SUMMARY A velocity model is generally an imperfect representation of the subsurface, which cannot precisely account for the 3-D inhomogeneities of Earth structure. We present a Bayesian moment tensor inversion framework for applications where reliable, tomography-based, velocity model reconstructions are not available. In particular, synthetic data generated using a 3-D model (SEG-EAGE Overthrust) are inverted using a layered medium model. We use a likelihood function derived from an optimal transport distance—specifically, the transport-Lagrangian distance introduced by Thorpe et al.—and show that this formulation yields inferences that are robust to misspecification of the velocity model. We establish several quantitative metrics to evaluate the performance of the proposed Bayesian framework, comparing it to Bayesian inversion with a standard Gaussian likelihood. We also show that the non-double-couple component of the recovered mechanisms drastically diminishes when the impact of velocity model misspecification is mitigated.

Previous studies on the association between ENO1 expression and breast cancer prognosis had yielded inconsistent results. In the present study, we assessed the prognostic effect of ENO1 in breast cancer using Guangzhou Breast Cancer Study (GZBCS) cohort with full consideration of the potential confounders and the modification effects. The results were further validated in the TCGA-BRCA cohort and explained by tumor immunity.Methods: ENO1 protein expressions were evaluated by immunohistochemistry in tissue microarrays from 961 patients with primary invasive breast cancer. Chi-square tests were used to assess the association of ENO1 levels with the patient’s characteristics. Cox regression models were applied to assess the prognostic effects. The TCGA-BRCA cohort was utilized to validate the results and explore the potential mechanisms. The immune infiltration was determined using the CIBERSORT and ssGSEA algorithms; the correlation between ENO1 expression and the abundance of tumor-infiltrating immune cells (TIICs) and scores of immune-related functions was evaluated by Wilcoxon signed-rank tests and Spearman’s rank test.Results: ENO1 protein expression exerted a protective effect on OS in stage I/II patients (HR=0.58, 95% CI: 0.35– 0.96) but not in stage III patients (HR=1.42, 95% CI: 0.81– 2.49, P interaction=0.04) in GZBCS; consistent results were obtained at mRNA levels in TCGA cohort. Immune infiltration analyses revealed that ENO1 was positively correlated with multiple antitumor TIICs (including M1 macrophages, B cells, CD8 T cells, T helper 2 cells, and NK cells) only in stage I/II but not stage III patients.Conclusion: A higher expression of ENO1 was associated with a better prognosis only in early-stage breast cancer, which may be related to the different effects of ENO1 on immune infiltration, suggesting that ENO1 may be a promising target for precision immunotherapy in breast cancer.Keywords: ENO1, breast cancer, prognosis, tissue microarray, tumor-infiltrating immune cells

Background: Cervical spondylotic radiculopathy (CSR) is the most common type of cervical spondylosis, which mainly presents as radioactive neuralgia. Neuralgia is closely related to gliocyte in the central nervous system. Therefore, to investigate the therapeutic effect and mechanism of LAA (astrocyte inhibitor) electro-acupuncture on neuralgia in rats with cervical spondylotic radiculopathy based on the neuron-gliocyte-chemokine signaling pathway. Material & Methods: The models of rats with neuralgia in cervical spondylotic radiculopathy were established and the intervention of electro-acupuncture and LAA were carried out. All rats were randomly divided into 4 groups (12 rats in each group). Control group, Model group, Sham operation group and Electro-acupuncture group. Results: The rats were bound to the plate in a prone position and treated with electro-acupuncture at Jiaji point, with dense wave frequency of 50 Hz and wave repacking frequency of 10 Hz, once a day, 20 min each time, for 14 consecutive days. The results found that the content of calcitonin gene-related peptides (CGRP), adenylate cyclase (AC), substances P in C6–C7 primary sensory neuron terminal of the spinal cord segment were significantly decreased in electro-acupuncture group compared with model group. Relative expression of RNA levels of protein kinase (PKC), Ca2+ channel protein (VGCC, CCL2 and CXC3L1) in electro-acupuncture group were significantly decreased compared with model group. Westernblot detected that the expression of IL-1β, IL-6, IL-18, TNF-α, NMDA and AMPA proteins in LAA+electro+acupuncture group were obvioulsy decreased. It indicates that neuralgia is closely related to gliocyte in the central nervous system, and also proves that LAA+electro acupuncture can effectively relieve neuralgia caused by cervical spondylosis.

