The suicide rate is increasing at an alarming pace, even among university students. Thus, this study attempted to identify suicidal ideation among university students through the perspective of love relationships (love satisfaction, love styles) and gender differences. A random sampling method has been carried out to collect the data. Analysis of the responses demonstrated that love relationship satisfaction was not associated with suicidal ideation among university students. On the other hand, there is a positive relationship between love styles and suicidal ideation among university students. Furthermore, love styles have a positive association with love relationship satisfaction. In addition, there is a significant difference between genders in suicidal ideation among university students, with females having higher suicidal ideation than males.

This study aimed to examine the effect of venetoclax coupled with azacytidine in treating older adults with relapsed and refractory (R/R) acute myeloid leukemia (AML). The clinical data of 10 senior patients with AML over 65 years old who were treated with venetoclax and azacytidine, including six patients with R/R AML, were retrospectively evaluated. This study comprised seven males and three females with a median age of 71 years. Five patients had at least one relapse, and one patient did not achieve remission after four cycles of azacytidine monotherapy, considering it resistant. AML with myelodysplasia-related changes was found in four cases. One of the 10 patients died early after 1-13 cycles of venetoclax plus azacytidine treatment due to a protracted period of neutropenia and severe lung infection induced by medications. Six of the remaining nine patients, including six R/R patients, achieved a complete remission (CR) or a CR with incomplete hematologic recovery (CRi). After two cycles of therapy, one patient did not react. Neutropenia lasted an average of 10.5 (6-15) days in all patients, with the most severe cases occurring in the second and third weeks of therapy. Three patients who tested positive for the TP53 gene mutation had the following outcomes: One relapsed patient has been in progression-free remission (PFS) for the past 24 months, whereas another has been in full remission but relapsed 2 months later. Another patient experienced complete remission in myelology for 4 months, but the variable allele fraction (VAF) value steadily rose, suggesting that the illness was on the verge of progressing. IDH2 gene alterations were found in three of four patients who obtained maintained CR for more than 18 months following recurrence. Venetoclax in combination with azacytidine is a successful and well-tolerated therapy for R/R AML in the elderly. Venetoclax and azacytidine may help patients with TP53 mutations and reduce VAF. The IDH2 mutation might be a good predictor of veneclax sensitivity. A notable adverse response in the treatment phase of the regimen is severe infection induced by neutropenia.

In convective quasi-equilibrium theory, tropical tropospheric temperature perturbations are expected to follow vertical profiles constrained by convection, referred to as A-profiles here, often approximated by perturbations of moist adiabats. Differences between an idealized A-profile based on moist-static energy conservation and temperature perturbations derived from entraining and nonentraining parcel computations are modest under convective conditions—deep convection mostly occurs when the lower troposphere is close to saturation, thus minimizing the impact of entrainment on tropospheric temperature. Simple calculations with pseudoadiabatic perturbations about the observed profile thus provide useful baseline A-profiles. The first EOF mode of tropospheric temperature (TEOF1) from the ERA-Interim and AIRS retrievals below the level of neutral buoyancy (LNB) is compared with these A-profiles. The TEOF1 profiles with high LNB, typically above 400 hPa, yield high vertical spatial correlation (∼0.9) with A-profiles, indicating that tropospheric temperature perturbations tend to be consistent with the quasi-equilibrium assumption where the environment is favorable to deep convection. Lower correlation tends to occur in regions with low climatological LNB, less favorable to deep convection. Excluding temperature profiles with low LNB significantly increases the tropical mean vertical spatial correlation. The temperature perturbations near LNB exhibit negative deviations from the A-profiles—the convective cold-top phenomenon—with greater deviation for higher LNB. In regions with lower correlation, the deviation from A-profile shows an S-like shape beneath 600 hPa, usually accompanied by a drier lower troposphere. These findings are robust across a wide range of time scales from daily to monthly, although the vertical spatial correlation and TEOF1 explained variance tend to decrease on short time scales.

