104. Effects of Different Pretreatment Methods on Browning and Drying Characteristics of Hot Air-Dried Daylily Buds

Abstract

This experiment investigated the effects of soaking treatment (for 45 min) with different color fixatives (0.1 g/100 mL citric acid, 0.2 g/100 mL citric acid, 0.2 g/100 mL sodium bicarbonate, and 1.0 g/100 mL sodium chloride solution) after blanching for two minutes on the browning inhibition and drying characteristics of daylily buds during hot air drying. The color, nutrient composition and microstructure of dried products were characterized, and the chemical structure was analyzed by Fourier transform infrared (FTIR) spectroscopy. The pretreatment conditions for dried daylily buds with improved commodity value were determined using principal component analysis (PCA). Results showed that the browning inhibitory effect of steam blanching was better than that of hot water blanching. The decreasing order of the browning inhibitory effects of color fixatives was 0.2 g/100 mL citric acid solution > 0.1 g/100 mL citric acid solution > 1.0 g/100 mL sodium chloride solution > 0.2 g/100 mL sodium bicarbonate solution. Daylily buds pretreated with 0.2 g/100 mL citric acid solution showed the highest drying rate, and the dried product had high rehydration ratio. Collectively, the dried product prepared by steaming followed by soaking with 0.2 g/100 mL citric acid solution had good quality with respect to color, flavonoid and polyphenol contents and microstructure. PCA indicated that the contents of polyphenols and 5-hydroxymethylhydrate were highly correlated with color and browning degree, and chewiness had the highest correlation with hardness. Out of the eight principal components, two (the first one included polyphenol content, browning degree and color difference, and the second one included hardness and 5-hydroxymethylfurfural content) were extracted, which could well reflect the comprehensive information on the quality of dried daylily buds. This study can provide theoretical and technical references for the deep processing of daylily buds.

Abstract

One of the malignant transformation hallmarks is metabolism reprogramming, which plays a critical role in the biosynthetic needs of unchecked proliferation, abrogating cell death programs, and immunologic escape. However, the mechanism of the metabolic switch is not fully understood. Here, we found that the S-nitrosoproteomic profile of endogenous nitrogen oxide in ovarian cancer cells targeted multiple components in metabolism processes. Phosphofructokinase (PFKM), one of the most important regulatory enzymes of glycolysis, was S-nitrosylated by nitric oxide synthase NOS1 at Cys351. S-nitrosylation at Cys351 stabilized the tetramer of PFKM, leading to resist negative feedback of downstream metabolic intermediates. The PFKM-C351S mutation decreased the proliferation rate of cultured cancer cells, and reduced tumor growth and metastasis in the mouse xenograft model. These findings indicated that S-nitrosylation at Cys351 of PFKM by NOS1 contributes to the metabolic reprogramming of ovarian cancer cells, highlighting a critical role of endogenous nitrogen oxide on metabolism regulations in tumor progression.

124. Accelerating Early Math Learning with Research-Based Personalized Learning Games: A Cluster Randomized Controlled Trial

Abstract

This study examines whether "My Math Academy," a digital game-based learning environment that provides personalized content and adaptive embedded assessments, can improve math knowledge of Transitional Kindergarten and Kindergarten students. A cluster randomized study was conducted with 20 classrooms, 10 of which were randomly assigned to use "My Math Academy" for 12-14 weeks. Results of hierarchical linear models indicated that after using the app for about 5 hours, treatment students significantly outperformed the control group in math knowledge and skills as measured by TEMA-3. "My Math Academy" produced the greatest learning gains in students who began with a moderate level of math knowledge and on the most difficult skills assessed. Additionally, the more games students played, the greater the learning gains they experienced. Teacher surveys revealed that "My Math Academy" produced positive impacts on students' interest and self-confidence in learning math. Teachers also recognized "My Math Academy" as a user-friendly, valuable learning resource that they will continue to use. The study results will inform ongoing development of "My Math Academy," including tools to enable teachers to help children with low levels of prior math knowledge.

Abstract

School closures because of natural phenomena, such as COVID-19, underscore long-standing gaps in access to science education in the United States of America, particularly for young students. When educators have to pivot to deliver virtual instruction, it is important to identify feasible remote learning strategies for science content across formal and informal learning environments. This article discusses two evaluation studies of a multiplatform science learning program was originally developed for an in-person, formal learning environment that was modified based on infrastructure, preparations, and resource availability to meet the needs of distance learning in formal and informal learning environments due to COVID-19.
