Eosinophilia is common in low-resource countries and usually implies helminthiasis. Since helminthiasis is a common cause of eosinophilia and its diagnosis is cumbersome, we hypothesized that broad-spectrum anthelmintic therapy may decrease the eosinophil count and eventually cure helminthiasis, whether microbiologic diagnosis is established or not. We recruited patients with eosinophilia aged 5 years and older who presented to Dhulikhel hospital, Nepal. Patients were treated with albendazole and ivermectin. A stool sample for microscopy was obtained. Of a total of 113 patients, 106 had a follow-up visit and 56 were classified as responders to treatment (defined as a decrease in eosinophil count to below 500 cells/µL, or an absolute decrease of more than 1000 cells/µL). For all patients, we found an absolute decrease in the eosinophil count and for the responding group (more than 50% of the whole cohort), the eosinophil count decreased substantially. All stool samples were negative. The reason for a lack of response in the remaining patients is unclear. In order to ascertain whether eosinophilia should be an indication for anthelmintic treatment, a randomized controlled study of empirical treatment after a thorough microbiologic workup is needed.

The identification of gastrointestinal helminth infections of humans and livestock almost exclusively relies on the detection of eggs or larvae in faeces, followed by manual counting and morphological char-acterisation to differentiate species using microscopy-based techniques. However, molecular approaches based on the detection and quantification of parasite DNA are becoming more prevalent, increasing the sensitivity, specificity and throughput of diagnostic assays. High-throughput sequencing, from single PCR targets through to the analysis of whole genomes, offers significant promise towards providing information-rich data that may add value beyond traditional and conventional molecular approaches; however, thus far, its utility has not been fully explored to detect helminths in faecal samples. In this study, low-depth whole genome sequencing, i.e. genome skimming, has been applied to detect and char-acterise helminth diversity in a set of helminth-infected human and livestock faecal material. The strengths and limitations of this approach are evaluated using three methods to characterise and differ-entiate metagenomic sequencing data based on (i) mapping to whole mitochondrial genomes, (ii) whole genome assemblies, and (iii) a comprehensive internal transcribed spacer 2 (ITS2) database, together with validation using quantitative PCR (qPCR). Our analyses suggest that genome skimming can success-fully identify most single and multi-species infections reported by qPCR and can provide sufficient cov-erage within some samples to resolve consensus mitochondrial genomes, thus facilitating phylogenetic analyses of selected genera, e.g. Ascaris spp. Key to this approach is both the availability and integrity of helminth reference genomes, some of which are currently contaminated with bacterial and host sequences. The success of genome skimming of faecal DNA is dependent on the availability of vouchered sequences of helminths spanning both taxonomic and geographic diversity, together with methods to detect or amplify minute quantities of parasite nucleic acids in mixed samples.(c) 2023 The Author(s). Diagnosis of soil-transmitted helminth (STH) and schistosome infections relies largely on conventional microscopy which has limited sensitivity, requires highly trained personnel and is error-prone. Rapid advances in miniaturization of optical systems, sensors and processors have enhanced research and development of digital and automated microscopes suitable for the detection of these diseases in resource-limited settings. While some studies have reported proof-of-principle results, others have evaluated the performance of working prototypes in field settings. The extensive commercialization of these innovative devices has, however, not yet been achieved. This review provides an overview of recent publications (2010–2022) on innovative field applicable optical devices which can be used for the diagnosis of STH and schistosome infections. Using an adapted technology readiness level (TRL) scale taking into account the WHO target product profile (TPP) for these diseases, the developmental stages of the devices were ranked to determine the readiness for practical applications in field settings. From the reviewed 18 articles, 19 innovative optical devices were identified and ranked. Almost all of the devices (85%) were ranked with a TRL score below 8 indicating that, most of the devices are not ready for commercialization and field use. The potential limitations of these innovative devices were discussed. We believe that the outcome of this review can guide the end-to-end development of automated digital microscopes aligned with the WHO TPP for the diagnosis of STH and schistosome infections in resource-limited settings.

Much of our understanding of the targets of IgE comes from studies of allergy, though little is known about the natural immunogenic targets seen after parasitic worm infections.We used human monoclonal antibodies (mAbs) for an unbiased and comprehensive characterization of the immunodominant antigens targeted by IgE in conditions like allergy or helminth infection that are associated with elevated levels of IgE.Using human hybridoma technology to immortalize IgE encoding B-cells from peripheral blood of subjects with filarial infections and elevated IgE, we generated naturally occurring human IgE mAbs. B-cell cultures were screened in an unbiased manner for IgE production without regard to specificity. Isolated IgE mAbs were then tested for binding to Brugia malayi somatic extracts using ImmunoCAP, immunoblot, and ELISA. Immunoprecipitation followed by mass spectrometry proteomics was used to identify helminth antigens that were then expressed in Escherichia coli for IgE binding characterization.We isolated 56 discrete IgE mAbs from 7 individuals with filarial infections. From these mAbs, we were able to definitively identify 19 filarial antigens. All IgE mAbs targeted filarial excreted/secretory proteins, including a family of previously uncharacterized proteins. Interestingly, the transthyretin-related antigens acted as the dominant inducer of the filaria-specific IgE antibody response. These filaria-specific IgE mAbs were potent inducers of anaphylaxis when passively administered to human FcεRI-expressing mice.We generated human hybridomas secreting naturally occurring helminth-specific IgE mAbs from filarial-infected subjects. This work provides much-needed insight into the ontogeny of helminth-induced immune response and IgE antibody response.

Helminths are remarkably successful parasites that can invade various mammalian hosts and establish chronic infections that can go unnoticed for years despite causing severe tissue damage. To complete their life cycles, helminths migrate through multiple barrier sites that are densely populated by a complex array of hematopoietic and non-hematopoietic cells. While it is clear that type 2 cytokine responses elicited by immune cells promote worm clearance and tissue healing, the actions of non-hematopoietic cells are increasingly recognized as initiators, effectors and regulators of anti-helminth immunity. This review will highlight the collective actions of specialized epithelial cells, stromal niches, stem, muscle and neuroendocrine cells as well as peripheral neurons in the detection and elimination of helminths at mucosal sites. Studies dissecting the interactions between immune and non-hematopoietic cells will truly provide a better understanding of the mechanisms that ensure homeostasis in the context of helminth infections.

Background: MicroRNAs (miRNAs) are about 22-nucleotide, small, noncoding RNAs that control gene expression post-transcriptionally. Helminth parasites usually express a unique repertoire of genes, including miRNAs, across different developmental stages with subtle regulatory mechanisms. Objective: There is a necessity to investigate the involvement of miRNAs in the development of parasites, host-parasite interaction, immune evasion and their abilities to govern infection in hosts. MiRNAs present in helminth parasites have been summarized in the current systematic review (SR). Methods: Electronic databases, including PubMed, Scopus, ProQuest, Embase, and Google Scholar search engine, were searched to identify helminth miRNA studies published from February 1993 till December 2019. Only the published articles in English were included in the study. Results: A total of 1769 articles were preliminarily recorded. Following the strict inclusion and exclusion criteria, 105 studies were included in this SR. Most of these studies focused on the identification of miRNAs in helminth parasites and/or probing of differentially expressed host miRNA profiles in specific relevant tissues, while 12 studies aimed to detect parasite-derived miRNAs in host circulating system and 15 studies characterized extracellular vesicles (EV)-derived miRNAs secreted by parasites. Conclusion: In the current SR, information regarding all miRNAs expressed in helminth parasites has been comprehensively provided and the utility of helminth parasitesderived miRNAs in diagnosis and control of parasitic infections has been discussed. Furthermore, functional studies on helminth-derived miRNAs have also been presented.

