This paper summarizes simulation based technical feasibility and cost benefit analyses of a range of off-the-shelf electrical losses reduction intervention solutions in the Isle of Wight (IoW) 11 kV distribution network in the United Kingdom (UK), assessed over a period of 45 years. Based on presented results, the paper provides a brief commentary on the merits, drawbacks, risks and recommendations associated with each considered network intervention. Among considered network interventions, the transformer auto stop-start scheme, where one of the transformers among a pair at a primary substation would be switched depending on the substation's varying load, was found to be the optimal solution, i.e., in reducing technical losses significantly and may also have a reasonable Net Present Value (NPV) with a good rate of Return on Investment (RoI) when fully implemented. The British seismologist John Milne (1850-1913) is a dominant and celebrated figure in historical accounts of seismology. In this paper I use evidence from seismographs preserved in the collections of the Science Museum Group to reconsider his contribution, particularly to the progress of international collaboration in seismology. These instruments were installed at Eskdalemuir Observatory in Dumfriesshire, Scotland between 1908 and 1925; not only were they situated far from Milne's private observatory on the Isle of Wight, but they represented examples of Russian, Japanese and German seismograph design as well as Milne's own. No other site in Britain boasted such an international suite of earthquake instruments. Together, they reveal an alternative vantage-point from which to view his influence. By attending closely to the instruments' provenance, I show how the collection can be a powerful tool for decentring the hagiography that surrounds the so-called 'Father of Modern Seismology'. Simple Summary: The family Cydnidae (so-called burrower bugs) contains 76 fossil species primarily assigned to two subfamilies, Amnestinae and Cydninae. Only five fossil species were undoubtedly classified within the subfamily Sehirinae. Eocenocydnus lisi, described from the Late Eocene of the Isle of Wight, United Kingdom, was also tentatively placed in this subfamily. All other burrower bug subfamilies have no described fossil species to date. To verify the systematic position of E. lisi within the family Cydnidae, a parallel, cross-eyed viewing method was used. It is the first case incorporating this method into insect fossil morphological studies. It allows the recognition of many more morphological details than directly viewing a two-dimensional flat image. The fossil burrower bug Eocenocydnus lisi described from the Late Eocene of the Isle of Wight, UK, is analysed using a parallel, cross-eyed viewing method. The species, tentatively placed in the subfamily Sehirinae, is redescribed and its systematic position is discussed. Background: DNA methylation is a biochemical process in which a methyl group is added to the cytosine-phosphate-guanine (CpG) site on DNA molecules without altering the DNA sequence. Multiple CpG sites in a certain genome region can be differentially methylated across phenotypes. Identifying these differentially methylated CpG regions (DMRs) associated with the phenotypes contributes to disease prediction and precision medicine development. Results: We propose a novel DMR detection algorithm, gbdmr. In contrast to existing methods under a linear regression framework, gbdmr assumes that DNA methylation levels follow a generalized beta distribution. We compare gbdmr to alternative approaches via simulations and real data analyses, including dmrff, a new DMR detection approach that shows promising performance among competitors, and the traditional EWAS that focuses on single CpG sites. Our simulations demonstrate that gbdmr is superior to the other two when the correlation between neighboring CpG sites is strong, while dmrff shows a higher power when the correlation is weak. We provide an explanation of these phenomena from a theoretical perspective. We further applied the three methods to multiple real DNA methylation datasets. One is from a birth cohort study undertaken on the Isle of Wight, United Kingdom, and the other two are from the Gene Expression Omnibus database repository. Overall, gbdmr identifies more DMR CpGs linked to phenotypes than dmrff, and the simulated results support the findings. Conclusions: Gbdmr is an innovative method for detecting DMRs based on generalized beta regression. It demonstrated notable advantages over dmrff and traditional EWAS, particularly when adjacent CpGs exhibited moderate to strong correlations. Our real data analyses and simulated findings highlight the reliability of gbdmr as a robust DMR detection tool. Most aircraft in the world are tracked by various surveillance radar systems. Currently there is no legal requirement for light aircraft to be fitted with a transponder; however, this does not mean light aircraft should not be tracked. By adding a cheap, live tracking