Carbohydrate is the cheapest source of energy among the three major nutrient groups, an appropriate amount of carbohydrates can reduce feed cost and improve growth performance, but carnivorous aquatic animals cannot effectively utilize carbohydrates. The objectives of the present study are aimed at exploring the effects of dietary corn starch levels on glucose loading capacity, insulin-mediated glycemic responses, and glucose homeostasis for Portunus trituberculatus. After two weeks of feeding trial, swimming crabs were starved and sampled at 0, 1, 2, 3, 4, 5, 6, 12, and 24 hours, respectively. The results indicated that crabs fed diet with 0% corn starch exhibited lower glucose concentration in hemolymph than those fed with the other diets, and glucose concentration in hemolymph remained low with the extension of sampling time. The glucose concentration in hemolymph of crabs fed with 6% and 12% corn starch diets reached the peak after 2 hours of feeding; however, the glucose concentration in hemolymph of crabs fed with 24% corn starch attained the highest value after 3 hours of feeding, and the hyperglycemia lasted for 3 hours and decreased rapidly after 6 hours of feeding. Enzyme activities in hemolymph related to glucose metabolism such as pyruvate kinase (PK), glucokinase (GK), and phosphoenolpyruvate carboxykinase (PEPCK) were significantly influenced by dietary corn starch levels and sampling time. Glycogen content in hepatopancreas of crabs fed with 6% and 12% corn starch first increased and then decreased; however, the glycogen content in hepatopancreas of crabs fed with 24% corn starch significantly increased with the prolongation of feeding time. In the 24% corn starch diet, insulin-like peptide (ILP) in hemolymph reached a peak after 1 hour of feeding and then significantly decreased, whereas crustacean hyperglycemia hormone (CHH) was not significantly influenced by dietary corn starch levels and sampling time. ATP content in hepatopancreas peaked at 1 h after feeding and then decreased significantly in different corn starch feeding groups, while the opposite trend was observed in NADH. The activities of mitochondrial respiratory chain complexes I, II, III, and V of crabs fed with different corn starch diets significantly increased first and then decreased. In addition, relative expressions of genes related to glycolysis, gluconeogenesis, glucose transport, glycogen synthesis, insulin signaling pathway, and energy metabolism were significantly affected by dietary corn starch levels and sampling time. In conclusion, the results of the present study reveal glucose metabolic responses were regulated by different corn starch levels at different time points and play an important role in clearing glucose through increased activity of insulin, glycolysis, and glycogenesis, along with gluconeogenesis suppression.

Printed cutting and repositioning templates could bring superior accuracy when transferring a maxillary plan to the operating room compared to a wafer-based method. However, the effect of these methods in different types of cases is inconclusive. The objective of the study was to compare the accuracy when using printed occlusal splints versus templates in simple and complicated cases.A retrospective cohort study design was used. Complicated cases were defined as cases involving impaction movement of more than 2 mm, occlusal plane canting of more than 3°, or midline discrepancies of more than 2.5 mm. Other cases were simple cases. Enrolled patients were randomly allocated into the digital occlusal splint (DOS) cohort and the digital templates (DT) cohort. The outcome variable was surgical accuracy, defined as the average deviation between the planned and postsurgical locations of bilateral maxillary central incisors, canines, first premolars, and first molars. Predictor variables were 1) operative complexity, simple versus complicated; and 2) technique for positioning the maxilla, DOS versus DT. Covariates were age and planned surgical movement. Two-way analysis of variance was used.Seventy patients were included in this study. Thirty-three were in the DOS cohort, and 37 in the DT cohort. The average deviation was significantly smaller in the complicated cases in the DT cohort (1.37 mm; 95% confidence interval, 1.08-1.66 mm) than that in the DOS cohort (2.47 mm; 95% confidence interval, 1.92-3.02 mm) (P = .002). The deviations in anteroposterior direction of complicated cases in the DT cohort were smaller than the corresponding values of the DOS cohort (P = .035). There is no significant difference between the deviation values of simple and complicated cases using templates (P = .116).These results indicate that in complicated cases, printed guiding templates exhibit better accuracy for repositioning the maxilla than printed occlusal splints, and the effect of templates in different cases proved to be stable.