7 páginas., In 2020, WHO recognised the importance of strongyloidiasis alongside soil-transmitted helminths (STH) in their 2021–30 roadmap, which aspires to target Strongyloides stercoralis with preventive chemotherapy by use of ivermectin. Combination treatment with both albendazole, the primary drug used to treat STH, and ivermectin, would improve the efficiency of mass drug administration targeting both STH and S stercoralis. In this Personal View, we discuss the challenges and opportunities towards the development of an efficient control programme for strongyloidiasis, particularly if it is to run concurrently with STH control. We argue the need to define the prevalence threshold to implement preventive chemotherapy for S stercoralis, the target populations and optimal dosing schedules, and discuss the added benefits of a fixed-dose coformulation of ivermectin and albendazole. Implementation of an efficient control programme will require improvements to current diagnostics, and validation of new diagnostics, to target and monitor S stercoralis infections, and consideration of the challenges of multispecies diagnostics for S stercoralis and STH control. Finally, the evolution of ivermectin resistance represents a credible risk to control S stercoralis; we argue that genome-wide approaches, together with improved genome resources, are needed to characterise and prevent the emergence of resistance. Migrations performed by helminths are impressive and diverse, and accumulating evidence shows that many are controlled by sophisticated sensory programs. The migrations of vector-borne helminths are particularly complex, requiring precise, stage-specific regulation. We review the contrasting states of knowledge on snail-borne schistosomes and mosquito-borne filarial nematodes. Rich observational data exist for the chemosensory behaviors of schistosomes, while the molecular sensory pathways in nematodes are well described. Recent investigations on the molecular mechanisms of sensation in schistosomes and filarial nematodes have revealed some features conserved within their respective phyla, but adaptations correlated with parasitism are pronounced. Technological developments are likely to extend these advances, and we forecast how these technologies may be applied.

Demarginalization through initiation of resettlement program since 1978 is an inevitable progress faced by the indigenous Orang Asli (OA) population in Peninsular Malaysia. As Malaysian huntergatherers, the Negrito has been exposed to various environmental-cultural variations. These changes may influence the pattern of soil-transmitted helminth (STH) infections, the common malady amongst OA. This study evaluated the deworming effects of single-dosage albendazole (400 mg) and STH-reinfection rate between Negritos who are still living in the inland jungle versus those living in resettlements at town peripheries (RPS). Stool samples from the consented participants were first examined using the direct faecal smear, formalin-ether sedimentation and Kato Katz techniques. Subsequently, stool collections were carried out in three time points following treatment (i.e., 21 days, 3 months and 6 months). In brief, a total number of 54 Negritos (inland: 24; RPS: 30) with a complete set of stool collection was included in this longitudinal study. This study revealed 72.2% cure rate against T. trichiura in the inland but only 15.0% in the RPS. Although the efficacy of albendazole against T. trichiura was ultimately low in the RPS, 62.6% egg reduction rate (ERR) (arithmetic mean) was noted (p = 0.001). For A. lumbricoides and hookworm, high cure rates were found in both communities (85.7-100.0%). Reinfection for T. trichiura was seen in less than 1 month with higher rate in the RPS (90.0%) as opposed to the inland (44.4%) at 21 days following treatment. This study found that the inland OA had better tolerability to single-dosage albendazole and experienced slower STH reinfection rates versus the RPS. Hence, the selection of albendazole dosage should be targeted and the use of single- dosage albendazole (biannually) would be more suitable for the inland OA. Conversely, we propose the use of 3-days albendazole regimens in the resettled RPS population.

Background. Intestinal helminth infections are still public health problems in tropical and subtropical countries including Ethiopia. This review and meta-analysis aimed to produce the pooled prevalence and associated risk factors of human intestinal helminth parasitic infections (HIHPIs) in Ethiopia. Methods. Articles written in English were searched from online databases. Sixty-seven studies were included. Meta-analysis was computed using STATA version 14. Result. The pooled prevalence of HIHPIs was (33.35%, 95% CI: 28.85%, 37.86%). Ascaris lumbricoides (10.84%, 95% CI: 9.34, 12.34), hookworm spp. (8.89%, 95% CI: 7.75, 10.04), Schistosoma mansoni (4.22%, 95% CI: 3.64, 4.81), Trichuris trichiura (2.51%, 95% CI: 2.17, 2.86), Hymenolepis nana (2.29%, 95% CI: 1.96, 2.63), Taenia species (1.01%, 95% CI: 0.80, 1.22), Strongyloides stercoralis (1.17%, 95% CI: 0.92, 1.41), and Enterobius vermicularis(0.71%, 95% CI: 0.52, 0.90) were recorded. Handwashing before food (OR: 5.22,95% CI: 3.49, 6.94), handwashing after toilet (OR: 3.03, 95%; CI: 1.01, 5.05), age (OR: 1.66, 95% CL. 1.09, 2.23), open defecation (OR: 2.42, 95% CI: 1.60, 3.24), eating raw and unwashed vegetables/fruits (OR: 1.98, 95%; CI: 1.30, 2.66), maternal education (OR: 1.81, 95% CI: 0.91, 2.72), family income (OR: 2.00, 95% CI: 0.87, 3.31), source of drinking water (OR: 3.12, 95% CI: 1.96, 4.27), swimming/contact with river water (OR: 1.90, 95% CI: 1.11, 2.69), barefoot (OR: 3.28, 95% CI: 1.67, 4.88), playing with soil (OR: 2.64, 95% CI: 1.40, 3.88), and family size (OR: 3.75, 95% CI: 2.03, 5.46) were factors associated with HIHPIs in Ethiopia. High heterogeneity of the prevalence of HIHPIs was observed among the studies within and among regions (I2 > 99.6% and P ≤ 0.001 ). Conclusion. HIHPIs in Ethiopia were significantly high. Therefore, special attention should be given by all stakeholders to minimize HIHPIs in Ethiopia.

Infection with helminth parasites affects more than 1.5 billion people and is concentrated in global areas of extreme poverty, having a significant impact on public health, social life and the economy. Upon entry into the host, helminth parasites often migrate through specific tissues triggering host immunity. The immune response triggered by helminth infections is complex and depends on parasite load, site of infection, acuteness/chronicity of the infection and is species-dependent. In general, susceptibility or resistance to the infection involves the participation of the innate immune response and then the balance between several effector CD4

The health consequences of soil-transmitted helminth (STH) infections are often attributed to parasite-caused tissue damage and nutrient loss, combined with immune energy costs. However, this view overlooks additional pathways by which infection can alter host energetics. Here, we take a first step toward defining this suite of energetic pathways and clarifying their cumulative impact on health. We consider how STH characteristics and human variation influence host-parasite interactions, as well as the initial and downstream energetic costs of infection. We conclude by discussing how complex biological, cultural, and temporal factors may affect host energetics, and explore promising avenues for future research. Our goal is to provide an integrative framework for more comprehensively measuring and addressing the energetic consequences of STH infection.

Helminth infections are detrimental to the overall health of dogs; therefore, this study aimed to identify antiparasitic-resistant helminths and evaluate the infection rate and risk factors for parasitism in canines. For this purpose, a parasitological evaluation of 38 randomly selected animals was performed, followed by the evaluation of the anthelminthic efficacy of three drugs: pyrantel pamoate with praziquantel (Canex Composto®), fenbendazole (Fenzol Pet®), and milbemycin oxime with praziquantel (Milbemax C®). Among the evaluated animals, 22/38 (57.89%) tested negative and 16/38 (42.71%) tested positive for Ancylostoma caninum infection. Evaluation of the efficacy of antiparasitic drugs showed that 12/16 (75%) dogs were infected with helminths that were susceptible to pyrantel pamoate with praziquantel. Among those for which pyrantel pamoate with praziquantel was not effective, 3/4 (75%) were susceptible to fenbendazole, while the remaining case resistant to both pyrantel pamoate with praziquantel and fenbendazole was sensitive to milbemycin oxime with praziquantel (100%). The odds ratio of infection in dogs inhabiting environments containing soil or grass was 6.67 times higher than that in dogs inhabiting impermeable environments. Mixed-breed dogs (SRD) were 6.54 times more likely to be infected compared to purebred dogs. A. caninum resistant to pyrantel pamoate with praziquantel (4/16, 25%) and fenbendazole (1/4, 25%) were detected. The results of this study demonstrated the importance of coproparasitological monitoring by professionals before and after treatments to assess antiparasitic drug effectiveness, ensure animal health and welfare, and minimize animal exposure to risk factors.