solution for light aircraft, the safety of low-flying aircraft pilots can be greatly increased. The radio operators who coordinate the aircraft can have an improved understanding of the air traffic and in the event of an emergency, the position of the aircraft can be relayed to emergency services. This paper proposes an approach to use a smartphone as an aircraft transponder to improve the radar tracking capabilities of low-flying aircraft. This study presents a practical and effective approach as well as a prototype implementation. The study includes the development of the three main components: (1) A mobile application that transforms a smartphone into an aircraft transponder; exploiting the GPS functionalities, (2) a desktop application that visualizes the aircraft data in real time on a map, and (3) a backend that bridges the mobile and the desktop application. The ant Tapinoma ibericum Santschi, 1925 is native to the Iberian Peninsula. This species, as well as other species from the Tapinoma nigerrimum complex, could form supercolonies that make these species potentially invasive and could give rise to pests. Recently a mature colony from this species has been found in the Isle of Wight (United Kingdom). Mitogenomes have been used to study the taxonomy, biogeography and genetics of species, improving the development of strategies against pest invasion. However, the number of available mitogenomes from the subfamily Dolichoderinae is still scarce and only two of these mitogenomes belong to Tapinoma species. Herein, the complete mitogenome of T. ibericum is presented in order to increase the molecular information of the genus. The T. ibericum mitogenome, retrieved by Next-Generation Sequencing data, is 15,715 bp in length. It contains the typical set of 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNAs and the A + T-rich control region. Comparisons of the T. ibericum mitogenome with other dolichoderine mitogenomes revealed the existence of four gene rearrangements in relation with the ancestral insect mitogenome. One of these rearrangements, involving the tRNA-Ile, tRNA-Gln and tRNA-Met genes, was found in most of the analyzed ant mitogenomes. Probably this rearrangement was an ancestral or plesiomorphic character in Formicidae. Interestingly, another rearrangement that affects to tRNA-Trp, tRNA-Cys and tRNA-Tyr genes was found only in Tapinoma species. This change could be a synapomorphic character for the genus Tapinoma, and could be used as a phylogenetic marker. Additionally, a phylogenetic analysis was performed using the protein-coding gene sequences from available Dolichoderinae mitogenomes, as well as mitogenomes from representative species from other Formicidae subfamilies. Eotyrannus lengi Hutt et al., 2001 from the Lower Cretaceous Wessex Formation (part of the Wealden Supergroup) of the Isle of Wight, southern England, is described in detail, compared with other theropods, and evaluated in a new phylogenetic analysis. Eotyrannus is represented by a single individual that would have been c. 4.5 m long; it preserves the anterior part of the skull, a partial forelimb and pectoral girdle, various cervical, dorsal and caudal vertebrae, rib fragments, part of the ilium, and hindlimb elements excluding the femur. Lack of fusion with regard to both neurocentral and sacral sutures indicates subadult status. Eotyrannus possesses thickened, fused, pneumatic nasals with deep lateral recesses, elongate, tridactyl forelimbs and a tyrannosaurid-like scapulocoracoid. The short preantorbital ramus of the maxilla and nasals that are approximately seven times longer than they are wide show that Eotyrannus was not longirostrine. A posterodorsally inclined ridge on the ilium's lateral surface fails to reach the dorsal margin: a configuration seen elsewhere in Juratyrant. Eotyrannus is not arctometatarsalian. Autapomorphies include the presence of curving furrows on the dentary, a block-like humeral entepicondyle, and a distoproximally aligned channel close to the distolateral border of the tibia. Within Tyrannosauroidea, E. lengi is phylogenetically intermediate between Proceratosauridae and Yutyrannus and the clade that includes Xiongguanlong, Megaraptora, Dryptosaurus and Tyrannosauridae. We do not find support for a close affinity between Eotyrannus and Juratyrant. Our analysis supports the inclusion of Megaraptora within Tyrannosauroidea and thus increases Cretaceous tyrannosauroid diversity and disparity. A proposal that Eotyrannus might belong within Megaraptora, however, is based on character states not present in the taxon. Several theropods from the Wessex Formation are based on material that overlaps with the E. lengi holotype but none can be shown to be synonymous with it. [ABSTRACT FROM AUTHOR]