The biosynthesis and termination of insect sex pheromones should be accurately regulated. In most moths, the biosynthesis and release of sex pheromones are regulated by a class of neuropeptides known as pheromone biosynthesis activating neuropeptides (PBANs). However, endogenous mechanisms underlying the termination of sex pheromone biosynthesis in moths remain elusive. In the present study, Helicoverpa armigera was employed as a model to investigate the role of octopamine (OA) in the inhibition of sex pheromone biosynthesis. Results demonstrated that the release of sex pheromones decreased with an increase in OA titres in older female moths. Moreover, OA treatment led to a significant decrease in sex pheromone production, female capability to attract male counterparts and subsequent female acceptance, indicating its inhibitory role in sex pheromone release. Drought detection is crucial for water resources management and global food security. Drought is typically detected using empirical indices, such as the Palmer Drought Severity Index (PDSI). However, those indices lack objectivity and are therefore suboptimal. Machine learning has been proven to be a powerful tool to define objective and optimal regressions, especially in hydrology. Here, we developed a machine learning (ML) model using Long Short-Term Memory (LSTM), which include memory effects, to predict the Evaporative Fraction (EF), an indicator of water stress at the surface, based on FLUXNET2015 Tier 1 eddy-covariance dataset. Compared to the widely used PDSI, EF is a more direct drought index to indicate water stress conditions. The results show that, firstly, with some routinely available variables, e.g., precipitation, net radiation, air temperature, relative humidity and other static variables like, Plant Functional Type (PFT) and soil property, the model can capture the EF dynamics, especially during the dry season. Secondly, we found there were different LSTM memory lengths across different Plant Functional Types. This indicates different rooting depth and different plant water use strategies that regulate the time scales of droughts. Our results have important implications for future water stress estimation, e.g., drought detection, in order to obtain a more direct and more accurate estimate of water stress.

Background and purposeClinical outcome in patients who received thrombectomy treatment is time-dependent. The purpose of this study was to evaluate the efficacy of the one-stop stroke management (OSSM) platform in reducing in-hospital workflow times in patients receiving thrombectomy compared with the traditional model.MethodsThe data of patients who received thrombectomy treatment through the OSSM platform and traditional protocol transshipment pathway were retrospectively analyzed and compared. The treatment-related time interval and the clinical outcome of the two groups were also assessed and compared. The primary efficacy endpoint was the time from door to groin puncture (DPT).ResultsThere were 196 patients in the OSSM group and 210 patients in the control group, in which they were treated by the traditional approach. The mean DPT was significantly shorter in the OSSM group than in the control group (76 vs. 122 min; P < 0.001). The percentages of good clinical outcomes at the 90-day time point of the two groups were comparable (P = 0.110). A total of 121 patients in the OSSM group and 124 patients in the control group arrived at the hospital within 360 min from symptom onset. The mean DPT and time from symptom onset to recanalization (ORT) were significantly shorter in the OSSM group than in the control group. Finally, a higher rate of good functional outcomes was achieved in the OSSM group than in the control group (53.71 vs. 40.32%; P = 0.036).ConclusionCompared to the traditional transfer model, the OSSM transfer model significantly reduced the in-hospital delay in patients with acute stroke receiving thrombectomy treatment. This novel model significantly improved the clinical outcomes of patients presenting within the first 6 h after symptom onset.

Evaporative loss of interception (Ei) is the first process occurring during rainfall, yet its role in large-scale surface water balance has been largely underexplored. Here we show that Ei can be inferred from flux tower evapotranspiration measurements using physics-informed hybrid machine learning models built under wet versus dry conditions. Forced by satellite and reanalysis data, this framework provides an observationally constrained estimate of Ei, which is on average 84.1 ± 1.8 mm per year and accounts for 8.6 ± 0.2% of total rainfall globally during 2000–2020. Rainfall frequency regulates long-term average Ei changes, and rainfall intensity, rather than vegetation attributes, determines the fraction of Ei in gross precipitation (Ei/P). Rain events have become less frequent and more intense since 2000, driving a global decline in Ei (and Ei/P) by 4.9% (6.7%). This suggests that ongoing rainfall changes favor a partitioning towards more soil moisture and runoff, benefiting ecosystem functions but simultaneously increasing flood risks.

