Several previous studies on YouTube™ on urological field have already been published. The aim of the current study was to evaluate the quality information of YouTubeTM videos on testicular pain. Using Google Trends tool, the frequency of worldwide YouTubeTM and Google Search on testicular pain was examined from 2010 to 2020. The keywords “testicular pain”, “testicular ache” and “scrotal pain” were used on the YouTube platform and the first 100 YouTubeTM videos were analyzed for each one. The Patient Education Materials Assessment Tool (PEMAT) for Audiovisual (A/V) Materials, the DISCERN score and Misinformation tool were used to assess video quality. According to YouTube™ Search the mean relative frequency search for “testicular pain” ranged from 10.5 to 30.0%. According to GoogleTM Search it ranged from 73.7 to 91.0%, Of all 300 videos, 117 were eligible for the analysis. The median number of views, thumbs-up and thumbs-down was respectively: 47060 (interquartile range [IQR] = 6297.0–144188.0), 289 (IQR = 40–912) and 19 (IQR = 4–53). Of all videos, 68.4% and 31.6% were produced respectively by Medical Doctors and Other. The median PEMAT Actionability and Understandability scores were 66.7% and 66.7%, respectively. The median DISCERN score ranged from 1 to 5, with an overall median score of 3, defined from question 16. The median misinformation score ranged from 2 to 5. In conclusion, an increased interest on testicular pain was recorded on both YouTubeTM and Google search during the last decade. However, according to the quality assessment tools used, YouTube™ users cannot get trustful and exhaustive information on testicular pain. Therefore, authors with/without medical background should improve the quality of information on YouTube™ videos.

The objective of the current research was to explore the potential prognostic value of readily available clinical and pathologic variables in bladder cancer. The novel association found between cholesterol levels and prognosis may provide the rationale for exploring novel treatments. Patients included had histologically confirmed urothelial bladder cancer and were treated with at least 3 cycles of cisplatin-based neoadjuvant chemotherapy before radical cystectomy with lymphadenectomy. A total of 245 patients at low, intermediate and high risk, presenting with 0-1, 2 or 3-4 risk factors, including positive lymph nodes, Hb 12.8, NLR ≥2.7 and cholesterol levels ≥199, were included. Five-year cancer-specific survival rate was 0.67, 0.78 and 0.94 at high, intermediate and low risk, respectively. Total cholesterol levels at the time of cystectomy may represent a commonly assessable prognostic factor and may be incorporated in a clinically meaningful risk-group classification model.Lay abstract This present study assessed a large group of patients with urothelial bladder cancer treated with chemotherapy followed by radical cystectomy, to capture the predictive power of commonly collected clinical, pathological and biochemical factors. The design of the study highlighted that higher cholesterol levels at the time of cystectomy were associated with shorter cancer-specific survival. This finding suggests that high blood-cholesterol levels truly have a negative influence on surviving cancer. Wild boar Sus scrofa populations have increased dramatically in recent decades throughout Europe. While hunting is widely used in management activities; it rarely has an important role in regulating and reducing wild boar populations. Therefore, increasing the efficiency of hunting is a compelling issue. In this study, we used a three-year dataset (2016–2018) on a wild boar population living in Campania (southern Italy) as a case study to explore how the hunting effort made in collective drive hunts affected the hunting rate, estimated as the number of individuals culled per day. We fitted a Linear Mixed Model, in which we included the number of wild boars culled per drive hunt as the dependent variable, and the number of beaters, shooters and dogs and the month during which hunting occurred as the predictors. A mean of 1.81 wild boars were culled per drive hunt. The number of culled animals per hunt increased with the increasing number of hunting dogs and with the progression of the hunting season (i.e., from October to December), whereas the number of beaters and shooters had no effect. Overall, we observed a low hunting rate. We suggest that adjusting the hunting calendar and reorganising wild boar collective hunts, e.g., through an appropriate management of the number and training of hunting dogs, are essential to increase the hunting rate. Our results can be useful for wildlife managers to enhance hunting contribution in counteracting the negative impact of wild boar.

Background: Prostate volume (PV) is a useful tool in risk stratification, diagnosis, and follow-up of numerous prostatic diseases including prostate cancer and benign prostatic hypertrophy. There is currently no accepted ideal PV measurement method. Objective: This study compares multiple means of PV estimation, including digital rectal examination (DRE), transrectal ultrasound (TRUS), and magnetic resonance imaging (MRI), and radical prostatectomy specimens to determine the best volume measurement style. Methods: A retrospective, observational, single-site study with patients identified using an institutional database was performed. A total of 197 patients who underwent robot-assisted radical prostatectomy were considered. Data collected included age, serum PSA at the time of the prostate biopsy, clinical T stage, Gleason score, and PVs for each of the following methods: DRE, TRUS, MRI, and surgical specimen weight (SPW) and volume. Results: A paired t test was performed, which reported a statistically significant difference between PV measures (DRE, TRUS, MRI ellipsoid, MRI bullet, SP ellipsoid, and SP bullet) and the actual prostate weight. Lowest differences were reported for SP ellipsoid volume (M = −2.37; standard deviation [SD] = 10.227; t[167] = −3.011; and p = 0.003), MRI ellipsoid volume (M = −4.318; SD = 9.53; t[167] = −5.87; and p = 0.000), and MRI bullet volume (M = 5.31; SD = 10.77; t[167] = 6.387; and p = 0.000). Conclusion: The PV obtained by MRI has proven to correlate with the PV obtained via auto-segmentation software as well as actual SPW, while also being more cost-effective and time-efficient. Therefore, demonstrating that MRI estimated the PV is an adequate method for use in clinical practice for therapeutic planning and patient follow-up.

