PURPOSE: To identify the value of a computed tomography (CT)-based radiomics model to predict probability of early recurrence (ER) in patients diagnosed with laryngeal squamous cell carcinoma (LSCC) after surgery. MATERIALS AND METHOD: Pre-operative CT scans of 140 LSCC patients treated by surgery are reviewed and selected. These patients are randomly split into the training set (n = 97) and test set (n = 43). The regions of interest of each patient were delineated manually by two senior radiologists. Radiomics features are extracted from CT images acquired in non-enhanced, arterial, and venous phases. Variance threshold, one-way ANOVA, and least absolute shrinkage and selection operator algorithm are used for feature selection. Then, radiomics models are built with five algorithms namely, k-nearest neighbor (KNN), logistic regression (LR), linear support vector machine (LSVM), radial basis function SVM (RSVM), and polynomial SVM (PSVM). Clinical factors are selected using univariate and multivariate logistic regressions. Last, a radiomics nomogram incorporating the radiomics signature and clinical factors is built to predict ER and its efficiency is evaluated by receiver operating characteristic (ROC) curve and calibration curve. Decision curve analysis (DCA) is also used to evaluate clinical usefulness. RESULTS: Four features are remarkably associated with ER in patients with LSCC. Applying to test set, the area under the ROC curves (AUCs) of KNN, LR, LSVM, RSVM, and PSVM are 0.936, 0.855, 0.845, 0.829, and 0.794, respectively. The radiomics nomogram shows better discrimination (with AUC: 0.939, 95% CI: 0.867–0.989) than the best radiomics model and the clinical model. Predicted and actual ERs in the calibration curves are in good agreement. DCA shows that the radiomics nomogram is clinically useful. CONCLUSION: The radiomics nomogram, as a noninvasive prediction tool, exhibits favorable performance for ER prediction of LSCC patients after surgery.

For higher vocational colleges, skills competition is an important indicator of professional personnel training quality. In this paper, the architectural interior design professional skills competition in Chongqing Energy Vocational College is taken as an example, the many years of teaching and competition experience are taken into account, the problems existing in the cultivation of architectural interior design professionals in higher vocational colleges are analyzed, and corresponding solutions are proposed.

BACKGROUND: In recent years, inductively coupled plasma mass spectrometry (ICP-MS) has been widely used in the field of molecular biology because of its unique advantages. Anthrax is a widespread and long-standing infectious disease, which affects and restricts people’s work and life seriously. OBJECTIVE: The study goal is to develop a new method for the detection of anthrax. METHODS: A rapid, sensitive and accurate method for the detection of anthrax characteristic DNA was proposed by combing gold nanoparticles (AuNPs) and inductively coupled plasma mass spectrometry. RESULTS: The linear range of this method is 100–2500 pmol/L and the limit of detection of 16.61 pmol/L. CONCLUSION: The proposed method has numerous advantages, including simplicity of operation, high sensitivity, and specificity, which provides a new idea for the detection of anthrax. Importantly, this methodology has good potential for the detection of other biological substances such as bacteria and viruses by changing the modification sequence on the nanoparticle probe.

Viruses are important components of the human body. Growing evidence suggests that they are engaged in the physiology and disease status of the host. Even though the vaginal microbiome is involved in human papillomavirus (HPV) infection and cervical cancer (CC) progression, little is known about the role of the vaginal virome. In this pilot exploratory study, using unbiased viral metagenomics, we aim to investigate the vaginal eukaryotic virome in women with different levels of cervical lesions, and examine their associations with different cervical disease status. An altered eukaryotic virome was observed in women with different levels of lesions and Lactobacillus profiles. Anelloviruses and papillomaviruses are the most commonly detected eukaryotic viruses of the vaginal virome. Higher abundance and richness of anelloviruses and papillomaviruses were associated with low-grade squamous intraepithelial lesion (LSIL) and CC. Besides, higher anellovirus abundance was also associated with lactobacillus-depleted microbiome profiles and bacterial community state (CST) type IV. Furthermore, increased correlations between Anelloviridae and Papillomaviridae occurred in the women with increased cervical disease severity level from LSIL to CC. These data suggest underlying interactions between different microbes as well as the host physiology. Higher abundance and diversity of both anelloviruses and papillomaviruses shared by LSIL and CC suggest that anellovirus may be used as a potential adjunct biomarker to predict the risk of HPV persistent infection and/or CC. Future studies need to focus on the clinical relevance of anellovirus abundance with cervical disease status, and the evaluation of their potential as a new adjunct biomarker for the prediction and prognoses of CC.