Background: Few randomized controlled trials (RCTs) investigated the effects of mindfulness intervention on affective and cognitive symptoms in older adults with mild cognitive impairment (MCI). Furthermore, no RCTs on mindfulness followed participants beyond two years. Objective: To examine the longitudinal effects of a mindful awareness practice (MAP) intervention on depressive, anxiety, and cognitive symptoms in MCI. Methods: In this parallel-arm and assessor-blinded RCT, 55 community-dwelling older adults with MCI were randomized into the MAP or active control, i.e., health education program (HEP). Intervention sessions were conducted weekly for three months and monthly for the subsequent six months. Assessments and follow-up were conducted at baseline, 3-month, 9-month, and 5-year time points. Depressive, anxiety, and cognitive symptoms were measured using the Geriatric Depression Scale-15 (GDS-15), Geriatric Anxiety Inventory-20 (GAI-20), and Mini-Mental State Examination (MMSE), respectively. Linear-mixed models, following the intention-to-treat principle, were used for data analyses. Results: A total of 55 participants aged 60 to 86 (Mean age: 71.3±6 years old) was recruited, with n = 28 allocated to the MAP arm and n = 27 allocated to the HEP arm. Compared to HEP, GDS-15, GAI-20, and MMSE scores did not differ significantly in MAP during follow-ups. Conclusion: Compared to HEP, MAP did not improve affective symptoms nor delay deteriorations in general cognition in community-dwelling older adults with MCI. Compared to our previous findings showing domain-specific improvements in MAP over HEP in attention and memory up to 9 months, this study highlights the importance of examining domain-specificity using detailed cognitive measures in non-pharmacological intervention with MCI.

Persistent byte-addressable memory (PM) is poised to become prevalent in future computer systems. PMs are significantly faster than disk storage, and accesses to PMs are governed by the Memory Management Unit (MMU) just as accesses with volatile RAM. These unique characteristics shift the bottleneck from I/O to operations such as block address lookup—for example, in write workloads, up to 45% of the overhead in ext4-DAX is due to building and searching extent trees to translate file offsets to addresses on persistent memory. We propose a novel contiguous file system, ctFS, that eliminates most of the overhead associated with indexing structures such as extent trees in the file system. ctFS represents each file as a contiguous region of virtual memory, hence a lookup from the file offset to the address is simply an offset operation, which can be efficiently performed by the hardware MMU at a fraction of the cost of software-maintained indexes. Evaluating ctFS on real-world workloads such as LevelDB shows it outperforms ext4-DAX and SplitFS by 3.6× and 1.8×, respectively.

Emotion is a feeling that can be expressed by different mediums. Emotion analysis is a key task in NLP which is responsible for judging the emotional tendency of texts. Currently, in a complex multi-semantic environment, it still suffers from poor performance. Traditional methods usually require human intervention, while deep learning always has a trade-off between local and global features. To solve the problem that deep learning models generalize poorly for emotion analysis, this article proposed a semantic-enhanced method called RA-CNN, a classification model under a multi-semantic environment. It integrates CNN for local feature extraction, RNN for global feature extraction, and attention mechanism for feature scaling. As a result, it can acquire the correct meaning of sentences. After experimenting with the hotel review dataset, it has an improvement in positive feeling classification compared with the baseline model (3%~13%), and it showed a competitive performance compared with ordinary deep learning models (~1%). On negative feeling classification, it also performed well close to other models.