Two satellite DNAs (satDNAs) have been isolated and characterized from three populations of Atlantolacerta andreanskyi. One satDNA (AAN-TaqI) has been isolated here from the first time. It is characterized by a tendency to AT enrichment (AT = 54.2%) and monomer length ranging from 187 to 199 bp. FISH experiments showed that this element occurs in subterminal position on the short arms of all chromosomes of the complement. The analyses of genetic variability of AAN-TaqI showed that the concerted evolution is acting effectively on these repeats that form separate clusters consistent with the geographic origin in the phylogenetic tree, thus supporting the hypothesis that A. andreanskyi would be a species complex. In addition, in the population from Jbel Aoulime this satDNA is already differentiated into two subfamilies. The other satDNA belongs to the family of IMO-TaqI already isolated in other lacertids. Differently from AAN-TaqI, concerted evolution does not seem to act effectively on this element that is not differentiated between populations. These results confirm that IMO-TaqI (AT = 53.4%) is conserved in both chromosomal position and most of its sequence in the lacertids from which it has been characterized so far. Its remarkable evolutionary conservation for about 45 million years could indicate that this satDNA may have a functional role that future investigations could unveil. Once again, this study shows how satDNAs coexisting in the same genome may differ in their evolutionary pattern, even though the reasons underlying this phenomenon in the species here studied have still to be fully understood.

The brown trout Salmo trutta is characterised by both anadromous (sea trout) and resident populations, naturally occurring in Atlantic and Ponto‐Caspian rivers. Sea trout are currently considered absent from rivers of the Mediterranean area, probably because of the non‐optimal chemical–physical characteristics of the Mediterranean Sea. However, the occasional bycatch of smoltified S. trutta in the Adriatic Sea is well known among fishermen and the biological explanation of this phenomenon is still controversial. The aim of this study was to compare the genetic diversity of freshwater and marine brown trout to try to understand the factors underlying the presence of putative anadromous brown trout in the Adriatic Sea. In this study, we analysed the genetic diversity of: (1) wild brown trout collected from the Esino River (central Italy); (2) a domestic strain of brown trout used for stocking the study area; and (3) a sample of Adriatic sea trout collected near the outlet of the Esino River. Together with genetic analysis, we carried out scale analysis in order to track the freshwater/marine stages of the life cycle in the sea trout samples. The genetic characterisation was carried out by polymerase chain reaction–restriction fragment length polymorphism analysis of the mtDNA fragment ND‐5/6 and the nuclear locus LDH‐C1* and by genotyping 15 microsatellite loci. The genetic polymorphism obtained was used to investigate intra‐ and inter‐population genetic diversity, rates of genetic introgression between wild and domestic samples and the origin of sea trout specimens by using assignment tests. Our genetic analyses demonstrated that the sea trout analysed in this study are from the domestic strain of Atlantic origin used in central Italy for stocking activities. The level of genetic introgression between native and domestic samples is high in the Esino River. The populations more resilient to introgressive hybridisation appeared to be those living in the portion of the river network dominated by carbonate rocks. Assignment tests (GeneClass) suggest the existence of a link between stocking efforts and the freshwater origin of the sea trout. In addition, data obtained from the analysis of scales, size measurement, and sex determination showed a pattern of smolt age, size, and sex ratio very similar to those observed in other anadromous populations. In conclusion, the present study highlighted that sea trout from the central Adriatic Sea originated from brown trout of Atlantic origin inhabiting the Esino River. Their seaward migratory behaviour could represent a consequence of an active migration instead of a passive displacement by water flow. Our results also showed that traditional stocking practices represent a negative activity for the conservation of the last Mediterranean native S. trutta populations.

We investigated the relationship between age and body length, and age at sexual maturity of Physeter macrocephalus individuals stranded along the Italian coast. Our molecular analysis shows that all our samples belong to the C.001.002 haplotype, shared between Atlantic and Mediterranean populations. We show that males attain sexual maturity at 10 years, similar to those from other marine areas. However, considering the same body length class, Mediterranean males are older than Atlantic ones. Our finding of a Mediterranean pregnant female of only 6.5 m in length and an assessed age of 24–26 years is particularly noteworthy, considering that females reach sexual maturity at about 9 years and 9 m of total length in other regions. Comparing our results with the literature data, we highlight the positive correlation between lifespan, adult body length and weight of males from the Mediterranean and Atlantic Ocean. Regardless of whether the relatively small size of Mediterranean specimens is a consequence of an inbreeding depression or an adaptation to less favorable trophic conditions, we recommend to closely monitor this population from a conservation perspective. In fact, its low genetic diversity likely corresponds to a relatively limited ability to respond to environmental changes compared with other populations.

Mediterranean brown trout is subject to several serious threats such as pollution, water abstraction, habitat alteration and especially genetic introgression with domestic strains used for stocking activities. Despite this latter issue has largely been debated by scientists, local managers and stakeholders for decades, official stocking practices with domestic trout still persists in several countries (Italy included), even if there are laws explicitly prohibiting introduction of organisms of non-local origin. Probably, the last opportunity to conserve native brown trout populations is represented by protected areas. Therefore, in the present study, we aimed to verify the role of the Nature 2000 network and a national park as valid tools to guarantee the survival of native brown trout in the Apennines. Partial mitochondrial DNA control region sequence analysis and genotyping of the locus LDH-C1* and 11 microsatellites were used to investigate the genetic diversity of three rivers from central Italy. For all rivers investigated a temporal analysis of introgression was also carried out. The genetic diversity of three domestic stocks was included in the sampling design for comparison. The main results of this study indicated that: (i) the genetic diversity of brown trout in central Italy is very complex and (ii) its conservation is seriously threatened by genetic introgression phenomena still ongoing. The only samples showing no introgression or a decrease in genetic introgression were those isolated by the presence of natural and/or artificial barriers to fish movements rather than protected by inhabiting rivers within the Natura 2000 network. This observation prompts an important reflection on issues concerning fluvial continuity restoration and suggests that barrier removal should be undertaken with caution in order to avoid the concrete risk of domestic trout spreading that could promote additional loss of native brown trout biodiversity.

