Helicobacter pylori, a dominant member of the gastric microbiota, shares co-evolutionary history with humans. This has led to the development of genetically distinct H. pylori subpopulations associated with the geographic origin of the host and with differential gastric disease risk. Here, we provide insights into H. pylori population structure as a part of the Helicobacter pylori Genome Project (HpGP), a multi-disciplinary initiative aimed at elucidating H. pylori pathogenesis and identifying new therapeutic targets. We collected 1011 well-characterized clinical strains from 50 countries and generated high-quality genome sequences. We analysed core genome diversity and population structure of the HpGP dataset and 255 worldwide reference genomes to outline the ancestral contribution to Eurasian, African, and American populations. We found evidence of substantial contribution of population hpNorthAsia and subpopulation hspUral in Northern European H. pylori. The genomes of H. pylori isolated from northern and southern Indigenous Americans differed in that bacteria isolated in northern Indigenous communities were more similar to North Asian H. pylori while the southern had higher relatedness to hpEastAsia. Notably, we also found a highly clonal yet geographically dispersed North American subpopulation, which is negative for the cag pathogenicity island, and present in 7% of sequenced US genomes. We expect the HpGP dataset and the corresponding strains to become a major asset for H. pylori genomics.

Abstract Background Histone ubiquitination modification is emerging as a critical epigenetic mechanism involved in a range of biological processes. In vitro reconstitution of ubiquitinated nucleosomes is pivotal for elucidating the influence of histone ubiquitination on chromatin dynamics. Results In this study, we introduce a Non-Denatured Histone Octamer Ubiquitylation (NDHOU) approach for generating ubiquitin or ubiquitin-like modified histone octamers. The method entails the co-expression and purification of histone octamers, followed by their chemical cross-linking to ubiquitin using 1,3-dibromoacetone. We demonstrate that nucleosomes reconstituted with these octamers display a high degree of homogeneity, rendering them highly compatible with in vitro biochemical assays. These ubiquitinated nucleosomes mimic physiological substrates in function and structure. Additionally, we have extended this method to cross-linking various histone octamers and three types of ubiquitin-like proteins. Conclusions Overall, our findings offer an efficient strategy for producing ubiquitinated nucleosomes, advancing biochemical and biophysical studies in the field of chromatin biology.

Abstract Background Diabetes mellitus (DM) can impair driving safety due to hypoglycemia, hyperglycemia, diabetic peripheral neuropathy, and diabetic eye diseases. However, few studies have examined the association between DM and driving safety in older adults based on naturalistic driving data. Methods Data for this study came from a multisite naturalistic driving study of drivers aged 65–79 years at baseline. Driving data for the study participants were recorded by in-vehicle recording devices for up to 44 months. We used multivariable negative binomial modeling to estimate adjusted incidence rate ratios (aIRRs) and 95% confidence intervals (CIs) of hard braking events (HBEs, defined as maneuvers with deceleration rates ≥ 0.4 g) associated with DM. Results Of the 2856 study participants eligible for this analysis, 482 (16.9%) reported having DM at baseline, including 354 (12.4%) insulin non-users and 128 (4.5%) insulin users. The incidence rates of HBEs per 1000 miles were 1.13 for drivers without DM, 1.15 for drivers with DM not using insulin, and 1.77 for drivers with DM using insulin. Compared to drivers without DM, the risk of HBEs was 48% higher for drivers with DM using insulin (aIRR 1.48; 95% CI: 1.43, 1.53). Conclusion Older adult drivers with DM using insulin appear to be at increased proneness to vehicular crashes. Driving safety should be taken into consideration in DM care and management.

