Histones serve many purposes in eukaryotic cells in the regulation of diverse genomic processes, including transcription, replication, DNA repair, and chromatin organization. As such, experimental systems to assess histone function are fundamental resources toward elucidating the regulation of activities occurring on chromatin. One set of important tools for investigating histone function are histone replacement systems, in which endogenous histone expression can be partially or completely replaced with a mutant histone. Histone replacement systems allow systematic screens of histone regulatory functions and the direct assessment of functions for histone residues. In this review, we describe existing histone replacement systems in model organisms, the benefits and limitations of these systems, and opportunities for future research with histone replacement strategies.

Despite the broad array of roles for epigenetic mechanisms on regulating diverse processes in eukaryotes, no experimental system is currently available in plants for the direct assessment of histone function. In this work, we present the development of a genetic strategy in Arabidopsis (Arabidopsis thaliana) whereby modified histone H4 transgenes can completely replace the expression of endogenous histone H4 genes. Accordingly, we established a collection of plants expressing different H4 point mutants targeting residues that may be post-translationally modified in vivo. To demonstrate its utility, we screened this new H4 mutant collection to uncover substitutions in H4 that alter flowering time. We identified different mutations in the H4 tail (H4R17A) and the H4 globular domain (H4R36A, H4R39K, H4R39A, and H4K44A) that strongly accelerate the floral transition. Furthermore, we identified a conserved regulatory relationship between H4R17 and the ISWI chromatin remodeling complex in plants: As with other biological systems, H4R17 regulates nucleosome spacing via ISWI. Overall, this work provides a large set of H4 mutants to the plant epigenetics community that can be used to systematically assess histone H4 function in Arabidopsis and a roadmap to replicate this strategy for studying other histone proteins in plants.

Transformation viaAgrobacterium tumefaciens(Agrobacterium) is the predominant method used to introduce exogenous DNA into plants. Transfer DNA (T-DNA) originating from Agrobacterium can be integrated as a single copy or in concatenated forms in plant genomes, but the mechanisms affecting final T-DNA structure remain unknown. In this study, we demonstrate that the inclusion of retrotransposon (RT)-derived sequences in T-DNA can increase transgene copy number by more than 50-fold inArabidopsis thaliana(Arabidopsis). RT-mediated amplification of T-DNA results in large concatemers in the Arabidopsis genome, which are primarily induced by the long terminal repeats (LTRs) of RTs. T-DNA amplification is dependent on the activity of DNA repair proteins associated with theta-mediated end joining (TMEJ). Finally, we show that T-DNA amplification can increase the frequency of targeted mutagenesis and gene targeting. Overall, this work uncovers molecular determinants that modulate T-DNA copy number in Arabidopsis and demonstrates the utility of inducing T-DNA amplification for plant gene editing.

The tail of replication-dependent histone H3.1 varies from that of replication-independent H3.3 at the amino acid located at position 31 in plants and animals, but no function has been assigned to this residue to demonstrate a unique and conserved role for H3.1 during replication. We found that TONSOKU (TSK/TONSL), which rescues broken replication forks, specifically interacts with H3.1 via recognition of alanine 31 by its tetratricopeptide repeat domain. Our results indicate that genomic instability in the absence of ATXR5/ATXR6-catalyzed histone H3 lysine 27 monomethylation in plants depends on H3.1, TSK, and DNA polymerase theta (Pol θ). This work reveals an H3.1-specific function during replication and a common strategy used in multicellular eukaryotes for regulating post-replicative chromatin maturation and TSK, which relies on histone monomethyltransferases and reading of the H3.1 variant.

Genetic variants affectingHeterogeneous Nuclear Ribonucleoprotein U (HNRNPU)have been identified in several neurodevelopmental disorders (NDDs). HNRNPU is widely expressed in the human brain and shows the highest postnatal expression in the cerebellum. Recent studies have investigated the role ofHNRNPUin cerebral cortical development, but the effects ofHNRNPUdeficiency on cerebellar development remain unknown. Here, we describe the molecular and cellular outcomes ofHNRNPUlocus deficiency duringin vitroneural differentiation of patient-derived and isogenic neuroepithelial stem cells with a hindbrain profile. We demonstrate thatHNRNPUdeficiency leads to chromatin remodeling of A/B compartments, and transcriptional rewiring, partly by impacting exon inclusion during mRNA processing. Genomic regions affected by the chromatin restructuring and host genes of exon usage differences show a strong enrichment for genes implicated in epilepsies, intellectual disability, and autism. Lastly, we show that at the cellular level.HNRNPUdownregulation leads to altered neurogenesis and an increased fraction of neural progenitors in the maturing neuronal population. We conclude that,HNRNPUlocus is involved in delayed commitment of neural progenitors to neuronal maturation in cell types with hindbrain profile.

