Honey bees are facing many serious threats, and Varroa destructor is of major worldwide concern. V. destructor was first observed in 2010 in the south–west Inidan Ocean island of Madagascar. Varroa spp. was first suspected in Mauritius in late July 2014. Two surveys were conducted in September and November 2014 in Mauritius and Rodrigues. The aims of these surveys were to confirm the presence of Varroa on Mauritius, determine the species and strain and to study its spread and interactions with the bee louse Braula pretoriensis. Honey bees were sampled from different sites. V. destructor and the bee louse Braula pretoriensis infestation rates were measured for each colony and apiary. V. destructor presence was confirmed and observed in the Central and Northern part of Mauritius, but was not detected in Rodrigues. B. pretoriensis was observed on both islands. V. destructor partial mitochondrial region of the CO1 showed that the Mauritius K strain found was different from the one from Madagascar. In Mauritius, B. pretoriensis numbers were significantly higher in colonies infested by V. destructor; however, bee louse infestation rates were similar in both islands in sites without the varroa mite. Although bee louse population regulation by honey bees is unknown, individual and social behaviors regulating V. destructor populations within colonies are not sufficient to maintain low levels of both B. pretoriensis and V. destructor. Sanitary measures were proposed, some of which are currently being applied to limit the spread of V. destructor across the island.

International audience; A substantial honeybee decline is being observed worldwide. Agricultural intensification and loss of wild floral resources rank among the main factors contributing to this decline. Landscape enhancement of floral resources has been proposed as an agro-environmental scheme intended to provide honeybees with compensatory food sources in intensive agrosystems. Floral scheme efficiency has rarely been evaluated with respect to landscape context. In this study, we developed and validated a modeling tool to delineate the landscape areas likely to be associated with higher efficiency of floral enhancement schemes. In particular, the proximity of some landscape elements used by honeybees, either as foraging habitat or as visual landmark for orientation, may partly determine floral scheme efficiency. We investigated this issue using resource selection functions (RSFs), i.e. models that aim to predict the occurrence of foraging honeybees at floral patches as a function of the presence of keystone landscape elements in their proximity. However, deciding which landscape elements are effectively in the proximity or not is mostly a matter of subjectivity. The novelty of our approach resides in its use of a distance-weighting function to explicitly account for the spatial location of surrounding landscape elements. In that respect, a distance function should be scaled on movement patterns of foraging organisms. Herein, we inferred movement patterns from the autocorrelative properties of honeybee foraging activity. This modeling approach was developed on Phacelia (Phacelia tanacetifolia) field margin strips, a typical “honeybeefriendly” floral scheme. A foraging survey conducted on 170 Phacelia plots (2 m × 2 m) from 17 Phacelia strips, all positioned within the foraging range of an experimental apiary, revealed that (i) the floral scheme efficiency is positively influenced by the presence of linear landscape elements such as hedgerows and forest edges, but negatively affected by the presence of alternative floral resources, and that (ii) weighting the relative importance of those landscape elements by incorporating a distance function into models considerably improved their predictive power. This modeling tool has the potential to help land managers optimizing their financial investment by avoiding low-efficiency landscape areas, or favoring high-efficiency ones, at the time of planning floral enhancement schemes

