Cervical cancer (CC) is the primary cancer-related cause of morbidity and mortality in women. Previous studies have shown that placenta-specific 8 (PLAC8) has different functions in multiple malignancies. This study aimed to explore the function and regulatory mechanism of PLAC8 in CC. Bioinformatics and immunohistochemical analyses demonstrated that PLAC8 was significantly upregulated in CC tissues compared with normal tissues. Gain/loss-of-function experiments showed that siRNA-mediated knockdown of PLAC8 suppressed cell migration and invasion, while PLAC8 overexpression promoted cell motility. Moreover, PLAC8 was revealed to affect the epithelial-mesenchymal transition (EMT) process by upregulating epithelial (E)-cadherin and decreasing the expression of mesenchymal markers of EMT, including vimentin, zinc finger E-box binding homeobox 1 (ZEB1), neural (N)-cadherin, matrix metalloproteinase-9 (MMP-9), and MMP-2 in PLAC8-silenced cells. Nowadays, data analysis is been the most important means to realize power scheduling in smart grids. However, the sharp increase in business data of grids has posed great challenges for this purpose. To deal with such issue, this paper utilizes deep learning to discover hidden rules from massive large-scale big data and particle swarm optimization (PSO) algorithm for generation of control decision. Therefore, an intelligent scheduling control method for smart grid based on deep learning is proposed in this paper. By modeling the historical data of the power company, the long short-term memory algorithm can effectively extract the effective features and realize the prediction of the coal consumption of the unit under certain conditions. At the same time, a kind of intelligent power scheduling algorithm is designed by using PSO, so as to save energy and reduce emissions as much as possible while fulfilling the real-time power generation task. Experiments on a real-world smart grid dataset show that the proposal can achieve a relatively good performance with respect to intelligent scheduling.

High-performance flexible conductive films are highly promising for the development of wearable devices, artificial intelligence, medical care, etc. Herein, a three-step procedure was developed to produce electromagnetic interference (EMI) shielding, Joule heating, and a hydrophobic nanofiber film based on hydrolysate of waste leather scraps (HWLS): (i) electrospinning preparation of the HWLS/polyacrylonitrile (PAN)/zeolitic imidazolate framework-67 (ZIF-67) nanofiber film, (ii) carbonization of the HWLS/PAN/ZIF-67 nanofiber film, and (iii) coating of the carbon nanofiber@cobalt (Co@CNF) nanofiber film with perfluorooctyltriethoxysilane (POTS). The X-ray diffraction results showed that metal nanoparticles and amorphous carbon had obvious peaks. The micromorphology results showed that metal nanoparticles were coated with carbon nanofibers. Aim: To explore the appropriate triage methods for women infected with high-risk human papillomavirus (hrHPV). Materials & methods: A total of 424 out of 872 hrHPV-infected women were divided into cervicitis (n = 123), cervical intraepithelial neoplasia grade 1 (CIN1; n = 89), CIN2 (n = 72), CIN3 (n = 87) and cervical cancer (n = 53) groups. Results: The sensitivity/specificity of ZNF582m, PAX1m and liquid-based cytology (LBC) for hrHPV-infected women with transformation zone 3 CIN3+ was 83.9/93.1, 77.4/90.6 and 80.6/58.5%, respectively. The ZNF582m/ PAX1m test had a higher specificity than LBC (p 0.05). ZNF582m/ PAX1m improved the positive predictive value of CIN3+ (64.7/60.0%) in low-grade LBC (negative predictive value: 91.7/88.7%). Conclusion: ZNF582m was superior to PAX1m and LBC tests in detecting CIN3+ in hrHPV-infected women.

