Nonalcoholic fatty liver disease and its progressive form, nonalcoholic steatohepatitis (NASH), are rapidly becoming the top causes of hepatocellular carcinoma (HCC). Currently, there are no approved therapies for the treatment of NASH. DEAD-box protein 5 (DDX5) plays important roles in different cellular processes. However, the precise role of DDX5 in NASH remains unclear.DDX5 expression was downregulated in patients with NASH, mouse models with diet-induced NASH (high-fat diet [HFD], methionine- and choline-deficient diet, and choline-deficient HFD), mouse models with NASH-HCC (diethylnitrosamine with HFD), and palmitic acid-stimulated hepatocytes. Adeno-associated virus-mediated DDX5 overexpression ameliorates hepatic steatosis and inflammation, whereas its deletion worsens such pathology. The untargeted metabolomics analysis was carried out to investigate the mechanism of DDX5 in NASH and NASH-HCC, which suggested the regulatory effect of DDX5 on lipid metabolism. DDX5 inhibits mechanistic target of rapamycin complex 1 (mTORC1) activation by recruiting the tuberous sclerosis complex (TSC)1/2 complex to mTORC1, thus improving lipid metabolism and attenuating the NACHT-, leucine-rich-repeat (LRR)-, and pyrin domain (PYD)-containing protein 3 inflammasome activation. We further identified that the phytochemical compound hyperforcinol K directly interacted with DDX5 and prevented its ubiquitinated degradation mediated by ubiquitin ligase (E3) tripartite motif protein 5, thereby significantly reducing lipid accumulation and inflammation in a NASH mouse model.These findings provide mechanistic insight into the role of DDX5 in mTORC1 regulation and NASH progression, as well as suggest a number of targets and a promising lead compound for therapeutic interventions against NASH.

Cilia are fundamental functional structures in natural biology. As the primary option of artificial cilia, magnetic cilia have been drawing extensive attention due to their excellent biocompatibility, sensitive response, and contactless actuation. However, most of the ordered magnetic cilia are fabricated by molds, suffering from high cost and low efficiency. In this paper, an ultrafast fabrication method of ordered cilia array using the micro-cavity inducing effect was proposed. With the impact of static and dynamic magnetic fields, the fine cilia were first formed in out-cavity area and then converged above cavities forming complete cilia structures. The mechanism of the micro-cavity inducing effect was further revealed. Finally, the ordered cilia array was used to develop the pressure sensor with variable stiffness, making the in situ adjustment of the sensor performance possible. The ordered cilia array was applied as a micro-mixer and largely improved the mixing efficiency for different mediums. The ordered cilia array also successfully served as the info carrier for rapid sub-encryption. This method allows the fast and controlled forming of ordered cilia arrays within 30 s, and the cilia structure can be adjusted in a large range of aspect ratios (1-9), providing an approach to large-scale producing the magnetic cilia for different applications.

Iridoviruses are emerging pathogens widely spread in diverse environments and hosts. Numerous species of the iridoviridae family are known to cause severe disease in freshwater and marine organisms. Here, we report the complete genome sequence and phylogenetic analysis of the iridovirus strain LPIV-ZS-2021 isolated from a small yellow croaker (Larimichthys polyactis) in China. The genome sequence comprises 110,560 bp with a G+C content of 53.42%, has 104 putative open reading frames (ORFs) and shares highest sequence homology with Red red seabream iridovirus (RSIV) isolated in Japan (98.61%). Phylogenetic analysis revealed that it is clustered within RSIV clade1. This is the first fully sequenced RSIV genomes detected in the small yellow croaker. The host range expansion of Megalocytivirus genus iridovirus warrants further attention to determine its potential economic and ecological impact.

Untargeted liquid chromatography-tandem mass spectrometry (LC-MS/MS) is a widely used method for discovering natural products (NPs); however, automatic MS/MS data mining for the discovery of NPs remains a challenge. In this work, LindenaneExtractor, a program based on characteristic MS/MS ions of lindenane sesquiterpenoids (LSs) was developed to automatically extract the LSs features for target LS discovery in plant extracts. To build this program, fragmentation mechanisms of characteristic ions of LSs were elucidated and confirmed by quantum chemical calculation and deuterium-labeled compounds. Subsequently, the information of characteristic ions was integrated and coded to develop LindenaneExtractor, which was further examined by standards and several public databases. Animal cloning can be achieved by somatic cell nuclear transfer (SCNT), but the resulting live birth rate is relatively low. We previously improved the efficiency of bovine SCNT by exogenous melatonin treatment or by overexpression of lysine-specific demethylase 4D (KDM4D) and 4E (KDM4E). In this study, we revealed abundant alternative splicing (AS) transitions during fertilization and embryonic genome activation, and demonstrated abnormal AS in bovine SCNT embryos compared with in vitro fertilized embryos. We used the CRISPR-Cas13d RNA-targeting system to target cis-elements of ABI2 and ZNF106 pre-mRNA to modify AS, thus reducing the ratio of abnormal-isoform SCNT embryos by nearly 50% and achieving a high survival rate (11%-19%). These results indicate that this system may provide an efficient method for bovine cloning, while also paving the way for further improvements in the efficiency of SCNT.