The H3 methyltransferases ATXR5 and ATXR6 deposit H3.1K27me1 to heterochromatin to prevent genomic instability and transposon reactivation. Here, we report that atxr5 atxr6 mutants displayed robust resistance to Geminivirus. The viral resistance correlated with activation of DNA repair pathways, but not with transposon reactivation or heterochromatin amplification. We identified RAD51 and RPA1A as partners of virus-encoded Rep protein. The two DNA repair proteins showed increased binding to heterochromatic regions and defense-related genes in atxr5 atxr6 vs wild type plants. Consequently, the proteins had reduced interactions to viral DNA in the mutant, thus hampering viral replication. Additionally, RAD51 recruitment to the host genome arose via BRCA1, HOP2 and CYCB1, and this recruitment was essential for viral resistance in atxr5 atxr6. Thus, Geminiviruses adapt to healthy plants by hijacking its DNA repairing pathways for replication, but the host could retain DNA repairing proteins via sacrificing its genome stability to suppress viral infection.

Flowering of the reference legume Medicago truncatula is promoted by winter cold (vernalization) followed by long-day photoperiods (VLD) similar to winter annual Arabidopsis. However, Medicago lacks FLC and CO, key regulators of Arabidopsis VLD flowering. Most plants have two INHIBITOR OF GROWTH (ING) genes (ING1 and ING2), encoding proteins with an ING domain with two anti-parallel alpha-helices and a plant homeodomain (PHD) finger, but their genetic role has not been previously described. In Medicago, Mting1 gene-edited mutants developed and flowered normally, but an Mting2-1 Tnt1 insertion mutant and gene-edited Mting2 mutants had developmental abnormalities including delayed flowering particularly in VLD, compact architecture, abnormal leaves with extra leaflets but no trichomes, and smaller seeds and barrels. Mting2 mutants had reduced expression of activators of flowering, including the FT-like gene MtFTa1, and increased expression of the candidate repressor MtTFL1c, consistent with the delayed flowering of the mutant. MtING2 overexpression complemented Mting2-1, but did not accelerate flowering in wild type. The MtING2 PHD finger bound H3K4me2/3 peptides weakly in vitro, but analysis of gene-edited mutants indicated that it was dispensable to MtING2 function in wild-type plants. RNA sequencing experiments indicated that7000 genes are mis-expressed in the Mting2-1 mutant, consistent with its strong mutant phenotypes. Interestingly, ChIP-seq analysis identified5000 novel H3K4me3 locations in the genome of Mting2-1 mutants compared to wild type R108. Overall, our mutant study has uncovered an important physiological role of a plant ING2 gene in development, flowering, and gene expression, which likely involves an epigenetic mechanism.

Epigenetic marks are reprogrammed in the gametes to reset genomic potential in the next generation. In mammals, paternal chromatin is extensively reprogrammed through the global erasure of DNA methylation and the exchange of histones with protamines1,2. Precisely how the paternal epigenome is reprogrammed in flowering plants has remained unclear since DNA is not demethylated and histones are retained in sperm3,4. Here, we describe a multi-layered mechanism by which H3K27me3 is globally lost from histone-based sperm chromatin in Arabidopsis. This mechanism involves the silencing of H3K27me3 writers, activity of H3K27me3 erasers and deposition of a sperm-specific histone, H3.10 (ref. 5), which we show is immune to lysine 27 methylation. The loss of H3K27me3 facilitates the transcription of genes essential for spermatogenesis and pre-configures sperm with a chromatin state that forecasts gene expression in the next generation. Thus, plants have evolved a specific mechanism to simultaneously differentiate male gametes and reprogram the paternal epigenome.

