Background: Phenolic compounds (PC) are found in various foods that are part of the human diet, such as mushrooms. The identification of PC is crucial due to their bioactive properties and their role in disease prevention. This study aimed to assess the antioxidant and antidiabetic properties of the phenolic component extract (PCE) derived from the Pleurotus ostreatus mushroom. The extract was separated using high-performance liquid chromatography (HLPC). The PCE was then tested on diabetic rats fed a high-fat diet (HFD). Also, this study was an attempt to treat infertility caused by type 2 diabetes in male rats. Methods: Streptozotocin (STZ) was administered to Wistar rats after a 4-week HFD as a model of type 2 diabetes (D2). Animals were given phenols orally on a high-fat diet for four weeks after being diagnosed with diabetes. Results: PCE constituents were 48.02% carbohydrates, 21.15% protein, 6.16% fiber, 2.06% fat, and 6.93% ash. The evaluation of the PCE was carried out by assessing its retention rates and subsequently comparing the UV data to standards. Total phenols and flavonoids in the crude extract were 23.04 mg/g gallic acid equivalent and 8.14 mg/g quercetin equivalent, respectively. Catechin was found as the significant PC (121.53 µg/mL), while gallic acid was the second PC (11.64 µg/ml) in the PCE of mushrooms. PCE showed high radical scavenging activity on DPPH. With dosage and incubation duration in dark-dependent conditions, it was discovered to be 93.23% at a 500 µg/ml concentration after 120 min in the dark. PCE reduces blood glucose levels while elevating insulin levels and antioxidant activity indicators. Our results show that phenols and their bioactive components can change glucose metabolism in STZ-induced diabetic HFD rats (D2). Conclusions: This study supports PCE's therapeutic potential in treating hyperglycemia, increasing insulin secretion, and addressing diabetes-induced infertility. [ABSTRACT FROM AUTHOR]

Inflammation is a part of the body's intricate biological reaction to noxious stimuli and defensive reactions. So, the aim of this investigation was to study the anti-inflammatory activity of exopolysaccharide (EPSSM) using carrageenan-induced paw edema in rats. A halophilic bacterial strain was isolated from marine sediments in the Red Sea in Egypt. The isolate has been visually and physiologically recognized, as well as by analyzing its 16S rRNA gene, which confirms Kocuria sp. clone Asker4. This particular isolate can be referenced using the accession number OL798051.1. EPSSM was subjected to purification and fractionation by a DEAE-cellulose column. Preliminary chemical analysis of EPSSM indicated that the monosaccharides were fructose, glucuronic acid, and xylose, with 2.0, 0.5, and 1.0, respectively. The antioxidant potential of EPSSM was investigated, and it was discovered that the level of activity increased independently of the concentrations, reaching a maximum threshold of 94.13% at 100 µg/mL of EPSSM for 120 min. Also, EPSSM at 50 mg/kg orally produced a significant anti-inflammatory effect on the carrageenan model at 2, 3, and 4 intervals. The EPSSM intervention resulted in reductions in the levels of catalase and superoxide dismutase enzymes, as well as a decrease in glutathione. Furthermore, the levels of nitric oxide, lipid peroxidation, and reactive oxygen species resulting from carrageenan-induced edema showed a significant reduction subsequent to the administration of EPSSM. Moreover, the findings indicated that the protein expression levels of cyclooxygenase-2 and interleukin-6 were reduced following treatment with EPSSM, resulting in a reduction of paw edema. Background: L-arginase, is a powerful anticancer that hydrolyzes L-arginine to L-ornithine and urea. This enzyme is widely distributed and expressed in organisms like plants, fungi, however very scarce from bacteria. Our study is based on isolating, purifying, and screening the marine bacteria that can produce arginase. Results: The highest arginase producing bacteria will be identified by using microbiological and molecular biology methods as Bacillus licheniformis