The cache-enabling unmanned aerial vehicle (UAV) non-orthogonal multiple access (NOMA) networks for mixture of augmented reality (AR) and normal multimedia applications are investigated, which is assisted by UAV base stations. The user association, power allocation of NOMA, deployment of UAVs and caching placement of UAVs are jointly optimized to minimize the content delivery delay. A branch and bound (BaB) based algorithm is proposed to obtain the per-slot optimization. To cope with the dynamic content requests and mobility of users in practical scenarios, the original optimization problem is transformed to a Stackelberg game. Specifically, the game is decomposed into a leader level user association sub-problem and a number of power allocation, UAV deployment and caching placement follower level sub-problems. The long-term minimization was further solved by a deep reinforcement learning (DRL) based algorithm. Simulation result shows that the content delivery delay of the proposed BaB based algorithm is much lower than benchmark algorithms, as the optimal solution in each time slot is achieved. Meanwhile, the proposed DRL based algorithm achieves a relatively low long-term content delivery delay in the dynamic environment with lower computation complexity than BaB based algorithm.

This paper studies the collision avoidance and interference mitigation for unmanned aerial vehicle (UAV) swarm where many UAVs track a common target. The considered problem is formulated to jointly minimize the average interference and ensure the collision avoidance among UAVs. By exploiting the problem characteristics, our major contributions are summarized as follows. First, the singular case tolerance (SCT)-artificial potential field (APF) is proposed to overcome the failure of traditional APFs in collision avoidance, where the repulsive force gain coefficient among UAVs is dynamically controlled by the corresponding interferences. Second, the mean field game (MFG) model is established to control communication power, where the instantaneous interferences among flying UAVs are represented by the mean field approximation. Third, considering the tight coupling of trajectory and interference of UAVs, a cooperative control approach enabled by dual-fields is proposed to jointly adjust the trajectories and power of UAVs. Simulation results validate the significant performance improvement of the cooperative control approach enabled by dual-fields. Compared with separate APF and MFG, the proposed dual-field-cooperation approach can achieve about 117% throughput gain and 88% interference reduction when the UAV swarm is close to the target.

The anisotropy of organic cations and Δ-machine learning were considered in predicting bandgaps of double hybrid organic–inorganic perovskites (DHOIPs). 19 promising DHOIPs with appropriate bandgaps for solar cells were screened out from 78 400 DHOIP.

The sixth generation (6G) mobile networks will reshape the world by offering instant, efficient, and intelligent hyper-connectivity, as envisioned by the previously proposed Ubiquitous-X 6G networks. Such hyper-massive and global connectivity will introduce tremendous challenges into the operation and management of 6G networks, calling for revolutionary theories and technological innovations. To this end, we propose a new route to boost network capabilities toward a wisdom-evolutionary and primitive-concise network (WePCN) vision for the Ubiquitous-X 6G network. In particular, we aim to concretize the evolution path toward the WePCN by first conceiving a new semantic representation framework, namely semantic base, and then establishing an intelligent and efficient semantic communication (IE-SC) network architecture. In the IE-SC architecture, a semantic intelligence plane is employed to interconnect the semantic-empowered physical-bearing layer, network protocol layer, and application-intent layer via semantic information flow. The proposed architecture integrates artificial intelligence and network technologies to enable intelligent interactions among various communication objects in 6G. It features a lower bandwidth requirement, less redundancy, and more accurate intent identification. We also present a brief review of recent advances in semantic communications and highlight potential use cases, complemented by a range of open challenges for 6G.

