To investigate the residual stress relaxation behaviors and fatigue strength, EA4T steel specimens are subjected to micro-shot peening (MSP), and part of them are isothermally annealed at 250 °C for 2 hours. Fatigue experiments are conducted on a four-point rotating bending machine under constant and variable loads. Surface compressive residual stress (CRS) and full width at half maximum (FWHM) before the experiment and during fatigue loading are analyzed using the x-ray diffraction method. The results indicate that the CRS of the MSP specimens are relaxed after the isothermal annealing, leading to a decrease in their fatigue strengths. Variable loading slightly increased the fatigue life of the un-peened specimen, but significantly reduced that of the MSP specimens. The effect of CRS relaxation on fatigue strength is analyzed, and the relationship between CRS and FWHM is also discussed. [ABSTRACT FROM AUTHOR]

Purpose: Study aims to investigate the consistency of delayed enhancement cardiac magnetic resonance imaging (DE-CMR) and 18F-FDG PET myocardial imaging in evaluating myocardial viability before CABG. Methods: The study analyzed data from 100 patients who were examined with DE-CMR, PET imaging, and echocardiography before and after CABG. All subjects were followed up for 6–12 month post- CABG. Results: DE-CMR and PET imaging have high consistency (90.1%; Kappa value = 0.71, p < 0.01) in determining myocardial viability. The degree of delayed enhancement was negatively correlated with the improvement in myocardial contractile function in this segment after revascularization (P < 0.001). The ratio of scarred myocardial segments and total DE score was significantly lower in the improvement group than non-improvement group. Multivariate regression identified that hibernating myocardium (OR = 1.229, 95%CI: 1.053–1.433, p = 0.009) was influencing factor of LVEF improvement after CABG. Conclusion: Both imaging techniques are consistent in evaluating myocardial viability. To investigate the thermal stress field and microstructure evolution of Ti-6Al-4V alloy component produced by laser engineered net shaping (LENS), numerical simulations are performed at both macroscale and mesoscale. At the macroscale, a finite element model is developed and validated by comparing the simulated residual stress with the measured residual stress. Meanwhile, the effect of scanning strategies on residual stress is analyzed. At the mesoscale, a three-dimensional (3D) cellular automaton model is established to analyze the growth mechanism of β-columnar grains. The results show that the maximum residual stress locates at the surrounding area at the interface between the substrate and the cladding layer. Based on the results under the unidirectional and reciprocating scanning strategies, the residual stress of the component is minimized by using the reciprocating alternating scanning strategy. Due to the higher temperature gradient at the bottom of the molten pool, β-columnar grains grow epitaxially from the pre-existing grains in the substrate. When the direction of dendritic growth is consistent with the direction of the temperature gradient, grain growth is faster and there is a clear preferred orientation, which ultimately forms β-columnar grain structures that tilt towards the scanning direction and penetrate multiple cladding layers. In this work, the influence of Si concentration on the microstructure and growth of plasma electrolytic oxidation (PEO) coatings on two types of Al–Si–Mg aluminum alloys were investigated. we utilized a multiscale correlative methodology to analyze the microstructure and micro-defect characteristics of the PEO coatings formed on A357 and 6082 aluminum alloys. The results demonstrate that both coatings exhibit a porous structure and are primarily composed of alumina and amorphous phases. Notably, the PEO coating formed on A357 alloy exhibits lower thickness, higher porosity, and more non-uniform barrier layer compared to that on 6082 alloy due to a higher concentration of Si in the substrate. Advanced X-ray computed tomography effectively characterizes micro-defects with different morphologies within the two coatings. Different maize varieties respond differentially to cadmium (Cd) stress. As the first organ in contact with the soil, the response of the root is particularly important. However, the physiological mechanisms that determine the response are not well defined. Here, we compared the differences in Cd-induced related gene expression, ionic homeostasis, and ultrastructural changes in roots of Cd-tolerant maize variety (XR57) and Cd-sensitive maize variety (LY296), and assessed their effects on Cd uptake and accumulation. Our findings indicate that XR57 absorbed a significantly lower Cd than LY296 did, and that the expression levels of genes related to Cd uptake (ZmNRAMP5 and ZmZIP4) and efflux (ZmABCG4) in the root were consistent with the Cd absorption at the physiological levels. Compared with LY296, the lower Cd concentration in the roots of XR57 caused less interference with the ion balance. Transmission electron microscope images revealed that the roots from XR57 exposed to Cd had developed thicker cell walls than LY296. In addition, the large increase ZmABCC1 and ZmABCC2 expression levels in XR57 mediated the appearance of numerous electron-dense granules in the vacuoles present in the roots. As a result, the high Cd tolerance of XR57 is the result of a multi-level response that involves increased resistance to Cd uptake, a stronger capacity for vacuolar regionalization, and the formation of thicker cell walls. Purpose: The developmental status of the male and female spike determines grain number (GN) per ear and yield. A better understanding of the effect of N on male and female spike differentiation and GN formation could help to gain insight into the mechanism of N regulation on yield. Methods: A field experiment was conducted with four nitrogen rates: 0 (N0), 129 (N1), 185 (N2), and 300 kg N ha−1 (N3) with a single summer maize hybrid to determine tassel and ear development and the resulting GN. Results: The expression of key proteins and genes related to fatty acid metabolism of N0 were down-regulated and the tapetum was abnormally degraded. This inhibited the synthesis of long-chain fatty acids (LCFAs) in the tassel and ear. Both pollen and silk development at the mononucleate stage were abnormal and presented as misshapen pollen, uneven silk tube, reduced silk, and poor pollination. Finally, anthesis-silking interval (ASI) was increased and the length of male and female spikes, fertilized florets, floret fertility percentage, and grain set rate were reduced in N0 treatment resulting in reduced GN. N application promoted fatty acid metabolism but N application greater than 185 kg N ha−1 did not increase LCFA of either male or female spike traits. Conclusions: A nitrogen rate of 185 kg N ha−1 was sufficient to promote lipid