Postcranial elements (cervical, sacral and caudal vertebrae, as well as ilium, rib and limb bone fragments) belonging to a gigantic tetanuran theropod were recovered from the basal unit (the White Rock Sandstone equivalent) of the Vectis Formation near Compton Chine, on the southwest coast of the Isle of Wight. These remains appear to pertain to the same individual, with enormous dimensions similar to those of the Spinosaurus holotype and exceeding those of the largest European theropods previously reported. A combination of features--including the presence of spinodiapophyseal webbing on an anterior caudal vertebra--suggest that this is a member of Spinosauridae, though a lack of convincing autapomorphies precludes the identification of a new taxon. Phylogenetic analysis supports spinosaurid affinities but we were unable to determine a more precise position within the clade weak support for a position within Spinosaurinae or an early-diverging position within Spinosauridae were found in some data runs. Bioerosion in the form of curved tubes is evident on several pieces, potentially related to harvesting behaviour by coleopteran bioeroders. This is the first spinosaurid reported from the Vectis Formation and the youngest British material referred to the clade. This Vectis Formation spinosaurid is unusual in that the majority of dinosaurs from the Lower Cretaceous units of the Wealden Supergroup are from the fluviolacustrine deposits of the underlying Barremian Wessex Formation. In contrast, the lagoonal facies of the upper Barremian--lower Aptian Vectis Formation only rarely yield dinosaur material. Background: Young adult offenders represent a third of the UK prison population and are at risk of poor health outcomes including drug and alcohol misuse, self-harm and suicide. Court diversion interventions aim to reduce the negative consequences of formal criminal justice sanctions and focus resources on addressing the root causes of offending. Although diversions are widely used, evidence of their effectiveness has not yet been established. Hampshire Constabulary, working together with local charities, have developed the Gateway programme, an out-of-court intervention aimed at improving the life chances of young adults. Issued as a conditional caution, participants undertake a health and social care needs assessment, attend workshops encouraging analysis of own behaviour and its consequences and agree not to re-offend during the 16-week caution.Methods: This is a pragmatic, multi-site, parallel-group, superiority randomised controlled trial with a target sample size of 334. Participants are aged 18-24, reside in Hampshire and Isle of Wight and are being questioned for an eligible low-level offence. Police investigators offer potential participants a chance to receive the Gateway caution, and those interested are also invited to take part in the study. Police officers obtain Stage 1 consent and carry out an eligibility check, after which participants are randomised on a 1:1 basis either to receive Gateway or follow the usual process, such as court appearance or a different conditional caution. Researchers subsequently obtain Stage 2 consent and collect data at weeks 4 and 16, and 1 year post-randomisation. The primary outcome is the Warwick-Edinburgh Mental Well-being Scale (WEMWBS). Secondary outcomes include health status, alcohol and drug use, recidivism and resource use. The primary analysis will compare the WEMWBS score between the two groups at 12 months.Discussion: This pioneering trial aims to address the evidence gap surrounding diversion in 18-24-year-olds. In the area of greenhouse operation, yield prediction still relies heavily on human expertise. This paper proposes an automatic tomato yield predictor to assist the human operators in anticipating more effectively weekly fluctuations and avoid problems of both overdemand and overproduction if the yield cannot be predicted accurately. The parameters used by the predictor consist of environmental variables inside the greenhouse, namely, temperature, CO2, vapour pressure deficit (VPD), and radiation, as well as past yield. Greenhouse environment data and crop records from a large scale commercial operation,Wight Salads Group (WSG) in the Isle of Wight, United Kingdom, collected during the period 2004 to 2008, were used to model tomato yield using an Intelligent System called "Evolving Fuzzy Neural Network" (EFuNN). Our results show that the EFuNN model predicted weekly fluctuations of the yield with an average accuracy of 90%. The contribution suggests that the multiple EFUNNs can be mapped to respective task-oriented rule-sets giving rise to adaptive knowledge bases that could assist growers in the control of tomato supplies and more generally could inform the decision making concerning overall crop management practices. Background. Recent UK government documents have stated that, within the National Health Service, consumers should be more involved in decision-making. This study considered the technique of discrete choice conjoint analysis (DCCA) for eliciting community views regarding the importance of reducing waiting times. More specifically, the study aimed to establish whether residents living on the Isle of Wight are prepared to travel to the mainland for elective surgery where waiting times are