Epigenetics play a role in the development of hypertension. However, the precise mechanisms of how histone modification in renal tubule cells regulates blood pressure is largely undiscovered. In this study, we investigated the role of renal Kdm6a in salt-sensitive hypertension using a kidney tubular cell-specific Kdm6a gene knockout (Kdm6a cKO) mouse model. First, we demonstrated that knockout of Kdm6a in the renal tubular cells resulted in an increase of H3K27me3 associated with the development of hypertension in both male and female Kdm6a cKO mice. To explore the mechanism of H3K27me3-mediated hypertension, we performed a ChIP seq to identify the target genes of H3K27me3 in the renal tubular cells. Data showed that H3K27me3 regulated several physiological pathways in the renal tubular cells, including sodium transport in proximal convoluted tubule (PCT), and blood pressure regulation in distal convoluted tubule (DCT). Indeed, we found that the expressions of NCC, NKCC2, and AQP2 were upregulated in kidneys of Kdm6a cKO mice. Furthermore, ChIP seq data reveals that FBLN2, an extracellular matrix protein, appears to be a target of H3K27me3, which was downregulated in the kidney of Kdm6a cKO mice determined by real-time PCR, Western, and IHC. Notably, we demonstrated that FBLN2 abated high salt-induced hypertension through downregulation of NCC, NKCC2, and AQP2 in the tubular cells of Kdm6a cKO mice. Interestingly, pre-treatment of FBLN2 prevented salt-sensitive hypertension in these mice. Additionally, we have provided evidence that FBLN2 may regulate blood pressure through mediating the NCC cell membrane trafficking. Using a super high-resolution microscopy, we demonstrated that FBLN2 reduced levels of cell surface NCC in DCT cells cultured in hypotonic medium. Here, we have first demonstrated that FBLN2 may be a target gene of H3K27me3 in epigenetic-mediated hypertension. FBLN2 treatment may prevent and treat salt-sensitive hypertension. Aspergillus genus is a key component in fermentation and food processing. However, sterigmatocystin (STE)-a mycotoxin produced by several species of Aspergillus-limits the use of some Aspergillus species (such as Aspergillus versicolor, Aspergillus inflatus, and Aspergillus parasiticus) because of its toxicity and carcinogenicity. Here, we engineered an STE-free Aspergillus versicolor strain based on genome mining techniques. We sequenced and assembled the Aspergillus versicolor D5 genome (34.52 Mb), in which we identified 16 scaffolds and 54 biosynthetic gene clusters (BGCs). We silenced cytochrome P450 coding genes STC17 and STC27 by insertional inactivation. The production of STE in the Δstc17 mutant strain was increased by 282% but no STE was detected in the Δstc27 mutant. Metabolites of Δstc27 mutant exhibited growth-promoting effect on plants. Our study makes significant progress in improving the application of some Aspergillus strains by restricting their production of toxic and carcinogenic compounds.

Tobacco bacterial wilt caused by Ralstonia solanacearum is one of the most devastating diseases. Microbial keystone taxa were proposed as promising targets in plant disease control. In this study, we obtained an antagonistic Bacillus isolate EM-1 from bacterial wilt-suppressive soil, and it was considered rhizosphere-resident bacteria based on high (100%) 16S rRNA gene similarity to sequences derived from high-throughput amplicon sequencing. According to 16S rRNA gene sequencing and MLSA, strain EM-1 was identified as Bacillus velezensis. This strain could inhibit the growth of R. solanacearum, reduce the colonization of R. solanacearum in tobacco roots, and decrease the incidence of bacterial wilt disease. In addition, strain EM-1 also showed a strong inhibitory effect on other phytopathogens, such as Alternaria alternata and Phytophthora nicotianae, indicating a wide antagonistic spectrum. The antimicrobial ability of EM-1 can be attributed to its volatile, lipopeptide and polyketide metabolites. Iturin A (C14, C15, and C16) was the main lipopeptide, and macrolactin A and macrolactin W were the main polyketides in the fermentation broth of EM-1, while heptanone and its derivatives were dominant among the volatile organic compounds. Among them, heptanones and macrolactins, but not iturins, might be the main potential antibacterial substances. Complete genome sequencing was performed, and the biosynthetic gene clusters responsible for iturin A and macrolactin were identified. Moreover, strain EM-1 can also induce plant resistance by increasing the activity of CAT and PPO in tobacco. These results indicated that EM-1 can serve as a biocontrol Bacillus strain for tobacco bacterial wilt control. This study provides a better insight into the strategy of exploring biocontrol agent based on rhizosphere microbiome.