metabolism, and increase the content of LCFAs. Introduction: Splice modulators have been assessed clinically in treating haematologic malignancies exhibiting splice factor mutations and acute myeloid leukaemia. However, the mechanisms by which such modulators repress leukaemia remain to be elucidated.Objectives: The primary goal of this assessment was to assess the molecular mechanism by which the natural splice modulator GEX1A kills leukaemic cells in vitro and within in vivo mouse models.Methods: Using human leukaemic cell lines, we assessed the overall sensitivity these cells have to GEX1A via EC50 analysis. We subsequently analysed its effects using in vivo xenograft mouse models and examined whether cell sensitivities were correlated to genetic characteristics or protein expression levels. We also utilised RT-PCR and RNAseq analyses to determine splice change and RNA expression level differences between sensitive and resistant leukaemic cell lines.Results: We found that, in vitro, GEX1A induced an MCL-1 isoform shift to pro-apoptotic MCL-1S in all leukaemic cell types, though sensitivity to GEX1A-induced apoptosis was negatively associated with BCL-xL expression. In BCL-2-expressing leukaemic cells, GEX1A induced BCL-2-dependent apoptosis by converting pro-survival BCL-2 into a cell killer. Thus, GEX1A + selective BCL-xL inhibition induced synergism in killing leukaemic cells, while GEX1A + BCL-2 inhibition showed antagonism in BCL-2-expressing leukaemic cells. In addition, GEX1A sensitised FLT3-ITD+ leukaemic cells to apoptosis by inducing aberrant splicing and repressing the expression of FLT3-ITD. Consistently, in in vivo xenografts, GEX1A killed the bulk of leukaemic cells via apoptosis when combined with BCL-xL inhibition. Furthermore, GEX1A repressed leukaemia development by targeting leukaemia stem cells through inhibiting FASTK mitochondrial isoform expression across sensitive and non-sensitive leukaemia types.Conclusion: Our study suggests that GEX1A is a potent anti-leukaemic agent in combination with BCL-xL inhibitors, which targets leukaemic blasts and leukaemia stem cells through distinct mechanisms. Our understanding of MM genomics has expanded rapidly in the past 5–10 years as a consequence of cytogenetic analyses obtained in routine clinical practice as well as the ability to perform whole-exome/genome sequencing and gene expression profiling on large patient data sets. This knowledge has offered new insights into disease biology and is increasingly defining high-risk genomic patterns. In this manuscript, we present a thorough review of our current knowledge of MM genomics. The epidemiology and biology of chromosomal abnormalities including both copy number abnormalities and chromosomal translocation are described in full with a focus on those most clinically impactful such as 1q amplification and del(17p) as well as certain chromosome 14 translocations. A review of our ever-expanding knowledge of genetic mutations derived from recent whole-genome/exome data sets is then reviewed including those that drive disease pathogenesis from precursor states as well as those that may impact clinical outcomes. We then transition and attempt to elucidate how both chromosomal abnormalities and gene mutations are evolving our understanding of disease risk. We conclude by offering our perspectives moving forward as to how we might apply whole-genome/exome-level data in addition to routine cytogenetic analyses to improve patient outcomes as well as further knowledge gaps that must be addressed. The ten–eleven translocation (TET) family of dioxygenases consists of three members, TET1, TET2, and TET3. All three TET enzymes have Fe+2 and α-ketoglutarate (α-KG)-dependent dioxygenase activities, catalyzing the 1st step of DNA demethylation by converting 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC), and further oxidize 5hmC to 5-formylcytosine (5fC) and 5-carboxylcytosine (5caC). Gene knockout studies demonstrated that all three TET proteins are involved in the regulation of fetal organ generation during embryonic development and normal tissue generation postnatally. TET proteins play such roles by regulating the expression of key differentiation and fate-determining genes via (1) enzymatic activity-dependent DNA methylation of the promoters and enhancers of target genes; and (2) enzymatic activity-independent regulation of histone modification. Interacting partner proteins and post-translational regulatory mechanisms regulate the activities of TET proteins. Mutations and dysregulation of TET proteins are involved in the pathogenesis of human diseases, specifically cancers. Here, we summarize the research on the interaction partners and post-translational modifications of TET proteins. We also discuss the molecular mechanisms by which these partner proteins and modifications regulate TET functioning and target gene expression. Multiple myeloma (MM) is an acquired malignant plasma cell disorder that develops late in life. Although progression free and overall survival has improved across all age, race, and ethnic groups, a subset of patients have suboptimal outcomes and are labeled as having high risk disease. A uniform approach to risk in NDMM remains elusive despite several validated risk stratification systems in clinical use. While we attempt to capture risk at diagnosis, the reality is that many important prognostic characteristics remain ill-defined as some patients relapse early who were defined as low risk based on their genomic profile at diagnosis. It is critical to establish a definition of high risk disease in order to move towards risk-adapted treatment approaches. Defining risk at diagnosis is important to both effectively design future clinical trials and guide which clinical data is needed in routine practice. The goal of this review paper is to summarize and compare the various established risk stratification systems, go beyond the R-ISS and international myeloma working group risk stratifications to evaluate specific molecular and cytogenetic abnormalities and how they impact prognosis independently. In addition, we explore the wealth of new genomic information from recent whole genome/exome sequencing as well as gene expression data and review known clinical factors affecting outcome such as disease burden and early relapse as well as patient related factors such as race. Finally, we provide an outlook on developing a new high risk model system and how we might make sense of co-occurrences, oncogenic dependencies, and mutually exclusive mutations. High temperature superconducting (HTS) maglev transportation has provided a new development direction for high-speed and ultra-high-speed rail transit. In order to promote the development of HTS materials in this area, the dynamic response of the vehicle superconductor under an alternating magnetic field with high frequency is well-worth studying. However, limited by some experimental