and signalling pathways, such as cytokine secretion and leucocyte chemotaxis. A ceRNA consisting of two lncRNAs, two miRNAs and five mRNAs was constructed. Through network construction via bioinformatic analysis, we identified some already approved drugs (such as metformin) that could target some molecules in the network as potential drug candidates for PCOS. LARGE SCALE DATA Public sequencing data were obtained from GSE34526, GSE84376, GSE102293, GSE106724 and GSE114419, which have been deposited in the Gene Expression Omnibus database. LIMITATIONS, REASONS FOR CAUTION Experiments, such as immunoprecipitation, luciferase reporter assays and animal model studies, are needed to validate the potential targets in the ceRNA network before the identified drug candidates can be tested using cellular and animal model systems. WIDER IMPLICATIONS OF THE FINDINGS Our findings provide new bioinformatic insight into the possible pathogenesis of PCOS from ceRNA network analysis, which has not been previously studied in the human reproductive field. Our study also reveals some potential drug candidates for the future development of possible therapies against PCOS. Understanding the occupational characteristics of patients is not only related to patients' life and health, but also conducive to improving their happiness. However, there were no studies that had been conducted on the relationship between occupation characteristic and postoperative recovery in patients with spinal fractures. The purpose of this study was to explore the relationship between the occupation characteristics of patients with thoracolumbar fracture and the characteristics of disease injury, treatment, and recovery so as to reduce the incidence and improve postoperative rehabilitation.Patients (n = 719) with thoracolumbar fractures were recruited. Patients were grouped according to the characteristic of occupations: unemployed group (n = 299), white-collar worker group (n = 20), and blue-collar worker group (n = 400). Data were collected, including the characteristics, injury and treatment information, and the recovery records for 1 year after operation. One-way ANOVA analysis, χMale, high-falling injuries and single segment injury (mainly T 11, T 12 and L2) were common in patients with thoracolumbar fractures, especially in the blue-collar worker group (70.8%, 78.3%, and 85.4%). Compared with the unemployed group, the patients in the white-collar worker group and blue-collar worker group had a higher proportion of young patients, a higher height and weight, a lesser rate of hypertension or diabetes. One week after injury, 73.4% of patients underwent surgery, with the blue-collar worker group accounted for the largest proportion. One month after surgery, 77.1% of patients were able to get out of bed, with the white-collar worker group accounted for the largest proportion. In the postoperative recovery information, patients in the blue-collar worker group were more likely to have severe low back pain (OR = 2.023, 95% CI: 1.440-2.284) and pain-disturbed sleep (OR = 2.287, 95% CI: 1.585-3.299) than those who in the unemployed group.Blue-collar workers, with a high risk of thoracolumbar fracture, have a higher incidence of low back leg pain and pain-disturbed sleep in the recovery after thoracolumbar fracture surgery, and this requires more attention.