Type 2-high asthma is a chronic inflammatory disease of the airways which is increasingly prevalent in countries where helminth parasite infections are rare, and characterized by T helper 2 (Th2)-dependent accumulation of eosinophils in the lungs. Regulatory cytokines such as TGF-β can restrain inflammatory reactions, dampen allergic Th2 responses, and control eosinophil activation. The murine helminth parasite Heligmosomoides polygyrus releases a TGF-β mimic (Hp-TGM) that replicates the biological and functional properties of TGF-β despite bearing no structural similarity to the mammalian protein. Here, we investigated if Hp-TGM could alleviate allergic airway inflammation in mice exposed to Alternaria alternata allergen, house dust mite (HDM) extract or alum-adjuvanted ovalbumin protein (OVA). Intranasal administration of Hp-TGM during Alternaria exposure sharply reduced airway and lung tissue eosinophilia along with bronchoalveolar lavage fluid IL-5 and lung IL-33 cytokine levels at 24 h. The protective effect of Hp-TGM on airway eosinophilia was also obtained in the longer T-cell mediated models of HDM or OVA sensitisation with significant inhibition of eotaxin-1, IL-4 and IL-13 responses depending on the model and time-point. Hp-TGM was also protective when administered parenterally either when given at the time of allergic sensitisation or during airway allergen challenge. This project has taken the first steps in identifying the role of Hp-TGM in allergic asthma and highlighted its ability to control lung inflammation and allergic pathology. Future research will investigate the mode of action of Hp-TGM against airway allergic eosinophilia, and further explore its potential to be developed as a biotherapeutic in allergic asthma.

Morocco has a great diversity of reptiles; more than 26% of the herpetofauna species are considered to be endemic. Nevertheless, there is little information available on helminth parasites of Moroccan lizards. The purpose of this article is to establish a helminth list using non-invasive methods for natural populations of seven lizard species: Acanthodactylus maculatus, Chalcides mionecton, Chalcides montanus, Chalcides polylepis, Quedenfeldtia moerens, Quedenfeldtia trachyblepharus, and Tarentola mauritanica. For each species, prevalence and intensity of infection were given as well as their relationship to some determinant parameters (sex, age class, size, altitude, and seasons). Four species of nematodes, Parapharyngodon micipsae, Pharyngodon mamillatus, Spauligodon auziensis, Thelandros alatus, and unassigned Spauligodon were found. Mean helminth intensity for the seven lizard species was 7.3 ± 16.2 SD (range = 1-92 in infected lizards), with total infection prevalence of 5.6%. Mixed infections were observed in Chalcides montanus and Chalcides polylepis co-infected by Pharyngodon mamillatus and Spauligodon sp. Furthermore, the intensity of infection by nematodes were significantly different among the seven studied species. The nematode infection in Chalcides mionecton were related to sex, host size, and altitude; in Chalcides polylepis to the host age; in Quedenfeldtia trachyblepharus to elevation, age, and host size; and in Tarentola Mauritanica to age, body size, seasons, and altitude. In conclusion, our study provides data about nine new host and locality records and the effect of some determinant factors on host parasitism.

INTRODUCTION: There is considerable research interest in the role of helminth infections in the development of allergic diseases. However, findings from previous studies are mixed. Existing systematic reviews of these studies are outdated. We performed a systematic review of the global literature on the association between helminth infections and development and clinical outcomes of allergic diseases. METHODS: We searched Cochrane Library, MEDLINE, EMBASE, ISI Web of Science, PubMed, Global Index Medicus, Scielo, KoreaMed, Google Scholar, and Lilacs for studies published up to January 2020. We included observational epidemiological studies (cohort, case-control and cross-sectional studies) of children and adults reporting associations between helminth infections and asthma, allergic rhinitis, eczema and atopy. We performed random-effects meta-analysis to summarize the effect estimates. RESULTS: We included 80 studies with 99,967 participants. In the meta-analyses, we did not observe an overall association between helminth infections and allergic diseases. There was, however, evidence that A. lumbricoides infections was associated with an increased risk of bronchial hyperreactivity in children (RR:1.41, 95%CI: 1.17-1.70; I2=50, p for I2=0.09), and was associated with an increased risk of atopy among helminth-infected adults (RR:1.37, 95%CI: 1.18-1.61; I2=52, p for I2=0.02). We found no study that addressed the association between helminth infection and clinical outcomes of allergic diseases. The overall strength of the underlying evidence was low to moderate. CONCLUSION: Helminth infections may increase the risk of bronchial hyperreactivity in children and atopy in adults. Well-designed longitudinal cohorts may help clarify potential causal associations between chronic helminth infections and allergic diseases.

In recent years, several authors have warned that the number of trained experts in parasite taxonomy and systematics is declining rapidly, and that the whole field is at risk. However, to date there has been no quantitative analysis to support these claims. Here, we provide the first such assessment, focusing on helminths parasitic in fish as an example, and using a representative dataset comprising over 2000 helminth species described in the past two decades. Based on standard indices of inequality, we demonstrate that a small group of highly prolific taxonomists are associated with the vast majority of new species descriptions, indicating that the research output in parasite discovery is concentrated in the hands of a small number of individuals. This situation has not improved over time. Furthermore, there has been no turnover over time, i.e., no replacement of the most prolific taxonomists: the individual researchers ranking among the most prolific describers of new parasite helminth species in the past decade were generally also the most prolific in the decade before that. Finally, based on the year in which these most prolific taxonomists published their first species description, we estimate that a large proportion of them are in the latter stages of their career. Inequalities in research output are the norm across scientific disciplines. However, persistent inequality in the number of species description per author, coupled with the same individuals ranking as most prolific over time and a majority of them in late career, all combine to support earlier claims that parasite taxonomy may well face a crisis in the form of an impending loss of taxonomic expertise.

Background Millions of people throughout the world suffer from parasite infections. Traditionally, technicians use manual eye inspection of microscopic specimens to perform a parasite examination. However, manual operations have limitations that hinder the ability to obtain precise egg counts and cause inefficient identification of infected parasites on co-infections. The technician requirements for handling a large number of microscopic examinations in countries that have limited medical resources are substantial. We developed the helminth egg analysis platform (HEAP) as a user-friendly microscopic helminth eggs identification and quantification platform to assist medical technicians during parasite infection examination. Methods Multiple deep learning strategies including SSD (Single Shot MultiBox Detector), U-net, and Faster R-CNN (Faster Region-based Convolutional Neural Network) are integrated to identify the same specimen allowing users to choose the best predictions. An image binning and egg-in-edge algorithm based on pixel density detection was developed to increase the performance. Computers with different operation systems can be gathered to lower the computation time using our easy-to-deploy software architecture. Results A user-friendly interface is provided to substantially increase the efficiency of manual validation. To adapt to low-cost computers, we architected a distributed computing structure with high flexibilities. Conclusions HEAP serves not only as a prediction service provider but also as a parasitic egg database of microscopic helminth egg image collection, labeling data and pretrained models. All images and labeling resources are free and accessible at http://heap.cgu.edu.tw . HEAP can also be an ideal education and training resource for helminth egg examination.

Durrington Walls was a large Neolithic settlement in Britain dating around 2500 BCE, located very close to Stonehenge and likely to be the campsite where its builders lived during its main stage of construction. Nineteen coprolites recovered from a midden and associated pits at Durrington Walls were analysed for intestinal parasite eggs using digital light microscopy. Five (26%) contained helminth eggs, 1 with those of fish tapeworm (likely Dibothriocephalus dendriticus) and 4 with those of capillariid nematodes. Analyses of bile acid and sterol from these 5 coprolites show 1 to be of likely human origin and the other 4 to likely derive from dogs. The presence of fish tapeworm reveals that the Neolithic people who gathered to feast at Durrington Walls were at risk of infection from eating raw or undercooked freshwater fish. When the eggs of capillariids are found in the feces of humans or dogs it normally indicates that the internal organs (liver, lung or intestines) of animals with capillariasis have been eaten, and eggs passed through the gut without causing disease. Their presence in multiple coprolites provides new evidence that internal organs of animals were consumed. These novel findings improve our understanding of both parasitic infection and dietary habits associated with this key Neolithic ceremonial site.

Understanding the epidemiology of canine parasitic infections, gastrointestinal helminthic infections in particular, is crucial for designing an efficient control programs targeted at minimizing the risks of zoonotic transmission. A total of 470 dogs attending veterinary clinics in Northeastern Nigeria were screened for gastrointestinal helminths using standard parasitological techniques. Epidemiological risk factors associated with the canine parasitic infections were assessed using multivariable logistic regression analysis. In total, 77.9% (366/470) of dogs had harboured one or more species of the gastrointestinal helminths: Ancylostoma caninum (40.2%), Toxocara canis (35.1%) Trichuris vulpis (26.6%), Spirocerca lupi (5.7%), Taenia/Echinococcus (12.3%), Dipylidium caninum (10.9%) and Diphyllobothriid ova (2.8%). Female dogs (OR=2.1; p=0.02), local breeds (OR=2.3; p0.01), younger dogs (OR=3.2; p0.001), dogs adopted as security guards (OR=1.8; p0.01) and poor body condition score (OR=3.1; p0.001) were strongly associated with A. caninum infection. For T. canis and T. vulpis, younger dogs (OR=2.5; p=0.02 and OR=2.3; p0.01) and poor body condition score (OR=4.2; p0.001 and OR=2.2; p0.01) were the strong predictors for infection, whereas female dogs (OR=2.8; p=0.02), younger dogs (OR=4.1; p0.001) and poor body condition score (OR=4.0; p0.001) were strongly associated with D. caninum infection in dogs. The results revealed high prevalence of gastrointestinal helminths-some of which have zoonotic potential-among dogs underlying huge public health risk.