Background. Kidney renal clear cell carcinoma (KIRC) is considered as a highly immune infiltrative tumor. Necroptosis is an inflammatory programmed cell death associated with a wide range of diseases. Long noncoding RNAs (lncRNAs) play important roles in gene regulation and immune function. lncRNA associated with necroptosis could systematically explore the prognostic value, regulate tumor microenvironment (TME), etc. Method. The patients’ data was collected from TCGA datasets. We used the univariate Cox regression (UCR) to select prediction lncRNAs that are related to necroptosis. Meanwhile, risk models were constructed using LASSO Cox regression (LCR). Kaplan–Meier (KM) analysis, accompanied with receiver operating characteristic (ROC) curves, was performed to assess the independent risk factors of different clinical characteristics. The evaluated factors are age, gender, disease staging, grade, and their related risk score. Databases such as Gene Ontology (GO), Kyoto encyclopedia of genes and genomes (KEGG), and Gene set enrichment analysis (GSEA) were used to search the probable biological characteristics that could influence the risk groups, containing signaling pathway and immue-related pathways. The single-sample gene set enrichment analysis (ssGSEA) was chosen to perform gene set variation analysis (GSVA), and the GSEABase package was selected to detect the immune and inflammatory infiltration profiles. The TIDE and I C 50 evaluation were used to estimate the effectiveness of clinical treatment on KIRC. Results. Based on the above analysis, we have got a conclusion that patients who show high risk had higher immune infiltration, immune checkpoint expression, and poorer prognosis. We identified 19 novel prognostic necroptosis-related lncRNAs, which could offer opinions for a deeper study of KIRC. Conclusion. The risk model we constructed makes it possible to predict the prognosis of KIRC patients and offers directions for further research on the prognostication and treatment strategies for KIRC.

In this paper, the stabilization of singularly perturbed systems by means of observer-based event-triggered control is studied. To obtain the ultimately bounded stability of the closed-loop system, two event-triggering conditions are designed, respectively, for the slow and the fast subsystems of the singularly perturbed systems based on the state errors between the observer and models. In each event-triggering condition, there are two different stages to be designed. In the first stage, the event-triggering conditions are given based on the observer error dynamics. While in the second stage, the absolute type event-triggering conditions are used. Under the designed event-triggering conditions, Zeno behavior can be eliminated from the closed-loop systems. Finally, numerical examples are provided to illustrate the efficiency and feasibility of the theoretical results.

In this work, we utilized polycyclic aromatic hydrocarbon (PAH) derivatives as ligands to develop a zinc-based metal-organic framework (Zn-MOF) as an effective detection probe to construct an electrochemiluminescence (ECL) sensor for trenbolone detection. As traditional ECL emitters, PAHs and their derivatives have limited luminescence efficiency because of the aggregation-induced quenching (ACQ) effect. Therefore, Zn-PTC was designed by the coordination of 3,4,9,10-perylenetetracarboxylic (PTC) in the MOF to eliminate the ACQ effect. Meanwhile, Zn-PTC formed based on an aromatic ligand possessed the metal-to-ligand charge-transfer (MLCT) effect, which could transfer the energy of Znsup2+/supto the aromatic ligand for strong luminescence. The ECL efficiency of Zn-PTC was calculated to be approximately 2.2 times that of the ligand (Ksub4/subPTC). Second, the Ag@Fe core-shell bimetallic nanocrystal was prepared for efficient activation of persulfate (Ssub2/subOsub8/subsup2-/sup), thereby generating more sulfate radicals (SOsub4/subsup•-/sup) to further promote ECL emission. According to ECL characterizations, UV-vis and fluorescence spectra, and density functional theory calculations, the luminescence and signal amplification mechanisms were investigated. In addition, NKFRGKYKC (NKF) was introduced as an affinity ligand to directionally immobilize the target antibodies, thus releasing specific sites in their Fab fragment to enhance binding activity. Based on the above strategies, the constructed biosensor exhibited high sensitivity, realizing trace detection of TBE with a wide detection range (10 fg/mL-100 ng/mL) and a low detection limit (3.28 fg/mL). This study provided an important reference for sensitive monitoring of steroid pollutants in the environment.

