Objective: This study used network pharmacology and molecular docking technology to explore the molecular mechanism of Panax notoginseng in the treatment of disseminated intravascular coagulation. Methods: The main active components and targets of Panax notoginseng were screened by the TCMSP database, and DIC-related targets were obtained from the GeneCards database. PPI network was constructed by String and Cytoscape, GO gene analysis and KEGG pathway enrichment analysis were performed by DAVID, and molecular docking was performed by AutoDock software. Results: Eight active compounds and 51 potential therapeutic targets of Sanqi were screened. The key targets include VEGF, MAPK3, EGFR, STAT3 and so on. Beta-sitosterol, Stigmasterol, quercetin, DFV, and Diop were identified as potential candidate ingredients. There are 95 KEGG enrichment pathways. The metabolic pathways involving a large number of genes mainly include PI3K-Akt signaling pathway, Rap1 signaling pathway, VEGF signaling pathway and TNF signaling pathway. Conclusion: Eight active compounds and 51 potential therapeutic targets of Sanqi were screened. The key targets include VEGF, MAPK3, EGFR, STAT3 and so on. Normal ECG standards in newborns, infants, children and adolescents have been collected and published by many authors. Only those by Davignon et al., Rijinbeek et al. and our two studies covered all ages from birth to adolescence. The standards reflecting the growth and development of the heart in infants, children and adolescents remained to be studied and explored.We selected from our ECG database, after discussions and consultation, 15 key ECG parameters and analyzed for their age- and sex-specific mean, standard deviation and 2nd to 98th percentiles and their percentile charts were constructed.The ranges and distributions of the normal ECG standards, means and 2nd to 98th percentiles of 15 key parameters were established.A complete set of normal ECG standards of 15 key parameters from birth to adolescents is available for clinicians and researchers.

In this paper, we describe an innovative design for a multidisciplinary service-learning course on supply chain management in the industrial distribution program housed in the College of Engineering at a large research university in Texas. The purpose of this course was to increase students’ technical expertise and build a mindset and skills needed to tackle current societal issues, raising their social responsibility and enhancing civic-mindedness. The course combined curricular learning with community service, applying the content of academic disciplines to practice. It brought students from industrial distribution and sociology together to help food pantries to improve their operational efficiencies so that the food pantries can serve more people and work toward their goal. In this project, student groups identified the specific needs of community food pantries. They recommended solutions to improve operational efficiency so that they could use their technical skills to address the challenges and make an impact on the communities. In addition, student groups interacted with partner agencies via technologies that aided distance communication, which facilitated partnerships in remote locations. We also evaluated this project. The results reveal that this project had an impact on students’ personal and professional development as well as the community. The patients in the remazolam group received 0.3 μg/kg sufentanil, 0.3 mg/kg remazolam and 1.2 mg/kg remifentanil, whereas the patients in the propofol group received 0.3 μg/kg sufentanil, 2.0 mg/kg propofol and 1.2 mg/kg remifentanil for insertion of the LMA. The primary endpoint was the summed score of the insertion conditions. The secondary endpoints included hemodynamics, the duration of induction, the duration of insertion, tidal volume, plateau pressure and adverse events.Results: No difference was identified between the propofol group and remazolam group in the median summed score [18.0 (18.0, 18.0), 18.0 (17.0, 18.0), respectively, P > 0.05]. The induction duration was significantly longer (P < 0.05) in the remazolam group than propofol group. The cost of dopamine (P < 0.05) was significantly lower in the remazolam group compared with the patients in the propofol group, while the plateau pressure (P < 0.05) and the incidence of transient mild laryngospasm (P < 0.05) were significantly higher in the remazolam group. No differences were identified between the two groups in terms of heart rate, tidal volume, injection pain or hiccups (P > 0.05).Conclusion: Remazolam provided similar insertion conditions and better hemodynamic stability than propofol during LMA insertion without the use of muscle relaxant agents. However, a higher incidence of transient mild laryngospasm was found in the remazolam group, which should be considered.Keywords: remazolam, propofol, laryngeal mask airway, muscle relaxants