Summary Avoiding toxins in food is as important as obtaining nutrition. Conditioned food aversions have been studied in animals as diverse as nematodes and humans [1, 2], but the neural signaling mechanisms underlying this form of learning have been difficult to pinpoint. Honeybees quickly learn to associate floral cues with food [3], a trait that makes them an excellent model organism for studying the neural mechanisms of learning and memory. Here we show that honeybees not only detect toxins but can also learn to associate odors with both the taste of toxins and the postingestive consequences of consuming them. We found that two distinct monoaminergic pathways mediate learned food aversions in the honeybee. As for other insect species conditioned with salt or electric shock reinforcers [4–7], learned avoidances of odors paired with bad-tasting toxins are mediated by dopamine. Our experiments are the first to identify a second, postingestive pathway for learned olfactory aversions that involves serotonin. Since 2006, disastrous colony losses have been reported in Europe and North America. The causes of the losses were not readily apparent and have been attributed to overwintering mortalities and to a new phenomenon called Colony Collapse Disorder. Most scientists agree that there is no single explanation for the extensive colony losses but that interactions between different stresses are involved. As the presence of Varroa in each colony places an important pressure on bee health, we here address the question of how Varroa contributes to the recent surge in honey bee colony losses, Seit 2006 werden in Europa und Nordamerika katastrophale Völkerverluste gemeldet. Die Ursachen dieser Verluste waren nicht leicht zu erklären, sie wurden als überwinterungsbedingte Mortalitäten bewertet und unter dem Begriff Colony Collapse Disorder (CCD) zusammengefasst. Die meisten Wisenschaftler stimmen dahingehend überein, dass diesen Völkerverlusten keine Einzelursache zugrunde liegt, sondern, dass vermutlich verschiedene Stressfaktoren zusammenwirken. Da der Varroa-Befall einen wichtigen Druck auf die Gesundheit der Bienen in den Völkern darstellt, gingen wir der Frage nach, inwiefern Varroa an den seit kurzem beobachteten Völkerverlusten beteiligt sein kann. Verschiedene physikalische und physiologische Faktoren mit negativer Wirkung der Varroa-Milbe auf die Gesundheit der einzelnen Biene und den Volkszusammenhang sind bereits bekannt. So führt das wiederholte Saugen von Hämolymphe zu Verletzungen der Bienen, zu erniedrigten Werten im Proteingehalt, sowie dem Lebend- und Trockengewicht und zur Behinderungen in der Organentwicklung. Die parasitische Milbe und die von ihr übertragenen Viren führen zu morphologischen Fehlentwicklungen, reduzierter Widerstandskraft und Lebenserwartung und zu negativen Effekten auf die Flugdauer und Heimfindungsfähigkeit der Sammlerinnen. Die Milbe schwächt das Immunsystem der Bienen, indem die Expression von Genen der Immunantwort reduziert wird. Zusammen mit erhöhten DWV-Viren-Titern reduziert sie damit die Lebensfähigkeit der Arbeiterinnen und die Koloniefitness. Die Rolle der Varroa-Milbe als Vektor in der horizontalen und vertikalen Übertragung von Viren ist hingehend bekannt, und Ko-Infektionen von V. destructor mit verschiedenen Viren wurden bereits als wichtige Faktoren im Varroa-bedingten Zusammenbruch von Völkern beschrieben. V. destructor muss deshalb weiterhin als eine ernsthafte Bedrohung der Honibiene gelten, und weiterhin werden Honigbienenpopulationen weltweit durch diese parasitische Milbe dezimiert. Die Hypothese, dass CCD durch eindringende Varroa-Milben und ihre immunsuppressiven Fähigkeiten hervorgerufen wird, ist damit nicht auszuschliessen und wird durch die Befunde von vanEngelsdorp et al. (2009) gestärkt. Die Varroa-Milbe ist seit Jahren in den meisten Ländern vertreten, aus denen auch Völkerverluste gemeldet werden. Obwohl die Milbenpopulationen kontrolliert werden können, kann trotzdem spekulativ postuliert werden, dass die Verluste zumindest teilweise durch einen Varroa-Befall der Völker bedingt sein können. Welche Veränderungen in der Varroa-Biologie können also die jetzigen Verluste im Vergleich zu denen vor 20 Jahren erklären? Wir diskutieren potentielle Ursachen, einschliesslich Veränderungen in der Biologie und Populationsdynamik von Varroa und ihrem Wirt, der Kontrolle von Varroa durch Akarizide, indirekte Effekten der Varroa-Behandlungen, sowie synergistische Effekte multipler Faktoren