Background The three-dimensional nuclear arrangement of chromatin impacts many cellular processes operating at the DNA level in animal and plant systems. Chromatin organization is a dynamic process that can be affected by biotic and abiotic stresses. Three-dimensional imaging technology allows to follow these dynamic changes, but only a few semi-automated processing methods currently exist for quantitative analysis of the 3D chromatin organization. Results We present an automated method, Nuclear Object DetectionJ (NODeJ), developed as an imageJ plugin. This program segments and analyzes high intensity domains in nuclei from 3D images. NODeJ performs a Laplacian convolution on the mask of a nucleus to enhance the contrast of intra-nuclear objects and allow their detection. We reanalyzed public datasets and determined that NODeJ is able to accurately identify heterochromatin domains from a diverse set of Arabidopsis thaliana nuclei stained with DAPI or Hoechst. NODeJ is also able to detect signals in nuclei from DNA FISH experiments, allowing for the analysis of specific targets of interest. Conclusion and availability NODeJ allows for efficient automated analysis of subnuclear structures by avoiding the semi-automated steps, resulting in reduced processing time and analytical bias. NODeJ is written in Java and provided as an ImageJ plugin with a command line option to perform more high-throughput analyses. While the links between wellbeing and performance are well demonstrated, the topic of CEO wellbeing has received little attention in the research literature. This study offers insights into how CEOs experience and recover from a crisis of wellbeing. Three male CEOs from the professional services industry were interviewed. Three themes emerged from an interpretative phenomenological analysis of the data: energy management, increased responsibility and relationships. Results demonstrate the potential for post-traumatic growth and highlight the need for updated models of resilience and leadership. Pragmatic elements were added to the IPA process in order to arrive at suggestions on how CEOs may better manage and sustain their wellbeing. Implications for the organisation as well as suggested future research are discussed.

Introduction Le diagnostic de melanome est l’un des points les plus cruciaux de la prise en charge dermatologique. Recemment, le developpement d’algorithmes de deep learning pour le diagnostic de tumeurs cutanees malignes et en particulier de melanome amorce une revolution dans le diagnostic par le dermatologue. Cependant, la plupart des algorithmes des etudes publiees ont ete developpes sur des bases de donnees publiques. Nous avons developpe un algorithme destine au diagnostic de tumeurs cutanees malignes a partir des photographies de vie reelle, prises dans le cadre de soin. Materiel et methodes Les donnees utilisees pour le developpement et la validation de l’algorithme sont la banque d’images dermoscopiques constituees dans le cadre du soin a l’hopital Saint-Louis, comprenant 985 images dermoscopiques annotees pour le diagnostic par 2 medecins diplomes (aspect clinique, et resultat histologique pour les tumeurs malignes). La seconde base est la base publique de photos dermoscopiques ISIC comprenant 24 464 images dermoscopiques. Nous avons entraine un algorithme (un reseau de neurones convolutif de type EfficientNet-B1) a l’identification des tumeurs malignes (TM) (melanome, carcinomes) versus benignes (naevus, HCF, tumeurs vasculaires benignes) (TB), et au diagnostic de lesions melanocytaires ou non melanocytaires, au sein de la base locale. Nous avons evalue la performance de l’algorithme entre differentes bases de donnees en l’entrainant sur la base de donnees ISIC et en le testant sur la base locale. Resultats Notre algorithme entraine sur la base ISIC a une precision diagnostique (aire sous la courbe ROC) de 0,90 sur la base ISIC, et de 0,79 pour le diagnostic de TM vs TB sur notre base locale. Pour identifier les facteurs participant a cette baisse de performance, nous avons analyse 33 photos « mal classees » pour le diagnostic de tumeurs melanocytaires ou non par l’algorithme. Les principales sources d’erreur sont les lentigos (classes en non melanocytaire) et les CBC tatoues et les keratoses seborrheiques (classe en melanocytaire). L’analyse de 33 photos mal classees a permis d’identifier des erreurs recurrentes: biopsies anterieures remaniant la lesion (7), trait de crayon sur la photo (4), erreur d’annotation dans l’image (4), localisation muqueuse (1), erreur de diagnostic identique a celle du medecin prescripteur (8). Discussion Le developpement d’un algorithme sur une base de photos issues de la vie reelle permet d’identifier les facteurs pouvant alterer ses performances, et qu’il faudra prendre en compte lors du developpement de ce type d’outils aupres des dermatologues. Le developpement de l’algorithme va etre poursuivi par un apprentissage federe entre differentes bases de donnees hospitalieres pour ameliorer ses performances.