Trehalase (Treh) hydrolyzes trehalose to generate glucose and it plays important role in many physiological processes. Acetyl-CoA, the precursor of sex pheromone biosynthesis in the pheromone gland (PG) of Helicoverpa armigera, originates from glucose during glycolysis. However, the function of Treh in sex pheromone biosynthesis remains elusive. In the present study, H. armigera was used as a model to investigate the function of two Trehs (Treh1 and Treh2) in sex pheromone biosynthesis. Results demonstrated that knockdown of HaTreh1 or HaTreh2 in female PGs led to significant decreases in Z11-16:Ald production, female ability to attract males, and successful mating proportions. Pheromone biosynthesis activating neuropeptide (PBAN) treatment triggered HaTreh1 and HaTreh2 activities in the isolated PGs and Sf9 cells. However, the activities of HaTreh1 and HaTreh2 triggered by PBAN were offset by H-89, the specific inhibitor of protein kinase A (PKA). Furthermore, the H-89 treatment significantly decreased the phosphorylation level of Trhe2, which was induced by PBAN. In addition, sugar feeding (5% sugar) increased the enzyme activities of Treh1 and Treh2. In summary, our findings confirmed that PBAN activates Treh1/2 activities by recruiting cAMP/PKA signalling, promotes glycolysis to ensure the supply of acetyl-CoA, and ultimately facilitates sex pheromone biosynthesis and mating behaviour.

Bitter melon extract (BME), which contains a variety of natural bioactive substances, has diversified biological functions. The present study is aimed at evaluating the effects of dietary supplementation of BME on growth performance, antioxidant capacity, inflammatory cytokine expression, and intestinal microbiota in common carp (Cyprinus carpio L.). An 8-week feeding trial was conducted using four isonitrogenous and isoenergetic diets containing different levels of BME (0%, 0.25%, 0.5%, and 1% for control, LBME, MBME, and HBME groups, respectively). The results showed that there were no significant differences in the growth performance of common carp among all groups, including final body weight (FBW), weight gain rate (WGR), specific growth rate (SGR), feed conversion ratio (FCR), and viscera index (VSI) ( p > 0.05 ), but the condition factor (CF) was decreased by all BME treatments ( p < 0.05 ). Serum superoxide dismutase (SOD) activity was significantly improved in all BME groups ( p < 0.05 ), and serum catalase (CAT) activity was significantly higher in the MBME group ( p < 0.05 ). Serum malondialdehyde (MDA) level was lowest while serum total antioxidant capacity (T-AOC) was highest in the HBME group ( p < 0.05 ). Dietary BME supplementation inhibited proinflammatory cytokine (il-1β and il-8) mRNA expression, while anti-inflammatory cytokine (il-6 and il-10) mRNA expressions were promoted ( p < 0.05 ). Furthermore, principal coordinate analysis (PCoA) showed that there were no differences in bacterial communities among different groups, and Shannon index and Simpson index of intestinal microbiota were unaffected ( p > 0.05 ). Whereas dietary BME reduced Firmicutes abundance and the ratio of Firmicutes to Bacteroidetes at phylum level, the relative abundance of streptococcaceae at family level, Streptococcus and Exiguobacterium at genus level were also decreased ( p < 0.05 ). Overall, these results suggested that dietary supplementation of BME improved serum antioxidant capacity, anti-inflammatory activity, and modified intestinal microbiota in common carp.

Plant root morphology is constantly reshaped in response to triggers from the soil environment. Such modifications in root system architecture involve changes in the abundance of reactive oxygen species (ROS) in the apoplast and in cell wall (CW) composition. The hybrid proline-rich proteins (HyPRPs) gene family in higher plants is considered important in the regulation of CW structure. However, the functions of HyPRPs remain to be characterized. We therefore analysed the functions of OsR3L1 (Os04g0554500) in rice. qRT-PCR and GUS staining revealed that OsR3L1 is expressed in roots. While the r3l1 mutants had a defective root system with fewer adventitious roots (ARs) and lateral roots (LRs) than the wild type, lines overexpressing OsR3L1 (R3L1-OE) showed more extensive LR formation but with a shorter root length. The expression of OsR3L1 was initiated by the OsMADS25 transcription factor. Moreover, the abundance of OsR3L1 transcripts was increased by NaCl. The R3L1-OE-3 line exhibited enhanced salt tolerance, whereas the r3l1-2 mutant showed greater salt sensitivity. The addition of H2 O2 increased the levels of OsR3L1 transcripts. Data are presented indicating that OsR3L1 modulates H2 O2 accumulation in the apoplast. We conclude that OsR3L1 regulates salt tolerance through regulation of peroxidases and apoplastic H2 O2 metabolism.