Abstract Objectives To investigate the utility of deep learning (DL) automated segmentation-based MRI radiomic features and clinical-radiological characteristics in predicting early recurrence after curative resection of single hepatocellular carcinoma (HCC). Methods This single-center, retrospective study included consecutive patients with surgically proven HCC who underwent contrast-enhanced MRI before curative hepatectomy from December 2009 to December 2021. Using 3D U-net-based DL algorithms, automated segmentation of the liver and HCC was performed on six MRI sequences. Radiomic features were extracted from the tumor, tumor border extensions (5 mm, 10 mm, and 20 mm), and the liver. A hybrid model incorporating the optimal radiomic signature and preoperative clinical-radiological characteristics was constructed via Cox regression analyses for early recurrence. Model discrimination was characterized with C-index and time-dependent area under the receiver operating curve (tdAUC) and compared with the widely-adopted BCLC and CNLC staging systems. Results Four hundred and thirty-four patients (median age, 52.0 years; 376 men) were included. Among all radiomic signatures, HCC with 5 mm tumor border ex tension and liver showed the optimal predictive performance (training set C-index, 0.696). By incorporating this radiomic signature, rim arterial phase hyperenhancement (APHE), and incomplete tumor “capsule,” a hybrid model demonstrated a validation set C-index of 0.706 and superior 2-year tdAUC (0.743) than both the BCLC (0.550; p < 0.001) and CNLC (0.635; p = 0.032) systems. This model stratified patients into two prognostically distinct risk strata (both datasets p < 0.001). Conclusion A preoperative imaging model incorporating the DL automated segmentation-based radiomic signature with rim APHE and incomplete tumor “capsule” accurately predicted early postsurgical recurrence of a single HCC. Critical relevance statement The DL automated segmentation-based MRI radiomic model with rim APHE and incomplete tumor “capsule” hold the potential to facilitate individualized risk estimation of postsurgical early recurrence in a single HCC. Key Points A hybrid model integrating MRI radiomic signature was constructed for early recurrence prediction of HCC. The hybrid model demonstrated superior 2-year AUC than the BCLC and CNLC systems. Abstract Objective Osteosarcopenia adversely affects the quality of life and physical health of older adults. We sought to explore the association between dietary patterns and osteosarcopenia in community-dwelling older adults. Methods This is a cross-sectional study from Northeast China, in which, we included older community adults aged 60 and above. Through face-to-face interviews, we collected dietary information from participants using a food frequency questionnaire. Subsequently, principal component analysis (PCA) was used to obtain the dietary patterns of the participants. Through physical examination, we obtained the participants’ information on osteosarcopenia, which was defined by the coexist of osteopenia and sarcopenia. We analysed the association between dietary patterns and dietary compositions with ostesarcopenia. Results In this study, a total of 9429 participants were included, and the prevalence of osteosarcopenia in community-dwelling older adults was 6.2%. PCA identified three main dietary patterns, and the lacto-ovo-vegetarian dietary pattern was inversely associated with osteosarcopenia. Compared to the lowest lacto-ovo-vegetarian quartile (Q1), the Q4 group (OR = 0.64, 95% CI:0.49–0.83) was inversely associated with osteosarcopenia. Through the weighted quantile sum regression model, we also found that the overall effect of the lacto-ovo-vegetarian dietary components was inversely associated with osteosarcopenia (OR = 0.58, 95% CI: 0.37–0.92); the largest contributors were vegetables, fresh milk, eggs, and dairy products. Conclusion Overall, we found that a lacto-ovo-vegetarian dietary pattern, particularly the consumption of vegetables, fresh milk, eggs, and dairy products, was inversely associated with osteosarcopenia in older adults. And this might provide new insights for the prevention and treatment of osteosarcopenia.

