Objective: The relationships between childhood weight self-misperception and obesity-related factors particularly health markers have not been extensively discussed. This study aims to examine the associations between weight self-misperception and obesity-related knowledge, attitudes, lifestyles and cardio-metabolic markers among Chinese paediatric population. Design: Cross-sectional study. Setting: Data sourced from a national survey in Chinese seven provinces in 2013. Participants: Children and adolescents aged 5–19 years. Results: Of the total 14 079 participants, there were 14·5 % and 2·2 % participants over-estimated and under-perceived their weight, respectively. Multi-variable logistic regression was applied to calculate OR and 95 % CI (95 % Cl) of obesity-related behaviours and cardio-metabolic markers by actual and perceived weight status. Individuals who perceived themselves as overweight/obese were more likely to have prolonged screen time, insufficient dairy intake and over sugar-sweetened beverages consumption (all P < 0·05), regardless of their weight. Furthermore, actual overweight/obese individuals had higher odds of abnormal cardio-metabolic markers, but a smaller magnitude of association was found among weight under-estimators. Among non-overweight/obese individuals, weight over-estimation was positively associated with abdominal obesity (OR: 10·49, 95 % CI: 7·45, 14·76), elevated blood pressure (OR: 1·30, 95 % CI: 1·12, 1·51) and dyslipidemia (OR: 1·43, 95 % CI: 1·29, 1·58). Conclusions: Weight over-perception was more prevalent than under-estimation, particularly in girls. Weight over-estimators tended to master better knowledge but behave more unhealthily; both weight over-perception and actual overweight/obesity status were associated with poorer cardio-metabolic markers. Future obesity intervention programmes should additionally pay attention to the population with inaccurate estimation of weight who were easily overlooked.

To stop the COVID-19 epidemic from spreading among their populations, several countries have implemented lockdowns. Students are being forced to stay at home during these lockdowns, which is causing them to use mobile phones, social media, and other digital technologies more frequently than ever. Their poor utilization of these digital tools may be detrimental to their emotional and mental health. In this study, we implement an Artificial Intelligence (AI) approach named Hierarchy-based K-Means Clustering (HKMC) algorithm to group individuals with comparable Twitter consumption habits to detect addictive Twitter activity during the epidemic. The effectiveness of the suggested HKMC is evaluated in terms of accuracy, precision, recall, and f1-score in respect to the association between students’ mental health and mobile phone dependency. Additionally, this study offers a comparative examination of both the suggested and existing procedures.

Wound treatment remains one of the most prevalent healthcare issues. Tylotoin is a skin repair peptide identified from salamander (Tylototriton verrucosus) and exhibits skin wound healing properties. Noticeably, the easy degradation and frequent administration limit its application in wound healing. Chitosan (CS) -PLGA-Tylotoin nanoparticles (CPT NPs) were prepared to circumvent this limitation and deliver Tylotoin for the promotion of the healing of skin wounds. Results showed that optimized CPT NPs particle size, zeta potential, encapsulation efficiency and drug loading were 297.80 ± 5.37 nm, 20.37 ± 0.83 mV, 81.00 % and 1.74 %, respectively. In vitro, CPT NPs exhibited good antibacterial properties and biocompatibility and persistently promoted the cell migration of HaCaT cells and HUVECs due to the long-term sustained release of Tylotoin within 14 days (64.81 %). In vivo, the scarless healing of skin wound promotion was evaluated in mouse back full-thickness wound models. We demonstrated that mouse back full-thickness wounds topically treated with CPT NPs once every two weeks exhibited better scarless healing than those treated with Tylotoin once daily. We envision that CPT NPs, as a Tylotoin delivery platform might, may be potentially utilized to in skin wounds healing in clinics in the future.