conditions and other factors, the effective modelling method becomes an indispensable and cost-effective tool to predict the electromagnetic properties of the superconductors. In this paper, based on the single-peaked permanent magnet guideway (PMG)-HTS bulks maglev configuration, the modelling using three different E-J relationships include the power law model (PLM), the flux flow and creep model (FFCM), and the flux flow model (FFM) are compared. Results indicate that the PLM has the highest computational efficiency and good reliability. Then, using the PLM, we further study the dynamic response of the HTS bulks over a Halbach-typed PMG under higher vibration frequencies. The effect of the magnet failure in the middle of the PMG on the levitation system is also discussed. A finite element method is applied with the H formulation. The moving mesh is used to realize the relative displacement between the PMG and HTS bulks. To investigate the temperature rise and the AC loss inside the HTS bulks, the Heat Transfer module is coupled. Results show that the temperature inside the HTS bulks will decay exponentially with time if the magnitude and frequency of the external magnetic field do not continue to increase. Aims: To realize the so-called "Second Green Revolution", it is imperative to study the roots of crop plants, and identify those traits that improve the efficiency of nitrogen (N) acquisition. We aimed to evaluate how the N acquisition efficiency of six hybrid maize lines commonly grown in northern China depends on their root anatomy. Methods: Maize hybrids classified as having high nitrogen uptake efficiency (HNUE) and low nitrogen uptake efficiency (LNUE) were grown under high-N and low-N conditions in the greenhouse and the field. Results: Under N stress in the field and the greenhouse, HNUE increased shoot dry weight, root length density, N content and nitrogen use efficiency compared to LNUE. Low N availability increased the percentage of root cortical aerenchyma and the size of cortical cells. Root anatomy, with greater formation of root cortical aerenchyma and larger cortical cell size, was associated with increased specific nitrogen absorption efficiency (SNAE) and shoot biomass under N stress. Under low N availability, the percentage of aerenchyma and their total area had significant positive correlations with the shoot dry weight, total N uptake, SNAE. Conclusions: The results suggest that plants in limited N availability form more root cortical aerenchyma and have larger cortical cells, which is of benefit to root growth, soil exploration, N acquisition, and shoot biomass. These observations support the hypothesis that root anatomical phenotypes that affect the metabolic and construction costs of producing root length merit consideration as selection criteria for breeding to improve N acquisition in hybrid maize. RIPK3 (receptor-interacting protein kinase 3) is a serine/threonine-protein kinase. As a key component of necrosomes, RIPK3 is an essential mediator of inflammatory factors (such as TNFα-tumor necrosis factor α) and infection-induced necroptosis, a programmed necrosis. In addition, RIPK3 signaling is also involved in the regulation of apoptosis, cytokine/chemokine production, mitochondrial metabolism, autophagy, and cell proliferation by interacting with and/or phosphorylating the critical regulators of the corresponding signaling pathways. Similar to apoptosis, RIPK3-signaling-mediated necroptosis is inactivated in most types of cancers, suggesting RIPK3 might play a critical suppressive role in the pathogenesis of cancers. However, in some inflammatory types of cancers, such as pancreatic cancers and colorectal cancers, RIPK3 signaling might promote cancer development by stimulating proliferation signaling in tumor cells and inducing an immunosuppressive response in the tumor environment. In this review, we summarize recent research progress in the regulators of RIPK3 signaling, and discuss the function of this pathway in the regulation of mixed lineage kinase domain-like (MLKL)-mediated necroptosis and MLKL-independent cellular behaviors. In addition, we deliberate the potential roles of RIPK3 signaling in the pathogenesis of different types of cancers and discuss the potential strategies for targeting this pathway in cancer therapy. As with most organ systems that undergo continuous generation and maturation during the transition from fetal to adult life, the hematopoietic and immune systems also experience dynamic changes. Such changes lead to many unique features in blood cell function and immune responses in early childhood. The blood cells and immune cells in neonates are a mixture of fetal and adult origin due to the co-existence of both fetal and adult types of hematopoietic stem cells (HSCs) and progenitor cells (HPCs). Fetal blood and immune cells gradually diminish during maturation of the infant and are almost completely replaced by adult types of cells by 3 to 4 weeks after birth in mice. Such features in early childhood are associated with unique features of hematopoietic and immune diseases, such as leukemia, at these developmental stages. Therefore, understanding the cellular and molecular mechanisms by which hematopoietic and immune changes occur throughout ontogeny will provide useful information for the study and treatment of pediatric blood and immune diseases. In this review, we summarize the most recent studies on hematopoietic initiation during early embryonic development, the expansion of both fetal and adult types of HSCs and HPCs in the fetal liver and fetal bone marrow stages, and the shift from fetal to adult hematopoiesis/immunity during neonatal/infant development. Ceramic micro-shot peening (CMSP) and steel micro-shot peening (SMSP) were utilized to investigate the effect of micro-shot peening (MSP) on the high-cycle fatigue properties of Al-7Si-0.3Mg casting aluminum alloy in a previous study. However, the improvement effects of CMSP and SMSP on the fatigue strength (at 5 × 107 cycles) were only 33% because the depth of harden layers was only 20 and 55 μm while the depth of compressive residual stress affected layers was only 37 and 68 μm. In this study, conventional shot peening (CSP) was utilized, and the results were compared with those of MSP, with the expectation that CSP would provide a greater improvement in the fatigue strength. The affected surface layers of the shot-peened specimens were characterized using surface morphology, microhardness, and residual stress analyses. In addition, the effect of CSP on the fatigue strength at 5 × 107 cycles was investigated using a rotating bending fatigue test (R = − 1). An investigation of the extensive surface compressive residual stress relaxation process for the three different shot-peened specimens during cyclic loading was conducted using x-ray diffraction. In addition, the initiation sites for fatigue cracks on the fracture surface were