Background Typhoid fever and paratyphoid fever are one of the most criticial public health issues worldwide, especially in developing countries. The incidence of this disease may be closely related to socio-economic factors, but there is a lack of research on the spatial level of relevant determinants of typhoid fever and paratyphoid fever. Methods In this study, we took Hunan Province in central China as an example and collected the data on typhoid and paratyphoid incidence and socio-economic factors in 2015–2019. Firstly spatial mapping was made on the disease prevalence, and again using geographical probe model to explore the critical influencing factors of typhoid and paratyphoid, finally employing MGWR model to analysis the spatial heterogeneity of these factors. Results The results showed that the incidence of typhoid and paratyphoid fever was seasonal and periodic and frequently occurred in summer. In the case of total typhoid and paratyphoid fever, Yongzhou was the most popular, followed by Xiangxi Tujia and Miao Autonomous Prefecture, Huaihua and Chenzhou generally focused on the south and west. And Yueyang, Changde and Loudi had a slight increase trend year by year from 2015 to 2019. Moreover, the significant effects on the incidence of typhoid and paratyphoid fever from strong to weak were as follows: gender ratio(q = 0.4589), students in ordinary institutions of higher learning(q = 0.2040), per capita disposable income of all residents(q = 0.1777), number of foreign tourists received(q = 0.1697), per capita GDP(q = 0.1589), and the P values for these factors were less than 0.001. According to the MGWR model, gender ratio, per capita disposable income of all residents and Number of foreign tourists received had a positive effect on the incidence of typhoid and paratyphoid fever. In contrast, students in ordinary institutions of higher learning had a negative impact, and per capita GDP shows a bipolar change. Conclusions The incidence of typhoid and paratyphoid fever in Hunan Province from 2015 to 2019 was a marked seasonality, concentrated in the south and west of Hunan Province. Attention should be paid to the prevention and control of critical periods and concentrated areas. Different socio-economic factors may show other directions and degrees of action in other prefecture-level cities. To summarize, health education, entry-exit epidemic prevention and control can be strengthened. This study may be beneficial to carry out targeted, hierarchical and focused prevention and control of typhoid fever and paratyphoid fever, and provide scientific reference for related theoretical research.

PurposeLower urinary symptoms (LUTS) may persist in a proportion of patients with benign prostatic hyperplasia (BPH) following transurethral resection of prostate (TURP), which is a major cause of reduced quality-of-life. We aimed to investigate the effect of frailty on LUTS in patients with BPH treated with TURP.MethodsWe longitudinally evaluated LUTS and health-related quality-of-life (HRQOL) in patients with BPH treated with TURP from February 2019 and January 2022 using International Prostate Symptom Score (IPSS) and Short Form-8 (SF-8), respectively. Patients were divided into frail and non-frail groups according to the Fried phenotype (FP). The primary purpose was comparing the outcomes of LUTS and HRQOL between two groups. Secondary purposes were investigating the frailty as a preoperative predictor of postoperative adverse LUTS outcomes following TURP using logistic regression analysis. A 1:2 propensity score matching (PSM) was performed to reduce the effects of selection bias and potential confounders.ResultsOf the 567 patients enrolled, 495 (87.3%) patients were non-frail (FP = 0–2), and the remaining 72 (12.7%) patients were classified into the frail group. There were no significant differences in body mass index (BMI), urine white blood cell (UWBC), creatinine, prostate-specific antigen (PSA) and prostate volume in both groups at baseline (all p > 0.05). However, patients with frailty were older, higher comorbidity rates, lower peak flow rates and lower HRQOL. In the frail group, although LUTS and HRQOL at 6 months following TURP improved significantly compared to those at baseline, it did not show a significant improvement compared with the non-frail group (both p p ConclusionIn patients with frail or non-frail, TURP for BPH provides overall good results. However, frail individuals are at higher risk of postoperative adverse LUTS outcomes. Frailty has the potential to be a strong objective tool for risk stratification and should be considered during the perioperative evaluation.

