The imbalance between the supply and demand of shared bikes is prominent in many urban rail transit stations, which urgently requires an efficient vehicle deployment strategy. In this paper, we propose an integrated model to optimize the deployment of shared bikes around urban rail transit stations, incorporating a seasonal autoregressive integrated moving average with long short-term memory (SARIMA-LSTM) hybrid model that is used to predict the heterogeneous demand for shared bikes in space and time. The shared bike deployment strategy was formulated based on the actual deployment process and under the principle of cost minimization involving labor and transportation. The model is applied using the big data of shared bikes in Xicheng District, Beijing. Results show that the SARIMA-LSTM hybrid model has great advantages in predicting the demand for shared bikes. The proposed allocation strategy provides a new way to solve the imbalance challenge between the supply and demand of shared bikes and contributes to the development of a sustainable transportation system.

Functional genomics, synthetic biology and metabolic engineering require efficient tools to deliver long DNA fragments or multiple gene constructs. Although numerous DNA assembly methods exist, most are complicated, time-consuming and expensive. Here, we developed a simple and flexible strategy, unique nucleotide sequence-guided nicking endonuclease (UNiE)-mediated DNA assembly (UNiEDA), for efficient cloning of long DNAs and multigene stacking. In this system, a set of unique 15-nt 3' single-strand overhangs were designed and produced by nicking endonucleases (nickases) in vectors and insert sequences. We introduced UNiEDA into our modified Cre/loxP recombination-mediated TransGene Stacking II (TGSII) system to generate an improved multigene stacking system we call TGSII-UNiE. Using TGSII-UNiE, we achieved efficient cloning of long DNA fragments of different sizes and assembly of multiple gene cassettes. Finally, we engineered and validated the biosynthesis of betanin in wild tobacco (Nicotiana benthamiana) leaves and transgenic rice (Oryza sativa) using multigene stacking constructs based on TGSII-UNiE. In conclusion, UNiEDA is an efficient, convenient and low-cost method for DNA cloning and multigene stacking, and the TGSII-UNiE system has important application prospects for plant functional genomics, genetic engineering and synthetic biology research.

BackgroundWhile the contralateral seventh cervical nerve (CC7) cross transfer was designed to reconstruct paralyzed arm function after stroke, improvement in language function was found in patients combined with aphasia.ObjectiveTo evaluate the effect of improvement in language function after CC7 cross transfer in stroke patients with chronic aphasia and explore its potential mechanism.MethodsIn a prospective observative cohort, patients diagnosed with hemiplegia combined with aphasia were included. The language function was evaluated through the changes of Aphasia Quotient evaluated by Western Aphasia Battery (WAB-AQ) as well as its four subtests from baseline to 1 week and 6 months after the surgery. Patients also received oral agility test by Boston Diagnostic Aphasia Examination (BDAE-OA). Purpose:IDH1/2-mutant gliomas harbor a distinct glioma-CpG island methylation phenotype (G-CIMP) that may promote the initiation and progression of secondary pathway gliomas by silencing tumor-suppressive genes. The potential role of tumor-suppressive microRNAs (miRNA; miR) in this process is not understood.Experimental Design: To identify potential tumor-suppressive miRNA hypermethylated in glioma, the methylation profiles of IDH1/2WT gliomas (n = 11) and IDH1MUT glioma (n = 20) were compared by using massively parallel reduced representation bisulfite sequencing (RRBS). The methylation status of selected miRNA was validated by using targeted bisulfite sequencing (BiSEQ) in a large cohort of glioma tissue samples including 219 IDH1WT and 72 IDH1/2MUT samples. The expression of selected miRNAs was determined by using the TaqMan qPCR. Functional analyses of miR148a were conducted and target genes were identified.Results: We identify miR148a as a novel, G-CIMP–associated miRNA whose methylation is tightly correlated with IDH1 mutation and associated with improved survival in patients with malignant glioma. We confirm that downregulation of miR148a can occur via DNA methylation. We demonstrate that IDH1 mutation provides a mechanism of miR148a methylation and downregulation, and that restoration of miR148a reduced tumorigenic properties of glioma cells, possibly by targeting DNMT1.Conclusions: We identify miR148a as a novel G-CIMP–associated miRNA, and provide results suggesting that miR148a restoration may have therapeutic implications. BackgroundPost-stroke aphasia is a common but intractable sequela which still needs new and more effective treatments. Evidence from follow-ups after contralateral seventh cervical nerve transfer surgery indicated that nerve transection leads to immediate language improvements in patients with right post-stroke aphasia.ObjectiveThrough a prospective cohort design, this study aims to prove that C7 neurotomy at the intervertebral foramen (NC7) combined with a 3-week intensive speech and language therapy (iSLT) can improve the language function in post-stroke aphasia patients.MethodsIn this study, patients aged over 18 years old and had been diagnosed with post-stroke aphasia for 1 year or longer were included. Primary outcomes were the change in the ability to retrieve personally relevant words in Boston Naming Test (BNT) with follow-up assessment after three-weeks’ iSLT post-operatively. As well as several secondary outcome measures including the Western Aphasia Battery (WAB), daily communication abilities (measured by the Communication Activities of Daily Living Third Edition [CADL-3]) and Fugl-Meyer of upper limb part (UEFM).ResultsThe average increase of BNT score was 11.2 points from baseline to 3 weeks post-operatively (P=0.001, 95%CI: 8.1-14.1). The WAB and CADL-3 assessment showed 9.4, 10.4 points increasing in average (PConclusionsNC7 plus iSLT significantly improved the language function in patients with post-stroke aphasia, and did not significantly affect the motor function of the right limb. The mechanism of this surgery needs to be further explored.