This study investigated the effects of adding biocontrol microbes on metabolites and pathogenic microorganisms during mushroom residue composting and the relationships of metabolite changes with microbes and material transformation. The results showed that the addition of Bacillus subtilis (BS) and Trichoderma harzianum (TH) with mushroom residue promoted the conversion of organic carbon and nitrogen. The abundance of pathogenic microbes was increased in biocontrol microbial treatments. BS or TH treatments increased the levels of amino acids, carbohydrates, and bacteriostatic alkaloid metabolites. Extracts from plants used in Chinese medicine can be good sources of fungicides for agricultural applications. In this study, we separated and identified antifungal compounds from four traditional Chinese medicine extracts and evaluated their antifungal activities in vitro and in vivo. In vitro, honokiol extracted from Artemisia argyi showed broad-spectrum antimicrobial and mycelial inhibitory activity with EC50 in the range 3.56 − 33.85 μg/mL against eight plant pathogens. q-PCR indicated that honokiol might induce cell cancerisation and inhibit cellular respiration, which provided significant insights into honokiol function in tobacco resistance to molecular mechanisms of the phytopathogenic fungus Phytophthora nicotianae. In vivo, honokiol significantly decreased the rate of fungal infection in eggplants, potatoes, grapes, cherry tomatoes, and cucumbers, and enhanced disease resistance in tobacco. Overall, our results indicate that honokiol has the potential to control a variety of fungal and oomycete diseases, and A. argyi could be a source of honokiol.

Fungi have been proved as promising and prolific sources of functional secondary metabolites with potent agricultural applications. In this study, 14 xanthone derivatives (1-14), including six new ones, versicones I-N (1-4, 7, 11), and a biogenetically related derivative (15), were isolated from the alga-derived fungus Aspergillus versicolor D5. Their structures were elucidated by comprehensive spectroscopic methods. Versicone L (4) exhibited a broad antifungal spectrum and prominent inhibitory effects on Botrytis cinerea at a minimum inhibitory concentration (MIC) of 152 μM, 7-fold stronger than that of the positive control, carbendazim (MIC = 1.05 × 103 μM). Dihydrosterigmatocystin (13) showed strong antifungal activity toward B. cinerea at MIC = 38.3 μM, almost 30-fold stronger than that of carbendazim. Meanwhile, 13 exhibited potent herbicidal activity toward Amaranthus retroflexus L. with an MIC of 24.5 μM, approximately 4-fold stronger than that of the positive control, glyphosate (MIC = 94.7 μM). Additionally, 13 also displayed remarkable activity against other weeds belonging to Amaranth sp. Analysis of the structure-herbicidal activity relationship indicated that the bifuranic ring played an important role in xanthone phytotoxicity and the presence of a double bond in the furan ring could decrease phytotoxicity. This study indicated that xanthones can be served as promising candidates for lead compounds of agrochemicals.

Bi2Te3-based materials have been widely utilized for commercial bulk thermoelectric (TE) devices. For the manufacture of miniaturized TE devices, the large-scale deposition of high-performance Bi2Te3-based thin films is crucial. However, it has not yet been effectively resolved. Herein, the Bi0.5Sb1.5Te3 films with different thicknesses were grown on sapphire substrates by controlling the co-sputtering time. We reveal that the film thickness has a significant impact on the electrical transport properties. Due to the mutual influence of electrical conductivity and Seebeck coefficient, there exists an optimal thickness with the maximum power factor is as high as 2900 μW m−1 K−2. Therefore, systematic research on the thickness-dependent TE characteristics of Bi2Te3-based films and the deposition of high-performance films will provide important information for the large-scale development of high-performance micro-TE devices.

KL (klotho) levels decline with age, which is an important mechanistic driver of aging. KL gene deficiency is associated with hypertension. The purpose of this study is to investigate the potential role of H3K27me3 (histone 3 lysine [K] 27 trimethylation) in the regulation of KL gene expression and examine the related molecular pathways that may drive kidney cell aging. Kidneys were collected from 6-month-old WT (wild type; young WT), 30-month-old WT (aged WT), and 6- (young) and 20-month-old (aged) KL mutant mice, respectively. We demonstrated that the H3K27me3 level was increased in kidneys of aged WT and KL mutant mice versus young WT mice. Elevation of H3K27me3 levels was likely due to downregulation of the H3K27 (histone H3 Lys 27)-specific demethylase JMJD3 (the Jumonji domain containing-3) in the aged kidneys. Inhibition of PRC2 (polycomb repressive complex C2; histone trimethyltransferase) decreased the H3K27me3 levels leading to an increase in the expression of KL in cultured primary renal tubule cells assessed by Western blot and KL promoter activity assays. The chromatin immunoprecipitation qPCR assay revealed that H3K27me3 was physically associated with the KL promoter region. Furthermore, aging impaired the SGK1 (serum- and glucocorticoid-induced protein kinase 1)/FOXO3a (the forkhead box class O 3a) signaling leading to upregulation of p53 and p16 (aging markers) in the kidney of aged WT mice. KL may regulate the SGK1/FOXO3 signaling, which was decreased due to KL deficiency. Thus, aging-associated downregulation of KL gene expression may be partly attributed to upregulation of H3K27me3 levels. Downregulation of KL may impair the SGK1/FOXO3 signaling contributing to kidney cell aging.