STUDY QUESTION Does cancer itself, before any gonadotoxic treatment, affect ovarian function in reproductive-aged patients? SUMMARY ANSWER Our study revealed that women with cancer may have decreased ovarian reserve markers even before cancer therapy. WHAT IS KNOWN ALREADY With the field “oncofertility” improving rapidly, cancer therapy-mediated ovarian damage is well characterised. However, there is a controversy about whether cancer itself affects ovarian function before gonadotoxic treatment. STUDY DESIGN, SIZE, DURATION We conducted a systematic meta-analysis investigating the association between cancer and ovarian function prior to gonadotoxic treatment. Titles or abstracts related to ovarian reserve [e.g., anti-Müllerian hormone (AMH), antral follicle count (AFC) or basal follicle-stimulating hormone (FSH)] combined with titles or abstracts related to the exposure (e.g., cancer*, oncolog* or malignan*) were searched in PubMed, Embase and Web of Science databases from inception to Feb 1, 2022. PARTICIPANTS/MATERIALS, SETTING, METHODS We included cohort, case-control and cross-sectional studies in English that examined ovarian reserve in reproductive-aged patients (18-45 years) with cancer compared to age-matched controls before cancer treatment. The quality of the included studies was assessed by ROBINS-I. Fixed or random effects were conducted to estimate standard or weighted mean difference (SMD or WMD, respectively) and confidence interval (CI). Heterogeneity was assessed by the Q test and I2 statistics, and publication bias was evaluated by Egger’s and Begg’s tests. MAIN RESULTS AND THE ROLE OF CHANCE The review identified 17 eligible studies for inclusion. The results showed that cancer patients had lower serum AMH levels compared to healthy controls (SMD = -0.19, 95% CI: -0.34 to -0.03, P = 0.001), especially women with hematological malignancies (SMD = -0.62, 95% CI: -0.99 to -0.24, P = 0.001). The AFC was also decreased in patients with cancer (WMD = -0.93, 95%CI: -1.79 to -0.07, P = 0.033) compared to controls, while inhibin B and basal FSH levels showed no statistically significant differences. LIMITATIONS, REASONS FOR CAUTION Serum AMH and basal FSH levels in this meta-analysis showed high heterogeneity, and the small number of studies contributing to most subgroup analyses limited the heterogeneity analysis. Moreover, the studies for specific cancer subtypes may be too small to draw conclusions; more studies are needed to investigate the possible impact of cancer type and stage on ovarian function. WIDER IMPLICATIONS OF THE FINDINGS Our study confirmed the findings that cancer per se, especially hematological malignancies, negatively affects serum AMH level and AFC values of reproductive-aged women. However, the lower AMH levels and AFCs may also be due to the changes in ovarian physiology under oncological conditions, rather than actual lower ovarian reserves. Based on the meta-analysis, clinicians should raise awareness about the possible need for personalized approaches for young women with cancer who are interested in pursuing fertility preservation strategies before anticancer treatments. Over the past decades, the incidence of cancer in young women has increased constantly. Cancer therapy-mediated damage to the ovary is well characterized, but whether cancer itself, prior to cytotoxic treatment, affects ovarian function is controversial. Ovarian reserve is generally defined as the quantity of oocytes remaining in the ovary, and markers of ovarian reserve include the hormone levels of anti-Müllerian hormone (AMH), inhibin B and basal follicle-stimulating hormone (FSH) and the sonographically measured antral follicle count (AFC). Among these, AMH and AFC are the most favourable indicators of ovarian reserve. Based on the results of this meta-analysis, patients with cancer had significant decreased serum AMH levels and AFC values before cancer treatment, while basal FSH levels and inhibin B levels showed no difference between cancer patients and control groups. From these results, we suggest that cancer itself may have negative effects on ovarian reserve. These findings may provide valuable information to clinicians and patients for choosing effective fertility preservation measures before cancer treatment.

OBJECTIVE: This study aimed to explore the clinical detection and prognosis of coagulation function in patients with liver failure and sepsis. METHODS: The plasma fibrinogen (FIB), factor II, factor VII, factor V, factor IV, antithrombin III (ATIII), platelet (PLT), mean PLT volume (MPV), D-dimer, prothrombin activity (PTA), and fibrin degradation product (FDP) levels and thromboelastogram values were detected in patients with liver failure complicated with sepsis and compared with those in the liver failure and liver cirrhosis groups. The patients with liver failure complicated with sepsis were analyzed by univariate and multivariate logistic regression, and the regression equation was established. Ovarian aging is a pacemaker with multiple organ dysfunction. Recently, stem cells with the ability to generate new oocytes have been identified, which provides the possibility of stem cell therapy for ovarian aging. Several studies have revealed the existence of stem cells in the human postmenopausal ovary. In this study, we describe a new method using magnetic-activated cell sorting combined with differential adhesion to isolate DDX4+ stem cells from ovaries of postmenopausal women and show that the cells exhibit similar gene expression profiles and growth characteristics with primitive germ cells. Furthermore, the DDX4+ stem cells could enter the meiosis stage and differentiation into oocytes. The RNA-seq data of the differentiated oocytes shows that mitochondrial metabolism may play an important role in the oogenesis process of the DDX4+ stem cells. Through using the human ovarian cortical fragments transplantation model, we indicated that the GFP-DDX4+ stem cells differentiated into some GFP positive oocyte-like structure in vivo. Our study provided a new method for the isolation of DDX4+ stem cells from the ovaries of postmenopausal women and confirmed the ability of these stem cells to differentiate into oocytes.