The rich variety of native honeybee subspecies and ecotypes in Europe offers a good genetic resource for selection towards Varroa resistance. There are some examples of mite resistance that have developed as a consequence of natural selection in wild and managed European populations. However, most colonies are influenced by selective breeding and are intensively managed, including the regular use of miticides. We describe all characters used in European breeding programs to test for Varroa resistance. Some of them (e.g., mite population growth, hygienic behavior) have been implemented in large-scale selection programs and significant selection effects have been achieved. Survival tests of pre-selected breeder colonies and drone selection under infestation pressure are new attempts to strengthen effects of natural selection within selective breeding programs. Some perspectives for future breeding activities are discussed, Seit Einzug der Varroa Milbe in Europa sind in einigen Regionen Völker aufgetreten, die langfristig ohne Behandlungsmaßnahmen überleben. Eine weitergehende Untersuchung von 82 derartiger Völker (VSB) in Frankreich zeigte eine mittlere Überlebensdauer von 7,8 Jahren. Der Varroa-Befallsanstieg während der Brutsaison verlief signifikant schwächer als bei normal anfälligen Kontrollvölkern (Abb. 1). Dabei spielen offenbar ein intensives Putzabwehr- und Bruthygieneverhalten eine Rolle, möglicherweise aber auch häufige Schwarmereignisse, spezifische Umweltbedingungen, Veränderungen der Milbenpopulation und die Virusprävalenz. Durch einen völligen Verzicht auf Bekämpfungsmaßnahmen ist es innerhalb weniger Generationen sowohl in einem kommerziellen Zuchtbetrieb in Frankreich als auch in einer isolierten Versuchs-population in Schweden zur Auslese resistenter Bienen gekommen. Als mögliche Ursachen der abgeschwächten Befallsentwicklung wurden u.a. eine geringere Brutdynamik und ein geringer Brutbefall bezogen auf die Gesamtmilbenzahl beobachtet. Aufgrund umfangreicher Untersuchungen liegen heute standardisierte Methoden zur Beurteilung der Befallsentwicklung und der Bruthygiene vor, die dank guter Heritabilitäten und effizienter Zuchtwertschätzverfahren in vielen europäischen Ländern Eingang in die Zuchtpraxis gefunden haben. Zuchtprogramme zur Selektion auf Putzabwehrverhalten, kürzere Verdecklungsdauer oder beeinträchtigte Varroa-Reproduktion scheiterten hingegen an ineffizienten Messverfahren, geringer genetischer Variabilität oder unzureichend geklärter biologischer Zusammenhänge. Beispielhaft wird die Organisation eines umfangreichen Zuchtprogramms zur Steigerung der Resistenz leistungsfähiger Zuchtlinien vorgestellt. Die Kooperation zahlreicher Zuchtbetriebe, einheitliche Prüfstandards und eine gemeinsame Zuchtwertschätzung ermöglichen eine Auslese in großer Population. Potenzielle Zuchtvölker werden auf ihre Überwinterungsfähigkeit ohne vorangehende Behandlung gegen Varroa geprüft und durch den Betrieb von Belegstellen mit unbehandelten Drohnenvölkern werden Effekte unterschiedlicher Varroa-Anfälligkeit auf den Paarungserfolg von Drohnen (Abb. 2) in die Selektion einbezogen. Aufgrund schlechterer Honigleistung und Sanftmut konnten US-Linien Russischer Bienen (RHB) trotz relativ hoher Widerstandsfähigkeit in Europa keine Bedeutung erlangen. Die meisten europäischen Zuchtprogramme zielen auf einer stetige Verbesserung einheimischer Populationen. Inwieweit es auf diesem Weg möglich ist, hochgradig resistente und zugleich leistungsfähige Zuchtlinien zu entwickeln, bleibt offen. Immerhin zeigte sich in einem mehrjährigen Vergleichsversuch eine höhere Überlebensfähigkeit der bereits auf Resistenz gegen Varroa ausgelesenen Linien. Wichtige Zukunftsperspektiven ergeben sich aus der Weiterentwicklung der Selektionsmethoden, der Berücksichtigung spezifischer Umweltanpassungen und verbesserter Haltungstechniken