RhRab5ip belongs to the Rab5-interacting protein (Rab5ip) family and may function in vesicle trafficking pathway. By using split ubiquitin yeast two-hybrid (SUY2H) system, bimolecular fluorescence complementation (BiFC) and subcellular colocalization we confirmed the existence of physical interaction between RhPIP1;1 and RhRab5ip in yeast and plant cell. The interaction of these two proteins happened at the small punctate structures in the cytoplasm. Implantable bioelectronics provide unprecedented opportunities for real-time and continuous monitoring of physiological signals of living bodies. Most bioelectronics adopt thin-film substrates such as polyimide and polydimethylsiloxane that exhibit high levels of flexibility and stretchability. However, the low permeability and relatively high modulus of these thin films hamper the long-term biocompatibility. In contrast, devices fabricated on porous substrates show the advantages of high permeability but suffer from low patterning density. Here, we report a wafer-scale patternable strategy for the high-resolution fabrication of supersoft, stretchable, and permeable liquid metal microelectrodes (μLMEs). We demonstrate 2-μm patterning capability, or an ultrahigh density of ~75,500 electrodes/cm 2 , of μLME arrays on a wafer-size (diameter, 100 mm) elastic fiber mat by photolithography. We implant the μLME array as a neural interface for high spatiotemporal mapping and intervention of electrocorticography signals of living rats. The implanted μLMEs have chronic biocompatibility over a period of eight months.

As an immune enhancer, Nocardia rubra cell-wall skeleton (Nr-CWS) has been used to treat persistent human papillomavirus infection and cervical precancerous lesions. However, it is still unclear whether it can be used to treat cervical carcinoma.In our study, the aim was to determine whether Nr-CWS affects the apoptosis of cervical carcinoma cells by enhancing the antitumor effect of dendritic cells and macrophages in vivo and in vitro.The experimental results showed that Nr-CWS can promote the activity of dendritic cells and macrophages and reduce their apoptosis. It also increased the cytokines IL-6, IL-12, TNF-ɑ, and IL-1β secreted by dendritic cells and macrophages and reduced their PD-L1 expression. In vitro, Nr-CWS inhibited the proliferation, colony forming ability of HeLa and SiHa cervical carcinoma cell lines cultured with macrophages, and more cells were blocked in G2/M phase. Nr-CWS promoted TNF-ɑ/TNFR1/caspase-8-mediated apoptosis by increasing macrophages secretion of TNF-ɑ and inhibited cell migration and invasion regulated by the WNT/β-catenin-EMT pathway. Nr-CWS also reduced the expression of the cervical carcinoma genes E6 and E7 thereby increasing expression of p53 gene and decreasing expression of PD-L1 gene. In vivo, Nr-CWS inhibited tumor growth and decreased the expression of E6, E7, PD-L1, P16, Ki67, and PCNA in tumors.Therefore, our results suggest that Nr-CWS can promote apoptosis of cervical carcinoma cells by enhancing the antitumor effect of dendritic cells and macrophages.

For enhanced mechanical and microwave absorption properties at the same time, the SiCf/hybrid matrix composites were fabricated by precursor infiltration and pyrolysis (PIP) method with polysiloxane (PSO) ethanol solution, alumina sols and silica sols. As the first layer of the hybrid matrix, the SiOC ceramic was pyrolyzed from PSO solution. The remained hybrid matrix was mullite, which sintered from alumina sols and silica sols. The effects of different content of PSO solution on the morphologies, flexure strength and reflection loss values of composites were studied. Additionally, the XRD patterns, Fourier Transform Infrared (FTIR) and Raman spectrum of hybrid matrix were also investigated. With the increasingly content of PSO solution from 0% to 10 % and 20%–60% in the first infiltration-pyrolysis process, the flexure strength of composites was increased from 175.18 MPa to 301.94 MPa and decreased from 263.33 MPa to 221.30 MPa, respectively. The complex permittivity was increased with the increasing content of PSO solution from 0%–40% due to the free carbon conductive network from excessive SiOC. Moreover, the complex permittivity of SiCf/hybrid matrix composites with 50 % and 60 % content of PSO solution was reduced due to more open porosity and broken free carbon conductive network. Additionally, the maximum reflection loss values of SiCf/hybrid matrix composite with 50 % PSO solution were over -60 dB and the effective absorption bandwidth (EAB) of this composite reaches 3.89 GHz in the X band.