Intestinal helminth management in horses has both clinical and epidemiologic relevance, in additional association with anthelmintic resistance. The Swiss Army employs military owned riding horses and privately owned pack horses, which are brought together for service periods up to 12 weeks. We compared husbandry conditions and intestinal helminth management of both groups via questionnaire and analysed faecal samples of 53 riding horses and 130 pack horses using combined sedimentation/flotation, the McMaster method and larval cultures. Riding horses only had cyathostomin infections (prevalence: 60,4 %), while pack horses harboured cyathostomins (71,5 %), Parascaris sp. (6,9 %) and Strongylus vulgaris (1,5 %). Regression models combining faecal sample results with questionnaire data unveiled correlations of husbandry practices with parasite frequencies identifying risk and protective factors. Pasture management, hygiene and deworming practices were highly variable for pack horses, while for riding horses there was an overall concept. This included a selective deworming strategy with faecal egg counts (FECs) of strongyles prior to deworming, applying a threshold of 200 eggs per gram of faeces (epg). Anthelmintic treatments based on FECs, weekly faeces removal on pastures (pastures), the use of macrocyclic lactones and deworming horses regularly were identified as protective factors regarding the 200 epg threshold for strongyle eggs. Accordingly, the mean epg for strongyle eggs between the groups (111 and 539 in riding and pack horses, respectively) was significantly different (p0,001). Overall, intestinal helminth management in pack -horses showed room for improvement regarding pasture hygiene, the used anthelmintics and the frequency of deworming, from which all Swiss Army horses would benefit, as they share pastures during their -service, therefore entailing the risk of parasite transmission.Das Management intestinaler Helminthen bei Pferden hat sowohl klinische als auch epidemiologische Relevanz, ebenso im Zusammenhang mit Anthelminthika-­Resistenz. Die Schweizer Armee setzt Armee-eigene Reitpferde und privat gehaltene Trainpferde ein, welche für Diensteinsätze von bis zu 12 Wochen zusammengebracht werden. Wir haben die Haltungsbedingungen und das intestinale Helminthen-Management beider Gruppen mittels Fragebogen verglichen und haben Kotproben von 53 Reitpferden und 130 Tragpferden mittels kombiniertem Sedimentations-/Flotationsverfahren, McMaster Methode und Larvenkulturen analysiert. Die Reitpferde zeigten lediglich Befall mit kleinen Strongyliden (Prävalenz: 60,4 %), während die Trag­pferde kleine Strongyliden (71,5 %), Parascaris sp. (6,9 %) und Strongylus vulgaris (1,5 %) beherbergten. Regres­sionsmodelle, in welchen Kotproben-Befunde mit den Fragebogendaten kombiniert wurden, zeigten Zusammenhänge zwischen Haltungspraktiken und der Häufigkeit bestimmter Parasiten und führten zur ­Iden­tifizierung von Risiko- und Schutzfaktoren. Weidemanagement, Hygiene und Entwurmungspraktiken variierten stark für die Tragpferde, während für die Reitpferde ein einheitliches Konzept vorhanden war. Dieses beinhaltete eine selektive Entwurmungsstra­tegie mit quantitativen Kotuntersuchungen auf Strongy­liden vor Entwurmung (Schwellenwert von 200 Eiern pro Gramm Kot, EpG). Entwurmungen basierend auf quantitativen Kotuntersuchungen, die wöchentliche Beseitigung von Kot auf der Weide, die Verabreichung von makrozyklischen Laktonen und eine regelmässige Entwurmung der ­Pferde erwiesen sich als Schutzfaktoren bezüglich des Schwellenwertes von 200 EpG für Strongyliden. Dementsprechend waren die durchschnittlichen Strongyliden-Eizahlen zwischen den zwei Gruppen (111 bei den Reitpferden und 539 bei den Tragpferden) signifikant unterschiedlich (p0,001). Insgesamt wies das Endoparasitenmanagement bei den Tragpferden Verbesserungspotential auf bezüglich Weidehygiene, verwendeten Anthelminthika und Entwurmungsfrequenz. Hiervon würden alle Armeepferde profitieren, da sie während ihres Dienstes die Weiden teilen und somit das Risiko einer Parasiten-Übertragung besteht.La gestion des helminthes intestinaux chez les chevaux présente une importance clinique et épidémiologique, en association avec la résistance aux anthelminthiques. L’armée suisse emploie des chevaux de selle militaires et des chevaux de bât privés qui sont réunis pour des périodes de service allant jusqu’à 12 semaines. Nous avons comparé la détention animale et la gestion des helminthes intestinaux des deux groupes par le biais d’un questionnaire et analysé les échantillons fécaux de 53 chevaux de selle et 130 chevaux de bât en utilisant la sédimentation/flottation combinée, la méthode McMaster et les cultures larvaires. Les chevaux de selle ne présentaient que des infections à cyathostomes (prévalence: 60,4 %), tandis que les chevaux de bât hébergeaient des cyathostomes (71,5 %), Parascaris sp. (6,9 %) et Strongylus vulgaris (1,5 %). Des modèles de régression combinant les résultats des échantillons fécaux et les données du questionnaire ont révélé des corrélations entre les pratiques de détention animale et la fréquence des parasites, identifiant les facteurs de risque et de protection. La gestion des pâturages, l’hygiène et les pratiques en matière de vermifugation étaient très variables pour les chevaux de bât, tandis que pour les chevaux de selle, il existait un concept unitaire. Ceci comprenait stratégie de vermifugation sélective avec comptage des œufs fécaux (CEF) de strongles avant la vermifugation, en appliquant un seuil de 200 œufs par gramme de fèces (opg). Les traitements anthelminthiques basés sur les CEF, l’enlèvement hebdomadaire des crottins sur le pâturage, l’utilisation de lactones macrocycliques et la vermifugation régulière des chevaux ont été identifiés comme des facteurs de protection concernant le seuil de 200 opg pour les œufs de strongles. En conséquence, l’iog moyen pour les œufs de strongles entre les groupes (111 et 539 chez les chevaux de selle et de bât, respectivement) était significativement différent (p0,001). Globalement, la gestion des helminthes intestinaux chez les chevaux de bât a montré qu’il est possible d’améliorer l’hygiène des pâturages, le choix des anthelminthiques et la fréquence des vermifuges, ce dont tous les chevaux de l’armée suisse bénéficieraient, car ils partagent les pâturages pendant leur service, ce qui entraîne un risque de transmission des parasites.La gestione degli elminti intestinali nei cavalli ha una rilevanza sia clinica che epidemiologica, in associazione alla resistenza agli antielmintici. L’esercito svizzero impiega cavalli da equitazione di proprietà dell’esercito e cavalli da soma di proprietà privata, che vengono riuniti per periodi di servizio fino a 12 settimane. Abbiamo confrontato le condizioni d'allevamento e la gestione degli elminti intestinali di entrambi i gruppi tramite un questionario e abbiamo analizzato campioni fecali di 53 cavalli da equitazione e 130 cavalli da soma utilizzando la sedimentazione/flottazione combinata, il ­metodo McMaster e le colture larvali. I cavalli da equitazione avevano solo infezioni da cyathostomi (prevalenza: 60,4 %), mentre i cavalli da soma ospitavano cyathostomi (71,5 %), Parascaris sp. (6,9 %) e Strongylus vulgaris (1,5 %). I modelli di regressione che combinano i risultati dei campioni fecali con i dati del questionario hanno rivelato correlazioni tra le pratiche di allevamento animale e la frequenza dei parassiti, identificando fattori di rischio e di protezione. La gestione del pascolo, l’igiene e le pratiche di sverminazione sono risultate molto variabili tra i cavalli da soma, mentre per i cavalli da equitazione vi è stato un approccio più uniforme. I trattamenti antielmintici basati sulle CEF, la rimozione settimanale delle feci sul pascolo, l’uso di lattoni macrociclici e la sverminazione regolare dei cavalli sono stati identificati come fattori protettivi rispetto alla soglia di 200 upg per le uova di strongili. Di conseguenza, l’upg medio per le uova di strongili tra i gruppi (111 e 539 nei cavalli da equitazione e da soma, rispettivamente) era significativamente diverso (p0,001). Parasitic diseases are considered to be one of the most prevalent pathologies worldwide. They are characterized as one of the most critical sanitary problems in cattle, causing a decrease in the productive capacity of parasitized animals, which translates into economic losses. Intestinal parasitism in cattle is caused by protozoa and helminths, and its manifestation is generally multi-etiological. Clinical signs in gastrointestinal parasitism may vary depending on parasite load, parasite species, and host immunity. This research aimed to determine the prevalence of the main parasitic families affecting cattle in the central province of the department of Boyacá. A cross-sectional study with simple random sampling was carried out, where 716 fecal samples were taken and processed using a modified Ritchie technique. An overall prevalence of 95,6% was determined, and the most prevalent families were Trichostrongylidae, Eimeriidae, Taeniidae, and Trichuridae. The age showed no significant statistical association with most of the parasitic families, except for the Strongyloididae family. The breeds showed a correlation with the Trichostrongylidae, Eimeriidae, Strongylidae, Chabertiidae, and Taeniidae families. The results show the high prevalence of GIP (gastrointestinal parasites) in cattle of the central province of the department of Boyacá. Resumen Las enfermedades parasitarias son consideradas como una de las patologías más prevalentes alrededor del mundo. Se caracterizan por ser uno de los problemas sanitarios de mayor importancia en los bovinos, causando una disminución en la capacidad productiva de los animales parasitados, lo cual se traduce en pérdidas económicas. El parasitismo gastrointestinal en bovinos es causado por protozoos y helmintos, y generalmente su presentación es multietiológica. Los signos clínicos en el parasitismo gastrointestinal pueden variar dependiendo de la carga parasitaria, la especie parasitaria y la inmunidad del huésped. El objetivo de esta investigación fue determinar la prevalencia de las principales familias parasitarias que afectan a los bovinos de la provincia central del departamento de Boyacá. Se realizó un estudio de corte transversal con muestreo aleatorio simple, en el cual se tomaron 716 muestras de materia fecal que fueron procesadas mediante la técnica de Ritchie modificada. Se determinó una prevalencia general de 95,6 %, en donde las familias más prevalentes fueron Trichostrongylidae, Eimeriidae, Taeniidae y Trichuridae. La edad no presentó asociación estadística significativa con la mayoría de las familias parasitarias, a excepción de la familia Strongyloididae. La raza mostró asociación con las familias Trichostrongylidae, Eimeriidae, Strongylidae, Chabertiidae y Taeniidae. Los resultados muestran una alta prevalencia de PGI (parásitos gastrointestinales) en la provincia central de Boyacá. Resumo As doenças parasitárias são consideradas uma das patologias mais prevalentes em todo o mundo, caracterizam-se por ser um dos mais importantes problemas de saúde dos bovinos, apresentando uma diminuição da capacidade produtiva dos animais parasitados, que se traduzem em perdas econômicas. O parasitismo gastrointestinal em bovinos é causado por protozoários e helmintos e sua apresentação é geralmente multe etiológica. Os sinais clínicos no parasitismo gastrointestinal podem variar dependendo da carga do parasita, da espécie parasitária e da imunidade do hospedeiro. O objetivo desta pesquisa foi determinar a prevalência das principais famílias de parasitas que afetam o gado na província Central do departamento de Boyacá. Foi realizado um estudo transversal com amostragem aleatória simples, no qual 716 amostras de fezes foram retiradas e processadas pela técnica de Ritchie modificada. Foi determinada uma prevalência geral de 95,6%, onde as famílias mais prevalentes foram Trichostrongylidae, Eimeriidae, Taeniidae e Trichuridae. A idade não apresentou associação estatística significativa com a maioria das famílias de parasitas, com exceção da família Strongyloididae. A raça apresentou associação com as famílias Trichostrongylidae, Eimeriidae, Strongylidae, Chabertiidae e Taeniidae. Os resultados mostram uma alta prevalência de IGP na província central de Boyacá.