Abstract Objectives To examine the relationships between adverse childhood experiences (ACEs) and developing sarcopenia in older adults and the modifying effects of active social participation. Methods This prospective cohort study used survey data from the China Health and Retirement Longitudinal Study, including baseline surveys from 2011, follow-up data from 2013, follow-up data from 2015, and information on ACEs from the 2014 Life History Survey. Information concerning 10 ACEs, including five threat-related ACEs and five deprivation-related ACEs before 17 years of age was obtained by questionnaires through face-to-face interviews. Sarcopenia status was assessed according to the Asian Working Group for Sarcopenia 2019 algorithm, consisted of low muscle mass, and low muscle strength, or poor physical performance. The relationship between ACEs, social participation, and sarcopenia was evaluated using Cox proportional hazard regression models. Results The study population comprised 6859 older adults in main analyses. Having experienced ≥ 3 ACEs led to an increased 31% risk of developing sarcopenia (hazard ratio [HR]:1.31, 95% confidence interval [CI]:1.10–1.56). Participants having experienced ≥ 2 threat-related ACEs (HR:1.22, 95%CI:1.04–1.43) or deprivation-related ACEs (HR:1.22, 95%CI:1.02–1.46) had a 22% higher risk of developing sarcopenia. The myosin heavy chain 9 (MYH9) gene, located on human chromosome 22, encodes non-muscle myosin heavy chain IIA (NM IIA). This protein is essential to various cellular events, such as generating intracellular chemomechanical force and facilitating the movement of the actin cytoskeleton. Mutations associated with thrombocytopenia in autosomal dominant diseases first highlighted the significance of the MYH9 gene. In recent years, numerous studies have demonstrated the pivotal roles of MYH9 in various cancers. However, its effects on cancer are intricate and not fully comprehended. Furthermore, the elevated expression of MYH9 in certain malignancies suggests its potential as a target for tumor therapy. Nonetheless, there is a paucity of literature summarizing MYH9’s role in tumors and the therapeutic strategies centered on it, necessitating a systematic analysis. This paper comprehensively reviews and analyzes the pertinent literature in this domain, elucidating the fundamental structural characteristics, biological functions, and the nexus between MYH9 and tumors. The mechanisms through which MYH9 contributes to tumor development and its multifaceted roles in the tumorigenic process are also explored. Additionally, we discuss the relationship between MYH9-related diseases (MYH9-RD) and tumors and also summarize tumor therapeutic approaches targeting MYH9. The potential clinical applications of studying the MYH9 gene include improving early diagnosis, clinical staging, and prognosis of tumors. This paper is anticipated to provide novel insights for tumor therapy.

The human gut microbiome (GM) impacts various physiological processes and can lead to pathological conditions and even carcinogenesis if homeostasis is disrupted. Recent studies have indicated a connection between the GM and prostatic disease. However, the underlying mechanisms are still unclear. This review aims to provide a summary of the existing information regarding the connection between the GM and various prostatic conditions such as chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS), benign prostatic hyperplasia (BPH), and prostate cancer (PCa). Furthermore, the review aims to identify possible pathogenic mechanisms and suggest potential ways of targeting GM to prevent and treat prostatic disease. Due to the complexity of the mechanism between GM and prostatic diseases, additional research is required to comprehend the association between the two. This will lead to more effective treatment options for prostatic disease.

Abstract Background This study was performed to analyze the clinical effect of different concentrations of ropivacaine in the labor analgesia of the dural puncture epidural (DPE) technique for obese puerperae. Methods One hundred and fifty first-term obese women who received vaginal delivery and required labor analgesia in our hospital were selected prospectively for this study, and divided into groups A, B, and C. The three groups of puerpera were given epidurals with different concentrations of ropivacaine (0.075%, 0.10%, and 0.125%) with sufentanil (0.5 μg/ml) for the labor analgesia regimen. The visual analog scale (VAS), Ramsay scale, and Bromage scale of puerperae before analgesia and at different time points after anesthesia, and analgesic onset time, analgesia time, first PCEA time, PCEA pressing time, ropivacaine consumption, labor time, maternal blood pressure and heart rate, maternal adverse reactions, blood gas analysis in the neonatal umbilical artery, and Apgar score were observed. Results The analgesia onset time, PCEA pressing time, and ropivacaine consumption in group C were lower and the analgesia time and the first PCEA time were longer than those in groups A and B. At T1-T3 and T5, VAS scores of group A were higher than those in groups B and C, Ramsay score of group A was lower than that of groups B and C at T2–T3, and Bromage score of group C at any time point was higher than other two groups. The time of the second stage of labor in groups B and C was longer than that in group A, which in group C was longer than that in group B. Compared with groups A and C, the blood pressure and heart rate of puerperae in group B were closer to normal values. Three different concentrations of ropivacaine had no significant effect on the umbilical artery blood gas analysis indices and Apgar scores at 1st minute and 5th minute in neonates. The incidence of maternal adverse reactions in group C was lower than those in groups A and B. Conclusion 0.1% ropivacaine combined with 0.5 μg/ml sufentanil through DPE technique has good analgesic efficacy and few adverse effects in obese puerperae.

