IntroductionThe development of high-quality stated preference (SP) surveys requires a rigorous design process involving engagement with representatives from the target population. However, while transparency in the reporting of the development of SP surveys is encouraged, few studies report on this process and the outcomes. Recommended stages of instrument development includes both steps for stakeholder/end-user engagement and pretesting. Pretesting typically involves interviews, often across multiple waves, with improvements made at each wave; pretesting is therefore resource intensive. The aims of this paper are to report on the outcomes of collaboration with a Lewy body dementia research advisory group during the design phase of a SP survey. We also evaluate an alternative approach to instrument development, necessitated by a resource constrained context.MethodThe approach involved conducting the stages of end-user engagement and pretesting together during a public involvement event. A hybrid approach involving a focus group with breakout interviews was employed. Feedback from contributors informed the evolution of the survey instrument.ResultsChanges to the survey instrument were organized into four categories: attribute modifications; choice task presentation and understanding; information presentation, clarity and content; and best-best scaling presentation. The hybrid approach facilitated group brainstorming while still allowing the researcher to assess the feasibility of choice tasks in an interview setting. However, greater individual exploration and the opportunity to trial iterative improvements across waves was not feasible with this approach.DiscussionInvolvement of the research advisory group resulted in a more person-centered survey design. In a context constrained by time and budget, and with consideration of the capacity and vulnerability of the target population, the approach taken was a feasible and pragmatic mechanism for improving the design of a SP survey.

The dearth of land in high-density urban centres necessitates constructing structures with deep interiors, with little or no connection to the external environment. Additionally, the negative relationships between high-rise structures in the city centresin recent times are increasingly making contact with the outdoor environments undesirable. Whether in the built environment, decision sciences, healthcare, or technological disciplines, there is a need to understand the factors necessary for windowless officeimplementation, its prominent researchers, and geographical contexts, among other conditions for practical application. These background variables could assist in arriving at emerging themes, problems and gaps for concrete windowless office development in Malaysia. In this systematic review, the bibliographic information of title, abstract and keywords was mined from 255 documents on the Scopus database to generate scientometric maps and links showing relationships on co-authorship, co-citation, co-occurrences, and bibliometric coupling. The top trending variables in windowless office research are COVID-19, energy efficiency, indoor air quality, heart rate variability, performance assessment, visual reality, and workplace environment. Like other advanced economies in Asia, Malaysia is seen to attain a satisfactory level of development to implement the windowless office design module, with stable electricity power, technological advancement, and psychological stability of the people. The emerging variables in this review will give novel directions in exploring the developmental, psychological and health conditions for effective windowless office implementation in the country and similar economies in Asia, Africa and South America, where metropolises are fast becoming compacted due to ever-growing city centres.

It is generally accepted that the human abdominal wall comprises skin, subcutaneous tissues, muscles and their aponeuroses, and the parietal peritoneum. Understanding these layers and their mechanical properties provides valuable information to those designing procedural skills trainers, supporting surgical procedures (hernia repair), and engineering-based work (in silico simulation). However, there is little literature available on the mechanical properties of the abdominal wall in layers or as a composite in the context of designing a procedural skills trainer. This work characterizes the tensile properties of the human abdominal wall by layer and as a partial composite. Tissues were collected from fresh-never-frozen and fresh-frozen cadavers and tested in uniaxial tension at a rate of 5 mm/min until failure. Stress–strain curves were created for each sample, and the values for elastic moduli, ultimate tensile strength, and strain at failure were obtained. The experimental outcomes from this study demonstrated variations in tensile properties within and between tissues. The data also suggest that the tensile properties of composite abdominal walls are not additive. Ultimately, this body of work contributes to a deeper comprehension of these mechanical properties and will serve to enhance patient care, refine surgical interventions, and assist with more sophisticated engineering solutions.