Recovery from gastrointestinal (GI) surgery is often interrupted by the unpredictable occurrence of postoperative complications, including infections, anastomotic leak, GI dysmotility, malabsorption, cancer development, and cancer recurrence, in which the implication of gut microbiota is beginning to emerge. Gut microbiota can be imbalanced before surgery due to the underlying disease and its treatment. The immediate preparations for GI surgery, including fasting, mechanical bowel cleaning, and antibiotic intervention, disrupt gut microbiota. Surgical removal of GI segments also perturbs gut microbiota due to GI tract reconstruction and epithelial barrier destruction. In return, the altered gut microbiota contributes to the occurrence of postoperative complications. Therefore, understanding how to balance the gut microbiota during the perioperative period is important for surgeons. We aim to overview the current knowledge to investigate the role of gut microbiota in recovery from GI surgery, focusing on the crosstalk between gut microbiota and host in the pathogenesis of postoperative complications. A comprehensive understanding of the postoperative response of the GI tract to the altered gut microbiota provides valuable cues for surgeons to preserve the beneficial functions and suppress the adverse effects of gut microbiota, which will help to enhance recovery from GI surgery.

Assembly is a critical step in the manufacturing process. Robotic assembly technology in automatic production lines has greatly improved the production efficiency. However, in assembly process, dynamic disturbances such as processing time change and advance delivery may occur, which cause the scheduling deviation. Traditional scheduling methods are not sufficient to meet the real-time and adaptive requirements in smart manufacturing. Digital twin (DT) has the characteristics of virtual-reality interaction and real-time mapping. In this paper, we propose a DT-based framework of task rescheduling for robotic assembly line (RAL) and its key methodologies, thus to realize the timely and dynamic adjustment of scheduling plan under uncertain interferences. First, a DT model of RAL task rescheduling composed of physical entity (PE), virtual entity (VE), and virtual-reality interaction mechanism is proposed. Then, a mathematical model is established. By analyzing the adaptive objective thresholds from the perspectives of event trigger and user demand trigger, a DT-driven multi-level (production unit level and line level) rescheduling strategy is proposed. Taking both the computing time and solution quality into consideration, the precedence graph is introduced to propose a rescheduling approach based on an improved discrete fireworks algorithm. Finally, the effectiveness of the proposed model and approach are verified by task scheduling experiments of RAL.

Non-methane hydrocarbons (NMHCs) can serve as precursors of ozone and photochemical smog, and hence their highly efficient detection is of great importance for air quality monitoring. Here, we synthesized a new fluorescent perylene bisimide (PBI)-cored metallacycle complex through coordination-driven self-assembly and used it for the production of a fluorescent film sensor. The unique rectangular structure of the developed fluorophore endows the sensor with enhanced sensing performance and discriminability to n-alkanes (C5-10). Specifically, the experimental detection limits for n-pentane, n-hexane, and n-decane are 39, 7, and 1.4 mg/m3, respectively, and the corresponding linear ranges are from 39 to 2546, 7 to 1745, and 1.4 to 85 mg/m3, respectively. Moreover, the sensing is fully reversible. In tandem with a gas chromatographic separation system, the film sensor showed comparable detection ability for the n-alkanes with a commercial flame ionization detector (FID), while the film sensor needs no hydrogen; it occupies a much smaller size (30 × 30 × 44 mm3) and consumes less energy (0.215 W). Further studies demonstrated that the developed sensor can be used for on-site and real-time quantification of NHMCs, laying the foundation for developing into a portable detector.

In this article, we propose the Raptor coded random access (RCRA) scheme to enable reliable transmission in the decentralized unmanned aerial vehicle (UAV) network. The considered network is composed of several overlapped random access systems with interference nodes, and the proposed RCRA scheme reduces bit-error ratio (BER) of the random access systems by three steps. First, we choose the number of slots based on a derived lower bound, which is necessary for reliable random access. Second, error-correcting codes are incorporated as the precode before random access, and then the access probability is optimized to achieve the minimum BER. Third, by correlating two consecutive slots, an idle-slot-filling approach is designed to further improve the efficiency of the random access systems. Numerical results show that the proposed RCRA scheme reduces significantly both block-error ratio (BLER) and BER at moderate- and high-signal-to-noise ratio (SNR) region. With $E_{s}/N_{0}$ equal to 0 dB, the RCRA scheme saves 20% slots, compared with the existing frameless ALOHA scheme, to achieve a target BER of 10−4.