Blockchain technology has been widely applied in numerous industries with its decentralization, verifiability, distributivity, and immutability. However, the identity privacy security of blockchain users is facing serious threats because of the openness of traditional blockchain transaction information. Moreover, numerous traditional cryptographic algorithms used by blockchain transaction networks are difficult to attack quantum computing. In this paper, we propose a new lattice-based blind ring signature scheme in allusion to completely anonymous blockchain transaction systems. There into, the blind ring signature can implement the complete anonymity of user identity privacy in blockchain transactions. Meanwhile, lattice cryptography can availably resist quantum computing attacks. Firstly, the proposed signature scheme has strong computational security based on the small integer solution (SIS) problem and a high sampling success rate by utilizing the techniques of rejection sampling from bimodal Gaussian distribution. Secondly, the proposed signature scheme can satisfy the correctness and security under the random oracle model, including anonymity, blindness, and one-more unforgeability. Thirdly, we construct a blockchain transaction system based on the proposed blind ring signature algorithm, which realizes the completely anonymous and antiquantum computing security of the blockchain users’ identity privacy. Finally, the performance evaluation results show that our proposed blind ring signature scheme has lower latency, smaller key size, and signature size than other similar schemes.

The Madden-Julian oscillation (MJO) has profound impacts on weather and climate phenomena, and thus changes in its activity have important implications under human-induced global climate change. Here, the time at which the MJO change signal emerges from natural variability under anthropogenic warming is investigated. Using simulations of the Community Earth System Model version 2 large ensemble forced by the shared socioeconomic pathways SSP370 scenario, an increase in ensemble mean MJO precipitation amplitude and a smaller increase in MJO circulation amplitude occur by the end of the 21st century, consistent with previous studies. Notably, the MJO precipitation amplitude change signal generally emerges more than a decade earlier than that of MJO wind amplitude. MJO amplitude changes also emerge earlier over the eastern Pacific than other parts of the tropics. Our findings provide valuable information on the potential changes of MJO variability with the aim of improving predictions of the MJO and its associated extreme events.

Self-assembled systems have recently attracted extensive attention because they can display a wide range of phase morphologies in nanocomposites, providing a new arena to explore novel phenomena. Among these morphologies, a bicontinuous structure is highly desirable based on its high interface-to-volume ratio and 3D interconnectivity. A bicontinuous nickel oxide (NiO) and tin dioxide (SnO2) heteroepitaxial nanocomposite is revealed here. By controlling their concentration, we fabricated tuneable self-assembled nanostructures from pillars to bicontinuous structures, as evidenced by TEM-energy-dispersive X-ray spectroscopy with a tortuous compositional distribution. The experimentally observed growth modes are consistent with predictions by first-principles calculations. Phase-field simulations are performed to understand 3D microstructure formation and extract key thermodynamic parameters for predicting microstructure morphologies in SnO2:NiO nanocomposites of other concentrations. Furthermore, we demonstrate significantly enhanced photovoltaic properties in a bicontinuous SnO2:NiO nanocomposite macroscopically and microscopically. This research shows a pathway to developing innovative solar cell and photodetector devices based on self-assembled oxides.

The traditional paradigm of viscosity-dominated evolution of protoplanetary discs has been recently challenged by magnetized disc winds. However, distinguishing wind-driven and turbulence-driven accretion through observations has been difficult. In this study, we present a novel approach to identifying their separate contribution to angular momentum transport by studying the gap and ring morphology of planet-forming discs in the ALMA continuum. We model the gap-opening process of planets in discs with both viscous evolution and wind-driven accretion by 2D multi-fluid hydrodynamical simulations. Our results show that gap-opening planets in wind-driven accreting discs generate characteristic substructures that differ from those in purely viscous discs. Specifically, we demonstrate that discs, where wind-driven accretion dominates the production of substructures, exhibit significant asymmetries. Based on the diverse outputs of mock images in the ALMA continuum, we roughly divide the planet-induced features into four regimes (moderate-viscosity dominated, moderate-wind dominated, strong-wind dominated, inviscid). The classification of these regimes sets up a potential method to constrain the strength of magnetized disc wind and viscosity based on the observed gap and ring morphology. Inspired by Roush's pioneering work on rare sugars, we have developed a scalable, stereoselective, de novo synthesis of orthogonally protected C2-fluoro digitoxose and cymarose, utilizing Sharpless kinetic resolution and organocatalytic fluorination as key steps. The utility of this strategy is demonstrated by the synthesis of a fluorinated analogue of digoxin, which indicates the fluorine on the sugar ring may have a significant impact on biological activity.

