Direct discharge of aquaculture wastewater may have toxic effects, due to the presence of heavy metals, antibiotics, and even resistant pathogens, but little attention has been given. Here, tanks simulating a wild ecosystem were built to study the effects of long-term exposure to duck wastewater containing oxytetracycline (OTC) and/or arsenic (As) on the growth, physiological function, and gut microbiota evolution of Xenopus tropicalis. The results showed that duck wastewater had no apparent impact on X. tropicalis, but the impact increased significantly (P0.05) with exposure to OTC and/or As, especially the impact on body weight and growth rate. Biochemical indicators revealed varying degrees of oxidative stress damage, hepatotoxicity (inflammation, necrosis, and sinusoids), and collagen fibrosis of X. tropicalis in all treated groups after 72 days of exposure, which indirectly inhibited X. tropicalis growth. Moreover, 16S rDNA amplicon sequencing results showed that the gut microbiota structure and metabolic function were perturbed after chronic exposure, which might be the leading cause of growth inhibition. Interestingly, the abundance of intestinal resistance genes (RGs) increased with exposure time owing to the combined direct and indirect effects of stress factors in duck wastewater. Moreover, once the RGs were expressed, the resistance persisted for at least 24 days, especially that conferred by tetA. These results provide evidence of the toxic effects of DW containing OTC (0.1-4.0 mg/L) and/or As (0.3-3.5 µg/L) on amphibians and indicate that it is vital to limit the usage of heavy metals and antibiotics on farms to control the biotoxicity of wastewater.

Oral squamous cell carcinoma (OSCC), the most common malignancy of the oral and maxillofacial region, severely affects human health. However, current treatments for OSCC commonly show only a ~60% 5-year survival rate of patients with distant metastases, indicating an urgent need for targeted treatments for patients with advanced metastases. Here, we report a survival-related long non-coding RNA, CYTOR, which is highly expressed in the lesions of oral cancer patients. We found that CYTOR can promote both migration and invasion in oral cancer cells as well as the epithelial–mesenchymal transition (EMT). RNA-sequencing of CYTOR-knockdown oral cancer cells revealed that CYTOR can regulate mitochondrial respiration and RNA splicing. Mechanistically, we found that nuclear-localized CYTOR interacts with HNRNPC, resulting in stabilization of ZEB1 mRNAs by inhibiting the nondegradative ubiquitination of HNRNPC. By synthesizing CYTOR-targeting small interfering RNAs (siRNAs) encapsulated in Nanoscale Metal Organic Frameworks (NMOFs), we demonstrate the targeted suppression of CYTOR to inhibit invasion and metastasis of oral cancer cells in a nude mouse model. Cumulatively, this study reveals the potential role of the CYTOR-HNRNPC-ZEB1 axis in regulating mitochondrial metabolism and glycolysis of oral cancer cells, and illustrates the effective use of lncRNA targeting in anti-metastatic cancer therapies.

The performance of a mixotrophic photoelectroactive biofilm reactor (MPBR) was improved in order to achieve enhanced simultaneous removal of multiple aqueous pollutants and the production of valuable biomass. The MPBR was optimized by integrating the regulation of light intensity (3000, 8000 and 23000 lux) and microbial extracellular electron extraction (using an electrode at -0.3, 0 and 0.3 V). To investigate the role of oral microbiome in promoting oral squamous cell carcinoma (OSCC) development.We investigated the salivary microbiome of 108 controls and 70 OSCC cases by16S rRNA gene sequencing and detected the fluorescence signal of OSCC-related pathological bacteria by fluorescence in situ hybridization assay (FISH). The invasion and migration assays were used to show the differences of invasive and migrative abilities between control and experimental groups. Quantitative real-time PCR and Western blotting were used to verify the epithelial-to-mesenchymal transition (EMT).In our study, the overall microbiome abundance and composition were richer in the 108 controls than in the 70 OSCC cases. We demonstrated that Streptococcus, Capnocytophaga, Peptostreptococcus, and Lactobacillus were highly abundant in the saliva of OSCC patients by 16S rDNA sequencing and FISH. Moreover, we found that Capnocytophaga gingivalis (C. gingivalis) was highly presented in OSCC tissues by FISH. We focused on C. gingivalis and found that its supernatant induced OSCC cells to undergo EMT, causing the cells to acquire a mesenchymal phenotype associated with highly invasive and metastatic properties.Taken together, these results indicated that C. gingivalis might invade OSCC tissues and played an important role in OSCC by promoting OSCC invasion and metastasis by inducing EMT. Hence, the role of C. gingivalis in cancer progression revealed a new direction for the research of OSCC.