observed using scanning electron microscopy. Furthermore, the fatigue life of the samples with the internal casting defect failure mode was predicted using linear elastic fracture mechanics, while that for samples with the surface crack initiation failure mode was predicted using the modified Morrow model considering the residual stress. Improved the utilization of fertilizer while maintaining the increased of grain yield was the focus of Chinese researchers. Nutrient uptake, distribution, and remobilization are important factors affecting the fertilizer utilization and grain yield of maize. This study aimed to provide a theoretical and practical basis for science-based, high-yielding, and high-efficiency cultivation practices by examining differences in biomass and nutrient uptake, distribution, and remobilization characteristics under three cultivation patterns. We set 12 treatments as follows: super high-yielding cultivation pattern (SH), optimized nutrient management cultivation pattern (ONM), local farmer's practice cultivation pattern (FP), and a series of nutrient omission plots, which excluded nitrogen (N), phosphorus (P), or potassium (K) from the three patterns. The results demonstrated that SH and ONM increased the yield and actual harvested ears by 35.4, 20.7 and by 20.2, 17.6%, respectively. Compared with the FP, SH and ONM increased biomass, N, P, and K accumulation at silking (R1 stage) by 24.4, 31.2, 39.4, and 34.8%, and by 21.7, 22.2, 31.7, and 34.8%, respectively. SH and ONM significantly increased biomass and nutrient distribution to the grains. ONM significantly increased N use efficiency. P and K use efficiency under the ONM pattern was significantly higher than under SH, but was lower than under the FP pattern over two years. This research demonstrates that ONM may significantly reduce fertilizer rates, effectively improve the nutrient remobilization efficiency and uptake at post-silking without negatively affecting grain yield, thereby increasing N use efficiency. This long-term study used a lysimeter platform to monitor the NH3 and N2O emissions of summer maize resulting from various fertilization treatments in the Huanghuaihai area, with the goal to assess the efficiency of fertilization measures aimed at reducing NH3 and N2O losses during the production of summer maize; the results provide a theoretical basis for synergistically improving maize yield and fertilizer utilization efficiency. A 2-year field trial was conducted. The trial included a no-N-fertilizer treatment as a control to study the following three fertilizer treatments: the exclusive application of urea, the exclusive application of cattle manure, and the combined application of organic and inorganic fertilizers. The results show that during the two maize growth seasons included in the trial, the average N2O losses associated with exclusively applying urea, exclusively applying cattle manure, and the combined application of organic and inorganic fertilizers were 273%, 542%, and 376% higher than those associated with the control treatment, respectively. During the same period, the average accumulative ammonia volatilization losses were 311%, 542% and 376% higher than those for the control treatment, respectively. The average nitrogen accumulation resulting from the combined application of organic and inorganic fertilizers was 82% higher than that for the control treatment, 12% higher than that for the exclusive application of cattle manure, and 6% higher than that for the exclusive application of urea. The average grain yield for the combined application of organic and inorganic fertilizers, the exclusive application of urea and the exclusive application of cattle manure were 76%, 68% and 61% higher than that for the control treatment, respectively. Overall, the combined application of organic and inorganic fertilizers showed a lower ammonia volatilization loss than the exclusive application of urea, which resulted in a higher ammonium nitrogen and nitrate nitrogen content in the soil, an increased nitrogen uptake, an increased dry matter accumulation of maize, and a high grain yield and nitrogen recovery efficiency. Purpose: This long-term study used a lysimeter platform to monitor the changes of organic matter and total nitrogen content in deep soils of various fertilization treatments of wheat-maize in the Huanghuaihai area, with the goal to improve the soil environment and increase the quantity and diversity of soil microbes, and then provide a theoretical basis for synergetic improvement of production and resource utilization efficiency. Materials and methods: The experiment included a no-N-fertilizer treatment as a control (CK) to study the following three fertilizer treatments: the exclusive application of urea (U), the exclusive application of cattle manure (M), and the combined application of organic and inorganic fertilizers (UM). Results and discussion: These results showed that fertilization significantly increased the soil organic matter content. Compared with CK, the average soil organic matter content of each soil layer treated with M, UM, and U was increased by 25.00%, 22.81%, and 8.96% in wheat growing season, respectively. The average soil organic matter content of each soil layer in the 2-year average M, UM, and U treatments increased by 148.70%, 16.81%, and − 7.18% maize growing season, respectively. In the 2-year wheat field, in the 0–20 cm soil layer, the effects of each treatment on the total nitrogen content of the soil were significant, and compared with the CK treatment, the UM, M, and U treatments increased by 105.72%, 75.07%, and 47.13%. For maize field, compared with the CK treatment, the total nitrogen content of M, UM, and U increased by 83.34%, 50.27%, and 7.10%. The addition of organic fertilizer can increase the number and diversity of bacteria and fungi in the soil, and the application of urea significantly reduced the quantities and diversities of bacteria. The combined application of organic and inorganic fertilizers can increase the number and diversity of soil actinomycetes. The combined application of organic and inorganic fertilizers could significantly increase the number of maize roots. Compared with CK treatment, the number of roots increased by 66.67%. After long-term experimental treatment, the yields of wheat and maize were all expressed as UM > U > M > CK. Conclusions: The combination of organic and inorganic N fertilizers increased the soil organic matter content, improved the soil environment, increased the quantity and diversity of soil microbes, promoted the growth and development of crop roots, and increased the yield of wheat and maize. An alloyed ferritic-pearlitic steel is studied after micro-shot peening with balls from a high-speed steel. The morphology and the roughness of the surface of the specimens are determined. Specimens with smooth and notched surfaces are subjected to multi-cycle fatigue tests. The fatigue