This study investigated the immediate biomechanical effects of a biter-width partial medial meniscectomy. Small tears on the free edge of the meniscus are occasionally discovered during arthroscopic knee procedures. Removal of these tears often requires no more width than that of a biter from the edge. While past cadaveric partial meniscectomy studies have reported on the effects of meniscectomies from resection of one-third to three-quarters of meniscal width, the biomechanical sequelae of substantially smaller meniscectomies remain unknown. This study was performed to determine how single-biter-width meniscectomies affect the pressure distribution and peak pressure on the tibial plateau, as these parameters are correlated with increased likelihood of development of osteoarthritis. Eight cadaveric knees were instrumented with a thin-film pressure sensor underneath the menisci while retaining the major ligaments and most of the capsule. Knees were loaded to 1000 N at angles of flexion of 0, 15, 30, and 45 in normal tibio-femoral angles and in 7° of varus. Pressure, peak pressure, force and contact area were recorded. Afterwards, biter-width meniscectomies were performed, and measures repeated. There were significant changes in contact area at most flexion and tibio-femoral angles. There were no changes in the amount of force that was transmitted through the medial compartment. The overall pressure on the tibial plateau did not change in most knee conditions and peak pressure did not change significantly in any condition. The changes in contact area were consistent with our expectations. Similarly, the lack of change in force was also expected. However, the lack of significant changes in pressure and peak pressure represent findings indicating that a biter sized meniscectomy may have clinically insignificant effects on biomechanics.

Treadmill exercise and mesenchymal stem cell transplantation are both practical and effective methods for the treatment of cerebral ischemia. However, whether there is a synergistic effect between the two remains unclear. In this study, we established rat models of ischemia/reperfusion injury by occlusion of the middle cerebral artery for 2 hours and reperfusion for 24 hours. Rat models were perfused with bone marrow mesenchymal stem cell-derived exosomes (MSC-exos) via the tail vein and underwent 14 successive days of treadmill exercise. Neurological assessment, histopathology, and immunohistochemistry results revealed decreased neuronal apoptosis and cerebral infarct volume, evident synaptic formation and axonal regeneration, and remarkably recovered neurological function in rats subjected to treadmill exercise and MSC-exos treatment. These effects were superior to those in rats subjected to treadmill exercise or MSC-exos treatment alone. Mechanistically, further investigation revealed that the activation of JNK1/c-Jun signaling pathways regulated neuronal apoptosis and synaptic-axonal remodeling. These findings suggest that treadmill exercise may exhibit a synergistic effect with MSC-exos treatment, which may be related to activation of the JNK1/c-Jun signaling pathway. This study provides novel theoretical evidence for the clinical application of treadmill exercise combined with MSC-exos treatment for ischemic cerebrovascular disease.