Multidrug-resistant bacteria, especially pathogens, pose a serious threat to disease treatment and recovery, but their potential toxicity to animal development is not entirely clear. As the most important site for nutrient absorption, we studied the intestinal microbiome of Xenopus tropicalis by analyzing the effect of multidrug-resistant Shigella on its intestinal health. Unlike in the control, Shigella intake promoted the secretion of neutral mucus and inhibited intestinal development and weight gain. Following 60 days of exposure, intestinal crypt atrophy, intestinal villus shortening, internal cavity enlargement, and external mucosal muscle disintegration were observed. The circular and longitudinal intestinal muscles became thinner with increasing pathogen exposure. In addition, the presence of Shigella altered the expression of multiple cytokines and classic antioxidant enzyme activities in the gut, which may have caused the intestinal lesions that we observed. 16 S rDNA sequencing analysis of intestinal samples showed that exposure to Shigella destroyed the normal gut microbial abundance and diversity and increased the functional bacterial ratio. Notably, the increased abundance of intestinal antibiotic resistance genes (ARGs) may imply that the resistance genes carried by Shigella easily migrate and transmit within the intestine. Our results expand existing knowledge concerning multidrug-resistant Shigella-induced intestinal toxicity in X. tropicalis and provide new insights for the threat assessment of resistance genes carried by drug-resistant pathogens.

Traditionally, toxicity of microplastics is ascribed to the chemicals adsorbed on them. However, microplastics can also interact with biomolecules, such as secretory proteins from aquatic organisms, and form protein-coated microplastics corona complex with unknown toxic effects. Here, we investigated the toxic effects of polystyrene microplastics (PS) and bovine serum albumin (BSA) coated PS corona complex (PS + BSA) on adult zebrafish (Danio rerio) intestines. The food intake ratio, accumulation and distribution of microplastics, histopathological changes, and molecular effects related to the antioxidant system in the intestine were studied. For the first time, we observed that PS + BSA aggregated on the inner surface of the zebrafish intestine, whereas PS dispersed. The aggregation of PS + BSA resulted in increased microplastics accumulation and longer residence time in the zebrafish intestine, which inhibited food intake and generated reactive oxygen species (ROS) in the intestine. Furthermore, the functions of the Keap1-Nrf2-ARE antioxidant signaling pathway and the activation of antioxidant enzymes were significantly affected by PS + BSA after a 21-day exposure. Ultimately, a higher accumulation of ROS and stronger inhibition of antioxidants led to more severe intestinal injury. These results suggest that the increased toxicity of protein-coated microplastics corona complex may be affected by oxidative damage and can result in the inhibition of digestion due to their aggregation and longer residence time in the intestine. Therefore, the ecological risk of microplastics may be underestimated owing to the interactive mechanisms of microplastics and protein coronas.

Di-(2-ethylhexyl) phthalate (DEHP), used as a popular plasticizer to enhance the flexibility of plastics, is a major pollutant in aquatic environments. DEHP poses severe risks to aquatic organisms since it is an endocrine-disrupting compound. To comprehensively evaluate the toxicity of DEHP on the growth and livers of male Xenopus tropicalis(X. tropicalis), sexually mature male X. tropicalis were exposed to environmentally relevant concentrations of DEHP, 0.2, 0.6, 1.8, 5.4 mg/L, for 49 days. The results showed that DEHP had a severe toxic effect on the livers of male X. tropicalis. Histopathological analysis of livers in all the DEHP-exposed groups showed changes in terms of vacuolization, loose cell cords, and an increasing amount of melanin. Large lipid droplets were markedly formed, and there were changes in the mitochondrial morphology upon DEHP exposure. In addition, oxidative stress (excessive ROS and inhibition of antioxidants) was induced through the suppression of biochemical indicators and the downregulation in the mRNA expression of genes (nrf2, cat, sod, gst, and gpx) related to oxidative stress. A reduction in expression of fatty acid metabolism-related gene (pparα) was seen post-DEHP exposure. Thus, our study suggests that the hepatotoxicity induced by DEHP could be attributed to oxidative stress and disordered fatty acid metabolism. In conclusion, long-term exposure to DEHP at environmentally relevant concentrations poses ecological risks to aquatic organisms, which serves as a reminder that the application of DEHP and other plasticizers should be limited.

