Osteosarcoma (OS) is a malignant tumor with an extremely poor prognosis, especially in progressive patients. Immunotherapy based on immune checkpoint inhibitors (ICIs) is considered to be a promising treatment option for OS. Due to tumor heterogeneity, only a minority of patients benefit from immunotherapy. Therefore, it is urgent to explore a model that can accurately assess the response of OS to immunotherapy. In this study, we obtained the single-cell RNA sequencing datasets of OS patients from public databases and defined 34 cell clusters by dimensional reduction and clustering analysis. PTPRC was applied to identify immune cell clusters and nonimmune cell clusters. Next, we performed clustering analysis on the immune cell clusters and obtained 25 immune cell subclusters. Immune cells were labeled with CD8A and CD8B to obtain CD8+ T cell clusters. Meanwhile, we extracted the differentially expressed genes (DEGs) of CD8+ T cell clusters and other immune cell clusters. Furthermore, we constructed a prognostic model (CD8-DEG model) based on the obtained DEGs of CD8+ T cells, and verified the excellent predictive ability of this model for the prognosis of OS. Moreover, we further investigated the value of the CD8-DEG model. The results indicated that the risk score of the CD8-DEG model was an independent risk factor for OS patients. Finally, we revealed that the risk score of the CD8-DEG model correlates with the immune profile of OS and can be used to evaluate the response of OS to immunotherapy. In conclusion, our study revealed the critical role of CD8 cells in OS. The risk score model based on CD8-DEGs can provide guidance for prognosis and immunotherapy of OS. [ABSTRACT FROM AUTHOR]

The helium coolant in the primary circuit of the high-temperature gas-cooled reactor (HTGR) contains traces of impurities, which can induce the corrosion of superalloys when exposed to elevated temperatures. The superalloy damage caused by the corrosion could threaten the safe operation of the reactor. In this work, the corrosion behavior of a representative superalloy (chromium-rich iron base alloy Incoloy 800H) was investigated under the impure helium at different typical temperatures of HTGR. An experimental setup developed for studying the high-temperature corrosion of superalloys was used to investigate the chemical reactions and corrosion behaviors of Incoloy 800H. It was found that CO2 is an important oxygen source in the reaction with chromium, and CO is released as the product. In addition, the observation and computation of the critical temperature (TC) of the reaction between CO2 and carbon in the alloy show that TC is much lower than that (TA) of the microclimate reaction, which indicates that CO2 can protect the scale from destruction. Furthermore, the slight decarbonization of the alloy was found above TC. Also, a model developed by the thermodynamic analysis was proposed to explain the mechanism of slight decarbonization and predict the critical temperature when the CO2-C reaction occurs. Objective. To systematically evaluate the efficacy, safety, recovery speed, and long-term visual quality of excimer laser corneal refractive surgery and to provide evidence-based medicine for the promotion and use of excimer laser corneal refractive surgery. Methods. Randomized controls on excimer laser refractive surgery in Web of science, PubMed, EMBASE, ScienceDirect, Cochrane Library, China Knowledge Network (CNKI), China VIP Database, Wan Fang Database, and China Biomedical Literature Database (CBM) were searched by the computer. Randomized controlled trial (RCT) data were extracted independently by two researchers, and the risk of bias of each included RCT was assessed according to the Cochrane Handbook 5.1.0 criteria. Meta-analysis of the collected data was performed using RevMan5.4 statistical software. Results. In the end, 9 high-quality literatures were included, with a total of 4366 samples, and meta-analysis was used. There was no significant difference in uncorrected visual acuity WMD after excimer laser keratorefractive surgery, but there was a statistically significant difference in WMD in the safety of excimer laser keratorefractive surgery. The results of uncorrected visual acuity (close) indicated the following: Chi2 = 13.56, DF = 5, P = 0.02, and I2 = 100%; the results of uncorrected visual acuity (distance) indicated the following: Chi2 = 34.44, DF =5 (P < 00000), and I2 = 85%; the results of best corrected visual acuity (myopia) indicated the following: Chi2 = 0.65, DF = 3, P = 088 > 0.05, and I2 = 0%; the results of best corrected visual acuity (hyperopia) indicated the following: Chi2 = 1.80, DF = 3, P = 0.61 > 0.05, and I2 = 0%. Conclusion. Excimer laser corneal refractive surgery is safe and effective, with faster recovery and better long-term visual acuity treatment effect. At present, FPGA (field-programmable gate array) architecture has made great progress in the requirements of hardware volume, which can meet common needs. However, for the increasing number of resources, it is difficult to significantly reduce the delay of process mapping. Therefore, this paper proposes FDMAP (fit descending map) algorithm from the perspective of the LUT number to reduce the delay. This paper proposes a method of FPGA mapping and debugging for heterogeneous multicore high-performance processors based on isomorphic symmetric FPGA architecture, which effectively utilizes the architectural features of heterogeneous multicore processors and the symmetric features of isomorphic FPGA, divides FPGA functions from top to bottom in a hierarchical way, and constructs FPGA architecture from bottom to top. Using differential bridge and adaptive delay adjustment sampling technology, combined with the embedded virtual logic analyzer debugging tool, FPGA architecture can be lightened and deployed quickly. Multicore complementary core-to-core