Environmental polarity is an important factor that drives biomolecular interactions to regulate cell function. Herein, a general method of using the fluorogenic probe NTPAN-MI is reported to quantify the subcellular polarity change in response to protein unfolding. NTPAN-MI fluorescence is selectively activated upon labeling unfolded proteins with exposed thiols, thereby reporting on the extent of proteostasis. NTPAN-MI also reveals the collapse of the host proteome caused by influenza A virus infection. The emission profile of NTPAN-MI contains information of the local polarity of the unfolded proteome, which can be resolved through spectral phasor analysis. Under stress conditions that disrupt different checkpoints of protein quality control, distinct patterns of dielectric constant distribution in the cytoplasm can be observed. However, in the nucleus, the unfolded proteome was found to experience a more hydrophilic environment across all the stress conditions, indicating the central role of nucleus in the stress response process.

After the accidental discovery of cis-platinum, extensive attempts have centralized on the rational design of metallic compounds for cancer treatment. Here a solvent-dependent complex of nickel (II) with 1,10-phenanthroline and naproxen, [Ni(1,10-phenanthroline)(naproxen)2(solvent)], solvent = 83% H2O and 17% EtOH in the crystal structure, has been synthesized and specified by the X-ray structure analysis. It’s in vitro DNA binding was inspected by the multispectroscopic methods and gel electrophoresis. The data of DNA-viscosity and competition fluorimetric test by methylene blue (MB) and Hoechst 33258 confirm groove binding mode of the complex to CT-DNA. Comparison of the results of this binding study with previous work revealed that the mode of binding of small compounds to DNA is highly influenced by the structure of the compounds. The DNA cleavage potency of the complex was appraised by the agarose gel electrophoretic and it was found that the complex does not have any momentous cleavage potency on the pUC18 plasmid DNA. The cytotoxicity of the complex on HT 29, HepG2 and HEK-293 cell lines by MTT method indicates that %inhibition of the complex on HT 29 is better than HepG2, compared with cisplatin drug. On HEK-293 cells, %inhibition growth of normal cells of the complex is less than cisplatin. Flow cytometry analysis of the complex on the HT 29 cells indicated the apoptosis cell death. RT-PCR studies revealed down-regulation of BCL2 expression, while the expression of BAX, caspase 3 and BAX/BCL2 genes was up-regulated in HT 29 cells by the complex. The intramolecular Diels-Alder reaction has been used as a powerful method to access the tricyclic core of the eunicellin natural products from a number of 9-membered-ring precursors. The endo/exo selectivity of this reaction can be controlled through a remarkable organocatalylic approach, employing MacMillan's imidazolidinone catalysts, although the mechanistic origin of this selectivity remains unclear. We present a combined experimental and density functional theory investigation, providing insight into the effects of medium-ring constraints on the organocatalyzed intramolecular Diels-Alder reaction to form the isobenzofuran core of the eunicellins.

In this manuscript we report the synthesis and X-ray characterization of four Zn(II) complexes and three Cd(II) complexes with an asymmetrical hydrazone–pyridine based ligand {HL = 2-acetyl-pyridyl-isonicotinoylhydrazone (HAPIH)}; i.e. {[Zn(HL)2](NO3)2·H2O} (1), [Zn(HL)Br2] (2), [Zn(HL)I2] (3), [Zn(HL)(NCS)2] (4), [Cd(L)2] (5), {[Cd(HL)Br2]·CH3OH} (6) and {[Cd(HL)I2]·2CH3OH} (7). The Schiff base acts as a tridentate N2O-donor ligand through the oxygen, the imine and pyridine nitrogen atoms in all the complexes. In most complexes, the ligand is observed to coordinate as a zwitterion since the proton in the hydrazine group ([double bond, length as m-dash]N–NH–C[double bond, length as m-dash]O) shifted to the uncoordinated pyridine ring, except in 7. On the other hand, in 5 the ligand acts as a negatively charged species and is bound to the cadmium center in the enolic form ([double bond, length as m-dash]N–N[double bond, length as m-dash]C–O−). In complexes 2–4, 6 and 7, the coordination geometry around each metal center is distorted trigonal bipyramidal, with the coordination sphere of the metal completed by two halide or NCS anions. On the other hand, in homoleptic complexes 1 and 5, the metal, chelated by two tridentate Schiff base ligands, exhibits an octahedral geometry. In the crystal packing of all compounds, the pyridine rings favour π–π interactions among the symmetry related complexes. The noncovalent interactions among the complexes have been analyzed using Hirshfeld surface analysis and DFT calculations using Grimme's D3 dispersion correction to properly describe the π–π interactions., We are grateful to the University of Tabriz for generous financial support to this research. S-Benzylisothiouronium halides are used as shelf-stable, odorless thiol equivalents. The method developed is used to access 2-(benzylthio)-4-(trifluoromethyl)thiazole carboxyl building blocks. Compared to tris(2-phenylpyridine)iridium(III) ([Ir(ppy)3]), iridium(III) complexes containing difluorophenylpyridine (df-ppy) and/or an ancillary triazolylpyridine ligand [3-phenyl-1,2,4-triazol-5-ylpyridinato (ptp) or 1-benzyl-1,2,3-triazol-4-ylpyridine (ptb)] exhibit considerable hypsochromic shifts (ca. 25-60 nm), due to the significant stabilising effect of these ligands on the HOMO energy, whilst having relatively little effect on the LUMO. Despite their lower photoluminescence quantum yields compared with [Ir(ppy) 3] and [Ir(df-ppy)3], the iridium(III) complexes containing triazolylpyridine ligands gave greater electrogenerated chemiluminescence (ECL) intensities (using tri-n-propylamine (TPA) as a co-reactant), which can in part be ascribed to the more energetically favourable reactions of the oxidised complex (M+) with both TPA and its neutral radical oxidation product. The calculated iridium(III) complex LUMO energies were shown to be a good predictor of the corresponding M+ LUMO energies, and both HOMO and LUMO levels are related to ECL efficiency. The theoretical and experimental data together show that the best strategy for the design of efficient new blue-shifted electrochemiluminophores is to aim to stabilise the HOMO, while only moderately stabilising the LUMO, thereby increasing the energy gap but ensuring favourable thermodynamics and kinetics for the ECL reaction. Of the iridium(III) complexes examined, [Ir(df-ppy) 2(ptb)]+ was most attractive as a blue-emitter for ECL detection, featuring a large hypsochromic shift (λmax=454 and 484 nm), superior co-reactant ECL intensity than the archetypal homoleptic green and blue emitters: [Ir(ppy)3] and [Ir(df-ppy)3] (by over 16-fold and threefold, respectively), and greater solubility in polar solvents.