This work presents facile preparation processes of multiphosphonic acid functionalised multi-walled carbon nanotubes (HEDP-MWCNTs and ATMP-MWCNTs), which have been characterised by FT-IR, XPS, X-ray diffraction, TGA, and SEM, and have employed for heavy metal ions adsorption. The adsorption properties of MWCNTs, HEDP-MWCNTs, and ATMP-MWCNTs for gold ions, cadmium ions, copper ions, lead ions, nickel ions, cobalt ions, zinc ions, and chromium ions have been investigated, the experimental results reveal that the functionalised samples HEDP-MWCNTs and ATMP-MWCNTs have excellent adsorption properties for Pb(II) and Au(III), respectively. The investigation results on their adsorption selectivity represent that in the binary heavy metal ion systems, HEDP-MWCNTs/ATMP-MWCNTs has displayed a strong affinity for Pb(II)/Au(III) and even could exhibit 100% selectivity while coexisting with copper ions, nickel ions, cobalt ions, zinc ions, chromium ions, or cadmium ions. Furthermore, the adsorption kinetics of HEDP-MWCNTs for Pb(II) and ATMP-MWCNTs for Au(III) have also been studied at different solution temperatures. All these research work results show that two novel functionalised adsorbents HEDP-MWCNTs and ATMP-MWCNTs have shown excellent adsorption properties for heavy metal ions and they can be used in wastewater treatment, especially for lead/gold ions removal.

Objective Head and neck squamous cell carcinoma (HNSCC) is a major threat to public health. Pyroptosis is a form of inflammatory programmed cell death that is still incompletely understood. The role of pyroptotic cell death in HNSCC remains to be fully defined. As such, the present study was developed to explore the potential prognostic utility of a pyroptosis-related gene (PRG) signature in HNSCC. Methods PRG expression patterns and the associated mutational landscape in HNSCC were analyzed, after which a 6-gene prognostic model was constructed through least absolute shrinkage and selection operator (LASSO) and Cox regression analyses using the TCGA dataset, followed by validation with two GEO datasets (GSE41643 and GSE65858). The relative expression of the genes in the prognostic model was assessed via RT-qPCR in tumor and paired adjacent normal tissue samples from a 32-patient cohort. Potential predictors of patient outcomes associated with this 6-gene model were identified through topological degree analyses of a protein–protein interaction network. Moreover, the prognostic value of NLRP3 as a predictor of HNSCC patient prognosis was established through immunohistochemical (IHC) analyses of samples from 176 HNSCC patients. Lastly, in vitro studies were performed to further demonstrate the relevance of NLRP3 in the context of HNSCC development. Results Differentially expressed PRGs were able to readily differentiate between HNSCC tumors and normal tissues. Risk scores derived from the 6-gene PRG model were independent predictors of HNSCC patient prognosis, and genes that were differentially expressed between low- and high-risk groups were associated with tumor immunity. RT-qPCR assays also showed the potential protective role of NLRP3 in HNSCC patients. IHC analyses further supported the value of NLRP3 as a predictor of HNSCC patient outcomes. Invasion and migration assays demonstrated the potential role of NLRP3 in the inhibition of HNSCC development. Conclusions Overall, these results highlight a novel prognostic gene signature that offers value in the context of HNSCC patient evaluation, although additional research will be essential to elucidate the mechanisms linking these PRGs to HNSCC outcomes.