Purpose To investigate the role of high mobility group protein A2 (HMGA2) in the epithelial-mesenchymal transition (EMT) of kidney cancer. Methods The renal carcinoma cell line ACHN were transfected using RNA interference technology (siRNA) to develop a cell line with low HMGA2 expression. The expression of E-cadherin, N-cadherin, and zinc finger protein (Snail) was detected using quantitative real-time polymerase chain reaction and Western blot at the mRNA and protein levels. Results The results show that the mechanism of HMGA2 action on kidney cancer cells is related to EMT, and this action is achieved by influencing the EMT marker proteins E-cadherin and N-cadherin. The mechanism where HMGA2 exerts its biological function is related to the transforming growth factor-β (TGF-β) signaling pathway, where HMGA2 is able to interact with Smad2 and Smad3. In the TGF-β signaling pathway, HMGA2 can bind and form a complex with Smad2 and Smad3 to induce the expression of the target gene, Snail, promote the production of Snail protein, and induce EMT in kidney cancer cells, thereby promoting the occurrence and development and metastasis of kidney cancer cells. Conclusion HMGA2 is closely related to the migration and invasive ability of kidney cancer cells and can promote EMT of kidney cancer cells by activating the intracellular TGF-β signaling pathway. In kidney cancer cells, the intracellular TGF-signaling pathway promotes EMT.

This article presents a novel high temperature heat flux (HTHF) sensor based on micromachined thermocouple (MTC) to meet the needs of transient high heat flux detection in ultrahigh temperature environments, such as fire research, high-power pulsed lasers testing, and aerospace engineering. The targeted design, fabrication, and package of an MTC-based HTHF sensor with excellent heat dissipation capacity are proposed, and relevant experiments were conducted in two aspects. First, powerful laser tube-based tests at ambient temperature show that such a sensor can detect a heat flux high to 10.8 MW/m2 with an extremely fast response; its response time, in this case, is 1.9 ms. And then, turbine engine test pipe based tests at high temperature reveal that the sensor with water cooling could survive at 1200 °C for at least 100 s. Compared to conventional heat flux sensors, it is more sensitive to transient heat flux changes and has a faster response speed. Such satisfactory performance makes the sensor extremely competitive in ultrahigh heat flux detection in extremely harsh environments.

The developed biosensor detection system can complete the detection of air samples by collecting exhaled breath condensate, greatly reducing the time to diagnose tuberculosis.

The steel turnout is one of the key components in the medium–low-speed maglev line system. However, the vehicle under active control is prone to vehicle–turnout coupled vibration, and thus, it is necessary to identify the vibration characteristics of this coupled system through field tests. To this end, dynamic performance tests were conducted on a vehicle–turnout coupled system in a medium–low-speed maglev test line. Firstly, the dynamic response data of the coupled system under various operating conditions were obtained. Then, the natural vibration characteristics of the turnout were analysed using the free attenuation method and the finite element method, indicating a good agreement between the simulation results and the measured results; the acceleration response characteristics of the coupled system were analysed in detail, and the ride quality of the vehicle was assessed by Sperling index. Finally, the frequency distribution characteristics of the coupled system were discussed. All these test results could provide references for model validation and optimized design of medium–low-speed maglev transport systems.

Coal mass is subjected to cyclic loading during pulsating hydraulic fracturing (PHF), and changes in its gas desorption properties affect gas drainage. Therefore, it is of great importance to correctly understand the influences of cyclic loading on the gas desorption properties of coal mass. Firstly, loading tests with different frequencies and amplitudes were performed on anthracite from Qinshui Basin (Shanxi Province, China) using a fatigue testing machine. Secondly, gas desorption tests were performed to determine the associated curves for each test group at different equilibrium pressures, and the initial desorption capacity and diffusion coefficient of the gas were calculated. Finally, the influence of different loading conditions on the gas desorption laws were analyzed. The test results demonstrate that a greater loading frequency increases the ratio of the initial desorption capacity so that the desorption rate of coal samples is higher, and the gas desorption properties become increasingly better in the initial stage. However, variations in the amplitude have minimal impact on the ratio of the initial desorption capacity. When the amplitude is too large in the initial stage, the diffusion coefficient decreases and the gas desorption properties worsen. In addition, the above test results are used to discuss the selection of the amplitude and frequency in the PHF process from a macroperspective. The contained research results provide an important theoretical basis for the field application of PHF technologies in coal mines.