Background Soil-transmitted helminths (STH) and schistosome parasites are highly aggregated within the human population. The probability distribution of worms per person is described well by the negative binomial probability distribution with aggregation parameter, k, which varies inversely with parasite clustering. The relationship between k and prevalence in defined populations subject to mass drug administration is not well understood. Methods and Results We use statistical methods to estimate k using two large independent datasets for STH and schistosome infections from India and Niger, respectively, both of which demonstrate increased aggregation of parasites in a few hosts, as the prevalence of infections declines across the dataset. Conclusions A greater attention needs to be given in monitoring and evaluation programmes to find and treat the remaining aggregates of parasites.

Background: Several in vitro and in vivo biological activities of serotonin, 5- hydroxytryptamine (5-HT), as a bioactive molecule, and its transporter (5-HT-Tr) were evaluated in parasitic infections. Objective: Herein, the roles of 5-HT and 5-HTR in helminths and protozoan infections with medical and veterinary importance are reviewed. Methods: We searched information in 4 main databases and reviewed published literature about the serotonin transporter's role as a promising therapeutic target against pathogenic parasitic infections between 2000 and 2021. Results: Based on recent investigations, 5-HT and 5-HT-Tr play various roles in parasite infections, including biological function, metabolic activity, organism motility, parasite survival, and immune response modulation. Moreover, some of the 5-HT-TR in Schistosoma mansoni showed an excess of favorite substrates for biogenic amine 5-HT compared to their mammalian hosts. Furthermore, the main neuronal protein related to the G protein-coupled receptor (GPCR) was identified in S. mansoni and Echinococcus granulosus, playing main roles in these parasites. In addition, 5-HT increased in toxoplasmosis, giardiasis, and Chagas disease. On the other hand, in Plasmodium spp., different forms of targeted 5-HTR stimulate Ca2+ release, intracellular inositol triphosphate (ITP), cAMP, and protein kinase A (PKA) activity. Conclusion: This review summarized the several functional roles of the 5-HT and the importance of the 5-HT-TR as a drug target with minimal harm to the host to fight against helminths and protozoan infections. Hopefully, this review will shed light on research regarding serotonin transporter-based therapies as a potential drug target soon.

Almost 80% of health problems in the developing world are due to malnutrition and infectious diseases, which are mainly parasitic. Updated records on the prevalence of parasitic infections and the potential risk factors are essential to enhancing control strategies. Therefore, this study was conducted to evaluate the current situation of intestinal parasitism among patients attending teaching hospitals in Zagazig district, Northeastern Egypt. The study involved five hundred cases. They were all subjected to faecal examination using direct smear measure and two commercial faecal concentrators: Mini-Parasep® solvent-free and Mini-FLOTAC procedures. Mini-FLOTAC was performed with two solutions (FS2: saturated sodium chloride and FS7: zinc sulphate). The overall prevalence of intestinal parasitic infections was 56%. Different species were identified, like Giardia lamblia (12.6%), Entamoeba histolytica/dispar (10%), Ascaris lumbricoides (8.8%) and Hymenolepis nana (8.6%). Data analyses revealed a significant association of intestinal parasitism with different socio-demographic features of the participants. Our results showed a better diagnostic performance of Mini-Parasep® in the overall recovery of intestinal parasites. It was more accurate than Mini-FLOTAC in diagnosing both helminths and protozoan infections. Mini-FLOTAC (FS2) exhibited a higher sensitivity than FS7 for helminth recovery (74.6% vs 53.4%), while FS7 was more sensitive for protozoan infections (50.6% vs 43.8%). Intestinal parasitosis remains a challenging health problem in Zagazig city, wherever reliable diagnostic approaches are limited. Thus, our study has proposed the value of the commercial concentrators (Mini-Parasep® and Mini-FLOTAC) as alternative techniques for diagnosing a large variety of parasite species in resource-constrained settings.

As our inventory of Earth's biodiversity progresses, the number of species given a Latin binomial name is also growing. While the coining of species names is bound by rules, the sources of inspiration used by taxonomists are an eclectic mix. We investigated naming trends for nearly 2900 new species of parasitic helminths described in the past two decades. Our analysis indicates that the likelihood of new species being given names that convey some information about them (name derived from morphology, host or locality of origin) or not (named after an eminent scientist, or for something else) depends on the higher taxonomic group to which the parasite or its host belongs. We also found a consistent gender bias among species named after eminent scientists, with male scientists being immortalized disproportionately more frequently than female scientists. Finally, we found that the tendency for taxonomists to name new species after a family member or close friend has increased over the past 20 years. We end by offering suggestions for future species naming, aimed at honouring the scientific community's diversity and avoiding etymological nepotism and cronyism, while still allowing for creativity in crafting new Latin species names.