Objective: To improve the prognosis of patients with gastric cancer (GC), more effective therapeutic targets are urgently needed. Increasing evidence indicates that miRNAs are involved in the progression of various tumors, and RAS-associated protein in the brain 31 (RAB31) is upregulated and promotes the progression of multiple malignant tumors. Here, we focused on identifying RAB31-targeted miRNAs and elucidating their potential mechanism in the progression of GC. Methods: RAB31 and miR-378a-3p expression levels were detected in paired fresh GC tissues and GC cell lines. Bioinformatics analysis was used to predict the miRNAs targeting RAB31 and the relationships between RAB31 and other genes. Dual-luciferase reporter assays were applied to verify the targeted interaction relationship. CCK-8, colony formation, flow cytometry, wound healing, and Transwell assays were performed to assess the proliferation, apoptosis, migration, and invasion of GC cells. Tumorsphere formation assays were performed to assess the stemness of gastric cancer stem cells. Related proteins were detected by Western blot. Xenograft assays in nude mice were performed to explore the effect of miR-378a-3p in vivo. Results: We report the first evidence that miR-378a-3p is downregulated in GC, whereas its overexpression inhibits proliferation, invasion, and migration as well as promotes apoptosis in GC cells. Mechanistically, miR-378a-3p inhibits the progression of GC by directly targeting RAB31. Restoring RAB31 expression partially offsets the inhibitory effect of miR-378a-3p. Further research revealed that miR-378a-3p inhibits GLI1/2 in the Hedgehog signaling pathway and attenuates the stemness of gastric cancer stem cells. Finally, xenograft assays showed that miR-378a-3p inhibits GC tumorigenesis in vivo. Conclusions: MiR-378a-3p inhibits GC progression by directly targeting RAB31 and inhibiting the Hedgehog signaling pathway proteins GLI1/2.

Although clinical outcomes of CLL have improved with the use of BCL-2 inhibitor, ABT-199, acquired resistance eventually occurs in many cases, which leads to CLL disease progression. Thus, understanding the mechanisms that mediate this relapse is important to design improved therapies. Herein, we report that cytokine IFN-γ, secreted by dysfunctional T cells, enhanced CLL cells resistance to ABT-199. IFN-γ stimulation significantly increased the expression of BCL-2, MCL-1 and BCL-xL. Blocking JAK1/2-STAT3 signaling pathway impaired the expression of these anti-apoptotic proteins after IFN-γ stimulation. The combination of ABT-199 with JAK1/2 inhibitor Ruxolitinib or STAT3 inhibitors Stattic and C188-9 increased malignant B cell death. In summary, we show that IFN-γ enhanced CLL cells resistance to ABT-199 at least in part by up-regulating BCL-2, MCL-1 and BCL-xL expression

RNA polymerase II–mediated eukaryotic transcription starts with the assembly of the preinitiation complex (PIC) on core promoters. The +1 nucleosome is well positioned about 40 base pairs downstream of the transcription start site (TSS) and is commonly known as a barrier of transcription. The +1 nucleosome–bound PIC-Mediator structures show that PIC-Mediator prefers binding to T40N nucleosome located 40 base pairs downstream of TSS and contacts T50N but not the T70N nucleosome. The nucleosome facilitates the organization of PIC-Mediator on the promoter by binding TFIIH subunit p52 and Mediator subunits MED19 and MED26 and may contribute to transcription initiation. PIC-Mediator exhibits multiple nucleosome-binding patterns, supporting a structural role of the +1 nucleosome in the coordination of PIC-Mediator assembly. Our study reveals the molecular mechanism of PIC-Mediator organization on chromatin and underscores the significance of the +1 nucleosome in regulating transcription initiation.