Pleurodont lizards are characterized by an ancient system of sex chromosomes. Along with stability of the central component of the system (homologous to the X chromosome of Anolis carolinensis [Dactyloidae], ACAX), in some genera the ancestral sex chromosomes are fused with microautosomes, forming neo-sex chromosomes. The genus Ctenonotus (Dactyloidae) is characterized by multiple X1X1X2X2/X1X2Y sex chromosomes. According to cytogenetic data, the large neo-Y chromosome is formed by fusion of the ancestral Y chromosome with 2 microautosomes (homologous to ACA10 or ACA11 and ACA12), the X1 chromosome is formed by fusion of the ancestral X chromosome with the autosome homologous to ACA10 or ACA11, and the X2 chromosome is homologous to autosome ACA12. To determine more precisely the content and evolution of the Ctenonotus sex chromosomes, we sequenced flow-sorted chromosomes (both sex chromosomes and microautosomes as control) of 2 species with a similar system: C. pogus and C. sabanus. Our results indicate that the translocated part of the X1 is homologous to ACA11, X2 is homologous to ACA12, and the Y contains segments homologous to both ACA11 and ACA12. Molecular divergence estimates suggest that the ancestral X-derived part has completely degenerated in the Y of Ctenonotus, similar to the degeneration of the Norops sagrei Y chromosome (Dactyloidae). The newly added regions show loss of DNA content, but without degeneration of the conserved regions. We hypothesize that the translocation of autosomal blocks onto sex chromosomes facilitated rapid degeneration of the pseudoautosomal region on the ancestral Y.

Introduction: The recent advances in technology and miniaturization of endoscopic devices have permitted the use of retrograde intra renal surgery (RIRS) to treat large and complicated kidney stones as first line therapy in alternative to percutaneous nephrolithotomy (PCNL). Systematically review the efficacy and safety of RIRS for large renal stones over 2 cm versus the current gold standard, the percutaneous nephrolithotomy. Evidence acquisition: A large search was effected in PubMed, Cochrane Library, Embase, Ovid and Scopus regarding the treatment of renal stones over 2 cm with RIR S versus PCNL. Articles not in English and not regarding adult population were excluded. The retrieval time included a time span from 2000 to 2019. All clinical trials were further evaluated about quality and references. The eligible studies were included and analysed with RevMan 5.2 Software. Evidence synthesis: Two randomized and nine non-randomized studies were included for a total of 1618 patients involved. Our meta-analysis showed no difference in SFR (RR =0.92, 95% CI : 0.86-0.99, P=0.03) and in mean operation time (WMD=6.34 min, 95% CI : -4.98 to 17.65, P=0.27) while shorter hospital stay was reported for RIR S (WMD=-2.15 days, 95% CI: -3.04 to -1.25, P≤0.00001). We reported moreover lower Hb drop (WMD=-0.83 g/dL, 95% CI: -1.20 to -0.45, P≤0.00001) and complications rate in favor of RIRS (RR=0.88, 95% CI: 0.71-1.09, P=0.23). Conclusions: RIRS is challenging PCNL for the treatment of large renal stones over 2cm, becoming a safe and effective alternative with a comparable stone free rate, lower complication rate and lower hospitalization time. It is, however, of the uttermost importance to share the treatment decision with the patient due to the possibility of requiring multiple RIR S session to completely clear larger stone burdens.

Evolutionary studies suffer of the impossibility of directly observing events that occurred far back in time, on a temporal scale much greater than human life. In our paper we describe and discuss the approaches that different branches of natural sciences have adopted to overcome this inherent and unsurpassable difficulty. We, for instance, explain how chemical features of geological stratigraphy can be used to deduce the conditions in which they formed, using known chemical and physical processes as models. We provide examples of how structural and compositional similarities are used by molecular biologists to infer ancestral traits. We describe how ancient DNA can be used to determine changes in genetic structure of organisms over evolutionary times. We provide instances of reconstruction of paleoenvironments to investigate the relationship between organismal functions and ecosystems in remote times. All these approaches allow to circumvent the unobservability of the past through rigorous approaches and solid assumptions, which, although they may not satisfy a strict Galilean method, constitute a scientifically sound way to investigate the biological past.

Small pelagic fish play a key role in the marine ecosystem, controlling predator abundance and regulating primary production rates by foraging on plankton. Alterations induced at different ecological levels by fishing activities and/or environmental changes are affecting the reproductive success of several small pelagics, including European anchovy (Engraulis encrasicolus), which is a major target of Adriatic mid-water trawl and purse-seine fisheries. In this study, we evaluated short-term genetic changes of the species in the Adriatic Sea by applying molecular markers in samples of three generations of European anchovy. Thirteen polymorphic microsatellite markers and a mitochondrial gene were used in cohorts of adults and larvae, collected at three sites in the north-east, central, and southern Adriatic Sea in 2015. Furthermore, temporal, and spatial genetic variation was assessed by comparing the above dataset with a sample of adult anchovy collected in 2012 at three sites close to those sampled in 2015. Expected heterozygosity was higher in adults than in larvae, suggesting a loss of genetic diversity and uneven reproduction. In addition, a comparison of the two datasets demonstrated a change in the anchovy population structure from 2012 to 2015. In the reproductive event of 2015, this change led the two main genetic stocks described in the Adriatic Sea to merge into one. We suggest that the population structure of European anchovy in the north-eastern Adriatic may be influenced by changes in environmental parameters and by periodic alterations in the temporal pattern of population connectivity.

Acanthodactylus lineomaculatus is now regarded as an ecotype of A. erythrurus with which it has been recently synonymized. Despite the wide range of A. erythrurus, karyological data for this species are scarce and limited to classical cytogenetic studies carried out in individuals from only 2 locations (central Spain and Spanish enclave of Melilla on the northwestern Mediterranean Moroccan coast). Here, for the first time, we cytogenetically characterized individuals of A. lineomaculatus from the southwestern Moroccan Atlantic coast with the aim to increase the karyological knowledge of this wide-ranging species and to assess if any chromosomal changes can be found in this ecotype in comparison to other populations of this species. The diploid number of the individuals investigated is 2n = 38 which is typical of most lacertids. Active NORs were located telomerically in a medium-small pair of chromosomes, and no inactive NORs were detected. C-banding revealed an intensely heterochromatic W chromosome composed of AT-rich (centromere and long arm telomeric region) and GC-rich (most of the long arm) regions, with extended interstitial telomeric sequences. These telomere-like repeats occupy the GC-rich heterochromatin of the W. The DNA composition of the W represents a trait distinguishing A. lineomaculatus (southwestern Morocco) from A. erythrurus from Spain that possess a DAPI-positive (AT-rich) W chromosome. In conclusion, these results add further evidence to the remarkable karyotype conservation in lacertid lizards, although differences in NOR location and in W chromosome structure among populations could suggest an incipient speciation mediated by chromosome changes in this wide-ranging lizard species.