The prevalence of soil-transmitted helminths (STH) is high in communities within the Peruvian Amazon despite repeated mass-drug administration, demanding alternative strategies of control. Smartphone-attached microscopy (SAM) permits visualization of STH from a small portable microscope through a smartphone screen, potentially providing an inexpensive and rapid method of STH visualization in communities where diagnostic laboratories with microscopes are inaccessible. In this study, a total of 45 community health workers who work within the health systems of Loreto, Peru, attended a 1-day training session with lectures and practicums on STH and SAM. Participants received a pre- and post-intervention questionnaire. Post-intervention, participants were significantly more confident using SAM and identifying parasite images, symptoms, transmission, and treatment (P ≤ 0.0045). Post-intervention, participants correctly labeled a median of five of seven SAM apparatus components and five of eight steps of Kato–Katz technique, were less likely to choose taking medicine to prevent parasite infection (P = 0.0075), and were more likely to select Kato–Katz technique as a type of diagnostic test (P < 0.0001). Most participants felt ready to use SAM in their communities and stated that it could help rural communities far from health centers or laboratories (24%); provide faster identification, results, diagnosis (19%); permit at-home or on-the-spot visualization (14%); and save money (14%). Results show that community health workers show a high level of willingness and competency to learn about both STH and SAM and may be a yet-unexplored practical method of augmenting STH visualization, bringing healthcare to communities in Loreto with poor access to diagnostic laboratories and clinics.

Helminths secrete a plethora of proteins involved in parasitism-related processes such as tissue penetration, migration, feeding and immunoregulation. Astacins, a family of zinc metalloproteases belonging to the peptidase family M12, are one of the most abundantly represented protein families in the secretomes of helminths. Despite their involvement in virulence, very few studies have addressed the role of this loosely defined protein group in parasitic helminths. Herein, we have analysed the predicted proteomes from 154 helminth species and confirmed the expansion of the astacin family in several nematode taxa. The astacin domain associated with up to 110 other domains into 145 unique domain architectures, where CUB and ShK constitute the principal and nearly independent bi-domain frameworks. The presence of co-existing domains suggests promiscuous adaptable functions to several roles. These activities could be related either to substrate specificity or to higher-order functions, such as anti-angiogenesis and immunomodulation, where the astacin domain would play an accessory role. Furthermore, some phylogenetically restricted mutations in the astacin domain affected residues located at the active cleft and binding sub-pockets, suggesting adaptation to different substrate specificities. Altogether, these findings suggest the astacin domain is a highly adaptable module that fulfills multiple proteolytic needs of the parasitic lifestyle. This study contributes to the understanding of helminth-secreted astacins and, ultimately, provides the foundation to guide future investigations about the role of this diverse family of proteins in host-parasite interactions.

A 20-year-old female resident of Beijing intended to consume the eggs of the parasitic worm, Taenia saginata, for weight loss; however, she apparently inadvertently ingested Taenia solium (pork tapeworm) eggs, which resulted in disseminated cysticercosis. Cysticerci developed in the brain, tongue, muscles, liver, peritoneum, and subcutaneous tissues. She was administered oral albendazole and praziquantel. After four 10-day courses of treatment, most of the cysts disappeared and she recovered. After 3 years, the patient remains in good health.

Intestinal nematode infections caused by soil-transmitted helminths (STH), such as the roundworm Ascaris lumbricoides, the whipworm Trichuris trichiura, and the hookworms Ancylostoma duodenale, and Necator americanus, infect more than 1 billion people throughout the world. School-aged children tend to harbor the greatest numbers of intestinal worms, and as a result, experience more adverse health consequences, such as poor growth, anemia, and cognitive decline. Clinicians should maintain a high degree of suspicion in endemic areas when patients present with surgical abdomens, particularly children. Current antihelminthic drugs are moderately effective, but reinfection is possible. Global efforts are needed to eradicate STH infections.

Host manipulation by parasites can shape host behaviour, community structure, and the flow of energy through food webs. A well-known example of host manipulation comes from hairworms (phylum Nematomorpha), which somehow cause their terrestrial insect definitive hosts to enter water, a phenomenon that has received lots of attention in recent years. However, little focus has been directed towards the interactions between hairworms and their aquatic insect hosts and the return of dormant hairworms from water to land. Here, we ask whether hairworm cyst infections impact, either directly or indirectly, the life history of their aquatic transport hosts. By observing the development of last-instar Olinga jeanae (Trichoptera: Conoesucidae) caddisfly larvae naturally infected with Gordius-type hairworm cysts under controlled conditions, we found that higher numbers of cysts per infected caddisfly correlated with a decrease in time to pupation. These new observations suggest that, apart from the striking host manipulation that brings the parasite from land to water, the presence of dormant hairworms is associated with changes in the development of their aquatic hosts, either through direct or indirect mechanisms, which may accelerate their transition from water to land.

Maternal infection during pregnancy is known to alter the development and function of offspring’s immune system, leading to inappropriate immune responses to common childhood infections and immunizations. Although this is an expanding field, maternal parasitic infections remain understudied. Millions of women of reproductive age are currently at risk for parasitic infection, whereas many pregnant, chronically infected women are excluded from mass drug administration due partially to a lack of resources, as well as fear of unknown adverse fetal developmental outcomes. In areas endemic for multiple parasitic infections, such as sub-Saharan Africa, there are increased rates of morbidity and mortality for various infections during early childhood in comparison with nonendemic areas. Despite evidence supporting similar immunomodulatory effects between various parasite species, there is no clear mechanistic understanding of how maternal infection reprograms offspring immunity. This brief review will compare the effects of selected maternal parasitic infections on offspring immunity.

Parasites are often distributed heterogeneously across host populations, but the controls of this heterogeneity across regional scales often remain unclear. Here, we test the relative importance of natural and anthropogenic environmental factors and biological attributes of host populations on the large-scale variability in infection probability and parasite abundance. We quantified a larval acanthocephalan parasite (Profilicollis altmani), as well as the trematodeMaritremasp. and the nematodeProleptussp., infection of mole crabsEmerita analogausing a hierarchical design at 8 sites spanning 500 km of the south-central shoreline of Chile. At each site, we measured the variables of beach size, morphodynamics, distance to nearest port, distance to tributaries, mole crab density, seagull density, and host body size. Using mixed-effects models, we analysed the associations of acanthocephalan infection probability and parasite abundance in mole crabs as functions of body size and the environmental site-level factors. Models accounted for 20 and 61% of variation in acanthocephalan infection probability and abundance, respectively, with mole crab body size exclusively accounting for nearly all of the model fits. Also, acanthocephalan abundance decreased with increasing mole crab density, suggesting a possible encounter-dilution effect. Co-infection was strong, with the 2 other larval parasite species correlated positively with acanthocephalan parasites. The low influence of environmental variables on acanthocephalan infection could be because spatial variability in these factors is relatively low or their influences are quick to saturate. Therefore, in this system, parasite infection is apparently more strongly related to the duration of host exposure than spatially variable environmental factors, even across large spatial scales.

Over millions of years, helminths have shared their host habitat with a myriad of other microorganisms, collectively known as the ‘microbiota’. Of the microbial communities that inhabit host organs and tissues, those residing in the gastrointestinal tract are known to play active roles in several key biological processes, including but not limited to, nutrient processing and absorption, defence against pathogens and, crucially, development and maintenance of innate and adaptive immunity. Over the past decade, the tight association between parasitic worms and the gut microbiota has attracted significant interest, as the concept of ‘host-parasite interactions’ has progressively shifted towards acknowledging the former as ‘holobiont’, i.e. the host in association with its associated mutualistic symbionts. Thus, a deep knowledge of worm-holobiont interactions is essential to better understand worm biology and mechanisms of helminth disease, and ultimately to identify parasite ‘Achille’s heels’ that may represent useful targets of helminth control.