The purpose of this study was to focus on the relationship between career-development training programs and the motivation of employees. The study used a qualitative method and a phenomenological design using semistructured interviews conducted with a script of open-ended questions. Two main components of the research design were the modified van Kaam method for exploration of experiences and the purposive selection of the participants. The 20 participants in the sample were project managers and consultants from south-central Texas, all of whom had experience with governmental projects. Based on interview responses, the findings of this study highlighted a concern from the participants that training programs have a major influence on workplace productivity and morale. Furthermore, the findings suggest the training program's quality may influence workplace productivity. The authors conclude by pointing out the analysis of the answers showed that not only does workplace productivity decline from the changes in a training program, but employee motivation is altered as well.

This study focused on the factors that affect motivation of faculty in Saudi Arabia. It included two surveys and open-ended queries to a focus group of five academic managers and 25 faculty members of varying nationalities, rank, and institutes in Saudi Arabia. The research showed that the faculties in Saudi Arabia's highest education industry feel disconnected from the program development. The faculty members did not feel motivated to participate in the development and improvement of the academic program due to: (a) lack of monetary and non monetary incentives, (b) management not involving faculty in decision-making, and (c) lack of recognition and moral support. However, the faculties were intrinsically motivated to perform their greater interest in intrinsic motivation as a personal measure for success inside the classroom, but extrinsic motivation was a factor that needed greater improvement from the management of the universities for faculty to partake in development of the program.

Abstract Delirium is an acute change in attention and cognition occurring in ~ 65% of severe SARS-CoV-2 cases. It is also common following surgery and an indicator of brain vulnerability and risk for the development of dementia. In this work we analyzed the underlying role of metabolism in delirium-susceptibility in the postoperative setting using metabolomic profiling of cerebrospinal fluid and blood taken from the same patients prior to planned orthopaedic surgery. Distance correlation analysis and Random Forest (RF) feature selection were used to determine changes in metabolic networks. We found significant concentration differences in several amino acids, acylcarnitines and polyamines linking delirium-prone patients to known factors in Alzheimer’s disease such as monoamine oxidase B (MAOB) protein. Subsequent computational structural comparison between MAOB and angiotensin converting enzyme 2 as well as protein–protein docking analysis showed that there potentially is strong binding of SARS-CoV-2 spike protein to MAOB. The possibility that SARS-CoV-2 influences MAOB activity leading to the observed neurological and platelet-based complications of SARS-CoV-2 infection requires further investigation.

Current trends in post-secondary education enrollment indicate that colleges and universities are likely to experience an increase in the number of online students. The purpose of this study was to ascertain the type of resources and support features online faculty need, desire, and expect in a support website. The method used to collect research findings was an online survey. The participants for this study consisted of the online faculty population at an institution of higher learning in the southwestern United States. Participants were invited by email to participate in a 13 question survey which asked participants to rate the importance of the questions listed. Of the 2,522 survey invitations e-mailed to potential participants, 380 responded with completed surveys, providing a response rate of 15.06%. Although this was a relativity low response rate, researchers felt the demographics of the respondents provided an accurate representation of the population studied. Findings from the survey indicated participants agree there is a need to implement a support website. Participants indicated the support website should provide support resources, communication forums, and resources to increase connectivity to the institution. The authors note providing online faculty with support websites could be a differentiation strategy to recruit and retain quality online faculty.

Abstract Background Type 2 diabetes is a risk factor for Alzheimer’s disease (AD), and AD brain shows impaired insulin signalling. The role of peripheral insulin resistance on AD aetiopathogenesis in non-diabetic patients is still debated. Here we evaluated the influence of insulin resistance on brain glucose metabolism, grey matter volume and white matter lesions (WMLs) in non-diabetic AD subjects. Methods In total, 130 non-diabetic AD subjects underwent MRI and [18F]FDG PET scans with arterial cannula insertion for radioactivity measurement. T1 Volumetric and FLAIR sequences were acquired on a 3-T MRI scanner. These subjects also had measurement of glucose and insulin levels after a 4-h fast on the same day of the scan. Insulin resistance was calculated by the updated homeostatic model assessment (HOMA2). For [18F]FDG analysis, cerebral glucose metabolic rate (rCMRGlc) parametric images were generated using spectral analysis with arterial plasma input function. Results In this non-diabetic AD population, HOMA2 was negatively associated with hippocampal rCMRGlc, along with total grey matter volumes. No significant correlation was observed between HOMA2, hippocampal volume and WMLs. Conclusions In non-diabetic AD, peripheral insulin resistance is independently associated with reduced hippocampal glucose metabolism and with lower grey matter volume, suggesting that peripheral insulin resistance might influence AD pathology by its action on cerebral glucose metabolism and on neurodegeneration.