The silver nanowire (AgNWs) / cotton fiber was used as a conductive substrate with high conductivity charge transfer. Then the polyaniline (PANI) molecular chain was fixed on the AgNWs / cotton fiber by in-situ polymerization to prevent its dedoping and improve its cyclic properties. Thus the flexible capacitor electrode material with conductive, specific capacity and cyclic properties was obtained. The results showed that when the mass ratio of KH-560 to AgNWs/cotton fiber was 3:1, the grafting rate of epoxide group on the modified AgNWs/cotton fiber was the highest. When the mass ratio of aniline to modified AgNWs cotton fabric was 3:1, the PANI content of in-situ polymerization on the fabric surface was the highest, 20.83%. The specific capacity of PANI/AgNWs/cotton fiber electrode material was the highest, 154 F/g, after 5000 cycles of charging and discharging, the specific capacity could keep 96%. It is worth mentioning, the introduction of PANI and AgNWs could significantly improve the wear resistance of cotton fiber, the friction resistance of the cotton fabric increased to 36,000 times. It is provided a new idea for design a flexible capacitor electrode material integrating electrical conductivity and cyclic stability.

Today, corresponding to the new wave of internationalisation and integration, education is also subject to rapid and innovative changes. Accordingly, detailed research is undertaken in the present study on innovative teaching strategies that can be developed, under the background of artificial intelligence, to familiarise college students with the ability to fluently express their ideas in English. The research shows that among the factors affecting the quality of English teaching, teachers’ teaching methods, English teaching environment and students’ independent learning ability account for high weightage, which are 54%, 31% and 11%, respectively. Through the model of college students’ English teaching innovation strategy, it can be seen that the teaching method, teaching environment and autonomous learning method are the most feasible methods that can be employed in crafting a students’ English teaching innovation strategy.

Background Competing endogenous RNAs (CeRNAs) play an important role in maintenance of ovarian cancer stem cells (OCSCs) characteristics. Objective To isolate a new OCSC line and construct a ceRNA network for maintenance of OCSCs characteristics by the cell line and GEO Database. Methods We isolated OCSCs from ovarian cancer cell line COC1 by chemotherapy drugs and growth factors. We identified the DEMs, DELs and DEGs between OCSCs and COC1 by microarray and combined them with representative microarray profiles in GEO Database. The ceRNA network was constructed by STRING and Cytoscape. QPCR and western blot were used to verify the expression of several genes that contained in the predicted network. Results According to the combination, 28 DEMs were identified at first, and 452 DEGs were obtained combining with the predicted targets of these miRNAs and our mRNA microarray results. Up-regulated DEGs of them were significantly enriched in ‘p53 signaling pathway’, ‘FoxO signaling pathway’, et al, whereas down-regulated DEGs were significantly enriched in ‘Adherens junction’ and ‘Hepatitis C’ pathway. Finally, we obtained OCSCs characteristics related ceRNA network with 10 DEMs, 21 DEGs, and 25 transcripts of 13 DELs. We verified that LINC00665-miR-146a-5p-NRP2 should be one of the pathways of the constructed ceRNA network. Conclusion The ceRNA network we constructed may be involved in the stem cell characteristics maintenance of OCSCs and provide directions for further OCSCs research in the future.

Administration of cisplatin, a common chemotherapeutic drug, has an inevitable side effect of sensorineural hearing loss. The main etiologies are stria vascularis injury, spiral ganglion degeneration, and hair cell death. Over several decades, the research scope of cisplatin-induced ototoxicity has expanded with the discovery of the molecular mechanism mediating inner ear cell death, highlighting the roles of reactive oxygen species and transport channels for cisplatin uptake into inner ear cells. Upon entering hair cells, cisplatin disrupts organelle metabolism, induces oxidative stress, and targets DNA to cause intracellular damage. Recent studies have also reported the role of inflammation in cisplatin-induced ototoxicity. In this article, we preform a narrative review of the latest reported molecular mechanisms of cisplatin-induced ototoxicity, from extracellular to intracellular. We build up a signaling network starting with cisplatin entering into the inner ear through the blood labyrinth barrier, disrupting cochlear endolymph homeostasis, and activating inflammatory responses of the outer hair cells. After entering the hair cells, cisplatin causes hair cell death via DNA damage, redox system imbalance, and mitochondrial and endoplasmic reticulum dysfunction, culminating in programmed cell death including apoptosis, necroptosis, autophagic death, pyroptosis, and ferroptosis. Based on the mentioned mechanisms, prominent therapeutic targets, such as channel-blocking drugs of cisplatin transporter, construction of cisplatin structural analogues, anti-inflammatory drugs, antioxidants, cell death inhibitors, and others, were collated. Considering the recent research efforts, we have analyzed the feasibility of the aforementioned therapeutic strategies and proposed our otoprotective approaches to overcome cisplatin-induced ototoxicity.