There is controversy regarding whether excess FGF23 causes left ventricular hypertrophy (LVH) directly through activation of fibroblast growth factor receptor 4 (FGFR4) in cardiomyocytes or indirectly through reductions in soluble Klotho (sK). We investigated the respective roles of myocardial FGFR4 and sKL in mediating FGF23-induced LVH using mouse genetic and pharmacological approaches. To investigate a direct role of myocardial FGFR4 in mediating the cardiotoxic effects of excess circulating FGF23, we administered rFGF23 to mice with cardiac-specific loss of FGFR4 (FGFR4 heart-cKO). We tested a model of sKL deficiency, hypertension and LVH created by the conditional deletion of FGFR1 in the renal distal tubule (FGFR1DT cKO mice). The cardioprotective effects of sKL in both mouse models was assessed by the systemic administration of recombinant sKL. We confirmed that FGF23 treatment activates PLCγ in the heart and induces LVH in the absence of membrane α-Klotho. Conditional deletion of FGFR4 in the myocardium prevented rFGF23-induced LVH in mice, establishing direct cardiotoxicity of FGF23 through activation of FGFR4. Recombinant sKL administration prevented LVH, but not HTN, in FGFR1DT cKO mice, consistent with direct cardioprotective effects. Co-administration of recombinant sKL with FGF23 in culture inhibited rFGF23-induced p-PLCγ signaling. Thus, FGF23 ability to include LVH represents a balance between FGF23 direct cardiac activation of FGFR4 and the modulating effects of circulating sKL to alter FGF23-dependent myocardial signaling pathways.

Microcapsules with toluene-di-isocyanate (TDI) as core and paraffin as shell for self-healing of concrete were prepared using melt condensation method, and the effects of preparation temperature, agitation rate and paraffin/TDI mass ratio on core fraction of microcapsules were studied. The size distribution and morphology of microcapsules were characterized by laser particle size analyzer and scanning electron microscopy (SEM). Components of the microcapsules were analyzed by Fourier transform infrared spectrometer (FTIR). Finally, the effect of microcapsules on self-healing ability of mortars was evaluated, which indicated that preparation temperature, agitation rate and paraffin/TDI mass ratio had significant effect on core fraction of microcapsules. The optimum microcapsules could be prepared with a paraffin/TDI mass ratio 1:2 and an agitation rate 600 rpm at 75 °C. The core fraction of microcapsules prepared with optimum parameters was 66.5%, and the particle size was between 30 and 300 µm, mainly concentrated on 90 μm. SEM showed that the microcapsules were regular spheres and the shell thickness was about 1/10 of the diameter. FTIR confirmed that the TDI was successfully encapsulated in the paraffin shell. Compared with the control mortar, compressive strength of the mortar with 3% microcapsules (by mass of cement) increased by 28.2%. The reserved ratio of compressive strength was 77.2% under 60% fc0 pre-load after 48 h self-healing. The cracks with a width of less 0.4 mm on the mortar were rapidly self-healed by the microcapsules in 6 h.

Phytophthora nicotianae is currently considered one of the most devastating oomycete plant pathogens, and its control frequently relies solely on the use of systemic fungicides. There is an urgent need to find environment-friendly control techniques. This study examined the chemical composition, inhibitory activity, and possible modes of action of the essential oil of Chrysanthemum indicum L. (EOC) flower heads against P. nicotianae. The EOC was obtained using hydrodistillation at a 0.15% yielded. It inhibited mycelial growth and spore germination of P. nicotianae at a minimum inhibitory concentration (MIC) of 200 μL/L, and exhibited fumigation effects (92.68% inhibition at 157.48 μL/L). Marked deformation of P. nicotianae mycelia included deformed tip enlargement, shrinkage, and rupture. Further, 55 and 47 compounds were identified using gas chromatography-mass spectrometry (GC-MS) and headspace solid-phase microextraction (HS-SPME) GC-MS analyses, representing 88.2% and 98.91% of the total EOC, respectively. Monoterpenes (25.77%) and sesquiterpenes (54.14%) were the major components identified using GC-MS, whereas monoterpenes were the main constituents in the HS-SPME GC-MS analysis. The higher proportions of sesquiterpenes and monoterpenes could be responsible for the inhibitory activity of EOC, which increased mycelia membrane permeability and the content of mycelial malondialdehyde (MDA) in a dose-dependent manner. Cell death also occurred. Thus, destruction of the cell wall and membrane might be two modes of action of EOC. Our results would be useful for the development of a new plant source of fungicide for P. nicotianae-induced disease.

