N/P dual-doped three-dimensional porous carbon was synthesized via nano-CaCO 3 template-assistant carbonization of intumescent flame retardants (IFRs) precursor. [Display omitted] Heteroatom-doped three-dimensional (3D) porous carbons possess great potential as promising electrodes for high-performance supercapacitors. Inspired by the inherent features of intumescent flame retardants (IFRs) with universal availability, rich heteroatoms and easy thermal-carbonization to form porous carbons, herein we proposed a self-assembling and template self-activation strategy to produce N/P dual-doped 3D porous carbons by nano-CaCO 3 template-assistant carbonization of IFRs. The IFRs-derived carbon exhibited large specific surface area, well-balanced hierarchical porosity, high N/P contents and interconnected 3D skeleton. Benefitting from these predominant characteristics on structure and composition, the assembled supercapacitive electrodes exhibited outstanding electrochemical performances. In three-electrode 6 M KOH system, it delivered high specific capacitances of 407 F g−1 at 0.5 A g−1, and good rate capability of 61.2% capacitance retention at 20 A g−1. In two-electrode organic EMIMBF 4 /PC system, its displayed high energy density of 62.8 Wh kg−1 at a power density of 748.4 W kg−1, meanwhile it had excellent cycling stability with 84.7% capacitance retention after 10,000 cycles. To our best knowledge, it is the first example to synthesize porous carbon from IFRs precursor. Thus, the current work paved a novel and low-cost way for the production of high-valued carbon material, and expanded its application for high-performance energy storage devices. Concessive relations, often indicated by conjunction words such as although, are semantically and pragmatically more complex than causal relations (expressed using because), as they involve more semantic features such as implicated meaning and negation. However, it remains unclear how linguistic-level complexity is manifested through different brain activities and functional connectivities. This fMRI study investigated how the neural underpinnings of concessive relations differ from those of causal relations. Pragmatically congruent and incongruent words were embedded in causal as well as concessive sentences. The whole-brain analysis revealed that relative to because-congruent sentences, although-congruent sentences evoked increased activations in a left network including IFG, bilateral MFG, mPFC, pMTG, and TPJ. DCM analysis showed that while the functional connectivity from IFG to MFG was commonly involved in processing concessive and causal relations, functional connectivities from pMTG to IFG and from pMTG to TPJ were involved in processing causal and concessive relations, respectively. First-person narratives are more attentively and emotionally engaging than third-person narratives. This study examined whether and how personal perspective modulates counterfactual processing. Participants read counterfactual and causal conditionals written from the first-person or third-person perspective (e.g., If/Because I/he had read enough literature before, I/he would have finished my/his thesis easily.), followed by factual consequences that contained a critical word either consistent or inconsistent with preceding contexts (e.g., Therefore, when I/he was about to defend the thesis I/he felt panicked/confident). In both perspectives, inconsistent words showed a prolonged N400 vs. consistent words in the counterfactual condition, but a larger P600 in the causal condition. The critical word showed a larger P600 in the first- than the third-person condition in counterfactual scenarios, but not in causal scenarios. These findings suggest that personal perspective exerts different influences on counterfactual processing, presumably by modulating the amount of attentional resources involved. Abstract: There have been a number of behavioral and neural studies on the processing of syntactic gender and number agreement information, marked by different morpho-syntactic features during sentence comprehension. By using the event-related potential (ERP) technique, the present study investigated whether the processing of semantic gender information and the processing of notional number information can be differentiated and to what extent they might interact during Chinese pronoun resolution. The pronoun (with singular form in Experiment 1 and with plural form in Experiment 2) in a sentence matched its antecedent or mismatched it with respect to either biological gender or notional number or both. While the number mismatch elicited a P600 effect starting from 550ms (for singular