Developing high-temperature microwave absorption material with light weight is crucial. In this work, Ti3SiC2/Na3Zr2Si2PO12 composites were exploited as light microwave absorption material at high temperature by plasma spraying and vacuum sintering for microstructure, phase, dielectric property, and microwave absorption property study. Results showed that both the preparation method and Ti3SiC2 concentration had vital influences on the dielectric properties and microwave absorption properties of Ti3SiC2/Na3Zr2Si2PO12 composites. The denser Ti3SiC2/Na3Zr2Si2PO12 ceramics had higher complex permittivity than that of Ti3SiC2/Na3Zr2Si2PO12 coatings with lower density, which could obtain better microwave absorption property in thinner thickness with less Ti3SiC2 content. As the content of Ti3SiC2 increased, the complex permittivity of both Ti3SiC2/Na3Zr2Si2PO12 coatings and ceramics improved due to the increased interfacial polarization and conduction loss. The optimum microwave absorption property was obtained in Ti3SiC2/Na3Zr2Si2PO12 ceramic with 10% Ti3SiC2 content, which had a bandwidth of 4.2 GHz and minimum reflection loss (RLm) of − 14.4 dB in 1.7 mm.

Cerebral ischemia/reperfusion (I/R) is a complicated brain disease involving severe inflammation, autophagy, apoptosis and necroptosis. Hippocampal CA1 neuron death in cerebral I/R injury has been proven to be a form of necroptosis that is regulated by the connection of receptor interacting protein kinase (RIP) 1 and RIP3. The RIP3 substrate molecule is mixed lineage kinase domain-like protein (MLKL). The downstream of this signal pathway triggers mitochondrial permeability transition pore (mPTP) channel opening. The occurrence of mitochondrial membrane potential disappearance and the reduction of ATP facilitate reactive oxygen species (ROS) generation. The high level of ROS is essential for engulfing damaged organelles and proteins. In many conditions, autophagy has a decisive function in determining cell outcome through intricate signaling pathway interactions with other cell death signaling mechanisms. Necroptosis also been demonstrated to be responsible for the induction of autophagy. Studies have confirmed that cyclophilin D (CypD), as a regulator of mPTP, has effects on the course of necroptosis, autophagy and apoptosis. Preliminary treatment with cyclosporine-A (CsA), an effective inhibiting agent of CypD, has partial but valid negatively regulated function in necroptosis-associated molecule markers. Administration of CsA is also attributed to inhibiting autophagy-related molecular marker expression. Cerebral I/R injury widely increased the expression levels of autophagy-associated proteins, such as light chain 3-II (LC3-II) and extracellular signal-regulated kinase (ERK), while a small molecule inhibitor of RIP1, necrostatin-1 (Nec-1) functions as a negative regulator to them. The molecular mechanisms and the interaction between necroptosis and autophagy might represent a promising therapeutic target for cerebral I/R injury.

SLF and KD-S SiC fibers were used to fabricate SiCf/SiC composites by precursor impregnation and pyrolysis (PIP) method. To simultaneously improve both the mechanical and electromagnetic wave absorption properties of SiCf/SiC composites, the SiO2 fillers were mixed into the PSC/xylene precursor solution as inert filler. In this research, the effects of SiO2 filler on the micro morphologies, XRD patterns, mechanical and dielectric property were investigated. The results exhibit that the flexural strength of the SLF and KD-S SiCf/SiC composites was improved from 167.69 MPa to 241.95 MPa and 248.16 MPa–324.87 MPa with SiO2 filler, respectively. The lower complex permittivity and better impedance matching are attributed to a broken conductive network in the SiC matrix, which is caused by SiO2 filler. Therefore, the reflection loss values of SLF SiCf/SiC composites with SiO2 filler were approach at −40 dB, and the KD-S SiCf/SiC composites with SiO2 filler of that over −12 dB. The effective absorption bandwidths of SLF and KD-S SiCf/SiC composites with SiO2 filler are 3.44 GHz and 2.75 GHz, respectively.