Canine vector-borne diseases (CVBD) are an important and emerging health concern for humans and animals worldwide. The purpose of the presented study was to assess, from 2016 to 2019, the seroprevalence of CVBD agents and clarify the epidemiology of tick-borne disease in stray dogs living in the Campania Region, Southern Italy. For this purpose, blood samples were collected from January 2016 to December 2019 from 1023 dogs in authorized kennels located in the five municipalities of the Campania Region. SNAP&reg, 4DX&reg, from IDEXX&reg, Laboratories was used for detection of Ehrlichia spp., Anaplasma spp., Borrelia burgdorferi sensu lato (s.l.), and Dirofilaria immitis antibodies. The overall seroprevalence of CVBD in stray dogs was 19.6% (95% Confidence Intervals (CI): 17.2&ndash, 22.8%, 201/1023). The most common pathogen was Ehrlichia spp., with a percentage of positivity of 16.03%, followed by Anaplasma spp. with 7.8%. B. burgdorferi s.l. and D. immitis were detected in only 0.2% of dogs, co-infection was detected in 4.5% of stray dogs tested. No link was detected between the gender, age, location, and CVBD seropositivity, except for Ehrlichia spp. for which location (Avellino Province, p = 0.007) and gender (male, p = 0.002) were risk factors for seropositivity. Our results demonstrated that animals are exposed to at least one of the four etiological agents (Ehrlichia spp., Anaplasma spp. Borrelia burgdorferi s.l., and Dirofilaria immitis) transmitted by vectors. Finally, this study highlighted the utility of serological monitoring in stray dogs, housed in kennels, given the threat posed by CVBD to animals and the zoonotic implications of these etiological agents and their vectors on human health.

The Mediterranean brown trout (Salmo trutta complex) is one of the freshwater fish species complex at greater risk of extinction in the Mediterranean area. The introduction of alien invasive species and their interaction with the native fauna represent some of the major threats to the survival of this species. Currently, the genetic variability of the Mediterranean trout (Salmo trutta complex) is being compromised by the introgressive hybridization with the Atlantic trout (Salmo trutta Linnaeus, 1758). Therefore, it is necessary to gain further knowledge on genetic and demographic characteristics of Mediterranean trout populations, that, combined with environmental and angling data, will allow to undertake sound conservation strategies. The aims of the present study were to analyze: i) the demographic characteristics of the Mediterranean trout in seven central Apennine river basins where native populations are threatened by stocking with the domestic trout of north Atlantic origin; ii) the influence of both environmental parameters and different fishery management strategies on the status of native trout populations. The project focuses on 14 sites included in the Natura 2000 EU wide network of nature protection areas, established under the 1992 Habitat Directive. A total of 25 watercourses were investigated for a total of 32 sampling sites. A census of the fish fauna was carried out by electrofishing at each sampling location. Fish and environmental data were collected in spring and autumn 2014. The results obtained in the present research allowed us to detect the presence of three residual Mediterranean trout populations with a high degree of genetic integrity in the study area. The native trout populations were characterized by higher abundances and higher adult density values. Well-structured native populations with higher legal-size specimens density and poor body conditions in terms of relative weight were observed in the no-fishing areas, probably due to the presence of intraspecific competition phenomena. The environmental analysis confirmed the key role played by the geological characteristics of the central Apennines in shaping the introgression levels observed in the wild trout populations, while the recovery of the water quality appears decisive in the development of management strategies aimed at their conservation.

A microbiological survey of 73 pellets collected from different birds of prey species housed at the Wildlife Rescue and Rehabilitation Center of Napoli (southern Italy) was performed. Pellets were analyzed by culture and biochemical methods as well as by serotyping and polymerase chain reaction. We isolated a wide range of bacteria some of them also pathogens for humans (i.e. Salmonella enterica serotype Typhimurium, Campylobacter coli, Escherichia coli O serogroups). This study highlights the potential role of birds of prey as asymptomatic carriers of pathogenic bacteria which could be disseminated in the environment not only through the birds of prey feces but also through their pellets.

The DNA composition of the W represents a trait distinguishing A. lineomaculatus (southwestern Morocco) from A. erythrurus from Spain that possess a DAPI-positive (AT-rich) W chromosome. In conclusion, these results add further evidence to the remarkable karyotype conservation in lacertid lizards, although differences in NOR location and in W chromosome structure among populations could suggest an incipient speciation mediated by chromosome changes in this wide-ranging lizard species.

Anchovies represent the largest world’s marine fish catches and the current threats on their populations impose a sustainable exploitment based on sound scientific information. In the European anchovy (Engraulis encrasicolus), the existence of several populations has been proposed but a global view is missing. Using a multidisciplinary approach, here we assessed the divergence among different ecotypes and its possible causes. SNPs have revealed two functionally distinct ecotypes overlapping in the Central Mediterranean, with one ecotype confined near the river estuaries. The same SNPs outliers also segregated two distinct populations in the near Atlantic, despite their large spatial distance. In addition, while most studies suggested that adaptation to low salinity is key to divergence, here we show that the offshore ecotype has higher environmental tolerance and an opportunistic feeding behaviour, as assessed by the study of environmental conditions, anchovy diet and trophic levels, and passive egg dispersal. These results provide insights into the anchovy evolutionary history, stressing the importance of behaviour in shaping ecotypes.

In this study, IMO-TaqI satDNA, previously isolated in several species of Lacertidae, was isolated and characterized from four species of the genus Lacerta and three of the genus Timon. The aim was to gain further insights into the evolutionary dynamics of this satDNA, its occurrence among lacertids and to understand if it plays any role in sex chromosome evolution in these seven species. The results here obtained highlighted the presence of this repetitive element in the genome of all the species investigated, thus indicating that IMO-TaqI satDNA is evolutionary conserved among a wide variety of lacertids. In addition, this element was found to be very abundant in the constitutive heterochromatin of the W-sex chromosome of the four Lacerta species investigated. The occurrence of IMO-TaqI satDNA on Lacerta heterochromosome suggests that it is involved in the differentiation of the W chromosome by heterochromatinization, and the fact that it is absent in the W of other lacertids investigated seems to confirm that repetitive DNA sequences would remain randomly trapped into the sex chromosomes, undergoing amplification as a consequence of the suppression of recombination.