shorter but travel costs may be greater, and, further, if residents are willing to travel, what reduction in waiting time and increase in travel costs would be acceptable. Balanomorph cirripedes from the Eocene-Oligocene of the Hampshire Basin (United Kingdom) and the Middle Eocene of the Cotentin Peninsula, Manche (France) are described. A new genus, Vectibalanus, is founded, with the type species Balanus unguiformis J. de C. Sowerby, 1846; assigned to this are also Balanus erisma J. de C. Sowerby, 1846 and Vectibalanus mortoni sp. nov. In addition, a new species of Lophobalanus Zullo, 1984, L. fresvillensis sp. nov., is described. This is the first record of that genus from outside the eastern USA and the oldest species known to date. Cladistic analysis of 24 morphological characters suggests that Vectibalanus unguiformis is sister taxon to a group comprising the most derived balanomorph taxa, and thus represents an important transition in the evolution of the group, with the initiation of development of a complex parietal wall structure. Vectibalanus unguiformis was evidently adapted to low salinity habitats (10-30 ppt), and is the oldest known brackish water barnacle. The other species (V. erisma, V. mortoni sp. nov.) occupied more clearly marine environments (>30 ppt). Balanomorph barnacles appeared simultaneously in the Priabonian (Upper Eocene) of the Gulf and Atlantic seaboards of the USA and northwest Europe, which probably represents a northerly migration from Tethys. Background: Previous studies have shown that DNA methylation (DNAm) is associated with body mass index (BMI). However, it is unknown whether DNAm at pre-adolescence is associated with BMI status transition from pre- to post-adolescence. In the Isle of Wight (IoW) birth cohort, genome-wide DNA methylation in whole blood was measured using Illumina Infinium Human450 and EPIC BeadChip arrays in n = 325 subjects, and pre- to post-adolescence BMI transition was classified into four groups: (1) normal to normal, (2) normal to overweight or obese, (3) overweight or obese to normal, and (4) persistent overweight or obese. We used recursive random forest to screen genome-wide Cytosine-phosphate-Guanine (CpG) sites with DNAm potentially associated with BMI transition for each gender, and the association of BMI status transition with DNAm at an earlier age was assessed via logistic regressions. To evaluate gender specificity, interactions between DNAm and gender were included in the model. Findings in the IoW cohort were further tested in an independent cohort, the Avon Longitudinal Study of Parents and Children (ALSPAC). Results: In total, 174 candidate CpGs were selected including CpGs from screening and CpGs previously associated correctionally with BMI in children and adults. Of these 174 CpGs, pre-adolescent DNAm of 38 CpGs in the IoW cohort was associated with BMI status transition, including 30 CpGs showing gender-specific associations. Thirteen CpGs showed consistent associations between the IoW cohort and the ALSPAC cohort (11 of which were gender-specific). Background: Adolescence is a significant period for the gender-dependent development of lung function. Prior studies have shown that DNA methylation (DNA-M) is associated with lung function and DNA-M at some cytosine-phosphate-guanine dinucleotide sites (CpGs) changes over time. This study examined whether changes of DNA-M at lung-function-related CpGs are associated with changes in lung function during adolescence for each gender, and if so, the biological significance of the detected CpGs.Methods: Genome-scale DNA-M was measured in peripheral blood samples at ages 10 (n = 330) and 18 years (n = 476) from the Isle of Wight (IOW) birth cohort in United Kingdom, using Illumina Infinium arrays (450 K and EPIC). Spirometry was conducted at both ages. A training and testing method was used to screen 402,714 CpGs for their potential associations with lung function. Linear regressions were applied to assess the association of changes in lung function with changes of DNA-M at those CpGs potentially related to lung function. Adolescence-related and personal and family-related confounders were included in the model. The analyses were stratified by gender. Multiple testing was adjusted by controlling false discovery rate of 0.05. Findings were further examined in two independent birth cohorts, the Avon Longitudinal Study of Children and Parents (ALSPAC) and the Children, Allergy, Milieu, Stockholm, Epidemiology (BAMSE) cohort. Pathway analyses were performed on genes to which the identified CpGs were mapped.Results: For females, 42 CpGs showed statistically significant associations with change in FEV1/FVC, but none for change in FEV1 or FVC. No CpGs were identified for males. In replication analyses, 16 and 21 of the 42 CpGs showed the same direction of associations among the females in the ALSPAC and BAMSE cohorts, respectively, with 11 CpGs overlapping across all the three cohorts. Through pathway analyses, significant biological processes were identified that have