LZTFL1 is a tumor suppressor located in chromosomal region 3p21.3 that is deleted frequently and early in various cancer types including the kidney cancer. However, its role in kidney tumorigenesis remains unknown. Here we hypothesized a tumor suppressive function of LZTFL1 in clear cell renal cell carcinoma (ccRCC) and its mechanism of action based on extensive bioinformatics analysis of patients’ tumor data and validated it using both gain- and loss-functional studies in kidney tumor cell lines and patient-derive xenograft (PDX) model systems. Our studies indicated that LZTFL1 inhibits kidney tumor cell proliferation by destabilizing AKT through ZNRF1-mediated ubiquitin proteosome pathway and inducing cell cycle arrest at G1. Clinically, we found that LZTFL1 is frequently deleted in ccRCC. Downregulation of LZTFL1 is associated with a poor ccRCC outcome and may be used as prognostic maker. Furthermore, we show that overexpression of LZTFL1 in PDX via lentiviral delivery suppressed PDX growth, suggesting that re-expression of LZTFL1 may be a therapeutic strategy against ccRCC.

This study aimed to investigate the effects of feed containing yak meat on scalded rats’ body condition and wound healing.Methods: Adopting a two-factor factorial design, the growth performance, food intake, body weight, and Lee’s index of rats were measured. The wound conditions of scalded rats with different feeds (basic, basic + yak meat, and basic + yellow beef) were observed at different periods, and their wounds’ hematoxylin and eosin (H&E) staining states were detected. The proliferating cell nuclear antigen (PCNA)-positive cells and apoptosis were analyzed to evaluate the effects of feed on the wound healing of scalded rats.Results: The feed intake was the highest in the yellow beef feed group and the lowest in the yak meat feed group. The body weight was the highest in the yak meat feed group and the lowest in the yellow beef feed group. Furthermore, 45 days after scalding, the obesity index in the yak beef feed group was the closest to that of the rats before scalding. The wound recovery of the rats in the yak meat feed group was the best at 30 days, and the H&E staining results also proved that the recovery effect of the scalded rats in the yak meat feed group was better than other two groups. According to the results of PCNA and apoptosis, the yak meat feed group had lower positive cell rate and faster wound healing.Conclusion: The rats in the yak meat feed group recovered better than those in the other groups, and the yak beef feed had the best effect on the wound healing of the scalded rats.Keywords: scalded rats, yak meat feed, yellow beef feed, basic feed, wound healing

Phosphorus (P) is a macronutrient necessary for plant growth and development. Inorganic phosphate (Pi) deficiency modulates the signaling pathway of the phytohormone jasmonate in Arabidopsis thaliana, but the underlying molecular mechanism currently remains elusive. Here, we confirmed that jasmonate signaling was enhanced under low Pi conditions, and the CORONATINE INSENSITIVE1 (COI1)-mediated pathway is critical for this process. A mechanistic investigation revealed that several JASMONATE ZIM-DOMAIN (JAZ) repressors physically interacted with the Pi signaling-related core transcription factors PHOSPHATE STARVATION RESPONSE1 (PHR1), PHR1-LIKE2 (PHL2), and PHL3. Phenotypic analyses showed that PHR1 and its homologs positively regulated jasmonate-induced anthocyanin accumulation and root growth inhibition. PHR1 stimulated the expression of several jasmonate-responsive genes, whereas JAZ proteins interfered with its transcriptional function. Furthermore, PHR1 physically associated with the basic helix–loop–helix (bHLH) transcription factors MYC2, MYC3, and MYC4. Genetic analyses and biochemical assays indicated that PHR1 and MYC2 synergistically increased the transcription of downstream jasmonate-responsive genes and enhanced the responses to jasmonate. Collectively, our study reveals the crucial regulatory roles of PHR1 in modulating jasmonate responses and provides a mechanistic understanding of how PHR1 functions together with JAZ and MYC2 to maintain the appropriate level of jasmonate signaling under conditions of Pi deficiency.