replacement simulation mapping methods such as debug shells can be used to effectively complete the mapping of the target's high-performance heterogeneous multicore processor to the entire SOC (system on-ship) chip system-level FPGA. In the aspect of algorithm, the fdmap algorithm is mainly implemented, and the low latency mapping of resources is realized with FPGA architecture. In order to verify the effectiveness of mapping the fdmap algorithm, this paper compares the fdmap algorithm with the vector VM algorithm. The research shows that when the wavelength resolution is 7 pm and the temperature error is less than 1°C, the shell is debugged, and 10 mapping examples are simulated with the fdmap algorithm. In the experiment, the LUT with the most critical 20% is selected, and the closed value of the LUT search type is set to 0.86. Compared with the original data, the number of LUTs increased by 15.2%, and the criticality decreased by 35.21%. Compared with the vector VM algorithm with the biggest gap, the number of LUTs decreased by 14.25%, the criticality improved by 14.21%, and the overall delay decreased by 65%. Therefore, the isomorphic symmetric FPGA architecture proposed in this paper can improve the structural criticality and significantly reduce the latency while reducing the number of LUTs. Robot will be used in the grinding industry widely to liberate human beings from harsh environments. In the grinding process, optimal trajectory planning will not only improve the processing quality but also improve the machining efficiency. The aims of this study are to propose a new algorithm and verify its efficiency in achieving the optimal trajectory planning of the grinding robot. An objective function has been defined terms of both time and jerk. Improved whale optimization algorithm (IWOA) is proposed based on whale optimization algorithm (WOA) and differential evolution algorithm (DE). Mutation operation and selection operation of DE are imitated in the part of initialization to process the population initialized by WOA, and then, the search tasks of WOA are performed. Motion with a constant velocity of the end-effector is considered during fine grinding. The continuity of acceleration and velocity will be achieved by minimizing jerk, and at the same time, smooth robot movement can be obtained. Cubic spline interpolation is implemented. A six-axis industrial robot is used for this research. Results show that optimal trajectory planning based on IWOA is more efficient than others. The clinical value of carbohydrate antigen (CA) 19-9 in gastric cancer is controversial. We evaluated the clinicopathologic and prognostic value of CA 19-9 in gastric cancer. Methods. A literature search was conducted in PubMed and Embase databases. Odds ratios (ORs), risk ratios (RR), hazard ratios (HRs), and 95% confidence intervals (CIs) were used as effect measures. Results. Thirty-eight studies were included. Results showed that there were significant differences in the incidence of high CA 19-9 levels between stages III/IV and I/II groups (OR = 3.36; 95% CI = 2.34-4.84), the pT3/T4 and pT1/T2 groups (OR = 2.40; 95% CI = 1.60-3.59), the lymph node-positive and node-negative groups (OR = 2.91; 95% CI = 2.21-3.84), the metastasis-positive and metastasis-negative groups (OR = 2.76; 95% CI = 1.12-6.82), and vessel invasion-positive and invasion-negative groups (OR = 1.66; 95% CI = 1.11-2.48). Moreover, CA 19-9 was significantly associated with poor overall survival (HR = 1.83; 95% CI = 1.56-2.15), disease-free survival (HR = 1.85; 95% CI = 1.16-2.95), and disease-specific survival (HR = 1.33; 95% CI = 1.10-1.60) in gastric cancer. Influenza A (H1N1) virus was the most common cause of human influenza (flu) in 2009. We report a patient infected with influenza A virus who showed improvement in his pulmonary inflammation and fibrosis progression after switching to Chinese herbal medicine (CHM) therapy. Although antiviral drug and corticosteroids were administered, some disturbing respiratory illness and immune imbalance attributed to corticosteroids taken prompted him to seek for CHM therapy. After CHM treatment for three months, significant recovery in chest radiograph was noted, and the result of subsequent pulmonary function test showed improvement. [ABSTRACT FROM AUTHOR]

The objective of this clinical study is to examine the effects of a Chinese herbal medicine formula (Jia Wei Liu Jun Zi Tang: JWLJZT) on motor and non-motor symptoms, and on complications of conventional therapy in idiopathic Parkinson's disease (PD), using an add-on design. Fifty-five patients with PD were randomly allocated to receive either Chinese herbal medicine or placebo for 24 weeks. Primary outcome measure was the 39-item Parkinson's Disease Questionnaire (PDQ-39). Secondary outcome measures included the Unified Parkinson's Disease Rating Scale (UPDRS), Short-Form-36 Health Survey (SF-36), Geriatric Depression Scale (GDS), home diaries, and a range of category rating scales. JWLJZT resulted in a significant improvement in the UPDRS IVC when compared with placebo at 12 weeks (P = .039) and 24 weeks (P = .034). In addition, patients in the Chinese herbal medicine group also showed significant improvement in PDQ-39 communication scores at 12 weeks (P = .024) and 24 weeks (P = .047) when compared with the placebo group. There were no significant differences between treatment and control groups for SF-36 variables, GDS score or the mean daily "on-off" time. One case of mild diarrhea was noted in the treatment group. The findings suggest that JWLJZT can relieve some non-motor complications of conventional therapy and improve the communication ability in patients with PD. The results of this pilot study warrant larger multi-center clinical studies to assess long-term efficacy and tolerability of JWLJZT, and to elucidate the mechanisms by which it affects PD function.