pronoun) or 400ms (for plural pronoun) post-onset of the pronoun, the gender mismatch elicited an earlier (for singular) and larger (for both singular and plural) P600 effect. More importantly, the double mismatch produced a P600 effect identical to the effect elicited by the single gender mismatch. These results demonstrate that biological gender information and notional number information are processed differentially and have different processing priorities during Chinese pronoun resolution. Abstract: Mesh is an important and popular interconnection network topology for large parallel computer systems. A mesh can be divided into submeshes to obtain the upper bounds on the connection probability for the mesh. Combinatorial techniques are used to get closer upper bounds on the connection probability for 2-D meshes compared with the existing upper bounds we have known. Simulation results of meshes of various sizes show that our upper bounds are close to the exact connection probability. The combinatorial methods and tools used in this paper can be used to study the connection probabilities for other networks. [Copyright &y& Elsevier]

Abstract: A novel transglycosylating β-galactosidase was purified from Enterobacter agglomerans B1. It was a homodimer of ∼248kDa. The optimal pH and temperature for oNPGal hydrolysis were 7.5–8.0 and 37–40°C, respectively. The K m values for oNPGal and lactose were 0.06 and 114mM, respectively. The enzyme produced galacto-oligosaccharides in a 38% yield at the lactose concentration of 12.5% (w/v). When using oNPGal as donor, the enzyme was able to catalyze glycosyl transfer to a series of acceptors, including hexose, pentose, β- or α-disaccharides, hexahydroxy alcohol, cyclitol, and aromatic glycosides. This suggested the enzyme to be a potential synthetic tool for preparing galactose-containing chemicals. The gene encoding this enzyme was cloned by degenerate PCR and TAIL-PCR. It revealed an ORF of 3090 nucleotides encoding a 1029 amino-acid protein, which had been expressed in Escherichia coli. Transferase activities in both recombinant and natural enzymes were similar. Cyprinid herpesvirus 2 (CyHV-2) is the causative pathogen of herpesviral haematopoietic necrosis disease, which has caused huge economic losses to aquaculture industry in China. In this study, nine truncated CyHV-2 membrane glycoproteins (ORF25, ORF25C, ORF25D, ORF30, ORF124, ORF131, ORF136, ORF142A, ORF146) and a GFP reporter protein were respectively expressed using baculovirus surface displaying system. Western blot showed that the proteins were successfully packaged in the recombinant virus particles. In baculovirus transduced gibel carp kidney cells, the target proteins were expressed and displayed on the fish cell surface. Healthy gibel carp were immunized by immersion with the recombinant baculoviruses and the fish treated with phosphate-buffered saline (PBS) were served as mock group. The expression of interleukin-11 (IL-11), interferon α (IFNα) and a complement component gene C3 were significantly up-regulated in most experimental groups, and interferon γ (IFNγ) expression in some groups were also induced after immunization. Subsequently, the immunized gibel carp were challenged by intraperitoneal injection of CyHV-2 virus. All the immunized groups exhibited reduced mortality after CyHV-2 challenge. In the groups immunized with baculoviruses displaying and expressing ORF25, ORF25C and ORF146, the relative percentage survival values reached 83.3%, 87.5% and 70.8%, respectively. Podocyte injury plays a critical role in diabetic kidney disease (DKD). Our previous work demonstrated a protective role of tyrosine-protein kinase receptor TYRO3 in glomerular disease; However, the downstream signaling of TYRO3 remains unclear. Our data showed that genetic ablation of tyro3 in zebrafish recapitulated a nephrotic syndrome phenotype. TYRO3 expression was suppressed by high glucose and TGF-β, which may contribute to the decreased TYRO3 expression in progressive DKD. Moreover, knockdown of TYRO3 expression with siRNA induced podocytes apoptosis and cytoskeleton rearrangement. Further study revealed that TYRO3 conferred antiapoptotic effects through the activation of JNK/c-jun-P53 in podocytes. Our results revealed a novel signaling module of TYRO3 in podocyte homeostasis, which provides a new molecular insight of TYRO3 effect in podocyte protection. Codon bias at the 5′ terminal of coding sequence (CDS) is known to be distinct from the rest of the CDS. A number of events occur in this short region to regulate early translation elongation and co-translational translocation. In the genes encoding secretory