We study the relation between the Galois group $G$ of a linear difference-differential system and two classes $\mathcal{C}_1$ and $\mathcal{C}_2$ of groups that are the Galois groups of the specializations of the linear difference equation and the linear differential equation in this system respectively. We show that almost all groups in $\mathcal{C}_1\cup \mathcal{C}_2$ are algebraic subgroups of $G$, and there is a nonempty subset of $\mathcal{C}_1$ and a nonempty subset of $\mathcal{C}_2$ such that $G$ is the product of any pair of groups from these two subsets. These results have potential application to the computation of the Galois group of a linear difference-differential system. This study aimed to investigate the effects of high load fast and medium tempo back squats using a low number of repetitions on maximal strength and power output. Seventeen participants completed a countermovement jump test and 1-repetition maximum (1-RM) assessment before and after an eight-week intervention. All participants were randomly divided into a fast tempo (FAS: 1/0/1/0) and a medium tempo (MED: 2/0/2/0) resistance training (RT) group and performed three repetitions per set of a Smith back squat exercise with 85% 1-RM intensity. Maximal strength, jump height, peak power and force of the two groups were significantly improved (p < 0.05). In addition, peak velocity significantly increased after the intervention in the FAS group (p < 0.05), but not in the MED group (p > 0.05). A significant interaction effect between training groups was observed for jump height (F (1, 30) = 5.49, p = 0.026, η2 = 0.155). However, no significant group by time interaction effects were found between training groups for maximal strength (F (1, 30) = 0.11, p = 0.742, η2 = 0.004). Therefore, the two groups showed similar effects in maximal strength, but, compared with the MED group, FAS resistance training with low repetitions caused favorable adaptations in power output in trained men.

Although traditional quantum-confined nanostructures e.g. regular quantum wells or quantum dots have achieved huge success in the field of semiconductor lasers for past decades, these traditional nanostructures are encountering the difficulty of enhancing device performance to a higher level due to their inherent gain bottleneck. In this paper, we are proposing a new super-gain nanostructure based on self-assembled well-wire complex energy-band engineering with InGaAs-based materials to break through the existing bottleneck. The nanostructure is constructed by utilizing the special strain-driven indium (In)-segregation and the growth orientation-dependent on-GaAs multi-atomic step effects to achieve the distinguished ultra-wide and uniform super-gain spectra. Participants were assigned to receive Longyizhengqi granule or conventional treatment. The primary outcome was the time for nucleic acid to turn negative and the secondary outcomes are hospital stay and changes in cycle threshold (Ct) values for N gene and Orf gene. Multilevel random-intercept model was performed to analyze the effects of treatment.Results: A total of 3243 patients were included in this study (Longyizhengqi granule 667 patients; conventional treatment 2576 patients). Age (43.5 vs 42.1, p< 0.01) and vaccination doses (not vaccinated: 15.8% vs 21.7%, 1 dose: 3.5% vs 2.9%, 2 doses: 27.9% vs 25.6%, 3 doses: 52.8% vs 49.8%. p< 0.01) show statistical difference between Conventional treatment group and LYZQ granules group. The use of Longyizhengqi granule could significantly reduce the time for nucleic acid to turn negative (14.2 days vs 10.7 days, p< 0.01), shorten hospital stay (12.5 days vs 9.9 days, p< 0.01), and increase the changes in Ct value for N gene (8.44 vs 10.33, p< 0.01) and Orf gene (7.31 vs 8.44, p< 0.01) to approximately 1.5. Moreover, the difference in the changes of Ct values on the 4th, 6th, 8th, and 10th days seem to increase between two groups. No serious adverse events were reported.Conclusion: Longyizhengqi granule might be a promising drug for the treatment of mild COVID-19, and it might be beneficial to effectively shorten the negative transition time of nucleic acid, the total days of hospitalization, and increase the changes of Ct values. Long-term randomized controlled trials with follow-up evaluations are required to confirm its long-term efficacy.Keywords: COVID-19, Omicron variant, traditional Chinese medicine, prospective study