Research background: Inflation has always been the core issue of economic research and there are many academic research achievements in this field. In recent years, global inflation has intensified, and many scholars focus on research in this field again, providing certain reference value for countries around the world to formulate corresponding macro policies. Purpose of the article: The five-year impact factors are used as the evaluation criteria in this paper, and 1,637 high-quality documents on inflation from 1906 to 2022 are collected from the Web of Science Core Collection database. Using bibliometrics, a comprehensive review of influential literature in the field of inflation is conducted to reveal the evolution and trends of the field. Methods: First, we focus on these high-quality documents about the descriptive statistical characteristics, high cited documents and high impact factor journals. Then, based on the visualization tool, the cooperative network of countries/regions, authors and institutions is depicted and the cooperative relationship between them is determined. At the same time, the most influential countries/regions, authors and institutions are identified by analyzing the citation structure. In addition, through thematic and keyword analysis, the topic hotspots and future research trends of high-quality literature in the field of inflation are deduced. Findings & value added: On the whole, the research on inflation in the United States is relatively mature, and has produced a large number of influential academic cooperation results. Finally, we have a series of discussions on the history of inflation in the United States and policy suggestions. In the future, governments of various countries, especially the United States, will still face certain challenges in how to formulate policies and measures to mitigate the impact of inflation.

College students' tourism group has always been an indispensable market segment of the tourism market. In order to study the outbreak period of normalized college students 'tourism consumption behavior change, this paper adopts the questionnaire survey and depth interview, in xi'an university students as a sample, from the outbreak of the tourism behavior change, tourism behavior intention and may influence its behavior decision factor selection of three dimensions, with the method of mathematical statistical method analysis. Studies show that the consumption behavior, destination choice, travel time and frequency, and travel willingness of college students have all changed during the normalization of the epidemic. And from the government, tourism enterprises, college students themselves and other three dimensions put forward solutions.

Phase separation (PS) proteins form droplets to regulate myriad membraneless organelles (MLOs) and cellular pathways such as transcription, signaling transduction and protein degeneration. PS droplets are usually liquid-like and can convert to hydrogel/solid-like under certain conditions. The PS behavior of proteins is regulated by co-PS partners and mutations, modifications, oligomerizations, repeat regions and alternative splicing of the proteins. With growing interest in PS condensates and associated proteins, we established PhaSepDB 1.0, which provided experimentally verified PS proteins and MLO-related proteins. The past few years witnessed a surge in PS-related research works; thus, we kept updating PhaSepDB. The current PhaSepDB contains 1419 PS entries, 770 low-throughput MLO-related entries and 7303 high-throughput MLO-related entries. We provided more detailed annotations of PS proteins, including PS verification experiments, regions used in experiments, phase diagrams of different experimental conditions, droplet states, co-PS partners and PS regulatory information. We believe that researchers can go further in studying PS proteins with the updated PhaSepDB (http://db.phasep.pro/).

