Microfluidic systems made out of polydimethylsiloxane (PDMS) offer a platform to mimic vascular flow conditions in model systems at well-defined shear stresses. However, extracellular matrix (ECM) proteins that are physisorbed on the PDMS are not reliably attached under high shear stress conditions, which makes long-term experiments difficult. To overcome this limitation, we functionalized PDMS surfaces with 3-aminopropyltriethoxysilane (APTES) by using different surface activation methods to develop a stable linkage between the PDMS surface and collagen, which served as a model ECM protein. The stability of the protein coating inside the microfluidic devices was evaluated in perfusion experiments with phosphate-buffered saline (PBS) at 10-40 dynes/cm 2 wall shear stress. To assess the stability of cell adhesion, endothelial cells were grown in a multi-shear device over a shear stress range of 20-150 dynes/cm 2 . Cells on the APTES-mediated collagen coating were stable over the entire shear stress range in PBS (pH 9) for 48 h. The results suggest that at high pH values, the electrostatic interaction between APTES-coated surfaces and collagen molecules offer a very promising tool to modify PDMS-based microfluidic devices for long-term endothelialization under high shear stress conditions., (Copyright © 2020 Elsevier B.V. Lignin-carbohydrate complexes, i.e. LCC-48 and LCC-72 were isolated with vibrational ball milling for 48 h and 72 h, respectively. Composite tubular carriers were prepared by casting film formation and crimping with the LCCs and cellulose nanofibrils (CNFs) as raw materials. The structures and chemical properties of different milling time LCCs were analyzed. The carriers were used to culture human coronary artery endothelial cells (HCAEC), and the activities of these cells were examined in vitro. The FT-IR and chemical composition results showed that the biocarriers were composed of lignin moieties and polysaccharides. The SEM and inverted microscope studies revealed that a large number of cells adhered to the porous carriers. HCAEC grown on the LCC-72/CNF carriers outperformed the LCC-48/CNF and control groups in every observed category, including cell proliferation rate and metabolic activity. Endothelial cells (ECs) are critical determinants of vascular homeostasis and inflammation, but transcriptional mechanisms specifying their identities and functional states remain poorly understood. Here, we report a genome-wide assessment of regulatory landscapes of primary human aortic endothelial cells (HAECs) under basal and activated conditions, enabling inference of transcription factor networks that direct homeostatic and pro-inflammatory programs. We demonstrate that 43% of detected enhancers are EC-specific and contain SNPs associated to cardiovascular disease and hypertension. We provide evidence that AP1, ETS, and GATA transcription factors play key roles in HAEC transcription by co-binding enhancers associated with EC-specific genes. We further demonstrate that exposure of HAECs to oxidized phospholipids or pro-inflammatory cytokines results in signal-specific alterations in enhancer landscapes and associate with coordinated binding of CEBPD, IRF1, and NFκB. Collectively, these findings identify cis-regulatory elements and corresponding trans-acting factors that contribute to EC identity and their specific responses to pro-inflammatory stimuli.

Vascular calcification is commonly seen in elderly people, though it can also appear in middle-aged subjects affected by premature vascular aging. The aim of this work is to test the involvement of microvesicles (MVs) produced by senescent endothelial cells (EC) and from plasma of elderly people in vascular calcification. The present work shows that MVs produced by senescent cultured ECs, plus those found in the plasma of elderly subjects, promote calcification in vascular smooth muscle cells. Only MVs from senescent ECs, and from elderly subjects' plasma, induced calcification. This ability correlated with these types of MVs' carriage of: a) increased quantities of annexins (which might act as nucleation sites for calcification), b) increased quantities of bone-morphogenic protein, and c) larger Ca contents. The MVs of senescent, cultured ECs, and those present in the plasma of elderly subjects, promote vascular calcification. The present results provide mechanistic insights into the observed increase in vascular calcification-related diseases in the elderly, and in younger patients with premature vascular aging, paving the way towards novel therapeutic strategies.

Aiming to mimic a blood vessel structurally, morphologically, and mechanically, a sequential electrospinning technique using a small diameter mandrel collector was performed and a three-layered tubular scaffold composed of nanofibers of polycaprolactone, collagen, and poly(l-lactic acid) as inner, intermediate, and outer layers, respectively, was developed. Biological performances of the scaffold in terms of compatibility with blood and endothelial cells were assessed to get some insights into its potential use as a tissue engineered small-diameter vascular replacement compared to an expanded polytetrafluoroethylene vascular graft. Due to direct contact of the blood and endothelial cells with inner surface of the scaffold, polycaprolactone fibers were characterized using SEM, water contact angle measurement, and ATR-FTIR. Despite similar surface wettability of the electrospun scaffold and the expanded polytetrafluoroethylene graft, the three-layered scaffold significantly reduced platelet adhesion and hemolysis ratio compared to expanded polytetrafluoroethylene graft while comparable blood clotting profiles were observed for both electrospun scaffold and expanded polytetrafluoroethylene graft. However, inflammatory response to nanofibrous surface of the scaffold was reduced compared to expanded polytetrafluoroethylene graft. The electrospun scaffold also presented a significantly more supportive substrate for endothelialization than the expanded polytetrafluoroethylene graft. The results described herein suggested that the three-layered scaffold has superior biological properties compared to an expanded polytetrafluoroethylene graft for vascular tissue engineering., (© The Author(s) 2016.)