Background: Endothelial injury induced by low shear stress (LSS) is an initiating factor in the pathogenesis of various cardiovascular diseases, including atherosclerosis, hypertension, and thrombotic diseases. Low shear stress activates the mammalian target of rapamycin complex 2 (mTORC2) signaling pathway. Rictor, the main constituent protein of mTORC2, is involved in vascular development. However, the impact of conditional Rictor ablation on endothelial homeostasis, especially on endothelial-specific markers, such as vascular endothelial-cadherin (VE-cadherin) and von Willebrand factor (VWF), under blood flow stimulation is unclear.Objective: We aimed to investigate whether endothelial Rictor is involved in maintaining vascular endothelial integrity and the potential role of Rictor in atheroprone blood flow-mediated endothelial injury.Methods and results: Immunofluorescence staining showed that endothelial Rictor was successfully knocked out in a mouse model. Scanning electron microscopy (EM) detection revealed disruption of the endothelial monolayer in the thoracic aorta of Rictor-deficient mice. Furthermore, scanning electron microscopy and transmission electron microscopy showed that Rictor deletion disrupted endothelial integrity and expanded cell junctions in the left common carotid artery region. In vitro, low shear stress disrupted actin filament polarity and the promoted the translocation of vascular endothelial-cadherin, the key component of adherens junctions (AJs) in human umbilical vein endothelial cells. After Rictor downregulation by small interfering RNA, the translocation of vascular endothelial-cadherin and stress fibers increased. Rictor knockdown inhibited low shear stress-induced von Willebrand factor upregulation, and downregulation of vascular endothelial-cadherin decreased low shear stress-induced von Willebrand factor expression. These results suggest that vascular endothelial-cadherin/von Willebrand factor is a possible mechanism mediated by Rictor in the pathological process of low shear stress-induced endothelial injury.Conclusion: Rictor is a key protein that regulates endothelial integrity under vascular physiological homeostasis, and Rictor mediates low shear stress-induced endothelial injury by regulating adherens junctions and von Willebrand factor.

Background Radix Sophorae Flavescentis (Kushen) is the principal herb consisting of Compound Kushen Injection (CKI), which has been approved for the clinical treatment of tumors. Although CKI has been widely used, especially on non-small cell lung cancer (NSCLC), its underlying mechanisms of Kushen regarding microRNA-target gene-pathway remain unclear. Methods A549 cell line was selected as the NSCLC model and treated with different concentrations of Kushen to obtain the mRNA and miRNA expression profiles. Then differentially expressed (DE) genes and miRNAs were analyzed with miRTarBase, miRDB, and TargetScan 7.0. A DE miRNAs-DEGs network was finally constructed to find the critical miRNA-mRNA interaction influenced by Kushen. Furthermore, clinical significance and prognosis analysis of hub genes based on TCGA and Oncomine datasets were performed further to narrow down the hub genes and corresponding miRNAs. Finally, the effects of Kushen on critical miRNA-mRNA interaction and related pathways were verified by in vitro experiments, including qPCR, Western blot, miRNA mimic transfection assay, and dual-reporter luciferase assay. Results A DE miRNAs-DEGs network was constructed based on miRNA-target databases screening, with 16 DE miRs and 68 DEGs identified. With the help of clinical evaluation and qPCR experiment of hub genes, we focused on miR-183-5p/EGR1 and miR221-3p/FOS interactions. The qPCR results indicated that Kushen induced down-regulation of miR-183-5p and miR-221-3p, and up-regulation of EGR1 and FOS. Dual-luciferase reporter assay results demonstrated that EGR1 and FOS were direct targets of miR-183-5p and miR-221-3p in A549 cells, respectively. Furthermore, the up-regulation of EGR1 by Kushen was also accompanied by an increase in PTEN expression and a decrease in AKt expression. And the opposite effects of EGR1 on A549 were eliminated by miR-183-5p overexpression. However, the overexpression of miR-221-3p cannot induce FOS down-regulation. Conclusions Altogether, the above results supported that miR-183-5p /EGR1 and the PTEN/AKT pathway played a crucial role in the beneficial effects of Kushen on A549, which highlights the therapeutic potential of Kushen in the treatment of NSCLC.

PurposeThe relationship between the CDK5RAP3 and UFM1 expression and the prolonged outcomes of patients who underwent gastric cancer (GC) surgery was investigated.MethodsSingle-sample gene set enrichment analysis (ssGSEA), unsupervised clustering and other methods were used to verify the relationship between CDK5RAP3 and UFM1 in GC through public databases. Additionally, CDK5RAP3 and UFM1 expression in cancerous and paracancerous tissues of GC was analysed in the context of patient prognosis.ResultsCDK5RAP3 and UFM1 expression was downregulated synchronously, the interaction was observed between the two proteins, and UFM1 and CDK5RAP3 expression was found to be inversely associated to AKT pathway activation. Prognostic analysis showed that the prognosis is poorer for low CDK5RAP3 and UFM1 patients, than for high CDK5RAP3 and/or UFM1 (pConclusionsIt is confirmed in our findings that a combination of CDK5RAP3 and UFM1 can produce a more precise prediction model for GC patients’ survival.