Meaning and purpose form a key pillar of psychological wellbeing. Individuals and organizations that score high on these metrics have been found to exhibit a range of positive effects which in turn impact the ‘triple bottom line’ (people, profits, planet). This chapter introduces meaning as a multi-faceted construct and outlines how Steger’s SPIRE and CARMA models can be applied in a one-to-one coaching setting to benefit individuals and help leaders create the conditions for meaningful work, respectively. Several practical methods and techniques on how to measure, identify, create and shape meaning through coaching are presented along with specific questions a coach may ask and a case study to demonstrate how the literature on meaning may inform coaching practice. Consideration also is given to whom meaning-centred coaching benefits most, placing meaning-oriented positive psychology coaching within a future-focused organizational context.

ObjectivesThis study aimed to explore how eight female leaders from four Arab countries experienced their leadership journey, and to consider a potential role for coaching in that developmental journey.Design and MethodA qualitative design was applied to explore the participants’ experiences. Data collection was through semi-structured interviews, and interpretative phenomenological analysis (IPA) was used to analyse the data.ResultsFour themes emerged: (i) a continuous, exhausting struggle; (ii) fulfilment and pride in achievements; (iii) adoption of coping strategies; (iv) engagement in personal and professional development.ConclusionThe findings suggest that coaching may be an appropriate intervention to increase the individual leadership potential of females in the Arab world.

ObjectivesCoaches have an unclear role and the industry shares a complex border with therapeutic practices. This study explored the nature of the relationship between coaching and therapeutic practices, how coaching professionals experience, navigate and manage this boundary, and sought to identify what roles they adopt.DesignSeven International Coaching Federation Master Certified Coaches self-selected to participate. Sixty-minute semi-structured interviews were conducted using an initial interview schedule comprised of 15 questions.MethodA qualitative methodology was adopted to explore the social reality and operation of coaches within their professional context. A grounded theory method of data collection and analysis was utilised to explore gaps in the profession’s current understanding of roles and boundaries.ResultsA range of borders and boundaries were identified. The nature of the relationship between coaching and therapeutic practices appears to hinge on each coach’s self-selected boundaries. Such boundaries appear to be delineated most often by a coach’s feelings of competency and their client’s ability to process and move forward within coaching.ConclusionsFindings suggest that the main difference between coaching and therapeutic practice is driven by how coaches perceive the work they do as opposed to differences in the processes, content and roles.

These assays provide information regarding the modification sites on histones, the product specificity, and the impact of other histone or nucleotide modifications on enzyme activity. In particular, histone methyltransferase (HMT) assays have been instrumental in elucidating the activity and site specificity of many plant HMT enzymes. In this chapter, we describe a general protocol that can be used to perform HMT assays using different chromatin substrates, detection methods, and enzymes directly purified from plant material or heterologous sources.

Summary The CRISPR/Cas9 system has greatly improved our ability to engineer targeted mutations in eukaryotic genomes. While CRISPR/Cas9 appears to work universally, the efficiency of targeted mutagenesis and the adverse generation of off-target mutations vary greatly between different organisms. In this study, we report that Arabidopsis plants subjected to heat stress at 37°C show much higher frequencies of CRISPR-induced mutations compared to plants grown continuously at the standard temperature (22°C). Using quantitative assays relying on green fluorescent protein (GFP) reporter genes, we found that targeted mutagenesis by CRISPR/Cas9 in Arabidopsis is increased by approximately 5-fold in somatic tissues and up to 100-fold in the germline upon heat treatment. This effect of temperature on the mutation rate is not limited to Arabidopsis, as we observed a similar increase in targeted mutations by CRISPR/Cas9 in Citrus plants exposed to heat stress at 37°C. In vitro assays demonstrate that Cas9 from Streptococcus pyogenes (SpCas9) is more active in creating double-stranded DNA breaks at 37°C than at 22°C, thus indicating a potential contributing mechanism for the in vivo effect of temperature on CRISPR/Cas9. This study reveals the importance of temperature in modulating SpCas9 activity in eukaryotes, and provides a simple method to increase on-target mutagenesis in plants using CRISPR/Cas9.