Glutaraldehyde and didecyldimethylammonium bromide (GD) is a disinfectant widely used to prevent African swine fever (ASF) in livestock farms. However, the effect of residual GD on the activated sludge microbial ecology of receiving wastewater treatment plants (WWTPs) remains largely unknown. In this study, seven simulated systems were established to research the effects of GD on WWTPs and reveal the underlying mechanisms of microecological responses to GD at different concentrations. Both the nitrogen and carbon removal rates decreased with increasing GD concentrations, and nitrogen metabolism was inhibited more obviously, but the inhibition weakened with increasing stress duration. Microorganisms activated their SoxRS systems to promote ATP synthesis and electron transfer to support the hydrolysis and efflux of GD by producing a small number of ROS when exposed to GD at less than 1 mg/L. The overproduction of ROS led to a decrease of antioxidant and nitrogen removal enzyme activities, and upregulation of the porin gene increased the risk of GD entering the intracellular space upon exposure to GD at concentrations higher than 1 mg/L. Some denitrifiers survived via resistance and their basic capabilities of sugar metabolism and nitrogen assimilation. Notably, low concentrations of disinfectants could promote vertical and horizontal transfer of multiple resistance genes, especially aminoglycosides, among microorganisms, which might increase not only the adaptation capability of denitrifiers but also the risk to ecological systems. Therefore, the risks of disinfectants targeting ASF on ecology and health as well as the effects of disinfectant residuals from the COVID-19 epidemic should receive more attention.

Subnanometric Pd clusters confined within zeolite crystals was fabricated using zeolitic seeds with premade [Pd3Cl(PPh2)2(PPh3)3]+ clusters under hydrothermal conditions. Characterization of the Pd3@Beta catalysts indicate that the Pd clusters confined in the channels of Beta zeolite exhibit better dispersion and stronger interaction with the zeolite support, leading to stabilized Pd species after heat treatment by high temperature. In the model reaction of toluene combustion, the Pd3@Beta outperforms both zeolite-supported Pd nanoparticles prepared by conventional impregnation of Pd3/Beta and Pd/Beta. Temperatures for achieving toluene conversion of 5%, 50% and 98% of Pd3@Beta are 136, 169 and 187 °C at SV = 60 000 mL g−1 h−1, respectively. Pd3@Beta could also maintain the catalytic reaction for more than 100 h at 230 °C without losing its activity, an important issue for practical applications. The metal-containing zeolitic seed directed synthesis of metal clusters inside zeolites endows the catalysts with excellent catalytic activity and high metal stability, thus providing potential avenues for the development of metal-encapsulated catalysts for VOCs removal.

Electrical resistance tomography (ERT) is a sensing technique for process monitoring and measurement, the image reconstruction algorithms are required fast and accurate. The OMP class algorithm has the property of reconstructing a higher quality signal by fewer iteration steps (shorter elapsed time) and faster convergence speed compared with other iterative algorithms. But the classical OMP is not self-adapted and may fall into a local optimal solution. In this paper, a modified orthogonal matching pursuit (MOMP) was presented by adding a continuous constraint to prevent local optimal solution and self-adaption process on classical OMP algorithm which finds the optimal number of iterations based on the information entropy of measurement to make it suitable for ERT inverse problem. The algorithm was experimentally validated versus other algorithms through a 16-electrode ERT system. Comparing with other algorithms, MOMP algorithm has a lower mean square error than non-iterative algorithms and a shorter time cost than iterative algorithms. Dynamic experiments showed that the MOMP algorithm is promising for industrial process monitoring.

The rapid removal of organic dyes from water under low temperature environment offers significant advantages when water purification is required in cold regions. In this work, porous lanthanum hexaboride (LaB6) nanoparticles were fabricated via a facile low-temperature molten metal (LTMM) method in Zn media under argon atmosphere. As is evidenced by the characterization results, porous LaB6 can be prepared at 400 °C. Elevated reaction temperature led to the growth of nanoparticles, serious aggregation and the residue of Zn element in the final product. Notably, LaB6 nanoparticles prepared at 600 °C by such a LTMM method presented ultrafast adsorption, with a maximum MB sorption capacity of 606.2 mg·g−1, which was competitive with most of reported sorbents. Owing to the photothermal conversion property, LaB6 powder not only accelerated the melting of iced MB solution, but also achieved the efficient adsorption of MB in cold weather. LaB6 nanoparticles could be used as an attractive sorbent for dye removal from contaminated water, because of its facile synthesis process, excellent sorption performance and photothermal conversion property.

