Purpose: To investigate the interventional effect of ShensongYangxin (SSYX) on heart failure (HF) using preserved ejection fraction (HFpEF) rat model. Methods: HFpEF rat model was established using abdominal aorta coarctation method and randomly divided into a positive drug control group; SSYX at high, medium, and low dosage groups; and normal control group. After 8 weeks oral treatment of SSYX, echocardiography and cardiac catheterization were used to investigate the effects of SSYX on HFpEF rat cardiac functions, including mean heart rate (HR), left ventricle anterior and posterior wall thicknesses at end diastole (LVAWd + LVPWd), left ventricular internal diameter at end diastole (LVIDd), and left ventricle mass (LVM). Results: SSYX markedly decreased heart weight and improved survival rate (p < 0.01) after 12 weeks of treatment. The expression of NT-proBNP decreased in a dose-dependent manner and was significantly lower in SSYX treatment groups (p < 0.01). Compared with normal control group, expression of CaMK II, PKA and RyR2 was significantly lower (p < 0.005), while expression level of SERCA2a significantly increased after 4 g/kg/day SSYX treatment (p < 0.01). Conclusion: SSYX significantly attenuates HFpEF-induced cardiac dysfunction and increases survival rate, suggesting that SSYX may prevent HF via regulation of cytoplasmic Ca2+ handling. Morphological and functional alterations of hepatic mitochondria have been docu- mented in patients with alcoholic liver disease (ALD). Our recent study demonstrated that zinc level was decreased in whole liver and mitochondria by chronic alcohol feeding. The present study was undertaken to determine whether zinc deficiency mediates alcohol- induced mitochondrial electron transport chain (ETC) defect and whether defective ETC function may lead to generation of reactive oxygen species (ROS). Male Wistar rats were pair fed with the Lieber-DeCarli control or ethanol diet for 5 mo. Chronic alcohol exposure increased hepatic triglyceride, free fatty acid, and 4-hy- droxynonenal (4HNE) levels; meanwhile hepatic mitochondrial 4HNE level was also increased. Moreover, hepatic mitochondrial respiratory complexes I, III, IV, and V and hepatic ATP production were decreased by chronic alcohol exposure. Chronic alcohol feeding decreased peroxisome proliferator-activated receptor gamma coacti- vator-1-alpha (PGC1a.), nuclear respiratory factor 1 (NRF1), mito- chondrial transcription factor A (TFAM), and mitochondrial DNA. HepG2 cells were treated with N,N,N=,N=-tetrakis (2-pyridylmethyl) ethylenediamine (TPEN) for 6 h. Zinc deficiency significantly de- creased mitochondrial respiratory complexes I, III, and IV. In addi- tion, PGC1a., NRF1, and TFAM levels as well as mitochondrial DNA were significantly decreased by TPEN treatment. Knockdown of mitochondrial respiratory complexes I, III, or IV by shRNA caused a decrease in mitochondrial membrane potential and an increase in ROS production. These results suggest that alcohol-induced hepatic zinc deficiency could inactivate mitochondrial biogenesis pathway and decrease mitochondrial DNA replication, which, in turn, decreases mitochondrial complex protein expression. Alcohol consumption causes nicotinic acid deficiency. The present study was undertaken to determine whether dietary nicotinic acid supplementation provides beneficial effects on alcohol-induced endotoxin signaling and the possible mechanisms at the gut-liver axis. Male Sprague-Dawley rats were pair-fed the Lieber-DeCarli liquid diets containing ethanol or isocaloric maltose dextrin for eight weeks, with or without dietary supplementation with 750 mg/liter nicotinic acid. Chronic alcohol feeding elevated the plasma endotoxin level and activated hepatic endotoxin signaling cascade, which were attenuated by nicotinic acid supplementation. Alcohol consumption remarkably decreased the mRNA levels of claudin-1, claudin-5, and ZO-1 in the distal intestine, whereas nicotinic acid significantly up-regulated these genes. The concentrations of endotoxin, ethanol, and acetaldehyde in the intestinal contents were increased by alcohol exposure, and niacin supplementation reduced the intestinal endotoxin and acetaldehyde levels. Nicotinic acid supplementation upregulated the intestinal genes involved in aldehyde detoxification via transcriptional regulation. These results demonstrate that modulation of the intestinal barrier function and bacterial endotoxin production accounts for the inhibitory effects of nicotinic acid on alcohol-induced endotoxemia and hepatic inflammation. The P2X7 inhibitor, brilliant blue G (BBG), has been reported as a neuroprotective drug against a variety of disorders, including neuropathic pain and brain ischemia. Currently, no studies have examined