In the present study the genetic variability of European sardine from Adriatic and Ionian seas was investigated in order to detect the occurrence of genetic structure within and between these basins. In several samples the analysis of genetic variability at eight microsatellite loci showed a number of homozygote individuals higher than expected at Hardy-Weinberg equilibrium. The inter-population differentiation level estimated by AMOVA, qST and r R ST and Bayesian descriptors detected no signs of population differentiation between the samples analysed. These results are consistent with previous studies based on allozymes and several mitochondrial DNA markers and add further evidence contradicting the early identification, based on morphological and reproductive data, of two sub-populations in the Adriatic Sea.

Satellite DNAs represent a large portion of all high eukaryotic genomes. They consist of numerous very similar repeated sequences, tandemly arranged in large clusters up to 100 million base pairs in length, usually located in the heterochromatic parts of chromosomes. The biological significance of satDNAs is still under discussion, but most of their proposed functions are related to heterochromatin and/or centromere formation and function. Because information about the structure of reptilian satDNA is far from exhaustive, we present a molecular and cytogenetic characterization of two satDNA families in four lacertid species. Two families of tandemly repeated DNAs, namely TaqI and HindIII satDNAs, have been cloned and sequenced from four species belonging to the genus Iberolacerta. These satDNAs are characterized by a monomer length of 171–188 and 170–172 bp, and by an AT content of 60.5% and 58.1%, respectively. FISH experiments with TaqI satDNA probe produced bright signals in pericentromeric regions of a subset of chromosomes whereas all the centromeres were marked by HindIII probe. The results obtained in this study suggest that chromosome location and abundance of satDNAs influence the evolution of these elements, with centromeric families evolving tenfold faster than interstitial/pericentromeric ones. L’intervento si pone l’obiettivo di analizzare lo scritto autobiografico del pittore Saverio Altamura (1896). Tra la fine dell’Ottocento e l’inizio del Novecento numerosi artisti, napoletani di nascita o d’adozione (Altamura era nato a Foggia, ma si era formato nella città partenopea), elaborano una serie di testi autobiografici, i quali risultano importanti per poter ricostruire le vicende culturali italiane all’altezza del XIX secolo (tra questi artisti ricordiamo, tra gli altri, Bernardo Celentano, Gioacchino Toma, Michele Cammarano, Domenico Morelli, Edoardo Dalbono). Nell’opera di Altamura, in particolar modo, gli interessi artistici si sovrappongono a quelli politici in un miscuglio nel quale grande importanza è affidata anche al tema amoroso. Trasporre sul piano letterario vicende avvenute decenni prima rispetto al tempo della scrittura significa, però, anche ripensare in maniera critica a quelle vicende. Nell’ultimo capitolo dell’autobiografia Altamura torna con la mente alla rivolta napoletana del 15 maggio 1848, alla quale egli aveva partecipato. Un dubbio lacera la coscienza dell’anziano artista. È il dubbio di aver generato la morte di due contadini che cercavano di abbattere la barricata di Largo Carità. Una macchia cruenta, così, finisce per imbrattare la tela del racconto risorgimentale disegnata con la penna dal pittore meridionale.

Squamate reptiles show a striking diversity in modes of sex determination, including both genetic (XY or ZW) and temperature-dependent sex determination systems. The genomes of only a handful of species have been sequenced, analyzed and assembled including the genome of Anolis carolinensis. Despite a high genome coverage, only macrochromosomes of A. carolinensis were assembled whereas the content of most microchromosomes remained unclear. Most of the Anolis species have homomorphic XY sex chromosome system. However, some species have large heteromorphic XY chromosomes (e.g., A. sagrei) and even multiple sex chromosomes systems (e.g. A. pogus), that were shown to be derived from fusions of the ancestral XY with microautosomes. We applied next generation sequencing of flow sorting-derived chromosome-specific DNA pools to characterize the content and composition of microchromosomes in A. carolinensis and A. sagrei. Comparative analysis of sequenced chromosome-specific DNA pools revealed that the A. sagrei XY sex chromosomes contain regions homologous to several microautosomes of A. carolinensis. We suggest that the sex chromosomes of A. sagrei are derived by fusions of the ancestral sex chromosome with three microautosomes and subsequent loss of some genetic content on the Y chromosome.

We report a case of polyostotic chondroblastic osteosarcoma in a kestrel ( Falco tinnunculus ) admitted to the Wildlife Rehabilitation and Rescue Center (Naples, Italy). A consolidated fracture of the left tibiotarsus bone and a deviation of the limb were evident. After radiographic, cytologic, and histopathologic examinations, a diagnosis of polyostotic chondroblastic osteosarcoma was made. To our knowledge, this is the first report on polyostotic chondroblastic osteosarcoma in a kestrel.

The divergence of lineages leading to extant squamate reptiles (lizards, snakes, and amphisbaenians) and birds occurred about 275 million years ago. Birds, unlike squamates, have karyotypes that are typified by the presence of a number of very small chromosomes. Hence, a number of chromosome rearrangements might be expected between bird and squamate genomes. We used chromosome-specific DNA from flow-sorted chicken (Gallus gallus) Z sex chromosomes as a probe in cross-species hybridization to metaphase spreads of 28 species from 17 families representing most main squamate lineages and single species of crocodiles and turtles. In all but one case, the Z chromosome was conserved intact despite very ancient divergence of sauropsid lineages. Furthermore, the probe painted an autosomal region in seven species from our sample with characterized sex chromosomes, and this provides evidence against an ancestral avian-like system of sex determination in Squamata. The avian Z chromosome synteny is, therefore, conserved albeit it is not a sex chromosome in these squamate species.