Systems archetypes are patterns of structure found in systems that are helpful in understanding some of the dynamics within them. The intent of this study was to examine educational attainment data using the success-to-the-successful archetype as a model to see if it helps to explain the inequality observed in the data. Data covering 1990 to 2009 was collected from the United States Census Bureau for the United States, as well as for the States of Georgia and Washington. Findings indicated that this archetype is useful in explaining the variation, and there are several other terms and concepts that are helpful as well as the terms/concepts: success-to-the successful, the rich get richer, Pareto's law, Zipf's law, and power law distributions are related and sometimes used interchangeably.

Abstract Objective The prevalence of pre-diabetes is increasing globally with more than 470 million people projected to develop pre-diabetes by 2030. In Africa, the average prevalence of pre-diabetes was estimated at 7.3% in 2015 and affected individual will develop type 2 diabetes mellitus within few decades. The aim of the study was to determine the prevalence of pre-diabetes and associated risk factors among residents of Iganga municipality. A cross-sectional study was conducted among males and females aged 13–60 years. District health office provided updated household list from which sampling of the villages was performed based on probability proportionate to population. Consented participants were prepared for the study, allowing fasting for 8 to 10 h before blood collection the next morning. Individuals with impaired fasting glucose, were subjected to OGTT. Results 130 participants were enrolled, of which 98 were women. The mean age of the participants was 35 years. The prevalence of pre-diabetes was 3.8%. Abstract Background Liraglutide is a glucagon-like peptide-1 (GLP-1) analogue currently approved for type 2 diabetes and obesity. Preclinical evidence in transgenic models of Alzheimer’s disease suggests that liraglutide exerts neuroprotective effects by reducing amyloid oligomers, normalising synaptic plasticity and cerebral glucose uptake, and increasing the proliferation of neuronal progenitor cells. The primary objective of the study is to evaluate the change in cerebral glucose metabolic rate after 12 months of treatment with liraglutide in participants with Alzheimer’s disease compared to those who are receiving placebo. Methods/design ELAD is a 12-month, multi-centre, randomised, double-blind, placebo-controlled, phase IIb trial of liraglutide in participants with mild Alzheimer’s dementia. A total of 206 participants will be randomised to receive either liraglutide or placebo as a daily injection for a year. The primary outcome will be the change in cerebral glucose metabolic rate in the cortical regions (hippocampus, medial temporal lobe, and posterior cingulate) from baseline to follow-up in the treatment group compared with the placebo group. The key secondary outcomes are the change from baseline to 12 months in z scores for clinical and cognitive measures (Alzheimer’s Disease Assessment Scale—Cognitive Subscale and Executive domain scores of the Neuropsychological Test Battery, Clinical Dementia Rating Sum of Boxes, and Alzheimer’s Disease Cooperative Study—Activities of Daily Living) and the incidence and severity of treatment-emergent adverse events or clinically important changes in safety assessments. Other secondary outcomes are 12-month change in magnetic resonance imaging volume, diffusion tensor imaging parameters, reduction in microglial activation in a subgroup of participants, reduction in tau formation and change in amyloid levels in a subgroup of participants measured by tau and amyloid imaging, and changes in composite scores using support machine vector analysis in the treatment group compared with the placebo group. Discussion Alzheimer’s disease is a leading cause of morbidity worldwide. As available treatments are only symptomatic, the search for disease-modifying therapies is a priority. If the ELAD trial is successful, liraglutide and GLP-1 analogues will represent an important class of compounds to be further evaluated in clinical trials for Alzheimer’s treatment. The duodenal-jejunal bypass liner (Endobarrier) is an endoscopic treatment for obesity and type 2 diabetes mellitus (T2DM). It creates exclusion of the proximal small intestine similar to that after Roux-en-Y Gastric Bypass (RYGB) surgery. The objective of this study was to employ a reductionist approach to determine whether bypass of the proximal intestine is the component conferring the effects of RYGB on food intake and sweet taste preference using the Endobarrier as a research tool. A nested mechanistic study within a large randomised controlled trial compared the impact of lifestyle modification with vs. without Endobarrier insertion in patients with obesity and T2DM. Forty-seven participants were randomised and assessed at several timepoints using direct and indirect assessments of food intake, food preference and taste function. Patients within the Endobarrier group lost numerically more weight compared to the control group. Using food diaries, our results demonstrated similar reductions of food intake in both groups. There were no significant differences in food preference and sensory, appetitive reward, or consummatory reward domain of sweet taste function between groups or changes within groups. In conclusion, the superior weight loss seen in patients with obesity and T2DM who underwent the Endobarrier insertion was not due to a reduction in energy intake or change in food preferences.