Gastrointestinal parasitism by helminths and protozoa poses risks to animal and human health owing to clinical changes and transmission of potentially zoonotic agents. Thus, the present study aimed to verify the occurrence of gastrointestinal parasites in dogs from the municipality of Cuiabá, Mato Grosso, Brazil, using coproparasitological tests. From June 2021 to April 2022, faecal samples were collected from domestic dogs at the Veterinary Hospital and Animal Protection Shelters in the Cuiabá municipality. A semi-structured questionnaire was applied to the owners and those responsible for the shelters to analyse the factors associated with gastrointestinal parasitism. A total of 353 faecal samples were collected and subjected to parasitological flotation and sedimentation techniques. Data were analysed using the chi-squared test and exploratory factorial analysis. The occurrence of gastrointestinal parasitism was 22.66% and the parasites found alone or in mixed infections were Ancylostoma spp., Trichuris vulpis, Toxocara spp., Dipylidium caninum, Cystoisospora spp., Giardia duodenalis, and coccidia. It was concluded that the occurrence of gastrointestinal parasites in dogs is frequent, and the variables associated with these infections were source origin, breed, age, coexistence with other animals, and dull fur. Resumo O parasitismo gastrointestinal por helmintos e protozoários apresenta riscos à saúde animal e humana devido às alterações clínicas e transmissão de agentes potencialmente zoonóticos. Assim, o presente estudo teve como objetivo verificar a ocorrência de parasitoses gastrointestinais em cães do município de Cuiabá, Mato Grosso, Brasil, utilizando-se exames coproparasitológicos. De junho de 2021 a abril de 2022, foram coletadas amostras fecais de cães domésticos no Hospital Veterinário e Abrigos de Proteção Animal do município de Cuiabá. Foi aplicado um questionário semiestruturado aos tutores e responsáveis pelos abrigos para analisar os fatores associados ao parasitismo gastrointestinal. Um total de 353 amostras fecais foram coletadas e submetidas às técnicas de flotação e sedimentação parasitológica. Os dados foram analisados, utilizando-se o teste Qui-quadrado e a análise fatorial exploratória. A ocorrência de parasitismo gastrointestinal foi de 22,66%, e os parasitos encontrados isoladamente ou em infecções mistas foram Ancylostoma spp., Trichuris vulpis, Toxocara spp., Dipylidium caninum, Cystoisospora spp., Giardia duodenalis e Coccidia. Conclui-se que a ocorrência de parasitoses gastrointestinais em cães é frequente, e as variáveis associadas a essas infecções foram procedência, raça, idade, convivência com outros animais e pelagem opaca.

The sex ratio (ratio of boys to girls) at birth (SRB) is about 1.05 (about 105 males to 100 females) under normal conditions and could be an indicator for monitoring demographic changes. Generally, in comparison with female fetuses, the male fetus is biologically weaker and more vulnerable to prenatal events. SRB is influenced by various factors, such as maternal malnourishment. Remarkably, maternal malnourishment and less energy intake are pivotal factors for declining SRB in humans. As estimates, the SRB is below the normal range in sub-Saharan Africa (1.03) than the normal range (1.05). On the other hand, both malaria and helminth infections are hyperendemic in sub-Saharan Africa and both diseases are associated with maternal malnourishment. Hence, an important question arises, could declining SRB in sub-Saharan Africa be influenced by malaria and helminth infections? Cumulative evidence suggests that malaria and helminth infections could influence SRB by induction of maternal malnourishment. This hypothesis provides new ideas about the variation of SRB in some regions of the world where helminths and malaria are endemic.

Long non-coding RNAs (lncRNAs) emerged in the past 20 years due to massive amounts of scientific data regarding transcriptomic analyses. They have been implicated in a plethora of cellular processes in higher eukaryotes. However, little is known about lncRNA possible involvement in parasitic diseases, with most studies only detecting their presence in parasites of human medical importance. Here, we review the progress on lncRNA studies and their functions in protozoans and helminths. In addition, we show an example of knockdown of one lncRNA in Schistosoma mansoni, SmLINC156349, which led to in vitro parasite adhesion, motility, and pairing impairment, with a 20% decrease in parasite viability and 33% reduction in female oviposition. Other observed phenotypes were a decrease in the proliferation rate of both male and female worms and their gonads, and reduced female lipid and vitelline droplets that are markers for well-developed vitellaria. Impairment of female worms' vitellaria in SmLINC156349-silenced worms led to egg development deficiency. All those results demonstrate the great potential of the tools and methods to characterize lncRNAs as potential new therapeutic targets. Further, we discuss the challenges and limitations of current methods for studying lncRNAs in parasites and possible solutions to overcome them, and we highlight the future directions of this exciting field.

Helminths are complex multicellular metazoan parasites. Approximately 300 species can infect humans, causing a wide array of pathology and disability. They also have a significant socioeconomic impact globally. Although mortality rates are generally low, the burden of disease they cause is considerable. They are often overlooked in temperate regions where the overall prevalence is low. However, they represent an important differential diagnosis in migrants and returning travellers, and should be considered in unexplained peripheral blood eosinophilia. Helminths can present a diagnostic challenge, but prompt identification usually leads to successful treatment with good outcomes.

The major soil-transmitted helminths that infect humans are the roundworms, whipworms and hookworms. Soil-transmitted helminth infections rank among the most important neglected tropical diseases in terms of morbidity, and almost one billion people are still infected with at least one species. While anthelmintic drugs are available, they do not offer long term protection against reinfection, precipitating the need for vaccines that provide long-term immunologic defense. Vaccine discovery and development is in advanced clinical development for hookworm infection, with a bivalent human hookworm vaccine in clinical trials in Brazil and Africa, but is in its infancy for both roundworm (ascariasis) and whipworm (trichuriasis) infections. One of the greatest hurdles to developing soil-transmitted helminth vaccines is the potent immunoregulatory properties of these helminths, creating a barrier to the induction of meaningful long-term protective immunity. While challenging for vaccinologists, this phenomenon presents unique opportunities to develop an entirely new class of anti-inflammatory drugs that capitalise on these immunomodulatory strategies. Epidemiologic studies and clinical trials employing experimental soil-transmitted helminth challenge models, when coupled with findings from animal models, show that at least some soil-transmitted helminth-derived molecules can protect against the onset of autoimmune, allergic and metabolic disorders, and several natural products with the desired bioactivity have been isolated and tested in pre-clinical settings. The yin and yang of soil-transmitted helminth infections reflect both the urgency for effective vaccines and the potential for new immunoregulatory molecules from parasite products.

This checklist of parasites of Kerodon rupestris, an endemic rodent from the Brazilian semiarid region, revealed records of 25 enteroparasite taxa comprising Cestoda (Anoplocephalidae), Trematoda, Acanthocephala and Nematoda (Ancylostomidae, Ascarididae, Heterakidae, Oxyuridae, Pharyngodonidae, Trichuridae, Capillariidae, Trichostrongylidae, and Strongyloididae), and two taxa of coccidian. Paleoparasitological and parasitological studies published until 2019 were assessed in the present study and locality information, site of infection, sample dating, and host data were summarized from each reference. Analyses of recent feces and coprolites revealed the highest species richness in the Piauí State. The chronological data corroborated that Trichuris spp. and oxyurids are part of the parasitic fauna of K. rupestris. This represents the first checklist of intestinal parasites from K. rupestris. Resumo Este levantamento de parasitos intestinais de Kerodon rupestris, um roedor endêmico da região semiárida brasileira, revelou registros de 25 táxons de enteroparasitos compreendendo Cestoda (Anoplocephalidae), Trematoda, Acanthocephala e Nematoda (Ancylostomidae, Ascarididae, Heterakidae, Oxyuridae, Pharyngodonidae, Trichuridae, Capillariidae, Trichostrongylidae, and Strongyloididae), e dois táxons de coccídios. Os estudos paleoparasitológicos e parasitológicos publicados até 2019 foram avaliados no presente trabalho e as informações de localização, local da infecção, datação da amostra e dados do hospedeiro, foram resumidas a partir de cada referência. Análises de fezes recentes e coprólitos revelaram a maior riqueza de espécies no estado do Piauí. Os dados cronológicos corroboram que Trichuris spp. e oxiurídeos fazem parte da fauna parasitária de K. rupestris. Essa é a primeira lista de verificação de parasitos intestinais de K. rupestris.

Anthelmintic resistance (AR) has thus far only rarely been reported for intestinal helminths of dogs and cats, in contrast to parasites of livestock and horses. We highlight possible reasons for this striking and important discrepancy, including ecological, biological and genetic factors and/or intervention regimens of key intestinal helminths concerning both host groups. In view of the current knowledge related to the genetics, mechanisms and principles of AR development, we point at issues which in our view contribute to a comparatively lower risk of AR development in intestinal helminths of dogs and cats. Finally, we specify research needs and provide recommendations by which, based on the available information about AR in ruminant and equine helminths, the development of AR in dog and cat helminths may best be documented, prevented or at least postponed., Graphical abstract Image 1, Highlights • Identifying causes of differences in anthelmintic resistance between livestock and dogs & cats. • Discussion of roles of AR mechanisms, genetics and husbandry in delayed AR evolution in dogs & cats. • Research needs for improved tools to monitor AR development in dogs & cats.