Background Tumor surveillance of isocitrate dehydrogenase (IDH) mutant gliomas is accomplished via serial contrast MRI. When new contrast enhancement (CEnew) is detected during postsurgical surveillance, clinicians must assess whether CEnew indicates pseudoprogression (PsP) or tumor progression (TP). PsP has been better studied in IDH wild-type glioblastoma but has not been well characterized in IDH mutant gliomas. We conducted a retrospective study evaluating the incidence, predictors, natural history, and survival of PsP patients in a large cohort of IDH mutant glioma patients treated at a single institution. Methods We identified 587 IDH mutant glioma patients treated at UCLA. We directly inspected MRI images and radiology reports to identify CEnew and categorized CEnew into TP or PsP using MRI or histopathology. Results Fifty-six percent of patients developed CEnew (326/587); of these, 92/326 patients (28% of CEnew; 16% of all) developed PsP and 179/326 (55%) developed TP. All PsP patients had prior radiation, chemotherapy, or chemoradiotherapy. PsP was associated with longer overall survival (OS) versus TP patients and similar OS versus no CEnew. PsP differs from TP based on earlier time of onset (median 5.8 vs 17.4 months from treatment, P < .0001) and MRI features that include punctate enhancement and enhancement location. Conclusion PsP patients represented 28% of CEnew patients and 16% of all patients; PsP patients demonstrated superior outcomes to TP patients, and equivalent survival to patients without CEnew. Crohn's disease (CD) is a chronic recurrent gastrointestinal disorder with a high incidence of inflammation due to intestinal inflammation. Acupuncture is commonly used as an alternative therapy for patients with CD. The aim of this study was to design a systematic review and meta-analysis protocol, to provide guidance for the establishment of systematic evaluation and meta-analysis on the efficacy and safety of acupuncture on CD.We will search PubMed, the Cochrane Library, Embase, Web of Science, and 4 Chinese databases: China National Knowledge Infrastructure (CNKI), Wanfang database, VIP database, and Chinese Biomedical Database to obtain randomized controlled trials of CD treated with acupuncture from inception to November 5, 2022. Primary outcome include CD symptoms severity and clinical efficacy, secondary outcome indicators include laboratory indicators or inflammatory markers, severity of endoscopic lesions, quality of life, and safety outcomes. We will analyze the data using RevMan V.5.4 software. Two reviewers will assess the risk of bias and study quality by the Cochrane Collaboration Risk of Bias Tool and GRADE methods, respectively.This systematic review and meta-analysis protocol will analyze the efficacy, symptom improvement, quality of life, and safety of acupuncture therapy for CD.This protocol outlines the planned scope and methodology of a forthcoming systematic review and meta-analysis to provide guidelines for a rigorous assessment of the efficacy and safety of acupuncture for the treatment of CD.

In order to improve the ability of community governance and reduce the occurrence of accidents, based on the entropy and self-organization theory, this paper first analyzes the influencing factors of community resilience, analyzes the process of emergency and the key factors of response from the change of entropy value of community security system, constructs the mathematical expression of community resilience and entropy value, extracts the evolution law of community resilience, and then constructs the decision theory of building entropy effect and improving community resilience from the perspective of input-output benefit. The results show that the community resilience is positively correlated with the absolute value of negative entropy; the community resilience is negatively correlated with the absolute value of positive entropy; the system resilience can be improved by allocating the limited degree and quantity value according to the importance of the impact on the correlation of each subsystem in the community system.

The presence of decentralized production in modern electrical systems precludes traditional automatic generation control (AGC) strategies. Renewable energy production is highly dependent on the environment; for example, photovoltaic (PV) system, but its power generation is very unstable, which makes power grids containing PV systems face greater challenges. In this paper, we propose a PV-grid control method based on self-gain system compensation to reduce the negative effects of large fluctuations and uncertainties in solar energy production. First, the performance characteristics of solar modules are discussed and a multi-region model with a system with high penetration of solar systems is built. The AGC power is then displayed with different power variations according to different maximum power point tracking methods to analyze the uncertainty and variation of the PV output power and its effects on the AGC. We have also developed and implemented a compensation unit to eliminate the negative effects of PV power on AGC. Finally, the proposed method was demonstrated using the load from the example step as a disturbance variable to obtain a dynamic model of a dual-zone switched grid AGC with two PV systems based on a DMPC-distributed model. As a result of the simulation, the proposed method in this paper can guarantee the excellent properties of the dynamic behavior of the AGC system with large variations and high uncertainty of the PV system, and the validity of the proposed method have also been confirmed.