resistance of the specimens with a smooth surface and with a notched surface increases by 22 and 27%, respectively, after the shot-peening. In pursuit of maximum grain yield farmers in the North China Plain usually apply excessive N fertilizer, resulting in wasted resources and environmental pollution. To assess the economic and environmental performances of different nitrogen rates will be conductive to sustain cleaner crop production. An 8-year field experiment was carried out with four treatments, N0 (0 kg ha−1 for winter wheat and summer maize), N1 (168 kg ha−1 for winter wheat and 129 kg ha−1 for summer maize), N2 (240 kg ha−1 for winter wheat and 185 kg ha−1 for summer maize) and N3 (300 kg ha−1 for winter wheat and summer maize), on the double cropping at Dawenkou research field (36°11'N, 117°06'E), Shandong Province, China. The crop production, soil physical-chemical parameters, and greenhouse gas emission are measured and the economic and environmental performances are assessed. The optimal nitrogen rate obtained the highest grain yield of summer maize in 4 of 8 year and was equivalent to conventional N rate in the other years. The nitrogen partial factor productivity and agronomic efficiency of optimal nitrogen rate was 63% and 58% higher than that of conventional nitrogen rate. The optimal nitrogen rate effectively decreased soil bulk density and increased weight percentage of water-stable aggregate and activities of urease and invertase compared to conventional nitrogen rate, which improved soil productivity. The fertilizer nitrogen loss and global warming potential of optimal nitrogen rate reduced by 76% and 35% compared to conventional nitrogen rate. The annual greenhouse gas intensity of optimal nitrogen rate decreased by 14–35% compared to others. The net ecosystem economic budget under optimal nitrogen rate is 252–604 $ ha−1 yr.−1 higher than other addition levels. The objective of this study was to evaluate the ability of the phytohormone S-abscisic acid (S-ABA) to protect maize seedlings grown under drought stress and to measure their increased drought tolerance. The maize hybrids 'Zhengdan 958' (ZD958; drought tolerant) and 'Xundan 20' (XD20; drought sensitive) were treated with nutrient solutions of different concentrations (1, 2, 4, 8, and 10 mg/kg) of S-ABA under polyethylene glycol (PEG, 15% w/v, MW 6000) simulated drought stress. Optimal concentrations of S-ABA were designed to be sprayed onto the leaves of seedlings, and their effect on endogenous ABA, malondialdehyde (MDA), osmotic substances, antioxidant enzyme activities, and Asr1 gene expression in seedlings were studied. Results indicated that, under drought stress, S-ABA treatment significantly improved maize seed germination rate (GR), germination energy (GE), and seedling biomass (p < 0.05). After spraying 4 mg/kg S-ABA onto leaves, the endogenous hormone ABA, osmotic substances, antioxidant enzyme activities, and expressive quantity of the Asr1 gene were extended and MDA content dropped significantly (p < 0.05). Moreover, ZD 958 endogenous ABA content, osmotic substances content, antioxidant enzyme activity and Asr1 gene expressive quantity were higher than that of XD 20 (p < 0.05). In conclusion, S-ABA treatment increased the content of endogenous ABA, induced an increase in antioxidant enzyme activity and Asr1 gene expression level, reduced the oxidative damage caused by drought to maize leaves, and improved the adaptability of maize seedlings to withstand drought stress. In this paper, EA4T alloy railway axle steel was shot peened with steel balls and ceramic balls with a diameter of 50 μm, and 109-cycle fatigue tests of unpeened and peened specimens were carried out. The fatigue properties and the surface compression residual stress relaxation processes were investigated. The results of surface fracture observation indicated that all the cracks initiated from the surface, and both peened and unpeened specimens have conventional fatigue limits in the very high-cycle fatigue regime. Moreover, compared with unpeened (UP) specimens, the fatigue limits were improved by 31 and 28% for ceramic shot-peened (CSP) and steel shot-peened (SSP) specimens, respectively. The improvement of fatigue limit is related to the surface compressive residual stresses (SCRS) and the surface-hardened layer. The SCRS and full-width half-maximum (FWHM) values during the relaxation processes have been analyzed. At stress amplitudes lower than the cyclic yield strength, the residual stress of SSP specimens relaxed quickly in the first loading cycle and then remained stable, while the residual stress of CSP specimens relaxed quickly in the first loading cycle and relaxed slowly afterward, then remained stable. For all SSP and CSP specimens with applied stress amplitudes larger than the cyclic yield strength, the residual stress relaxed quickly in the initial cycles, and then remained stable after the first loading cycle. Purpose: In this study, we analyzed the effects of different maize varieties with nitrogen utilization efficiency, fertilizer type, and rate on the ammonia volatilization emission of farmland. Aimed to seek the best matching method to improve grain yield and fertilizer utilization efficiency of summer maize simultaneously. Materials and methods: In field experiments, we choose two maize varieties with different nitrogen utilization efficiency (Zhengdan958, Z and Lainong14, L) as material. Set four different fertilizer treatments (200 kg N hm−2 inorganic fertilizer (U1), 100 kg N hm−2 inorganic fertilizer (U2), 200 kg N hm−2 organic fertilizer (M1), and 100 kg N hm−2 organic fertilizer (M2) to study their effect on NH3 emission and loss, maize grain yield, and nitrogen accumulation. Results and discussion: Ammonia volatilization accounted for 8.61–21.68% of applied N. Under the same variety, ammonia volatilization accumulation after fertilization was as follows: U1 > U2 > M1 > M2. Ammonia volatilization rates increased first and then gradually decreased after the fertilization. The ammonia volatilization loss and cumulative loss increased due to increased nitrogen fertilizer application rate. The average nitrogen accumulation and harvest index of 200 kg N hm−2 N treatments were higher than 100 kg N hm−2 N treatments, and the difference between the inorganic fertilizer and organic fertilizer was not significant. In 2016 and 2017, the average yield of Zhengdan958 was 11,758.79 kg hm−2, which was 15.78% higher than that of Lainong14, and the difference between the two fertilizer types was not significant. The average yield of 200 kg N hm−2 N treatment was 11,959.42 kg hm−2, which was 20.13% higher than those of 100 kg N hm−2 N treatment. Conclusions: By changing the type of fertilizer, replacing chemical fertilizers with organic fertilizer can reduce the loss of ammonia volatilization and promote the synergistic improvement to yield and resource utilization efficiency. Among