Background: Hand grip strength is a well validated measure of physical strength, while anthropometric measures such as weight, waist, hip and mid upper arm circumference, are well validated to determine physical health. The present analysis investigated the relationship between hand grip strength, anthropometric measures and dementia incidence in 70–90‐year‐old men residing in Northern Ireland (NI). Method: The PRIME‐COG study is nested in PRIME (Prospective Epidemiological Study of Myocardial Infarction), a cohort study of men in NI. In the most recent follow up for PRIME‐COG, men underwent a comprehensive cognitive assessment from which a consensus diagnosis of normal cognitive function, MCI or dementia was made by experienced clinical researchers. Hand grip strength and anthropometrics measures were also recorded. Logistic regression models were performed to assess the association between hand grip strength and anthropometric measures and cognitive endpoints. Result: The analysis identified that low hand grip strength was associated with dementia and/or MCI, although this did not reach statistical significance (OR 1.39, p = 0.18). In the anthropometric measures, a significant relationship was observed between hip circumference and dementia and/or MCI at PRIME‐COG follow up (OR 0.93, p = 0.01), whereas BMI was non‐significantly associated (OR 1.22, p = 0.58). The remaining anthropometric measures investigated were not found to hold any significant relationships with the cognitive outcomes. Conclusion: Within a cohort of older men residing in NI, low hand grip strength was associated with a 39% increased risk of dementia or MCI, although this was not statistically significant. In anthropometric measures, hip circumference was found to be significantly negatively associated with a 7% reduced risk of dementia or MCI, while BMI was associated with a 22% increased risk, which again was not of statistical significance. Background: Between 10% and 15% of hypothyroid patients experience persistent symptoms despite achieving biochemical euthyroidism. Unexplained persistent symptoms can be a sign of somatization. This is associated with distress and high health care resource use and can be classified as somatic symptom disorder (SSD). Prevalence rates for SSD differ depending on classification criteria and how they are ascertained, varying between 4% and 25%. As this has not been studied in hypothyroid patients before, the aim of this study was to document somatization in people with hypothyroidism and to explore associations with other patient characteristics and outcomes. Methods: Online, multinational cross-sectional survey of individuals with self-reported, treated hypothyroidism, which included the validated Patient Health Questionnaire-15 (PHQ-15) for assessment of somatization. Chi-squared tests with the Bonferroni correction were used to explore outcomes for respondents with a PHQ-15 score ≥10 (probable somatic symptom disorder [pSSD]) versus a PHQ-15 score <10 (absence of SSD). Results: A total of 3915 responses were received, 3516 of which contained the valid PHQ-15 data (89.8%). The median score was 11.3 (range 0–30 [confidence interval 10.9–11.3]). The prevalence of pSSD was 58.6%. Associations were found between pSSD and young age (p < 0.001), women (p < 0.001), not working (p < 0.001), having below average household income (p < 0.001), being treated with levothyroxine (LT4) (rather than combination of LT4 and L-triiodothyronine [LT3], LT3 alone, or desiccated thyroid extract) (p < 0.001), expression of the view that the thyroid medication taken did not control the symptoms of hypothyroidism well (p < 0.001), and with number of comorbidities (p < 0.001). pSSD was associated with respondent attribution of most PHQ-15 symptoms to the hypothyroidism or its treatment (p < 0.001), dissatisfaction with care and treatment of hypothyroidism (p < 0.001), a negative impact of hypothyroidism on daily living (p < 0.001), and with anxiety and low mood/depression (p < 0.001). Conclusions: This study demonstrates a high prevalence of pSSD among people with hypothyroidism and associations between pSSD and negative patient outcomes, including a tendency to attribute persistent symptoms to hypothyroidism or its treatment. Opening the sky to new classes of airspace user is a political and economic imperative for the European Union. Drone industries have a significant potential for economical growth according to the latest estimations. To enable this growth safely and efficiently, the CORUS project has developed a concept of operations for drones flying in Europe in very low-level airspace, which they have to share that space with manned aviation, and quite soon with urban air mobility aircraft as well. U-space services and the development of smart, automated, interoperable, and sustainable traffic management solutions are presented as the key enabler for achieving this high level of integration. In this paper, we present the U-space concept of operations (ConOps), produced around three new types of airspace volume, called X, Y, and Z, and the relevant U-space services that will need to be supplied in each of these. The paper also describes the reference high-level U-space architecture using the European air traffic management architecture methodology. Finally, the paper proposes the basis for the aircraft separation standards applicable by each volume, to be used by the conflict detection and resolution services of U-space.