Karyotype and other chromosomal characteristics in the Adriatic brook lamprey Lampetra zanandreai, representative of one of the most ancestral group of vertebrates, were examined using conventional (Ag-staining, C-banding as well as CMA(3) and DAPI fluorescence) and molecular (FISH with 18/28S rDNA and EcoRI satDNA as probes) protocols with metaphase chromosomes derived from whole blood cultures. The chromosome complement had a modal diploid chromosome number of 2n = 164, as in other petromyzontid lamprey species. Ag-staining and CMA(3) fluorescence, as well as FISH with 18/28S rDNA probes, detected nucleolar organizer regions (NORs) close to the centromeres of the biarmed chromosomes of pairs 1 and 2, the largest chromosome pairs of the complement. In addition to NORs, CMA(3) fluorescence revealed positive signals in approximately 40 other chromosomes. DAPI stained mostly centromeric regions of many chromosomes as well as conspicuously massive blocks overlapping NOR sites. C-banding evidenced a large amount of constitutive heterochromatin in somatic chromosomes, with approximately 40 C-positive acrocentric elements completely heterochromatic, corresponding with the 40 CMA(3)+ chromosomes and positive heterochromatic blocks in pericentromeric regions of chromosome pairs 1 and 2. Polymerase chain reaction (PCR)-based cloning of satellite DNA with primers derived from Petromyzon marinus centromeric sequences was successful for L. zanandreai genomic DNA. The sequence was AT-rich (59%) and characterized by short consensus motifs similar to other centromeric satellite motifs. FISH using satDNA clones as a probe produced a fluorescent signal on a single pair of small chromosomes. This sequence was PCR-amplified also in L. planeri and P. marinus genomic DNA, and the evolution of this repetitive element in the above species was analysed.

The genetic variability of the wall lizard, Podarcis muralis, was analysed in that part of its range regarded as its area of origin and also its expansion centre. The analysis was performed on partial cytochrome b sequences (405 bp) of 117 specimens representing 31 sampling locations from southern, central and northern Italy. The results of this study revealed 30 haplotypes in the Italian sample belonging to three main genetic lineages. The genetic divergence among haplotypes ranged from 0.2 to 6.6%, whereas the estimation of divergence times among mtDNA lineages highlighted a complex pattern of genetic differentiation with both Pleistocene and Pliocene divergences (3.2–0.7 Ma). These results put in evidence the role played by the pre-Pleistocene and Pleistocene palaeogeographic and palaeoclimatic events in shaping the genetic diversity of the wall lizard in Italy and confirm the role of this peninsula as glacial refuge. Identification of potential spawning and nursery areas of European anchovy (Engraulis encrasicolus) represents an essential step in the management of a resource which is of fundamental importance both for fishery and pelagic trophic web. Egg and larvae occurrence from ichthyoplankton surveys (2012 - 2015) in the South Western Adriatic Sea were examined to understand the mechanisms that control their distribution. Egg and larvae densities varied through the years with the highest values recorded in 2012 and the lowest in 2014. Positive correlations between eggs and larvae with zooplankton were observed. When quotient analysis was used to find relations with environmental and biological variables, the results pointed out an egg preference for depth between 91 and 120 m and an avoidance between 11 and 30 m. Moreover, egg avoidances for high values of chlorophyll-a (> 0.52 mg m-3) and low values of zooplankton biomass (< 199 mg m-2) were highlighted. On the other hand, different ranges of habitat preferences and avoidances were observed for larvae: preference range of depth between 11 and 60 m and avoidance > 151 m; preference for high zooplankton biomass (> 1000 mg m-2) and avoidance for low biomass (< 299 mg m-2). These correlations and the quotient values suggest that egg and larvae distribution in the South-Western Adriatic Sea is mainly driven by food availability and depth.

This study deals with the gonadal development and breeding season of a brown trout population in the river Metauro (Central Italy), a typical Apennine river with a short course and a highly unpredictable water flow. The results obtained showed that most individuals spawn between November and January, with a peak in December. This trend is similar to that observed in populations from Central Europe, but differs considerably from other populations at similar latitudes, which show a spawning peak between February and March. As brown trout in the river Metauro are heavily stocked with farmed strains of Atlantic origin, it is likely that in this population the reproductive characteristics have changed according to the pattern of North European populations. On the other hand, the unusual presence of mature trout in June could be the result of stocking with farmed individuals whose reproductive characteristics have been modified by breeders.

In the Mediterranean area, one of the major threats to freshwater fish fauna is represented by the introduction of alien species. The bullhead, Cottus gobio, is a species of great conservation interest threatened by the massive introduction of hatchery-reared brown trout, Salmo trutta, for angling purposes. The aims of this research were: i) to present a case-study of interspecific and intraspecific competition in dwelling fish from two Mediterranean streams (central Italy); ii) to evaluate the bullhead population status before and after the alien brown trout removal, and iii) to investigate the influence of the environmental parameters on the abundance of both species. For the bullhead populations the abundances and the young-of-the-year percentage significantly increased after the removal activities. The growth analysis showed that, after the trout removal, bullhead were distinguished by greater performance and larger sizes in all age classes in comparison to the pre-removal period, while the condition factor (Kn) showed no differences, suggesting that the effects of trout population control acted mainly through predator-prey dynamics. An inverse correlation between the abundance of bullhead and its body condition suggested that the beneficial effects due to the reduction of predation could be compensated by the intraspecific competition due to the increase of the bullhead population density.

The North Adriatic Sea is considered a critical feeding and developmental area for Mediterranean loggerhead turtles. In this study, a comparative analysis of mitochondrial DNA control region sequences was carried out on sixty-five loggerhead individuals stranded and caught as bycatch in the Adriatic Sea from 1999 to 2002. We demonstrated the existence of genetic relationships between the North-Central Adriatic (NCA) aggregate and the Central-Eastern Mediterranean rookeries. Short-range and long-range migrations towards the Adriatic Sea were hypothesized by the finding of individuals bearing haplotypes endemic to the nesting populations of Greece and Turkey. The occurrence of individuals belonging to dimensional classes from juveniles to mature adults strongly support both the feeding and developmental role of the NCA.