The invasion of the Western honey bee, Apis mellifera, by Varroa destructor is attributed to two mitochondrial haplotypes (K and J) that shifted last century from their primary host the Eastern honey bee, A. cerana, in north-east Asia. Here, mitochondrial DNA sequences (cox1, cox3, atp6 and cytb: 2700 base pairs) were obtained from mites infesting both Eastern and Western honeybees (respectively 21 and 11 colonies) from Asia including regions where the shifts first occurred. A total of eighteen haplotypes were uncovered in Asia (11 on A. cerana and 7 on A. mellifera). Two new variants of the K haplotype and two of the J haplotype were found on Western honeybees in what appeared to be well-established infestations. New haplotypes may represent a potential threat to A. mellifera worldwide. The extreme lack of polymorphism in the K and J haplotypes outside of Asia, can now be plausibly explained as being due to genetic 'bottlenecks' that occurred in Asia before and after mites shifted from their original Eastern honeybee host. International audience; Pheromones are very important in animal communication. To learn more about the molecular basis of pheromone action, we studied the effects of a potent honey bee pheromone on brain gene expression. Brood pheromone (BP) caused changes in the expression of hundreds of genes in the bee brain in a manner consistent with its known effects on behavioral maturation. Brood pheromone exposure in young bees causes a delay in the transition from working in the hive to foraging, and we found that BP treatment tended to upregulate genes in the brain that are upregulated in bees specialized on brood care but downregulate genes that are upregulated in foragers. However, the effects of BP were age dependent; this pattern was reversed when older bees were tested, consistent with the stimulation of foraging by BP in older bees already competent to forage. These results support the idea that one way that pheromones influence behavior is by orchestrating large-scale changes in brain gene expression. We also found evidence for a relationship between cis and BP regulation of brain gene expression, with several cis-regulatory motifs statistically overrepresented in the promoter regions of genes regulated by BP. Transcription factors that target a few of these motifs have already been implicated in the regulation of bee behavior. Together these results demonstrate strong connections between pheromone effects, behavior, and regulation of brain gene expression.

The purpose of this study is to investigate the possibility to use acetylcholinesterase (AChE) as a biomarker of exposure to deltamethrin insecticide in the honeybee, Apis mellifera and to test its reliability in the presence of other contaminants, as carbamate insecticide. Joined actions of deltamethrin (pyrethroid) and pirimicarb (carbamate), alone or in association, are investigated on AChE activity in surviving and dead honeybees, with a special focus on the relative proportions of its membrane and soluble forms. At the 0.5X dose (12.5 ng of deltamethrin and/or 2.5 microg of pirimicarb per bee), the residual tissue AChE activity in dead bees was 78% with deltamethrin, 43% with pirimicarb and 33% with dual treatment. In surviving bees, tissue AChE activity represented 250%, and 270% of control AChE activity with deltamethrin and dual treatment, respectively. The analysis of membrane and soluble AChE forms revealed an increase in the soluble form in dead bees after deltamethrin and dual treatment. However, in vitro investigations showed no direct interaction of deltamethrin on soluble and membrane AChE activity. The results suggest that the action of deltamethrin on AChE activity, in honeybee intact organisms, could be due to indirect mechanisms. The duality of AChE response to deltamethrin exposure, exhibited by the possibility of increase (surviving bees) or decrease (dead bees) of its activity has been pointed out for the first time. The important increase in AChE activity in response to deltamethrin, not altered by pirimicarb treatment, suggests that AChE activity could represent a robust biomarker specific to deltamethrin exposure in living bees.

International audience; Two acetylcholinesterase (EC 3.1.1.7) membrane forms AChEm1 and AChEm2, have been characterised in the honey bee head. They can be differentiated by their ionic properties: AChEm1 is eluted at 220 mM NaCl whereas AChEm2 is eluted at 350 mM NaCl in anion exchange chromatography. They also present different thermal stabilities. Previous processing such as sedimentation, phase separation, and extraction procedures do not affect the presence of the two forms. Unlike AChEm1, AChEm2 presents reversible chromatographic elution properties, with a shift between 350 to 220 mM NaCl, depending on detergent conditions. Purification by affinity chromatography does not abolish the shift of the AChEm2 elution. The similar chromatographic behaviour of soluble AChE strongly suggests that the occurrence of the two membrane forms is not due to the membrane anchor. The two forms have similar sensitivities to eserine and BW284C51. They exhibit similar electrophoretic mobilities and present molecular masses of 66 kDa in SDS-PAGE and a sensitivity to phosphatidylinositol-specific phospholipase C in non-denaturing conditions, thus revealing the presence of a glycosyl-phosphatidylinositol anchor. We assume that bee AChE occurs in two distinct conformational states whose AChEm2 apparent state is reversibly modulated by the Triton X-100 detergent into AChEm1