Graphene oxide was prepared using the modified Hummers method and reduced graphene oxide (rGO) - titanium dioxide (TiO2) nanocomposite was synthesised using the one-step hydrothermal treatment. The synergistic effects on adsorption and photocatalytic properties of the rGO-TiO2 nanocomposite for the humic acid removal were systematically investigated. The results of X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), and Raman and infrared (IR) spectroscopy indicate that GO was partially reduced to reduced graphene oxide (rGO) in the hydrothermal synthesis process and anatase TiO2 nanoparticles uniformly grew on the surface of rGO. The photoelectron and photohole generated under visible light irradiation were effectively separated on the surface of rGO-TiO2. The rGO-TiO2 nanocomposite exhibited higher photocatalytic activity as a result of the synergistic effects of surface functional groups for adsorption and the excellent conductivity for photocatalytic reaction. The effect of rGO-TiO2 nanocomposite dosage, light intensity and system temperature on the removal of humic acid solution was investigated. The results show that the removal efficiency of humic acid increased with system temperature and light intensity. When the dosage of rGO-TiO2 nanocomposite was 1.2 g/L, the temperature, the light intensity and the pH of this system was 303 K, 4.37 Wm−2 and 7, respectively, the removal efficiency of humic acid reached 88.7% under visible light irradiation.

Livestock farms are a propagating source of antibiotic resistance genes (ARGs) via the discharge of animal waste carrying resistant bacteria. Groundwater is widely used in traditional livestock farming. However, there is little available information on ARGs in pumped well water. To investigate the dissemination of ARGs in well water and to assess the impact of rainfall on ARGs, the abundance and diversity of ARGs on a long-term co-livestock feedlot farm where both pig and fish farming is conducted were analysed during the dry and wet seasons in this study, and twenty ARGs were detected. The total concentrations of the target ARGs (∑ARGs) in piggery wastewater and fishponds were approximately 109 copies/mL and 106 copies/mL, respectively. The wastewater treatment plant at this farm failed to avoid ARG contamination, and the biogas digester and lagoon treatment showed low abilities to eliminate ∑ARGs. The open operation mode of the lagoon and fishpond facilitated the dissemination of ARGs, which was assisted by the nearby stream, especially in the wet season. Additionally, the tetracycline resistance gene tetB showed the highest levels among the target ARGs in well water, which were higher in the wet season than the dry season. Affected by rainfall, the concentrations of ARGs in the samples rose, and the correlation between surface water and groundwater increased. These results suggested that both the surface water and the well water in the livestock farming area harboured serious ARGs contamination, which could pose ecological risks and cause food safety problems.

Melon, Cucumis melo L., is an important horticultural crop with abundant morphological variability, but the genetic diversity and relationships within wild and cultivated melons remain unclear to date. In this study, thick-skinned (TC) (cultivated subspecies melo), thin-skinned (TN) (cultivated subspecies agrestis), and wild accessions were analyzed for genetic diversity and relationships using 36 microsatellite markers. A total of 314 alleles were detected with a mean allelic number of 8.72 and polymorphism information content of 0.67. Cluster analysis of the accessions resulted in four distinct clusters (I, II, III, and IV) broadly matching with the TC, TN, and wild groups. Cluster I contained only two Indian wild accessions. Cluster II was consisted of 49 South Asian accessions, 34 wild accessions, and 15 TN accessions. Cluster III was a typical TC group including 51 multiorigin TC accessions and one wild accession. The remaining 88 accessions, including 75 TN accessions, 6 wild accessions, and 7 TC accessions, formed the cluster IV, and all the TN and wild accessions in this cluster were from China. These findings were also confirmed by Principal component analysis and STRUCTURE analysis. The South Asian subspecies agrestis accessions, wild and cultivated, had close genetic relationships with a distinctive genetic background. Chinese wild melons showed closeness to cultivated subspecies agrestis landraces and could be a return from the indigenous cultivated melons. The AMOVA and pairwise F statistics (FST) presented genetic differentiation among the three groups, with the strongest differentiation (FST = 0.380) between TC and TN melons. These results offer overall information on genetic diversity and affiliations within a variety of melon germplasms and favor efficient organization and utilization of these resources for the current breeding purpose.