Purpose: We evaluated the feasibility and safety of using a new unibody outer double-branched stent-graft system to reconstruct the canine ascending aorta, aortic arch, and supra-aortic vessels. Materials and methods: The outer-branched stent-graft was a unibody design. The branched stent-graft consisted of a main stent-graft and 2 branches. The introducer system included a tri-channel catheter, 2 detachable sleeves, a front fixing device, a constraining wire, and a curved outer sheath. The branched stent-graft was loaded into the introducer system. Ten adult mongrel dogs underwent general anesthesia, and the branched stent-grafts were deployed into the canine ascending aorta, aortic arch, and supra-aortic vessels by the introducer system. All animals were followed up for 3 months. At the end of the follow-up period, computed tomographic angiography (CTA) was performed to observe the patency of the branched stent-grafts. Results: The mean operation time was 142.7±13.7 minutes. The mean fluoroscopy time was 20.73±2.22 minutes. The mean dosage of contrast agent was 95.9±8.7 mL. During the operation, the tri-channel catheters successfully paralleled the wires in the aorta. All 10 branched stent-grafts were successfully implanted into the canine ascending aorta and aortic arch. There were no symptoms of cerebral embolization and no incision infection during the follow-up period. Computed tomographic angiography and specimens showed that the branched stent-grafts and native vessels were patent, the inner surfaces of the branched stent-grafts were covered by neointima, and there was no retrograde aortic dissection in the ascending aorta. Conclusions: This animal research demonstrated that the unibody outer double-branched stent-graft system could be applied to reconstruct the canine ascending aorta, aortic arch, and supra-aortic vessels. Clinical Impact Thoracic endovascular aortic repair has been the main treatment method for aortic aneurysms or dissections involving the descending thoracic aorta. However, the aortic arch and ascending aorta remain the last segments of the aorta without a validated and routinely used endovascular option. In this research, we designed a new unibody outer branched stent-graft system to reconstruct the distal ascending aorta, aortic arch and supra-aortic vessels. The unibody outer branched stent-graft system could be applied to treat aortic pathologies which involve the middle and distal proximal ascending aorta, aortic arch and proximal descending aorta.

Glyceraldehyde-3-phosphate dehydrogenase (GAPDH), a key enzyme in glycolysis, is commonly used as an internal reference gene in humans, mice, and insects. However, the function of GAPDH in insect development, especially in metamorphosis, has not been reported. In the present study, Helicoverpa armigera and Spodoptera frugiperda ovarian cell lines (Sf9 cells) were used as materials to study the function and molecular mechanism of GAPDH in larval metamorphosis. The results showed that HaGAPDH was more closely related to GAPDH of S. frugiperda and Spodoptera litura. The transcript peaks of HaGAPDH in sixth instar larvae were 6L-3 (epidermal and midgut) and 6L-1 (fat body) days, and 20E and methoprene significantly upregulated the transcripts of HaGAPDH of larvae in qRT-PCR. HaGAPDH–GFP–His was specifically localized in mitochondria in Sf9 cells. Knockdown of HaGAPDH by RNA interference (RNAi) in sixth instar larvae resulted in weight loss, increased mortality, and decreases in the pupation rate and emergence rates. HaGAPDH is directly bound to soluble trehalase (HaTreh1) physically and under 20E treatment in yeast two-hybrid, coimmunoprecipitation, and colocalization experiments. In addition, knockdown of HaGAPDH increased the Treh1 activity, which in turn decreased the trehalose content but increased the glucose content in larvae. Therefore, these data demonstrated that GAPDH controlled the glucose content within the normal range to ensure glucose metabolism and metamorphosis by directly binding with HaTreh1.