Purpose Diabetic kidney disease (DKD) is an inflammatory disease. This study aimed to investigate the association of fibrinogen to albumin ratio (FAR) with DKD. Patients and Methods A total of 1022 type 2 diabetes mellitus (T2DM) patients with DKD and 1203 T2DM patients without DKD were enrolled in this study. Laboratory values including blood cell count, hemoglobin A1c, biochemical parameters, and fibrinogen and albumin creatinine ratio were recorded. Patients were classified according to tertile of admission FAR. Clinical parameters were compared between groups. Logistic regression, linear regression, ROC analysis and spline regression were carried out. Results FAR in the DKD group was significantly higher than that in the non-DKD group. FAR had the highest odds ratio as an independent risk factor for the development of DKD and the highest area under ROC curve for predicting DKD compared with albumin (ALB) or fibrinogen (FIB) alone. Simple linear regression analyses revealed a significant and linear correlation of FAR with neutrophil and neutrophil-to-lymphocyte ratio. FAR was an independent risk factor for development of DKD. Spline regression showed that there was a significant linear association between DKD incidence and continuous FAR value when it exceeded 67.3mg/g. Conclusion FAR is a stronger independent predictor of DKD than FIB and ALB. FAR is an independent risk factor for DKD development when it exceeded 67.3mg/g. FAR might be one of novel diagnostic biomarkers to predict and prevent DKD progression. However, a prospective study to validate the prognostic model is still needed., Graphical Abstract

Unmanned aerial vehicles (UAVs) have extensive civilian and military applications, but establishing a UAV network providing high data rate communications with low delay is a challenge. Millimeter wave (mmWave), with its high bandwidth nature, can be adopted in the UAV network to achieve high speed data transfer. However, it is difficult to establish and maintain the mmWave communication links due to the mobility of UAVs. In this paper, a beam management scheme utilizing angular domain information (ADI) is proposed to rapidly establish and reliably maintain the communication links for the mmWave UAV network. Firstly, Gaussian process machine learning (GPML)-enabled position prediction is proposed to facilitate coarse-ADI acquisition through the proposed UAV clustering algorithm. Then, with the proposed confined-ADI acquisition which removes the redundancy in the coarse-ADI acquisition, fast beam tracking with respectively the single-beam pattern and the multi-beam pattern is achieved. Finally, a data-driven beam pattern selection scheme is proposed for improving the spectrum efficiency. Simulation results verify the outstanding performance of the proposed beam management for mmWave UAV networks.

The widespread application in displays, information encryption, and sensors has boosted studies of electrochromic (EC) systems combining large contrast, fast response, high robustness, and low-cost properties. Herein, we report a film-type new EC system with a non-planar perylene bisimide-carborane derivative (PBI-CB) as the electroactive materials. It was revealed that the film demonstrated outstanding EC properties with response times of 1.18 and 0.94 s for the coloration and bleaching processes, respectively, large transmittance variation around 630 nm (45.7%), and superior stability for more than 200 coloration-bleaching cycles. Moreover, the film also showed precious electrofluorochromic (EFC) properties. The emission around 650 nm at the "on" state could be more than 24.5 times than that at the "off" state, and the response times of the off and on processes could reach 2.2 s and 4.3 s, respectively. Considering the facts that the film was fabricated via simple drop-coating, the EC/EFC operation was performed via a routine three-electrode system and the voltage applied is only -1.3 V, we believe that the EC/EFC system as developed would find applications in smart windows, information encryption, optoelectrical sensing, etc. In addition, the work could also pave the way for developing combined EC/EFC systems via employing known organic fluorophores as the electrochemical active materials, which are not only abundant in numbers but also solution-processable.

Gastric cancer is the leading cause of cancer-related death worldwide. The aim of present study was to investigate the anti-tumor effect of purified Omphalia lapidescens protein (pPeOp) in gastric cancer. Microarray analysis was performed to find out differentially expressed genes in pPeOp-treated MC-4 gastric cancer cells. The Janus kinase (JAK)/signal transducer and activator of transcription (STAT) three signaling pathway was most likely to be altered based on bioinformatics analysis. Interleukin-6 (IL-6) and NSC74859 were used as the agonist and inhibitor of the JAK/STAT3 signaling pathway, respectively. Flow cytometry and MTS assay were used for cell proliferation and viability analysis in pPeOp-treated gastric cancer cell lines with IL-6 or NSC74859. The anti-tumor effect was increased when pPeOp were co-treated with IL-6, while decreased in inhibitor treatment. The expression of the crucial members in the pathway of MC-4 cells, including glycoprotein 130 (GP130), JAK1, JAK2, STAT3, p-STAT3, suppressor of cytokine signaling SOCS1 and SOCS3, was detected by western blotting. pPeOp exhibited promising anticancer effect in the xenograft nude mice model, established by STAT3 knock down gastric cancer cells.Thus, JAK/STAT3 inhibition partially contributed to the anticancer effect of pPeOp, which may serve as a novel strategy for gastric cancer.Supplemental data for this article is available online at https://doi.org/10.1080/01635581.2021.1960385.

Seven lindenane-type sesquiterpenoid trimers, including four new ones (