proteins, there is a special signal sequence which has a higher occurrence of rare codons. In this study, we analyzed codon bias of secretory genes in several eukaryotes. The results showed that secretory genes in the species except mammals had a higher occurrence of rare codons in the 5′ terminal of CDS, and the bias was greater than the same region of non-secretory genes. GO analysis revealed that secretory genes containing rare codon clusters in different regions were responsible for various roles in gene functions. Moreover, codon bias in the region encoding the hydrophobic region of protein is similar in secretory and non-secretory genes, indicating that codon bias in secretory genes was partly influenced by amino acid bias. Rare codon clusters are found more frequently in specific regions, and continuous rare codons are not favoured probably because they will increase the probability of ribosome collision and drop-off. Based on ribosome profiling data, there is no significant difference in the average translation efficiencies between rare and optimal codons. Higher ribosomal density in the 5′ terminal may result from ribosome pausing which could be involved in different translation events. These findings collectively provided rich information on codon bias in secretory genes, which may shed light on the co-effect of codon bias, mRNA structure and tRNA abundance in translational regulations. Dysregulation of mammalian target of rapamycin (mTOR) in hepatocellular carcinoma (HCC) represents a valuable treatment target. Recent studies have developed a highly-selective and potent mTOR kinase inhibitor, CZ415. Here, we showed that nM concentrations of CZ415 efficiently inhibited survival and induced apoptosis in HCC cell lines (HepG2 and Huh-7) and primary-cultured human HCC cells. Meanwhile, CZ415 inhibited proliferation of HCC cells, more potently than mTORC1 inhibitors (rapamycin and RAD001). CZ415 was yet non-cytotoxic to the L02 human hepatocytes. Mechanistic studies showed that CZ415 disrupted assembly of mTOR complex 1 (mTORC1) and mTORC2 in HepG2 cells. Meanwhile, activation of mTORC1 (p-S6K1) and mTORC2 (p-AKT, Ser-473) was almost blocked by CZ415. In vivo studies revealed that oral administration of CZ415 significantly suppressed HepG2 xenograft tumor growth in severe combined immuno-deficient (SCID) mice. Activation of mTORC1/2 was also largely inhibited in CZ415-treated HepG2 tumor tissue. Together, these results show that CZ415 blocks mTORC1/2 activation and efficiently inhibits HCC cell growth in vitro and in vivo . [ABSTRACT FROM AUTHOR]

Abstract: Recombinant expression systems differ in the type of glycosylation they impart on expressed antigens such as the human immunodeficiency virus type 1 (HIV-1) envelope glycoproteins, potentially affecting their biological properties. We performed head-to-head antigenic, immunogenic and molecular profiling of two distantly related Env surface (gp120) antigens produced in different systems: (a) mammalian (293 FreeStyle™ cells; 293F) cells in the presence of kifunensine, which impart only high-mannose glycans; (b) insect cells (Spodoptera frugiperda, Sf9), which confer mainly paucimannosidic glycans; (c) Sf9 cells recombinant for mammalian glycosylation enzymes (Sf9 Mimic™), which impart high-mannose, hybrid and complex glycans without sialic acid; and (d) 293F cells, which impart high-mannose, hybrid and complex glycans with sialic acid. Molecular models revealed a significant difference in gp120 glycan coverage between the Sf9-derived and wild-type mammalian-cell-derived material that is predicted to affect ligand binding sites proximal to glycans. Modeling of solvent-exposed surface electrostatic potentials showed that sialic acid imparts a significant negative surface charge that may influence gp120 antigenicity and immunogenicity. Gp120 expressed in systems that do not incorporate sialic acid displayed increased ligand binding to the CD4 binding and CD4-induced sites compared to those expressed in the system that do, and imparted other more subtle differences in antigenicity in a gp120 subtype-specific manner. Non-sialic-acid-containing gp120 was significantly more immunogenic than the sialylated version when administered in two different adjuvants, and induced higher titers of antibodies competing for CD4 binding site ligand–gp120 interaction. These findings suggest that non-sialic-acid-imparting systems yield gp120 immunogens with modified antigenic and immunogenic properties, considerations that should be considered when selecting expression systems for glycosylated antigens to be used for structure–function studies and for vaccine use.