The rise in mortgage defaults in the banking industry highlights the connections between financial and macroeconomic shocks as well as the risk of financial instability created by credit market friction. The percentage of non-performing loans in the banking industry is assessed in this study in connection to the effects of the global economy. In order to do a regression analysis, three American banks are chosen, and the variables GDP growth, unemployment rate, real interest rate, and CPI are taken into account. The study found a significant correlation between the percentage of non-performing loans and the unemployment rate (NPL). The results of Wells Fargo indicates that there is no obvious link between GDP and NPL. There is an association between GDP expansion and the percentage of non-performing loans for Citi Bank and Bank of America. Real interest rates and NPL do not significantly affect one another. Therefore, even though some factors might not affect such probabilities, changes in the economic climate may have an impact on banks' likelihood of non-performing loans.

Background There remain concerns regarding the technical feasibility of endoscopic resection for large gastrointestinal stromal tumors (GISTs), mainly relating to the risk of tumor rupture and the adequacy of the resection margins. This study aimed to evaluate the feasibility and therapeutic outcomes of the newly developed no-touch endoscopic full-thickness resection (NT-EFTR) technique for GISTs. Methods In this retrospective study, 92 patients with gastric GISTs undergoing NT-EFTR were included. Clinicopathological, endoscopic, and follow-up data were collected and analyzed. Results The median tumor size was 2.5 cm and en bloc resection was achieved in all patients with negative surgical margins. The median time of the NT-EFTR procedure was 59.5 minutes. Large tumors (> 3.0 cm), extraluminal tumor growth pattern, and large gastric defects were significant contributors to long operative times. Patients were discharged within 4 days postoperatively. During follow-up, all patients were free from local recurrence and distant metastasis. Conclusions NT-EFTR was a feasible method for the resection of gastric GISTs and can be expected to achieve complete radical resection. Large tumors with extraluminal growth and large gastric defects impact procedural difficulty.

In this paper, two group experiments were conducted to study the effect of the main stage flow velocity on thermoacoustic instability and flame macrostructure. The experiments were carried out under atmosphere condition, and namely the pilot stage flame mode and the stratified swirl flame mode. The experimental results show that in the pilot stage flame mode, the thermoacoustic oscillation amplitude remains constant (around 1000 Pa) with the decrease of the main stage flow velocity. But when the velocity is zero (without main stage air), the thermoacoustic instability disappeared. In the stratified swirl flame mode, the amplitude of thermoacoustic instability is the largest (around 300 Pa) when the main stage flow velocity is 6.2 m/s, and the amplitude is slightly smaller in other working conditions. The time-averaged flame shape under the two flame modes is recorded and discussed. This paper highlights the effect of interactions between the pilot stage flame and main stage air or flame on thermoacoustic instability.

In cold and humid temperate forests, low temperature, late frost and frequent freeze–thaw cycles are the main factors limiting tree growth and survival. Ring- and diffuse-porous tree species differing in xylem anatomy coexist in these forests, but their divergent adaptations to these factors have been poorly explored. To fill this knowledge gap, we compared four ring-porous and four diffuse-porous tree species from the same temperate forest in Northeast China by quantifying their leaf and stem functional traits, their stem growth rates using tree ring analysis and their resistance to cold represented by upper altitude species distribution borders from survey data. We found that the ring-porous trees were characterized by traits related to more rapid water transport, carbon gain and stem growth rates than those of the diffuse-porous species. Compared with the diffuse-porous species, the ring-porous species had a significantly higher shoot hydraulic conductance (Ks-shoot, 0.52 vs 1.03 kg m−1 s−1 MPa−1), leaf photosynthetic rate (An, 11.28 vs 15.83 μmol m−2 s−1), relative basal area increment (BAIr, 2.28 vs 0.72 cm year−1) and stem biomass increment (M, 0.34 vs 0.09 kg year−1 m−1). However, the observed upper elevational distribution limit of the diffuse-porous species was higher than that of the ring-porous species and was associated with higher values of conservative traits, such as longer leaf life span (R2 = 0.52). Correspondingly, BAIr and M showed significant positive correlations with acquisitive traits such as Ks-shoot (R2 = 0.77) and leaf photosynthetic rate (R2 = 0.73) across the eight species, with the ring-porous species occurring at the fast-acquisitive side of the spectrum and the diffuse-porous species located on the opposite side. The observed contrasts in functional traits between the two species groups improved our understanding of their differences in terms of growth strategies and adaptive capabilities in the cold, humid temperate forests.