Transmembrane protein, also known as integral membrane protein, can be distributed in the lipid bilayer or across the entire membrane, and it plays an important role in cell signal transduction. It has been discovered that multiple transmembrane proteins are involved in the regulation of tumor signals. Recent studies have revealed that the abnormal expression of some transmembrane protein is closely related to the occurrence, development and prognosis of non-Hodgkin's lymphoma (NHL), including programmed cell death protein 1 (PD-1), TMEM30A, NOTCH1, TOLL-like receptor (TLR), sphingosine-1-phosphate receptor, TRAIL, etc. The study on these transmembrane proteins and related genes has important clinical significance for the treatment and prognosis of NHL, and it may become a new therapeutic target. At present, there have been some research results in this field at home and abroad. The novel amphiphilic acylated dextran-g-polyisobutylene (AcyDex-g-PIB) graft copolymers with different branch lengths (Mn,PIB, 2600–5800 g/mol) and grafting numbers (GN, 5–28 per 1000 Dex monosaccharide) were successfully synthesized via the nucleophilic substitution of the hydroxyl (―OH) side groups along AcyDex backbone by the living PIB-THF4+ chains prepared through cationic polymerization. The crystallization of AcyDex backbone in AcyDex-g-PIB graft copolymers was confined due to the presence of PIB branches and the morphology changed from short rod-like crystals to fragment-like crystals with increasing Mn,PIB and GN. The obvious microphase separation occurred due to the incompatibility between hard AcyDex backbone and soft PIB branches. AcyDex-g-PIB graft copolymers exhibit excellent biocompatibility towards HeLa cells and good hemocompatibility with red blood cells (RBCs), both of which increase with increasing GN. The increases of water contact angle and roughness on the surface of the graft copolymers with increasing Mn,PIB and GN manifest the anti-protein adsorption performance. The amphiphilic AcyDex-g-PIB graft copolymers could self-assemble in aqueous solution into nanospheres, which can be used as pH-sensitive drug carriers and can release 100% of the loaded drug within 72 h at pH=7.4. AcyDex-g-PIB graft copolymers bearing silver nanoparticles (Ag-NPs, 0.8 wt%–3.9 wt%, 4.5–9.5 nm) show good antibacterial properties. This kind of amphiphilic graft copolymer would have a promising prospect in biological and medical fields.

Blind quantum computing (BQC) ensures that a classical client could delegate complex computing tasks to a remote quantum server safely. In order to detect the dishonest behavior of the participants, we present a verifiable multi-party universal BQC protocol in distributed networks. By using the stabilizer formalism, we propose an honesty check method to test the correctness of the graph states generated by the servers. The honesty of both the clients and the servers can be judged fairly with the help of the arbitrator. Moreover, a load balancer is introduced to control the possible breakdown of servers in the network. No-signaling principle ensures the unconditional security of the protocol. Through the use of universal resource states, our protocol can be applied in more multi-party verifiable universal BQC protocols. The failure management and workload balancing of the protocol also provide a promising prospect for the future BQC network.

Chelatococcus daeguensis strain M38T9 is a thermotolerant bacterium isolated from a hot-spring in Malaysia. The draft genome of C. daeguensis strain M38T9 consists of 4,218,658 bp assembled into 50 scaffolds. The GC content of the genome is 67.91 %, and the sequencing coverage of 184×. There are 4,046 predicted genes, 3,962 protein-coding genes, and 53 RNA-coding genes (tRNA: 45, rRNA: 4). The draft genome has been deposited at DDBJ/ENA/GenBank under the BioProject accession number PRJNA668056. The raw reads were deposited in the Sequence Read Archive (SRA) under accession number SRR13805582. Here we report the draft genome of this strain to expand our understanding of the genomic information available on the genus Chelatococcus.

