Emotion perception is essential to affective and cognitive development which involves distributed brain circuits. The ability of emotion identification begins in infancy and continues to develop throughout childhood and adolescence. Understanding the development of brain's emotion circuitry may help us explain the emotional changes observed during adolescence. Our previous study delineated the trajectory of brain functional connectivity (FC) from late childhood to early adulthood during emotion identification tasks. In this work, we endeavour to deepen our understanding from association to causation. We proposed a Bayesian incorporated linear non-Gaussian acyclic model (BiLiNGAM), which incorporated our previous association model into the prior estimation pipeline. In particular, it can jointly estimate multiple directed acyclic graphs (DAGs) for multiple age groups at different developmental stages. Simulation results indicated more stable and accurate performance over various settings, especially when the sample size was small (high-dimensional cases). We then applied to the analysis of real data from the Philadelphia Neurodevelopmental Cohort (PNC). This included 855 individuals aged 8-22 years who were divided into five different adolescent stages. Our network analysis revealed the development of emotion-related intra- and inter- modular connectivity and pinpointed several emotion-related hubs. We further categorized the hubs into two types: in-hubs and out-hubs, as the center of receiving and distributing information. Several unique developmental hub structures and group-specific patterns were also discovered. Our findings help provide a causal understanding of emotion development in the human brain.

The dynamic characteristics of functional network connectivity have been widely acknowledged and studied. Both shared and unique information has been shown to be present in the connectomes. However, very little has been known about whether and how this common pattern can predict the individual variability of the brain, i.e. "brain fingerprinting", which attempts to reliably identify a particular individual from a pool of subjects. In this paper, we propose to enhance the individual uniqueness based on an autoencoder network. More specifically, we rely on the hypothesis that the common neural activities shared across individuals may lessen individual discrimination. By reducing contributions from shared activities, inter-subject variability can be enhanced. Results show that that refined connectomes utilizing an autoencoder with sparse dictionary learning can successfully distinguish one individual from the remaining participants with reasonably high accuracy (up to 99:5% for the rest-rest pair). Furthermore, high-level cognitive behavior (e.g., fluid intelligence, executive function, and language comprehension) can also be better predicted using refined functional connectivity profiles. As expected, the high-order association cortices contributed more to both individual discrimination and behavior prediction. The proposed approach provides a promising way to enhance and leverage the individualized characteristics of brain networks.

Multi-modal brain functional connectivity (FC) data have shown great potential for providing insights into individual variations in behavioral and cognitive traits. The joint learning of multi-modal imaging data can utilize the intrinsic association, and thus can boost the learning performance. Although several multi-task based learning models have already been proposed by viewing the feature learning on each modality as one task, most of them ignore the geometric structure information inherent in the modalities, which may play an important role in extracting discriminative features. In this paper, we propose a new manifold regularized multi-task learning model by simultaneously considering between-subject and between-modality relationships. Besides employing a group-sparsity regularizer to jointly select a few common features across multiple tasks (modalities), we design a novel manifold regularizer to preserve the structure information both within and between modalities in our model. This will make our model more adaptive for realistic data analysis. Our model is then validated on the Philadelphia Neurodevelopmental Cohort dataset, where we regard our modalities as functional MRI (fMRI) data collected under two paradigms. Specifically, we conduct experimental studies on fMRI based FC network data in two task conditions for intelligence quotient (IQ) prediction. The results demonstrate that our proposed model can not only achieve improved prediction performance, but also yield a set of IQ-relevant biomarkers.