them, using nitrogen-efficient varieties and using organic fertilizer instead of chemical fertilizer was beneficial to reduce the loss of ammonia volatilization, increase the accumulation of nitrogen, and promote the growth of maize to obtain high yield. Low light is a type of abiotic stress that seriously affects plant growth and production efficiency. We investigated the response mechanisms of summer maize to low light by measuring the changes in endogenous hormones in the grains and during grain filling in summer maize at different light intensities to provide a theoretical basis for the production and management of summer maize under light stress. We applied different light treatments in a field experiment as follows: S, shading from tassel stage (VT) to maturity stage (R6); CK, natural lighting in the field; and L, increasing light from VT to R6. The shading level was 60%, and the maximum illumination intensity of the increasing light treatment on cloudy days was 1600-1800 μmol m−2 s−1. Compared with the control, shading significantly increased the grain abscisic acid (ABA) content at 5-20 days after pollination and decreased the indole acetic acid (IAA), zeatin riboside (ZR), and gibberellin (GA) contents (P < 0.05). The grain-filling rate decreased under shading conditions. Meanwhile, the grain volume, grain weight, and yield all decreased; the yields in 2013 and 2014 decreased by 61 and 60%, respectively. The grain IAA, ZR, and GA contents were increased by increasing light. The grain ABA content at 5-20 days after pollination did not significantly differ from that of CK (P < 0.05). After 20 days after pollination, the ABA content decreased, the grain-filling rate and the filling duration increased, and the yield increased. However, shading after anthesis increased the grain ABA content and reduced the IAA, ZR, and GA contents. Grain growth and development were inhibited, and the yield decreased. The grain ABA content decreased; the IAA, ZR, and GA contents increased; and the yield increased after increasing light. The results indicate that different light intensities regulated the levels of grains endogenous hormones, which influenced the grain-filling rate and duration, and consequently, regulated grain weight and yield. The effect of shear-affected zone (SAZ), with a stress-concentration source induced by the punching process, on tensile properties was investigated. Tests using honed specimens (which have the same shapes and stress-concentration without any SAZ) and smooth specimens were conducted to compare the effect with that of the punched specimens. Dual-phase steel, which has a high work-hardening ability and low yield strength, and precipitation-hardened steel, which has a low work-hardening ability and high yield strength, were used in the tests. Materials with two tensile strength grades were prepared from both types of steel. Only the precipitation-hardened steel with higher strength grade punched specimen showed a brittle fracture with extremely short fracture-elongation, whereas the other specimens showed a ductile fracture. The fracture surface analysis revealed that cracks initiated in the maximum shear stress plane of the SAZ under tensile loading at first. We call the crack “shear crack.” The steel which showed brittle fracture used in this study easily exhibited plastic-strain localization compared with the other steels. If the shear crack is sharp, then the transition from ductile to brittle failure tends to occur. Furthermore, the strength characteristics of the punched specimen depend on the crack length dependency of the strength resistance and the failure phenomenon of the original material. To investigate the interactive effects of water and N from controlled release urea (CRU) on N metabolism, accumulation, translocation, and yield in Zhengdan958 (a summer maize cultivar planted widely in China), three water levels (adequate water W3, mild water stress W2, severe water stress W1) and four amounts of CRU (N) (N0, N1, N2, and N3 were 0, 105, 210, and 315 kg N ha, respectively) were carried out under the waterproof shed and soil column conditions. The results showed that yield, N metabolism, accumulation, and translocation were significantly affected by water, CRU, and their interactions after tasseling. Yields showed an increasing trend in response to N rates from 100.2 to 128.8 g plant under severe water stress (W1), from 124.7 to 174.6 g plant under mild water stress (W2), and from 143.7 to 177.0 g plant under adequate water conditions (W3). There was an associated optimum amount of N for each water level. Under W1 and W2, N3 treatments showed significant advantages in three N metabolism enzymes' activities and the N accumulations, and yield and its components were highest. But the nitrogen harvest index (NHI) of N3 had no significant difference with other nitrogen treatments. Under W3, the N translocation efficiency (NTE) and N translocation conversion rate (NTCR) of N2 in stem and leaf were higher than those of N3, but the N metabolism enzymes' activities and yields of N2 and N3 had no significant difference, which indicated that N2 was superior to N3. The N3 treatment under W2 and N2 under W3 increased the N accumulation capacity in maize grain as well as the N translocation to grain that contributed to the increase of 1000-gain weight and grains per ear after tasseling. Under this experimental condition, a CRU rate of 225 kg ha was the best treatment when the soil moisture content was 75 ± 5% of field capacity, but an N rate of 300 kg ha was superior when soil moisture content was maintained at 55 ± 5% of field capacity during the entire growing season. A field experiment was performed to study the effects of spraying exogenous 6-benzyladenine (6-BA) after waterlogging for 6 days at the third leaf stage on the leaf mesophyll cell ultrastructure, chlorophyll content, gas exchange parameters, chlorophyll fluorescence, and malondialdehyde (MDA) content of summer maize ( Zea mays L.) hybrids Denghai605 (DH605) and Zhengdan958 (ZD958), to determine the photosynthetic mechanism for the regulation of 6-BA to waterlogged summer maize. Results showed that exogenous application of 6-BA effectively alleviated the adverse effects of waterlogging on photosynthetic characteristics by increasing leaf area index and chlorophyll content. Treatment with spraying exogenous 6-BA resulted in significant and substantial (in the range of 9-37%) increases of the net photosynthetic rate compared to waterlogging treatments. In addition, spraying 6-BA effectively alleviated the adverse effects of waterlogging on leaf ultrastructure by increasing the numbers of grana and grana lamella, decreasing MDA content, and improving membrane structure and chloroplast structure. Visibly, spraying exogenous 6-BA effectively alleviated the adverse effects of waterlogging on leaf ultrastructure of summer maize led to the improvement of photosynthetic