Layered double hydroxides (LDHs) with different particle sizes (75 nm, 115 nm, 180 nm and 300 nm which were marked with LDHs-75, LDHs-115, LDHs-180 and LDHs-300) were characterized by scanning electron microscope, UV–vis reflection and absorption spectrophotometer and laser scatter meter. Furthermore, this paper investigated the influence of LDHs with different particle sizes on the rheological properties of bitumen before and after ultraviolet aging by temperature susceptibility test, dynamic shear rheometer test (DSR) and IR spectra test (FTIR). The researches reveal that LDHs-180 hold the strongest UV shielding performance. The temperature susceptibility of pristine bitumen and LDHs-75, LDHs-115, LDHs-180, LDHs-300 modified bitumen after aging decreased by 9.01%, 6.36%, 3.98%, 2.15% and 5.49%, respectively, which indicate LDHs-180 is more beneficial to weaken the decrease of bitumen temperature susceptibility after aging. Similarly, LDHs-180 modified bitumen show less variation in rheological properties than other bitumen samples after aging. This implies that LDHs-180 could mitigate the influences of UV aging on the rheological properties of bitumen more availably. In addition, the FTIR analysis further shows that the carbonyl and sulfoxide index of LDHs-180 modified bitumen is the smallest after aging, which indicates LDHs-180 is more helpful for improving bitumen anti-UV aging properties than that of LDHs-75, LDHs-115 and LDHs-300.

Lignosulfonate (LS) grafted layered double hydroxides (LDHs) were prepared through covalent bonding with diisocyanate (diphenyl-methane-diisocyanate (MDI), toluene-2,4-diisocyanate (TDI) or isophorone diisocyanate (IPDI)), and were characterized by Fourier transform infrared (FTIR) spectrometer, X-ray diffraction (XRD), scanning electron microscopy (SEM) and ultraviolet and visible (UV–vis) spectroscopy. The degree of grafting (Dg) and mass proportion (Mp) had also been calculated. Then LS grafted LDHs modified bitumen was investigated using high temperature storage stability, physical properties measurement and thin-layer chromatography with flameionization detection test. FTIR and XRD showed that LS was successfully grafted to LDHs via covalent linked by diisocyanate. SEM displayed that LS grafted LDHs represented a stable cross-linked structure and had weaker surface agglomeration compared to LDHs. Dg and Mp of LS grafted LDHs linked by MDI (LS-m-LDHs) respectively reached up to 71.68% and 2.53:1, representing excellent grafting effect. The UV–vis analyses displayed that LS grafted LDHs had better UV absorbing ability than LDHs, which had a combined effect of chemically absorbing and physically reflecting UV light. Storage stability test expressed that the compatibility between bitumen and LDHs was improved prominently by the grafting of LS. Before and after aging, the variations of retained penetration ratio (RPR), ductility retention rate (DRR), softening point increment (SPI), viscosity aging index (VAI) and the colloid stable index (CI) of LS grafted LDHs modified bitumen were smaller compared to LS modified bitumen and LDHs modified bitumen, which illustrated that LS grafted LDHs exhibited better anti-UV aging ability of bitumen in comparison with LDHs and LS. In improving the aging resistance of bitumen. LS-m-LDHs were superior to LS grafted LDHs linked by TDI and LS grafted LDHs linked by IPDI.

Objective: Aging-associated downregulation of Klotho is partly due to epigenetic upregulation of H3K27me3 in the kidney. The purpose of this study is to investigate the potential role of KDM6a, a demethylase of histone 3 lysine (K) 27 trimethylation (H3K27me3), in the regulation of blood pressure and explore the related molecular pathways that may influence the kidney function during aging. Methods and Results: Wild type and Ksp-Cre/KDM6a-floxed mice were divided into vehicle control and tamoxifen (10 mg/kg/mouse/7days) treated cohorts (WT mice: KDM6a floxed or Ksp-Cre only and Ksp-Cre/KDM6a -loxed mice). Blood pressure (BP) was measured by tail cuff method and confirmed by carotid artery cannulation at the end of study. Deletion of the KDM6a gene (KDM6a-cKO) significantly increased BP at day 7. BP remained elevated throughout the study. We found that the H3K27me3 level was significantly increased in the kidneys of KDM6a-cKO mice compared to control mice confirming that KDM6a is an important H3K27 demethylase in the control of methylation levels in the kidney. Elevation of BP was likely due to upregulation of the Na + :K + :2Cl - co-transporter (NKCC2) in the thick ascending limb of Henle's loop and NaCl co-transporter (NCC) in the distal convoluted tubule confirmed by qPCR and immunohistochemistry. Accordingly, we found that the urine sodium level was decreased in KDM6a-cKO mice compared to KDM6a-con mice. We showed that expression of aquaporin 2 (AQP2) was increased in the kidney of KDM6a-cKO mice, suggesting that AQP2 may also contribute to increased BP through modulation of body water homeostasis. Furthermore, we demonstrated that expression of Klotho was downregulated by 50%, which blocked FGFR1/Klotho/ERK signaling in the kidney of KDM6a-cKO mice. Notably, expression of aging markers including p53, p21, and p16 was significantly increased in the kidney of KDM6a-cKO mice, indicating that deletion of KDM6a in the renal tubule results in kidney aging. Conclusion: KDM6A is essential to the maintenance of normal kidney function and blood pressure. Renal-specific KDM6A knockout-induced hypertension is likely attributed to increased H3K27me3 levels and the resultant dowregulation of Klotho gene expression which impairs the FGFR1/Klotho/ERK signaling.