OF2. Characterization of arginase is the objective of this study. The activity of enzyme was screened, and estimated beside partial sequencing of arginase gene was analyzed. In silico homology modeling was applied to generate the protein's 3D structure, and COACH and COFACTOR were applied to determine the protein's binding sites and biological annotations based on the I-TASSER structure prediction. The purified enzyme was undergone an in vitro anticancer test. Conclusions: L-arginase demonstrated more strong anti-cancer cells with an IC50 of 21.4 ug/ml in a dose-dependent manner. L-arginase underwent another investigation for its impact on the caspase 7 and BCL2 family of proteins (BCL2, Bax, and Bax/Bcl2). The purpose of this investigation is the supportive functions of L-carnitine when co-administered with the DDP-4 inhibitor member and the nephroprotective impact of alogliptin in nephrotoxicity induced by gentamicin in male Wistar rats. A total number of 25 male Albino rats weighed 130.0±5.7 g, and divided into five equal groups, G1: (Control negative), G2: (Control positive, has nephrotoxicity), G3: (Treated group with L-carnitine), G4: (Treated with Alogliptin) and G5: (Treated with alogliptin and L-carnitine). The biochemical results of serum total proteins, albumin, urea, uric acid, and creatinine were significantly changed (P ≤ 0.05) among different groups while serum globulin was not significant, and the mean values of these parameters showed significant variations in G5 than G3 and G4. The following measures were made in order to identify the oxidative/antioxidant cascades: glutathione peroxidase (GPx), catalase (CAT);, concentration of malondialdehyde (MDA) and Super Oxide Dismutase (SOD) and these parameters were showed significant changes (P ≤ 0.05) among different groups and the mean value showed the significant of G5 than G3 and G4. The results of the histological and immunohistochemical analyses point to a possible function for alogliptin and L-carnitine in preventing renal tissue destruction in rat models of gentamicin-induced nephrotoxicity. According to the improvement of various biochemical indicators and oxidative state as well as the restoration of the renal structural integrity and function, treatment with alogliptin or L-carnitine protects the kidney through their antioxidant action. Chemotherapeutic medications, including 5 – fluorouracil (5FU), are the same old technique to most cancers and are associated with numerous peripheral toxicities. We investigated exopolysaccharide (EPSST) produced from the isolated streptomycete of the Mediterranean Sea for the capability to lower the severity of mucositis in vivo. The streptomycete was isolated from Mediterranean Sea sediment from the beaches of Port Said Governorates, Egypt and identified morphologically, physiologically, and biochemically and confirmed by molecularly 16S rDNA analysis. The EPSST was extracted from the supernatant of streptomycete by using 4 volumes chilled ethanol and then the functional groups, MW, and chemical evaluation have been detected via Fourier-transform infrared (FTIR), and high-performance liquid chromatography (HPLC). In addition, antioxidant activity was measured through the usage of 2, 2-diphenyl-1-picrylhydrazyl (DPPH). Thirty-two male rats (180–200 g) were randomly divided into a control group (normal saline), intraperitoneal injection of 5-fluorouracil (5-FU, 150 mg/kg), normal rats were treated with EPSST and 5-FU + EPSST group. These groups were continued up to the day of sacrifice (28 days post treatments). The isolated strain became recognized based totally on 16S rDNA sequence as Streptomyce sp. with accession number SAMN08349905. The chemical evaluations of EPSST were galacturonic, glucose, galactose, mannose, and arabinose with a relative ratio of 2.1: 1: 5.37: 1.62: 1.29 individually, with an average molecular weight (Mw) 9.687 × 103 g/mol. Also, the EPSST contained uronic acid (16%) and sulfate (12.149%) and no protein was detected. EPSST inhibited the DPPH radical activity. The findings of this study propose that EPSST inhibits 5-FU-induced mucositis through