AimsTirzepatide is a novel dual glucose-dependent insulinotropic peptide (GIP) and glucagon-like peptide-1 receptor agonist (GLP-1 RA). At present, there is no controversy over its effectiveness, but its safety. We conducted a systematic review to assess the safety of tirzepatide.MethodsWe searched PubMed, Embase and Cochrane databases for randomized controlled trials (RCTs) of tirzepatide from databases inception to August 28, 2022 and used the Cochrane Systematic Assessment Manual Risk of Bias Assessment Tool (version 5.1) and modified Jadad scale to assess risk of bias. The systematic review was conducted via Revman5.4.ResultsNine RCTs with a total of 9818 patients were included. The overall safety profile of tirzepatide is similar to GLP-1RAs, except for the hypoglycemia (tirzepatide 15mg, pooled RR=3.83, 95% CI [1.19- 12.30], P=0.02) and discontinuation (tirzepatide 10mg, pooled RR=1.75,95%CI[1.16-2.63], P=0.007 and 15mg, pooled RR=2.03, 95%CI [1.37-3.01], P=0.0004). It also showed that the dose escalation could not rise the occurrence rates of total, severe, gastrointestinal adverse events and hypoglycemia (P>0.05); Compared with 5mg, tirzepatide 10mg and 15mg were associated with more frequent nausea (P), discontinuation (P) and injection-site reaction (P); The rates of vomiting and diarrhea were dose-dependence at the range of 5-15mg.ConclusionThe safety profile of tirzepatide is generally acceptable, similar to GLP-1 RAs. It is necessary to pay attention to its specific adverse events (hypoglycemia and discontinuation) at high doses (10mg or higher). Nausea, vomiting, diarrhea, discontinuation and injection-site reaction were dose-dependence among specific dose ranges.As the heterogeneity in different studies by interventions, the results may be with biases and the further confirmation is needed. Meanwhile, more well-designed trials are needed to control the confounding factors and ensure adequate sample size.

Semaphorins are a family of evolutionarily conserved morphogenetic molecules that were initially found to be associated with axonal guidance. Semaphorin 4C (Sema4C), a member of the fourth subfamily of semaphorins, has been demonstrated to play multifaceted and important roles in organ development, immune regulation, tumor growth, and metastasis. However, it is completely unknown whether Sema4C is involved in the regulation of ovarian function. We found that Sema4C was widely expressed in the stroma, follicles, and corpus luteum of mouse ovaries, and its expression was decreased at distinct foci in ovaries of mice of mid-to-advanced reproductive age. Inhibition of Sema4C by the ovarian intrabursal administration of recombinant adeno-associated virus-shRNA significantly reduced oestradiol, progesterone, and testosterone levels in vivo. Transcriptome sequencing analysis showed changes in pathways related to ovarian steroidogenesis and the actin cytoskeleton. Similarly, knockdown of Sema4C by siRNA interference in mouse primary ovarian granulosa cells or thecal interstitial cells significantly suppressed ovarian steroidogenesis and led to actin cytoskeleton disorganization. Importantly, the cytoskeleton-related pathway RHOA/ROCK1 was simultaneously inhibited after the downregulation of Sema4C. Furthermore, treatment with a ROCK1 agonist after siRNA interference stabilized the actin cytoskeleton and reversed the inhibitory effect on steroid hormones described above. In conclusion, Sema4C may play an important role in ovarian steroidogenesis through regulation of the actin cytoskeleton via the RHOA/ROCK1 signaling pathway. These findings shed new light on the identification of dominant factors involved in the endocrine physiology of female reproduction.