Chub mackerel (Scomber japonicus) is a pelagic fish widely distributed in temperate and subtropical zones throughout the Indian and Pacific Oceans and is commercially exploited, particularly in the North Pacific. Although highly targeted in this region, little is known about their life history aspects. The objectives of this study are to evaluate the growth heterogeneities and ageing analysis of this species. We describe the length-at-age, weight-at-length, relative condition factor relationships, spatiotemporal heterogeneity and compare estimated growth parameter values to those reported from other regions. This study used data obtained from Chinese fishing vessels collected from 2016–2020 in the northwest Pacific Ocean. Length-weight data from 2686 specimens (40–294 mm, fork length; 0.8–311.8 g body weight) were analyzed, and the Length-weight relationship was W = (1.41 × 10−6) × FL3.37. Seven linear mixed-effects models (LMEM) were used to analyze the heterogeneity of length-weight relationships of Chub mackerel. The Length-weight relationships for Chub mackerel were best described by a model with random effects with both year and season (spring, summer, autumn) with the scalar parameter a. Age estimates were obtained from 175 specimens, and the length-at-estimated ages relationship was described using three non-linear candidate growth models. The von Bertalanffy growth model fit the data best for Chub mackerel in the northwest Pacific Ocean. Comparing the results to that of previous studies, we observed that individual Chub mackerel exhibited a slower growth rate than that observed in previous studies. In addition, relative condition factors varied among years, seasons, and regions. Information presented in this study provides an effective scientific basis for stock assessment and fishery management of Chub mackerel in the northwest Pacific Ocean.

Diatomite is a kind of natural material with adsorption capacity and it’s widely used in industry. The main purpose of this paper is to study the influence of hydrochloric acid concentration and modification time on the modification effect of diatomite. This article describes a modified method of diatomite, which can not only reduce the purification cost, but also improve the separation efficiency of solanesol. The results show that when the concentration of hydrochloric acid is 3mol/L and the treatment time is 50 minutes, the modification effect of diatomite is the best. The determination of the optimum technological conditions is conducive to the separation and purification of Solanesol by diatomite, and lays a foundation for the further development and utilization of diatomite.

P. polymyxa YC0136 inoculation had beneficial effect on growth promotion and biological control of tobacco (Nicotiana tabacum L.) under field conditions. This study aimed to reveal the growth-promoting mechanisms of strain YC0136. In growth-promotion assays, tobacco plant height was increased by 8.42% and 8.25% at 60 and 90 days, respectively, after inoculation with strain YC0136. Strain YC0136 also promoted the accumulation of tobacco biomass in varying degrees. Following inoculation with strain YC0136, 3,525 and 4,368 tobacco genes were up-regulated and down-regulated, respectively. Strain YC0136 induced the expression of plant hormone-related genes in tobacco, including auxin, cytokinin, and gibberellin, as well as transcription factors related to stress resistance such as WRKY and MYB. In addition, strain YC0136 induced the up-regulation of genes in the phenylpropanoid biosynthesis pathway by 1.51–4.59 times. Interaction with tobacco also induced gene expression changes in strain YC0136, with 286 and 223 genes up-regulated and down-regulated, respectively. Tobacco interaction induced up-regulation of the ilvB gene related to auxin biosynthesis in strain YC0136 by 1.72 times and induced expression of some nutrient transport genes. This study contributes to our understanding of the growth-promoting mechanisms of strain YC0136 on tobacco and provides a theoretical basis for the application of P. polymyxa YC0136 as a biological fertilizer.

Interaction with tobacco also induced gene expression changes in strain YC0136, with 286 and 223 genes up-regulated and down-regulated, respectively. Tobacco interaction induced up-regulation of the ilvB gene related to auxin biosynthesis in strain YC0136 by 1.72 times and induced expression of some nutrient transport genes. This study contributes to our understanding of the growth-promoting mechanisms of strain YC0136 on tobacco and provides a theoretical basis for the application of P. polymyxa YC0136 as a biological fertilizer.