Recently, design of cost-effective multifunctional electromaterials for supercapacitors and oxygen evolution reaction (OER) and enhancing their functionalities have become an emphasis in energy storage and conversion. Herein, a series of cheap and functional phosphate composites with different ratios of cobalt and nickel are synthesized using a simple polyalcohol refluxing method, and their excellent capacity and OER properties are systematically studied. Notably, owing to the different major role of Co and Ni elements in the phosphate composites for capacity and OER, the optimal electroconductibility, structural adjustment, electrochemical active sites, and activities for capacity and OER are obtained from the composites with the different ratios of Co/Ni. In addition, using high-capacity BiPO4 (BPO) as the negative electrodes, the new type of all-phosphate asymmetric supercapacitor (CNPO-40//BPO) shows a high energy density and reaches 36.84 W h kg-1 at a power density of 254.52 W kg-1. Its cyclic stability is also more excellent than that of the CNPO-40//AC device using commercial activated carbon as the negative electrodes. This study is beneficial to the more in-depth research on efficient dual-function electromaterials in capacity and OER and provides a high-efficient way to improve the practicality of asymmetric supercapacitors using the high-capacity Bi-based electromaterials as the negative electrodes.

Acoustic radiation force impulse (ARFI) imaging, shear wave elasticity imaging (SWEI), high intensity focused ultrasound (HIFU) and other ultrasonic diagnostic and treatment techniques can be realized according to different intensities of ultrasonic excitation of biological tissues. A multi-physics coupling simulation platform has been established, which allows researchers in academia and industry to simulate the response of different intensity ultrasonic excitations in isotropic linear elasticity media. Combining the acoustic module, solid mechanics module, electrical module and thermal module, the displacement response caused by low-intensity focused ultrasound excitation and the temperature response caused by high-intensity focused ultrasound and the corresponding conductivity change are simulated. The simulation platform developed is suitable for ARFI, SWEI, HIFU as well as non-invasive temperature monitoring and efficacy evaluating of HIFU. It provides guidance for understanding how the characteristics and parameters may affect the response of ultrasound excitation with different intensity.

Background: Antibiotic resistance bacteria (ARB) and antibiotic resistance genes (ARGs) have been considered as emerging contaminants, which even might be closely related to human health.Methods: To investigate the disease-producing risk of ARB and the horizontal gene transfer (HGT) risks of both extracellular ARGs (eARGs) and intracellular ARGs (iARGs), an in vitro digestion model was established to simulate the process of ARB and ARGs going through digestive tract. CTC/DAPI-FCM assay was used to study the survival of ARB during digestion, and the changes of genes (including tetA, tetG, tetM, sul1, sul2, bla EBC, bla FOX, intI1 and 16S rRNA) were determined by QPCR.Results: The results showed that ARB were mostly affected by pH of gastric juice. About 99% ARB (total population of 2.45 × 109–2.54 × 109) were killed by the gastric juice of pH 2.0 for the severely damage of bacterial cell membrane, but more than 80% ARB (total population of 2.71 × 109–3.90 × 109) were still alive with intact cell membrane when the pH of gastric juice increased to 3.0 and above. ARGs, intI1 and 16S rRNA could be detectable even at extreme pH when most bacteria died. The eARGs (accounting for 0.03%–24.56% of total genes) were less than iARGs obviously. The eARGs showed greater HGT potential than that of iARGs, suggesting transformation occurs more easily than conjugation. The transfer potential followed the order as: tet (100%) > sul (75%) > bla (58%), related to the high correlation of intI1 with tetA and sul2 (p < 0.01). Moreover, gastric juice of pH 1.0 could decrease the transfer frequency of ARGs by 2–3 order of magnitude compared to the control, but still threatening human health.Conclusions: Under the treatment of digestive juice, ARGs still have high gene horizontal transfer potential, suggesting that food-borne ARB pose a risk of ARGs horizontal transfer to intestinal bacteria.