Eukaryotic centromeres contain the kinetochore, which connects chromosomes to the spindle allowing segregation. During meiosis centromeres are suppressed for crossovers, as recombination in these regions can cause chromosome mis-segregation. Plant centromeres are surrounded by repetitive, transposon-dense heterochromatin that is epigenetically silenced by histone 3 lysine 9 dimethylation (H3K9me2), and DNA methylation in CG and non-CG sequence contexts. Here we show that disruption of Arabidopsis H3K9me2 and non-CG DNA methylation pathways increases meiotic DNA double strand breaks (DSBs) within centromeres, whereas crossovers increase within pericentromeric heterochromatin. Increased pericentromeric crossovers in H3K9me2/non-CG mutants occurs in both inbred and hybrid backgrounds, and involves the interfering crossover repair pathway. Epigenetic activation of recombination may also account for the curious tendency of maize transposonDsto disruptCHROMOMETHYLASE3when launched from proximal loci. Thus H3K9me2 and non-CG DNA methylation exert differential control of meiotic DSB and crossover formation in centromeric and pericentromeric heterochromatin.

Making a Histone Mark The covalent marks on histones (the principal components of chromatin) play a critical role in the regulation of gene expression. Somehow these marks are preserved when a cell in a tissue divides so that the daughter cells maintain the gene expression program and tissue identity of the parent cell. Jacob et al. (p. 1249 ) show that the Arabidopsis histone methylase ATXR5 is specific for the replication-dependent histone variant H3.1 and maintains the repressive histone H3 lysine-27 methyl mark on the H3.1 variant during genome replication, thus, preserving cell-type–specific regions of heterochromatin and gene repression through cell division and beyond.

In this paper, we present good practices of applying and extending Random Decision Forests (RDFs) for the 3D modelling of scenes where humans interact with moving, deformable and revolving objects in a professional context. We apply our method to two use-cases; the first is in the industrial context of the luxury leather good production while the second is in an atelier specialised in the wheel-throwing art of pottery. In the first use-case we use a single RDF, while for the second one of pottery, we extend the typical application of RDFs, by introducing the Hierarchical Random Decision Forests (HRDFs). More precisely, we use three RDFs in a tree structure architecture. The parent RDF is used to create a rough initial segmentation of the scene, while the two children RDFs are used to further classify the regions of the left and right arm, hand and fingers respectively. Results demonstrate that the proposed algorithm is sufficient for the accurate classification of scenes where humans interact with objects by using hand gestures in both simple and complex scenarios.

Germline development and early embryogenesis in eukaryotes are characterized by large-scale genome reprogramming events. In companion cells of the Arabidopsis male gametophyte, epigenome reorganization leads to loss of heterochromatin and production of a distinct small RNA (sRNA) population. A specific class of sRNA derived from transposons appears to be mobile and can accumulate in germ cells. In the germline of maize, rice, and Arabidopsis, specific ARGONAUTE-sRNA silencing complexes appear to play key roles in reproductive development, including meiosis and regulation of germ cell fate. These results reveal new roles for sRNAs during plant reproduction and suggest that mobility of sRNAs could be critical for some of these functions.

Post-translational modifications of histones play key roles in the regulation of gene expression and chromatin structure in eukaryotes. Methylation of histone 3 on lysine 27 (H3K27) is one of the most common and well-studied histone post-translational modifications. The vast majority of research on this histone residue, however, has focused on the trimethylated form (H3K27me3). Despite occurring at higher levels than H3K27me3 in animals and plants, the monomethylated form of H3K27 (H3K27me1) remains relatively poorly characterized. The absence of information concerning H3K27me1 is due in large part to the fact that the enzymes catalyzing this epigenetic mark were only recently identified. In this article, we highlight new findings concerning H3K27me1, including the identification of two plant-specific H3K27 monomethyltransferases that are required for gene silencing and heterochromatin condensation. We also discuss the emerging similarities and differences in H3K27 methylation in plant and animal systems.