Scanning laser optical tomography (SLOT) is a 3D imaging technique, based on the principle of computed tomography to visualize samples up to magnitude of several centimeters. Intrinsic contrast mechanisms as absorption, scattering and autofluorescence provide information about the 3D architecture and composition of the sample. Another valuable intrinsic contrast mechanism is second harmonic generation (SHG), which is generated in noncentrosymmetric materials and commonly used to image collagen in biological samples. The angular dependence of the SHG signal, however, produces artifacts in reconstructed optical tomography datasets (OPT, SLOT). Thus, successful use of this intrinsic contrast mechanism is impaired. We investigate these artifacts by simulation and experiment and propose an elimination procedure that enables successful reconstruction of SHG-SLOT data. Nevertheless, in many cases specific labeling of certain structures is necessary to make them visible. Using multiple dyes in one sample can lead to crosstalk between the different channels and reduce contrast of the images. Also autofluorescence of the sample itself can account for that. By using multispectral imaging in combination with spectral unmixing techniques, this loss can be compensated. Therefore either a spectrally resolved detection path, or spectrally resolved excitation is required. Therefore we integrated a white supercontinuum light source in our SLOT-setup that enables a spectral selection of the excitation beam and extended the detection path to a four channel setup. This enables the detection of three fluorescence channels and one absorption channel in parallel, and increases the contrast in the reconstructed 3D images significantly. © COPYRIGHT SPIE. Phthalocyanines (Pc) are promising second-generation photosensitizers for the photodynamic therapy (PDT) of cancer. We report on the tetrasulfonated aluminum phthalocyanine (AlPcS4) localization in cultured Chinese hamster lung cells studied by means of confocal laser scanning microscopy (CLSM). In these cells AlPcS4 was found in granules surrounding Golgi apparatus and in the peripheral cytoplasmic region. Peripheral Pc-containing granules partially coincided with the acidic cellular compartments. The effect of irradiation with light on Pc intracellular distribution was also studied. In the Pc-free medium disruption of some Pc- containing granules was observed followed by appearance of Pc fluorescence in the cell plasma membrane, the nuclear envelope, and the near-nuclear region. When cells were irradiated in the presence of Pc in external medium a drastic increase of membrane permeability to Pc was observed, followed by Pc binding the cell plasma membrane, nuclear envelope, and some structures in the cytoplasm. Diffusive Pc fluorescence in the nucleus was also observed. A V-shape Bio-Transfer-Standard (V-BTS), developed and produced at the University of Helsinki (UH), was measured in two laboratories. In comparison to Siemens Star calibration specimens, the V-BTS performs better at high lateral frequencies close to the diffraction limit of the optical instrument. This peunits deteunining of the Instrument Transfer Function (ITF). The V-BTS features two lipid bilayer steps that partly overlap each other at an angle of 20, with an average height of 4.6 +/- 0.1 nm. The Round Robin (RR) test aims to determine whether the V-BTS and the developed application protocol work with different optical profilers in different laboratories. First the artefact was measured at Sensofar-Tech, S.L. using an S-neox profiler working in Phase Shifting Interferometry mode. Then V-BTS was measured at UH using a custom-built Scanning White Light Interferometer. All measurements done by four different operators at the two laboratories have a range or standard deviation of 0.1 nm which agrees with the theoretical estimates and with measurements done using an atomic force microscope and with a surface plasmon resonance based instrument. The RR results show the applicability of the V-BTS for calibration and for ITF characterization of 3D optical profilers.

Light-sheet microscopy (LSM) has received great interest for fluorescent imaging applications in biomedicine as it facilitates three-dimensional visualisation of large sample volumes with high spatiotemporal resolution whilst minimising irradiation of, and photo-damage to the specimen. Despite these advantages, LSM can only visualize superficial layers of turbid tissues, such as mammalian neural tissue. Propagation-invariant light modes have played a key role in the development of high-resolution LSM techniques as they overcome the natural divergence of a Gaussian beam, enabling uniform and thin light-sheets over large distances. Most notably, Bessel and Airy beam-based light-sheet imaging modalities have been demonstrated. In the single-photon excitation regime and in lightly scattering specimens, Airy-LSM has given competitive performance with advanced Bessel-LSM techniques. Airy and Bessel beams share the property of self-healing, the ability of the beam to regenerate its transverse beam profile after propagation around an obstacle. Bessel-LSM techniques have been shown to increase the penetration-depth of the illumination into turbid specimens but this effect has been understudied in biologically relevant tissues, particularly for Airy beams. It is expected that Airy-LSM will give a similar enhancement over Gaussian-LSM. In this paper, we report on the comparison of Airy-LSM and Gaussian-LSM imaging modalities within cleared and non-cleared mouse brain tissue. In particular, we examine image quality versus tissue depth by quantitative spatial Fourier analysis of neural structures in virally transduced fluorescent tissue sections, showing a three-fold enhancement at 50 μm depth into non-cleared tissue with Airy-LSM. We describe work on producing a selective plane illumination microscope for cardiac imaging in zebra fish embryos. The system has a novel synchronization system for imaging oscillating structures (e.g. the heart) and will have adaptive optics for image optimization.

We have developed a novel microscope technique that can achieve wide field-of-view (FOV) imaging and yet possess resolution that is comparable to conventional microscope. The principle of wide FOV microscope system breaks the link between resolution and FOV magnitude of traditional microscopes. Furthermore, by eliminating bulky optical elements from its design and utilizing holographic optical elements, the wide FOV microscope system is more cost-effective. In our system, a hologram was made to focus incoming collimated beam into a focus grid. The sample is put in the focal plane and the transmissions of the focuses are detected by an imaging sensor. By scanning the incident angle of the incoming beam, the focus grid will scan across the sample and the time-varying transmission can be detected. We can then reconstruct the transmission image of the sample. The resolution of microscopic image is limited by the size of the focus formed by the hologram. The scanning area of each focus spot is determined by the separation of the focus spots and can be made small for fast imaging speed. We have fabricated a prototype system with a 2.4-mm FOV and 1-μm resolution. The prototype system was used to image onion skin cells for a demonstration. The preliminary experiments prove the feasibility of the wide FOV microscope technique, and the possibility of a wider FOV system with better resolution.