Alternaria alternata causes serious brown spot disease in tobacco. To effectively control the disease, we tested the antifungal effects of the essential oil from the buds of Syringa oblata Lindl. Twelve different compounds were identified by gas chromatography–mass spectrometry. The major constituents were eugenol (40.426%), eugenol acetate (28.784%), β-caryophyllene (21.989%), and α-caryophyllene (3.456%). Among these, the most active antifungal component was eugenol, exhibiting complete inhibition of mycelial growth. The minimum inhibitory concentration of eugenol against A. alternata was 150 μg/mL, and the minimum fungicidal concentration was 250 μg/mL. The effect of eugenol on the surface morphology and membrane integrity of A. alternata was studied by scanning electron microscopy (SEM) and fluorescent microscopy (FSM). SEM observations revealed shriveled hyphae, while FSM observations revealed a disrupted membrane in eugenol-treated samples. Further experiments confirmed that eugenol activated a membrane-active mechanism that increased the membrane permeability, as evidenced by extracellular conductivity measurements. Under field conditions, an in vivo assay indicated that eugenol emulsion was effective in protecting tobacco from brown spot infection. This study suggests that eugenol has the potential to be used as a fungicide to control tobacco brown spot disease caused by A. alternata.

Self-healing microcapsules show great potential in extending the service life of concrete, but it is a challenge to achieve the smart release of the healing agent from the microcapsules. In this research, Novel Microcapsules Raptured by Microwave (NMRM) with unique core–shell structure for self-healing concrete were successfully produced. The core content was calculated and the surface morphology and size distribution of NMRM were explored by Scanning Electron Microscope (SEM) and Dynamic Light Scattering (DLS). The temperature variations of specimens were monitored by a thermal infrared imager. Furthermore, the effects of NMRM dosage on the fundamental performance of concrete was investigated and self-healing behavior of the mortar and concrete were evaluated. As a result, rapture of microcapsules with 5% graphite can be quickly triggered under the microwave irradiation. The 60% fc0 pre-damaged concrete with 5% NMRM possessed 92.6% of CSRR. The purpose of this investigation is to provide a new trigger for microcapsule rupture on the specific location with cracks at the right time for self-healing concrete.

Phytophthora nicotianae causes serious black shank disease in tobacco. Syringa oblata essential oil and its main components were evaluated to develop an effective and environmentally friendly biocontrol agent. Eugenol, which exhibited the strongest activity, was intensively investigated in vitro and in vivo. The mycelial growth of P. nicotianae was inhibited by eugenol at a minimum inhibitory concentration of 200μgmL-1, and inhibition occurred in a dose-dependent manner. Extracellular pH and extracellular conductivity results indicated that eugenol increased membrane permeability. Flow cytometry and fluorescent staining results further showed that eugenol disrupted mycelial membranes but did not affect spore membrane integrity. The in vivo results confirmed that treatment of tobacco with various concentrations of eugenol formulations reduced disease incidence and better controlled against the disease. Our results suggested that the ability of eugenol to control tobacco black shank depended on its ability to damage mycelial membranes and that eugenol formulations have potential as an eco-friendly antifungal agent for controlling tobacco blank shank.

This study explored the chemical compositions of garlic essential oil, the inhibitory activity of garlic essential oil and diallyl disulfide (DADS) against Phytophthora nicotianae, and the effects on mycelial plasma membrane permeability and P. nicotianae inhibition. In total, 29 compounds were detected in garlic essential oil, of which 26 were detected by gas chromatography‒mass spectrometry (GC-MS) and 21 by headspace solid-phase microextraction (HS-SPME) GC-MS. DADS (60.12% and 19.09%) and trisulfide di-2-propenyl (14.18% and 17.98%) were the major components identified by HS-SPME GC-MS and GC-MS analysis, respectively. Half-inhibitory concentration (Ec50, antagonism) and minimum inhibitory concentration (MIC, fumigation) of DADS against P. nicotianae were 150.83 μL/L and 20 μL/L, respectively, while Ec50 of garlic essential oil was 1108.25 μL/L. Mycelial membrane permeability gradually increased in a concentration-dependent manner, and cell death increased at 450 μL/L DADS. Furthermore, DADS treatment significantly reduced the incidence of tobacco black shank and the number of P. nicotianae pathogens in rhizosphere soil. DADS also promoted root development of tobacco seedlings at low concentrations, which was inhibited at high concentrations. Therefore, DADS may play an important role in the antifungal effect against P. nicotianae by destroying mycelial cell membrane integrity, causing an increase in cell membrane permeability, and leading to cell death.