Nuclear pore complexes (NPCs) mediate the transport of RNA and other cargo between the nucleus and the cytoplasm. In vertebrates, the NPC protein TRANSLOCATED PROMOTER REGION (TPR) is associated with the inner filaments of the nuclear basket and is thought to serve as a scaffold for the assembly of transport machinery. In a screen for mutants that suppress the expression of the floral inhibitor FLOWERING LOCUS C, we identified lesions in the Arabidopsis (Arabidopsis thaliana) homolog of TPR (AtTPR). attpr mutants exhibit early-flowering and other pleiotropic phenotypes. A possible explanation for these developmental defects is that attpr mutants exhibit an approximately 8-fold increase in nuclear polyA RNA. Thus AtTPR is required for the efficient export of RNA from the nucleus. Microarray analysis shows that, in wild type, transcript abundance in the nuclear and total RNA pools are highly correlated; whereas, in attpr mutants, a significantly larger fraction of transcripts is enriched in either the nuclear or total pool. Thus AtTPR is required for homeostasis between nuclear and cytoplasmic RNA. We also show that the effects of AtTPR on small RNA abundance and auxin signaling are similar to that of two other NPC-associated proteins, HASTY (HST) and SUPPRESSOR OF AUXIN RESISTANCE3 (SAR3). This suggests that AtTPR, HST, and SAR3 may play related roles in the function of the nuclear pore.

Stalled bacterial ribosomes are freed when they switch to the translation of transfer-messenger RNA (tmRNA). This process requires the tmRNA-binding and ribosome-binding cofactor SmpB, a beta-barrel protein with a protruding C-terminal tail of unresolved structure. Some plastid genomes encode tmRNA, but smpB genes have only been reported from bacteria. Here we identify smpB in the nuclear genomes of both a diatom and a red alga encoding a signal for import into the plastid, where mature SmpB could activate tmRNA. Diatom SmpB was active for tmRNA translation with bacterial components in vivo and in vitro, although less so than Escherichia coli SmpB. The tail-truncated diatom SmpB, the hypothetical product of a misspliced mRNA, was inactive in vivo. Tail-truncated E. coli SmpB was likewise inactive for tmRNA translation but was still able to bind ribosomes, and its affinity for tmRNA was only slightly diminished. This work suggests that SmpB is a universal cofactor of tmRNA. It also reveals a tail-dependent role for SmpB in tmRNA translation that supersedes a simple role of linking tmRNA to the ribosome, which the SmpB body alone could provide.

It has been postulated that a highly reduced form of transfer messenger RNA (tmRNA), a bacterial molecule involved in the rescue of stalled ribosomes during translation, is expressed in the mitochondrion of the jakobid Reclinomonas americana. Here we show that genes encoding both one-piece and two-piece tmRNAs are present in six different jakobid mitochondrial DNAs. Mitochondrial tmRNAs have retained the highly conserved tRNAAla-like domain, but they apparently lack the mRNA-like region present in all bacterial tmRNAs. Comparative analysis of jakobid mitochondrial genomes shows that a potential mRNA-like region in R. americana (orf64) is located at distant genomic positions in other jakobids. Our results strongly suggest that orf64 is a tatA homolog. Through Northern hybridization we confirm the postulated reduced size of both a one-piece tmRNA in Jakoba libera and a two-piece tmRNA in Seculamonas ecuadoriensis. The J. libera tmRNA is post-transcriptionally modified by addition of a 3′ CCA tail, processed in vitro by RNase P RNA, and specifically charged with alanine in vitro by alanyl-tRNA synthetase. Our results strongly support the functionality of these reduced mitochondrial tmRNAs.