Objectives: To investigate the performance of Pulsatilla saponin A (PsA) in Multiple Myeloma (MM) cells. Methods: Proliferation, cell cycle analysis, apoptosis and TUNEL assays were conducted to detect the growth and apoptosis in MM cells. Western blotting was used to identify the change in the protein. Results: In cell assays, PsA significantly inhibited the growth and apoptosis in MM cells. Cyclin B1, caspase-3, cleaved-caspase-3, PARP, cleaved-PARP, p-ERK increased, while Bcl-2 decreased after PSA treatment. The CD49e positive rate of U266 cells was increased after 96h PsA treatment. At the same time, immunoglobulin and the Free Light Chain (FLC) ratio in the culture supernatant obviously increased. Also, the differentiation induced by PsA was confirmed in the primary myeloma cells. Conclusions: Our findings reveal that PsA may exert its antitumor effect by causing G2 arrest and apoptosis in myeloma cells. And low-dose PsA can induce the differentiation of myeloma cell lines and primary myeloma cells, probably through the MEK/ERK signaling pathway in vitro.

Recently, high-temperature stability is a challenge in a number of microwave absorption materials. Hence, researchers are still searching for a novel material system preferably with a high-temperature resistance to be applied in the field of microwave absorption. Here, in the current study, toward this aim, lanthanum (La)-doped strontium titanate (SrTiO3) blended with TiO2 were fabricated by hot-press sintering in a vacuum. The as-prepared samples are denoted as TiO2–Sr1−xLaxTiO3 with x varying from 0.1 to 0.3 in steps of 0.1. Scanning electron microscope (SEM), X-ray diffraction (XRD), X-ray photoelectron spectroscope (XPS), and microwave vector network analyzer were carried out to study their morphology, phase composition, structure, and electromagnetic and microwave absorption properties, respectively. It is revealed that the La atom was efficiently doped at the Sr-site in SrTiO3. Benefiting from the tunability of its dielectric and impedance properties, TiO2–Sr1−xLaxTiO3 can be utilized in a highly efficient way to absorb microwave radiations with a decent design. Results illustrated that TiO2–Sr1−xLaxTiO3 (x = 0.2) with a thickness of only 0.42 mm exhibits a high microwave absorption efficiency of −40.89 dB and can achieve a 2.82 GHz bandwidth of reflection loss value below −5 dB. Thus, TiO2–Sr1−xLaxTiO3 composites ceramics can be served as an opening opportunity for the application of high-temperature stability and tunable high-performance effectiveness microwave absorption materials in stealth technology and information security.

Purple-stem Brassica napus (B. napus) is a phenotype with unique color because of its high anthocyanins content. Anthocyanins are naturally occurring plant pigments that have antioxidants activity and play important role in plant defense against abiotic and biotic stresses. In the present study, drought induced effects on plants were investigated in hydroponically grown seedlings of green stem (GS) and purple stem (PS) genotypes of B. napus. The results of this study showed that the major function of anthocyanins accumulation during drought was to enhance the antioxidant capability and stress tolerance in B. napus plants. Our results showed that drought significantly inhibited the plant growth in terms of decreased biomass accumulation in both genotypes, although marked decline was observed in GS genotype. The reduction in photosynthetic attributes was more noticeable in the GS genotype, whereas the PS genotype showed better performance under drought stress. Under stressful conditions, both the genotype showed excessive accumulation of reactive oxygen species (ROS) and malondialdehyde (MDA), as well as higher levels of antioxidant enzymes activities. Under drought conditions, the GS genotype showed apparent damages on chloroplast deformation like in thylakoid membrane and grana structural distortion and fewer starch grains and bigger plastoglobuli. Moreover, during drought stress, the PS genotype exhibited maximum expression levels of anthocyanins biosynthesis genes and antioxidant enzymes accompanied by higher stress tolerance relative to GS genotype. Based on these findings, it can be concluded that GS genotype found more sensitive to drought stress than the PS genotype. Furthermore this research paper also provides practical guidance for plant biologists who are developing stress-tolerant crops by using anthocyanin biosynthesis or regulatory genes.