Two novel 3-decalinoyltetramic acid (3DTA) derivatives, namely fusarisetins C and D (1 and 2), and four known derivatives (3–6) were isolated from the marine-derived fungus Fusarium equiseti D39. Their structures were determined by spectroscopic data, vibrational circular dichroism (VCD) calculations, and X-ray crystallography. Compound 2 was identified as the first fusarisetin to possess an unprecedented carbon skeleton with a tetracyclic ring system comprised of a decalin moiety (6/6) and a tetramic acid moiety. A plausible biosynthetic pathway for the isolated compounds was proposed. All 3DTAs derivatives exhibited a potent phytotoxicity, and 5 also displayed a remarkable anti-phytopathogenic activity superior to the positive control resulting in damage of the cell membrane of Pseudomonas syringae and ensuing leakage of the intracellular components. Here, the phytotoxicity of fusarisetins has been reported for the first time. The OSMAC fermentation optimization approach to give 5 was performed by varying the culture media and salinities. The results showed that potato liquid medium with 1% salinity is the most favorable condition for the production of 5.

Taurine transporter (TauT) has been identified as a target gene of p53 tumor suppressor. TauT is also found to be overexpressed in variety type of human cancers, such as leukemia. This study showed that expression of TauT was upregulated by c-Myc and c-Jun oncogenes. To explore whether blocking of TauT inhibits tumor development, the RNA interference (RNAi) and immune targeting approaches were tested in tumor cells in vitro and in p53 mutant mice in vivo. Knockdown of TauT expression by RNAi resulted in cell cycle G2 arrest and suppressed human breast cancer MCF-7 cells proliferation determined by colonies production and cell migration assays. Knockdown of TauT also rendered MCF-7 cells more susceptible to chemotherapeutic drug-induced apoptosis. An antibody specifically against TauT blocked taurine uptake and induced cell cycle G2 arrest leading to cell death of variety type of tumor cells without affecting the viability of normal mammalian cells. TauT peptide vaccination significantly increased median lifespan (1.5-fold) of the p53 null mice and rescued p53+/− mice by extending the median lifespan from 315 days to 621 days. Furthermore, single dose treatment of tumor-bearing (thymic lymphoma) p53 null mice with TauT peptide reduced tumor size by about 50% and significantly prolonged survival of these mice from average 7 days (after observing the thymic lymphoma) to 21 days. This finding demonstrates that a novel TauT peptide vaccine can delay, inhibit, and/or treat p53 mutation related spontaneous tumorigenesis in vivo. Therefore, TauT peptide may be used as a universal cancer vaccine to prevent and/or treat patients with p53 mutation-mediated cancers.

The surface free energy and related parameters of asphalt binder affect the wettability of asphalt binder to aggregate and its moisture sensitivity. In this research, the surface free energy and related parameters of SBS modified bitumen (SMB) after aging/rejuvenating were tested and analyzed. And then based on this data, the effect of common rejuvenator cashew shell oil and active isocyanate rejuvenators on the water damage resistance and moisture sensitivity were evaluated and predicted. The results indicated that the surface free energy of SMB decreased after aging, while rejuvenators all can enhance the surface free energy. The cohesion work of aged SMB increased after rejuvenating, and the adhesion work between aged SMB and granite were enhanced by 12.4% at least with the addition of rejuvenators, namely, rejuvenators can improve its cracking resistance and interface stability, and active rejuvenator hexamethylene diisocyanate showed the best performance. Water boiling test showed that the adhesion grade of SBS modified bitumen on granite decreased from adhesion grade 5 to adhesion grade 3 after aging, while active rejuvenators can bring the adhesion grade of aged SMB on granite to the original level. This paper would be helpful to understand the moisture resistance mechanism of rejuvenated SBS modified asphalt mixture with active regeneration technology.

The regeneration and reusing of reclaimed asphalt mixture can promote the recycling of waste building materials and the sustainable development of asphalt pavement. To further improve the properties of rejuvenated SBS modified asphalt, the effects of structural changes of SBS, and the colloidal stability of asphalt on the properties of binder were studied. And based on the results, bio-rejuvenator and active rejuvenator were applied to rejuvenate ultraviolet aged SBS modified asphalt (USMA), the impacts of rejuvenators on the composition, structure, physical and rheological properties of USMA were systematically investigated. The results show the decrease of colloidal index and the damage of SBS structure all have a serious adverse influence on the properties of binder. Bio-rejuvenator can rebalance the colloidal stability of ultraviolet (UV) aged binders, meanwhile active rejuvenator could react with the active terminal groups of broken SBS segments and partially rebuilds its crosslinking structure. And the physical and rheological properties of UV aged asphalt can be all basically restored by adding bio-rejuvenator; however bio-rejuvenator cannot restore the properties of USMA, which can only improve the low-temperature performance of USMA. While the addition of active rejuvenator greatly enhances the high-temperature performance of rejuvenated SBS modified asphalt. Furthermore, the combination of bio-rejuvenator and active rejuvenator can both recover the colloidal structure and damaged SBS structure of USMA, which obviously improves its high- and low-temperature properties overall. This paper would contribute to the realization of high-quality regeneration of reclaimed SBS modified asphalt.