Activated phosphoinositide 3-kinase (PI3K) δ Syndrome (APDS), caused by autosomal dominant mutations in PIK3CD (APDS1) or PIK3R1 (APDS2), is a heterogeneous primary immunodeficiency. While initial cohort-descriptions summarized the spectrum of clinical and immunological manifestations, questions about long-term disease evolution and response to therapy remain. The prospective European Society for Immunodeficiencies (ESID)-APDS registry aims to characterize the disease course, identify outcome predictors, and evaluate treatment responses. So far, 77 patients have been recruited (51 APDS1, 26 APDS2). Analysis of disease evolution in the first 68 patients pinpoints the early occurrence of recurrent respiratory infections followed by chronic lymphoproliferation, gastrointestinal manifestations, and cytopenias. Although most manifestations occur by age 15, adult-onset and asymptomatic courses were documented. Bronchiectasis was observed in 24/40 APDS1 patients who received a CT-scan compared with 4/15 APDS2 patients. By age 20, half of the patients had received at least one immunosuppressant, but 2–3 lines of immunosuppressive therapy were not unusual before age 10. Response to rapamycin was rated by physician visual analog scale as good in 10, moderate in 9, and poor in 7. Lymphoproliferation showed the best response (8 complete, 11 partial, 6 no remission), while bowel inflammation (3 complete, 3 partial, 9 no remission) and cytopenia (3 complete, 2 partial, 9 no remission) responded less well. Hence, non-lymphoproliferative manifestations should be a key target for novel therapies. This report from the ESID-APDS registry provides comprehensive baseline documentation for a growing cohort that will be followed prospectively to establish prognostic factors and identify patients for treatment studies.