Brachial plexus root avulsion (BPRA) is frequently caused by high-energy trauma including traffic accident and birth trauma, which will induces massive motoneurons (MNs) death as well as loss of motor and sensory function in the upper limb. The death of MNs is attributed to energy deficiency, neuroinflammation and oxidative stress at the injured ventral horn of spinal cord triggered by BPRA injury. It has been reported which aldose reductase (AR), an endogenous enzyme that catalyzes fructose synthesis, positively correlates with the poor prognosis following cerebral ischemic injury, diabetic retinopathy and diabetic peripheral neuropathy. However, the role of AR in BPRA remains unknown. Herein, we used a mouse model and found that in the spinal cord of BPRA mice, the upregulation of AR correlated significantly with (1) an inactivated SIRT1–AMPK–mTOR pathway and disrupted autophagy; (2) increased byproducts accumulation of lipid peroxidation metabolism and neuroinflammation; and (3) increased MNs death. Furthermore, our results demonstrated the role of AR in BPRA injury whereby the absence of AR (AR knockout mice, AR−/−) prevented the hyper-neuroinflammation and disrupted autophagy as well as motor neuron death caused by BPRA injury. Finally, we further demonstrate that AR inhibitor epalrestat is neuroprotective against BPRA injury by increasing autophagy level, alleviating neuroinflammation and rescuing MNs death in mice. Collectively, our data demonstrate that the AR upregulation in the spinal cord is an important factor contributing to autophagy disruption, neuroinflammation and MNs death following brachial plexus roots avulsion in mice. Our study also provides a promising therapy drug to assist re-implantation surgery for the treatment of BPRA.

Gadolinium (Gd)-doped perovskite SrTiO3 combining with Al2O3, Al2O3−Sr(1−x)GdxTiO3 (x varying from 0 to 0.3 in steps of 0.1) composite ceramics was synthesized by hot-press sintering in a vacuum. X-ray diffraction (XRD), scanning electron microscopy (SEM), energy-dispersive spectrometer (EDS), four-probe testing instrument, and vector network analyzer were utilized to study their phase and composition characteristics, micromorphology, electrical conductivity, electromagnetic and microwave-absorbing properties, respectively. The XRD, SEM, and EDS results demonstrated that Gd atoms were successfully doped into SrTiO3 crystal and substituted the Sr sites. As a result, Al2O3−Sr(1−x)GdxTiO3 with x = 0.2, which has the advantage of an ultra-thin thickness (0.341 mm), exhibits excellent absorbing properties with a broad bandwidth (90% microwave absorption) of 4.07 GHz in the X band. Furthermore, the present investigation illustrated that Al2O3−Sr(1−x)GdxTiO3 (x = 0.1, 0.2, and 0.3) could be applied in the X band for microwave absorption, with broadband width and ultra-thin thickness (≤ 0.35 mm), by controlling the molar ratios of Gd and the thickness.

Background: Metastasis and paclitaxel (PTX) resistance are the main reason for the poor prognosis of ovarian cancer (OC). Evidence has shown that RNA-binding proteins (RBPs) and long noncoding RNAs (lncRNAs) can modulate post-transcriptional regulation. The aim of this study was to determine the relationship among RBP, lncRNA and OC and to further guide clinical therapy. Methods: The expression of pre‐mRNA processing factor 6 (PRPF6) in OC tissues form 68 patients were detected by Immunohistochemistry. The expression of small nucleolar RNA host gene 16(SNHG16) transcripts-SNHG16-L/S in OC cell lines were analyzed by real-time PCR (RT-PCR). Transwell, CCK8 assays and flow cytometry analyses were used to detected effects of PRPF6 and SNHG16 on tumorigenesis and PTX-resistance. Molecular interactions were examined by dual‐luciferase reporter gene assay, RNA immunoprecipitation and chromatin immunoprecipitation. OC cells that knockdown PRPF6 were injected subcutaneously in nude mice.Results: The results showed that PRPF6 was upregulated in OC chemoresistant tissues and was closely related to advanced FIGO stages. PRPF6 promoted progression, and PTX resistance in vitro and in vivo. SNHG16-L/S were differentially expressed in OC cells and tissues as detected through real-time PCR (RT-PCR). SNHG16-L/S had opposite effects on progression and PTX resistance in OC. Mechanistically, SNHG16-L inhibited GATA-binding protein 3 (GATA3) transcription by binding to CCAAT/enhancer-binding protein B (CEBPB). Moreover, PRPF6 induced the alternative splicing of SNHG16, causing downregulation of SNHG16-L and, leading to the upregulation of GATA3 expression to further promote metastasis and PTX-resistance in OC.Conclusions: Totally, these data unveiled that PRPF6 promotes metastasis and PTX resistance of OC through SNHG16-L/CEBPB/GATA3 axis, which provides a new direction for OC treatment.