Fifty years ago, two distinct evolutionary lineages (Korean and Japanese) invaded the Western honeybee Apis mellifera. This haplo–diploid parasite species reproduces mainly through brother–sister matings, a system which largely favors the .xation of new mutations. In a worldwide sample of 225 individuals from 21 locations collected on Western honeybees and analyzed at 19 microsatellite loci, a series of de novo mutations was observed. Using historical data concerning the invasion, this original biological system has been exploited to compare three mutation models with allele size constraints for microsatellite markers: stepwise (SMM) and generalized (GSM) mutation models, and a model with mutation rate increasing exponentially with microsatellite length (ESM). Posterior probabilities of the three models have been estimated for each locus individually using reversible jump Markov Chain Monte Carlo. The relative support of each model varies widely among loci, but the GSM is the only model that always receives at least 9% support, whatever the locus. The analysis also provides robust estimates of mutation parameters for each locus and of the divergence time of the two invasive lineages (67,000 generations with a 90% credibility interval of 35,000–174,000). Recent studies have demonstrated a remarkable and unexpected complexity in social insect pheromone communication, particularly for honeybees (Apis mellifera L.). The intricate interactions characteristic of social insects demand a complex language, based on specialized chemical signals that provide a syntax that is deeper in complexity and richer in nuance than previously imagined. Here, we discuss this rapidly evolving field for honeybees, the only social insect for which any primer pheromones have been identified. Novel research has demonstrated the importance of complexity, synergy, context, and dose, mediated through spatial and temporal pheromone distribution, and has revealed an unprecedented wealth of identified semiochemicals and functions. These new results demand fresh terminology, and we propose adding “colony pheromone” and “passenger pheromone” to the current terms sociochemical, releaser, and primer pheromone to better encompass our growing understanding of chemical communication in social insects.

Bad News for Bees Neonicotinoid insecticides were introduced in the early 1990s and have become one of the most widely used crop pesticides in the world. These compounds act on the insect central nervous system, and they have been shown to be persistent in the environment and in plant tissues. Recently, there have been controversial connections made between neonicotinoids and pollinator deaths, but the mechanisms underlying these potential deaths have remained unknown. Whitehorn et al. (p. 351 , published online 29 March) exposed developing colonies of bumble bees to low levels of the neonicotinoid imidacloprid and then released them to forage under natural conditions. Treated colonies displayed reduced colony growth and less reproductive success, and they produced significantly fewer queens to found subsequent generations. Henry et al. (p. 348 , published online 29 March) documented the effects of low-dose, nonlethal intoxication of another widely used neonicotinoid, thiamethoxam, on wild foraging honey bees. Radio-frequency identification tags were used to determine navigation success of treated foragers, which suggested that their homing success was much reduced relative to untreated foragers.

In social insects, larvae are completely dependent on adults to complete their development. In honey bees (Apis mellifera L.), larval development requires that larvae engage workers in nursing tasks rather than allocating energy in other activities (Le Conte and Hefetz, 2008). Indeed, in the absence of any constraint, workers tend to become active reproductive individual. The queen inhibits worker ovary development via pheromones (Hoover et al., 2003; Wossler and Crewe, 1999), but the inhibition of worker ovary development seems superior when bees are exposed to unsealed brood (Kropacova and Haslbachova, 1970, 1971). Components of the brood pheromone have already been identified and consist of a blend of 10 esters (Le Conte et al., 1989, 1990). Two of these low volatility compounds have been demonstrated to partially decrease worker ovary development (Mohammedi et al., 1998; Pankiw and Garza, 2007). Thus, we asked whether brood emits volatile compounds that could also have an effect on worker ovary activation. We identified a new highly volatile molecule from the larvae E-β-ocimene, that inhibits worker ovary maturation. We used Solid Phase Microextraction (SPME) (65 μm Carbowax) to sample volatiles emitted by different larval instars. We analyzed 20 larvae at stage 1, 2–3 or 10 larvae at stage 4–5. Background Malnutrition is a major factor affecting animal health, resistance to disease and survival. In honey bees (Apis mellifera), pollen, which is the main dietary source of proteins, amino acids and lipids, is essential to adult bee physiological development while reducing their susceptibility to parasites and pathogens. However, the molecular mechanisms underlying pollen's nutritive impact on honey bee health remained to be determined. For that purpose, we investigated the influence of pollen nutrients on the transcriptome of worker bees parasitized by the mite Varroa destructor, known for suppressing immunity and decreasing lifespan. The 4 experimental groups (control bees without a pollen diet, control bees fed with pollen, varroa-parasitized bees without a pollen diet and varroa-parasitized bees fed with pollen) were analyzed by performing a digital gene expression (DGE) analysis on bee abdomens. Results Around 36, 000 unique tags were generated per DGE-tag library, which matched about 8, 000 genes (60% of the genes in the honey bee genome). Comparing the transcriptome of bees fed with pollen and sugar and bees restricted to a sugar diet, we found that pollen activates nutrient-sensing and metabolic pathways. In addition, those nutrients had a positive influence on genes affecting longevity and the production of some antimicrobial peptides. However, varroa parasitism caused the development of viral populations and a decrease in metabolism, specifically by inhibiting protein metabolism essential to bee health. This harmful effect was not reversed by pollen intake. Conclusions The DGE-tag profiling methods used in this study proved to be a powerful means for analyzing transcriptome variation related to nutrient intake in honey bees. Ultimately, with such an approach, applying genomics tools to nutrition research, nutrigenomics promises to offer a better understanding of how nutrition influences body homeostasis and may help reduce the susceptibility of bees to (less virulent) pathogens.

