Abstract

IgG4-related disease (IgG4-RD) can cause fibroinflammatory lesions in nearly any organ. Correlation among clinical, serological, radiological and pathological data is required for diagnosis. This work was undertaken to develop and validate an international set of classification criteria for IgG4-RD. An international multispecialty group of 86 physicians was assembled by the American College of Rheumatology (ACR) and the European League Against Rheumatism (EULAR). Investigators used consensus exercises; existing literature; derivation and validation cohorts of 1879 subjects (1086 cases, 793 mimickers); and multicriterion decision analysis to identify, weight and test potential classification criteria. Two independent validation cohorts were included. A three-step classification process was developed. First, it must be demonstrated that a potential IgG4-RD case has involvement of at least one of 11 possible organs in a manner consistent with IgG4-RD. Second, exclusion criteria consisting of a total of 32 clinical, serological, radiological and pathological items must be applied; the presence of any of these criteria eliminates the patient from IgG4-RD classification. Third, eight weighted inclusion criteria domains, addressing clinical findings, serological results, radiological assessments and pathological interpretations, are applied. In the first validation cohort, a threshold of 20 points had a specificity of 99.2% (95% CI 97.2% to 99.8%) and a sensitivity of 85.5% (95% CI 81.9% to 88.5%). In the second, the specificity was 97.8% (95% CI 93.7% to 99.2%) and the sensitivity was 82.0% (95% CI 77.0% to 86.1%). The criteria were shown to have robust test characteristics over a wide range of thresholds. ACR/EULAR classification criteria for IgG4-RD have been developed and validated in a large cohort of patients. These criteria demonstrate excellent test performance and should contribute substantially to future clinical, epidemiological and basic science investigations.

Abstract

The KamLAND-Zen 800 experiment is searching for the neutrinoless double-beta decay of 136Xe by using 136Xe-loaded liquid scintillator. The liquid scintillator is enclosed inside a balloon made of thin, transparent, low-radioactivity film that we call Inner Balloon (IB). The IB, apart from guaranteeing the liquid containment, also allows to minimize the background from cosmogenic muon-spallation products and 8B solar neutrinos. Indeed these events could contribute to the total counts in the region of interest around the Q-value of the double-beta decay of 136Xe. In this paper, we present an overview of the IB and describe the various steps of its commissioning minimizing the radioactive contaminations, from the material selection, to the fabrication of the balloon and its installation inside the KamLAND detector. Finally, we show the impact of the IB on the KamLAND background as measured by the KamLAND detector itself.

Abstract

Histone H3 lysine‐9 di‐methylation (H3K9me2) and lysine‐27 tri‐methylation (H3K27me3) are linked to repression of gene expression, but the functions of repressive histone methylation dynamics during inflammatory responses remain enigmatic. Here, we report that lysine demethylases 7A (KDM7A) and 6A (UTX) play crucial roles in tumor necrosis factor (TNF)‐α signaling in endothelial cells (ECs), where they are regulated by a novel TNF‐α‐responsive microRNA, miR‐3679‐5p. TNF‐α rapidly induces co‐occupancy of KDM7A and UTX at nuclear factor kappa‐B (NF‐κB)‐associated elements in human ECs. KDM7A and UTX demethylate H3K9me2 and H3K27me3, respectively, and are both required for activation of NF‐κB‐dependent inflammatory genes. Chromosome conformation capture‐based methods furthermore uncover increased interactions between TNF‐α‐induced super enhancers at NF‐κB‐relevant loci, coinciding with KDM7A and UTX recruitments. Simultaneous pharmacological inhibition of KDM7A and UTX significantly reduces leukocyte adhesion in mice, establishing the biological and potential translational relevance of this mechanism. Collectively, these findings suggest that rapid erasure of repressive histone marks by KDM7A and UTX is essential for NF‐κB‐dependent regulation of genes that control inflammatory responses of ECs.

Abstract

Global hydrological models (GHMs) have become an established tool to simulate water resources worldwide. Most of the GHMs are however uncalibrated and typically use a set of basic hydrological parameters, that could potentially lead to unrealistic projections of the terrestrial water cycle. The calibration of hydrological models is usually performed by using and comparing modeled to observed discharge. Accurate station data and reliable time series data of discharge are, however, often not available for many parts of the world and classic calibration approaches are therefore not feasible. In this paper, we aim to develop a new calibration approach that requires no additional data, is easy to implement, and substantially improves model performance, especially in regions where uncalibrated model performance is rather poor. This is achieved by using the Budyko framework, which provides a conceptual representation of the long‐term water and energy balance. We use a state‐of‐the‐art GHM and calibrate the model within nine river catchments of different sizes and characteristics. Since observed river discharge is available for these catchments, we are able to compare the Budyko‐based calibration approach to a classic discharge‐based calibration scheme and the uncalibrated model version. In all catchments, the Budyko‐based calibration approach decreases biases and increases model performance compared to the uncalibrated model version although performance improvements obtained through a classic calibration approach are greater. Nonetheless, a Budyko‐based calibration is a valuable, intermediate approach between use of a basic set of a priori hydrological parameters and classical calibration against discharge data.