previously been related to lung function development.Conclusions: The detected 11 CpGs in all three cohorts have the potential to serve as the candidate epigenetic markers for changes in lung function during adolescence in females. Objective: The aims of this study are to examine the potential association between grandmaternal BMI and grandchild's birth weight (BW) and whether maternal BW and BMI mediate this association.Methods: Data of 209 grandmother-mother pairs and 355 grandchildren from the Isle of Wight birth cohort in the UK were analyzed using path analysis.Results: An indirect effect of grandmaternal BMI on increasing grandchild's BW was mediated by maternal BW and BMI at age 18 years (indirect effects: β = 2.3 g/unit increase in grandmaternal BMI via maternal BW and β = 4.4 g via maternal BMI; P = 0.04). These two mediating effects of maternal BW and BMI confounded one another. Grandmaternal smoking during pregnancy had an indirect effect on decreasing grandchild's BW, dependent on maternal smoking during pregnancy and BW (indirect effects: β = -36.1 g compared with nonsmoking grandmothers via maternal smoking during pregnancy and β = -27.2 g via maternal BW; P = 0.005). Neither direct effect between grandmaternal BMI and grandchild's BW nor that between grandmaternal smoking during pregnancy and grandchild's BW was statistically significant.Conclusions: Larger grandmaternal BMI indirectly increased grandchild's BW via maternal BW and BMI. Archaeological evaluations in advance of the installation of a new garden in the area previously known as the Privy Garden, now the Princess Beatrice garden, at Carisbrooke Castle revealed the first prehistoric feature from the site and established a northern continuation of a Conquest-period ringwork ditch. The main discovery, a 12th century high status, masonry, cellared structure, probably a chamber block of two-storeys, is identified as the 'chamber next the chapel' mentioned in a 13th century account. Further structures added in the 13th-15th century are discussed. The demolition of these buildings in the late 16th century and the re-ordering of the site are seen in the context of major works by Sir George Carey. Environmental sampling, particularly of yard surfaces and a midden developed to the east and south of the chamber block, has greatly enhanced understanding of the medieval and early post-medieval diet and economy at the castle. Background: Patients often report adverse reactions to wheat. Interpretation of sensitization to wheat pollen and flour with/without sensitization to grass pollen is a clinical problem. Aim: We set out to determine the prevalence of wheat allergy in a birth cohort (10/11 year olds) and investigate the usefulness of performing skin prick tests (SPT), specific IgE tests and component resolved diagnostics to wheat pollen and flour. Methods: The Food Allergy and Intolerance Research (FAIR) birth cohort included babies born on the Isle of Wight (UK) between September 2001–August 2002 (n = 969). Children were followed up at 1, 2, 3 and 10/11 years. 588 children had SPTs to wheat pollen and grass during the 10 year follow‐up. 294 children underwent further SPT to wheat flour and 246 had specific IgE testing to wheat and grass. Results: Eight children underwent oral food challenges (OFC). We diagnosed 0.48 % (4/827; 95 % CI 0–1 %) children with wheat allergy based on OFC. 16.3 % (96/588) were sensitized to grass pollen, 13.4 % (79/588) to wheat pollen; 78 % (75/96) sensitized to both. Only one child was sensitized to wheat flour and wheat pollen, but not grass pollen. For specific IgE, 15.0 % (37/246) and 36.2 % (89/246) were sensitized to wheat and grass pollen, with 40.5 % (36/89) sensitized to both. Of the 37 children sensitized to wheat, 3 (8.1 %) were sensitized to omega 5 gliadin, 1 (2.7 %) to wheat lipid transfer protein and 1 to wheat gliadin. Conclusion: Clinicians should be aware of the high level of cross‐sensitization when performing tests to wheat and grass pollen i.e. sensitisation to wheat specific IgE and wheat pollen SPT should be assessed in the presence of grass pollen SPT and/or specific IgE. Background: Pterosaurs have been known from the Cretaceous sediments of the Isle of Wight (southern England, United Kingdom) since 1870. We describe the three-dimensional pelvic girdle and associated vertebrae of a small near-adult pterodactyloid from the Atherfield Clay Formation (lower Aptian, Lower Cretaceous). Despite acknowledged variation in the pterosaur pelvis, previous studies have not adequately sampled or incorporated pelvic characters into phylogenetic analyses. Methodology/Principal Findings: The new specimen represents the new taxon Vectidraco daisymorrisae gen. et sp. nov., diagnosed by the presence of a concavity posterodorsal to the acetabulum and the form of its postacetabular process on the ilium. Several characters suggest that Vectidraco belongs to Azhdarchoidea. We constructed a pelvis-only phylogenetic analysis to test whether the pterosaur pelvis carries a useful phylogenetic