Numerous plant compounds and their metal-ion complexes exert antioxidative, anti-inflammatory, anticancer, and other beneficial effects. This review highlights the different bioactivities of flavonoids, chromones, and coumarins and their metal-ions complexes due to different structural characteristics. In addition to insight into the most studied antioxidative properties of these compounds, the first part of the review provides a comprehensive overview of exogenous and endogenous sources of reactive oxygen and nitrogen species, oxidative stress-mediated damages of lipids and proteins, and on protective roles of antioxidant defense systems, including plant-derived antioxidants. Additionally, the review covers the anti-inflammatory and antimicrobial activities of flavonoids, chromones, coumarins and their metal-ion complexes which support its application in medicine, pharmacy, and cosmetology.

A chemical and biological preliminary study of the species Vismia cayennensis, collected in the Amazonas state, Venezuela. The antibacterial test of plant bark extract showed significant inhibition in Escherichia coli, Shigella sp and Staphylococcus aureus. In addition, three of the five soluble fractions of different polarity solvents, specifically those of chloroform, acetone and water, maintained moderately active against Shigella sp strain. The hydroalcoholic extract of the plant and the fraction soluble in chloroform, exhibited a significant antiinflammatory effect. Cytotoxicity tests performed by the methods of (3-(4,5-dimetiltiazol-2-yl)-5-(3-carboximetoxifenil)-2-(4-sulfophenyl)-2H-tetrazolium)/phenazine methosulfate of (MTS/PMS) and sulforhodamine B , revealed that only has water soluble cytotoxic effect. Additionally, a study phytochemical obtained information on the presence of some families of secondary metabolites such as flavonoids, saponins, tannins, polyphenols, anthraquinones, triterpenes and sterols. It can be inferred that the stem bark of the plant V. cayennensis, is a promising source of bioactive secondary metabolites

Two new labdane-type diterpenes, named viterotulin C (1) and vitexilactone D (2), together with five known diterpenes (3–7), were isolated from the fruits of Vitex trifolia L. var. simplicifolia Cham. Their structures were elucidated by detailed analysis of spectroscopic data. All the compounds were evaluated for their inhibitory effects on nuclear factor-kappa B (NF-κB) pathway in HEK 293 cell line. These compounds presented inhibition on TNF-α-induced NF-κB activation, with inhibition rates ranging from 42.52 ± 10.69% to 68.86 ± 10.76% at the concentration of 50 μM. [ABSTRACT FROM AUTHOR]

Abstract: BACKGROUND: Recent epidemiological studies have suggested that phenolic compounds present in grapefruit play an important role in the bioactive properties of this fruit. However, the consumption of fresh grapefruit is low. Freeze‐dried powdered grapefruit can be an alternative to promote this fruit consumption. To improve the quality and stability of the powdered fruit, encapsulating and anticaking agents can be added. In the present study, different grapefruit powders obtained by freeze‐drying with the addition of gum arabic (1.27 g per 100 g) and bamboo fibre (0.76 g per 100 g) with and without a pre‐drying microwave treatment were compared with the fresh and freeze‐dried fruit with no carriers added, aiming to evaluate the effect of these preservation processes on phenolics content and on its antioxidant [1,1‐diphenyl‐2‐picrylhydrazyl (DPPH), 2,2′‐azinobis(3‐ethylbenzothiazoline‐6‐sulphonic acid) (ABTS) and ferric reducing ability of plasma (FRAP)] and anti‐inflamatory (evaluated in RAW 264.7 macrophages) capacities. RESULTS: Freeze‐drying and gum arabic and bamboo fibre addition significantly increased total phenolics, as well as the antioxidant and anti‐inflammatory activities (by inhibiting nitric oxide production of lipopolysaccharide activated RAW 264.7 macrophages), of grapefruit. An additional increase in these parameters was obtained with microwave pretreatment before freeze‐drying. CONCLUSIONS: The combined addition of gum arabic and bamboo fibre to grapefruit puree and the application of a microwave pretreatment improve the functional properties of the fruit without showing cytotoxicity in vitro. © 2017 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

Abstract: Two cyclodidepsipeptides, 3-(2-methylpropyl)-6-(propan-2-yl)-4-methyl-morpholine-2,5-dione (1) and 3,6-di(propan-2-yl)-4-methyl-morpholine-2,5-dione (2), were evaluated for inhibitory activity against commercial enzyme xanthine oxidase (XO) in vitro and XO in rat liver homogenate as well as for anti-inflammatory response on human peripheral blood mononuclear cells (PBMCs). Both of cyclodidepsipeptides were excellent inhibitors of XO and significantly suppressed the nuclear factor of κB (NF-κB) activation. Allopurinol, a widely used XO inhibitor and drug to treat gout, relevated stronger inhibitory effect on rat liver XO activity than those of compounds 1 and 2. Molecular docking studies were performed to gain an insight into their binding modes with XO. The studied morpholine-diones derivatives exerting XO inhibition and anti-inflammatory effect may give a promise to be used in the treatment of gout and other excessive uric acid production or inflammatory conditions.