Plants produce a wide variety of secondary metabolites in the process of evolution. Secondary metabolites have highly diverse structures due to the modification of the basic skeletons of metabolites. They are required for interaction with the environment and are produced in response to abiotic/biotic stress. Characterization of secondary metabolic pathways is significant to plant molecular breeding and natural product biosynthesis. The liquid chromatography-high resolution tandem mass spectrometry (LC-HRMS/MS) is one of the major techniques for untargeted metabolomics study. The LC-HRMS/MS method could detect tens of thousands of metabolic features and provide abundant structural information. It has been widely used in the discovery and characterization of the secondary metabolome. However, due to the largely diverse structure and limited records in the mass spectral library, the annotation of the secondary metabolome is very difficult. To address the analytical challenges associated with the vast structural diversity and the large numbers of secondary metabolites, particularly those previously unknown structural metabolites, a novel method for the efficient characterization of pathway-associated metabolites was developed. Modification reactions and MS/MS spectral information were collected using the metabolic pathways database and mass spectral library. Screening and annotation of metabolites involved in phenylpropanoid metabolism in maize leaves were used as an example. First, a database of modified groups was established via pathway-associated modifications from open access metabolic pathway database and literature. Here, pathway databases included the Kyoto Encyclopedia of Genes and Genomes (KEGG) and Plant Metabolic Pathways (PlantCyc). A total of 61 modification types were enrolled, including 10 generic and 51 pathway-specific modifications. Modified metabolomes were filtered from untargeted LC-HRMS/MS metabolomics data. Next, MS/MS spectra of the pathway-associated compounds (probe molecules) were collected in the Global Natural Products Social Molecular Networking (GNPS) MS/MS spectral library. The MS/MS of compounds assigned to chemical classes of phenylpropanoids were kept. An MS/MS spectral database of the probe molecules was constructed. It included 2677 spectra of 1542 phenylpropanoid compounds in the positive mode and 814 spectra of 661 phenylpropanoid compounds in the negative mode. Then, an MS/MS molecular network was generated by modified metabolome and probe molecules. The clusters comprising both probe molecules and modified metabolites were kept. To explore more previously unknown structural metabolites, the clusters with one more pathway-specific modified metabolite were retained even though they didn't contain any probe molecule. A total of 392 and 417 phenylpropanoid pathway-related metabolic metabolites were obtained in positive and negative ion modes, respectively. The pathway-associated metabolites were annotated based on the propagation of the molecular network. For the metabolites within the co-cluster, annotations were performed using the probe molecules as the initial seed. The modification group's substructure information was used for network propagation annotation. For the clusters containing only pathway-specific modified metabolites, the annotation is similar to the above process if identified nodes were present within the cluster. Otherwise, de novo annotation was manually executed based on substructure information. Finally, 129 unique metabolites were annotated after integration and removal of redundancy. Ten annotated metabolites were validated using commercially available or synthesized reference compounds. The other annotation results were validated using predicted chemical classes

The disorder flow of seeds caused by the collision in the seed-guiding tube is one of the important reasons for the reduction of seeding uniformity. For this problem, a seed-guiding tube with brachistochrone considering the influence of friction was presented in this paper. By analyzing kinetics and rotating the equation coordinate system, brachistochrone under the condition of friction was obtained and applied to the design of the structural parameters of the grain conductor of T-groove furrowing device, and a discrete element model of the grain conductor and wheat was established, thus single-factor simulation analysis and quadratic regression orthogonal rotation combination simulation tests were conducted to explore the optimal combination of parameters. The test results showed that the seed-guiding tube had a brachistochrone radius of 14.708 mm, a linear length of 500 mm and a working speed of 4 km/h, and the seed spacing offset dispersion degree and the seed lateral offset dispersion degree were 12.26 and 8.53 respectively, which had a high seeding uniformity. The results of the bench validation test showed that the coefficients of variation of the two test evaluation indexes between the bench test results and the virtual simulation results were 10.20% and 8.62% respectively, therefore the simulation results could be used for the optimization of the seed guide tube.

Data and knowledge of surface water bodies (SWB), including large lakes and reservoirs (surface water areas > 1 km2) are critical for the management and sustainability of water resources. However, the existing global or national dam datasets have large georeferenced coordinate offsets for many reservoirs, and some datasets have not reported reservoirs and lakes separately. In this study, we generated China’s surface water bodies, Large Dams, Reservoirs, and Lakes (China-LDRL) dataset by analyzing all available Landsat imagery in 2019 (19,338 images) in Google Earth Engine and very-high spatial resolution imagery in Google Earth. There were ~3.52 × 106 yearlong SWB polygons in China for 2019, only 0.01 × 106 of them (0.43 %) were of large size (> 1 km2). The areas of these large SWB polygons accounted for 83.54 % of the total 214.92 × 103 km2 yearlong surface water area (SWA) in China. We identified 2,140 large dams, including 1,494 reservoir dams and 646 river dams, 1,976 large reservoirs (16.42 × 103 km2), and 3,508 large lakes (75.97 × 103 km2). In general, most of the dams and reservoirs in China were distributed in South China, East China, and Northeast China, whereas most of lakes were located in West China, the Lower Yangtze River Basin, and Northeast China. The provision of the reliable, accurate China-LDRL dataset on dams, large reservoirs and lakes will enhance our understanding of water resources management and water security in China.