A forearc environment is usually characterised by a relatively low geothermal gradient and hence little magmatic activity occurs. However, S-type granites were discovered within the forearc accretionary complex of the East Kunlun Orogenic Belt. The S-type granites intruded into an upper amphiolite facies partially migmatitic crystalline basement in form of dikes and sills at ca. 440 Ma which corresponds to the transition of the Proto-Tethyan to the Paleo-Tethyan realm in the northern Tibetan Plateau. The observed granites contain either garnet + biotite + muscovite or garnet + muscovite: (1) muscovite granite is strongly peraluminous with an aluminous saturation index (ASI) of more than 1.1 (ASI = molar [Al2O3/(Na2O+K2O+CaO]) and has high-K calc-alkaline characteristics, low Sr/Y (1.9–16.1) and LaN/YbN (1.85–13.2) ratios. (2) Two-mica granite is moderately peraluminous (ASI = 1.02–1.09), has high Ca and low K contents as well as high Sr/Y (16.8–67.7) and LaN/YbN(10.9–33.3) ratios. Other trace element contents and their ratios also show striking differences with high Sr (207–324 ppm) content and CaO/Na2O (0.47–0.96) ratio, and a low Rb/Sr (0.04–0.32) ratio for two-mica granite, but low Sr (63–126 ppm) content and CaO/Na2O (0.08–0.20) ratio, and a high Rb/Sr (0.56–2.53) ratio for muscovite granite. The observed differences are due to different protolith chemistries and melting mechanisms. Based on melting experiments of metasedimentary rocks (Patiño Douce and Harris, 1998), muscovite granite was most likely produced by dehydration melting of a metapelitic source and the two-mica granite by H2O-fluxed melting of a metagreywacke. Zircon Hf isotopes of the two S-type granites have εHf(440 Ma) values of -6.85 to +12.02 indicating the involvement of a mantle-derived magma which probably triggered the anatexis of supracrustal rocks deposited in a forarc regime. Coveal adakites with a younging westward trend as well as mafic rocks have been reported in this accretionary complex, which together with anatexis and metamorphism of accreted material support the occurrence of a slab window beneath the forearc accretionary complex of the East Kunlun Orogenic Belt during subduction of the Tethyan oceanic slab. Background Certain bacterial infectious diseases are categorized as notifiable infectious diseases in China. Understanding the time-varying epidemiology of bacterial infections diseases can provide scientific evidence to inform prevention and control measures. Methods Yearly incidence data for all 17 major notifiable bacterial infectious diseases (BIDs) at the province level were obtained from the National Notifiable Infectious Disease Reporting Information System in China between 2004 and 2019. Of them 16 BIDs are divided into four categories, respiratory transmitted diseases (RTDs, 6 diseases), direct contact/fecal-oral transmitted diseases (DCFTDs, 3 diseases), blood-borne/sexually transmitted diseases (BSTDs, 2 diseases), and zoonotic and vector-borne diseases (ZVDs, 5 diseases), and neonatal tetanus is excluded in the analysis. We characterized the demographic, temporal, and geographical features of the BIDs and examined their changing trends using a joinpoint regression analysis. Results During 2004‒2019, 28 779 thousand cases of BIDs were reported, with an annualized incidence rate of 134.00 per 100 000. RTDs were the most commonly reported BIDs, accounting for 57.02% of the cases (16 410 639/28 779 000). Average annual percent changes (AAPC) in incidence were − 1.98% for RTDs, − 11.66% for DCFTDs, 4.74% for BSTDs, and 4.46% for ZVDs. Females had a higher incidence of syphilis than males, and other BIDs were more commonly reported in males. Among 0-5-year-olds, the diseases with the largest increases in incidence were pertussis (15.17% AAPC) and scarlet fever (12.05%). Children and students had the highest incidence rates of scarlet fever, pertussis, meningococcal meningitis, and bacillary dysentery. Northwest China had the highest incidence of RTDs, while South and East China had the highest incidences of BSTDs. Laboratory confirmation of BIDs increased from 43.80 to 64.04% during the study period. Conclusions RTDs and DCFTDs decreased from 2004 to 2019 in China, while BSTDs and ZVDs increased during the same period. Great attention should be paid to BSTDs and ZVDs, active surveillance should be strengthened, and timely control measures should be adopted to reduce the incidence.

Background: Previous studies haveverified the dysfunction of mesenchymal stem cells (MSCs) for immunoregulation in acquired aplastic anemia (AA) patients. Exosomes derived from MSCs can partially substitute MSCs acting as immune regulator. Dysfunction of exosomes (Exos)derived from AA-MSC (AA-Exos) may play a key role in immunologic dissonance. Method: In this study, CD3+ T cells were collected and cocultured with AA-Exos and exosomes derived from MSC of healthy donors(HD-Exos). The proliferation, differentiation and activation of CD3+ T cells were detected to compare the immunosuppressive effects between AA-Exos and HD-Exos. An immune-mediated murine model of AA was structured to compare the therapeutic effect of AA-Exos and HD-Exos. Furthermore, total RNA including miRNA from exosomes we purified and total RNA of CD3+ T cells were extracted for RNA-seq in order to construct the miRNA–mRNA network for interactions and functional analysis. Results: AA-Exos had impaired inhibition effects on CD3+ T cells in terms of cell proliferation, activation and differentiation compared with exosomes from HD-Exos. HD-Exos other than AA-Exos can rescued the AA mice. Importantly, we found some differentially expressed miRNA involved in immune response, such as miR-199, miR-128 and miR-486. The Gene Ontology analysis of differentially expressed genes (DEGs) revealed involvement of various cellular processes, such as lymphocyte chemotaxis, lymphocyte migration and response to interferon-gamma, etc. The Kyoto Encyclopedia of Genes and Genomes analysis illustrated upregulation of critical pathways associated with T cell function after co-culturing with AA-Exos compared with HD-Exos, such as graft-versus-host disease, Th17 cell differentiation, and JAK-STAT signaling pathway, A miRNA–mRNA network was established to visualize the interaction between them. Conclusion: In summary, AA-Exos had impaired immunosuppressive effect on T cells, less ability to rescue AA mice and differently expressed miRNA profile, which might partly account for the pathogenesis of AA as well as provide a new target of AA treatment.