This paper is aimed at estimating Radio Frequency (RF) radiation absorbed by humans around Base Transceiver Stations (BTS) within Kaduna metropolis. RF radiation absorbed by humans from forty two (42) selected (BTS) tagged M1 to M42 of the four major network providers (MTN, 9mobile, Airtel and Globacom) within Kaduna metropolis, Kaduna State, Nigeria within radial distance intervals of 20 m to 100 m from the foot of each BTS were estimated. The Average Power Density (PDAve) and Specific Absorption Rate of data obtained from these BTS were computed, and the results indicated that the average amount of SAR for the selected forty two (42) base transceiver BTS facilities were within the range of 16.9 - 331.7 × 10-9 W/kg and 0.648-10.659 × 10-6 W/m² respectively. These values are lower than the recommended limit by International Commission on Non Ionization Radiation Protection (ICNIRP) which is 0.08 W/kg for the human body. This study has shown that RF radiation absorbed by people either residing or transiting the study area has negligible effect on their health. [ABSTRACT FROM AUTHOR]

Lysosomes are acidic cytoplasmic organelles that are present in all nucleated mammalian cells and are involved in a variety of cellular processes including repair of the plasma membrane, defense against pathogens, cholesterol homeostasis, bone remodeling, metabolism, apoptosis and cell signaling. Defects in lysosomal enzyme activity have been associated with a variety of neurological diseases including Parkinson's Disease, Lysosomal Storage Diseases, Alzheimer's disease and Huntington's disease. Fluorogenic lysosomal staining probes were synthesized for labeling lysosomes and other acidic organelles in a live-cell format and were shown to be capable of monitoring lysosomal metabolic activity. The new targeted substrates were prepared from fluorescent dyes having a low pKa value for optimum fluorescence at the lower physiological pH found in lysosomes. They were modified to contain targeting groups to direct their accumulation in lysosomes as well as enzyme-cleavable functions for monitoring specific enzyme activities using a live-cell staining format. Application to the staining of cells derived from blood and skin samples of patients with Metachromatic Leukodystrophy, Krabbe and Gaucher Diseases as well as healthy human fibroblast and leukocyte control cells exhibited localization to the lysosome when compared with known lysosomal stain LysoTracker® Red DND-99 as well as with anti-LAMP1 Antibody staining. When cell metabolism was inhibited with chloroquine, staining with an esterase substrate was reduced, demonstrating that the substrates can be used to measure cell metabolism. When applied to diseased cells, the intensity of staining was reflective of lysosomal enzyme levels found in diseased cells. Substrates specific to the enzyme deficiencies in Gaucher or Krabbe disease patient cell lines exhibited reduced staining compared to that in non-diseased cells. The new lysosome-targeted fluorogenic substrates should be useful for research, diagnostics and monitoring the effect of secondary therapeutic agents on lysosomal enzyme activity in drug development for the lysosomal storage disorders and allied diseases.

This study combined high resolution mass spectrometry (HRMS), advanced chemometrics and pathway enrichment analysis to analyse the blood metabolome of patients attending the memory clinic: cases of mild cognitive impairment (MCI; n = 16), cases of MCI who upon subsequent follow-up developed Alzheimer's disease (MCI_AD; n = 19), and healthy age-matched controls (Ctrl; n = 37). Plasma was extracted in acetonitrile and applied to an Acquity UPLC HILIC (1.7μm x 2.1 x 100 mm) column coupled to a Xevo G2 QTof mass spectrometer using a previously optimised method. Data comprising 6751 spectral features were used to build an OPLS-DA statistical model capable of accurately distinguishing Ctrl, MCI and MCI_AD. The model accurately distinguished (R2 = 99.1%; Q2 = 97%) those MCI patients who later went on to develop AD. S-plots were used to shortlist ions of interest which were responsible for explaining the maximum amount of variation between patient groups. Metabolite database searching and pathway enrichment analysis indicated disturbances in 22 biochemical pathways, and excitingly it discovered two interlinked areas of metabolism (polyamine metabolism and L-Arginine metabolism) were differentially disrupted in this well-defined clinical cohort. The optimised untargeted HRMS methods described herein not only demonstrate that it is possible to distinguish these pathologies in human blood but also that MCI patients 'at risk' from AD could be predicted up to 2 years earlier than conventional clinical diagnosis. Blood-based metabolite profiling of plasma from memory clinic patients is a novel and feasible approach in improving MCI and AD diagnosis and, refining clinical trials through better patient stratification.