the potential for BBG to provide neuroprotection in animal models of TBI. The aim of the present study was to investigate the neuroprotective effect of BBG on TBI and to determine the underlying mechanisms. The rats were subjected to a diffuse cortical impact injury caused by a modified weight-drop device, and then divided randomly into three groups: the sham-operated, BBG treatment and vehicle groups. In the BBG treatment group, 50 mg/kg brilliant blue G (BBG; 100% pure), a highly specific and clinically useful P2X7 antagonist, was administered via the tail vein 15 min prior to or up to 8 h following TBI. The co-localization of NeuN and protein kinase Cγ (PKCγ) was followed with immunofluorescent staining. The expression of P2X7, PKCγ and inflammatory cytokines was identified by western blot analysis. Wet-dry weight method was used to evaluate brain edema, and motor function outcome was examined using the neurological severity score. The present study demonstrated that the administration of BBG attenuated TBI-induced cerebral edema and the associated motor deficits. Following trauma, BBG treatment significantly reduced the levels of PKCγ and interleukin-1β in the cortex. The results provide in vivo evidence that BBG exerted neuroprotective effects by attenuating brain edema and improving neurological functions via reducing PKCγ and interleukin-1β levels following TBI. Hepatic zinc deficiency has been well documented in alcoholic patients, but the mechanisms by which zinc deficiency mediates cell death have not been well defined. The objectives of this study were to determine whether alcohol perturbs subcellular zinc homeostasis and how organelle zinc depletion may link with cell death pathways. Wistar rats were pair-fed with the Lieber-DeCarli control or ethanol diet for 5 mo. Chronic alcohol exposure significantly reduced zinc level in isolated hepatic endoplasmic reticulum (ER) and mitochondria. Among the detected zinc transporters, ER Zrt/Irt-like protein (ZIP)13 and mitochondrial ZIP8, which transport zinc from ER and mitochondria to cytosol, were significantly increased. Mitochondrial zinc transporter (ZnT) 4, which transports zinc from cytosol to mitochondria, was also increased. ER phosphorylated eukaryotic initiation factor 2α, activating transcription factor 4, and C/EBP homologous protein were significantly upregulated, and mitochondrial cytochrome c release and Bax insertion were detected in association with caspase-3 activation and apoptotic cell death. To define the role of zinc deficiency in ER and mitochondrial stress, H4IIEC3 cells were treated with 3 μM N,N,N'=,N'=-tetrakis (2-pyridylmethyl) ethylenediamine for 6 h with or without supplementation with zinc or N-acetylcysteine (NAC). The results demonstrated that zinc deprivation induced caspase-3 activation and apoptosis in association with ER and mitochondria dysfunction, which were inhibited by zinc as low as 10 μM but not by 2 mM NAC. These results suggest that chronic ethanol exposure induced in ER and mitochondrial zinc deficiency might activate intrinsic cell death signaling pathway, which could not be effectively rescued by antioxidant treatment. Synaptic scaling is a form of synaptic plasticity that contributes to the homeostatic regulation of neuronal activity both in vitro and in vivo, by bidirectionally and proportionally adjusting postsynaptic AMPA receptor (AMPAR) abundance to compensate for chronic perturbations in activity. This proportional regulation of synaptic strength allows synaptic scaling to normalize activity without disrupting the synapse-specific differences in strength thought to underlie memory storage, but how such proportional scaling of synaptic strength is accomplished at the biophysical level is unknown. Here we addressed this question in cultured rat visual cortical pyramidal neurons. We used photoactivation and fluorescence recovery after photobleaching of fluorescently tagged AMPAR to show that scaling down, but not up, decreases the steady-state accumulation of synaptic AMPAR by increasing the rate at which they unbind from and exit the postsynaptic density (Koff). This increase in Koff was not diffusion limited, was independent of AMPAR endocytosis, and was prevented by a scaffold manipulation that specifically blocks scaling down, suggesting that it is accomplished through enhanced dissociation of AMPAR from synaptic scaffold tethers. Finally, simulations show that increasing Koff decreases synaptic strength multiplicatively, by reducing the fractional occupancy of available scaffold "slots." These data demonstrate that scaling down is accomplished through a regulated increase in Koff, which in turn reduces the fractional occupancy of synaptic scaffolds to proportionally reduce synaptic strength.