The objective of this study is to develop a data-driven machine learning based tool to estimate the FPSO topsides weight. The data were collected from public sources including IHS, news and magazines, covering world-wide active FPSO geographic locations, topsides weights, and their production throughput. One of the challenges is that the size of the dataset is less than 200 data points, largely due to the limited total number of FPSOs worldwide. Another challenge is that there are missing values for gas production, as such, imputation of missing values becomes necessary. In this study, data imputation was conducted by incorporating geographic information and physics guided feature engineering, through which the imputation is more accurate compared to simple imputers. For machine learning algorithms, polynomial regression was first evaluated as the baseline model and various machine learning models were built and compared with the baseline, such as Gaussian process regressor, random forest, neural network, and natural gradient boosting, with the purpose of identifying the most accurate one. To solve the overfitting issue caused by the small size of the dataset, several strategies have been investigated and compared, such as k-fold cross validation, regularization and extensive hyper-parameter tuning via Bayesian optimization algorithm based on the Hyperopt library. Among all the machine learning models, it is found that the natural gradient boosting method is the best performer with a mean absolute percentage error (MAPE) of 24% on the blind testing data, which is 35% lower than the baseline model. Shapley Additive exPlanations (SHAP) analysis was also implemented for model interpretation and gas production was found to be the most influential feature. The trained gradient boosting model was deployed to an internal web application in which users could get a quick estimation of FPSO topsides weight by providing three features: gas production, oil production and water depth. The 2D and 3D cross plots with historic data and predicted value are also provided in the web-app for better results visualizations. The novelty of this paper is to develop a data-driven machine learning tool for FPSO topsides weight estimation on an early stage of a project, which can serve as an independent alternative to the traditional empirical based approaches to help pre-design the facilities and estimate the cost. In the back-end, the best machine learning model was identified, along with the best imputation strategy based on a physics guided feature engineering approach. In the front-end, a web application was developed for an interactive estimation of FPSO topsides weight. With continuous enrichment and validation of the collected data, the machine learning approach can serve as a trustworthy fast and early estimation for FPSO topsides weight.

Background Sclerotherapy is an effective treatment for CLMs of the head and neck. However, Management of intra-abdominal cystic lymphatic malformations(CLMs) remains debates. The purpose of this study was to examine safety and efficacy of laparoscopic assisted sclerotherapy for intra-abdominal CLMs in pediatric patients. Methods We retrospectively reviewed the patients treated with laparoscopic assisted sclerotherapy for intra-abdominal CLMs at a single tertiary medical center between July 2020 and July 2022.The basic information and clinical data of maximal diameter, dose, reduction of lesion’s volume and complications were analyzed. The efficacy was assessed by comparing the pre-operative and post-operative cross-sectional images. Results 7 patients were included in this study and underwent a total of 9 sclerotherapy sessions. 4 patients were symptomatic and majored in abdominal pain. The primary sclerotherapy did not response in better clinical outcome in 2 patients due to intra-cystic hemorrhage and additional procedure were performed. Lesions in 6 patients reduced ≥90% in volume which met the complete response criteria. No further complications were recorded during the follow-up. Conclusions Laparoscopic assisted sclerotherapy is a safe and effective method of managing intra-abdominal CLMs. Laparoscopy could localize the origin of lesions and provides more accuracy and stabilization during the injection procedure.