A cluster randomized controlled trial of endoscopy-based screening for esophageal cancer (EC) and gastric cancer (GC) was conducted to evaluate the efficacy and feasibility of this strategy in a non-high-incidence rural area of China. The trial design and baseline findings are presented here.A total of 33 eligible villages in Luoshan County in Henan Province were assigned randomly to the intervention or control group in a 1:1 ratio by a computer-generated randomization list. Local residents aged 40 to 69 years were enrolled from the villages. Participants in the intervention group were risk-stratified with a questionnaire, and high-risk individuals were subsequently screened by endoscopy. The primary outcomes were EC and GC mortality. The secondary outcomes comprised the detection rate, stage distribution, and the treatment rate. In this study, baseline characteristics were assessed by a questionnaire. Multivariate logistic regression analysis was performed to explore factors associated with endoscopy compliance.Trial recruitment was completed in 2017, and ultimately, there were 12,475 and 11,442 participants allocated to the intervention (17 clusters) and the control group (16 clusters), respectively. We included 23,653 participants in the analysis, with 12,402 in the intervention group and 11,251 in the control group. A total of 6,286 (50.7%) participants in the intervention group were estimated as high-risk individuals, and 2,719 (43.3%) underwent endoscopy. Multivariate logistic regression analysis demonstrated that some factors including age, gender, education, personality and mental health, and upper gastrointestinal diseases or symptoms might affect endoscopy compliance. The detection rates for positive cases of EC and GC were 0.22% and 0.55%, respectively. The rates for esophageal and gastric precancerous lesions were 0.70% and 2.35%, respectively. The early detection rates for EC and GC were 50.0% and 33.3%, respectively. Additionally, the overall treatment rate for positive cases was 90.0%.The diagnostic yield of endoscopy-based screening for EC and GC was relatively low in a non-high-incidence rural area. The study may offer clues for the improvement of endoscopy compliance and the optimization of screening strategies for upper gastrointestinal cancer in non-high-incidence areas.Chinese Clinical Trial Registry ChiCTR-EOR-16008577.

It is great of significance to figure the time-trend of esophageal cancer (EC) and its current status for effective prevention and control, especially in EC high risk areas. As one of world-renowned high-risk areas, the epidemiology of EC in Henan has not been recently updated. Therefore, we aimed to depict the status quo of EC and analyze its time-trend in Henan.The EC data were extracted from the Henan Provincial Cancer registry database derived from the population based cancer registry system, which covered 30.51% of the whole population in Henan and were qualified according to national and international guidelines. The incidence and mortality of EC were estimated by area (rural/urban), gender, and age groups. The age-standardized rates (ASRs) were calculated according to the Segi's population. Joinpoint regression was used to calculate annual percentage change (APC) and average annual percentage change (AAPC) to evaluate the time-trend of EC.As estimated, there were 29,913 new EC cases in Henan, 2018. The crude incidence and the age-standardized incidence rate by world standard population (ASIRW) was 27.43/10The EC incidence and mortality in Henan has exhibited a significant declining trend in past years. Nonetheless, the disease burden remains high, especially in males and rural areas. Therefore, the ongoing prevention and control strategies of EC should be maintained alongside the establishment of more effective strategies.

However, due to growing concerns about the antibiotic residue in meat and eggs, phytogenic formulations are becoming an attractive approach to manage this disease. In this study, we investigated the anti-coccidial function and mechanism of phytogenic formulations composed of Bidens pilosa, Artemisia indica, and both used in combination. We found that these formulations increased the survival rate and reduced body weight loss, the feed conversion ratio, oocyst excretion, bloody stools, and gut lesions of chickens. Mechanistic studies showed that A. indica, but not B. pilosa, reduced the survival of Eimeria oocysts. Accordingly, they both inhibited oocyst sporulation and sporozoite invasion into Madin-Darby bovine kidney (MDBK) cells. Overall, we demonstrate that these formulations protect chickens against coccidiosis. Moreover, a combination of B. pilosa and A. indica has an additive effect on coccidiosis control and growth performance in chickens compared to either one used alone.
