Small interfering RNA (siRNA) was utilized to silence FN1 in HNSCC cells. Cell counting kit-8 (CCK-8) assay, colony formation assay, Transwell assay and wound healing assay were performed to reveal the effect of FN1 on malignant behaviors of HNSCC cells. Moreover, a co-culture model of macrophages and HNSCC cells was established to investigate whether FN1 induce macrophage M2 polarization. Finally, we used bioinformatic methods to explore the possible FN1-related pathways in HNSCC.Results: FN1 is significantly overexpressed in HNSCC patients and has been obviously correlated with higher pathological stage and poor prognosis. Downregulation of FN1 suppressed the proliferation, migration and invasion of HNSCC cells, and inhibited macrophage M2 polarization in vitro. In addition, “PI3K-Akt” and “MAPK” signaling pathways may be involved in the malignant process of FN1 in HNSCC.Conclusion: The overexpression of FN1 promotes HNSCC progression and induces macrophages M2 polarization. The title compounds exist as equilibrium mixtures of the N-1 and N-2 isomers in solution. However, the crystal structure of 3-(1-hydroxycyclohexyl)-1-(α-pyrrolidinomethyl)-1,2,4-triazole discloses only this isomer in the solid state. Evidence for the nature of the isomerization processes in solution is provided. The kinetic and thermodynamic parameters are measured and qualitatively assessed.

The crystal structure of Pt(en) (mal) (OH)2H2O has been determined from three-dimensional X-ray data collected by Syntex P3/R3 four circle diffractometer. The crystal is triclinic with space group P1, α =8.4526(15)?, b=6.0947 (8)?, c=10.1854 (15)?, α = 106.48°(I), β= 103.65°(I), γ= 79.12° (I), and Z = 2. The crystal structure has been solved by Patterson method and refined by block least square refinement for positional parameters and anisotropic tempreture factors of nonhydrogen atoms. The final R factor is 0.055.The result of crystal structure analysis shows that the platinum atom is inovolved in a d2sp3 hybridization and an octahedral configuration.