Pheromones in social insects play a key role in the regulation of group homoeostasis. It is well-established that parasites can modify hormone signaling of their host, but less is known about the effect of parasites on pheromone signaling in insect societies. We, thus, tested in honey bees (Apis mellifera) the effect of the widespread parasite Nosema spp. on the production of ethyl oleate (EO), the only identified primer pheromone in honey bee workers. Since environmental stressors like pesticides also can weaken honey bees, we also analyzed the effect of imidacloprid, a neonicotinoid widely used in agriculture, on EO production. We show that, contrary to imidacloprid, Nosema spp. significantly altered EO production. In addition, the level of Nosema infection was correlated positively with the level of EO production. Since EO is involved in the regulation of division of labor among workers, our result suggests that the changes in EO signaling induced by parasitism have the potential to disturb the colony homoeostasis

The possibility to use acetylcholinesterase as biomarker of exposure to deltamethrin insecticide in the honeybee, Apis mellifera were considered. Joined actions of deltamethrin and pirimicarb (carbamate), alone or in association (dual treatment), were investigated on AChE activity in surviving and dead honeybees in order to test its reliability as biomarker. All treatments induced a reduction in tissue AChE activity in dead bees. In surviving bees, deltamethrin treatment induced an important increase of AChE activity that is not abolished by pirimicarb treatment. The analysis of AChE forms revealed an increase in the soluble form in surviving and dead bees and an increase of the membrane form in surviving bees. No direct effect of deltamethrin on soluble and membrane AChE was observed in vitro. The important increase in AChE activity in response to deltamethrin, not altered by pirimicarb treatment, suggests that AChE activity could represent a robust biomarker specific to deltamethrin exposure in living bees.

F1000 Factor 8.0 - Exceptional Faculty of 1000 Biology: Evaluated by Taylor Ricketts 14 nov.2006, Gary Luck 9 jan.2007; The extent of our reliance on animal pollination for world crop production for human food has not previously been evaluated and the previous estimates for countries or continents have seldom used primary data. In this review, we expand the previous estimates using novel primary data from 200 countries and found that fruit, vegetable or seed production from 87 of the leading global food crops is dependent upon animal pollination, while 28 crops do not rely upon animal pollination. However, global production volumes give a contrasting perspective, since 60% of global production comes from crops that do not depend on animal pollination, 35% from crops that depend on pollinators, and 5% are unevaluated. Using all crops traded on the world market and setting aside crops that are solely passively self-pollinated, wind-pollinated or parthenocarpic, we then evaluated the level of dependence on animal-mediated pollination for crops that are directly consumed by humans. We found that pollinators are essential for 13 crops, production is highly pollinator dependent for 30, moderately for 27, slightly for 21, unimportant for 7, and is of unknown significance for the remaining 9. We further evaluated whether local and landscape-wide management for natural pollination services could help to sustain crop diversity and production. Case studies for nine crops on four continents revealed that agricultural intensification jeopardizes wild bee communities and their stabilizing effect on pollination services at the landscape scale.

A brood pheromone identified in honeybee larvae has primer and releaser pheromone effects on adult bees. Using gas chromatography-mass spectrometry (GC-MS) to evaluate fatty acid esters--the pheromonal compounds--in different parts of the larvae, we have localized the source of the esters as the larval salivary glands. A histochemical study describes the glands and confirms the presence of lipids in the glands. Epithelial cells of the gland likely secrete the fatty acids into the lumen of the gland. These results demonstrate the salivary glands to be a reservoir of esters, components of brood pheromone, in honeybee larvae.