Myeloid-derived suppressor cells (MDSC) are a group of heterogeneous immature myeloid cells and display immunosuppressive function. In this study, MDSC populations were evaluated in acquired aplastic anemia (AA) (n=65) in which aberrant immune mechanisms contributed to bone marrow destruction. Our data demonstrate that both the proportion and immunosuppressive function of MDSC are impaired in AA patients. Decreased percentage of MDSC, especially monocytic MDSC, in the blood of AA patients (n=15) is positively correlated with the frequency of T-regulatory cells, bone marrow level of WT1 and decreased plasma level of arginase-1. RNA sequencing analyses reveal that multiple pathways including DNA damage, interleukin 4, apoptosis, and Jak kinase singnal transducer and activator of transcription are upregulated, whereas transcription, IL-6, IL-18, glycolysis, transforming growth factor and reactive oxygen species are downregulated in MDSC of AA (n=4), compared with that of healthy donors (n=3). These data suggest that AA MDSC are defective. Administration of rapamycin significantly increases the absolute number of MDSC and levels of intracellular enzymes, including arginase-1 and inducible nitric-oxide synthase. Moreover, rapamycin inhibits MDSC from differentiating into mature myeloid cells. These findings reveal that impaired MDSC are involved in the immunopathogenesis of AA. Pharmacologically targeting of MDSC by rapamycin might provide a promising therapeutic strategy for AA.

It is known that permanent magnet vernier machine has multiple flux field harmonics which can produce stable torque. However, some flux field harmonics would create torque that undermines the torque output. Such as the 6slots26poles split-tooth PMVM here, of which modulated flux field outputs torque while fundamental flux field undermines torque output. In this paper, a 26poles PMVM with coding-shaped tooth is proposed. The coding-shaped tooth can introduce permeance harmonics with specific phase and amplitude, thus modulating flux fields that all output torque. Air-gap permeance harmonics of the proposed and the regular split-tooth PMVM are obtained via FEA. Based on MMF-permeance model, flux fields of two machines and their contributions to average torque are investigated, proving that the proposed machine has multiple flux fields that all output torque. By optimizing geometry of coding-shaped tooth, the proposed machine obtains largest average torque and smallest torque ripple simultaneously. Electromagnetic performances of the optimized machines are studied in semi-analytical way and FEA. It is proved that with same overall size, PM usage and copper loss, the proposed machine achieves 30% larger torque density, 86% smaller cogging torque and 62% lower torque ripple than its counterpart. Finally, a 16Nm (rated)prototype is built and tested.

Permanent magnet vernier machines (PMVMs) have drawn more and more attention owing to their high torque density, whereas the power factor of which are lower than that of regular PM synchronous machines (PMSMs). At present, the analysis of PMVMs are mainly from the perspective of PM rotors, and the research results suggest that there seems a relationship between torque density and power factor. However, from PM rotor perspective, the relationship can’t be explained in principle. In this paper, the performance of PMVMs are investigated from the viewpoint of armature fields, where both the high torque density and low power factor have been clarified. It is indicated that the high torque density of PMVMs is attributed to high amplitudes and large variation gradient of armature working harmonic which has the same pole-pair of PMs. While the power factor suffers from the lower proportion of armature working harmonic to total harmonics. Meanwhile, the negative coupling between torque capability and power factor is first revealed from the armature field perspective. Finally, two prototypes of 18/14/4 and 15/14/1 with same 14-pole-pair PM rotors and different armature fields are built and tested, and the analysis results match well with FEA and experimental results.

The failure probability and strength of jointed and fractured rock mass under water pressure action place important constraints on the safety and stability of rock mass engineering. In this study, two strength calculation models for rock mass joint rupture failure and shear slip failure are established according to different relative sizes and working conditions, and the effects of fractal dimension, water pressure, and number of cracks on the strength and failure probability of rock mass are analyzed. The most unfavorable dip angle under shear-slip conditions is calculated. The proposed models accurately reflect the failure probability and strength of jointed rock mass, and different strength calculation methods should be selected according to the specific working conditions. The effect of water pressure on the strength of jointed rock mass is sensitive to the failure mode. Reasonable drainage should be carried out according to the different failure modes when constructing tunnels with abundant groundwater, and certain drainage or water plugging construction schemes are not necessarily applicable to all cases. The dip angle at which joint fissures are most prone to shear slip is given as π/4 + φ/2, where φ is the jointed rock mass strength parameter. An increase in the fractal dimensions, external stress, and number of fractures will enhance the rock mass failure probability regardless of failure mode. Controlled blasting and advanced grouting reinforcement measures are recommended when a tunnel traverses through a joint fissure zone to reduce the number of cracks generated in the surrounding rock and thus improve its strength and stability.