Digit identity in the avian wing is a classical example of conflicting anatomical and embryological evidence regarding digit homology. Anatomical in conjunction with phylogenetic evidence supports the hypothesis that the three remaining digits in the bird wing are digits 1, 2, and 3. At the same time, various lines of embryological evidence support the notion that these digits develop in positions that normally produce digits 2, 3, and 4. In recent years, gene expression as well as experimental evidence was published that supports the hypothesis that this discrepancy arose from a digit identity shift in the evolution of the bird wing. A similar but less well-known controversy has been ongoing since the late 19th century regarding the identity of the digits of the three-toed Italian skink, Chalcides chalcides. Comparative anatomy identifies these digits as 1, 2, and 3, while embryological evidence suggests their derivation from embryological positions 2, 3, and 4. Here we re-examine this evidence and add gene expression data to determine the identity of the three digits of C. chalcides. The data confirm that the adult and the embryological evidence for digit identity are in conflict, and the expression of Hoxd11 suggests that digits 1, 2, and 3 develop in positions 2, 3, and 4. We conclude that in C. chalcides, and likely in its close relatives, a digit identity frame shift has occurred, similar to the one in avian evolution. This result suggests that changes in of digit identity might be a more frequent consequence of digit reduction than previously assumed.

The pattern of genetic variability of two species of Mediterranean gobiids was compared, with reference to their different life history traits (Aphia minuta paedomorphic and pelagic; Gobiusniger metamorphosed and benthic). The aim was to evaluate how different life histories can affect the genetic structure in these marine teleosts. The study was carried out on populations of both species sampled in the western Mediterranean and in the Adriatic Sea. Seven restriction endonucleases were used for the RFLP analysis of a mitochondrial DNA segment comprising the NADH dehydrogenase subunits 3, 4L and 4. The results highlighted two different patterns of genetic variation, a weak genetic structure in A. minuta and population subdivision in G. niger. These observations may be explained not only in terms of the different dispersal capabilities of these species, but also considering that A. minuta is an abbreviate iteroparous spawner while G. niger is a protracted iteroparous spawner. Because abbreviate iteroparity is a reproductive strategy selected in stable environments with high resource availability, Pliocene and Pleistocene climate oscillations may have represented factors that negatively influenced the reproductive success of A. minuta, producing demographic fluctuations and bottlenecks, as suggested by the mismatch distribution analysis. The weak genetic structure of A. minuta populations seems to be therefore due to a more recent re-colonization of the Mediterranean basin after a severe population decline, rather than to the high vagility of this pelagic goby.

A hitherto undescribed satellite DNA family (AvaII satDNA) has been isolated and characterised in Eumeces schneideri, a squamate reptile belonging to the family Scincidae. AvaII satDNA is characterised by a monomer length of 208 bp, a GC content of 59% and exhibits a certain degree of CpG methylation. FISH experiments with AvaII satDNA probe produced bright signals (i) at the end of the short arms of all subtelocentric chromosomes except for pair 14, in which the signal was at the end of the long arms, (ii) at the ends of both arms of the small metacentric chromosomes 12, and (iii) in a terminal position on the acrocentric chromosomes 11 and 13. AvaII satDNA repeats were not found in the metacentric pair 3, whereas only a weak interstitial signal occurred in the metacentric pairs 1 and 2. C-banding showed that this satellite represents most of the constitutive heterochromatin in the genome of this skink, and chromomycin A3 staining produced a clear signal overlapping with the satellite, except for NOR-associated heterochromatin. In addition, quantitative dot blot analysis showed that these repetitive sequences constitute about 3% of the genomic DNA of this lizard. AvaII satDNA sequence analysis revealed the occurrence of short guanine residue stretches for which a function in structural stability of these sequences and a role in recombination with telomeric sequences can be hypothesised. Fibre FISH experiments showed that on some chromatin fibres telomeric sequences and AvaII satellite DNA repeats are intermingled or overlapping.

A cytogenetic analysis was carried out using conventional staining, banding techniques and fluorescence in situ hybridization (FISH) in Italian populations of brown trout (Salmo truttacomplex). All individuals analysed, belonging to the Atlantic (At), Marmoratus (Ma), Adriatic (Ad) and Mediterranean (Me) lineages, showed remarkable karyotype uniformity, with diploid complement of 2n = 80 chromosomes, arm number (NF) of 102 and invariable karyotype composition. Such uniformity was also observed with respect to the location of 5S rDNA and the active, i.e. silver-positive NOR sites. On the contrary, FISH with 28S ribosomal probe and fluorescent staining with CMA3 revealed that inactive NOR sites are more numerous in Ad and Me than in At and Ma lineages. A centromeric sequence was successfully isolated from Salmo trutta individuals by polymerase chain reaction (PCR)-based cloning, using primers designed from published Atlantic salmon (Salmo salar) satellite DNA sequences. This sequence had high AT content (65.3%) and short consensus motif (A/T)(G/C)AAA(T/C) similar to other centromeric satellite repeats. The isolated satellite DNA clones were localized with FISH in the centromeric regions of the brown trout chromosomes, showing lineage-specific patterns. Because it is well known that AT-rich sequences can induce a pronounced DNA curvature, which in turn would promote faster and higher chromatin spiralization, it may be hypothesised that the wide distribution of this satellite in the S. trutta genome may have played a role in its karyotype stability. The presence of this sequence in other salmonid species was also tested by Southern blot hybridization and used to analyze its evolution within salmonids.

Skinks represent the most diversified squamate reptiles with a great variation in body size and form, and are found worldwide in a variety of habitats. Their remarkable diversification has been accompanied by only a few chromosome rearrangements, resulting in highly-conservative chromosomal complements of these lizards. In this study cross-species chromosome painting using Scincus scincus (2n = 32) as the source genome, was used to detect the chromosomal rearrangements and homologies between the following skinks: Chalcides chalcides (2n = 28), C. ocellatus (2n = 28), Eumeces schneideri (2n = 32), Lepidothyris fernandi (2n = 30), Mabuya quinquetaeniata (2n = 32). The results of this study confirmed a high degree of chromosome conservation between these species. The main rearrangements in the studied skinks involve chromosomes 3, 5, 6 and 7 of S. scincus. These subtelocentric chromosomes are homologous to the p and q arms of metacentric pair 3 and 4 in C. chalcides, C. ocellatus, L. fernandi, and M. quinquetaeniata, while they are entirely conserved in E. schneideri. Other rearrangements involve S. scincus 11 in L. fernandi and M. quinquetaeniata, supporting the monophyly of Lygosominae, and one of the chromosomes S. scincus 12–16, in M. quinquetaeniata. In conclusion, our data support the monophyly of Scincidae and confirm that Scincus-Eumeces plus Chalcides do not form a monophyletic clade, suggesting that the Scincus-Eumeces clade is basal to other members of this family. This study represents the first time the whole genome of any reptile species has been used for cross-species chromosome painting to assess chromosomal evolution in this group of vertebrates.