Ion chelator can chelate calcium ions and migrate in cement-based materials to promote self-healing of cracks. This paper investigated the migration and self-healing abilities of ion chelator in cement-based materials by a novel method. In this method, control mortar without ion chelator and self-healing mortar with ion chelator are poured into the same mold, cracks at different locations of the two kinds of mortars are prefabricated, and the migration of ion chelator from self-healing mortar to the control mortar is assessed by observing the self-healing process of crack in control mortar. Meanwhile, the microstructure and chemical component of self-healing product in cracks at different locations of the control mortar was characterized through scanning electron microscope (SEM) and X-ray diffraction (XRD). The results show that the self-healing ability of control mortar is closely related to the migration distance of ion chelator. When the migration distance of the ion chelator was 2 cm, the crack with a width of 0.37 mm can be sealed after curing for 7 d. When the migration distance of the ion chelator was 5 cm, the crack with a width of 0.26 mm can also be repaired after curing for 7 d. Through SEM observation and XRD analysis, it can be found that the ion chelator promotes the formation of a large number of calcite crystals for sealing the cracks in the control mortar. However, the calcite crystals at the crack will become loose with the increase of the migration distance of the ion chelator.

To fully restore the performance of waste SBS modified asphalt mixture, better realize its recycling and application in high-grade pavement, achieve the concept of sustainable green development of asphalt pavement. In this paper, the synergistic effects of bio-rejuvenator and active isocyanate rejuvenators (including hexamethylene diisocyanate (HDI), diphenyl-methane-diisocyanate (MDI) and toluene-2, 6-diisocyanate (TDI)) on the chemical structure, physical and rheological properties of aged SBS modified bitumen were thoroughly investigated. Fourier infrared spectrum and Gel permeation chromatography analysis indicate the isocyanate groups of active rejuvenators react with hydroxyl or carboxyl group from the degraded SBS segments to form new amide groups that can reconnect the broken segments of aged SBS and partly restore its molecular weight distribution. From physical and rheological properties, bio-rejuvenator can restore the low-temperature properties by rebalancing the colloidal structure of aged binder, meanwhile, active rejuvenators can greatly improve the high-temperature performance of rejuvenated SBS modified bitumen with bio-rejuvenator through reconnecting the broken segments of aged modifier SBS. Furthermore, MDI with benzene ring structure is more conducive to enhancing the high temperature rutting resistance of rejuvenated SBS modified bitumen. While HDI can bring some flexible segments into the restored structure of SBS, which is more beneficial to improve its low-temperature cracking resistance of rejuvenated binder. And the fluorescence images further demonstrate isocyanate rejuvenators can partly restore the crosslinking network structure of aged modifier SBS in rejuvenated SBS modified bitumen. This research would be conductive to promoting the application of rejuvenated SBS modified bitumen into the construction of high standard pavement.

Ion chelator is a novel crystalline additive which can chelate Ca2+ and migrate in concrete to promote self-healing of surface crack and internal pore. In order to further explore the effect of ion chelator on hydration process of cement, the hydration heat, chemically bound water, compressive strength, hydration products, pore-size distribution and microstructure of cement paste with ion chelator were investigated in this paper. Results of the experiments showed that the ion chelator accelerated the hydration of the cement paste. Incorporation of ion chelator in cement greatly enhanced the chemically bound water content and the mechanical property. Compared with control cement paste, the chemically bound water content and compressive strength of cement paste with 0.5 wt% ion chelator increased by 13.3% and 16.8% after curing for 28 days. According to the DTA-TG and XRD tests, the ion chelator can promote the hydration of C3S and C2S, which was conducive to the generation of calcium silicate hydrate gels. Furthermore, the addition of ion chelator improved the pore-size distribution and microstructure of cement paste, and a large number of floccules and needles appeared in the pores of cement paste.

In order to evaluate the influence of silane coupling agent modified zeolite warm mix additives on the properties of asphalt, the physical and rheological properties as well as chemical structure of asphalt with zeolite, γ-ammoniapropyltriethoxysilane modified zeolite (KH550-zeolite) and γ-(2,3-epoxypropoxy)propytrimethoxysilane modified zeolite (KH560-zeolite) before and after thin film oven test (TFOT) aging were thoroughly investigated. The results showed that, compared with zeolite, KH550-zeolite and KH560-zeolite made the softening point of asphalt increase more, and whose penetration and ductility decrease less, which indicated that KH550-zeolite and KH560-zeolite (especially KH560-zeolite) could enhance the high-temperature properties and had less effect on the penetration and ductility of asphalt. The temperature susceptibility of asphalt was clearly reduced and the rutting factor (G*/sinδ) was enhanced after the introduction of silane coupling agent modified zeolites compared with zeolite. After the TFOT aging, the performance of asphalt with silane coupling agent modified zeolites (especially KH560-zeolite) had a little change. In a word, KH550-zeolite and KH560-zeolite as warm mix additives had a positive effect on the physical, rheological and anti-aging properties of asphalt.