A honey bee antennal water-soluble protein, APS2, was purified and characterized as the first Hymenoptera putative odorant-binding protein. Comparison of its measured Mr (13 695.2±1.6) to that of the corresponding cDNA clone shows it does not undergo any post-translational modification other than a 19-residue signal peptide cleavage and formation of three disulfide bridges. These biochemical features are close to those of Lepidoptera odorant-binding proteins. In situ hybridization experiments demonstrated its specific expression in olfactory areas. Based on its higher expression in the worker than in the drone, ASP2 might be more involved in general odorant than in sex pheromone detection.

International audience; The microsporidium Nosema ceranae is a newly prevalent parasite of the European honey bee (Apis mellifera). Although this parasite is presently spreading across the world into its novel host, the mechanisms by it which affects the bees and how bees respond are not well understood. We therefore performed an extensive characterization of the parasite effects at the molecular level by using genetic and biochemical tools. The transcriptome modifications at the midgut level were characterized seven days post-infection with tiling microarrays. Then we tested the bee midgut response to infection by measuring activity of antioxidant and detoxification enzymes (superoxide dismutases, glutathione peroxidases, glutathione reductase, and glutathione-S-transferase). At the gene-expression level, the bee midgut responded to N. ceranae infection by an increase in oxidative stress concurrent with the generation of antioxidant enzymes, defense and protective response specifically observed in the gut of mammals and insects. However, at the enzymatic level, the protective response was not confirmed, with only glutathione-S-transferase exhibiting a higher activity in infected bees. The oxidative stress was associated with a higher transcription of sugar transporter in the gut. Finally, a dramatic effect of the microsporidia infection was the inhibition of genes involved in the homeostasis and renewal of intestinal tissues (Wnt signaling pathway), a phenomenon that was confirmed at the histological level. This tissue degeneration and prevention of gut epithelium renewal may explain early bee death. In conclusion, our integrated approach not only gives new insights into the pathological effects of N. ceranae and the bee gut response, but also demonstrate that the honey bee gut is an interesting model system for studying host defense responses

Background In social insects, the queen is essential to the functioning and homeostasis of the colony. This influence has been demonstrated to be mediated through pheromone communication. However, the only social insect for which any queen pheromone has been identified is the honey bee (Apis mellifera) with its well-known queen mandibular pheromone (QMP). Although pleiotropic effects on colony regulation are accredited to the QMP, this pheromone does not trigger the full behavioral and physiological response observed in the presence of the queen, suggesting the presence of additional compounds. We tested the hypothesis of a pheromone redundancy in honey bee queens by comparing the influence of queens with and without mandibular glands on worker behavior and physiology. Results Demandibulated queens had no detectable (E)-9-oxodec-2-enoic acid (9-ODA), the major compound in QMP, yet they controlled worker behavior (cell construction and queen retinue) and physiology (ovary inhibition) as efficiently as intact queens. Conclusions We demonstrated that the queen uses other pheromones as powerful as QMP to control the colony. It follows that queens appear to have multiple active compounds with similar functions in the colony (pheromone redundancy). Our findings support two hypotheses in the biology of social insects: (1) that multiple semiochemicals with synonymous meaning exist in the honey bee, (2) that this extensive semiochemical vocabulary exists because it confers an evolutionary advantage to the colony.

International audience; Quantifying colony losses is a two-part process. First, colony loss data needs to be collected by surveying beekeepers and then it needs to be calculated and reported in a standardized way. We propose using two different ways to calculate and communicate colony losses. The first we term the total colony losses, sometimes referred to as cumulative loss rate in other systems, which aggregates all losses suffered by all beekeepers surveyed. While the total loss calculation is straightforward, calculating a 95% CI for this metric is complicated by the need to account for the varying sizes of responding beekeepers operations and the nested nature of colony losses within those operations. The second reporting method is average loss is the mean loss suffered by each responding beekeeper. The utility of these two reporting mechanisms differ, in that both are potentially biased by the demographics of the apicultural industry; total loss figures are more heavily influenced by the losses experienced by the few large operations, while average losses are more representative of the many small operations. Additionally, it is important to note that the results from this survey are representative of the responding population alone, and cannot be considered representative of the industry unless some means of identifying and adjusting for varying response is performed.