Water monitoring policies are recommended to ensure ecosystem preservation in industrial and agricultural development. Due to the reduction of water resources and the importance of sustainable rice production, it is necessary to develop relevant policies to promote the optimization of water and fertilizer distribution in rice paddy water environments. This article aims to plan the two objectives of water supply and reducing the environmental effects of nitrates in rice cultivation and analyzing the maximum and minimum benefits. By modifying the methods of fertilization and water distribution, the amount of production can be gradually increased by taking into account the harmful effects on the environment. Water and nitrate have a synergistic effect on the growth of paddy rice. However, the amount of runoff that is affected by nitrates is directly related to the pollution of aquatic environments. The results confirmed the correlation between nitrate concentration and production and reported the reduction of its negative effects depending on the management of inflow and outflow to rice fields. Changing the outflow according to the fertilization time and the inflow rate is an effective solution in the fertilizer nitrogen uptake and reducing the nitrate concentration in surface water sources in the long term. HIGHLIGHTS Achieving sustainability in the ecosystem and food security requires the development of research on the correlation between these factors and water resources.; The multi-objective model increased the sustainability of the ecosystem against pollutants by increasing benefits.;

AbstractContext Dan-Shen Decoction, which is composed of Danshen, Tanxiang and Sharen, has a good therapeutic effect on ischemic heart disease (IHD). However, systematic research on the exact mechanism of action of Dan-Shen Decoction is still lacking. The anti-IHD effect of Dan-Shen Decoction was examined in this study using a systematic pharmacological method.Objective This study validates the efficacy and explores the potential mechanisms of Dan-Shen Decoction in treating IHD by integrating network pharmacology analyses and experimental verification.Materials and methods The active components, critical targets and potential mechanisms of Dan-Shen Decoction against IHD were predicted by network pharmacology and molecule docking. H9c2 cells were pretreated with various 1 µg/mL Dan-Shen Decoction for 2 h before induction with 1000 µmol/L CoCl2 for 24 h. The cell viability was detected by CCK8, and protein expression was detected by western blots.Results The network pharmacology approach successfully identified 69 active components in Dan-Shen Decoction, and 122 potential targets involved in the treatment of IHD. The in vitro experiments indicate that the anti-IHD effect of Dan-Shen Decoction may be closely associated with targets such as AKT1 and MAPK1, as well as biological processes such as cell proliferation, inflammatory response, and metabolism.Conclusions This study not only provides new insights into the mechanism of Dan-Shen Decoction against IHD, but also provides important information and new research ideas for the discovery of anti-IHD compounds from traditional Chinese medicine.

To enhance the utilization of clean energy sources, such as wind power and photovoltaic within virtual power plants, and mitigate carbon emissions, this paper proposes a virtual power plant participation in the electricity carbon joint market optimization operation model based on ensemble empirical mode decomposition–improved bat algorithm (IBA)–Markov chain new energy output prediction. First, complementary set empirical mode decomposition is performed on historical data to construct a Markov chain based wind power and photovoltaic prediction model optimized by IBA. Second, this prediction model is used to predict the daily generation power of wind power and photovoltaic power. Finally, with the optimization goals of maximizing the benefits and minimizing the carbon costs of virtual power plants, a virtual power plant system participating in the electricity carbon joint market model based on wind power and photovoltaic output prediction results is constructed. At the same time, demand response factors are introduced and solved using the NSGA-II algorithm. Taking a certain park as an example for simulation analysis, the research results show that the combined effect of carbon market and demand response can achieve 99.82% of new energy consumption in virtual power plants without significantly reducing profits, basically achieving complete new energy consumption, demonstrating the effectiveness of the proposed model in this paper.

BackgroundThe impact of the COVID-19 pandemic on mental health among healthcare workers has been widely reported during the initial and ongoing phases of the COVID-19 pandemic. Yet, little remains known about the mental health status of primary healthcare workers in China during the post-pandemic era.MethodsA cross-sectional study was conducted between March 1, 2023, and May 31, 2023 in Anhui China. A total of 13,530 primary healthcare workers were recruited. Multiple logistic regression was used to identify potential factors associated with the incidence of depression and anxiety among primary healthcare workers.ResultsThe prevalence of depression and anxiety among primary healthcare workers was 50.7 and 26.4%, respectively. Multiple logistic regression revealed that female gender (OR = 1.345, 95%CI = 1.222–1.479), being divorced or widowed (OR = 1.432, 95%CI = 1.128–1.817), being a nurse (OR = 1.250, 95%CI = 1.126–1.388), and working more than 8 h per day (OR = 1.710, 95%CI = 1.583–1.846) were significantly associated with depression. A higher risk of anxiety among primary healthcare workers was associated with female gender (OR = 1.338, 95%CI = 1.198–1.494), being divorced or widowed (OR = 1.373, 95%CI = 1.056–1.770), being a nurse (OR = 1.139, 95%CI = 1.013–1.282), and working more than 8 h per day (OR = 1.638, 95%CI = 1.497–1.794). Better monthly income, more than 21 years of working experience and without experience of workplace violence were protective factors against depression and anxiety during the post-pandemic era.ConclusionDepressive symptoms are more common among primary healthcare workers in China during the post-pandemic era. Female gender, being divorced or widowed, being a nurse, working years, working seniority, monthly income, and experience of workplace violence were identified as associated factors. Targeted intervention is needed when developing strategies to reduce depression and improve primary healthcare workers’ wellness and mental health.