Trematomus eulepidotus and T. loennbergii are two of the most common epibenthic fish in the waters of the High Antarctic continental shelf. Since the reproductive biology of these species has not been studied in the Ross Sea, we provide a macroscopic and histological analysis of the reproductive effort and gonadal development in both sexes. Most samples were collected during benthic trawl surveys in the south-western Ross Sea in the 1996 and 1997 summer seasons. The aim of the study was to define the reproductive characteristics of these two sympatric species and to examine the hypothesis that different reproductive strategies mitigate interspecific competition. We found that, in common with most Antarctic notothenioids, both species possess a suite of similar reproductive strategies including delayed sexual maturity, prolonged gametogenesis, group-synchronous oocyte maturation, a single spawning event per year and iteroparity. Both species show a comparable reproductive effort in terms of potential fecundity with between 2000 and 20 000 eggs per female per season. Nevertheless, the two species exhibited a considerable difference in the timing of the breeding season, spawning in summer (T. eulepidotus) and in autumn (T. loennbergii). This gives rise to a mismatch in the time of appearance of larvae in the environment and probably leads to reduced competition.

E possibile, in questo senso, soffermarsi in maniera specifica su alcune comparazioni'. Nella biografia di Ferrante Gonzaga, scritta da Alfonso de' UUoa (1563), si accosta, ad esempio, la vicenda di Scipione a quella del capitano cinquecentesco, mentre nella vita di Cornelio Scipione l'Emiliano, scritta da Antonio Bendinelli (2 ed., 1568), si associano le gesta del 'maggiore' Africano con quelle dell'omonimo 'minore'. Il discorso, in realtà, finisce per coinvolgere diversi letterati (da Bernardo Segni e Antonio Benivieni il Giovane a Scipione Ammirato e Scipione de' Monti), nelle cui opere l'Africano diviene sicuramente un continuo, e nobilitante, termine di paragone.

We analysed histological characteristics of gonads and reproductive effort of the small deep-living dragonfishes Akarotaxis nudiceps (Waite) and Bathydraco marri Norman collected in the south-western Ross Sea. From a macroscopic point of view, most specimens of B. marri were juveniles in early stages of gonad maturity, except for a maturing female. Conversely, the sample of A. nudiceps was composed of both immature and adult fish in different stages of maturity. A single A. nudiceps female was mature with a gonadosomatic index of 9.8%. Its absolute and relative fecundity was 260 oocytes and 31.5 oocytes g−1 TW, respectively, with a mean size of ripe oocytes of 1.9 mm. Gametogenesis in both species closely resembled that observed in other notothenioids, with females possessing two well-defined groups of oocytes. One group consisted of previtellogenic oocytes as a reserve stock while the other group was maturing oocytes to be ovulated in the current spawning season. A distinctive feature of oogenesis in recovering and maturing females of A. nudiceps was the presence of both postovulatory follicles in different stages of reabsorption and atretic oocytes. Based on low absolute fecundity, it is possible that A. nudiceps provides parental care and egg guarding.

Genetic diversity was analyzed in Chalcides chalcides populations from peninsular Italy, Sardinia, Sicily and Tunisia by sequencing 400 bp at the 5' end of the mitochondrial gene encoding cytochrome b (cyt b) and by restriction fragment length polymorphism (RFLP) analysis of two mitochondrial DNA segments (ND-1/2 and ND-3/4). The results of the phylogenetic analysis highlighted the presence of three main clades corresponding with three of the four main geographical areas (Tunisia, Sicily and the Italian peninsula), while Sardinia proved to be closely related to Tunisian haplotypes suggesting a colonization of this island from North Africa by human agency in historical times. On the contrary, the splitting times estimated on the basis of cyt b sequence data seem to indicate a more ancient colonization of Sicily and the Italian Peninsula, as a consequence of tectonic and climatic events that affected the Mediterranean Basin during the Pleistocene. Finally, the analysis of the genetic variability of C. chalcides populations showed a remarkable genetic homogeneity in Italian populations when compared to the Tunisian ones. This condition could be explained by a rapid post-glacial expansion from refugial populations that implied serial bottlenecking with progressive loss of haplotypes, resulting in a low genetic diversity in the populations inhabiting the more recently colonized areas.

Genetic diversity was analysed in Salmo trutta populations living in an area of central Italy by sequencing 310 bp of the 5′ end of the mtDNA control region (D‐loop) and by Restriction Fragment Length Polymorphism (RFLP) analysis of three mtDNA segments. Data show a genetic structure profoundly altered by stockings with allochthonous material of Atlantic origin. In fact, 15 of the RFLP haplotypes detected are linked to an Atlantic sequence. The remaining 9 were instead coupled with sequences representing the three major phylogenetic lineages previously identified in the Mediterranean basin (Adriatic, marmoratus, and Mediterranean), representing the native genetic diversity of brown trout in that area. The close genetic affinity observed between these latter haplotypes and those found in the Balkan peninsula by other authors seems to be in accordance with a recent, natural, history of dispersion between the two borders of the Adriatic Sea. The Antarctic plunderfishes Artedidraco lönnbergi and A. skottsbergi are small, bottom dwelling species inhabiting the continental shelf of the High Antarctic Zone. During cruise 97–9 of the US RV Nathaniel Palmer during the summer in the south-western Ross Sea, samples of both species were collected by means of bottom trawling. On the basis of macroscopic and histological analysis, we present the first data on the reproductive characteristics of these two plunderfishes, including gametogenesis, spawning period and absolute fecundity. Histologically, we found immature (stage I and II) and mature (stage V) females in both species, whereas developing females (stage III) were found only in A. skottsbergi. All examined male specimens of A. skottsbergi were in the final stage of spermatogenesis (stage III), whereas male A. lönnbergi were immature (stage I), mature (stage IV) and post-reproductive (stage V) individuals. In both species, spawning takes place in summer during December and January. Absolute fecundity was very low, with less than 100 and 200 oocytes in A. lönnbergi and A. skottsbergi, respectively. These data are compared with those reported in literature for other artedidraconids.