signal. Resolution in recovered trees was poor, but they approximately matched trees recovered from analyses of total evidence. We also added Vectidraco and our pelvic characters to an existing total-evidence matrix for pterosaurs. Both analyses recovered Vectidraco within Azhdarchoidea. Conclusions/Significance: The Lower Cretaceous strata of western Europe have yielded members of several pterosaur lineages, but Aptian pterosaurs from western Europe are rare. With a pelvis length of 40 mm, the new animal would have had a total length of c. 350 mm, and a wingspan of c. 750 mm. Barremian and Aptian pterodactyloids from western Europe show that small-bodied azhdarchoids lived alongside ornithocheirids and istiodactylids. This assemblage is similar in terms of which lineages are represented to the coeval beds of Liaoning, China; however, the number of species and specimens present at Liaoning is much higher. While the general phylogenetic composition of western European and Chinese communities appear to have been approximately similar, the differences may be due to different palaeoenvironmental and depositional settings. Terpenoid resin is produced by all families and most genera of the order Coniferales (the conifers), and the distribution of terpenes present in most conifer resins is characteristic of the originating family. Analyses of early Cretaceous (Barremian) amber (fossil resin) from the English Wealden, Isle of Wight, southern England, by pyrolysis-gas chromatography-mass spectrometry (Py-GC-MS), indicate a terpene distribution dominated by abietane- and labdane-type terpenes. Similar distributions are observed in some species of the extant family Pinaceae. The Pinaceae are well represented within the Wealden deposits of southern England, by only one (known) species, Pityites solmsii (Seward) Seward, whereas the macro-fossil record of these deposits is dominated by the extinct conifer family Cheirolepidiaceae, for which no resin chemistry has been reported. By analogy with modern materials, it is probable that the ambers found in these deposits are derived from an extinct member of the Pinaceae, but given the absence of evidence concerning the chemotaxonomy of the Cheirolepidiaceae, this family cannot be excluded a priori as a possible paleobotanical source. These ambers may therefore be assigned to either the Pinaceae or to the Cheirolepidiaceae. These samples are the oldest ambers to date to yield useful chemotaxonomic data. [ABSTRACT FROM AUTHOR]

Abstract. A new fossil spider, Vectaraneus yulei gen. et sp. nov., from the Eocene Bembridge Marls Insect Bed of the Isle of Wight, shows internal anatomy, including book lungs and tracheae, preserved by calcium carbonate replacement. The wide, medially positioned, tracheal spiracle and large tracheae which enter the prosoma are adaptations for an amphibious mode of life. The spider is placed in Cybaeidae Simon, 1898, Argyronetinae Menge, 1869, a subfamily which includes the Recent European Water Spider, Argyroneta aquatica (Clerck, 1757). The only previously described Bernbridge Marls spider, Eoatypus woodwardii McCook, 1888, is redescribed; it is unrelated to Veclaraneus. The holotype of Argyroneta antiqua Von Heyden (1859) is redescribed; it is not an Argyroneta. Specimens referred to A. antiqua by Bertkau (1878) probably belong in Argyronetinae Menge, 1869, and this subfamily is emended herein. [ABSTRACT FROM AUTHOR]

Numerical modeling of currents and waves is used throughout the marine energy industry for resource assessment. This study compared the output of numerical flow simulations run both as a standalone model and as a two-way coupled wave–current simulation. A regional coupled flow-wave model was established covering the English Channel using the Delft D-Flow 2D model coupled with a SWAN spectral wave model. Outputs were analyzed at three tidal energy sites: Alderney Race, Big Roussel (Guernsey), and PTEC (Isle of Wight). The difference in the power in the tidal flow between coupled and standalone model runs was strongly correlated to the relative direction of the waves and currents. The net difference between the coupled and standalone runs was less than 2.5%. However, when wave and current directions were aligned, the mean flow power was increased by up to 7%, whereas, when the directions were opposed, the mean flow power was reduced by as much as 9.6%. The D-Flow Flexible Mesh model incorporates the effects of waves into the flow calculations in three areas: Stokes drift, forcing by radiation stress gradients, and enhancement of the bed shear stress. Each of these mechanisms is discussed. Forcing from radiation stress gradients is shown to be the dominant mechanism affecting the flow conditions at the sites considered, primarily caused by dissipation of wave energy due to white-capping. Wave action is an important consideration at tidal energy sites. Although the net impact on the flow power was found to be small for the present sites, the effect is site specific and may be significant at sites with large wave