In eukaryotes, most genetic material resides in a separate membrane-bound compartment known as the nucleus. Transport of cargo, such as RNA and protein, across this barrier is facilitated by the nuclear pore complex (NPC). In the July issue of Plant Physiology, we showed that a component of the NPC, Arabidopsis thaliana TPR (AtTPR), is required for normal development. Two striking phenotypes of attpr mutants are that they are early flowering and show an accumulation of polyadenylated RNA in the nucleus. In addition, the expression of several microRNAs (miRNAs) is reduced in attpr mutants. In this addendum, we have examined the effect of AtTPR on the expression of miRNA targets. Our results show that miRNA targets are more likely to be upregulated than other transcripts in attpr mutants. For example, when comparing the nuclear RNA pool between wild-type and attpr plants, we found that 75% of the miRNA targets showing a significant change in transcript level are upregulated in attpr mutants. Although the targets of some miRNAs were upregulated, other miRNA targets were relatively unaffected by attpr mutations. Thus it appears that AtTPR may be required for the proper expression or localization of a subset of miRNAs.

The autonomous floral promotion pathway plays a key role in the regulation of flowering in rapid-cycling Arabidopsis (Arabidopsis thaliana) by providing constitutive repression of the floral inhibitor FLOWERING LOCUS C (FLC). As a result, autonomous pathway mutants contain elevated levels of FLC and are late flowering. Winter annual Arabidopsis, in contrast, contain functional alleles of FRIGIDA (FRI), which acts epistatically to the autonomous pathway to up-regulate FLC and delay flowering. To further explore the relationship between FRI and the autonomous pathway, we placed autonomous pathway mutants in a FRI-containing background. Unexpectedly, we found that a hypomorphic allele of the autonomous pathway gene fy (fy null alleles are embryo lethal) displayed background-specific effects on FLC expression and flowering time; in a rapid-cycling background fy mutants contained elevated levels of FLC and were late flowering, whereas in a winter annual background fy decreased FLC levels and partially suppressed the late-flowering phenotype conferred by FRI. Because FY has been shown to have homology to polyadenylation factors, we examined polyadenylation site selection in FLC transcripts. In wild type, two polyadenylation sites were detected and used at similar levels. In fy mutant backgrounds, however, the ratio of products was shifted to favor the distally polyadenylated form. FY has previously been shown to physically interact with another member of the autonomous pathway, FCA. Interestingly, we found that fy can partially suppress FLC expression in an fca null background and promote proximal polyadenylation site selection usage in the absence of FCA. Taken together, these results indicate novel and FCA-independent roles for FY in the regulation of FLC.

It is important for the normal function of a cell that DNA replication takes place only once per cell cycle, and various mechanisms exist to prevent its repetition. Another mechanism has been shown to operate in Arabidopsis, this one surprisingly involving two histone (H3K27) monomethyltansferases, ATXR5 and ATXR6. Mutations in the genes encoding these two enzymes lead to re-replication of specific genomic locations, the majority of which correspond to transposons and other repetitive and silenced elements. ATXR5 and ATXR6 are proposed to be components of a pathway required to prevent over-replication of heterochromatin in Arabidopsis. DNA replication occurs only once per cell cycle, and numerous pathways prevent re-replication. Here it is shown that mutations in ARABIDOPSIS TRITHORAX-RELATED PROTEIN5 (ATXR5) and ATXR6 — which encode histone methyltransferases — lead to re-replication of specific genomic locations, notably those corresponding to transposons and other repetitive and silenced elements. ATXR5 and ATXR6 are proposed to be components of a pathway that prevents over-replication of heterochromatin in Arabidopsis. Multiple pathways prevent DNA replication from occurring more than once per cell cycle1. These pathways block re-replication by strictly controlling the activity of pre-replication complexes, which assemble at specific sites in the genome called origins. Here we show that mutations in the homologous histone 3 lysine 27 (H3K27) monomethyltransferases, ARABIDOPSIS TRITHORAX-RELATED PROTEIN5 (ATXR5) and ATXR6, lead to re-replication of specific genomic locations. Most of these locations correspond to transposons and other repetitive and silent elements of the Arabidopsis genome. These sites also correspond to high levels of H3K27 monomethylation, and mutation of the catalytic SET domain is sufficient to cause the re-replication defect. Mutation of ATXR5 and ATXR6 also causes upregulation of transposon expression and has pleiotropic effects on plant development. These results uncover a novel pathway that prevents over-replication of heterochromatin in Arabidopsis.

Depth information is provided by time of flight sensor with automotive certification. In particular, we develop our implementation of a Random Forest based posture classification in two subcases: micro gestures at the wheel and macro gestures in front of the touch screen.