performance, resulting in significant and substantial (around 20%) increases in grain yield compared to those of the waterlogging treatments. Plant density has been recognized as a major factor determining the grain yield. The photosynthetic performance changes as the density increases. The main objective of this research was to evaluate responses of photosynthetic performance and chloroplast ultrastructure to planting densities in two summer maize ( Zea mays L.) hybrids Denghai661 (DH661) and Nongda108 (ND108). DH661 was planted at densities of 30,000, 45,000, 60,000, 75,000, 90,000, 105,000, 120,000, or 135,000 plants ha. ND108 was planted at densities of 30,000, 45,000, 60,000, 75,000, or 90,000 plants ha. Research variables included leaf area, grain yield, chlorophyll content, leaf gas exchange parameters, number of chloroplasts, and chloroplast ultrastructure. As plant density increased, chlorophyll a and b content significantly decreased; carotenoids initially decreased and then increased; the net photosynthetic rate during each growth period significantly decreased; the membrane structure of mesophyll cells was gradually damaged; the number of chloroplasts significantly decreased; the external form of chloroplasts shifted from long and oval to elliptical or circular; the number of grana significantly decreased, while the number of grana lamellae increased; grana gradually became hypogenetic and eventually dissolved; plot yield increased; and yield per plant significantly decreased. The object of this paper is to propose a novel method to evaluate the maximum inclusion size in medium strength steel by ultrasonic fatigue testing. The inclusion sizes in the medium strength steel were evaluated by ultrasonic fatigue testing using the fatigue specimen with a large risk volume under water-cooling condition. To ensure fatigue specimens of medium strength steel fracture from the internal inclusion, heat treatment and oxynitrocarburization were conducted to increase the strength of the specimen and to protect the specimen from surface corrosion induced by cooling water. The results show that evaluation of the inclusion size by the proposed method is more accurate than traditional approaches, which are based on inclusion size characterization from arbitrary two dimensional cross sections. A field experiment was conducted to study the effects of shading on the photosynthetic characteristics and mesophyll cell ultrastructure of two summer maize hybrids Denghai605 (DH605) and Zhengdan958 (ZD958). The ambient sunlight treatment was used as control (CK) and shading treatments (40 % of ambient sunlight) were applied at different growth stages from silking (R1) to physiological maturity (R6) (S1), from the sixth leaf stage (V6) to R1 (S2), and from seeding to R6 (S3), respectively. The net photosynthetic rate ( P ) was significantly decreased after shading. The greatest reduction of P was found at S3 treatment, followed by S1 and S2 treatments. P of S3 was decreased by 59 and 48 % for DH605, and 39 and 43 % for ZD958 at tasseling and milk-ripe stages, respectively, compared to that of CK. Additionally, leaf area index (LAI) and chlorophyll content decreased after shading. In terms of mesophyll cell ultrastructure, chloroplast configuration of mesophyll cells dispersed, and part of chloroplast swelled and became circular. Meanwhile, the major characteristics of chloroplasts showed poorly developed thylakoid structure at the early growth stage, blurry lamellar structure, loose grana, and a large gap between slices and warping granum. Then, plasmolysis occurred in mesophyll cells and the endomembrane system was destroyed, which resulted in the dissolution of cell membrane, karyotheca, mitochondria, and some membrane structures. The damaged mesophyll cell ultrastructure led to the decrease of photosynthetic capacity, and thus resulted in significant yield reduction by 45, 11, and 84 % in S1, S2, and S3 treatments, respectively, compared to that of CK. In this study, Austempered ductile irons (ADIs) with two different strength grades were produced and the fatigue properties were measured at 10 cycles. The results show that the S-N curves give a typical step-wise shape and there is no fatigue limit in the very high cycle fatigue regime. The two grades ADI have the similar fracture behaviors and fatigue failure can initiate from defects at specimen surface and subsurface zone. On the fracture surfaces of some specimens, the 'granular-bright-facet' area with rich carbon distribution is observed in the vicinity of the defect. The microstructure affects the crack behaviors at the early propagation stage. The ADI with upper and lower bainite shows higher fatigue strength compared with the ADI with coarse upper bainite. A field experiment was conducted to study the effects of shading on tassel and ear development and yield formation of three summer maize hybrids Zhenjie 2 (ZJ2), Denghai 605 (DH605), and Zhengdan 958 (ZD958). The ambient sunlight treatment was used as control (CK) and shading treatments (40 % of ambient sunlight) were applied at different growth stages from silking stage (R1) to physiological maturity stage (R6) (treatment S1), from the sixth extended leaf stage (V6) to R1 (treatment S2) and from seeding to R6 (treatment S3). Shading had no significant effect on the time from seeding to shoot emergence (VE); however, subsequent growth and development were delayed with shading beyond this point. The differentiation time of both tassel and ear delayed, and female spike (tassel) floret differentiation, sexual organ formation time, and anthesis-silking interval (ASI) were lengthened. After shading, the total number of floret, silk, and fertilization floret reduced significantly; the number of abortive seeds increased, and the total setting percentage among different treatments showed that CK>S2>S1>S3; and the total setting percentages in S1, S2, and S3 of ZD958 were 44, 72, and 15 % respectively. The total floret number of tassel primordium differentiation, fertility rate, and seed setting rate of florets in S3 treatment was the minimum; kernels per ear decreased seriously and single ear setting percentage was only 16 %; although floret degeneration number of S2 during ear differentiation stages increased and floret fertility rate reduced than that of CK, fertilization flower seed production increased and abortive seed decreased after canceling shading. Aborted kernel of S1 increased and kernel dry weight reduced, resulting in a significant decrease of kernel number per ear and kernel weight, and the grain abortive rate of 40-62 %. In conclusion, shading changed the growth and development process and caused infertility of tassel and ear; tassel branches decreased, reducing pollen vitality and silks differentiation cut down; and grain dry matter accumulation and setting percentage decreased, causing yield reduction. Toll-like receptors (TLRs), which are found in innate immune