Active electronic states in transition metal dichalcogenides are able to prompt hydrogen evolution by improving hydrogen absorption. However, the development of thermodynamically stable hexagonal 2H-MoS2 as hydrogen evolution catalyst is likely to be shadowed by its limited active electronic state. Herein, the charge self-regulation effect mediated by tuning Mo−Mo bonds and S vacancies is revealed in metastable trigonal MoS2 (1T'''-MoS2) structure, which is favarable for the generation of active electronic states to boost the hydrogen evolution reaction activity. The optimal 1T'''-MoS2 sample exhibits a low overpotential of 158 mV at 10 mA cm−2 and a Tafel slope of 74.5 mV dec−1 in acidic conditions, which are far exceeding the 2H-MoS2 counterpart (369 mV and 137 mV dec−1). Theoretical modeling indicates that the boosted performance is attributed to the formation of massive active electronic states induced by the charge self-regulation effect of Mo−Mo bonds in defective 1T'''-MoS2 with rich S vacancies.

Determining the origin of bone metastatic cancer (OBMC) is of great significance to clinical therapeutics. It is challenging for pathologists to determine the OBMC with limited clinical information and bone biopsy.We designed a regional multiple-instance learning algorithm to predict the OBMC based on hematoxylin-eosin (HE) staining slides alone. We collected 1041 cases from eight different hospitals and labeled 26,431 regions of interest to train the model. The performance of the model was assessed by ten-fold cross validation and external validation. Under the guidance of top3 predictions, we conducted an IHC test on 175 cases of unknown origins to compare the consistency of the results predicted by the model and indicated by the IHC markers. We also applied the model to identify whether there was tumor or not in a region, as well as distinguishing squamous cell carcinoma, adenocarcinoma, and neuroendocrine tumor.In the within-cohort, our model achieved a top1-accuracy of 91.35% and a top3-accuracy of 97.75%. In the external cohort, our model displayed a good generalizability with a top3-accuracy of 97.44%. The top1 consistency between the results of the model and the immunohistochemistry markers was 83.90% and the top3 consistency was 94.33%. The COVID-19 infection adversely affects the cardiovascular system. Transthoracic echocardiography has demonstrated diagnostic, prognostic and therapeutic utility. We report biventricular myocardial strain in COVID-19. Methods: Biventricular strain measurements were performed for 12 patients. Patients who were discharged were compared with those who needed intubation and/or died. Results: Seven patients were discharged and five died or needed intubation. Right ventricular strain parameters were decreased in patients with poor outcomes compared with those discharged. Left ventricular strain was decreased in both groups but was not statistically significant. Conclusion: Right ventricular strain was decreased in patients with poor outcomes and left ventricular strain was decreased regardless of outcome. Right ventricular strain measurements may be important for risk stratification and prognosis. Further studies are needed to confirm these findings.

Accurate global terrestrial evapotranspiration (ET) estimation is essential to better understand Earth’s energy and water cycles. Although several global ET products exist, recent studies indicate that ET estimates exhibit high uncertainty. With the increasing trend of extreme climate hazards (e.g., droughts and heat waves), accurate ET estimation under extreme conditions remains challenging. To overcome these challenges, we used 3-hour and 0.25° Global Land Data Assimilation System (GLDAS) datasets (net radiation, land surface temperature (LST), and air temperature) and three-temperature (3T) model, without resistance and parameter calibration, in global terrestrial ET product development. The results demonstrated that the 3T model-based ET product agreed well with both global eddy covariance (EC) tower (root mean square error (RMSE) = 24.9 mm month-1) and basin-scale water balance observations (RMSE =116.0 mm yr-1). The 3T model-based global terrestrial ET product was comparable to other common ET products, i.e., MOD16, P-LSH, PML, GLEAM, GLDAS, and Fluxcom, retrieved from various models, but the 3T model performed better under extreme weather conditions in croplands than did the GLDAS, attaining 9.0–20 % RMSE reduction. The proposed ET product could provide periodic and large-scale information to support water cycle-related studies. As the only member of the CX3C chemokine receptor subfamily, CX3CR1 binds to its sole endogenous ligand CX3CL1, which shows notable potential as a therapeutic target in atherosclerosis, cancer, and neuropathy. However, the drug development of CX3CR1 is hampered partially by the lack of structural information. Here, we present two cryo–electron microscopy structures of CX3CR1-G i1 complexes in ligand-free and CX3CL1-bound states at 2.8- and 3.4-Å resolution, respectively. Together with functional data, the structures reveal the key factors that govern the recognition of CX3CL1 by both CX3CR1 and US28. A much smaller conformational change of helix VI upon activation than previously solved class A GPCR-G i complex structures is observed in CX3CR1, which may correlate with three cholesterol molecules that play essential roles in conformation stabilization and signaling transduction. Thus, our data deepen the understanding of cholesterol modulation in GPCR (G protein–coupled receptor) signaling and provide insights into the diversity of G protein coupling.