The rapid and efficient design strategy represents a highly intriguing research domain within the realm of metasurfaces. It necessitates minimizing the data load in coding metasurface design, facilitating the development of a precise approach to designing metasurfaces with specific characteristics. In this paper, we proposed a restrictive design strategy of metasurface for coding phase distributions based on the Fibonacci sequences, which is efficient to optimize designated radar cross section (RCS) reduction according to application requirements. Through the given restrictive strategy, the encoding range of the metasurfaces is reduced from the entire metasurfaces to the initial phase units in the corners, greatly reducing the total amount of data in the optimization database. In order to verify the availability of the strategy, RCS reduction metasurfaces with variable phase sequences in microwave region are analyzed. For broadband and wide-angle RCS reduction, the optimized Fibonacci discrete phase gradient (FDPG) metasurface is obtained by adjusting the phase sequences. Both the simulation and measured results show that the optimized FDPG metasurfaces by the proposed designing strategy have the RCS reduction of −10 dB in the frequency ranges of 6 GHz to 18 GHz under wide incidence angles from 0° to 30°. The simulated and measured results are good consistent over the whole frequency ranges and angle regions. This method combines mathematical sequences with physics organically, making it an important field in metasurfaces research and it is expected to play an important role in future studies.

Abstract Understanding the CO2 transformation mechanism on materials is essential for the design of efficient electrocatalysts for CO2 reduction. In aconventional adsorbate evolution mechanism (AEM), the catalysts encounter multiple high‐energy barrier steps, especially CO2 activation, limiting the activity and selectivity. Here, lattice carbonate from Cu2(OH)2CO3 is revealed to be a mediator between CO2 molecules and catalyst during CO2 electroreduction by a 13C isotope labeling method, which can bypass the high energy barrier of CO2 activation and strongly enhance the performance. With the lattice carbonate mediated mechanism (LCMM), the Cu2(OH)2CO3 electrode exhibited ten‐fold faradaic efficiency and 15‐fold current density for ethylene production than the Cu2O electrode with AEM at a low overpotential. Theoretical calculations and in situ Raman spectroscopy results show that symmetric vibration of carbonate is precisely enhanced on the catalyst surface with LCMM, leading to faster electron transfer, and lower energy barriers of CO2 activation and carbon–carbon coupling. This work provides a route to develop efficient electrocatalysts for CO2 reduction based on lattice‐mediated mechanism.

A broadband radar cross-section (RCS) reduction employed by the vortex metasurface is proposed. The underlying physics is associated with the Pancharatnam-Berry geometrical phases involved in spin-to-orbital angular momentum conversion. The proposed metasurface generates spin-controlled vortex waves with different topological charges. The vortex singularity implements specular extremely low RCS reduction. The prototypes are fabricated and measured to verify the wideband performance of the RCS reduction. The simulated results illustrate that −10 dB broadband RCS reduction from 9 GHz to 22.5 GHz and the RCS reduction has peak values −37.2 dB, −40.8 dB, and −42.5 dB at 9.8 GHz, 13.1 GHz, and 20.8 GHz, respectively. The measurement results illustrate that −10 dB RCS reduction over 9.1–18 GHz is realized, and the RCS reduction peak −53 dB at 10.1 GHz. The measured electric field intensities and phase distribution of the vortex metasurface also be displayed. Due to the limitation of experimental conditions, no test was done in the range greater than 18 GHz. Far-field results achieve good agreement between simulations and experiments, and the near-field measurements are a subtle difference in the experimental results. This work reveals the potential advantages of vortex waves in the fields of multifunctional electromagnetic stealth, beamforming for communication systems, etc.