This experiment tested the hypothesis that swimming performance in Atlantic salmon (Salmo salar) parr is connected to cardiorespiratory performance and morphology, as well as maximum heart rate (fHmax) related measures of thermal tolerance. Moreover, it was hypothesized that the cardiorespiratory differences between poor and strong swimmers will be retained in a later life stage, i.e., 15 weeks post-smoltification and seawater transfer. This experiment screened a population of 3,200 parr (11.2 ± 0.25 g) for their swimming performance, classifying them as poor and good swimmers based on their critical swimming speeds (4.4±0.1 body length s-1 and >6.8±0.1 body length s-1, respectively). Compared with poor performing parr, good swimmers had a significantly thicker compact myocardium (by 23.7%) and taller gill secondary lamellae (by 16.2%). In contrast, there was no significant difference in maximum oxygen consumption between the two groups as assessed using a ‘chase’ protocol, and the relationship between heart rate specific measures of thermal tolerance and swim performance was variable. For example, three measures did not differ between the two groups, whereas the Arrhenius breakpoint temperature for fHmax and fHmax were higher and lower, respectively, in the poor swimmers. Importantly, the identified morphological and fHmax differences at the parr stage persisted after 15 weeks of common garden rearing in seawater, and they were associated with an increase in relative ventricular mass and a small, but significant, improvement in growth rate. Therefore, it seems that an early assessment of swimming performance can effectively screen for morphological capacities related to oxygen supply and growth rate, but less so for heart rate related measures of thermal tolerance.

One of the challenges in the analysis of large data sets, particularly in a population-based setting, is the ability to perform comparisons across projects. This has to be done in such a way that the integrity of each individual project is maintained, while ensuring that the data are comparable across projects. These issues are beginning to be observed in human DNA methylation studies, as the Illumina 450k platform and next generation sequencing-based assays grow in popularity and decrease in price. This increase in productivity is enabling new insights into epigenetics, but also requires the development of pipelines and software capable of handling the large volumes of data. The specific problems inherent in creating a platform for the storage, comparison, integration and visualization of DNA methylation data include data storage, algorithm efficiency and ability to interpret the results to derive biological meaning from them. Databases provide a ready-made solution to these issues, but as yet no tools exist that that leverage these advantages while providing an intuitive user interface for interpreting results in a genomic context.We have addressed this void by integrating a database to store DNA methylation data with a web interface to query and visualize the database and a set of libraries for more complex analysis. The resulting platform is called DaVIE: Database for the Visualization and I of Epigenetics data. DaVIE can use data culled from a variety of sources, and the web interface includes the ability to group samples by sub-type, compare multiple projects and visualize genomic features in relation to sites of interest. We have used DaVIE to identify patterns of DNA methylation in specific project and across different projects, identify outlier samples, and cross-check differentially methylated CpG sites identified in specific projects across large numbers of samples.

With the rising use of facial recognition systems in a range of real-world scenarios and applications, attackers are also increasing their efforts, with a number of spoofing techniques emerging. As a result, developing a reliable spoof detection mechanism is critical. Active-based techniques have been shown to be good at finding spoofs, but they have a number of problems, such as being intrusive, expensive, hard to compute, not being able to be used in many situations, and usually needing extra hardware. This research presented an active-based robust spoof detection technique capable of detecting a wide range of media or 2D attacks while being less intrusive, less expensive, low in complexity, and more generalizable than other activebased techniques. It doesn't require any additional hardware, so it can easily be integrated into current systems. The distortion variations of video frames of the user's face collected at varying distances from the camera are analyzed to detect spoofing. Both the legitimate and spoof attack datasets were created using real-world facial photo and video data. The proposed approach achieved a spoof detection accuracy of 98.18% using both machine learning classifiers and a deep learning model, with an equal error rate and a half total error rate as low as 0.023 and 0.021, respectively.