Nucleobase mismatches can jeopardize DNA polymerization specificity, causing mutations and errors in DNA replication and detection. Herein we report the first synthesis of novel 2-Se-thymidine triphosphate (SeTTP), describe the single-selenium atom-specific modification strategy (SAM) against T/G mismatches, and demonstrate SAM-assisted polymerization and detection with much higher specificity and sensitivity. SAM can effectively suppress the formation of non-specific products in DNA polymerization and detection. Thus, SAM enhances the specificity of DNA synthesis by approximately 10 000 fold, and in turn, it allows the detection of clinical COVID-19 viral RNA in low copy numbers (single-digit copies), while the conventional RT-qPCR does not.

Objective: This retrospective study analyzed the factors affecting recurrence in patients after surgery with borderline ovarian tumors and postoperative recurrence and pregnancy after fertility-sparing surgery (FSS), to provide guidance for clinical treatment of borderline ovarian tumors and propose a therapeutic strategy for fertility protection. Methods: A total of 415 patients with borderline ovarian tumors were initially operated on in the gynecology ward of Shengjing Hospital Affiliated with China Medical University from September 1, 2013, to September 1, 2019. Central pathology review and prospective follow-up were carried out. The clinical and pathological data were consulted through the medical record query system of our hospital. The recurrence and pregnancy of the patients were investigated through telephone follow-up and outpatient and inpatient medical records. The influence of clinical and pathological variables on recurrence and pregnancy were evaluated using univariate/multivariate analyses. Results: In this study, 415 patients were collected, of which 21 lost follow-up, and a total of 394 eligible patients were included in the analysis. Among these patients, 25 patients relapsed with a recurrence rate of 6.3% and there were 196 patients with fertility-sparing surgery, of the 63 patients attempting to conceive, 35 were able to attain pregnancy with a pregnancy rate of 55.6%. All patients survived until the follow-up deadline. In univariate and multivariate analyses, FSS, FIGO stage, and micropapillary pattern were independent risk factors for recurrence of BOTs. Objective: Calcium channel blockers, such as dihydropyridines, are commonly used to inhibit enhanced activity of vascular Ca V 1.2 channels in hypertension. However, patients who are insensitive to such treatments develop calcium channel blocker-resistant hypertension. The function of Ca V 1.2 channel is diversified by alternative splicing, and the splicing factor PTBP (polypyrimidine tract-binding protein) 1 influences the utilization of mutually exclusive exon 8/8a of the Ca V 1.2 channel during neuronal development. Nevertheless, whether and how PTBP1 makes a role in the calcium channel blocker sensitivity of vascular Ca V 1.2 channels, and calcium channel blocker-induced vasodilation remains unknown. Approach and Results: We detected high expression of PTBP1 and, inversely, low expression of exon 8a in Ca V 1.2 channels (Ca V 1.2 E8a ) in rat arteries. In contrast, the opposite expression patterns were observed in brain and heart tissues. In comparison to normotensive rats, the expressions of PTBP1 and Ca V 1.2 E8a channels were dysregulated in mesenteric arteries of hypertensive rats. Notably, PTBP1 expression was significantly downregulated, and Ca V 1.2 E8a channels were aberrantly increased in dihydropyridine-resistant arteries compared with dihydropyridine-sensitive arteries of rats and human. In rat vascular smooth muscle cells, PTBP1 knockdown resulted in shifting of Ca V 1.2 exon 8 to 8a. Using patch-clamp recordings, we demonstrated a concomitant reduction of sensitivity of Ca V 1.2 channels to nifedipine, due to the higher expression of Ca V 1.2 E8a isoform. In vascular myography experiments, small interfering RNA-mediated knockdown of PTBP1 attenuated nifedipine-induced vasodilation of rat mesenteric arteries. Conclusions: PTBP1 finely modulates the sensitivities of Ca V 1.2 channels to dihydropyridine by shifting the utilization of exon 8/8a and resulting in changes of responses in dihydropyridine-induced vasodilation.