cells, are essential mediators of rapid inflammatory responses and appropriate T-cell activation in response to infection and tissue damage. Accumulating evidence suggests that TLR signaling is involved in normal hematopoiesis and specific hematologic pathologies. Particular TLRs and their downstream signaling mediators are expressed not only in terminally differentiated innate immune cells but also in early hematopoietic progenitors. Sterile activation of TLR signaling is required to generate early embryonic hematopoietic progenitor cells. In adult animals, TLR signaling directly or indirectly promotes differentiation of myeloid cells at the expense of that of lymphoid cells and the self-renewal of hematopoietic stem cells during infection and tissue damage. Activating mutations of the MyD88 gene, which codes for a key adaptor involved in TLR signaling, are commonly detected in B-cell lymphomas and other B-cell hematopathologies. Dysregulated TLR signaling contributes to the pathogenesis of many hematopoietic disorders, including bone marrow failure, myelodysplastic syndrome, and acute myeloid leukemia. Complete elucidation of the molecular mechanisms by which TLR signaling mediates the regulation of both normal and pathogenic hematopoiesis will prove valuable to the development of targeted therapies and strategies for improved treatment of hematopoietic disorders. Clarification of influencing factors (cultivar planted, cultivation management, climatic conditions) affecting yields of summer maize ( Zea mays L.) would provide valuable information for increasing yields further under variable climatic conditions. Here, we report actual maize yields in the Huanghuaihai region over the past 50 years (1957-2007), simulated yields of major varieties in different years (Baimaya in the 1950s, Zhengdan-2 in the 1970s, Yedan-13 in the 1990s, and Zhengdan-958 in the 2000s), and factors that influence yield. The results show that, although each variety change has played a critical role in increasing maize yields, the contribution of variety to yield increase has decreased steadily over the past 50 years (42.6%-44.3% from the 1950s to the 1970s, 34.4%-47.2% from the 1970s to the 1990s, and 21.0%-37.6% from the 1990s to the 2000s). The impact of climatic conditions on maize yield has exhibited an increasing trend (0.67%-22.5% from the 1950s to the 1970s, 2.6%-27.0% from the 1970s to the 1990s, and 9.1%-51.1% from the 1990s to the 2000s); however, interannual differences can be large, especially if there were large changes in temperature and rainfall. Among climatic factors, rainfall had a greater positive influence than light and temperature on yield increase. Cultivation measures could change the contribution rates of variety and climatic conditions. Overall, unless there is a major breakthrough in variety, improving cultivation measures will remain important for increasing future summer maize yields in the Huanghuaihai region. In humans, mutations in BMPR1A, SMAD4 and PTEN are responsible for juvenile polyposis syndrome, juvenile intestinal polyposis and Cowden disease, respectively. The development of polyposis is a common feature of these diseases, suggesting that there is an association between BMP and PTEN pathways. The mechanistic link between BMP and PTEN pathways and the related etiology of juvenile polyposis is unresolved. Here we show that conditional inactivation of Bmpr1a in mice disturbs homeostasis of intestinal epithelial regeneration with an expansion of the stem and progenitor cell populations, eventually leading to intestinal polyposis resembling human juvenile polyposis syndrome. We show that BMP signaling suppresses Wnt signaling to ensure a balanced control of stem cell self-renewal. Mechanistically, PTEN, through phosphatidylinosital-3 kinase-Akt, mediates the convergence of the BMP and Wnt pathways on control ofß-catenin. Agrotis ipsilon (Lepidoptera: Noctuidae) is a major underground pest that damages many agricultural crops in China and other countries. A diet-incorporation-based bioassay was conducted to evaluate the sublethal effects of the novel anthranilic diamide chlorantraniliprole on the nutritional physiology, enzymatic properties and population parameters of this cutworm. Chlorantraniliprole exhibited signs of active toxicity against third instar larvae of A. ipsilon, and the LC50 was 0.187 μg.g−1 of artificial diet after treatment for 72 h. The development time of the larval, pupal and adult stages was significantly affected after chlorantraniliprole exposure, compared to the control treatment. Relative to the control treatment, chlorantraniliprole decreased pupal and adult emergence rates, fecundity and fertility and increased the proportions of developmental deformities, the adult preoviposition period (APOP) and the total preoviposition period (TPOP). Furthermore, compared to those treated with the control, A. ipsilon larvae treated with low doses of chlorantraniliprole decreased food utilization and nutrient content (protein, lipid, carbohydrate, trehalose), showed lower pupal weights and growth rates. Compared with the control treatment, chlorantraniliprole significantly reduced digestive enzyme activities and observably increased detoxifying and protective enzyme activities and hormone titers. Importantly, these chlorantraniliprole-induced changes affected life table parameters of the cutworm. These results suggest that chlorantraniliprole at low concentrations can impair A. ipsilon development duration, normal food consumption and digestion process, enzymatic properties, hormone levels, fecundity and population levels. Various literatures have demonstrated that overexpression of Metadherin (MTDH) is correlated with tumor metastasis and it can predict poor survival outcomes in female reproduction malignancies. In order to enhance the statistical power and reach a recognized conclusion, we conducted a systematic review and meta-analysis to thoroughly investigate the association of MTDH expression with tumor metastasis and survival outcomes following PRISMA guidelines. Odds ratios (ORs) and hazard ratios (HRs) were used to demonstrate the impact of MTDH on tumor metastasis and prognosis respectively. Data were pooled with appropriate effects model on STATA12.0. Our results indicated that high MTDH expression is significantly correlated with higher mortality for breast, ovarian and cervical cancer. High immunohistochemical expression of MTDH is remarkably associated with shorter disease-free survival (DFS) in breast cancer but not in ovarian cancer. The pooled results suggested that high level of MTDH significantly predicted distant metastasis and lymph node metastasis in breast cancer. Strong associations were observed between MTDH expression and lymph node metastasis in ovarian and cervical cancer. In conclusion, MTDH might be a novel biomarker which can effectively reflect metastasis status and prognosis of breast cancer.