exposure or strong asymmetry in the flow conditions and should thus be considered for detailed resource and engineering assessments. Residential variable energy price schemes can be made more effective with the use of a demand response (DR) strategy along with smart appliances. Using DR, the electricity bill of participating customers/households can be minimised, while pursuing other aims such as demand-shifting and maximising consumption of locally generated renewable-electricity. In this article, a two-stage optimization method is used to implement a price-based implicit DR scheme. The model considers a range of novel smart devices/technologies/schemes, connected to smart-meters and a local DR-Controller. A case study with various decarbonisation scenarios is used to analyse the effects of deploying the proposed DR-scheme in households located in the west area of the Isle of Wight (Southern United Kingdom). There are approximately 15,000 households, of which 3000 are not connected to the gas-network. Using a distribution network model along with a load flow software-tool, the secondary voltages and apparent-power through transformers at the relevant substations are computed. The results show that in summer, participating households could export up to 6.4 MW of power, which is 10% of installed large-scale photovoltaics (PV) capacity on the island. Average carbon dioxide equivalent (CO2e) reductions of 7.1 ktons/annum and a reduction in combined energy/transport fuel-bills of 60%/annum could be achieved by participating households. Background: Adolescence is a period characterized by major biological development, which may be associated with changes in DNA methylation (DNA-M). However, it is unknown to what extent DNA-M varies from pre- to post-adolescence, whether the pattern of changes is different between females and males, and how adolescence-related factors are associated with changes in DNA-M. Methods: Genome-scale DNA-M at ages 10 and 18 years in whole blood of 325 subjects (n = 140 females) in the Isle of Wight (IOW) birth cohort was analyzed using Illumina Infinium arrays (450K and EPIC). Linear mixed models were used to examine DNA-M changes between pre- and post-adolescence and whether the changes were gender-specific. Adolescence-related factors and environmental exposure factors were assessed on their association with DNA-M changes. Replication of findings was attempted in the comparable Avon Longitudinal Study of Parents and Children (ALSPAC) cohort. Results: In the IOW cohort, after controlling for technical variation and cell compositions at both pre- and post-adolescence, 15,532 cytosine–phosphate–guanine (CpG) sites (of 400,825 CpGs, 3.88%) showed statistically significant DNA-M changes from pre-adolescence to post-adolescence invariant to gender (false discovery rate (FDR) = 0.05). Of these 15,532 CpGs, 10,212 CpGs (66%) were replicated in the ALSPAC cohort. Pathway analysis using Ingenuity Pathway Analysis (IPA) identified significant biological pathways related to growth and development of the reproductive system, emphasizing the importance of this period of transition on epigenetic state of genes. In addition, in IOW, we identified 1179 CpGs with gender-specific DNA-M changes. In the IOW cohort, body mass index (BMI) at age 10 years, age of growth spurt, nonsteroidal drugs use, and current smoking status showed statistically significant associations with DNA-M changes at 15 CpGs on 14 genes such as the AHRR gene. For BMI at age 10 years, the association was gender-specific. Findings on current smoking status were replicated in the ALSPAC cohort. Conclusion: Adolescent transition is associated with changes in DNA-M at more than 15K CpGs. Background: To identify novel epigenetic markers of adolescent asthma and replicate findings in an independent cohort, then explore whether such markers are detectable at birth, predictive of early-life wheeze, and associated with gene expression in cord blood. Methods: We performed epigenome-wide screening with recursive random forest feature selection and internal validation in the IOW birth cohort. We then tested whether we could replicate these findings in the independent cohort ALSPAC and followed-up our top finding with children of the IOW cohort. Results: We identified 10 CpG sites associated with adolescent asthma at a 5% false discovery rate (IOW, n = 370), five of which exhibited evidence of associations in the replication study (ALSPAC, n = 720). One site, cg16658191, within HK1 displayed particularly strong associations after cellular heterogeneity adjustments in both cohorts (ORIOW = 0.17, 95% CI 0.04–0.57) (ORALSPAC = 0.57, 95% CI 0.38–0.87). Additionally, higher expression of HK1 (OR = 3.81, 95% CI 1.41–11.77) in cord blood was predictive of wheezing in infancy (n = 82). Conclusion: We identified novel associations between asthma and wheeze with methylation at cg16658191 and the expression of HK1, which may serve as markers of, predictors of, and potentially etiologic factors involved in asthma and early life wheeze.