adjustment of oxidative stress, apoptosis, inflammatory factors, activation of antioxidant enzymes. The clinical administration of EPSST may recover the chemotherapy-induced intestinal dysfunction, consequently increasing the clinical efficiency of chemotherapy. In addition, the administration of EPSST reduced 5-FU-induced histopathological incongruities such as neutrophil infiltration, loss of cellular integrity, and villus and crypt distortion. The clinical administration of EPSST may recover the chemotherapy-induced intestinal dysfunction, consequently increasing the clinical efficiency of chemotherapy. [ABSTRACT FROM AUTHOR]

Optimization of the culture medium conditions for Bacillus thuringiensis var. israelensis mosquitocidal toxins production under solid state fermentation using Taguchi experimental design of surface response methodology was studied. The obtained results revealed that the optimum culture medium conditions for the maximum mosquitocidal activity against second instar Culex pipiens larvae were 6% substrate concentration, 40% initial moisture content, 2% inoculum size and initial pH 6.5 for 7 days incubation period. The obtained sporulation titer and larval mortality % were 2.2 × 1010 CFU/g final product and 90%, respectively. LC50 of this product was 3.2 ppm. [ABSTRACT FROM AUTHOR]

Mosquitocidal toxins ofBacillus thuringiensis israelensis(Bti) andLysinibacillus sphaericus14N1 (Ls14N1) were produced under solid-state fermentation using agro-industrial wastes. Sugar beet pulp–sesame meal (1:1) and wheat germ meal–linen meal (1:1) at 9% were the efficient substrate mixtures for the growth and toxin production of Bti and Ls14N1, respectively. Bti was more active after the addition of beef extract (0.2%) or yeast extract (0.5%) to the medium. On the other hand, the addition of yeast extract (0.2%) or NYSM salts (2%) significantly enhanced the toxicity produced by Ls14N1. The optimum conditions for the maximum toxicity of Bti were at pH 7–8, 20–30% moisture, 4–10% inoculum and 7 days incubation. For Ls14N1, the best conditions were pH 6.5–7.5, 20–30% moisture, 4–10% inoculum and 5 days incubation. It was found that the best thickness of carrier-substrates in the plate (15 cm in diameter) for the maximum mosquitocidal activity was about 0.5 cm for Bti and 0.5–1 cm for Ls14N1. Pilot-scale production in aluminium trays applying the above conditions showed a decrement of toxicity of fermented cultures and some plates were contaminated. These problems were dissolved by reducing the moisture content to 15%, increasing inoculum to 10% and manual agitation of trays every-day. [ABSTRACT FROM PUBLISHER]

Objectives: The first goal of this study was to synthesize the silver nanoparticles Alcaligenes xylosoxidans exopolysaccharide (Ag-AXEPS). The second objective was to analyse the role of Ag-AXEPS nanoparticles (NPS) in treating bleomycin (BLM)-induced lung fibrosis., Methods: Intratracheal bleomycin (2.5 U/kg) was administered to prompt pulmonary fibrosis in rats, and pulmonary fibrosis was treated with Ag-AXEPS nanoparticles (100 ppm/twice a week for four weeks)., Key Findings: Ag-AXEPS nanoparticles significantly decreased the diversity of pulmonary inflammatory agents in rats with BLM-induced fibrosis. Reduced levels of respiratory tumor necrosis factor-alpha, monocyte chemotactic protein-1, matrix metalloproteinases (MMP-2 and MMP-9) were observed on treatment with synthesized Ag-AXEPS. Similarly, the treatment decreased IL-12, mRNA levels of BAX and plasma fibrosis markers like N-terminal procollagen III propeptide and transforming growth factor-β1. On the other hand, the treatment increased mRNA BCL2 and total antioxidant capacity. It also lowered the level of fibrosis, as was shown by a quantified pathologic study of hematoxylin-eosin-stained lung parts. The treatment, however, ensured that lung collagen was restored, as assessed by Masson's trichrome stain, and that overall survival was increased and enhanced., Conclusions: Our work showed that nanoparticles could be obtained at 37°C and may be a possible pulmonary fibrosis therapeutic agent., (© The Author(s) 2021.