Burn injury leads to mitochondrial dysfunction and autophagy, also known as mitophagy. The alleviation of mitochondrial damage may be a potential method for the treatment of burn injury and complications. In this animal study, we analyzed the expression of mitochondrial damage- and mitophagy-related factors, specifically PINK1 and PRKN. The results showed mitochondria damage in the skin; compared with the normal control group, genes involved in the mitochondrial damage, such as Nrf-1, UQCRC2, CYC1, and NDUFA9, as well as in the mitophagy, including PINK1, PRKN, MFN1, and USP30, were differentially expressed. Furthermore, PINK1 interacted with PRKN and participated in mitophagy in the skin. In conclusion, our data reveal more about the mechanism underlying mitophagy in burns, providing a potential clinical treatment.

Alzheimer's disease (AD) is an irreversible and progressive neurodegenerative disorder featured by memory loss and cognitive default. However, there has been no effective therapeutic approach to prevent the development of AD and the available therapies are only to alleviate some symptoms with limited efficacy and severe side effects. Necroptosis is a new kind of cell death, being regarded as a genetically programmed and regulated pattern of necrosis. Increasing evidence reveals that necroptosis is tightly related to the occurrence and development of AD. This review aims to summarize the potential role of necroptosis in AD progression and the therapeutic capacity of targeting necroptosis for AD patients.

Oxygen electrochemistry plays a key role in renewable energy technologies such as fuel cells and electrolyzers, but its slow kinetics limits the performance and commercialization of such devices. Here, a novel triple-layer IrO6/MnO6 nanosheet was developed by a simple hydrothermal method. During the substitution of Mn by Ir, lattice compressive strain is introduced into the IrO6/MnO6 nanosheets, thereby tuning their electronic structure to suit the favorite oxygen evolution reaction (OER) activity. This compressively strained nanosheet catalyst exhibits an excellent mass activity of 7882 A g− 1 at an overpotential of 270 mV in 0.5 M H2SO4, and reaches 10, 50 and 100 mA cm− 2 at overpotentials of only 62 mV, 197 mV and 234 mV, respectively, ranking the highest OER activity among the state-of-the-art values in acid media. The catalyst exhibits a remarkable stability even at 300 mA cm− 2 in 0.5 M H2SO4. Using the nanosheets as OER catalyst and NiMo alloy as hydrogen evolution reaction catalyst, a two-electrode electrolyzer achieves 10 mA cm− 2 with only a cell voltage of 1.46 V for overall water splitting in 0.5 M H2SO4. This strategy enables the material with high feasibility for practical applications on hydrogen production.

To establish a canine model of aortic arch aneurysm that is suitable for research on new devices and techniques applied to the aortic arch.Fifteen mongrel dogs underwent surgery. The autologous pericardial patch was sewn on the aortotomy site in the anterior wall of the aortic arch. The animals were followed up for 3 months postoperatively. Computed tomography angiography was used to visualize and measure the aneurysm model. Hematoxylin and eosin staining was used to observe the histological characteristics of the aneurysm model. Changes in aneurysm diameter over time were analyzed using analysis of variance.One dog died of hemorrhage during surgery. Fourteen dogs survived the surgical procedure. Two of them died on the first postoperative day because of ruptures at the suturing margin. The diameter of the aneurysm model was twice as large as that of the aortic arch. There was no significant change in the maximum diameter of the aneurysm model during the follow-up period.We established a controllable and stable aortic arch aneurysm model created with an autologous pericardium patch. The aneurysm model can be used to research endoleaks after thoracic endovascular aortic repair and new endovascular techniques can be applied to the aortic arch.