Sarglamides A–E (1−5), representing the first example of heterodimers of a trans-N-cinnamoylindolidinoid and an α-phelladrene derivatives, were isolated from Sarcandra glabra subsp. brachystachys. Particularly, compounds 4 and 5 possess unprecedented cage-like 6/6/5/6/5- and 6/6/6/6/5-fused pentacyclic scaffolds, respectively. Their structures were established by spectroscopic analysis, X-ray crystallography, quantum chemical calculations, and chemical conversions. Plausible biosynthetic pathways of 1−5, involving the co-isolated enantiomers 6a and 6b were proposed. Compounds 3–7 showed inhibitory activity against lipopolysaccharide (LPS) induced inflammation in BV-2 microglial cells.

Social metaverse is a shared digital space combining a series of interconnected virtual worlds for users to play, shop, work, and socialize. In parallel with the advances of artificial intelligence (AI) and growing awareness of data privacy concerns, federated learning (FL) is promoted as a paradigm shift towards privacy-preserving AI-empowered social metaverse. However, challenges including privacy-utility tradeoff, learning reliability, and AI model thefts hinder the deployment of FL in real metaverse applications. In this paper, we exploit the pervasive social ties among users/avatars to advance a social-aware hierarchical FL framework, i.e., SocialFL for a better privacy-utility tradeoff in the social metaverse. Then, an aggregator-free robust FL mechanism based on blockchain is devised with a new block structure and an improved consensus protocol featured with on/off-chain collaboration. Furthermore, based on smart contracts and digital watermarks, an automatic federated AI (FedAI) model ownership provenance mechanism is designed to prevent AI model thefts and collusive avatars in social metaverse. Experimental findings validate the feasibility and effectiveness of proposed framework. Task allocation plays a vital role in multi-robot autonomous cleaning systems, where multiple robots work together to clean a large area. However, most current studies mainly focus on deterministic, single-task allocation for cleaning robots, without considering hybrid tasks in uncertain working environments. Moreover, there is a lack of datasets and benchmarks for relevant research. In this paper, to address these problems, we formulate multi-robot hybrid-task allocation under the uncertain cleaning environment as a robust optimization problem. Firstly, we propose a novel robust mixed-integer linear programming model with practical constraints including the task order constraint for different tasks and the ability constraints of hybrid robots. Secondly, we establish a dataset of \emph{100} instances made from floor plans, each of which has 2D manually-labeled images and a 3D model. Thirdly, we provide comprehensive results on the collected dataset using three traditional optimization approaches and a deep reinforcement learning-based solver. The evaluation results show that our solution meets the needs of multi-robot cleaning task allocation and the robust solver can protect the system from worst-case scenarios with little additional cost. To prevent implicit privacy disclosure in sharing gradients among data owners (DOs) under federated learning (FL), differential privacy (DP) and its variants have become a common practice to offer formal privacy guarantees with low overheads. However, individual DOs generally tend to inject larger DP noises for stronger privacy provisions (which entails severe degradation of model utility), while the curator (i.e., aggregation server) aims to minimize the overall effect of added random noises for satisfactory model performance. To address this conflicting goal, we propose a novel dynamic privacy pricing (DyPP) game which allows DOs to sell individual privacy (by lowering the scale of locally added DP noise) for differentiated economic compensations (offered by the curator), thereby enhancing FL model utility. Considering multi-dimensional information asymmetry among players (e.g., DO's data distribution and privacy preference, and curator's maximum affordable payment) as well as their varying private information in distinct FL tasks, it is hard to directly attain the Nash equilibrium of the mixed-strategy DyPP game. Alternatively, we devise a fast reinforcement learning algorithm with two layers to quickly learn the optimal mixed noise-saving strategy of DOs and the optimal mixed pricing strategy of the