The article provides data on the species composition of alien helminth species, which infest introduced fish species in pond fish farms, and also the literature data on natural reservoirs of Belarus. Seven alien helminth species have been revealed in introduced fish in reservoirs of Belarus ( Khawia sinensis Hsü, 1935, Schyzocotyle acheilognathi (Yamaguti, 1934) , Caryophyllaeus fimbriceps Annenkova-Chlopina, 1919, Philometroides cyprini (Ishii, 1931), Dactylogyrus dulkeiti Bychowsky, 1936, Bothriocephalus claviceps (Goeze, 1782) , Anguillicola crassus Kuwahara 1974). All species of helminths excluding the nematode A.crassus and the cestode B.claviceps - parasites of the eels Anguilla anguilla , were introduced to Belarus with the seeding of Cyprinus carpio haematopterus (Linnaeus, 1758) from the Far East. The invasive species of helminths ( K. sinensis, S. acheilognathi, P. cyprini, A. crassus ), which affect fishery by causing the death of farmed fish, are singled out.

A study was carried out in Kamuli and Hoima districts in Eastern and Western regions of Uganda to determine the Taenia solium porcine cysticercosis (PCC) and gastrointestinal (GI) parasites co-infection status in pigs. One hundred sixty-one households were selected randomly and visited between November and December 2019. A household questionnaire was administered, and faecal and blood samples were collected from at least one pig older than 3 months per household. A blood sample was obtained from a jugular venipuncture, and a rectal faecal sample was obtained. Taenia spp. circulating antigen levels in the sample sera were tested using a commercial enzyme-linked immunosorbent assay kit, apDia™ cysticercosis Ag ELISA. The modified McMaster technique was used to identify and quantify the GI parasites. The apparent animal-level seroprevalence for PCC was 4.8% (95% CI 2.7–7.1) and differed across the two districts (p = 0.018). At the pig herd level, the prevalence was 9.7% (95% CI 5.5–14.4). The prevalence of the different nematode eggs and coccidian oocysts in the two districts was as follows: strongyles 79.0% (95% CI 74.3–83.6), coccidia 73.3% (95% CI 68.3–78.6), Trichuris spp. 7.4% (95% CI 4.9–10.6), Strongyloides ransomi 2.1 (95% CI 0.7–3.5) and Ascaris spp. 4.9 (95% CI 2.8–7.4). Overall, across the two districts, the arithmetic mean for the oocysts per gram (OPG) for coccidia was 2042.2 ± 5776.1, and eggs per gram (EPG) were the highest in strongyles 616.1 ± 991. Overall, 57.4% of the porcine cysticercosis seropositive pigs were also positive for at least one of the gastrointestinal helminths which included strongyles, Strongyloides ransomi, Trichuris spp. and Ascaris spp. The co-infection status of pigs with both PCC and GI parasites demonstrated by this study can provide an incentive for integrating the control and management of both parasites with oxfendazole. Further studies are required to understand the feasibility of using oxfendazole including cost–benefit analysis and the acceptability by local stakeholders for the control of T. solium cysticercosis and gastrointestinal parasites in pigs.

Maintenance of the life cycle of a wide diversity of helminths depends on the aquatic environment. Freshwater turtles are considered one of the main groups in these ecosystems due to their high density and biomass. However, there is little information on the infection parameters of helminths associated with the Chelidae species Hydromedusa tectifera, Phrynops hilarii, and Acanthochelys spixii that occur in the Pampa biome. Therefore, this study aimed at analyzing the gastrointestinal helminth assemblage of the three species. Analyses showed that A. spixii was associated with 11 helminth taxa while P. hilarii and H. tectifera were parasitized by eight taxa each. The gastrointestinal helminth assemblage of A. spixii and H. tectifera showed high similarity by the Morisita index. Infections suggest that the helminth fauna is directly related to the diet of the hosts, since A. spixii, P. hilarii, and H. tectifera are generalist and/or opportunistic with diets composed of various invertebrates, which, in turn, are part of the life cycles as intermediate hosts of most of the identified helminths. Thus, this study highlights not only the importance of helminthological research for the understanding of the biology of host organisms, but also the need for conservation of flooded areas characteristic of the Pampa biome, which is essential to maintain the diversity of the ecosystem.

Fasciolosis caused by the liver flukes Fasciola hepatica and Fasciola gigantica is one of the most important neglected parasitic diseases of humans and animals. The ability of the parasites to infect and multiply in their intermediate snail hosts, and their adaptation to a wide variety of mammalian definitive hosts contribute to their high transmissibility and distribution. Within the mammalian host, the trauma caused by the immature flukes burrowing through the liver parenchyma is associated with most of the pathogenesis. Similarly, the feeding activity and the physical presence of large flukes in the bile ducts can lead to anaemia, inflammation, obstruction and cholangitis. The high frequency of non-synonymous polymorphisms found in Fasciola spp. genes allows for adaptation and invasion of a broad range of hosts. This is also facilitated by parasite's excretory-secretory (ES) molecules that mediate physiological changes that allows their establishment within the host. ES contains cathepsin peptidases that aid parasite invasion by degrading collagen and fibronectin. In the bile ducts, cathepsin-L is critical to haemoglobin digestion during feeding activities. Other molecules (peroxiredoxin, cathepsin-L and Kunitz-type inhibitor) stimulate a strong immune response polarised towards a Treg/Th2 phenotype that favours fluke's survival. Helminth defence molecule, fatty acid binding proteins, Fasciola-specific glycans and miRNAs modulate host pro-inflammatory responses, while antioxidant scavenger enzymes work in an orchestrated way to deter host oxidant-mediated damage. Combining these strategies Fasciola spp. survive for decades within their mammalian host, where they reproduce and spread to become one of the most widespread zoonotic worm parasites in the world.

Soil Transmitted Helminths (STH) are intestinal nematode worms that infect humans who ingest their eggs via the faecal-oral route. Soil Transmitted Helminths (STH) infection is transmitted through eggs and larvae found in human feces which then contaminate the soil in environmental areas that have poor sanitation. The impact caused by worms includes malnutrition, growth and development disorders and cognitive disorders in children, and in adults it can reduce work productivity. The incidence of worms is found mainly in people living in rural areas, especially farmers. STH transmission to vegetable farmers occurs through water and sludge used in vegetable cultivation. STH prevalence is supported by suitable natural conditions, personal hygiene and low environmental sanitation, especially in the vegetable farming environment. This study aims to determine the relationship between personal hygiene and intestinal worm egg infection (Soil Transmitted Helmint) vegetable farmers in Gianyar Regency. The type of research used in this study is a descriptive correlative study with a cross sectional study approach which aims to determine the relationship between personal hygiene and intestinal worm infection (Soil Transmitted Helmint) in vegetable farmers in Gianyar Regency. The results showed that the personal hygiene condition of vegetable farmers in Gianyar Regency was poor (83.3%) and the incidence of worm infection in vegetable farmers in Gianyar Regency was 20%. There is no relationship between personal hygiene personal hygiene with intestinal worm egg infections (Soil Transmitted Helmint) in vegetable farmers in Gianyar Regency (p-value> 0.05).

Ecologoparasitological research was done based on the helminths and helminth communities of the Prussian carp (Carassius gibelio (Bloch, 1782)) from the freshwater ecosystem of the Panicheri Reservoir, Aegean Water Basin, Bulgaria. As a result of the examined nine specimens of the Prussian carp, two species of helminths were found: Ligula intestinalis (Linnaeus, 1758), larvae and Pomphorhynchus laevis (Müller, 1776). The dominant structure of the helminth communities was determined. The Panicheri Reservoir is a new habitat in Bulgaria of P. laevis and L. intestinalis from C. gibelio. C. gibelio is a new host record for L. intestinalis in Bulgaria. The two helminth species are core species for the helminth communities of the examined species of the freshwater fish. P. laevis was distinguished with higher prevalence and mean intensity (P%=33.34; MI=1.34) than L. intestinalis (P%=22.23; MI=1.0). The circulatory pathways of the helminth flow were traced.