Highly repetitive sequences play a major structural and function role in the genome. In the present study, we developed Cas9-assisted cloning and SMRT sequencing of long repetitive sequences (CACS) to sequence and manipulate highly repetitive genes from eukaryotic genomes. CACS combined Cas9-mediated cleavage of a target segment from an intact genome, Gibson assembly cloning, and PacBio SMRT sequencing. Applying CACS, we directly cloned and sequenced the complete sequences of fibroin heavy chain (FibH) genes from 17 domesticated (Bombyx mori) and 7 wild (Bombyx mandarina) silkworms. Our analysis revealed the unique fine structure organization, genetic variations, and domestication dynamics of FibH. We also demonstrated that the length of the repetitive regions determined the mechanical properties of silk fiber, which was further confirmed by Cas9 editing of FibH. CACS is a simple, robust, and efficient approach, providing affordable accessibility to highly repetitive regions of a genome. STATEMENT OF SIGNIFICANCE: Silkworm silk is the earliest and most widely used animal fiber, and its excellent performance mainly depends on the fibroin heavy chain (FibH) protein. The FibH gene is the main breakthrough in understanding the formation mechanism and improvement of silk fiber. In the study, we developed a CACS method for characterizing the fine structure and domestication landscape of 24 silkworm FibH genes. We used CRISPR/Cas9 to edit the repetitive sequence of FibH genes, revealing the relationship between FibH genes and mechanical properties of silkworm silk. Our study is helpful in modifying silk genes to manipulate other valuable highly repetitive sequences, and provides insight for silkworm breeding.

This study employed the Taguchi method, analysis of variance, and response surface methodology for plastic gear rack injection molding parameters followed by a factorial quality validation. This study was expected to reduce the time cost of mold design and injection molding by making different combinations of the molding parameters, designing an experimental method, and performing the data simulation experiment by computer-aided engineering (CAE). With the research tool of polymer (polyacetal) for plastic material, computer-aided design mold design, and CAE mold flow analysis software, a numerical analysis of plastic molding flow was conducted. Taguchi L16 (45) orthogonal array designed 16 experimental combinations including injection molding conditions of filling time, holding pressure, holding time, plastic temperature, and mold temperature. The experimental results of molding analysis of software (Moldex3D) determined the optimum molding essentials of plastic injection: filling time 0.2 s, holding pressure 98 MPa, plastic temperature 195 °C, and mold temperature 65 °C. In this study, the parameters of the response surface method were used for the actual injection verification. The CAE simulation software can greatly improve the mold design and injection molding parameter testing time to enhance the overall working efficiency and cost control.

Hepatitis B virus (HBV) infection is a major driver of hepatocarcinogenesis. Ferroptosis is a type of iron-mediated cell death that can suppress liver transformation. Previous studies have linked HBV to ferroptosis in liver fibrosis and acute liver failure. However, whether ferroptosis is involved in HBV-mediated liver cancer is poorly understood. Here, we identified heat shock protein family A member 8 (HSPA8) as a crucial host factor that modulates HBV replication and ferroptosis in liver cancer. Hepatitis B X protein (HBx) upregulated HSPA8 by coactivating the transcription factor heat shock factor 1 (HSF1) in cells. HSPA8 enhanced HBV replication by recruiting hepatitis B core protein (HBc) to the HBV covalently closed circular DNA (cccDNA) minichromosome, forming a positive feedback loop. Moreover, HSPA8 suppressed ferroptosis in liver cancer cells by upregulating the expression of SLC7A11/GPX4 and decreasing erastin-mediated reactive oxygen species and Fe2+ accumulation in cells in vitro and in vivo. Inhibition of HSPA8 reduced the growth of HBV-positive liver tumors and increased sensitivity to erastin. In conclusion, HBx-elevated HSPA8 regulates both HBV replication and ferroptosis in liver cancer. Targeting HSPA8 could be a promising strategy for controlling HBV and hepatocarcinogenesis. Significance: HBV-induced upregulation of HSPA8 promotes hepatocarcinogenesis by suppressing ferroptosis and stimulating HBV replication, identifying HSPA8 as a potential therapeutic target in liver cancer.