ABSCISIC ACID-INSENSITIVE3 (ABI3) and ABI5 are 2 crucial transcription factors in abscisic acid (ABA) signaling, and their homeostasis at the protein level plays a decisive role in seed germination and subsequent seedling growth. Here, we found that PLANT U-BOX 8 (PUB8), a U-box E3 ubiquitin ligase, physically interacts with ABI3 and ABI5 and negatively regulates ABA responses during early Arabidopsis (Arabidopsis thaliana) seedling growth. Loss-of-function pub8 mutants were hypersensitive to ABA-inhibited cotyledon greening, while lines overexpressing PUB8 with low levels of ABI5 protein abundance were insensitive to ABA. Genetic analyses showed that ABI3 and ABI5 were required for the ABA-sensitive phenotype of pub8, indicating that PUB8 functions upstream of ABI3 and ABI5 to regulate ABA responses. Biochemical analyses showed that PUB8 can associate with ABI3 and ABI5 for degradation through the ubiquitin-mediated 26S proteasome pathway. Correspondingly, loss-of-function of PUB8 led to enhanced ABI3 and ABI5 stability, while overexpression of PUB8 impaired accumulation of ABI3 and ABI5 in planta. Further phenotypic analysis indicated that PUB8 compromised the function of ABI5 during early seedling growth. Taken together, our results reveal the regulatory role of PUB8 in modulating the early seedling growth by controlling the homeostasis of ABI3 and ABI5.

Accumulating evidence shows that PD-L1 expression on dendritic cells (DC) is critical for cancer immunotherapy and that Porphyromonas gingivalis (Pg) colonization aggravates the progression of upper gastrointestinal cancers. However, the effects of Pg infection on PD-L1 expression on DCs and related immune consequences in the infection milieu of oral cancer remain unexplored. Here, we found that Pg infection robustly enhanced PD-L1 expression on DCs in a gingipain-dependent manner in cultured cell and systemic infection assays. Pg infection suppressed antigen-specific CD8+ T cells through upregulation of PD-L1 expression on ovalbumin (OVA)-pulsed DCs. This suppression was manifested by decreased IFNγ, perforin, granzyme B, and CD107a. Further analysis showed that Pg drastically reduced CD8+ T cells’ ability to lyse OVA-pulsed target cells. Additionally, Pg infection increased the phosphorylation of Akt and STAT3, leading to a significant increase in PD-L1 expression. This was substantiated by using siRNA, overexpression plasmids, and pharmacologic inhibitors. Consistent with the in vitro observations, in a syngeneic mouse oral cancer model, Pg infection significantly enhanced PD-L1 expression on DCs from intratumoral tissues and cervical lymph nodes and exacerbated oral cancer progression, whereas a Pg lysine-specific, gingipain-defective mutant failed to do so. These influences of Pg were largely diminished when tumor cells were pretreated with antibiotics or a STAT3 inhibitor. Therefore, we demonstrated that Pg infection upregulates PD-L1 expression on DCs through Akt-STAT3 signaling, suppresses CD8+ T-cell cytotoxicity, and aggravates oral cancer growth, suggesting targeting Pg, and/or its mediated signaling, could be a therapeutic strategy to improve the efficacy of checkpoint blockade immunotherapy.

Reversible Data Hiding in Encrypted Image (RDHEI) schemes may redistribute the data hiding procedure to other parties and can preserve privacy of the cover image. Recently, cloud computing technology has led to the rapid growth of networked media, and many multimedia rights are owned by multiple parties, such as a film's producer and multiple distributors. Thus, the data hiding task could be distributed to multiple distributed servers. Multi-party data hiding has become an important demand for networked media. In addition, it is essential to preserve multi-server and multi-message privacy and data integrity. However, most of the RDHEI schemes involve only one data hider. That inspired us to design the secure multi-party embedding over distributed encrypted-image servers as a solution for multi-party RDHEI applications. In this article, we propose a novel Reversible Data Hiding over Distributed Encrypted-Image Servers (RDH-DES) based on secret sharing. The Chinese remainder theorem, secret sharing, and block-level scrambling are developed as a lightweight cryptography to generate the encrypted image shares. These shares are distributed to different image servers and are used to embed secret data in the proposed framework. The marked encrypted image can be constructed through the marked encrypted shares from different parties, and the decryption and extraction can be completed by the receiver. The experimental results and theoretical analysis have demonstrated that the proposed scheme is secure and effective.