curator without prior knowledge of players' private information. In commercial unmanned aerial vehicle (UAV) applications, one of the main restrictions is UAVs' limited battery endurance when executing persistent tasks. With the mature of wireless power transfer (WPT) technologies, by leveraging ground vehicles mounted with WPT facilities on their proofs, we propose a mobile and collaborative recharging scheme for UAVs in an on-demand manner. Specifically, we first present a novel air-ground cooperative UAV recharging framework, where ground vehicles cooperatively share their idle wireless chargers to UAVs and a swarm of UAVs in the task area compete to get recharging services. Considering the mobility dynamics and energy competitions, we formulate an energy scheduling problem for UAVs and vehicles under practical constraints. A fair online auction-based solution with low complexity is also devised to allocate and price idle wireless chargers on vehicular proofs in real time. We rigorously prove that the proposed scheme is strategy-proof, envy-free, and produces stable allocation outcomes. The first property enforces that truthful bidding is the dominant strategy for participants, the second ensures that no user is better off by exchanging his allocation with another user when the auction ends, while the third guarantees the matching stability between UAVs and UGVs. Due to the limited battery and computing resource, offloading unmanned aerial vehicles (UAVs)' computation tasks to ground infrastructure, e.g., vehicles, is a fundamental framework. Under such an open and untrusted environment, vehicles are reluctant to share their computing resource unless provisioning strong incentives, privacy protection, and fairness guarantee. Precisely, without strategy-proofness guarantee, the strategic vehicles can overclaim participation costs so as to conduct market manipulation. Without the fairness provision, vehicles can deliberately abort the assigned tasks without any punishments, and UAVs can refuse to pay by the end, causing an exchange dilemma. Lastly, the strategy-proofness and fairness provision typically require transparent payment/task results exchange under public audit, which may disclose sensitive information of vehicles and make the privacy preservation a foremost issue. To achieve the three design goals, we propose SEAL, an integrated framework to address strategy-proof, fair, and privacy-preserving UAV computation offloading. SEAL deploys a strategy-proof reverse combinatorial auction mechanism to optimize UAVs' task offloading under practical constraints while ensuring economic-robustness and polynomial-time efficiency. Based on smart contracts and hashchain micropayment, SEAL implements a fair on-chain exchange protocol to realize the atomic completion of batch payments and computing results in multi-round auctions. In addition, a privacy-preserving off-chain auction protocol is devised with the assistance of the trusted processor to efficiently protect vehicles' bid privacy. In order to save computing power yet enhance safety, there is a strong intention for autonomous vehicles (AVs) in future to drive collaboratively by sharing sensory data and computing results among neighbors. However, the intense collaborative computing and data transmissions among unknown others will inevitably introduce severe security concerns. Aiming at addressing security concerns in future AVs, in this paper, we develop SPAD, a secured framework to forbid free-riders and {promote trustworthy data dissemination} in collaborative autonomous driving. To defend against free-riding attacks,} we formulate the interactions between publisher AVs and subscriber AVs as a vehicular publish/subscribe game, {and incentivize AVs to deliver high-quality data by analyzing the Stackelberg equilibrium of the game. We also design a reputation evaluation mechanism in the game} to identify malicious AVs {in disseminating fake information}. {Furthermore, for} lack of sufficient knowledge on parameters of {the} network model and user cost model {in dynamic game scenarios}, a two-tier reinforcement learning based algorithm with hotbooting is developed to obtain the optimal {strategies of subscriber AVs and publisher AVs with free-rider prevention}. Extensive simulations are conducted, and the results validate that our SPAD can effectively {prevent free-riders and enhance the dependability of disseminated contents,} compared with conventional schemes.
