This study aimed at examining the structure-role modeling and antifatigue mechanism of polysaccharides, including M. androsaceus exopolysaccharide 3 (MEPS3), isolated from Marasmius androsaceus fermentation broth. The molecular weight of MEPS3 was 10.47 kDa. Furthermore, monosaccharide analysis showed the presence of mannose, glucose, and galactose in MEPS3 in a molar ratio of 0.08 : 0.34 : 1.46. Mannose, α-galactose, and α-d-glucose anomeric hydrogen signals were detected using nuclear magnetic resonance spectroscopy. MEPS3 was found to contain glyoxylic acid, forming rod-like chains that support high-purity polymerization. The weight-loaded swimming test results showed that MEPS3 treatment reduced lactic acid (LA) levels by 25.72% and increased the lactate dehydrogenase (LDH) activity by 5.67% in the plasma. Furthermore, it lowered malondialdehyde (MDA) levels by 47.09% and increased reactive oxygen species (ROS) and glutathione peroxidase (GSH-Px) levels by 52.42 and 97.03%, respectively, in the plasma. In addition, MEPS3 treatment reduced MDA and ROS levels in the liver by 28.85 and 18.64% while increasing superoxide dismutase (SOD) and GSH-Px levels by 17.41 and 38.13%, respectively. MEPS3 treatment increased the expression of nuclear factor-erythroid 2-related factor 2, glutamate-cysteine ligase, quinone oxidoreductase 1, and heme oxygenase 1 by 22.5, 24.8, 20.3, and 43.1%, respectively, in the liver. These findings demonstrate that MEPS3 effectively alleviates fatigue by removing harmful metabolites and indicate that the antifatigue mechanism is related to the Nrf-2 signaling pathway. Environmental risk of heavy metals and metalloids in athletic fields has raised people's attention in the recent years. Seven trace elements, including metals and metalloids, were detected in the runoff of five typical athletic fields in the university campus under three rainfall events. Except for Cr, the total concentrations of Zn, Pb, Cu, Mn, Cd, and As in artificial turf runoff are the highest among five athletic fields, followed by that of plastic runway. The concentration and first flush effect of trace elements are followed in the order of 1st > 2nd > 3rd rainfall events. The strongest correlations between metals and metalloids were observed in the tennis court runoff, while the artificial turf shows the least. The release of trace elements could be directly from the surface materials and particles on the athletic field and influenced by the comprehensive factors including surface materials, rainfall events, and pollutant characters. Pollution risk assessment shows that the pollution extent of the five types of athletic field is at least "moderate" and follows the order of artificial turf > basketball court > plastic runway > badminton court > tennis court. Pb shows the highest pollution level, while Cr shows the highest healthy risk. The results can provide a theoretical basis for runoff pollution control and safety use of athletic fields. Danhong Huayu Koufuye (DHK), a traditional Chinese prescription, is used to treat central retinal vein occlusion clinically. We previously reported that DHK prevented diabetic retinopathy (DR) in rats. Moreover, we found that it protected endothelial cells from hyperglycemia-induced apoptosis through antioxidation and anti-inflammation. Here, we investigated whether antioxidative and anti-inflammatory activities of DHK contributed to its therapeutic effect on DR in streptozotocin- (STZ-) induced diabetic rats. DHK significantly blocked the breakdown of the blood-retinal barrier (BRB) and increased the thickness of the inner nuclear layer (INL), as well as suppressed the swelling of the ganglion cell layer (GCL) in diabetic retinas. DHK remarkably increased the activities of superoxide dismutase (SOD) and glutathione peroxidase (GPx) in plasma, and decreased serum level of nitric oxide (NO). Moreover, DHK markedly reduced the serum levels of vascular endothelial growth factor (VEGF) and intercellular adhesion molecule-1 (ICAM-1). Furthermore, DHK significantly downregulated protein expressions of VEGF and inducible NO synthase (iNOS) and mRNA expression of ICAM-1 in retinas. These results suggest that the antioxidative and anti-inflammatory activities of DHK may be important mechanisms involved in the protective effect of DHK on DR in STZ-induced diabetic rats. On the basis of Internet of Things (IoT) technologies, Community Medical Internet of Things (CMIoT) is a new medical information system and generates massive multiple types of medical data which contain all kinds of user identity data, various types of medical data, and other sensitive information. To effectively protect users’ privacy, we propose a secure privacy data protection scheme including transmission protection and access control. For the uplink transmission data protection, bidirectional identity authentication and fragmented multipath data transmission are used, and for the downlink data protection, fine grained access control and dynamic authorization are used. Through theoretical analysis and experiment evaluation, it is proved that the community medical data can be effectively protected in the transmission and access process without high performance loss. To identify the distribution of heavy metals in sewer sediments and assess their potential harmfulness to the environment and human health, the occurrence of Pb, Zn, Cu, and Cd in the sewer sediment of six functional areas and two streets in an inner-city suburb of Beijing, China, was investigated by using a sequential extraction procedure. Results show that the concentrations of Cu, Zn, Cd, and Pb vary between 50 and 175, between 80 and 180, between 0.75 and 2.5, and between 20 and 110 mg/kg, respectively, and Fe-Mn oxide fraction is significant for all metals in sampling areas. Pollution assessment shows that 1–2% of Cu at Chegongzhuang Street and 1–3% of Zn at Fuwai Street in the exchangeable fractions are of low risk. 10–25% of Cd at six functional areas indicates medium risk. 40–60% of Pb at Fuwai Street existing in the exchangeable fractions is of high to very high risk. The sum of these metals associated with exchangeable, carbonate bound, and Fe-Mn oxide fractions is quite high; however, these three fractions represent the proportion of heavy metals that can be remobilized by changes in environmental conditions. Arteriogenesis is an inflammatory process associated with rapid cellular changes involving vascular resident endothelial progenitor cells (VR-EPCs). Extracellular cell surface bound 20S proteasome has been implicated to play an important role in inflammatory processes. In our search for antigens initially regulated during collateral growth mAb CTA 157-2 was generated against membrane fractions of growing collateral vessels. CTA 157-2 stained endothelium of growing collateral vessels and the cell surface of VR-EPCs. CTA 157-2 bound a protein complex (760 kDa) that was identified as 26 kDa α7 and 21 kDa β3 subunit of 20S proteasome in mass spectrometry. Furthermore we demonstrated specific staining of 20S proteasome after immunoprecipitation of VR-EPC membrane extract with CTA 157-2 sepharose beads. Functionally, CTA 157-2 enhanced concentration dependently AMC (7-amino-4-methylcoumarin) cleavage from LLVY (N-Succinyl-Leu-Leu-Val-Tyr) by recombinant 20S proteasome as well as proteasomal activity in VR-EPC extracts. Proliferation of VR-EPCs (BrdU incorporation) was reduced by CTA 157-2. Infusion of the antibody into the collateral circulation reduced number of collateral arteries, collateral proliferation, and collateral conductance in vivo. In conclusion our results indicate that extracellular cell surface bound 20S proteasome influences VR-EPC function in vitro and collateral growth in vivo. [ABSTRACT FROM AUTHOR]