Abstract

This project was funded as part of the National Geographic Society and Rolex partnership to support a Perpetual Planet. Mountains are the water towers of the world, supplying a substantial part of both natural and anthropogenic water demands1,2. They are highly sensitive and prone to climate change3,4, yet their importance and vulnerability have not been quantified at the global scale. Here, we present a global Water Tower Index, which ranks all water towers in terms of their water-supplying role and the downstream dependence of ecosystems and society. For each tower, we assess its vulnerability related to water stress, governance, hydropolitical tension and future climatic and socio-economic changes. We conclude that the most important water towers are also among the most vulnerable, and that climatic and socio-economic changes will affect them profoundly. This could negatively impact 1.9 billion people living in (0.3 billion) or directly downstream of (1.6 billion) mountain areas. Immediate action is required to safeguard the future of the world’s most important and vulnerable water towers. Postprint

Abstract

This study proposes a GAP/N2O direct-injection type gas-hybrid rocket system, aiming at a small satellite thruster with high-density specific thrust without gunpowder. This is a simple system that eliminates the pressurization mechanism of the oxidizing agent by using nitrous oxide with a high vapor pressure using the self-exothermic decomposition characteristics of GAP, which is a high-density and high-energy fuel, and achieves high-density specific thrust. Here, the result of the experiment in which an average thrust of 350 N was generated using the blowdown system of N2O with the future satellite installation in the visual field is shown.

Abstract

Water scarcity severely impairs food security and economic prosperity in many countries today. Expected future population changes will, in many countries as well as globally, increase the pressure on available water resources. On the supply side, renewable water resources will be affected by projected changes in precipitation patterns, temperature, and other climate variables. Here we use a large ensemble of global hydrological models (GHMs) forced by five global climate models and the latest greenhouse-gas concentration scenarios (Representative Concentration Pathways) to synthesize the current knowledge about climate change impacts on water resources. We show that climate change is likely to exacerbate regional and global water scarcity considerably. In particular, the ensemble average projects that a global warming of 2 °C above present (approximately 2.7 °C above preindustrial) will confront an additional approximate 15% of the global population with a severe decrease in water resources and will increase the number of people living under absolute water scarcity (3 per capita per year) by another 40% (according to some models, more than 100%) compared with the effect of population growth alone. For some indicators of moderate impacts, the steepest increase is seen between the present day and 2 °C, whereas indicators of very severe impacts increase unabated beyond 2 °C. At the same time, the study highlights large uncertainties associated with these estimates, with both global climate models and GHMs contributing to the spread. GHM uncertainty is particularly dominant in many regions affected by declining water resources, suggesting a high potential for improved water resource projections through hydrological model development.

Abstract

The supercritical water gasification (SCWG) of wet-biomass holds promise as a technology to convert biological waste (wet biomass) into valuable energy. The improvement of the equipment utilization efficiency of the SCWG facility on-site is important. Therefore, the 24 h continuous operation with the pilot plant (the treatment capacity is 1 t-wet/d) was conducted and it was successful with stable pressure loss of heat exchanger. However, SCWG plant might be operated by daily start and stop (DSS) on weekday daytime when shochu makers work on. The bad influence to tar characteristic was concerned about by the rise and drop of the temperature and the pressure with DSS. Therefore, the DSS gasification tests included 3 days holidays were conducted during one week. The heat exchanger tar plugging was concerned about; however the test results of the heat exchanger pressure loss indicated no tar plugging sign. The product gas elemental analysis results suggested the increase of gasification catalyst effects by gasification tests. Abstract

The expression of matrix metalloproteinases (MMPs) and their inhibitors (TIMPs) by human vascular smooth muscle cells (SMC) was monitored as a function of the phenotypic modulation in vitro. Cell phenotype was manipulated by varying serum concentration and cell density. Synthetic phenotype was characterized by a minimum expression of the contractile proteins and a maximal proliferation rate. Contractile phenotype was quiescent and expressed a maximal level of contractile proteins. Synthetic cells expressed the highest levels of both MMP-1 and TIMP-1 and displayed maximal collagenolytic activity. No significant change was detected in MMP-2 expression or catalytic activity. Enzyme immunoassays revealed that MMP-1 expression fell by 77+/-2.4-95+/-0.5%, and that of TIMP-1 by 34+/-0.5-59+/-1.9%, as the cells acquired a contractile phenotype. The level of the MMP-1/TIMP-1 complex was similarly reduced by 78+/-2.9-85+/-1.6%. These data demonstrate that the expression of MMP-1 and TIMP-1 are coordinately regulated with SMC phenotype.

Abstract

A 74-year-old woman with non-Marfan syndrome underwent aortic root replacement and mitral valve replacement simultaneously for annulo-aortic ectasia and mitral regurgitation. Postoperatively, she suffered from cholecystitis and underwent percutaneous transhepatic gull bladder drainage. After this intervention, her postoperative recovery was uneventful. The purpose of this study is to suggest the possibility of mitral regurgitation due to annular enlargement of the mitral valve as an outcome of deterioration of the annulo-aortic ectasia.

Abstract

There are many theories where a dark matter particle is part of a multiplet with an electrically charged state. If WIMP dark matter ($\chi^{0}$) is accompanied by a charged excited state ($\chi^{-}$) separated by a small mass difference, it can form a stable bound state with a nucleus. In supersymmetric models, the $\chi^{0}$ and the $\chi^{-}$ could be the neutralino and a charged slepton, such as the neutralino-stau degenerate model. The formation binding process is expected to result in an energy deposition of {\it O}(1--10 MeV), making it suitable for detection in large liquid scintillator detectors. We describe new constraints on the bound state formation with a xenon nucleus using the KamLAND-Zen 400 Phase-II dataset. In order to enlarge the searchable parameter space, all xenon isotopes in the detector were used. For a benchmark parameter set of $m_{\chi^{0}} = 100$ GeV and $\Delta m = 10$ MeV, this study sets the most stringent upper limits on the recombination cross section $\langle\sigma v\rangle$ and the decay-width of $\chi^{-}$ of $2.0 \times 10^{-31}$ ${\rm cm^3/s}$ and $1.1 \times 10^{-18}$ GeV, respectively (90\% confidence level).
